The regulatory effect of light quality on the photosynthetic apparatus in attached leaves of rice plants was investigated by keeping rice plants under natural light, in complete darkness, or under illumination with light of different colors. When leaves were left in darkness and far-red (FR)-light conditions for 6 days at 30°C, there was an initial lag in chlorophyll (Chl) content, Chl a/b ratio, and maximum photosystem (PS) II photochemistry that lasted until the second day; these then rapidly decreased on the fourth day. In contrast, ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) rapidly disappeared with no lag under low or zero light conditions. By using spectrophotometric quantitation, it was determined that the PSII and PSI reaction centers were regulated by light quality, but cytochrome (Cyt) f was regulated by light intensity. However, the PSII heterogeneity was also strongly modified by the light intensity; PSIIα with the large antenna decreased markedly both in content and in antenna size. Consequently, the PSIIα/PSI ratio declined under FR-light because the low intensity of FR-light dominated over its quality in the modulation of the PSIIα/PSI ratio. An imbalance between them induced the generation of reactive oxygen species (ROS), although the ROS were scavenged by stromal enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX), and glutathione reductase (GR). The activities of these stromal enzymes are also regulated by light quality. Thus, although the photosynthetic apparatus is regulated differently depending on light quality, light quality may play an important role in the regulation of the photosynthetic apparatus.  相似文献

It has been suggested that the function of the chloroplast-localized small heat shock protein (sHsp) is to protect photosystem II (PSII) from heat inactivation. This paper reports that addition of purified sHsp protein to isolated thylakoid membranes gave no protection of PSII and questions that there is any direct effect of the sHsp on PSII. The opinion is forwarded that the primary role for the chloroplast-localized sHsp may not even be protection of PSII.  相似文献

For investigating the protective roles of antioxidative system in desiccation tolerance of Caragana species as they adapt to arid environments, we monitored a variety of ecophysiological parameters in the leaves of Caragana arborescens (mesophyte), C. microphylla (semiarid species), C. roborovskyi, C. stenophylla, C. acanthophylla, and C. tragacanthoides (xerophyte) grown under a drying-rehydration cycle. Relative leaf water content and chlorophyll content were decreased by 17.4?C39.2?%, and by 14?C40?%, respectively, after exposure to 48?days of drought stress. Malondialdehyde did not increase in xeric Caragana species. Hydrogen peroxide concentrations increased by 13.1?C43.9?% except in C. acanthophylla. The activities of superoxide dismutase (SOD), guaiacol peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and reduced glutathione (GSH) in xeric Caragana species were significantly elevated with progressing drought stress. However, catalase in all species decreased markedly before drought stress treatment reached 40?days. The xeric Caragana species showed higher SOD, POD, APX, and GR activities, as well as ascorbate content, and more manganese SOD isoenzymes. C. arborescens and C. microphylla accumulated more free proline. Our data indicate that SOD and POD with the ascorbate?Cglutathione cycle have important protective effects in xeric Caragana species under drought stress. Free proline may be crucial in the resistance of C. arborescens and C. microphylla to drought stress.  相似文献

To explore the significance of the ascorbate–glutathione cycle under drought stress, the leaves of 2-year-old potted apple (Malus domestica Borkh.) plants were used to investigate the changes of each component of the ascorbate–glutathione cycle as well as the gene expression of dehydroascorbate reductase (DHAR, EC 1.8.5.1), ascorbate peroxidase (APX, EC 1.11.1.11) and glutathione reductase (GR, EC 1.6.4.2) under drought stress. The results showed that the malondialdehyde (MDA) and H2O2 concentrations in apple leaves increased during drought stress and began to decrease after re-watering. The contents of total ascorbate, reduced ascorbic acid (AsA), total glutathione and glutathione (GSH) were obviously upregulated in apple leaves when the soil water content was 40–45%. With further increase of the drought level, the contents of the antioxidants and especially redox state of AsA and GSH declined. However, levels of them increased again after re-watering. Moreover, drought stress induced significant increase of the activities of enzymes such as APX, scavenging H2O2, and also of monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), DHAR and GR used to regenerate AsA and GSH, especially when the soil water content was above 40–45%. During severe drought stress, activities of the enzymes were decreased and after re-watering increased again. Gene expression of cytoplasmic DHAR, cytoplasmic APX and cytoplasmic GR showed similar changes as the enzyme activities, respectively. The results suggest that the ascorbate–glutathione cycle is up-regulated in response to drought stress, but cannot be regulated at severe drought stress conditions.  相似文献

Woody tissue photosynthesis might play a key role in maintaining plant carbon economy and hydraulic function under unfavourable conditions such as drought stress.

Abstract

Within trees, a portion of respired CO2 is assimilated by bark and woody tissue photosynthesis, but its physiological role remains unclear, in particular under unfavour able conditions like drought stress. We hypothesised that woody tissue photosynthesis will contribute to overall tree carbon gain both under sufficient water supply and during drought, and plays a role in maintaining the hydraulic function. We subjected half of the trees to a stem and branch light-exclusion treatment to prevent bark and woody tissue photosynthesis. Then, we measured leaf gas exchange and stem growth in Populus deltoides x nigra ‘Monviso’ trees both under well-watered and dry conditions. We additionally measured cavitation using acoustic emission in detached control and light-excluded branches to illustrate the role of woody tissue photosynthesis in xylem embolism repair. Under well-watered conditions, light exclusion resulted in reduced stem growth relative to control trees by 30 %. In response to drought, stem shrinkage of light-excluded trees was more pronounced as compared to control trees. During drought stress also maximum photosynthesis and transpiration rate tended to decrease more rapidly in light-excluded trees compared to control trees. Leaf fall in light-excluded branches together with the larger number of acoustic emissions in control branches indicates that in the latter more xylem vessels were still hydraulically functional under drought. Therefore, our study highlights that photosynthesis at branch and stem level might be a key factor in the resilience of trees to drought stress by maintaining both the plant carbon economy and hydraulic function.

We compared the effect of photoinhibition by excess photosynthetically active radiation (PAR), UV-B irradiation combined with PAR, low temperature stress and paraquat treatment on photosystem (PS) II. Although the experimental conditions ensured that the four studied stress conditions resulted in approximately the same extent of PS II inactivation, they clearly followed different molecular mechanisms. Our results show that singlet oxygen production in inactivated PS II reaction centres is a unique characteristic of photoinhibition by excess PAR. Neither the accumulation of inactive PS II reaction centres (as in UV-B or chilling stress), nor photo-oxidative damage of PS II (as in paraquat stress) is able to produce the special oxidizing conditions characteristic of acceptor-side-induced photoinhibition.  相似文献

Osmotically disrupted chloroplasts catalyze a rapid, light and AMP and ATP dependent 32Pi incorporation into ATP. Light does not stimulate [14C] AMP incorporation into ATP in this system. AMP in the presence of Pi inhibits electron flow in a manner analogous to ADP inhibition in the absence of Pi. The inhibition of AMP + Pi is reversed on addition of ADP.  相似文献

African animal trypanosomosis is a major pathological constraint to cattle breeding across 10 million km2 of sub-Saharan West African countries infested by tsetse flies, their cyclic vectors. The release of sterile males (sterile insect technique [SIT]) is a potentially important control technique aimed at eliminating the vectors. Prior to release, tsetse are generally treated with isometamidium chloride, a trypanocide, to prevent them from transmitting parasites. The present study investigated the preventive action of isometamidium chloride (0.5 mg/L) on the subsequent susceptibility of tsetse released into the wild. A total of 1755 Glossina palpalis gambiensis Vanderplank and 744 Glossina tachinoides Westwood were released, of which 50 and 48, respectively, were recaptured 22-43 days after release. Their probosces were analysed by polymerase chain reaction to identify mature infections with three trypanosome species (Trypanosoma vivax, Trypanosoma brucei sensu lato and Trypanosoma congolense savannah type). Two mature infections with T. vivax and four with T. congolense were detected, indicating that the use of this treatment regimen in an SIT campaign would not totally prevent sterile males from transmitting trypanosomes.  相似文献

The control of reactive oxygen species (ROS) and the stability of photosynthetic pigments under stress conditions are hypothesized to contribute to drought tolerance. Here we studied how ascorbic peroxidase (APX), superoxide dismutase (SOD), catalase (CAT) isozyme activities and chlorophyll a, b (Chl a, b) and carotenoids (Car) contents responded to water stress and whether they related to presence of a terminal drought tolerance QTL in pearl millet. We used PRLT2/89-33 (QTL donor), H77/833-2 (sensitive), and near-isogenic lines (QTL-NILs) introgressed with the QTL in H77/833-2 background. Under water stress there was no significant change in the total APX activity; only the proportional APX5 activity increased, with higher band intensity in tolerant genotypes. There were no significant changes in total activities of CAT and SOD under water stress, with similar band intensities in all genotypes, and a new CAT isozyme was induced in all genotypes. The photosynthetic pigment content decreased under water stress, although not differently in any genotype. Under water stress, the activities of most APX, CAT and SOD isozymes were closely related to the total chlorophyll/carotenoids ratio. Overall, besides APX5, water stress did not lead to major changes in the profile of isoenzymes involved in ROS scavenging. Similarly, the pigment content under stress did not discriminate genotypes according to the presence/absence of the QTL. This absence of discrimination for the ROS scavenging enzymes and for the pigment content under stress suggests that these traits may not play a key role in terminal drought tolerance in pearl millet.  相似文献

In cell-free systems, stress-resistant proteins nonspecifically stabilize stress-susceptible proteins. This mechanism has been suggested to contribute to thermotolerance in cells (Minton et al.: Proc. Natl. Acad. Sci. USA, 79: 7107-7117, 1982). To test this hypothesis, red-blood-cell-mediated microinjection was used to transfer macromolecules into monolayers of CHO cells. We introduced the heat-stable proteins fetuin and ovomucoid into RBCs during hypotonic hemolysis and then fused the RBCs to CHO cells with polyethylene glycol as fusogen. Fetuin and ovomucoid were successfully transferred into 36-55% of the CHO cells as demonstrated by fluorescence of FITC-conjugated proteins. The plating efficiency of these CHO cells after fusion ranged from 35% to 60%. Three hours after fusion, CHO cells microinjected with fetuin or ovomucoid were exposed to 43 degrees C for 0-180 min or 45 degrees C for 0-40 min, and thermal survival was determined. There was no difference in cell survival between control untreated cells, control cells fused with nonloaded RBCs, and cells fused with RBCs loaded with fetuin or ovomucoid. While our results do not support the hypothesis that heat-stable proteins nonspecifically protect cells from thermal stress, several possible explanations are provided for this observation.  相似文献

The hemoflagellate protozoan parasite Trypanosoma cruzi is the causative agent of Chagas disease, a progressive fatal cardiomyopathy widespread in South and Central America. Here, we postulate that the preferential colonization of cardiomyocytes by T. cruzi may reflect the role of myoglobin (Mb) as a nitric oxide (NO) scavenger, protecting the parasite from the trypanocidal effects of NO. The proposal of this novel function of Mb is based on knowledge that ferrous oxygenated Mb reacts rapidly and irreversibly with NO yielding nitrate and ferric oxidized Mb, which is reduced back to the physiologically active form by an intracellular reductase. The postulated protective role of Mb on the viability of T. cruzi is reminiscent of that postulated for hemoglobin in protecting intraerythrocytic Plasmodia from the parasiticidal effect of NO.  相似文献

The water potential at turgor loss point (Ψtlp) has been suggested as a key functional trait for determining plant drought tolerance, because of its close relationship with stomatal closure. Ψtlp may indicate drought tolerance as plants, which maintain gas exchange at lower midday water potentials as soil water availability declines also have lower Ψtlp. We evaluated 17 species from seasonally dry habitats, representing a range of life‐forms, under well‐watered and drought conditions, to determine how Ψtlp relates to stomatal sensitivity (pre‐dawn water potential at stomatal closure: Ψgs0) and drought strategy (degree of isohydry or anisohydry; ΔΨMD between well‐watered conditions and stomatal closure). Although Ψgs0 was related to Ψtlp, Ψgs0 was better related to drought strategy (ΔΨMD). Drought avoiders (isohydric) closed stomata at water potentials higher than their Ψtlp; whereas, drought tolerant (anisohydric) species maintained stomatal conductance at lower water potentials than their Ψtlp and were more dehydration tolerant. There was no significant relationship between Ψtlp and ΔΨMD. While Ψtlp has been related to biome water availability, we found that Ψtlp did not relate strongly to stomatal closure or drought strategy, for either drought avoiders or tolerators. We therefore suggest caution in using Ψtlp to predict vulnerability to drought.  相似文献

Photosynthesis depends on the diffusion of gaseous CO(2) inside the leaf spaces from the stomatal entry point to the mesophyll cell walls. Although most research considers only the vertical diffusion from stomata on upper and/or leaf lower surfaces, some of the gas will diffuse in the lateral (paradermal) direction. The importance of lateral CO(2) diffusion is reviewed, and the anatomical characteristics of leaves, including the variation of air space volume between species and conditions are discussed. The contribution of the air space conductance to the limitation of photosynthesis by the overall CO(2) diffusion pathway is usually ignored. However, the need to consider three-dimensional diffusion at the small scale of a few stomata is emphasized because stomata are discrete, and separated by 20-300 microm. At the large scale of 100s of micrometres, there may be barriers to CO(2) caused by the vascular tissue, particularly if there are bundle sheath extensions. The possible extent and controls on CO(2) lateral and vertical diffusion in different species and conditions are illustrated using chlorophyll a fluorescence imaging techniques. It is clear that there is a range of effective lateral permeabilities depending on the particular vascular patterns and cell arrangements, and that species cannot be simply divided into homobaric and heterobaric anatomies. Lateral diffusion in more permeable leaves can be sufficient to affect measurements of leaf gas exchange, particularly when fluxes are low, although its contribution to leaf photosynthesis in natural conditions needs clarification.  相似文献

The effect of light on the rate of ethylene production was monitored using two different techniques—leaf segments incubated in closed flasks versus intact plants in a flow-through open system. Three different plants were used, viz sunflower (Helianthus annuus), tomato (Lycopersicon esculentum), and soybean (Glycine max). Experiments were conducted both in the presence and absence of 1-aminocyclopropane-1-carboxylic acid (ACC).

The results obtained indicate that, in all three species studied, light strongly inhibits ethylene production when cut leaf segments are incubated in the presence of ACC in closed flasks. When ethylene measurements are made with ACC-sprayed intact plants using a continuous flow system, the effect of light on ethylene production is only marginal. In leaf segments of sunflower and soybean incubated only in distilled H2O in closed flasks, light promotes ethylene production. In tomato, there is no difference between the rate of ethylene production between light and darkness under such conditions. When measurements are made with intact plants in a continuous flow system, the rate of ethylene production is almost identical in light and darkness, in the three plants studied.

It is concluded that the effect of light on cut leaf segments incubated in the presence of ACC in closed flasks can be attributed to the techniques used for these measurements. Light has little effect on ethylene production by intact plants in an open system.

There is a strong correlation between leaf thickness and the light‐saturated rate of photosynthesis per unit leaf area (Pmax). However, when leaves are exposed to higher light intensities after maturation, Pmax often increases without increasing leaf thickness. To elucidate the mechanism with which mature leaves increase Pmax, the change in anatomical and physiological characteristics of mature leaves of Chenopodium album, which was transferred from low to high light condition, were examined. When compared with leaves subjected to low light continuously (LL leaves), the leaves transferred from low to high light (LH leaves) significantly increased Pmax. The transfer also increased the area of chloroplasts facing the intercellular space (Sc) and maintained a strong correlation between Pmax and Sc. The mesophyll cells of LL leaves had open spaces along cell walls where chloroplasts were absent, which enabled the leaves to increase Pmax when they were exposed to high light (LH). However, the LH leaves were not thick enough to allow further increase in Pmax to the level in HH leaves. Thus leaf thickness determines an upper limit of Pmax of leaves subjected to a change from low to high light conditions. Shade leaves would only increase Pmax when they have open space to accommodate chloroplasts which elongate after light conditions improve.  相似文献

Toxoplasma gondii is a Apicomplexa obligate intracellular protozoan parasite that infects up to a third of the world's population. In most humans infected with T. gondii, the disease toxoplasmosis is asymptomatic. However, T. gondii causes blindness, severe neurological disorders, hepatitis, and pneumonia in immunocompromised patients, and severe damage to the fetus. Here, we postulate that the colonization of the retina, heart, and skeletal muscle by T. gondii may reflect the role of neuroglobin (Ngb) and myoglobin (Mb) to protect the parasite from the toxoplasmacidal effects of nitric oxide (NO). This is based on the knowledge that Ngb and Mb catalyzes NO oxidation yielding the harmless nitrate. The postulated protective role of Ngb and Mb on the viability of T. gondii is reminiscent of that postulated for hemoglobin (Hb) and Mb in protecting intraerythrocytic Plasmodia and T. cruzi in cardiomyocytes, respectively, from the parasiticidal effect of NO. Therefore, undesirable pathogen protection by pseudo-enzymatic NO scavenging may represent a new unexpected function of members of the Hb superfamily.  相似文献

7-day soil drought followed by 7-day rehydration was applied to potted German chamomile (Chamomilla recutita) plants at the beginning of their generative stage. Plants of a wild type (WT), plus two diploid (2n) and two tetraploid (4n) genotypes were studied, in order to examine the alterations in chlorophyll (Chl) and carotenoids (Car) contents, and chlorophyll fluorescence (CF) parameters during water shortage and rehydration.
