Fluorescent antibody staining indicated differences in surface antigenicity in Anabaena azollae cells fresh from the leaf cavities of the fern, Azolla caroliniana, and algae which were isolated and subcultured from this fern. Coryneform bacteria were found associated with the nitrogen fixing blue-green alga, Anabaena azollae in the leaf cavity of Azolla caroliniana. Plate counts indicated ca. 7,400±1,900 bacterial cells per mature leaf cavity or approximately 1 bacterial cell for every algal cell. No other type of bacterium was found in these cavities. 相似文献

Summary The development of spontaneous nodules, formed in the absence ofRhizobium and combined nitrogen, on alfalfa (Medicago sativa L. cv. Vernal) was investigated at the light and electron microscopic level and compared to that ofRhizobium-induced normal nodules. Spontaneous nodules were initiated from cortical cell divisions in the inner cortex next to the endodermis, i.e., the site of normal nodule development. These nodules, on uninoculated roots, were white multilobed structures, histologically composed of nodule meristems, cortex, endodermis, central zone and vascular strands. Nodules were devoid of intercellular or intracellular bacteria confirming microbiological tests. Early development of spontaneous nodules was initiated by series of anticlinal followed by periclinal divisions of dedifferentiated cells in the inner cortex of the root. These cells formed the nodular meristem from which the nodule developed. The cells in the nodule meristems divided unequally and differentiated into two distinct cell types, one larger type being filled with numerous membrane-bound starch grains, and the other smaller type with very few starch grains. There were no infection threads or bacteria in the spontaneous nodules at any stage of development. This size differentiation is suggestive of the different cell sizes seen inRhizobium-induced nodules, where the larger cell type harbours the invading bacteria and the smaller type is essential in supportive metabolic roles. The ontogenic studies further support the claim that these structures are nodules rather than aberrant lateral roots, and that plant possess all the genetic information needed to develop a nodule with distinct cell types. Summary The development of external glands on traps and stolons ofU. monanthos has been studied using transmission electron microscopy. During early differentiation of the epidermis some cells remain narrow and develop a protuberance which subsequently divides into a terminal and a pedestal cell, with the remainder of the original cell forming the basal epidermal cell of the gland. The lateral wall of the pedestal cell soon becomes densely impregnated throughout its thickness, and this is followed by the formation of discontinuous cuticular deposits within the primary wall of the terminal cell. The outer wall of the terminal cell then usually undergoes extensive secondary wall thickening beginning with the formation of ingrowths which for a period characterize the cell as a transfer cell. Later, at the stage when traps begin capturing prey, these ingrowths are overlain by further layers of secondary wall material. Concomitantly, in the pedestal cell, wall ingrowths become fully differentiated on the outer transverse wall and persist throughout the remaining life of the gland.The function of external glands during early ontogeny is discussed. At the stage when the terminal cell is differentiated as a transfer cell it is suggested that the gland is mainly responsible for absorbing solutes from the external medium. Once traps commence capturing prey the gland may become modified for a rôle in water secretion, facilitated by the differentiation of the pedestal cell as a transfer cell, and by the formation of a thick outer wall in the terminal cell. 相似文献

Random and site-directed Tn5-induced mutagenesis of Bradyrhizobium japonicum yielded two mutations, one in strain 2960 and the other in strain 2606::Tn5-20, which mapped close to each other but in separate genes. The corresponding wild-type genes were cloned, and their approximate location on the cloned DNA was determined. Mutant 2960 was Fix- and formed green nodules on soybean, whereas strain 2606::Tn5-20 had ca. 4% of wild-type Fix activity and formed white nodules. Cytochrome oxidase assays (Nadi tests) showed a negative reaction with both mutants, indicating a functional deficiency of cytochrome c or its terminal oxidase or both. However, the mutants grew well under aerobic conditions on minimal media with different carbon sources. Furthermore, mutant 2960 had a reduced activity in hydrogen uptake, was unable to grow anaerobically with nitrate as the terminal electron acceptor and 2960-infected soybean nodules contained little, if any, functional leghemoglobin. Southern blot analysis showed that a B. japonicum heme biosynthesis mutant [strain LO505: O'Brian MR, Kirshbom PM, Maier RJ (1987) Proc Natl Acad Sci USA 84: 8390–8393] had its mutation close to the Tn5 insertion site of our mutant 2606::Tn5-20. This finding, combined with the observed phenotypes, suggested that the genes affected in mutants 2960 and 2606::Tn5-20 were involved in some steps of heme biosynthesis thus explaining the pleiotropic respiratory deficiencies of the mutants. Similar to strain LO505, the mutant 2606::Tn5-20 (but not 2960) was defective in the activity of protoporphyrinogen IX oxidase which catalyzes the penultimate step in the heme biosynthesis pathway. The objective of the present study was to elucidate whether remobilized N from lower leaves is involved in causing the drop in N(2) fixation during pod-filling in common bean (Phaseolus vulgaris L). Moreover, we addressed the question of whether remobilized N from lower leaves would reach the nodules. Nodulated common bean plants were grown in a growth chamber in quartz sand. During a 2-week period, at vegetative and at reproductive growth, 50% of the leaves (lower part) were either excised or individually darkened, thereby removing the same photosynthetic capacity yet allowing N to be remobilized from the darkened leaves. Moreover, at the vegetative growth period, three lower leaves per plants were (15)N labelled by applying (15)NH(4)NO(3) prior to imposing the darkening treatment. Leaf darkening at vegetative growth induced N remobilization as well as reduced N(2)-fixation rates and growth. Leaf excision at reproductive growth enhanced N(2) fixation. Changes in N(2)-fixation rates were in all cases the result of altered growth rates, while the % N in the whole plant and in various plant parts remained conserved. Directly after leaf labelling, but also at the end of the vegetative growth period, substantial amounts of (15)N from the leaves could be recovered in nodules in the control, and in higher amounts in the leaf-darkening treatment. It is proposed that nitrogen from leaves circulates within the plant via nodules, and that the strength or composition of this circular flow may be the signal for a putative N-feedback effect. 相似文献

Formate metabolism supported nitrogen-fixation activity in free-living cultures of Rhizobium japonicum. However, formate0dependent nitrogense activity was observed only in the presence of carbon sources such as glutamate, ribose or aspartate which by themselves were unable to support nitrogenase activity. Formate-dependent nitrogenase activity was not detected in the presence of carbon sources such as malate, gluconate or glycerol which by themselves supported nitrogenase activity. A mutant strain of R. japonicum was isolated that was unable to utilise formate and was shown to lack formate dehydrogenase activity. This mutant strain exhibited no formate-dependent nitrogenase activity. Both the wild-type and mutant strains nodulated soybean plants effectively and there were no significant differences in the plant dry weight or total nitrogen content of the respective plants. Furthermore pea bacteroids lacked formate dehydrogenase activity and exogenously added formate had no stimulatory effect on the endogenous oxygen uptake rate. The role of formate metabolism in symbiotic nitrogen fixation is discussed.Abbreviation FDH

The hemocytes of different types encountered in the diamondback moth Plutella xylostella larvae of each instar and the development of the differential hemocytes counts were herein presented. Hemocytes classes/populations characterized based on their affinity with fluorescent dye (acridine orange) and ultrastructural differences comprised the prohemcoytes (<10–16%), plasmatocytes (22–65%), granulocytes (25–72%), oenocytoids (<1–9%), and spherulocytes (<1%). Prohemcoytes were the smallest cells with a comparatively tremendous nucleus. Plasmatocytes and granulocytes occupied the main proportion of total cell numbers. Oenocytoids were in a most stable presence, i.e. rotund in a diameter of 10 μm and with a nucleus deviated from the central location; however, sometimes with two nuclei which were adjoining with each other. Spherulocytes were rare and only could be observed occasionally. Ultrastructural investigation revealed that hemocytes in the diamondback moth larvae were of the typical model as in the Lepidoptera insect larvae. It is interesting to find that the cell which could phagocytize bacteria in vitro was granulocyte, not the other types of hemocytes, although plasmatocyte was usually declared to participate in this reaction in various previous studies. 相似文献

Incubation in the dark of photoautotrophically grown N2-fixing heterocystous cyanobacteria leads to a loss of nitrogenase activity. Original levels of nitrogenase activity are rapidly regained upon re-illumination of the filaments, in a process dependent on de novo protein synthesis. Ammonia, acting indirectly through some of its metabolic derivatives, inhibits the light-promoted development of nitrogenase activity in filaments of Anabaena sp. ATCC 33047 and several other cyanobacteria containing mature heterocysts. The ammonia-mediated control system is also operative in N2-fixing filaments in the absence of any added source of combined nitrogen, with the ammonia resulting from N2-fixation already partially inhibiting full expression of nitrogenase. High nitrogenase levels, about two-fold higher than those in normal N2-fixing Anabaena sp. ATCC 33047, are found in cell suspensions which have been treated with the glutamine synthetase inhibitor l-methionine-d,l-sulfoximine or subjected to nitrogen starvation. Filaments treated in either way are insensitive to the ammonia-promoted inhibition of nitrogenase development, although this insensitivity is only transitory for the nitrogen-starved filaments, which become ammonia-sensitive once they regain their normal nitrogen status.Abbreviations Chl

Tillers of Festuca arundinacea Schreb. were subjected to-8°C in a bath of methylated spirits for three-quarters of an hour. They were thawed at room temperature and some material taken from the shoot apical meristem and leaf blade for electron microscopy. Similar material was taken from control plants for electron microscopy. Nine tillers subjected to-8°C and thawed subsequently failed to regrow. Nine control tillers regrew. All the treated meristem cells and about half the treated leaf mesophyll cells were extensively altered. Their nuclei were contracted, organelles were swollen or partly disrupted, plasmalemma and nuclear membranes were broken or absent and vacuoles were sometimes disrupted. Strongly osmiophilic material accumulated in the vicinity of membranes. About half the leaf mesophyll cells differed from the control mesophyll cells only in having more spherosomes and narrower thylakoids. Parallels with other ultrastructural studies of stress damage and the indications the results give of possible primary damaging events are discussed.Abbreviations ER

Metabolite levels have been compared in the dark and during photosynthesis in leaves and protoplasts from spinach, pea, wheat and barley. In protoplasts the subcellular distribution was also studied. The levels of triose phosphates and sugar bisphosphates were high in the light and low in the dark. The hexose phosphates and 3-phosphoglycerate levels in the dark were very variable depending on the plant material. In most conditions, hexose phosphates and triose phosphates were mainly in the extrachloroplast compartment, while 3-phosphoglycerate and the sugar bisphosphates were mainly in the chloroplast compartment. Leaves always had a very low triose phosphate: 3-phosphoglycerate ratio in the dark, but in protoplasts this ratio was higher. Detailed studies with spinach showed that metabolite levels were very dependent on the availability of carbohydrate in the leaf, particularly starch. Starch mobilisation is not controlled just by the availability of inorganic phosphate and accumulation of phosphorylated intermediates. Hydrolysis of starch may provide precursors for sucrose synthesis while phosphorolysis leads to provision of substrates for respiration. Starch breakdown generates high enough levels of hexose phosphate to support substantial rates of sucrose synthesis in the dark. Respiration is not greatly increased when metabolite levels are high during starch mobilisation. Higher levels of metabolites shorten the length of the induction phase of photosynthesis.Abbreviations Chl

We used a split-root system to determine the timing for induction of the autoregulation of nodulation (AUT) in Lotus japonicus (Regel) Larsen after inoculation with Mesorhizobium loti. The signal took at least five days for full induction of AUT and inhibition of infection thread formation. Strain ML108
(able to nodulate but unable to fix nitrogen) induced full AUT, but ML101 (unable to nodulate or to fix nitrogen) did not
induce autoregulation. These results indicate that Nod factor-producing strains induce AUT, but that the nitrogen fixed by

Summary Southern blots of Frankia total DNAs were hybridized with nifHDK probes from Rhizobium meliloti, Klebsiella pneumoniae and Frankia strain Arl3. Differences between strains were noted in the size of the hybridizing restriction fragments. These differences were more pronounced among Elaeagnus-compatible strains than among Alnus- or Casuarina-compatible strains. Gene banks constructed for Frankia strains EUN1f, HRN18a, CeD and ACoN24d were used to isolate nif-hybridizing restriction fragments for subsequent mapping and comparisons. The nifH zone had the highest sequence conservation and the nifH and nifD genes were found to be contiguous. The complete nucleotide sequence of the nifH open reading frame (ORF) from Frankia strain Arl3 is 861 bp in length and encodes a polypeptide of 287 amino acids. Comparisons of these nucleic acid and amino acid sequences with other published nifH sequences suggest that Frankia is most similar to Anabaena and Azotobacter spp. and K. pneunoniae and least similar to the Gram-positive Clostridium pasteurianum and to the archaebacterium Methanococcus voltae. 相似文献

Summary Leaves of two field growing co-occuring perennial shrubs (drought-deciduous Diplacus aurantiacus and the evergreen Heteromeles arbutifolia) from the Californian chaparral were exposed to small doses of SO2. During this exposure the leaf environment was manipulated to determine how the presence of SO2 alters the response of gas exchange to other environmental stresses. The data show that no direct changes in stomatal conductance (g) or net assimilation rate (A) could be attributed to short-term (7 h) SO2 (4.2 mol m-3, 0.1 l l-1) exposure. D. aurantiacus leaves possessed features which demonstrate that they were sensitive to changes in environment e.g. light flux and atmospheric relative humidity. The interspecific differences in stomatal sensitivity to water vapour were extremely important, as relative humidity is a major factor influencing carbon fixation and the rate of pollutant absorption. Conditions of high relative humidity and high xylem water potentials are suggested to pre-dispose leaves of D. aurantiacus to greater pollutant doses than the more stomatally-conservative evergreen, H. arbutifolia. In the presence of SO2 there was some indication of increased g for both D. aurantiacus and H. arbutifolia as W became smaller. This SO2-effect was only obvious as increasing atmospheric humidity induced further stomatal opening. The important consequences of an SO2 enhanced g, were a reduction in WUE, which may cause earlier leaf abscission and a concomitant decline in productivity.Abbreviations

Summary The role ofFrankia vesicle envelope lipids in regulating oxygen diffusion of symbiotic nitrogen fixation inAlnus incana was examined. Total lipids of symbioticFrankia (vesicle clusters) that had been adapted to oxygen tensions of 5,21, or 40 kPa were analyzed with a normal phase HPLC system. During the oxygen treatment, nitrogenase activity was measured as hydrogen evolution in an open flow-through system. When plants were transferred to low oxygen (5 kPa) or high oxygen (40 kPa), nitrogenase activity dropped initially. Activity recovered in both treatments with a rate comparable to the controls (21 kPa O2). Both lipid content and lipid composition of vesicle clusters were affected by the oxygen treatments. With increasing oxygen tension, the vesicle cluster lipid content increased. This correlated with structural data (fluorescence microscopy and TEM) which showed a thicker vesicle envelope at higher oxygen tension. Three hopanoid lipids, bacteriohopanetetrol (bht) and two isomers of phenylacetyl monoester of bht, made up approximately 80% of the vesicle cluster lipids. With changing oxygen concentrations, the ratio of the two bht esters changed whereas the relative proportion of bht remained fairly constant. A mathematical model is described which allows the estimation of rates of O2, CO2, N2, and H2 exchange from legume nodules under steady state conditions of N2 fixation. Calculated rates of gas exchange under defined conditions of nodule size, relative growth rate (RGR), specific total nitrogenase activity (TNA), nitrogenase electron allocation coefficient (EAC), uptake-hydrogenase activity (HUP) and nature of the N export product compared favorably with experimentally-obtained rates reported in the literature. Therefore the model was used to predict the effects of varying each of these nodule characteristics on the rates of gas exchange, and on the apparent respiratory cost (CO2/NH3) and sucrose cost (sucrose consumed/NH3) of N2 fixation.The model predicted that, all other characters being equal, ureide-producing nodules would consume 8% less sucrose per N fixed than asparagine-producing nodules, but would display an apparent respiratory cost which would be 5% higher than that in asparagine-producing nodules. In both ureide-producing and asparagine-producing nodules, the major factor affecting the apparent respiratory cost of N2 fixation was predicted to be EAC, followed by TNA, nodule RGR and nodule size. The relative importance of HUP in improving the apparent respiratory cost of N2 fixation was predicted to be largely dependent upon its potential role in the regulation of EAC.

A Rhizobium trifolii symbiotic plasmid specific gene library was constructed and the physical organisation of regions homologous to nifHDK, nifA and nod genes was determined. These symbiotic gene regions were localised to u 25 kb region on the sym-plasmid, pPN1. In addition four copies of a reiterated sequence were identified on this plasmid, with one copy adjacent to nifH. No rearrangement of these reiterated sequences was observed between R. trifolii bacterial and bacteroid DNA. Analysis of a deletion derivative of pPN1 showed that these sequences were spread over a 110 kb region to the left of nifA. 相似文献

Disposal of sewage water in cultivated soils often containing considerable amount of potentially toxic metals such as Cu, Zn, Ni, Cd, Pb and Cr can be beneficial or harmful to plant growth, rhizobial survival, nodulation and nitrogen fixation. Soil samples from 14 such locations were collected. Symbiotic effectivity of host-Rhizobium leguminosarum symbiosis in these soils was assessed. The total metal contents of Cd, Cu, Zn and Ni in all the 14 samples collected from farmer's fields receiving sewage water ranged between 1.3 and 6.7, 55.8-353.2, 356.0-1028.0 and 90.0-199.7 mg kg(-1) of soil, respectively. In Rohtak 1 soil, levels of Cd, Cu and Zn were highest while Ni was highest in Sonipat 2 soil. The content of available Cd, Cu, Zn and Ni in these soils ranged from 1.0-29.3; 6.2-47.0; 2.4-13.5, respectively, and was 2-9 percent of their total metal contents. All the N2 fixing parameters in pea and Egyptian clover were adversely affected by the presence of heavy metals. Available Cd and Cu contents significantly affected the N contents of pea and Egyptian clover plants, whereas Ni contents were negatively correlated with the plant biomass of pea and Egyptian clover. 相似文献
