Inhibition of tumour vascular growth, destruction of the tumour associated vasculature (TAV), and manipulation of the endothelial lining of the TAV provide powerful tools for anti-tumour therapy. We previously demonstrated that addition of TNF to chemotherapy improved tumour response. The major effect of TNF is an increased permeability of the tumour vascular bed resulting in augmented accumulation of co-administered drug in the tumour. As the TAV is recognised as a major candidate in tumour therapy it is becoming important to understand anti-vascular effects better. In our laboratory we examine the effect of immunotherapy on the TAV, and the effect of anti tumour-vascular therapy on tumours. This is studied in animal models, which exhibit similarities with the clinical setting, such as tumour perfusion treatment.  相似文献

Improving cell-material interactions is a major goal in tissue engineering. In this regard, functionalization of biomaterials with cell instructive molecules from the extracellular matrix stands out as a powerful strategy to enhance their bioactivity and achieve optimal tissue integration. However, current functionalization strategies, like the use of native full-length proteins, are associated with drawbacks, thus urging the need of developing new methodologies. In this regard, the use of synthetic peptides encompassing specific bioactive regions of proteins represents a promising alternative. In particular, the combination of peptide sequences with complementary or synergistic effects makes it possible to address more than one biological target at the biomaterial surface. In this review, an overview of the main strategies using peptides to install multifunctionality on biomaterials is presented, mostly focusing on the combination of the RGD motif with other peptides sequences. The evolution of these approaches, starting from simple methods, like using peptide mixtures, to more advanced systems of peptide presentation, with very well defined chemical properties, are explained. For each system of peptide's presentation, three main aspects of multifunctionality—improving receptor selectivity, mimicking the extracellular matrix and preventing bacterial colonization while improving cell adhesion—are highlighted.  相似文献

Minimum counts are commonly used to estimate population size and trend for wildlife conservation and management; however, the scope of inference based on such data is limited by untestable assumptions regarding the detection process. Alternative approaches, such as distance sampling, occupancy surveys, and repeated counts, can be employed to produce detection-corrected estimates of population parameters. Unfortunately, these approaches can be more complicated and costly to implement, potentially limiting their use. We explored a conceptual framework linking datasets collected at different spatial scales under different survey designs, with the goal of improving inference. Specifically, we link landscape-scale distance sampling surveys with local-scale minimum counts in an integrated modeling framework to estimate mountain goat (Oreamnos americanus) abundance at both the local and regional scale in south-central Alaska, USA, and provide an estimate of detection probability (i.e., sightability) for the minimum counts. Estimated sightability for the minimum count surveys was 0.67 (95% credible interval [CrI] = 0.52–0.83) and abundance for the entire area was 5,600 goats (CV = 9%), both in broad agreement with estimates from previous studies. Abundance estimates at the local scale (i.e., individual min. count unit) were reasonably precise ( = 18%), suggesting the integrated approach can increase the amount of information produced at both spatial scales by linking minimum count approaches with more rigorous survey designs. We propose that our integrated approach may be implemented in the context of a modified split-panel monitoring design by altering survey protocols to include frequent minimum counts within local count units and intermittent but more rigorous survey designs with inference to the entire study area or population of interest. Doing so would provide estimates of abundance with appropriate measures of uncertainty at multiple spatial scales, thereby improving inference for population monitoring and management. © 2019 The Wildlife Society.  相似文献

Autophagy is well-known as an internal catabolic process that is evolutionarily conserved and performs the key biological function in maintaining cellular homeostasis. It is tightly controlled by several autophagy-related (ATG) proteins, which are closely associated with many types of human cancers. However, what has remained controversial is the janus roles of autophagy in cancer progression. Interestingly, the biological function of long non-coding RNAs (lncRNAs) in autophagy has been gradually understood in different types of human cancers. More recently, numerous studies have demonstrated that several lncRNAs may regulate some ATG proteins and autophagy-related signaling pathways to either activate or inhibit the autophagic process in cancer. Thus, in this review, we summarize the latest advance in the knowledge of the complicated relationships between lncRNAs and autophagy in cancer. Also, the in-depth dissection of the lncRNAs-autophagy-cancers axis involved in this review would shed new light on discovery of more potential cancer biomarkers and therapeutic targets in the future.  相似文献
