Using indirect immunofluorescence assay, we examined the sera of 561 patients from November 1984 to February 2005 to determine the incidence of tsutsugamushi disease (scrub typhus) in Oita Prefecture, Japan. The results obtained were positive in 384 individuals (68.4%). Municipalities where patients were presumed to have been infected with Orientia tsutsugamushi were Taketa City (41.7%), Oyama Town (13.5%), and Ogi Town (8.3%). Infections occurred most often in October, November, and December. A small number of cases occurred from January to May. The serotypes Kuroki (47.5%), Kawasaki (42.5%), and Karp (10.0%) were detected by genetic analysis of O. tsutsugamushi DNA extracted from the blood of 120 patients. The gene sequences of the Kuroki type were highly homologous to that of the Nishino strain. The gene sequences of the Kawasaki type were identical to that of the Kawasaki strain. The gene sequence of the Karp type was highly homologous to that of the JP-2 type. To determine the distribution of vector mites, 558 wild rodents were captured and 72010 mites attached to these rodents were collected from 1982 to 1998. Six genera and 16 species of trombiculid mites were collected. Leptotrombidium pallidum and L. scutellare, which are known to be mite vectors for tsutsugamushi disease, accounted for 20.5% and 5.9%, respectively, of all trombiculid mites collected. The geographical distribution of cases roughly coincided with the distribution of L. scutellare. In Oita Prefecture, L. scutellare is presumed to primarily transmit tsutsugamushi disease. In addition, our results also suggest that L. pallidum transmits the Karp type of the causative rickettsia in some municipalities.  相似文献

Orientia tsutsugamushi is the etiological agent of scrub typhus, a mite-borne, febrile illness that occurs in the Asia-Pacific region. We conducted strain characterization of O. tsutsugamushi isolates from chiggers obtained from rodents based the nucleotide sequence of the 56-kDa outer membrane protein gene. With the use of PCR, a total of 68 DNA sequences of 56-kDa antigen genes were amplified. Phylogenetic analysis revealed that there were at least six definable clusters among the 68 isolates: 37% Karp-related strains (25/68), 27% TA763 strains (18/68), 12% JG-related strains (8/68), 19% Kato-related strains (13/68), 4% divergent strains (3/68), and 1% representing a Gilliam prototype strain (1/68). Overall, the O. tsutsugamushi genotypes exhibited a high degree of diversity, similar to that seen in strains from the rest of the areas where scrub typhus is endemic. Moreover, the 56-kDa protein sequence similarity between O. tsutsugamushi isolates from mites and those from human patients (H. Y. Lu et al., Am. J. Trop. Med. Hyg. 83:658-663, 2010) were striking, thus highlighting potential risk factors for this emerging zoonotic disease.  相似文献

Owing to climate change, the global resurgence of vector‐borne infectious diseases has emerged as a critical public health issue. Orientia tsutsugamushi is the etiological agent of tsutsugamushi disease (scrub typhus) a mite‐borne acute febrile disease occurring in the Asia‐Pacific region. We investigated the prevalence of tsutsugamushi disease transmitted by chigger mite vectors living on rodents. Using sticky‐type chigger traps for three months during 2016–2018, 1,057 chigger mites were collected (chigger mite index, 1.31) from four locations in the Hwaseong‐si area of Gyeonggi‐do, Republic of Korea. Five species distributed among three genera were identified. In addition, 94 rodents were captured (collection rate: 7.83%) using Sherman live traps over the course of three months (April, October, and November) during 2016–2017. Three rodent species were captured and identified and the striped field mouse (Apodemus agrarius) was the dominant rodent host species in the surveyed area. A total of 10,469 ectoparasitic chigger mites were recovered from the 94 rodents, from which 13 species distributed among four genera were identified. Of the 5,250 chigger mites examined, Leptotrombidium pallidum was most abundant (n = 2,558), followed by L. orientale, L. scutellare, L. zetum, Euschoengastia koreaensis, L. subintermedium, and Neotrombicula tamiyai. Of the examined chigger mites, no groups recovered from rodent hosts tested positive for O. tsutsugamushi. This study provides fundamental regional information on vector‐borne disease data collection in the Hwaseong‐si area, Gyeonggi‐do, and will further contribute to formulating disease control and prevention strategies.  相似文献

In order to clarify the epidemiological background of the endemic occurrence of tsutsugamushi disease in Toyama Prefecture, Japan, since 1978, comparative surveys have been carried out between endemic and nonendemic areas. Rickettsia tsutsugamushi (Rt) was isolated at a rate of about 36% (158/439) from field rodents in the endemic area while it was not isolated from any of 280 in nonendemic areas. In all of six stations in the endemic area, a significantly high proportion of rodents were found to be Rt carriers. However, no Rt was isolated from rodents captured from July to September. The organism was isolated from rodents captured in the other months, especially in a high proportion in November when infestation of rodents with Leptotrombidium pallidum was at its peak. When the rodents were examined by indirect immunofluorescence staining, the rate of anti-Rt antibody-positive animals was about 55% (157/287) and about 17% (62/368) in endemic and nonendemic areas, respectively. Larvae of mites collected from the rodents were found to belong to four genera and 11 species. Among them L. pallidum was the only mite that had been known to be a vector of Rt. L. pallidum was found most frequently and in abundance from rodents in the endemic area, whereas it was present in very small numbers in rodents in nonendemic areas. The infestation of rodents with L. pallidum showed a seasonal variation, i.e. two peaks per year, in spring and autumn, and the number of mites detected was markedly greater in November than in spring. Rt was isolated from L. pallidum on rodents captured in the endemic area.  相似文献

Incidence of tsutsugamushi disease (scrub typhus) caused by Orientia tsutsugamushi, is steadily increasing. It is a mite-borne disease transmitted by chigger mites. In this study, the chigger mites were collected from field small mammals in Hwaseong-si (city), Gyeonggi-do (province), Korea, 2019 and 2020. The field small mammals captured were 56 Apodemus agrarius (94.9%) and 3 Crocidura lasiura (5.1%). A total of 7,531 chigger mites were collected from the captured small mammals. Using PCR test, 153 chigger mite pools were examined and 17 pools were reported positive for O. tsutsugamushi. The O. tsutsugamushi were identified to 5 strains; Jecheon strain was most prevalent, followed by Boryong strain. The other strains were OI011, Taguchi, and Shimokoshi. Collectively, these results provide essential regional information on mite-borne tsutsugamushi disease in the Hwaseong-si, and further contribute to bring awareness and rapid diagnosis for the tsutsugamushi disease.  相似文献

对山东费县秋冬型恙虫病疫源地恙螨进行了调查并进行恙虫病立克次体(Rt)分离。从352只活鼠体外收集到11 762只恙螨,隶属2属5种,太平洋无前恙螨数量最多,占36.73%;其次是临淮岗纤恙螨(24.04%);小盾纤恙螨(21.65%);须纤恙螨(13.57%)和泰山纤恙螨(3.96%)。小盾纤恙螨出现在9—12月,高峰在11月;须纤恙螨出现在10月一翌年4月,高峰在12月;临淮岗纤恙螨从5月到11月存在,高峰在8月;太平洋无前恙螨出现在4—12月,高峰在7月。从小盾纤恙螨、须纤恙螨、临淮岗纤恙螨及太平洋无前恙螨中共分离到12株Rt,血清分型结果分离株以Gilliam型为主,但存在Karp型Rt。这些结果表明,上述4种恙螨均能自然感染Rt,有在该地区充作不同季节Rt传播媒介的可能。结合以往的研究结果,当地小盾纤恙螨集中出现于发病季节,其消长与当地人群发病基本一致,且能叮刺、经卵传递Rt,从而证实小盾纤恙螨是引起该地区秋冬型恙虫病流行的最重要的媒介。  相似文献

Because no reference on trombiculid mites (Acarina: Trombiculidae) in Cheju Island where tsutsugamushi disease is highly endemic had been available, studies on trombiculid mites in Cheju Island were implemented during the period of August 1991-April 1992, and the results obtained are summarized as follows: (1) The species and numbers of the field rodents collected were 143 Apodemus agrarius chejuensis (92.3%), 11 Crocidura lasiura (7.1%) and 1 Micromys minutus (0.6%). From total 12,075 chiggers harvested, 9 species of 4 genera in Trombiculidae were identified. (2) The predominant species through all seasons was L. zetum (43.3%), followed by L. orientale (27.4%) and L. scutellare (26.6%). However, in autumn when the most cases of tsutsugamushi disease occur, L. scutellare was prominently predominant, having 79.8% of the collected chiggers. (3) Among 1,142 L. scutellare examined for Rickettsia tsutsugamushi by means of IFA test, 6 individuals were found positive showing 0.5% of infection rate. This is the first finding that L. scutellare is the second vector species of tsutsugamushi disease in Korea. (4) Antibody positive rate of A. agrarius chejuensis sera were 31.2% (44/139), and 1 M. minutus serum was also found positive. Abstract  Investigations on trombiculid mites and isolation of Rickettsia tsutsugamushi therefrom were carried out in Feixian County, Shandong Province. A total of 11 762 trombiculid mites, consisting of 5 species from two genera, were collected from 352 rodents (including 247 Apodemus agrarius , 80 Cricetulus triton , 23 Rattus noruegicus , 2 Crocidura suaveolens ), and Walchia pacifica was the most predominant (36.73%), followed by Leptotrombididium linhuaikonense (24.04%), L. scutellare (21.65%), L. palpale (13. 57%), and L. taishanium (3. 96%). L. scutellare was found from September to December with a remarkable peak in November, whereas L. palpale occurred from October to April (the second year) with peak in December. L. linhuaikonense was found from May to November, with peak in August. W. pacifica appeared from April to December with peak in July. R. tsutsugamushi was isolated from L. scutellare, L. palpale, L. linhuaikonense and W. pacifica . The main serotypes of R. tsutsugamushi isolated from the chigger mites were of the Gilliam type, but Karp type also existed in L. linhuaikonense . These results indicate that the surveyed area has a high probability of occurrence of tsutsugamushi disease, and L. scutellare, L. palpale, L. linhuaikonense and W. pacifica may serve as the vectors in this area. It is suggested that L. scutellare is the most important vector which has caused the endemic of this disease in Feixian County.  相似文献

Scrub typhus and tick-borne spotted fever group (SFG) rickettsioses are transmitted by chiggers (larval trombiculid mites) and hard ticks, respectively. We assessed exposure to these disease vectors by extensively sampling both chiggers and ticks and their small mammal hosts in eastern Taiwan during 2007 and 2008. The striped field mouse Apodemus agrarius Pallas (Rodentia: Muridae) was the most common of the small mammals (36.1% of 1393 captures) and presented the highest rate of infestation with both chiggers (47.8% of 110 760) and ticks (78.1% of 1431). Leptotrombidium imphalum Vercammen-Grandjean & Langston (Trombidiformes: Trombiculidae) and immature Rhipicephalus haemaphysaloides Supino (Ixodida: Ixodidae) were the most abundant chiggers (84.5%) and ticks (>99%) identified, respectively. Immunofluorescent antibody assay revealed high seropositive rates of rodents against Orientia tsutsugamushi Hyashi (Rickettsiales: Rickettsiaceae), the aetiological agent of scrub typhus (70.0% of 437 rodents), and tick-borne SFG rickettsiae (91.9% of 418 rodents). The current study represents a first step towards elucidating the potential hosts and vectors in the enzootic transmission of O. tsutsugamushi and tick-borne SFG rickettsiae in Taiwan. Further studies should focus on characterizing pathogens in L. imphalum and R. haemaphysaloides, as well as the proclivity of both vectors to humans. Uncovering the main hosts of adult ticks is also critical for the prevention of SFG rickettsial infections.  相似文献

We performed a detailed electron microscopic observation on the escaping process of Orientia tsutsugamushi from the salivary gland cells of naturally infected trombiculid larvae into the acinar lumen of the gland during feeding on mice. In unfed larvae, many O. tsutsugamushi were intermingled with secretory granules in the cytoplasm of the salivary gland cell. O. tsutsugamushi was neither found in the acinar lumen nor observed escaping from the apical surface of the gland cell. In contrast, in the larvae fed on mice, many O. tsutsugamushi were observable in the acinar lumen. They were enveloped with the host glandular cell membrane. In salivary gland cells, secretory granules changed the distribution and accumulated in the apical region. In such cells, the majority of O. tsutsugamushi were found at the base of the cell. Some O. tsutsugamushi were pushing the glandular cell membrane outward in various degrees, showing different stages of escape. These findings suggest that larval feeding induced O. tsutsugamushi escape from salivary gland cells, that the escape was by budding, during which O. tsutsugamushi were enveloped in the host cell membrane, and that O. tsutsugamushi would be injected into the mouse skin as a mixture with mite saliva. The study also revealed the presence of many small vesicles that had the same cell wall structure as O. tsutsugamushi in the cytoplasm of the salivary gland cell. Most of them seemed to be products from degenerated Orientia.  相似文献

In our attempts to isolate Orientia tsutsugamushi from trombiculid mites, a strain was successfully isolated from Leptotrombidium fuji collected in Aichi Prefecture, Japan. This is the first case of isolation of O. tsutsugamushi from L. fuji. A phylogenetic analysis based on the base-sequence homology of the 56-kDa type-specific antigen-gene indicated that the strain is a new type which is not closely related to any strains analyzed previously. Three strains isolated from Leptotrombidium pallidum harvested at the same area were identified as being closely related to the JP-2 type (subtype-2 of Karp type in Japan) by phylogenetic analysis.  相似文献

Rodents are the natural hosts for Leptotrombidium mites that transmit Orientia tsutsugamushi, the causative agent of scrub typhus, a potentially fatal febrile human disease. Utilizing mite lines that included O. tsutsugamushi infected and non-infected Leptotrombidium species we investigated the varied infection response of outbred mice (ICR) exposed to L. chiangraiensis (Lc), L. imphalum (Li) and L. deliense (Ld). Each of six mite lines (Lc1, Lc5, Li3, Li4, Li7 and Ld) was separately placed in the inner ears of ICR mice either as a single individual (individual feeding, IF) or as a group of 2-4 individuals (pool feeding, PF). The species of infected chigger feeding on mice significantly affected mortality rates of the mice, with mite lines of Lc causing higher mean (±SE) mortality (90.7 ± 3.6 %) than mite lines of Li (62.9 ± 5.6 %) or Ld (53.6 ± 5.8 %). Mouse responses which included time to death, food consumption and total mice weight change depended on mite species and their O. tsutsugamushi genotype, more than on feeding procedure (IF vs. PF) except for mite lines within the Lc. Infected mite lines of Lc were the most virulent infected mites assessed whereas the infected Ld species was the least virulent for the ICR. Mice killed by various mite lines showed enlarged spleens and produced ascites. The results of this investigation of the clinical responses of ICR mice to feeding by various infected mite lines indicated that the different species of infected mites and their O. tsutsugamushi genotype produced different clinical presentations in ICR mice, a scrub typhus mouse model which mimics the natural transmission of O. tsutsugamushi that is critical for understanding scrub typhus disease in terms of natural transmission, host-pathogen-vector interaction and vaccine development.  相似文献

We have developed 18 hybridoma cell lines which secrete murine monoclonal strain-specific antibodies to prototype strains of Rickettsia tsutsugamushi: nine anti-Gilliam, four anti-Karp and five anti-Kato antibodies. All the monoclonal antibodies reacted only with their homologous strains in direct and indirect immunofluorescence (IF), or indirect immunoperoxidase (IP) test. By IF and IP tests with the monoclonal antibodies, 22 strains of R. tsutsugamushi, which were newly isolated from mites, field rodents and patients with Tsutsugamushi disease (scrub typhus) in Japan, were all clearly identified as either Gilliam or Karp type. Analysis by polyacrylamide gel electrophoresis and immunoblotting techniques revealed that the monoclonal antibodies recognized primarily the polypeptides of an apparent molecular weight of 54 to 56 kilodaltons of the homologous rickettsial surface. The monoclonal antibodies produced in the present study should enhance the serotyping and further analytical investigation of the rickettsial antigens since they recognize the strain- or type-specific polypeptides and do not show any cross-reaction among strains.  相似文献

Examination of 165 rodents trapped in domestic, peridomestic, and feral biotopes of central and southern India revealed the presence of 1,359 mesostigmatid mites. Rodents in central India were infested with 1 species of mite, Laelaps nuttalli; 98% of these mites were recovered from the peridomestic rodent Bandicota bengalensis. Material from the burrow nest of a peridomestic rodent examined in central India revealed the mite, Hypoaspis miles. Rodents from the diverse biotopes of southern India were infested by 8 species of mites: L. nuttalli, Laelaps buxtoni, Laelaps myonyssognathus, Androlaelaps aduncus, Androlaelaps marshalli, Androlaelaps sp. A, Hypoaspis sp. 1, and Hypoaspis sp. 2. Sheep from nearby villages regularly graze in feral biotopes. From the hair of these sheep, 2 species of mesostigmatid mites were recovered, A. aduncus and A. marshalli. The sheep appeared to aid the dispersal of rodent-infesting mites passively. Laelaps nuttalli was the predominant mite species found on the peridomestic rodent B. bengalensis. In both central and southern India, the prevalence, mean intensity, and relative density of this mite were much higher on peridomestic rodents than on domestic and feral rodents. A significant negative correlation was found between the numbers of mites and fleas infesting rodents in central India. A highly significant positive association between A. marshalli and A. aduncus on the feral rodent Tatera indica was recorded. In southern India, the overall prevalence of the 8 species of mites was highest on Mus platythrix. However, the combined prevalence of these mites on 2 feral rodents T. indica and M. platythrix was lower than their prevalence on B. bengalensis. Similarly, the combined values for mean intensity and relative density of these mites on the 2 feral rodents were lower than on the peridomestic rodent B. bengalensis.  相似文献

The richness, diversity, abundance and prevalence of mite species associated with sigmodontine rodents of different species in Entre Ríos province, Argentina are studied. Five of the six species of mites were reported for the first time in the study area. The richness and diversity of mites was higher on Oligoryzomys flavescens and O. delticola than on Akodon azarae. Androlaelaps rotundus was dominant and exhibited higher values of mean abundance and prevalence on A. azarae, Mysolaelaps microspinosus on O. flavescens and Gigantolaelaps mattogrossensis on O. delticola.  相似文献

Outbreaks of vector‐borne diseases are dramatically increasing because of climate change, consequently increasing the importance of surveillance of endemic disease vectors. In this study, we surveyed chigger mites, vectors for Orientia tsutsugamushi—the bacteria that causes Tsutsugamushi disease—, and their rodent hosts in Gimcheon, central South Korea, in 2015–2018. A total of 225 rodents were collected, with trap rate and percentage of rodents infected by chigger mites of 9.8 and 72.4%, respectively. Six species of rodents from five genera were collected, the most common rodent being Apodemus agrarius (n = 153, infection rate = 90.8%). The highest number of rodents was collected in spring (trap rate = 10.3), but the rate of infected rodents was higher in fall (81.5%) than in spring (61.3%). Trap rate was highest for bank near waterway (17.9), but the chigger index (CI) was highest in hill (224.1). A total of 20,534 (CI 126.0) chigger mite individuals from 10 species and three genera were found on the collected rodents. The most common species was Leptotrombidium pallidum (n = 7,982, 83.6%, CI 49.0), followed by L. palpale and L. scutellare. Chigger mites were most frequent at banks near waterway (n = 11,093, CI 152.0) and hill (n = 2,017, CI 224.1). To detect O. tsutsugamushi in chigger mites, 450 pools of chigger mites (n = 10,991) were analyzed; 24 pools were positive—23 of A. agrarius, the most frequently collected species in South Korea, 1 of Micromys minutus—and the minimum positive rate (MPR) was 0.22. The detected strain types included Boryong (dominant in all years, seasons, and habitats), Jecheon, 07–489, and IIOC1202.  相似文献

The authors present the results of a 6-year study of tsutsugamushi fever at the Kuril islands. The area of the disease proved to include Southern Kuril islands - Shikotan and Kunashir. The principal natural carriers of tsutsugamushi among the mouse-like rodents and trombiculidae larvae were revealed; highly virulent strains were found to prevail among the tsutsugamushi rickettsia strains. Tsutsugamushi fever in the population is characterized by a benign course, pyrexia, primary affects and lymphadenitis.  相似文献

During October-December 1987, chigger mites infested on the striped field mice (Apodemus agrarius coreae) collected at Pochun-gun, Gyeonggi Province and Chinhae city, Kyongnam Province were identified. 1. Of 171 back-striped mice collected, chigger mites were found from 58 mice, showing 33.9% of infestation rate. 2. Total 865 chigger mites collected were classified into seven species; Leptotrombidium pallidum (4.3%), L. palpale(23.0%), L. orientalis(20.2), L. zetum(19.0%). Neotrombicula tamiyai(32.9), N. japonica(0.2%) and Euschongastia koreaensis(0.3%). Though the most dominant species in number was N. tamiyai(32.9% of the total), this species was collected at very limited locality and for a short period. Therefore, most common species in Korea seems to be L. palpale which was found at all localities throughout the whole survey period. L. pallidum which is known as the vector species of tsutsugamushi disease in Korea was collected in a very low number(4.3% of the total chiggers collected). 3. The number of chigger mites infested on a host animal showed great variations from one chigger up to 207 chiggers. The present study has shown that there may be other Leptotrombidium species mite(s) for the vector and host of tsutsugamushi disease in Korea.  相似文献
