解淀粉芽胞杆菌(Bacillus amyloliquefaciens)是芽胞杆菌属的一个种,广泛存在于自然界,具有丰富的生境多样性,其芽胞具有抗逆性,可抵抗不良环境,同时能产生多种抗菌物质,如脂肽、抗菌蛋白和多种酶类,能抗植物病原真菌和有害细菌,该菌能形成生物膜,定植于植物根系,促进植物生长,因此对该菌进行分离鉴定以及培养优化具有重要意义。截至目前,已经从各种生境中分离、鉴定了多种有应用价值的解淀粉芽胞杆菌,通过不同策略对该菌的培养基和培养条件进行优化,使该菌相关功能得以提高。本文综述了近年来对解淀粉芽胞杆菌的生境多样性、分离鉴定及培养基和培养条件优化的策略,简要归纳了国内外关于解淀粉芽胞杆菌重要的工业和商业产品,为更好地研究和应用解淀粉芽胞杆菌提供必要的参考和借鉴。  相似文献

植物病原真菌是农业生产的主要威胁之一,使用生物制剂防治病原真菌被认为是更安全和可持续的方式。芽胞杆菌能产生多种抗真菌活性物质(脂肽、细菌素和酶等),是目前应用最广泛的生防菌。基于芽胞杆菌及其代谢产物的生物防治剂,可有效防治植物病原真菌,在农业生产中发挥着重要的作用。本文聚焦于芽胞杆菌代谢产物的生物防治潜力及其对抗3种常见植物病原真菌(稻瘟菌、尖孢镰刀菌、灰葡萄孢菌)的拮抗属性和机理研究等,通过调研近年来已发表的芽胞杆菌代谢产物抗菌的相关文献,对几种重要的芽胞杆菌代谢产物进行介绍,并总结了芽胞杆菌代谢产物对重要植物病原真菌的抗菌效果及其机制,同时对芽胞杆菌代谢产物对病原真菌的壁膜损伤、抑制真菌孢子萌发和菌丝生长以及竞争性结合真菌DNA等机制的研究手段和效果进行了总结,期望为今后芽胞杆菌类生防制剂的制备和应用提供指导策略。  相似文献

【目的】地衣芽胞杆菌FJAT-4产生的脂肽能够有效抑制尖孢镰刀菌的生长,本研究的目的在于探究地衣芽胞杆菌FJAT-4脂肽结构,分析培养基组分和培养温度对FJAT-4产抑菌脂肽的影响,阐述脂肽对尖孢镰刀菌的抑制作用,为菌株抑菌机理的阐释及其在枯萎病防治中的推广应用奠定基础。【方法】通过酸沉醇提法提取地衣芽胞杆菌FJAT-4产生的脂肽;利用液相色谱串联四极杆飞行时间质谱进行地衣芽胞杆菌FJAT-4脂肽组成分析及结构鉴定;以抑菌圈大小为指标评估地衣芽胞杆菌FJAT-4脂肽对尖孢镰刀菌的抑制效果;通过扫描电镜观察地衣芽胞杆菌FJAT-4粗脂肽对尖孢镰刀菌的抑制作用。【结果】地衣芽胞杆菌FJAT-4产生的抑菌脂肽由C_(17)fengycin A、C_(17)fengycin B、C_(17)fengycin B_2、C_(16)fengycin A衍生物、C_(16)fengycin B衍生物、C_(13)–C_(15)surfactin及C_(13)–C_(15)surfactin衍生物组成,其中C_(13)–C_(15) surfactin衍生物(m/z[M+Na]~+=1048.6/1062.6/1076.6)为新化合物。培养基成分不同对菌株FJAT-4脂肽组成影响较小,但温度对菌株FJAT-4产生抑菌脂肽的影响很大,该菌株在较低温度(20–25°C)下培养不产生脂肽,30–40°C下培养能产生抑菌脂肽,且高温有利于提高脂肽中surfactin的比例。该脂肽类物质对辣椒、番茄、香蕉和甜瓜尖孢镰刀菌等多种植物病原真菌均具有很好的抑制效果,且呈剂量依赖性。扫描电镜结果表明地衣芽胞杆菌FJAT-4所产的脂肽会严重影响辣椒、番茄、香蕉和甜瓜尖孢镰刀菌菌丝的正常生长,导致菌丝断裂变形、孢子变形或显著抑制了孢子的生长。【结论】地衣芽胞杆菌FJAT-4产生的抑菌脂肽为fengycin和surfactin类物质,该抑菌脂肽会致使尖孢镰刀菌菌丝体发育畸形,影响尖孢镰刀菌的正常生长。  相似文献

芽胞杆菌是一类产生具有抗逆特性芽胞的重要微生物资源,研究其分类地位对挖掘和利用芽胞杆菌功能菌株具有重要科学意义。基于基因组学的多相分类法,使得芽胞杆菌分类地位更加精确。截至2016年6月,全世界芽胞杆菌种类达813种,来自于5个科,即芽胞杆菌科、脂环酸芽胞杆菌科、类芽胞杆菌科、动球菌科、芽胞乳杆菌科,74个属。中国芽胞杆菌研究从2004年开始发表新种,目前,中国学者发表中国分离的芽胞杆菌新种达176种。本文就当前芽胞杆菌主要分类鉴定方法及应用进展进行了描述,希望为国内外芽胞杆菌同行研究者提供一定的帮助。  相似文献

解淀粉芽胞杆菌(Bacillus amyloliquefaciens)具有很强的抑制植物病原真菌的能力。其菌体细胞能产生多种酶类、脂肽类抗生素、生物表面活性素、聚酮类化合物和抑菌蛋白,同时具有诱导植物产生系统抗性(ISR)的能力,因此在工农业、种植业、养殖业、食品加工业、果蔬的采后保鲜和饲料业等行业具有重要价值。本文对解淀粉芽胞杆菌抗真菌作用、抗真菌能力提高策略、抗菌化合物合成调节、抑制真菌机制及其引发的ISR等问题进行了深入探讨和综述。  相似文献

【背景】芽胞杆菌是农业上重要的微生物菌剂,对植物具有促生、防病、防虫等作用。【目的】了解亚热带植物内生和根际芽胞杆菌的种群分布,为其功能挖掘提供科学依据。【方法】采用可培养手段对甘蔗(Saccharum officinarum)、红麻(Hibiscus cannabinus L.)、黄麻(Corchorus capsularis L.)、黄秋葵(Abelmoschus esculentus)和玫瑰茄(Hibiscus sabdariffa)根际土壤和根内生芽胞杆菌进行分离,利用16S rRNA基因测序对分离菌株进行分类鉴定,并分析系统发育地位。【结果】共获得了可培养芽胞杆菌菌株144株,其中根内生芽胞杆菌82株,根际土壤62株。经16S rRNA基因测序鉴定为芽胞杆菌4个属37个种,分别为芽胞杆菌属、短芽胞杆菌属、类芽胞杆菌属和赖氨酸芽胞杆菌属。芽胞杆菌菌落数量和种类在根部及其根际土壤中差异较大,根际土壤中芽胞杆菌菌落数量远远大于根部,根际土壤中芽胞杆菌菌落含量范围为(0.2-370.0)×10~5CFU/g,而根部为(0.1-81.0)×10~3 CFU/g。根部分离获得的芽胞杆菌种类远大于根际土壤中,从作物根际土壤中共获得了芽胞杆菌19个种,从根部获得内生芽胞杆菌共32个种。阿氏芽胞杆菌(Bacillus aryabhattai)、仙草芽胞杆菌(Bacillus mesonae)和假蕈状芽胞杆菌(Bacillus pseudomycoides)同时存在于4种作物的根际土壤和根部,其他芽胞杆菌种类仅存在于1种作物的根际或者根部。【结论】亚热带作物根际土壤和根部芽胞杆菌种类和数量极为丰富,而且还存在可分离培养的芽胞杆菌潜在新物种,这为了解植物与根际微生物相互作用、根际环境生态平衡提供了理论基础和科学依据。  相似文献

【目的】从昆虫黑水虻分离的肠道细菌进行抗植物病原菌的拮抗菌筛选,对获得有拮抗活性的肠道细菌进行活性物质的分子鉴定。【方法】用稀释涂布法从水虻肠道中分离菌株,采用平板对峙法进行抗菌筛选,对有抗菌活性的菌株通过生理生化实验、16S rRNA鉴定和进化树分析确定其种属。参考已知脂肽合成关键基因设计引物,以拮抗菌总DNA为模板进行PCR扩增,对目的片段进行测序。【结果】通过抗菌筛选获得一株对水稻黄单胞菌以及小麦纹枯病病原菌等有很强抑制效果的水虻肠道细菌BSF-CL,经鉴定为枯草芽胞杆菌。脂肽合成关键基因PCR结果显示BSF-CL菌株具有脂肽Iturin和Surfactin合成的关键基因。推测BSF-CL很可能合成脂肽Iturin和Surfactin。【结论】从水虻肠道中分离出对水稻黄单胞菌有很强抑菌活性的菌株,分离菌被鉴定为一种枯草芽胞杆菌,通过活性物质的分子克隆鉴定初步推测其活性物质可能为脂肽Iturin和Surfactin。  相似文献

枯草芽孢杆菌JA产生的脂肽类抗生素对植物病原真菌有广谱抗性。将发酵液经过酸沉淀、甲醇抽提以及反相高效液相色谱等步骤, 分离得到脂肽类抗生素的纯品。经IC50实验和抗菌谱测定, 考察了脂肽类抗生素对多种植物病原菌的作用, 确定了脂肽类抗生素的抗菌谱。深入研究表明, 枯草芽孢杆菌JA还产生未知成分的挥发性抑菌物质, 能够抑制灰霉病菌孢子的萌发和菌丝的生长。脂肽类抗生素和挥发性抑菌物质的协同作用, 有助于提高枯草芽孢杆菌的生物防治效果。  相似文献

【背景】植物内生菌的次生代谢产物是新型天然活性物质的重要来源。【目的】从芍药内生细菌中筛选对金黄色葡萄球菌有抑菌活性的菌株和次生代谢产物。【方法】采用平板对峙法筛选拮抗菌株,根据形态学特征和分子生物学的方法鉴定菌株,PCR扩增检测合成脂肽类物质的功能基因;运用牛津杯法依次测定内生细菌发酵液和脂肽类粗提物的抑菌活性,利用Sephadex LH-20凝胶层析分离脂肽类物质,利用基质辅助激光解吸电离飞行时间质谱分析具有抑菌作用的分离组分。【结果】共筛选出13株对金黄色葡萄球菌具有不同程度抑制作用的内生菌株,其中菌株SY11的抑菌作用最为显著,其发酵液和脂肽类粗提物均具有较强的抑制作用。结合形态学鉴定以及16S r RNA基因序列分析,鉴定其为解淀粉芽孢杆菌(Bacillusamyloliquefaciens)。PCR扩增检测表明菌株SY11含有3个合成脂肽类物质的功能基因fenA、ituD和srfkn,推测该菌株可能具有合成脂肽类物质的能力。根据具有抑菌活性分离组分的质谱分析结果,推测其有效物质的主要成分为Bacillomycin D。【结论】解淀粉芽孢杆菌SY11对金黄色葡萄球菌有良好抑制效果,其脂肽类粗提物也具有较强的体外抑菌活性。本研究为芍药内生细菌的开发应用奠定了基础。  相似文献

A new family of lipopeptides produced by Bacillus thuringiensis, the kurstakins, was discovered in 2000 and considered as a biomarker of this species. Kurstakins are lipoheptapeptides displaying antifungal activities against Stachybotrys charatum. Recently, the biosynthesis mechanism, the regulation of this biosynthesis and the potential new properties of kurstakins were described in the literature. In addition, kurstakins were also detected in other species belonging to Bacillus genus such as Bacillus cereus. Lipopeptides are microbial surface active compounds produced by a wide variety of bacteria, fungi and yeast. They are characterized by highly structural diversity and have the ability to decrease the surface and interfacial tension at the surface and interface, respectively. Surfactin, iturin and fengycin of Bacillus subtilis are among the most studied lipopeptides. This review will present the main factors encountering lipopeptides production along with the techniques developed for their extraction and purification. Moreover, we will discuss their ability to form pores and destabilize biological membrane permitting their use as antimicrobial, hemolytic and antitumor agents. These open great potential applications in biomediacal, pharmaceutic and agriculture fields.  相似文献

Marine Bacillus circulans DMS-2 (MTCC 8281) was found to produce lipopeptides, which reduced the surface tension of the medium from 69 to 28 mNm−1. The methanol extracts of the lipopeptides were resolved in RP-HPLC, which resulted in the presence of two major surface active fractions corresponding to the peaks E and F were eluted at the retention times of 16.8 and 18 min, respectively. Fourier transform infrared spectroscopy and matrix assisted laser desorption ionization time of flight (MALDI-ToF) mass spectral analysis showed that the produced lipopeptides belonged to surfactin family. In the MALDI-ToF spectrum, the major molecular mass of the HPLC purified isoforms were identified at m/z 1,044 and 1,058 Da, respectively. In addition, this marine strain also produced new variants of surfactins with molecular weights ranging from m/z 1,066 to 1,098 Da, which have not been reported earlier. Both surface-active fractions were found to have potent antimicrobial activity against the Gram-positive and Gram-negative bacterial strains.  相似文献

多种芽孢杆菌为益生菌,能分泌多种天然抗菌活性物质,其中脂肽是重要的一类。目前已鉴定的脂肽约有90多种,多数为环脂肽。脂肽中表面活性素(surfactin)、伊枯草菌素(iturin)、芬原素(fengycin)、杆菌霉素(bacillomycin)、多粘菌素(polymyxins)等是研究最广泛的脂肽。其中surfactin、iturin、fengycin由于其具有表面活性剂特性及抗真菌、抗细菌、抗病毒、抗肿瘤、抗炎症等功能,应用潜力巨大。本文对surfactin、iturin及fengycin的结构、功能、合成调控及其分离纯化和生产等方面的研究进展进行了评述。合成生物学是提高脂肽产量的重要手段,未来脂肽可用于种植业、养殖业、食品、医药、石油工业和环保等领域,因此需要在新型脂肽的发现、高产活性脂肽的生产、脂肽低廉生产技术的研发及安全性的评估等方面加强研究。  相似文献

【目的】分析枯草芽孢杆菌纳豆菌亚种Bna05菌株代谢产物中脂肽类物质的存在情况,并探讨它们在抗霉功能中所发挥的作用。【方法】利用特异性引物对Bna05菌株进行脂肽合成酶类基因片段扩增、测序和BLAST比对分析;通过平板抑菌圈区域取样法获得Bna05菌株的高抗霉活性代谢产物,对该产物进行反相高效液相色谱(RP-HPLC)分离;用琼脂微稀释法测定分离物的抗霉活性,并对活性分离物进行质谱鉴定。【结果】Bna05菌株含有sfp和srf AA基因,未检测到itu C、itu D、fen D、fen ACE、bym B、bym C基因;RP-HPLC分离得到3组抗霉活性物质F_2、F_3和F_4,F_2中未检测到脂肽类物质,从F_3和F_4中分别鉴定出两类Surfactin同系物:V_7-surfactin和I/L_7-surfactin。两类Surfactin分别与F_2组合使用时,均表现出抗霉协同作用;此外,与Surfactin单独使用相比,两类Surfactin混合物与F_2组合后的协同抗霉活性得到进一步增强。【结论】Bna05菌株所产脂肽类物质主要是V_7-surfactin和I/L_7-surfactin,Surfactin与Bna05菌株所产其它活性物质之间存在抗霉协同作用,而V_7-surfactin和I/L_7-surfactin的同时存在,对于增强这种协同抗霉作用是有利的。  相似文献

Bacillus subtilis GLB191 (hereafter GLB191) is an efficient biological control agent against the biotrophic oomycete Plasmopara viticola, the causal agent of grapevine downy mildew. In this study, we show that GLB191 supernatant is also highly active against downy mildew and that the activity results from both direct effect against the pathogen and stimulation of the plant defences (induction of defence gene expression and callose production). High-performance thin-layer chromatography analysis revealed the presence of the cyclic lipopeptides fengycin and surfactin in the supernatant. Mutants affected in the production of fengycin and/or surfactin were thus obtained and allowed us to show that both surfactin and fengycin contribute to the double activity of GLB191 supernatant against downy mildew. Altogether, this study suggests that GLB191 supernatant could be used as a new biocontrol product against grapevine downy mildew.  相似文献

Cyclic lipopeptides are produced by a soil Bacillus megaterium strain and several other Bacillus species. In this work, they are detected both in the Bacillus intact cells and the cells culture medium by MALDI-TOF mass spectrometry. The cyclic lipopeptides self-assemble in water media producing negatively charged and large aggregates (300–800 nm of mean hydrodynamic radius) as evaluated by dynamic light scattering and zeta-potential analysis. The aggregate size depends on pH and ionic strength. However, it is not affected by changes in the osmolarity of the outer medium suggesting the absence of an internal aqueous compartment despite the occurrence of low molecular weight phospholipids in their composition as determined from inorganic phosphorus analysis. The activity against a sensitive Bacillus cereus strain was evaluated from inhibition halos and B. cereus lysis. Essential features determining the antibiotic activity on susceptible Bacillus cereus cells are the preserved cyclic moiety conferring cyclic lipopeptides resistance to proteases and the medium pH. The aggregates are inactive per se at the pH of the culture medium which is around 6 or below. The knock out of the sensitive cells only takes place when the aggregates are disassembled due to a high negative charge at pH above 6.  相似文献

Lipopeptides constitute a structurally diverse group of metabolites produced by various bacterial and fungal genera. In the past decades, research on lipopeptides has been fueled by their surfactant activities. However, natural functions of lipopeptides compounds have received considerably less attention. The aim of this study was to isolate and identify the lipopeptides from Bacillus amyloliquefaciens An6, and further evaluate their biological activities. An6 lipopeptides were detected by PCR using degenerated primers and MALDI‐TOF‐MS. An6 strain was found to produce surfactin, fengycin, and bacillomycin. Following their purification, the in vitro antioxidant activity of An6 lipopeptides was studied through different assays. The scavenging effect on 1,1‐diphenyl‐2‐picrylhydrazyl radicals at a dosage of 0.75 mg/mL was 81%. Its reducing power was concentration‐dependant and reached a maximum of 1.07 at 2.5 mg/mL. Moreover, they showed a strong inhibition of β‐carotene bleaching. An6 lipopeptides mixture was also found to display significant antimicrobial activity against several Gram‐positive, Gram‐negative bacteria, and fungal strains. An6 lipopeptides were insensitive to proteolytic enzymes, stable between pH 4.0 and 12.0, and resistant to high temperature. Our results provided enough evidence proving that An6 lipopeptides could be used as functional‐food components.  相似文献
