Solenopsis invicta virus 2 (SINV-2) is the second virus identified from the fire ant, S. invicta Buren. SINV-2 is unique among positive-strand RNA viruses from insects by possessing four cistrons in a monopartite genome. Fire ant colonies testing positive for SINV-2 by RT-PCR did not exhibit any discernable symptoms. RT-PCR-based surveys for SINV-2 among 688 fire ant mounds in Alachua County, Florida, sampled during the period January, 2006 through December, 2007 showed that the prevalence of SINV-2 among nests ranged from 1.6% to 16.4%. Unlike S. invicta virus 1, no seasonal-associated prevalence was observed with regard to SINV-2 infection among fire ant colonies. No social form specificity was evident; SINV-2 was found in both monogyne and polygyne S. invicta ants. Real-time quantitative PCR experiments showed that SINV-2 genome equivalents per individual ant ranged from 1.9 × 107 in. pupae to 4.3 × 1011 in. inseminated queens. The SINV-2 infection was detected in all ant stages examined (eggs, larvae, pupae, workers, and queens). Tissue tropism studies indicated that the alimentary canal (specifically the midgut) is most likely the susceptible tissue. SINV-2 was successfully transmitted to uninfected S. invicta ants by feeding a partially purified homogenate of SINV-2-infected ants. The SINV-2 transmission rate ranged from 30% to 80%, and both positive (genomic) and negative (replicative) SINV-2 RNA strands accumulated in recipient ants over the course of the experiment. The red imported fire ant Solenopsis invicta Buren (RIFA), an invasive pest that has diverse detrimental impacts on the communities it invades, was recently discovered in China and has the potential to colonize numerous other regions. Using the model of Korzukhin et al. as modified by Morrison et al. and the biological and ecological characteristics of RIFA, we show that Jiangsu Province is a potentially suitable establishment area of RIFA. An isotherm map made by ArcMap, a Geographic Information System, indicated that several regions of Jiangsu Province (Changzhou, Liyang, Wuxian Dongshan, Nanjing and Lvsi) are at higher risk of S. invicta infestation, especially from late July and early August. Quarantine officials should be vigilant for any accidental introductions of this pest in the susceptible regions and time.  相似文献

Red imported fire ants (RIFA), Solenopsis invicta Buren, are medical, urban, and agricultural pests from South America. They are successful invaders due to their preference for disturbed habitats, high reproductive rates, and the ability to feed on a wide variety of food items (omnivorous). Fourth-instar larvae are used by the colony to digest solid food and then regurgitate it for consumption by workers and queens. Larvae are an ideal source of investigations of endosymbiotic bacteria possibly involved in nutrient distributions. Our study utilized 16S rDNA sequencing to describe the composition of the bacterial community in fourth-instar ant larvae in order to identify possible endosymbiotic bacteria present therein. The 16S rRNA gene was directly amplified from mixed-population DNA of whole fire ant larval guts and cloned into Escherichia coli. Bacterial communities from three geographically separated RIFA colonies were examined. Sequenced bacterial clones from guts were determined to be predominantly from the phylum Proteobacteria and the family Enterobacteriaceae. Our results did not detect the presence of endosymbiotic bacteria in the guts of RIFA larvae among the colonies. In addition, minimal species overlap was found when bacterial inventories were compared among colonies. Thus, bacteria coadapted with red imported fire ant larvae were not detected. Identified bacteria were not closely affiliated with endosymbiotic bacteria common in other insect species. Bacteria communities appeared to be unique to each geographical location and were determined by the foods consumed by the ants.  相似文献

Bacteria were isolated and cultured from the red imported fire ant (Solenopsis invicta) midgut. The small-subunit ribosomal RNA gene, (16s rRNA gene, approximately 1500 bp) was amplified from bacterial genomic DNA using the polymerase chain reaction and consensus sequence primers. Restriction fragment length polymorphism analysis revealed 10 unique profiles, indicating that at least 10 different bacteria are present in red imported fire ant midguts. The 16s rRNA gene sequence was determined for these isolates and queried against the NCBI genetic database. The results identified all isolates to at least the genus level. Antibiotic resistance profiles and biochemical activities were also determined for these species. This work provides the basis for a wider characterization of bacterial distributions in fire ant colonies and provides strains suitable for genetic manipulation to develop novel methods of fire ant control.  相似文献

Colonies of the fire ant, Solenopsis invicta, can survive flood conditions by forming a raft of ants that floats on the water’s surface until the flood recedes or higher ground is found. Having been forced from the protection of their subterranean nests, rafting colonies are totally exposed and are without retreat. I tested the hypothesis that rafting S. invicta colonies would compensate for their elevated vulnerability by increasing their defensiveness. I measured defensiveness using the amount of venom workers delivered per sting (venom dose), since the repellent effects (i.e., pain and tissue damage) of fire-ant venom are dose-dependent. In the laboratory I assayed colony defensiveness before and after flooding colonies from their nests with water. Colonies were consistently and significantly more defensive while rafting (i.e., each colony’s workers delivered higher venom doses when their colony was rafting than they did when it was assayed pre-flood). The larger venom doses of rafting colonies may reduce their chances of being damaged by encounters with other animals by reducing the duration of such encounters through increased repellency. Encounters with S. invicta during flood conditions have the potential to be unusually dangerous; large concentrations of workers are exposed and available for defense, and they deliver significantly larger venom doses when they sting. We screened adult and larval secretions and midden piles for the presence of Thelohania solenopsae spores to decipher potential sources for the horizontal transmission of the pathogen in fire ants. Hemolymph samples from both adult and larvae were also screened to rule out hemolymph contamination of samples. In adults, Thelohania spores were found in the crop and the fecal fluids, although only free spores were found in the fecal fluids of adults. In fourth instar larvae, both free and octospores were seen in midgut and the meconium samples. All of the midden pile samples had T. solenopsae spores of both types. Based on these results, we theorize that the pathogen may be horizontally transmitted within a colony by the removal and sharing of meconium of prepupating fourth instar larvae by adult workers and by the adult fecal droppings, and intercolonially by contamination of midden piles or brood raiding.  相似文献

Analysis of extracts of hemolymph obtained from sexually mature alate females of Solenopsis invicta from monogyne colonies resulted in identification of juvenile hormone III (JH III). The average amount of JH III was 0.32±0.04 pmol/μmolof hemolymph. Topical application of 0.038 pmol of JH III was sufficient to stimulate alates to shed their wings in the presence of the queen. The time in which alates were induced to dealate decreased linearly with increasing concentrations of JH III from 0.038 to 3.8 pmol. However, higher JH III concentrations deviated from linearity and did not reach dealation times comparable with those that occur after mating flights. Thus, it appears that the mechanism of dealation that occurs when female alates are out of the influence of their queen is different from the one associated with mating flights. Application of 0.42 μmol of precocene II inhibited dealation of alates in queenless colonies. However, this inhibition was reversed after applying 38 pmol JH III to precocene-treated alates. The sizes of corpora allata (CA) from sexuals treated with JH III did not differ from those of controls. However, the sizes of CA were reduced in alates treated with precocene II. The results indicated that JH was important to dealation.  相似文献

Quantitative real-time PCR was employed to measure the Solenopsis invicta virus 1 (SINV-1) load in tissues, individuals, and among colonies of the red imported fire ant, S. invicta Buren. Among tissues examined from SINV-1-infected adults and larvae, the alimentary canal (specifically the midgut) consistently had the highest number of SINV-1 genome copies (91.1 and 99.9%, respectively). Negative staining of a supernatant of the gut homogenate demonstrated the presence of spherical virus particles with a diameter of 30-35 nm, consistent with SINV-1. The number of SINV-1 genome copies in infected larvae and workers from the same queenright colonies were similar to each other. In other words, the infection rate was consistent among both developmental stages. No significant differences were observed in SINV-1 genome copy number among infected colonies sampled during the winter and summer. Although the SINV-1 infection rate of summer-collected mounds was previously shown to be six-times higher than winter-collected mounds, the intra-colony infection rate appears to be unaffected by season. Perhaps less inter-mound interaction during the winter months among S. invicta restricts spread of the virus. A positive correlation between intra-colony infection rate and mean SINV-1 genome copy number per ant was also observed. Based on these results, it is likely that SINV-1 replicates in gut epithelia of S. invicta and virus is shed into the gut lumen where it may be transmitted to nestmates by trophallaxis.  相似文献

Studies were conducted to examine the phenology, geographic distribution, and host specificity of the Solenopsis invicta virus-1 (SINV-1). Two genotypes examined, SINV-1 and -1A, exhibited similar seasonal prevalence patterns. Infection rates among colonies of S. invicta in Gainesville, Florida, were lowest from early winter (December) to early spring (April) increasing rapidly in late spring (May) and remaining high through August before declining again in the fall (September/October). Correlation analysis revealed a significant relationship between mean monthly temperature and SINV-1 (p<0.0005, r=0.82) and SINV-1A (p<0.0001, r=0.86) infection rates in S. invicta colonies. SINV-1 was widely distributed among S. invicta populations. The virus was detected in S. invicta from Argentina and from all U.S. states examined, with the exception of New Mexico. SINV-1 and -1A were also detected in other Solenopsis species. SINV-1 was detected in Solenopsis richteri and the S. invicta/richteri hybrid collected from northern Alabama and Solenopsis geminata from Florida. SINV-1A was detected in S. geminata and Solenopsis carolinensis in Florida and the S. invicta/richteri hybrid in Alabama. Of the 1989 arthropods collected from 6 pitfall trap experiments from Gainesville and Williston, Florida, none except S. invicta tested positive for SINV-1 or SINV-1A. SINV-1 did not appear to infect or replicate within Sf9 or Dm-2 cells in vitro. The number of SINV-1 genome copies did not significantly increase over the course of the experiment, nor were any cytopathic effects observed. 采用系统调查的方法研究广东吴川红火蚁Solenopsis invicta Buren蚁巢数量、蚁丘体积和工蚁的年度变化规律,结果表明1年之中红火蚁活动蚁巢密度出现2个高峰期,分别在6~8月和10~12月;4~7月和10~11月新增活动蚁巢数量较大;蚁巢自然消亡出现在多个时间,其中以9月最多。4~11月红火蚁工蚁活动旺盛,用诱饵诱集法监测,5、6月和10、11月红火蚁工蚁出现2个明显盛期,而陷阱法中工蚁数量为单峰型,高峰出现在7月。2种监测方法获得的工蚁数量间相关系数为0.88,达到极显著相关。气温、地表温度、5cm土壤温度与2种方法获得的工蚁数量间相关系数分别为0.88,0.88,0.86和0.93,0.94,0.95,均达极显著水平。红火蚁蚁丘体积4~6月、9~12月增大幅度较大,其中6月份蚁巢增长最为明显。  相似文献

调查了广东广州、深圳40余个苗木花卉场74种带土园艺植物上红火蚁Solenopsis invicta Buren发生情况。调查结果表明,桉树苗、海枣、垂叶榕、黄金香柳、小叶榕、狗尾草、罗汉松等17种植物上红火蚁发生比率超过1%,是高风险带土植物。不同生产环境、不同栽培方式中盆/钵/袋栽红火蚁发生高于地面栽培方式,露天置于砂土、泥土、碎石上红火蚁发生风险较高,置于水泥地上的风险较低。提出了降低带土园艺植物传播红火蚁的管理措施。  相似文献

红火蚁Solenopsis invicta是世界上最具破坏力和威胁性的入侵检疫害虫之一,对农林业生产安全、公共设施以及人体健康都造成了巨大影响。为筛选对红火蚁有致病性的病原真菌,本研究通过组织分离法对自然罹病的红火蚁进行致病菌分离,结合病原形态学及分子生物学的方法对病原真菌进行鉴定并采用喷雾法测定了病原真菌对红火蚁的致病力。结果表明:从罹病红火蚁上分离出3株病原真菌HHYSFJ01、HHYSFJ02和HHYSFJ05,分别为红绶曲霉Aspergillus nomius、淡紫拟青霉Purpureocillium lilacinum和球孢白僵菌Beauveria bassiana。致病性测定,这3株病原菌对红火蚁均具有较强的致病性,且3株病原菌致病性间差异显著。经1.0×10^8孢子/mL孢子悬浮液喷雾处理红火蚁10 d后,球孢白僵菌菌株HHYSFJ05的致病性最强,红火蚁工蚁的校正死亡率为84.71%,LT50为4.67 d;红绶曲霉菌株HHYSFJ01和淡紫拟青霉菌株HHYSFJ02次之,红火蚁工蚁的校正死亡率分别为78.82%和73.29%,LT50分别为4.99 d和5.20 d。  相似文献

At the onset of mating flights inSolenopsis invicta, workers swarm excitedly over the mound as alates prepare to fly. Previous studies demonstrated that this excitement is stimulated by the male and female alates. We investigated the glandular source(s) of pheromones produced by the alates that cause excitement. The only common female and male alate body part that elicited excitement when crushed was the head. Within the head, excised mandibular glands were found to be responsible for worker excitement. Fire ant workers are very sensitive to external stimuli and some excitement was elicited by crushed female gasters and male thoraces, but the response was never as significant as with crushed heads. Tests with summer and winter alates revealed similar results, except that gasters of winter female alates had a greater excitant effect than did gasters of summer female alates. This may be due to the production of attractant pheromones by the poison glands of overwintering female alates. We conclude that the mandibular gland is the source of alate excitant pheromones.  相似文献

We surveyed 165 sites to determine the ecological factors that might influence the distribution and prevalence of Thelohania solenopsae, and its effect on the demography of the red imported fire ant (Solenopsis invicta) in Louisiana. The microsporidium was found in 9.9% of colonies and at 16% of sites. Its distribution was clumped within the state with the majority of infected colonies and sites occurring in two infection patches. The proportion of polygyne colonies was a strong (positive) predictor of the proportion of infected colonies at a site. Infected monogyne colonies, however, still accounted for nearly 20% of infected colonies, a much higher proportion than anticipated. Several other factors, including the numbers of colonies at a site, precipitation, proximity to commercial waterways and ports, and type of habitat were also retained in the multiple logistic regression model describing T. solenopsae prevalence. The microsporidium appears to adversely affect the occurrence of brood, and possibly the size of S. invicta colonies and the mass of workers. It, however, was not included in the multiple regression model of the number of colonies or the density of ants at a site. Although our findings do not imply causation, they have identified several variables that might influence the epizootiology of T. solenopsae. Future work should concentrate on experimentally manipulating these variables to confirm these relationships.  相似文献

本文调查研究了蚁丘被破坏的程度对红火蚁Solenopsis invicta Buren蚁群迁移的影响。结果表明,蚁巢地表部分被破坏可导致红火蚁蚁群发生分巢、搬巢等现象,蚁群的移巢比率随着蚁丘被破坏程度的增大而增大。当蚁丘体积被破坏75%时分巢比率最高,为14.44%;被破坏100%时搬巢比率、移巢比率均最高,分别为16.27%、27.00%。红火蚁蚁丘被破坏程度(D)与蚁群迁移比率(M)之间关系模型为M=4.3443+25.474D-3.4857D2。蚁丘受到破坏后,蚁群迁移距离平均为4.3m,迁移方向是随机的。  相似文献

A potentially important and understudied biological control agent in US agroecosystems is the red imported fire ant, Solenopsis invicta Buren. Red imported fire ants may be particularly important biological control agents because we can manipulate their abundance with changes in habitat complexity. The effect of habitat complexity on biological control by fire ants was determined using plots of collards intercropped with white clover (complex habitat) and simple collard monocrops. The most economically significant pests of collards are larvae of the diamondback moth (DBM), Plutella xylostella (L.). Predation of DBM larvae by fire ants was more rapid and efficient in the intercrop than the monocrop. Red imported fire ants were 23% less abundant in the intercrop than the monocrop, however, suggesting that fire ants had a greater per capita effect on DBM survival in the complex habitat. Red imported fire ant predation of DBM larvae was significantly affected by larval density. Red imported fire ants also reduced the survival of leaf beetles, another economically significant pest taxa, by 45%. Furthermore, collard leaf damage tended to be inversely related to fire ant density and fire ants were more effective at reducing crop damage in the complex intercrop. Our study indicates the ability of red imported fire ants to be effective biological control agents and suggests that increasing habitat complexity can enhance red imported fire ant efficacy and herbivore control.  相似文献

We have characterized 11 polymorphic microsatellite loci in the invasive ant Solenopsis invicta. Primer pairs were evaluated on fire ants collected from monogyne mounds in Lauderdale County, Mississippi. The observed and effective number of alleles ranged from two to six and from 1.31 to 2.64, respectively. The observed and expected heterozygosity values ranged from 0.1613 to 0.7826 and from 0.1491 to 0.6242, respectively. The polymorphism information content of the microsatellites ranged from 0.1482 to 0.6208. Probability tests indicated significant deviations from the Hardy-Weinberg equilibrium at three loci. Pairwise tests did not detect linkage disequilibrium between any pair of loci.  相似文献
