于2004年4月至2005年5月在西双版纳的3个不同地点(勐仑植物园、新山和补蚌)选择荒地、野芭蕉林和森林3种不同植被类型的生境,对2个重要的雨林种子传播类群--鼠类和蚂蚁的多样性进行了研究.采用铗日法捕获9种鼠共97只,采用陷阱法捕获104种蚂蚁共2 600头.结果表明,蚂蚁聚集生活在干燥的荒地生境中,而鼠类则集中生活在食物资源丰富的野芭蕉林生境中.不同地点鼠的种类和数量无显著差异(P>0.05),但不同地点蚂蚁的种类和数量差异显著(P<0.05);不同生境鼠和蚂蚁的种类和数量均差异显著(P<0.05).地点和生境对2个类群分布有不同的影响,这种影响也通过它们对种子散布的不同作用,最终影响地区内植被的更新和循环.  相似文献

通过排水采集器(Lysimeter)模拟试验,研究了太湖地区不同施肥水平下稻季农田养分淋洗特点。结果表明,施肥后田面水NH4^+-N浓度升高很快,2~3d达到峰值,最高值达26.2mg·L^-1,随后下降很快,这一周期约7~10d,渗漏水中NH4^+-N浓度很低,稻季NH4^+-N淋洗的氮仅占施肥量的0.008%~0.074%,渗漏液中NO3^--N含量随着氮肥用量的增加而增加,其浓度范围在0~7.14mg·L^-1,在土壤剖面中呈上低下高的趋势,稻季氮素的淋洗仍以NO3^--N为主,净淋洗量在3.2~8.3kg·hm^-2之间,占总施肥量的1.40%~2.78%,田面水磷浓度在施磷肥后1d即达最高值,随后下降,下层渗漏液中T-P含量很低,几乎不受施肥量的影响,猪粪能促进磷的迁移。  相似文献

报道了对太湖猪7个类群,红灯笼猪,约克夏种猪及太湖地区野猪共57个个体随机扩增多态DNA(RAPD)分析的结果,重建了太湖猪的系统发生树。实验从40个引物中选用重复性较好的13个引物对所有个体进行了随机扩增,共获得198个RAPD标记,单个引物获得的标记数在3-16间。  相似文献

根据核DNA的ITS序列的RFLP分析鉴定稻属CD染色体组物种   总被引:1,自引:0,他引:1  
稻属物种的染色体组类型有10种,其中具CD染色体组的物种有3种(Oryza alta Swallen,O.grandiglumis(Doell)Prod.和O.latifolia Desv.),仅分布在美洲中部和南部.这3个种在形态上和稻属其他染色体组的种比较容易区别,但它们彼此之间鉴别比较困难.近年来的研究表明,O.alta和O.grandiglumis应归并为1个种(O.grandiglumis),而O.latifolia应保持不变.本文基于代表不同分布区11个样品的核糖体转录间隔区(ITS)的77个克隆序列数据,利用DNA Striderl.2软件进行了限制性酶切位点分析,提出了一个鉴别CD染色体组物种的方法.方法的具体步骤是:(1)用通用引物扩增ITS片段;(2)用特异性的限制性内切酶Fok Ⅰ和/或Dra Ⅲ消化PCR扩增产物;(3)用1%的琼脂糖电泳并根据消化产物的多态性特征来鉴别不同物种.基于本文提出的核糖体ITS限制性片段多态性(RLFP)分析,可以快速和可靠地将稻属CD染色体组物种区别开来.  相似文献

洞庭湖浮游生物群落DNA指纹与理化因子的关系   总被引:12,自引:0,他引:12  
对东洞庭湖7个采样站点的浮游生物群落DNA多态性进行了RAPD指纹分析,并探讨了其DNA指纹与理化因子的关系。结果如下:(1)从80条随机引物中筛选出的11条引物共获得148条谱带,多态率为98.6%;所用引物在各站点可获得45—70条扩增谱带,平均为57.6条,其中Ⅳ站最少;(2)Ⅳ站的泥沙含量最高(75.5mg/L),其他各站在1.2—40.3mg/L;各站点间的pH值比较接近,都呈弱碱性;Ⅰ站的TN、TDN及TP含量最高,Ⅲ站的TDP最高,但各站点相差不大。相似性聚类分析表明,浮游生物群落DNA指纹将7个站点划分为两大类———Ⅳ站作为单独的一类而明显有别于其他6个站点,这与洞庭湖浮游生物分布的主要制约因子———泥沙含量的聚类结果是一致的。因此,研究表明浮游生物群落DNA指纹与环境主要限制因子是密切相关的,这类资料的积累将为建立简易、有效的水资源质量评价体系提供科学依据。  相似文献

为研究天坑负地形生境土壤酶活性、养分和木本植物多样性的分布特征和内在联系,该文以广西大石围天坑群天坑森林为研究区,基于天坑坑内—坑口—坑外群落多样性调查和土壤生境调查,采用相关性分析和冗余分析方法,探究木本植物物种多样性与土壤因子间的耦合关系。结果表明:(1)坑内木本植物有21种31属20科,坑口木本植物有91种58属58科,坑外木本植物有47种30属30科,天坑坑内Margalef指数和Patrick指数显著低于天坑坑口和坑外生境,沿天坑坑内—坑口—坑外生境变化,物种分布总体较为均匀,但物种数量增多。(2)天坑坑外土壤全氮含量显著高于坑内和坑口生境,坑内的土壤全镁、土壤全磷和土壤速效磷含量显著高于坑口和坑外生境,坑外的土壤碱性磷酸酶和土壤脲酶活性显著高于坑口和坑内生境,表明天坑坑内土壤磷含量较高,但土壤酶活性总体偏低。(3)物种多样性与土壤养分、土壤酶活性相关性强。综上表明,天坑生境对物种多样性、土壤酶活性及土壤养分含量的分布特征影响较大,土壤速效磷、土壤碱性磷酸酶、土壤含水量、土壤脲酶、土壤全镁和土壤全磷是影响天坑森林木本植物物种多样性的关键因子。该研究为喀斯特地区植物资源的保护与...  相似文献

旱区植物多样性、生产力与环境因子的关系是旱区生态学研究的重要课题,对于揭示该地区植被的环境适应机制有重要的参考价值。基于中国旱区东西样带的系统采样和原位调查,定量分析了各影响因素对旱区植物多样性和生产力变化的解释作用,阐明了旱区群落生产力的调控机制。结果表明:(1)旱区群落性状加权值与地上生物量间的关系(R2=0.46)相较于Shannon-Wiener指数与地上生物量的关系(R2=0.21)更为紧密。(2)旱区群落Shannon-Wiener指数、群落性状加权值、地上生物量与年均降水量、土壤有机碳含量、土壤总氮含量线性正相关,与土壤总磷含量无显著相关性;Shannon-Wiener指数、群落性状加权值与年均气温线性负相关,地上生物量与年均气温无显著相关性。(3)年均降水量对旱区群落性状加权值的解释率为40.9%,两者为正相关关系;年均气温对Shannon-Wiener指数的解释率为28.3%,两者为负相关关系。(4)群落性状加权值对地上生物量的直接路径显著,年均降雨量、年均气温和土壤有机碳通过群落性状加权值间接影响地上生物量。  相似文献

依据2006~2008冬季的自然雪被分布状况,在青藏高原东缘的高寒草甸中设置3条样带(即深雪、中雪和浅雪)。在2009年的生长季,在3个雪梯度样带中,分别测定了2种优势植物圆穗蓼(Polygonum macrophyllum)和黑褐穗苔草(Carex atrofusca subsp. Minor)生物量和碳氮营养积累及分配的动态特征。结果表明,深雪能够促进圆穗蓼和黑褐穗苔草生物量和碳氮养分的积累,可能使它们产生的凋落物数量更多且质量更好。深雪更有利于圆穗蓼(非禾本科草本植物)根系生物量、碳氮养分的积累;深雪不仅同时促进黑褐穗苔草(禾本科植物)地上部分和根系生物量、碳氮积累,而且还使其种子产量增加和质量提高,潜在地增强了黑褐穗苔草种子的繁殖能力,可能使得黑褐穗苔草的种间竞争能力增强。可以预见,未来季节性雪被的变化,必定会引起青藏高原东缘高寒草甸的初级生物量及其结构、植物群落物种组成等均发生相应变化。  相似文献

为了探究我国灰鹅品种的遗传多样性和系统进化,运用DNA测序技术测定了我国13个灰鹅品种mtDNAD-loop区521bp序列。结果表明:这13个灰鹅品种521bpD-loop区序列的T、C、A和G碱基含量分别为23.8%、29.0%、32.3%和15.0%;平均单倍型多样度和核苷酸多样度分别为0.19245和0.00036;13个灰鹅种间变异大于种内变异,且均未出现群体扩张现象;共享单倍型和系统进化树分析表明,伊犁鹅起源于灰雁(Anser anser),其余12个灰鹅品种起源于鸿雁(A.cygnoides)。  相似文献

本研究利用20对微卫星引物对鳜(Siniperca chuatsi)原种群体和养殖群体进行遗传多样性分析。结果表明,在鳜原种群体中检测到多态性位点14个,养殖群体11个。在两个群体中共检测到等位基因数96个,其中原种群体检测到等位基因数53个,每个位点的等位基因数在1~7之间,平均有效等位基因数为2.7390;养殖群体检测到等位基因数43个,每个位点的等位基因数在1~6之间,平均有效等位基因数为2.1284。原种群体的平均观察杂合度0.5708,Nei氏期望杂合度0.5295,平均多态信息含量PIC0.5353;养殖群体的平均观察杂合度0.3839,Nei氏期望杂合度0.4011,平均多态信息含量PIC0.5043。因此,与养殖群体相比,鳜原种群体仍有丰富的遗传多样性。本研究可为鳜种质资源的保护、监测和遗传育种提供分子水平上的数据。  相似文献

The full‐length complementary DNA (cDNA) sequences encoding cd8α and cd8β molecules were sequenced and characterized from mandarin fish Siniperca chuatsi. Conserved motifs and residues were found to be present in derived peptides of the Cd8 molecules. For example, WXR motif, DXGXYXC motif, and four cysteine residues were present in the extracellular region of the Cd8 protein. Threonine, serine and proline residues involved in multiple O‐linked glycosylation events were located in the membrane proximal hinge region. The common CPH motif in the cytoplasmic tail was detected similar to other teleost Cd8 molecules. Different from those in mammals, S. chuatsi Cd8 sequences have many extra cysteine residues (C149 in Cd8α sequence and C46, C51 and C158 in Cd8β sequence), which also exist in other teleost Cd8 molecules. Real‐time polymerase chain reaction (RT‐PCR) and Western blot analyses revealed that the thymus had the highest expression of cd8 messenger (m)RNA and protein. After stimulated with phytohaemagglutinin, polyriboinsine‐polyribocyaidylic acid and concanavalin A (ConA), the expression level of cd8 mRNA increased significantly in head‐kidney lymphocytes at 4 and 8 h, but decreased to normal level at 12 h. Similarly, stimulation with ConA in vivo also led to an increase in the cd8 mRNA level in the spleen. Immunohistochemistry analysis demonstrated that Cd8α‐positive cells can be detected in the thymus, spleen and intestine by using polyclonal anti‐Cd8α antibody.  相似文献

Thirty-one Naegleria fowleri strains from different continents were studied by using the Random Amplified Polymorphic DNA (RAPD) method. Five primers among the 40 examined generated eight RAPD variants corresponding partially to their geographic origin. Four characteristic variants are detected in Oceania. The four remaining ones are present in Europe, two of which are observed on other continents. One European variant is found to exhibit similarities with strains from Oceania. The genetic diversity found in Europe and Oceania as well as the existence of ubiquitous RAPD variants bring new insights about the dispersion of N. fowleri throughout the world.  相似文献

1. Aquatic macrophyte diversity and water quality of 55 ponds in western Japan were related to land use and morphometric variables to identify the environmental factors that sustain biodiversity and the spatial extent at which these factors operate. 2. The relevant spatial extent for floating‐leaved macrophyte richness (500 m from pond edge) was larger than that for submerged macrophyte occurrence (10, 75 and 100 m), whereas emergent macrophyte richness was best explained at much larger extents (1000 m). Total macrophyte richness was explained at the extent of 500, 750 and 1000 m. The extents relevant for explaining the physicochemical condition of pond water (100 and 250 m) were similar to those for submerged and floating‐leaved macrophytes, suggesting that these two growth forms are more sensitive to water quality compared to emergent macrophytes. 3. Diversity of all three growth forms and that of total macrophytes collectively were inversely related to turbidity and nutrient concentration; among the three growth forms, submerged macrophytes were most affected by water quality. 4. Negative relationships were found between the proportion of urban area and the diversity of the three growth forms and that of total macrophytes and water quality. Species richness of emergent, floating‐leaved and total macrophytes decreased with depth and increased with surface area up to about 5000 m2, above which it declined. 5. Urbanisation and enlargement of ponds were the two main factors that decreased aquatic macrophyte diversity in irrigation ponds. To alleviate the adverse effects of urban areas on aquatic macrophyte diversity, our results suggest that management efforts should focus on the creation of buffer zones within the relevant spatial extent from the pond edge.  相似文献

Eighteen microsatellite markers were isolated and characterized using an enrichment protocol in the Chinese mandarin fish Siniperca chuatsi (Basilewsky), a commercially important piscivorous fish in China. Out of 48 pairs of primers designed, 18 loci exhibited polymorphism with three to six alleles (mean 4.4 alleles/locus) and average observed heterozygosity ranged from 0.633 to 0.833 (mean 0.748) in a test population from Dongting Lake of China. Except for two loci, all other 16 loci were in the Hardy–Weinberg equilibrium. These markers would be useful for such studies as population genetics, ecology and selective breeding of the Chinese mandarin fish in future.  相似文献

The relationship between mercury content in fish (pike and perch), the different fractions of mercury in lake water and water color was investigated in 76, mainly oligotrophic lakes distributed over a large part of Sweden. The lakes were classified in terms of drainage area characteristics, lake morphometry and water chemistry. The dominant fraction of mercury in lake water was RIHg (fraction reducible to elemental mercury by NaBH4). RIHg and water color were strongly positively correlated. Water color (determined by the comparative method using colored disks) was used as a surrogate for the amount of humic matter in the water. Thus, humic matter appears to be acting as an important carrier of mercury. A positive relationship between mercury content in fish and water color was found only in deep lakes (average depth > 5 m). It is suggested that the bioavailability of mercury attached to humic matter increases due to anoxic conditions, common in the hypolimnion of deep lakes.  相似文献

通过整体连续切片,研究了鳜鱼不同发育时期的头肾结构,并利用原位PCR方法检测了B淋巴细胞在鳜鱼头肾中的分布。在孵化后第1d观察到了肾组织,主要由肾小管组成。尔后头肾的发育经历了三个结构和功能的转变。第一个阶段为孵化后第1d到第7d,头肾作为滤过性器官存在,由肾小管及少量淋巴细胞组成。第二个阶段从第8d到第36d,是一个功能混合型阶段,头肾中既有肾小管,又有造血组织;随时间推移,肾小管数量减少,淋巴细胞数量剧增。紧接着进入第三个阶段:肾小管完全消失,头肾中开始出现大量的嗜铬细胞,头肾作为淋巴-肾上腺组织而存在。肾上腺首先出现在头肾前端,随发育成熟,集中分布于头肾门静脉周围。IgM在鳜头肾中大量表达,IgM分泌细胞分布于整个头肾组织,在血管周围有集中趋势[动物学报51(3):440—446,20051。  相似文献
