用GenBank中飞蝗Locusta migratoriaL.的序列来设计微卫星引物,并对这些引物的有效性进行验证。结果表明,在所设计的3对引物中,只有1对为有效引物,可扩增出微卫星位点。序列分析表明本位点是一个不连续的重复微卫星位点。该多态微卫星位点含有14个等位基因,不同飞蝗地理种群的等位基因数目、大小和频率都存在较大的差异。对该位点各等位基因型进行χ2检验,基因型频率的观察和期望杂合度分别为0.4578和0.8836,该位点不属于Hardy-Weinberg平衡位点(χ2=733.12,P=0.0000)。该微卫星位点表现出高度的多态性说明是分析飞蝗种群遗传多样性的优良分子遗传标记。  相似文献

The migratory locust, Locusta migratoria, is the most widely distributed grasshopper species in the world. However, its global genetic structure and phylogeographic relationships have not been investigated. In this study, we explored the worldwide genetic structure and phylogeography of the locust populations based on the sequence information of 65 complete mitochondrial genomes and three mitochondrial genes of 263 individuals from 53 sampling sites. Although this locust can migrate over long distances, our results revealed high genetic differentiation among the geographic populations. The populations can be divided into two different lineages: the Northern lineage, which includes individuals from the temperate regions of the Eurasian continent, and the Southern lineage, which includes individuals from Africa, southern Europe, the Arabian region, India, southern China, South‐east Asia and Australia. An analysis of population genetic diversity indicated that the locust species originated from Africa. Ancestral populations likely separated into Northern and Southern lineages 895 000 years ago by vicariance events associated with Pleistocene glaciations. These two lineages evolved in allopatry and occupied their current distributions in the world via distinct southern and northern dispersal routes. Genetic differences, caused by the long‐term independent diversification of the two lineages, along with other factors, such as geographic barriers and temperature limitations, may play important roles in maintaining the present phylogeographic patterns. Our phylogeographic evidence challenged the long‐held view of multiple subspecies in the locust species and tentatively divided it into two subspecies, L. m. migratoria and L. m. migratorioides.  相似文献

为探讨群居型西藏飞蝗Locusta migratoria tibetensis Chen地理种群数量性状关系,从青藏高原9个地区采集到不同地理种群,以数值分析对形态指标及其比值进行了分析。结果表明,前翅长度(E)、后足股节长度(F)、头宽(C)形态指标和E/F、F/C两个比值可以作为分析种群关系的参数。聚类分析和主成分分析结果相似,能够较好地说明青藏高原9个种群的地理变异和种群相互关系,9个地理种群可以分三个类群,即第Ⅰ类群包括百巴(BB)与八美(BM)种群;第Ⅱ类群包括普兰(PL)、扎囊(ZN)、洛须(LX)种群;第Ⅲ类群包括噶尔(GE)、香孜(XZ)、日土(RT)、那嘎(NG)种群。  相似文献

采用水平淀粉凝胶电泳技术,分析了我国河北平山、辽宁葫芦岛两个蝗区东亚飞蝗自然种群的遗传结构。在酶谱分析的19个位点中,多数位点的等位基因数目较少,而位点Aat—l、Pgi和Mdh—2的等位基因数目相对较多。由于杂合子数目较少而使每个基因位点的平均杂合度降低(Ho=0.024和0.028)。对每个位点的各基因型进行x^2检验,绝大多数位点的基因型频率偏离Hardy—Weinberg平衡。较低的Fst值(Fst=0.021)表明两个种群的遗传分化程度不高。两个种群间较高的遗传一致度(I=0.991)和较小的遗传距离(D=0.092)也证实了上述结果。由此推测,该蝗虫较强的迁飞能力有可能增强种群间的基因交流,降低种群间的遗传分化。然而,在一些等位酶指标如位点Ao-2、Fbp—l、Mdh—2的等位基因频率分布、每个位点的平均等位基因数(A=2.5和2.7)和多态位点百分率(P=52.6%和57.9%)等,两个种群之间呈现出明显的差异。这也许与两个种群之间不同的生态环境条件和较远的地理距离有关。  相似文献

Abstract: Based on haploid males, random amplified polymorphic DNA (RAPD) markers were used to study genetic variation within and among four French populations and one Finnish outgroup population of the common pine sawfly, Diprion pini (L.), representing a severe European forest pest associated with mass outbreaks. Taking into account that all multilocus haplotypes were detected, a total of 140 individuals were completely discriminated by means of 17 polymorphic markers (present or absent), which were amplified from nine selected random-decamer primers. All populations shared the same genetic types, but pronounced population-specific frequency distributions were found, indicating that on average 84% of the present genetic variation exists within populations and the remaining smaller part counts for interpopulational variation. The haplotype differentiation registered was able to distinguish between the class of lowland populations and the class of highland populations. Reproductive isolation and therefore limited altitudinal gene flow, indicated in case of a French highland population by an enlarged number of fixed markers, and hitchhiking effects with respect to selective processes at certain loci following local adaptation and speciation are discussed to explain the population structures found. A possible selection is indicated by five markers, showing significantly different frequency distributions between the class of highland populations and the class of lowland populations.  相似文献

ABSTRACT. The course and projection areas of the tympanal receptor fibres in the thoracic ventral cord were revealed by iontophoresis in the last three larval instars. There were no significant differences between the arrangement in larvae and that in adults. The threshold curves of the auditory organ of the last three instars were measured by recording summed potentials in the tympanal nerve. In the frequency range tested (1–20 kHz), larvae and adults differed only in sensitivity. More detailed information was obtained by single-cell recordings from receptor neurones in the tympanal nerve of last instar larvae. No differences could be shown between the threshold curves, or the suprathreshold activity, of low frequency receptors of last instars and adults. However, the high frequency receptors of the last instars are far less sensitive in the frequency range above 12 kHz. This seems to depend on the different mechanical properties of the tympanum in larvae. The response patterns of some typical ventralcord neurones (G-, K-, B-type) were identified by extracellular single-cell recordings in last instar larvae. Convergence of auditory and vibratory inputs onto the G-neurone and the B-neurone (as is known to exist in the adult) was found in larvae in the final and penultimate instars to be causing similar response patterns.  相似文献

【目的】飞蝗Locusta migratoria(Linnaeus)是我国乃至全世界广泛发生的重要农业害虫,其种群暴发会给农作物造成重大为害和减产。飞蝗种群发生动态受低温及变温的影响,但低温和变温如何作用于胚胎发育和是否诱导胚胎滞育的发生,目前尚不清楚。【方法】本文在对飞蝗胚胎发育特点观察鉴定的基础上,研究了胚胎发育中期恒定低温诱导、诱导后恢复、发育中期变温诱导及发育早期低温诱导等条件对胚胎发育进程的影响。【结果】研究发现,胚胎发育起点温度下非致死温度7.5℃处理促使胚胎发育历期变异增加,而25~30℃高温下胚胎发育整齐。7.5℃处理时间长短对胚胎发育影响不明显,但该低温培育时间长短影响后期常温下的胚胎发育,即7.5℃下长期低温可能促进25℃或30℃下的胚胎的发育。25℃以下变温温度影响胚胎发育进展,但影响胚胎发育的限制因子主要是发育起点以上温度。【结论】因此,温度作为单一环境因子,其特定的低温和变温处理不仅没有诱导胚胎滞育的发生,反而促进后期胚胎在常温下的发育。本研究结果对揭示温度变化对个体发育和种群动态影响及预测飞蝗种群发生有重要的指导意义。  相似文献

Abstract The aggregation components from fecal volatiles of the oriental migratory locust, Locusta migratoria manilensis were identified with gas chromatographic/electroantennographic detector (GC‐EAD), GC‐MS (mass spectrometry) analyses and behavioral bioassays. Both last instar nymphs and adults of the oriental migratory locust have similar aggregation pheromones in their volatiles. A total of 11 electrophysiologically active compounds, namely, hexanal, cyclohexanol, heptanal, phenol, 2,5‐dimethyl‐pyrazine, benzyl alcohol, benzaldehyde, guaiacol, nonanal, 2,6,6‐trimethyl‐2‐cyclohexene‐1,4‐dione and decyl aldehyde were identified in the fecal volatiles of 2‐day‐old immature adult male locusts. Only hexanal, nonanal, benzaldehyde, cyclohexanol and 2,5‐dimethyl‐pyrazine elicited significant aggregation responses in immature 2‐day‐old adult males. However, adult males had significantly lower behavioral responses to synthetics of five single compounds than the blend of cyclohexanol, 2,5‐dimethyl‐pyrazine, benzaldehyde, nonanal, hexanal in ratios of 100: 100: 2: 60: 30 in the range of 30–60 μg/mL. We propose that it is the blend of these five compounds that plays a key role in eliciting and sustaining aggregation in gregarious oriental migratory locusts. These results also showed that the aggregation pheromones of the oriental migratory locust are significantly different from those found in the desert locust.  相似文献

对东亚飞蝗山西临猗和永济2个地理种群的酯酶特性进行了比较研究.非变性聚丙烯酰胺凝胶电泳图谱显示:以a-乙酸萘酯为底物染色,2个东亚飞蝗种群谱带差别不明显.但是,酯酶动力学研究结果表明:以a-乙酸萘酯和α-丁酸萘酯为底物时,永济种群的酯酶活性分别是临猗种群的1.81倍和1.20倍.永济种群酯酶活性的增高可能与非变性聚丙烯酰胺凝胶电泳图谱显示出较临猗种群多出的酶带有关.体外酯酶抑制动力学研究表明:永济和临猗2种群所含酯酶大都为B型酯酶,其含量分别为84.94%和91.47%.永济种群对对氧磷的耐受性要高于临猗种群,我们推测可能与2种群马拉硫磷使用背景不同有关.  相似文献

Single copy nuclear polymorphic (scnp) DNA is potentially a powerful molecular marker for evolutionary studies of populations. However, a practical obstacle to its employment is the general problem of haplotype determination due to the common occurrence of heterozygosity in diploid organisms. We explore here a 'consensus vote' (CV) approach to this question, combining statistical haplotype reconstruction and experimental verification using as an example an indel-free scnp DNA marker from the flanking region of a microsatellite locus of the migratory locust. The raw data comprise 251-bp sequences from 526 locust individuals (1052 chromosomes), with 71 (28.3%) polymorphic nucleotide sites (including seven triallelic sites) and 141 distinct genotypes (with frequencies ranging from 0.2 to 25.5%). Six representative statistical haplotype reconstruction algorithms are employed in our CV approach, including one parsimony method, two expectation-maximization (EM) methods and three Bayesian methods. The phases of 116 ambiguous individuals inferred by this approach are verified by molecular cloning experiments. We demonstrate the effectiveness of the CV approach compared to inferences based on individual statistical algorithms. First, it has the unique power to partition the inferrals into a reliable group and an uncertain group, thereby allowing the identification of the inferrals with greater uncertainty (12.7% of the total sample in this case). This considerably reduces subsequent efforts of experimental verification. Second, this approach is capable of handling genotype data pooled from many geographical populations, thus tolerating heterogeneity of genetic diversity among populations. Third, the performance of the CV approach is not influenced by the number of heterozygous sites in the ambiguous genotypes. Therefore, the CV approach is potentially a reliable strategy for effective haplotype determination of nuclear DNA markers. Our results also show that rare variations and rare inferrals tend to be more vulnerable to inference error, and hence deserve extra surveillance.  相似文献

An albino strain, which had originated from Okinawa, Japan, and a normally coloured strain, which had originated from West Africa, have been used to study density‐dependent morphometric phase characteristics and their changes in adults of the migratory locust, Locusta migratoria (L.). By repeated crossings we also obtained congenic albinos and normal phenotypes and investigated their morphometrics with increasing West African genome, eventually reaching 99.6% West African and 0.4% Okinawa gene pool. The data were analysed by the classical morphometric ratios (F/C and E/F; F = length of the hind femur, C = maximum width of the head, E = length of the fore wings), as well as by canonical discriminant (multivariate) analysis. The latter was based on measurements of F, C and E (as above), as well as of M (minimum width of the pronotum) and H (maximum height of the pronontum). Okinawa albinos showed more solitarious morphometrics and a smaller amplitude of morphometric phase change than West African normal phenotypes. Both the morphometric ratios and the canonical discriminant analysis demonstrated clearly that these differences were caused primarily by the strain (Okinawa vs. West African). However, the pigmentation (albino vs. normal colouration) also affected morphometric phase differences; albinos showed more solitarious morphometrics and somewhat more restricted morphometric phase change than congenic normal phenotypes. The effect of the pigmentation was considerably smaller than that of the strain. The results refute Nolte's claim that albino locusts constitute an extreme solitarious phase, even under crowding. However, Nolte's less extreme claim, that albino locusts have more solitarious morphometrics than normally coloured locusts, is validated by the present results.  相似文献

There is growing interest in quantifying genetic population structure across the geographical ranges of species to understand why species might exhibit stable range limits and to assess the conservation value of peripheral populations. However, many assertions regarding peripheral populations rest on the long-standing but poorly tested supposition that peripheral populations exhibit low genetic diversity and greater genetic differentiation as a consequence of smaller effective population size and greater geographical isolation relative to geographically central populations. We reviewed 134 studies representing 115 species that tested for declines in within-population genetic diversity and/or increases in among-population differentiation towards range margins using nuclear molecular genetic markers. On average, 64.2% of studies detected the expected decline in diversity, 70.2% of those that tested for it showed increased differentiation and there was a positive association between these trends. In most cases, however, the difference in genetic diversity between central and peripheral population was not large. Although these results were consistent across plants and animals, strong taxonomic and biogeographical biases in the available studies call for a cautious generalization of these results. Despite the large number of studies testing these simple predictions, very few attempted to test possible mechanisms causing reduced peripheral diversity or increased differentiation. Almost no study incorporated a phylogeographical framework to evaluate historical influences on contemporary genetic patterns. Finally, there has been little effort to test whether these geographical trends in putatively neutral variation at marker loci are reflected by quantitative genetic trait variation, which is likely to influence the adaptive potential of populations across the geographical range.  相似文献

Two unique serine proteinase isoenzymes (LmHP-1 and LmHP-2) were isolated from the hemolymph of African migratory locust (Locusta migratoria migratorioides) nymphs. Both have a molecular mass of about 23 kDa and are activated by thiol-reducing agents. PMSF abolishes enzymes activity only after thiol activation, while the cysteine proteinase inhibitors E-64, iodoacetamide, and heavy metals fail to inhibit the thiol-activated enzymes. The N-terminal sequence was determined for the more-abundant LmHP-2 isoenzyme. It exhibits partial homology to that of other insect serine proteinases and similar substrate specificity and inhibition by the synthetic and protein trypsin inhibitors pABA, TLCK, BBI, and STI. The locust trypsins LmHP-1 and LmHP-2 constitute a new category of serine proteases wherein the active site of the enzyme is exposed by thiol activation without cleavage of peptide bonds.  相似文献

Ooplasmic activities of potassium and sodium were measured with ion sensitive microelectrodes before and during the period of maximal water uptake which occurs 3–5 days after oviposition for eggs incubated at 37°C. Potassium activity increased from 84 mM in eggs before fertilization at 118 mM in eggs 1 day after fertilization (d1). Sodium activity increased from 8 mM to 29 mM over the same period. These changes exceeded those predicted from the decrease in water content (8%) during the first day after oviposition. Between d1 and d3, potassium and sodium activities decreased to values predicted on the basis of the 88% increase in egg water content. Although water content increased an additional 46% between d3 and d5, ooplasmic sodium activity remained constant at 11 mM and potassium activity increased from 64 mM to 74 mM during this time. Declines in concentrations of sodium and potassium measured in whole eggs by atomic absorption spectrometry mirrored the increase in egg water content. The results suggest that regulation of ooplasmic sodium and potassium activities is accomplished by release of these ions from internal stores, possibly the york spheres. © 1992 Wiley-Liss, Inc.  相似文献

本文研究东亚飞蝗Locusta migratoria manilensis(Meyen)雄性生殖系统组织解剖结构及三维可视化数字模型的构建。采用石蜡切片技术对东亚飞蝗进行组织切片,脱水、透明、HE染色和拍照;并应用冰冻切片技术将冰冻包埋剂包埋后的飞蝗雄性个体进行连续切片,进行截面图像信息采集,建立数据集;通过Photoshop、Image-Pro Plus(IPP)软件对雄性生殖器官截面图像进行分割、处理、序列化和三维重建。通过试验观察分析东亚飞蝗雄性生殖器官精巢、附腺、输精管、精球囊和射精囊及交配器的组织构造;并成功构建了飞蝗雄性生殖系统的三维结构可视化数字模型。该模型可以任意旋转,能从不同角度观察,该试验为研究和教学提供了理论基础。  相似文献

采用聚丙烯酰胺凝胶电泳技术,对分布于华中地区的11个小毛莨(Ranunculus ternatus)居群的遗传分化进行了检测。对8个酶系统17个酶位点上的分析结果表明,该种各居群的各项遗传多样性指标处于一个相对较低的水平:多态位点比率(P)为0-53.0%,平均每位点等位基因数(A)为1-1.647,平均预期杂合度(He)和观察杂合度(Ho)分别为0-0.108和0-0.102。居群间遗传一致度甚高(I=0.9754-0.9991)。根据Nei 's传距离所作出的聚灰分析表明,豫南信阳地区3个居群与湖北省武汉地区8个居群之间关系较远。而在武汉地区,长江以北的居群及长江以南的部分居群分别相聚在一起,用GPS定位方法得到居群间空间距离并据此聚类,该种的遗传分化与地理因素的相关性,并推测出长江的隔离作用加强了两岸居群间的遗传分化,同时发现一个生于独特生境的居群在表型和遗传结构上都已与其他邻近居群有了很大分异,由于该居群在所检测的酶位点上均无特有等位基因出现,作者认为不宜将其作为新种或新变种处理。  相似文献
