Prebiotic ribose synthesis: A critical analysis   总被引:3,自引:0,他引:3  
The discovery of catalytic ability in RNA has given fresh impetus to speculations that RNA played a critical role in the origin of life. This question must rest on the plausibility of prebiotic oligonucleotide synthesis, rather than on the properties of the final product. Many cliams have been published to support the idea that the components of RNA were readily available on the prebiotic earth. In this article, the literature cited in support of the prebiotic availability of one subunit, D-ribose, is reviewed to determine whether it justifies the claim.Polymerization of formaldehyde (the formose reaction) has been the single reaction cited for prebiotic ribose synthesis. It has been conducted with different catalysts: numerous basic substances, neutral clays and heat, and various types of radiation. Ribose has been identified (yields are uncertain, but unlikely to be greater than 1%) in reactions run with concentrated (0.15 M or greater) formaldehyde. It has been claimed in reactions run at lower concentration, but characterization has been inadequate, and experimental details have not been provided.The complex sugar mixture produced in the formose reaction is rapidly destroyed under the reaction conditions. Nitrogenous substances (needed for prebiotic base synthesis) would interfere with the formose reaction by reacting with formaldehyde, the intermediates, and sugar products in undesirable ways.The evidence that is currently available does not support the availability of ribose on the prebiotic earth, except perhaps for brief periods of time, in low concentration as part of a complex mixture, and under conditions unsuitable for nucleoside synthesis.  相似文献

A theoretical mechanism resulting in the prebiotic appearance of enantiopure ribose, which would be needed for the origin of RNA and the “RNA world” is proposed. The mechanism simultaneously explains the emergence of biological homochirality and could answer the question of why nucleic acids are based on ribose rather than another sugar. Cleavage of certain non-chiral mineral crystals is known to lead to formation of chiral surfaces. In a chromatography-like process a mixture of racemic carbohydrates originating from the formose reaction is proposed to have been separated on such a chiral surface. Monosaccharides interact with surfaces through their hydroxyl groups, either by hydrogen bond formation or complex formation with metal ions. α-Ribopyranose, which has all hydroxyl groups on one side of the ring, is known to interact more strongly than other sugars with surfaces, as corroborated by certain chromatographic and electrophoresis data. A similar scenario leading to enantiopure ribose is separation on a flat, but not necessarily chiral surface in the presence of a strong electric field capable of orienting highly polar derivatives of sugars. The prebiotic possibilities for the synthesis of interstellar carbohydrates through a protic variant of the formose reaction under gas phase conditions were studied. Ab initio calculations were used to evaluate potential mechanisms. Based on considerations of barrier heights and temperature variations in the Interstellar Medium the plausibility of extended sugar synthesis will be discussed.  相似文献

Formose syrup was studied as a carbon source for growth of a series of microorganisms obtained from various collections. Approximately 80 strains of bacteria, yeasts, and molds were inoculated into a medium containing formose syrup and mineral salts supplemented with small amounts of yeast extract and casein hydrolysate to supply accessory growth factors. Two preparations of formose syrup, produced by two different laboratories, were employed. Formose syrup I, characterized by a low sugar content, was poorly utilized; syrup II, containing a higher sugar concentration, was utilized to a greater extent. Two strains of Aerobacter acrogenes yielded 1.3 g dry cell mass from an initial charge of 10 g of formose II solids, whereas growth on 10 g of D -glucose amounted to 3.7 g. Klebsiella aerogenes MIT-B44, the best microbial strain isolated from soil by an enrichment technique, produced 1.3 g cells from 10 g fromose syrup II solids in supplemented medium; in direct comparisons, it produced 10–15% more cell 0.7–0.9 g cells per 10 g formose and grew with a doubling time of 55–70 min. Under such conditions, its macromolecular composition was 52% protein, 22% RNA, and 2% DNA. Although the apparent yield of cells from formose was only 8–11%, the actual yield based on formose utilized was 30%, the same as observed with glucose. A second strain was isolated from soil by enrichment with spent broth from K. aerogenes. This unidentified gram-negative, short rod-shaped bacterium grew in mixed culture with strain MIT-B44; in unsupplemented media they produced 1.55 g cells from 10 g formose II solids and 2.9 g cells from 10 g glucose.  相似文献

Pulse radiolysis studies of the hydrogen atom transfer ("repair") reaction from the sulfhydryl-containing (RSH) compound dithiothreitol (DTT) to the DNA sugar deoxyribose and to several related sugars have been undertaken. The H transfer reaction is measured by monitoring the transient absorbance of the radical-anion RSSR-. The H atom transfer reactions for some sugars were fitted by a single time exponential function, but other sugars exhibited both a fast and a slow component (approximately 10-fold difference in rates) to the reaction. The reaction rates for the slow stage of the reaction between DTT and the sugars ranged from 0.5 X 10(7) dm3 mole-1 sec-1 for ribose-5-phosphate to 9 X 10(7) dm3 mole-1 sec-1 for 2-deoxyglucose. The maximum extent of total repair varied from 60% for ribose-5-phosphate to 100% for 2-deoxyglucose. The rate of repair, the extent of repair, and the appearance of more than one component of repair seem to depend on several factors: The occurrence of radical-radical reactions in the system is indicated by the demonstration of a dose dependence of the reaction kinetics, and this affects the observed rate of formation of RSSR-. Sugars with a deoxy group on the 2-carbon atom seem to have enhanced rates and extents of repair and to exhibit both fast and slow components to the reaction. The presence of a phosphate group on the sugar causes a decrease in the rate and extent of repair. The biological relevance of the reactions studied herein is discussed and the rates obtained are compared with rates for repair of damage in certain radiobiological systems.  相似文献

In an effort to explore the contribution of the sugar constituents of pharmaceutically relevant glycosylated natural products, chemists have developed glycosylation methods for the generation of 'glycorandomized' libraries. Each member of these libraries is uniquely differentiated by an attached carbohydrate. Recently, two complementary glycorandomization strategies have emerged: chemoenzymatic glycorandomization, a biocatalytic approach dependent upon the substrate promiscuity of enzymes to activate and attach sugars to natural products, and neoglycorandomization, an efficient one-step chemical sugar ligation reaction that does not require prior sugar protection or activation. These strategies are likely to have a significant impact on fundamental glycoscience and drug discovery.  相似文献

The sandfly Phlebotomus orientalis Parrot (Diptera: Psychodidae) is the vector of visceral leishmaniasis in eastern and Upper Nile regions of Sudan, where vector infection rates of over 7% have been reported. Sugars are known to be important for development of the parasite and for increasing the survival and oviposition rates of several species of sandflies. In the present study we have analysed the sugars present in the guts of individuals and groups of male and female P. orientalis and compared these with sugars from several potential local plant sources: Acacia seyal, Balanites aegyptiaca and Combretum kordofanum. The distribution of these trees in Sudan is closely correlated with that of P. orientalis. Only 20% of individually analysed female sandflies had significant amounts of sugars present suggesting that P. orientalis either digest their sugar meal quickly or do not require regular sugar meals. Interestingly, the sugars present in the males were significantly different to those found in the females, indicating that they had fed on different sugar sources. There was evidence that fruit sugars from Balanites aegyptiaca, Combretum kordofanum and aphid or coccid honeydew are utilized by male and female P. orientalis. There was evidence to indicate that female P. orientalis feeds directly on honeydew. There was no evidence to indicate that direct feeding on leaves is a typical source for the sugar meal. There was no melizitose and only a very small amount of turanose present in the male, suggesting that honeydew was not an important sugar source for males.  相似文献

Simple and rapid methods for the preparation of phenylthiocarbamyl (PTC) derivatives of amino sugars and amino sugar alcohols and their quantitative determination with high sensitivity (less than 10 pmol) by C18 reversed-phase high-performance liquid chromatography are described. Rapid sample preparation of the phenyl isothiocyanate (PITC)-derivatized amino sugars and amino sugar alcohols was achieved by a simple extraction of the reaction mixture with chloroform to remove the excess PITC and its adducts. Baseline separation of the PTC derivatives of amino sugars and amino sugar alcohols was obtained within 30 min, using a simple solvent system consisting of 0.2% each of n-butylamine, phosphoric acid, and tetrahydrofuran. The mobile phase containing n-butylamine, in conjunction with a C18 stationary phase, mimics the conditions for the separation of carbohydrates on an amino-bonded column. GlcNH2 and GalNH2 derived from the initial protein-sugar linkages were also separated from the amino acids for quantitative estimation of sugar chains in glycoproteins. Amino sugar alcohols gave single reaction products with PITC while the reaction with amino sugars was accompanied by the formation of secondary products. Apparently the secondary products were formed in an acid-catalyzed intramolecular cyclization of the PTC-hexosamines involving the aldehyde functional group. Conditions were developed to stop the transformations and maintain the stability of PTC derivatives for their convenient determination by HPLC.  相似文献

Reaction of triose sugars with ammonia under anaerobic conditions yielded autocatalytic products. The autocatalytic behavior of the products was examined by measuring the effect of the crude triose–ammonia reaction product on the kinetics of a second identical triose–ammonia reaction. The reaction product showed autocatalytic activity by increasing both the rate of disappearance of triose and the rate of formation of pyruvaldehyde, the product of triose dehydration. This synthetic process is considered a reasonable model of origin-of-life chemistry because it uses plausible prebiotic substrates, and resembles modern biosynthesis by employing the energized carbon groups of sugars to drive the synthesis of autocatalytic molecules.  相似文献

Escherichia coli K-12 mutants lacking the adenosine 5'-monophosphate-activated pyruvate kinase have been isolated accidentally and used to prepare further mutants additionally devoid of the fructose bisphosphate-activated pyruvate kinase. Such double mutants totally devoid of pyruvate kinase activity still grow well under aerobic conditions on sugars that are catabolized by the phosphoenolpyruvate (PEP):sugar phosphotransferase system, but they grow poorly on non-phosphotransferase system sugars. This suggests that although pyruvate kinase plays a major role in the formation of pyruvate from PEP during growth on non-phosphotransferase system sugars, the operation of the PEP:sugar phosphotransferase system can contribute significantly to pyruvate production from PEP. In the absence of pyruvate kinase and an active PEP:sugar phosphotransferase system the methylglyoxal glycolytic bypass may also function to some extent for the formation of pyruvate during the catabolism of simple hexose sugars. No unique physiological role can yet be ascribed to the adenosine 5'-monophosphate-activated pyruvate kinase as a result of these studies.  相似文献

The use of chilled, extended semen in dog breeding is becoming increasingly popular as preparation and transportation is less expensive and regulations are often less complicated than for frozen semen. Sugar is one of the main constituents in semen extenders, and glucose and fructose are metabolized in separate pathways by freshly ejaculated dog sperm. In this study, glucose, fructose or an equal mixture of both were used in an egg-yolk-tris (EYT) extender at two different concentrations (10 and 70 mM). EYT extender without sugar supplementation, providing only the glucose (3-4 mM) originating from the egg-yolk, served as a control. The longevity of the chilled semen at 5 degrees C was 23 days: the quality of physical and functional characteristics decreasing with time. Glucose and fructose had a strong influence on motility and movement patterns of chilled canine semen. The beneficial effect of 70 mM sugar concentrations compared to 10 mM and the control was pronounced, and maintained sperm motility > or = 70% for 8 days of storage, compared to for 4 days in the control extender. Fructose maintained higher sperm motility than did glucose and the mixture. VAP values were higher in sugar-supplemented extenders (P < 0.05). Neither type nor concentration of the two sugars influenced sperm plasma membrane, acrosome integrity or the acrosome reaction following ionophore challenge (ARIC). Sugar consumption by dog sperm varied between the different periods of storage and with sugar concentrations provided in the extenders. Glucose consumption by dog sperm was greater than fructose consumption when both sugars were present in equal amounts, indicating that dog sperm used glucose in preference to fructose. In conclusion, the major influence of the two sugars on chilled semen was to support motility. EYT extender supplemented with fructose at a concentration of 70 mM was found to be the best of the tested extenders for long-term preservation of chilled canine semen.  相似文献

This paper describes the synthesis and antiproliferative activity of conjugates of genistein (1) and unsaturated pyranosides. Constructs linking genistein with a sugar moiety through an alkyl chain were obtained in a two-step synthesis: in a first step genistein was converted into an intermediate bearing an ??-hydroxyalkyl substituent, containing two, three or five carbon atoms, at position 7, while the second step involved Ferrier glycosylation reaction, employing glycals. Antiproliferative activity of several genistein derivatives was tested in cancer cell lines in vitro. The most potent derivative, Ram-3 inhibited the cell cycle, interacted with mitotic spindles and caused apoptotic cell death. Neither genistein nor the sugar alone were able to influence the mitotic spindle organization. Our results indicate, that conjugation of genistein with certain sugars may render the interaction of derivatives with new molecular targets.  相似文献

Apricot seeds could be obtained as a byproduct in different juice or conserve producing industries. Disposal of large amount of apricot seeds is wasting of potentially precious sources of phytochemicals. This study encompassed apricot cultivars that have different origin and flowering time with the aim of providing valuable information about the sugar content in its kernels. High-performance anion-exchange chromatography with pulsed amperometric detection was used for the determination of 22 sugars in the kernels of 70 apricot cultivars. The most common sugars in the apricot kernels are sucrose, fructose, and glucose whose ratio proved to be ripening time dependent (1:1:1 in the early, 1:2:3 in medium and 1:3:3 in the late cultivars). The kernels of the apricot with shorter vegetation period had the highest sum of glucose and fructose compared to other groups. Other sugar components were present in different levels as minor constituents depending on the ripening time. Also, most common sugars and some low-level sugars were strongly correlated to each other, indicating the normal metabolic carbohydrate pathway. To understand the distribution modes of sugars, a principal component analysis was performed.  相似文献

Nucleoside diphosphate sugars serve in essential roles in metabolic processes. They have, therefore, been used in mechanistic studies on glycosylation reactions, and their analogues have been synthesised as enzyme and receptor inhibitors. Despite extensive biochemical research, little is known about their chemical reactions. In the present work the chemical cleavage of two different types of nucleoside diphosphate sugars has been studied. UDP-Glc is phosphorylated at the anomeric carbon, whereas in ADP-Rib C-1 is unsubstituted, allowing hence the equilibrium between cyclic hemiacetal and acyclic carbonyl forms. Due to the structural difference, these substrates react via different pathways under slightly alkaline conditions: while UDP-Glc reacts exclusively by a nucleophilic attack of a glucose hydroxyl group on the diphosphate moiety, ADP-Rib undergoes a complex reaction sequence that involves isomerisation processes of the acyclic ribose sugar and results in a release of ADP.  相似文献

A spectrophotometric method is described for estimating the carbohydrate content of acid-hydrolyzed glycoproteins. The method is based on the alkaline ferricyanide reaction with reducing sugars and is accurate in the 1–25 nmole range. Using a simple two-step ion-exchange column procedure both the total sugar content and the proportion of neutral and amino sugars can be determined.  相似文献

Effects of zeatin on amino acid and sugar contents of detached radish (Raphanus sativus L.) cotyledons were investigated to determine if accumulation of these solutes contributes to cytokinin-enhanced growth. Protein and amino acid levels were not significantly affected, but in cotyledons incubated in light the hormone caused greater accumulations of reducing sugars than occurred in light controls. Continuous fluorescent light or a few minutes of red light increased both the growth rate and the reducing sugar levels compared to dark controls. A far red treatment following red light overcame the promoting effect of the latter. Amounts of reducing sugars were closely associated with growth under the above conditions. Activity of an unidentified amylase was elevated by continuous light or a red light treatment (nullifiable by far red), suggesting that reducing sugars were derived from starch. Zeatin-treated cotyledons exhibited less amylase activity than did light controls, perhaps implicating cytokinin-stimulated conversion of fats to sugars in light. In darkness zeatin promoted cotyledon growth but did not increase sugar levels nor amylase activity, suggesting that enhanced ion accumulation also contributes to the more rapid water uptake leading to growth.  相似文献

We review current knowledge of the most abundant sugars, sucrose, maltose, glucose and fructose, in the world's major crop plants. The sucrose‐accumulating crops, sugar beet and sugar cane, are included, but the main focus of the review is potato and the major cereal crops. The production of sucrose in photosynthesis and the inter‐relationships of sucrose, glucose, fructose and other metabolites in primary carbon metabolism are described, as well as the synthesis of starch, fructan and cell wall polysaccharides and the breakdown of starch to produce maltose. The importance of sugars as hormone‐like signalling molecules is discussed, including the role of another sugar, trehalose, and the trehalose biosynthetic pathway. The Maillard reaction, which occurs between reducing sugars and amino acids during thermal processing, is described because of its importance for colour and flavour in cooked foods. This reaction also leads to the formation of potentially harmful compounds, such as acrylamide, and is attracting increasing attention as food producers and regulators seek to reduce the levels of acrylamide in cooked food. Genetic and environmental factors affecting sugar concentrations are described.  相似文献
