[3H]Lysergic acid diethylamide (LSD) in the presence of 40 nM ketanserin labeled the 5-HT1A receptor subtype in rat hippocampal membranes. In the presence of guanosine triphosphate (GTP), the Bmax and affinity of [3H]LSD binding to the 5-HT1A binding site were significantly decreased. [3H]LSD in the presence of 40 nM WB4101 labeled the 5-HT2 receptor subtype in homogenates of rat frontal cortex. In contrast to the effect on [3H]LSD binding to the 5-HT1A binding site, GTP produced no significant effect on either the Bmax or the KD of [3H]LSD binding to the 5-HT2 binding site. Competition of 5-HT for [3H]LSD binding to the 5-HT2 binding site was best described by a computer-derived model assuming two binding sites. In the presence of GTP, the 5-HT competition curve was shifted significantly to the right with an approx. 3-fold increase in the IC50. These binding characteristics are consistent with [3H]LSD acting as an antagonist at the 5-HT2 receptor which has multiple affinity states for agonists and is coupled to a guanine nucleotide regulatory subunit. Thus, [3H]LSD has binding characteristics consistent with it acting as an agonist at the 5-HT1A receptor subtype but as an antagonist at the 5-HT2 receptor subtype in rat brain.  相似文献

The activity of the muscarinic cholinergic system (acetylcholine, ACh; acetylcholinesterase, AChE; choline acetyltransferase, ChAT; muscarinic acetylcholine receptors) was studied in the carp brain. The ACh content (13.9 ± 1.1 nmol/g wet tissue) was estimated by gas chromatography after microwave irradiation focused to the head. The AChE and ChAT activities were 153 ± 13 nmol/min/mg protein and 817 ± 50 pmol/min/mg protein, respectively. The characteristics of [3H](−)quinuclidinyl benzilate ([3H](−)QNB) and [3H]pirenzepine ([3H]PZ) binding were also studied in brain membranes. Their specific binding was linearly dependent on the protein content and they appeared to bind with high affinity to a single, saturable binding site. A dissociation constant (Kd) of 47 ± 6.3 pM and a maximum number of binding sites (Bmax) of 627 ± 65 fmol/mg protein were obtained for [3H](−)QNB, with a Kd value of 3.85 ± 0.67 nM and a Bmax value of 95.3 ± 6.25 fmol/mg protein for [3H]PZ binding. The [3H]PZ binding amounted to only 15% of the [3H](−)QNB-labeled sites, as estimated from the ratio of the Bmax values of [3H](−)QNB and [3H]PZ, suggesting a low density of M1 subtype. Atropine sulfate, atropine methylnitrate and PZ inhibited the binding of both radioligands with Hill slopes (nH) close to unity. The nH value of AF-DX 116 was close to 1 against [3H](−)QNB binding, while it was 0.75 against [3H]PZ binding. The displacement curves of oxotremorine and carbachol were shallow for the binding of both radioligands. The rank order of potency of muscarinic ligands against [3H](−)QNB binding (Ki nM) was atropine sulfate (0.55) > atropine methylnitrate (1.61) > PZ (61.19) > oxotremorine (156.3) > AF-DX 116 (307) > carbachol (1301), while in the case of [3H]PZ binding it was atropine sulfate (0.24) > atropine methylnitrate (0.34) > PZ (10.38) > AF-DX 116 (55.87) > oxotremorine (62.79) > carbachol (1696). The results indicate the presence of a well-developed muscarinic cholinergic system with predominantly M2 receptors in the carp brain.  相似文献

High affinity, specific [3H]5-hydroxytryptamine (5-HT) binding to spinal cord synaptosomes was examined to identify the 5-HT receptor subtypes present. Computer nonlinear regression analysis of competition studies employing 8-OH-DPAT indicated that this 5-HT1A selective agonist demonstrated high affinity competition (Ki = 1.3 nM) for 24.6 ± 0.7% of the total [3H]5-HT binding sites. Competition studies employing the 5-HT1B selective agonist RU24969, in the presence of 100 nM 8-OH-DPAT, indicated that RU24969 demonstrated high affinity (Ki = 1.1 nM) competitive inhibition for 26.2 ± 1.4% of all [3H]5-HT binding sites. Neither 5-HT1C, 5-HT1D, 5-HT2 nor 5-HT3 selective compounds demonstrated any high affinity competition for the residual 49% of specific [3H]5-HT binding. Therefore, three major classes of [3H]5-HT binding sites could be demonstrated in spinal cord synaptosomes: 5-HT1A, 5-HT1B and a novel [3H]5-HT binding site which respectively represented 25, 26 and 49% of spinal cord synaptosomal [3H]5-HT binding. Further studies focusing on the function of the latter binding site are needed to determine if the presently identified novel binding site is the major 5-HT1 receptor subtype present in spinal cord.  相似文献

In the absence of detergent, specific binding of [3H]GR65630, a 5-hydroxytryptamine3 (5-HT3) antagonist, determined in the presence of 5-HT3 receptor antagonist ICS205-930, was at most 30% of the total binding. To decrease the level of nonspecific binding, the effects of detergents on [3H]GR65630 binding to rat cortical membranes were investigated. The use of a detergent (0.1% Lubrol PX or Triton X-100) decreased nonspecific binding, increasing the proportion of specific binding to 70% of total binding. In the presence of 0.1% Triton X-100, binding of [3H]GR65630 was rapid, reversible and saturable at 25°C. The rank order of 5-HT3 receptor active drugs in inhibiting [3H]GR65630 binding was quipazine > ICS205-930 > 2-methyl-5-HT = 5-HT > metoclopramide, which confirmed that [3H]GR65630 efficiently labeled 5-HT3 receptors in the presence of Triton X-100. Triton X-100 improved 5-HT3 receptor binding with rat brain membranes.  相似文献

The interaction of the nicotinic agonist (R,S)-3-pyridyl-1-methyl-2-(3-pyridyl)-azetidine (MPA) with different nicotinic acetylcholine receptor (nAChR) subtypes was studied in cell lines and rat cortex. MPA showed an affinity (Ki = 1.21 nM) which was higher than anatoxin-a > (−)-nicotine > (+)-[R]nornicotine > (−)-[S]nornicotine > and (+)-nicotine, but lower than cytisine (Ki = 0.46 nM) in competing for (−)-[3H]nicotine binding in M10 cells, which stably express the recombinant 4β2 nAChR subtype. A one-binding site model was observed in all competing experiments between (−)-[3H]nicotine binding and each of the agonists studied in M10 cells. MPA showed a 13-fold higher affinity for (−)-[3H]nicotine binding sites compared to the [3H]epibatidine binding sites in rat cortical membranes. In human neuroblastoma SH-SY5Y cells, which predominantly express the 3 nAChR subunit mRNA, MPA displaced [3H]epibatidine binding from a single population of the binding sites with an affinity in the same nM range as that observed MPA in displacing [3H]epibatidine binding in rat cortical membranes. Chronic treatment of M10 cells with MPA significantly up-regulated the number of (−)-[3H]nicotine binding sites in a concentration dependent manner. Thus MPA appears to have higher affinity to 4-subunit containing receptor subtype than 3-subunit containing receptor subtype of nAChRs. Furthermore MPA binds to 4β2 receptor subtype with higher affinity than (−)-nicotine and behaves, opposite to cytisine, as a full agonist in up-regulating the number of nAChRs. © 1998 Elsevier Science Ltd. Total 5-HT binding sites and 5-HT1A receptor density was measured in brain regions of rats treated with imipramine (5 mg/kg body wt), desipramine (10 mg/kg body wt) and clomipramine (10 mg/kg body wt), for 40 days, using [3H]5-HT and [3H]8-OH-DPAT, respectively. It was observed that chronic exposure to tricyclic antidepressants (TCAs) results in significant downregulation of total [3H]5-HT binding sites in cortex (42–76%) and hippocampus (35–67%). The 5-HT1A receptor density was, however, decreased significantly (32–60%) only in cortex with all the three drugs. Interestingly, in hippocampus imipramine treatment increased the 5-HT1A receptor density (14%). The affinity of [3H]8-OH-DPAT was increased only with imipramine treatment both in cortex and hippocampus. The affinity of [3H]5-HT to 5-HT binding sites in cortex was increased with imipramine treatment and decreased with desipramine and clomipramine treatment. 5-HT sensitive adenylyl cyclase (AC) activity was significantly increased in cortex with imipramine (72%) and clomipramine (17%) treatment, whereas in hippocampus only imipramine treatment significantly increased AC activity (50%). In conclusion, chronic treatment with TCAs results in downregulation of cortical 5-HT1A receptors along with concomitant increase in 5-HT stimulated AC activity suggesting the involvement of cortical 5-HT1A receptors in the mechanism of action of TCAs.  相似文献

We have further characterized the 5-HT3 receptors in rat and rabbit tissues by evaluating the binding of the 5-HT3 receptor ligand, [3H]GR67330 to homogenates of rabbit ileum, rat ileum and rat brain (entorhinal cortex). In each tissue specific [3H]GR67330 binding represented a single saturable, high affinity site (Kd = 0.14, 0.18, 0.076 nM in rabbit ileum, rat ileum and rat brain respectively). The densities of sites present in rabbit and rat ileum were similar to that present in rat brain (Bmax = 63, 47, 72 fmol/mg protein in rabbit ileum, rat ileum and rat brain respectively).

In each tissue, 5-HT3 receptor agonists and antagonists potently competed for [3H]GR67330 binding. Derived inhibition constants were similar in rat ileum and brain. However marked differences in IC50s were apparent for rabbit ileum compared with rat brain or ileum. These were most apparent with agonists. Thus, mCPBG [1-(meta-chlorophenylbiguanide)], phenylbiguanide, 5-HT and 2-methyl 5-HT were at least 5 times less potent to inhibit [3H]GR67330 binding in rabbit ileum than rat brain. The most pronounced differences were evident with phenylbiguanide and mCPBG which were 70 and 300 times less potent in the rabbit ileum respectively compared with the rat tissues. These differences were unlikely to be due to depletion effects because tissue combination experiments (rabbit ileum and rat brain) yielded biphasic inhibition curves for phenylbiguanide with affinities for each component similar to those in the individual tissues. Antagonist affinities also varied between the rabbit and rat tissues, although less markedly. Amongst the antagonists, the most marked differences were apparent with SDZ 206–830 and quipazine each being 10 times less potent to inhibit binding to rabbit than rat tissue.

Hill coefficients for inhibition of binding varied with tissue. In rat brain, as previously described for [3H]GR67330, Hill coefficients for agonist (and quipazine) inhibition of binding were greater than unity. This was less marked in rat and rabbit ileum tissues.

The present studies provide further evidence for species variation in 5-HT3 receptors.  相似文献

Aromatic analogs of arcaine were shown to have inhibitory effects on the binding of the channel blocking drug [3H]MK-801 to the NMDA receptor complex. The most potent compound of the series was an N,N′-bis(propyl)guanidinium which inhibited [3H]MK-801 binding with an IC50 of 0.58 μM and an IC50 of 12.17 μM upon addition of 100 μM spermidine. The increase in IC50 upon addition of spermidine suggests competitive antagonism between the inhibitor and spermidine at the arcaine-sensitive polyamine site of the NMDA receptor complex.  相似文献

Eighteen corpora striata from normal human foetal brains ranging in gestational age from 16 to 40 weeks and five from post natal brains ranging from 23 days to 42 years were analysed for the ontogeny of dopamine receptors using [3H]spiperone as the ligand and 10 mM dopamine hydrochloride was used in blanks. Spiperone binding sites were characterized in a 40-week-old foetal brain to be dopamine receptors by the following criteria: (1) It was localized in a crude mitochondrial pellet that included synaptosomes; (2) binding was saturable at 0.8 nM concentration; (3) dopaminergic antagonists spiperone, haloperidol, pimozide, trifluperazine and chlorpromazine competed for the binding with IC50 values in the range of 0.3–14 nM while agonists—apomorphine and dopamine gave IC50 values of 2.5 and 10 μM, respectively suggesting a D2 type receptor.

Epinephrine and norepinephrine inhibited the binding much less efficiently while mianserin at 10 μM and serotonin at 1 mM concentration did not inhibit the binding. Bimolecular association and dissociation rate constants for the reversible binding were 5.7 × 108 M−1 min−1 and 5.0 × 10−2 min−1, respectively. Equilibrium dissociation constant was 87 pM and the KD obtained by saturation binding was 73 pM.

During the foetal age 16 to 40 weeks, the receptor concentration remained in the range of 38–60 fmol/mg protein or 570–1080 fmol/g striatum but it increased two-fold postnatally reaching a maximum at 5 years Significantly, at lower foetal ages (16–24 weeks) the [3H]spiperone binding sites exhibited a heterogeneity with a high (KD, 13–85 pM) and a low (KD, 1.2–4.6 nM) affinity component, the former accounting for 13–24% of the total binding sites. This heterogeneity persisted even when sulpiride was used as a displacer. The number of high affinity sites increased from 16 weeks to 24 weeks and after 28 weeks of gestation, all the binding sites showed only a single high affinity.

GTP decreased the agonist affinity as observed by dopamine competition of [3H]spiperone binding in 20-week-old foetal striata and at all subsequent ages. GTP increased IC50 values of dopamine 2 to 4.5 fold and Hill coefficients were also increased becoming closer to one suggesting that the dopamine receptor was susceptible to regulation from foetal life onwards.  相似文献

The binding of [3H]proctolin to oviduct membranes has been analyzed to investigate the nature of proctolin binding sites in the oviduct. Proctolin binding was found to be time dependent, proportional to concentration of membrane protein, saturable, specific and reversible. Two apparent proctolin binding sites were recognized. The first had a Kd of 400 ± 82 nM and a Bmax of 23.7 ± 6.7 pmol/mg protein. The second had a Kd of 2.4 ± 0.2 μM and a Bmax of 96.3 ± 16.7 pmo/mg protein.

Binding was specific in thatcompetition experiments with a wide range of peptides showed that only Arg-Tyr-Leu-Pro-Ala was an effective competitor at μM concentrations. All other peptides examined weekly reduced proctolin binding at concentrations above 50 μM. Certain peptides were found to potentiate [3]pproctolin binding at low μM concentrations (1–10 μM) and to inhibit proctolin binding at higher concentrations. The significance of these findings is discussed.  相似文献

The present study investigated the effects of damage to the basal forebrain cholinergic system upon [3H]ketanserin binding in the neocortex and hippocampus of monkeys. [3H]Ketanserin specifically binds to serotonin type-2 receptor sites. Lesions were placed in the medial septal area, nucleus basalis, or both regions. Ten months later, [3H]ketanserin binding was increased in the neocortex, but not in the hippocampus, while levels of choline acetyltransferase (acetyl-CoA: choline O-acetyltransferase, EC 2.3.1.6) activity decreased in the neocortex and hippocampus. Changes in the levels of choline acetyltransferase and [3H]ketanserin binding were correlated significantly in the neocortex (r = −0.64, P < 0.025), but not in the hippocampus. The data suggest that degeneration of the basal forebrain cholinergic system may alter serotonergic function in the neocortex.  相似文献

Previous studies delineated two classes of δ binding sites; a δ binding site not associated with the opioid receptor complex, termed the δncx site, and a δ site associated with the opioid receptor complex, termed the δcx site. The δncx site has high affinity for [ -Pen2, -Pen5]enkephalin, and is synonymous with what is now identified as the δ1 binding site. Pretreatment of membranes with the δ-selective acylating agents FIT, or (+)-trans-SUPERFIT, deplete membranes of the δncx binding site, which permits the selective labeling of the δcx binding site with [3H][ -Ala2,Leu5]enkephalin. The present study compared the properties of the δcx binding site present in brain membranes pretreated with (+)-trans-SUPERFIT with the properties of the δcx site present in untreated membranes. The major findings are: 1) pretreatment of membranes with (+)-trans-SUPERFIT decreased the IC50 values of δ-preferring drugs, and increased the IC50 values of μ-preferring drugs, for the δcx binding site; 2) the degree of δ selectivity was highly correlated with the magnitude of the (+)-trans-SUPERFIT-induced shift in the IC50 values; 3) the ligand-selectivity patterns of the μ and δcx sites present in (+)-trans-SUPERFIT-pretreated membranes were poorly correlated; 4) whereas μ-preferring drugs were noncompetitive inhibitors of [3H][ -Ala2,Leu5]enkephalin binding to the δcx site, δ-preferring drugs were competitive inhibitors. Viewed collectively, these data support the hypothesis that the μ and δcx binding sites are distinct, provide additional evidence for δ receptor heterogeneity, and suggest that ( (+)-trans-SUPERFIT-pretreated membranes will provide a useful preparation for studying the δcx binding site.  相似文献

[3H]Neurotensin (NT) was found to bind specifically and with high affinity to crude membranes prepared from rat uterus. Scatchard analysis of saturation binding studies indicated that [3H]NT apparently binds to two sites (high affinity Kd 0.5 nM; low affinity Kd 9 nM) with the density of high affinity sites (41 fmoles/mg prot.) being about one-third that of the low affinity sites (100 fmoles/mg prot.). In competition studies, NT and various fragments inhibited [3H]NT binding with the following potencies (IC50): NT 8–13 (0.4 nM), NT 1–13 (4 nM), NT 9–13 (130 nM), NT 1–11, NT 1–8 (>100 μM). Quantitatively similar results were obtained using brain tissue. These findings raise the possibility of a role for NT in uterine function.  相似文献

The affinity of amitriptyline for muscarinic receptors in rat brain areas was studied using autoradiographic techniques including image analysis. As shown by competitive inhibition of [3H]-l-quinuclidinyl benzilate binding, amitriptyline was found to be a potent inhibitor of muscarinic receptors throughout the rat brain. Muscarinic receptors in the external layers of the cortex displayed a high affinity for amitriptyline (IC50 = 65.8 ± 2.1 nM), while the hippocampal regions had somewhat lower affinities (e.g. IC50 = 96.3 ± 3.4 nM). Amitriptyline bound with lower affinity in the thalamus and various midbrain regions, such as the paraventricular nucleus of the thalamus and the superior colliculus, which had IC50 values of 112 ± 6.8 and 117 ± 32.6 nM, respectively. Other midbrain regions displayed higher affinities, for example, the substantia nigra had an IC50 value of 62.8 ± 0.9 nM. The data show that amitriptyline binds with high affinity to muscarinic receptors with a modest subtype selectivity that is unlike that of either pirenzepine or AF-DX 116. In addition, amitriptyline at concentrations of 10 nM-1 μM antagonized the oxotremorine-induced inhibition of acetylcholine release in cortical nerve endings, demonstrating activity at M2 autoreceptors.  相似文献

The first effects of 3,4-methylen-dioxy-metamphetamine (MDMA, “ecstasy”), on serotonin 1A (5-HT1A) receptors in rat hippocampus were determined by means of [3H]-8-hydroxy-dipropylamino-tetralin ([3H]-8-OH-DPAT) and 5′guanosine-(γ-[35S]-thio)triphosphate ([35S]-GTPγS) binding as well as inhibition of forskolin (FK)-stimulated adenylyl cyclase (AC) activity. The study was completed by [35S]-GTPγS functional autoradiography experiments carried out in frontal sections of rat brain, including the hippocampal region. Results showed that MDMA was either able to displace [3H]-8-OH-DPAT binding (Ki  500 nM) or to reduce the number of specific sites (Bmax) without affecting Kd. The drug also failed to change the [35S]-GTPγS binding or to inhibit AC velocity, underlying its behavior as a non-competitive 5-HT1A receptor antagonist. Further, MDMA (1 or 100 μM), partially antagonized either [35S]-GTPγS binding stimulation of the agonists 5CT and 8-OH-DPAT or the AC inhibition induced by 5CT and DP-5CT. However, in contrast to binding studies, in AC assays the amphetamine displayed an effect also on EC50, always being less potent than the reference antagonist WAY100,635. In functional autoradiography, MDMA behaved either as a partial 5-HT1A antagonist in limbic areas or, added alone, as an agonist, increasing the coupling signal presumably through 5-HT release from synapses. Interestingly, the selective 5-HT re-uptake inhibitor (SSRI) fluoxetine had no effect on MDMA [35S]-GTPγS binding activation. This latter finding indicates that the amphetamine can release 5-HT via alternative mechanisms to 5-HT transporter binding, probably via membrane synaptic receptors or vesicular transporters. The release of other transmitters is not excluded. Therefore, our results encourage at extending the study of MDMA biochemical profiles, in the attempt to elucidate those amphetamine-induced pathways with a potential for neurotoxicity or psycho-stimulant activity.  相似文献

A series of tris-azaaromatic quaternary ammonium salts has been synthesized and evaluated for their ability to inhibit neuronal nicotinic acetylcholine receptors (nAChRs) mediating nicotine-evoked [3H]dopamine release from superfused rat striatal slices and for inhibition of [3H]nicotine and [3H]methyllycaconitine binding to whole rat brain membranes. The 3-picolinium compound 1,3,5-tri-{5-[1-(3-picolinium)]-pent-1-ynyl}benzene tribromide (tPy3PiB), 3b, exhibited high potency and selectivity for nAChR subtypes mediating nicotine-evoked [3H]dopamine release with an IC50 of 0.2 nM and Imax of 67%.  相似文献

The formation of three [Tl(en)n]3+ complexes (n=1–3) in a pyridine solvent has been established by means of 205Tl and 1H NMR. Their stepwise stability constants based on concentrations, Kn=[Tl(en)n 3+]/{[Tl(en)n−1 3+]·[en]}, at 298 K in 0.5 M NaClO4 ionic medium in pyridine, were calculated from 205Tl NMR integrals: log K1=7.6±0.7; log K2=5.2±0.5 and log K3=2.64±0.05. Linear correlation between both the 205Tl NMR shifts and spin–spin coupling 205Tl–1H versus the stability constants has been found and discussed. A single crystal with the composition [Tl(en)3](ClO4)3 was synthesized and its structure determined by X-ray diffraction. The Tl3+ ion is coordinated by three ethylenediamine ligands via six N-donor atoms in a distorted octahedral fashion.  相似文献

Specific binding of [125I]-(−)-cyanopindolol to human tracheal smooth muscle membranes was saturable, stereo-selective and of high affinity (Kd=5.3±0.9 pmol/l and RT=78±7fmol/g tissue). The β1-selective antagonists atenolol and LK 203-030 inhibited specific [125I]-(−)-cyanopindolol binding according to a one binding site model with low affinity in nearly all subjects, pointing to a homogeneous β2-adrenoceptor population. In one subject using LK 203-030 a small β-adrenoceptor subpopulation could be demonstrated. The beta-mimetics isoprenaline, fenoterol, salbutamol and terbutaline recognized high and low affinity agonist binding sites. Isoprenaline's pKH- and pKL- values for the high and low affinity sites were 8.0±0.2 and 5.9±0.3 respectively. In functional experiments isoprenaline relaxed tracheal smooth muscle strips having intrinsic tone with a pD2-value of 6.63±0.19.  相似文献

Polyclonal antibodies were raised by the repeated injection of rabbits with synthetic peptides corresponding to selective portions (peptide 1: aminoacid residues 12–23, and peptide 2: aminoacid residues 243–268) of the aminoacid sequence of the rat 5-HT1A receptor. Both antisera allowed the immunoprecipitation of 5-HT1A receptors but not of other 5-HT receptor types and adrenergic receptors solubilized from rat hippocampal membranes. Immunoblots demonstrated that a single protein of 63 kDa, corresponding to the molecular weight of the rat 5-HT1A receptor binding subunit, was recognized by each antiserum. Immunoautoradiographic labelling of rat brain sections with the anti-peptide 2-antiserum exhibited the same regional distribution as 5-HT1A sites labelled by selective radioligands such as [3H]8-OH-DPAT and [125I]BH-8-MeO-N-PAT. However regional differences apparently existed between the respective intensity of labelling by the agonist radioligands and the antiserum, which might be explained by variations in the degree of coupling of 5-HT1A receptor binding subunits with G proteins from one brain area to another.  相似文献

Inhibitory effects of zotepine (Zot) on D-1, D-2, D-3 and D-4 subtypes of dopamine (DA) receptors were investigated in crude synaptic membranes of rat striatum and bovine caudate nucleus and compared to those of chlorpromazine (CPZ) and haloperidol (HAL). From the IC50-values of Zot, CPZ and HAL, the K-values of each drug are estimated as follows: 34.4, 152 and 244 nM (D-1, 3H-labeled cis-flupenthixol binding (1.0 nM) to rat membranes); 37.4, 7.1 and 2.4 nM (D-2, [3H]spiperone (Spi) binding (0.5 nM) to rat membranes in the presence of 0.1 μM ketanserin); 73.1, 15.2 and 22.4 nM (D-3, 3H-labeled N-propylapomorphine (NPA) binding (0.29 nM) to bovine membranes in the presence of 0.1 μM Spi); 9.5, 65.3 and 3.1 nM (D-4, [3H]NPA binding (0.29 nM) bovine membranes in the presence of 25 nM DA), respectively. Zot binds with higher affinity to D-4 but lower affinity to D-3 than to other subtypes. It is also presumed that Zot binds to D-1 with high affinity and D-2 and D-3 with low affinity compared to CPZ and HAL.  相似文献
