Myoglobin is a globin with heme as prosthetic group whose main known biological role is to bind to O2 reversibly. On account of their large diversity, globins from mollusks have contributed to the study of this protein class. The cDNA of the myoglobins from Biomphalaria straminea and Biomphalaria tenagophila, which have a glutamine as distal residue (E7), were constructed and analyzed by bioinformatic tools. Native (not recombinant) myoglobins of these two Biomphalaria species were purified and their experimental molecular mass (about 16 kDa) and pI (about (8.0) were provided. Data analysis showed that these proteins are monomers with the signature for the classic myoglobin fold and they are blocked in amino terminus probably by an acetyl group. Values of the autoxidation rates showed that these myoglobins oxidized slowly. About the primary sequences of the myoglobins, they turned out to be satisfactory to group mollusks in phylogenetic class.  相似文献

Of the several species of Biomphalaria snails worldwide that serve as the intermediate host for Schistosoma mansoni, Biomphalaria alexandrina is a species that is indigenous to Egypt. Recently, there has been much debate concerning the presence of Biomphalaria glabrata and the hybrid of the species with Biomphalaria alexandrina. Due to this debate, the absence of a clear explanation for the presence of B. glabrata in Egyptian water channels and the probability that they may be reintroduced, we conducted this field study to identify Biomphalaria species present in Alexandria water channels. Laboratory-adapted susceptible snails to Schistosoma mansoni of the following species were used as a reference; Biomphalaria alexandrina, Biomphalaria glabrata and their hybrid. These snails were used to perpetuate the Schistosoma life cycle at the Theodor Bilharz Research Institute (TBRI), Cairo, Egypt. Morphological and molecular studies were conducted on these reference snails as well as on the first generation of Biomphalaria snails from two areas in the Alexandria governorate. The morphological study included both external shell morphology and internal anatomy of the renal ridge. The molecular study used a species-specific PCR technique.The results demonstrated that there was an absence of Biomphalaria glabrata and the hybrid from Alexandria water channels. Moreover, the susceptibility patterns of these reference snails were studied by measuring the different parasitological parameters. It was found that Biomphalaria glabrata and the hybrid were significantly more susceptible than Biomphalaria alexandrina to the Egyptian strain of Schistosoma mansoni. The results demonstrated that if Biomphalaria glabrata was reintroduced and adapted to the local environment in Egypt, it would have important epidemiologic impacts that would have a serious effect on the health of Egyptian people.  相似文献

Schistosomiasis is a tropical disease caused by the digenean parasite Schistosoma mansoni. In South America it is transmitted to humans via freshwater snails of the genus Biomphalaria. In a global warming scenario, disease range expansions to subtropical countries are possible. For the first time, the distributions and genetic identification of Biomphalaria specimens in border regions of Uruguay are reported. The inclusion of Uruguayan samples allows a better understanding of the relationships between and within taxa of Biomphalaria. Samples were collected between 2015 and 2016 using hand nets. Initially, they were classified morphologically. They were then classified genetically by analysing a fragment of the cytochrome c oxidase subunit I gene. Phylogenetic analysis revealed the presence of B. peregrina and B. occidentalis. Species belonging to the B. tenagophila complex were also recognised. Individuals initially identified from their external morphology as B. tenagophila tenagophila showed inconsistencies with the analysis of COI sequences that assigned them to B. occidentalis. Since the presence of schistosomiasis in Uruguay is likely to occur in the next few years, an exhaustive population survey of Biomphalaria taxa should be urgently developed to identify the presence of S. mansoni and the places most susceptible to be colonised by these snails.  相似文献

This is done by applying Von Foerster equations with boundary conditions derived from experiment. Myoglobin was isolated from the radular muscle of the chitonLiolophura japonica, a primitive archigastropodic mollusc.Liolophura contains three monomeric myoglobins (I, II, and III), and the complete amino acid sequence of myoglobin I has been determined. It is composed of 145 amino acid residues, and the molecular mass was calculated to be 16,070 D. The E7 distal histidine, which is replaced by valine or glutamine in several molluscan globins, is conserved inLiolophura myoglobin. The autoxidation rate at physiological conditions indicated thatLiolophura oxymyoglobin is fairly stable when compared with other molluscan myoglobins. The amino acid sequence ofLiolophura myoglobin shows low homology (11–21%) with molluscan dimeric myoglobins and hemoglobins, but shows higher homology (26–29%) with monomeric myoglobins from the gastropodic molluscsAplysia, Dolabella, andBursatella. A phylogenetic tree was constructed from 19 molluscan globin sequences. The tree separated them into two distinct clusters, a cluster for muscle myoglobins and a cluster for erythrocyte or gill hemoglobins. The myoglobin cluster is divided further into two subclusters, corresponding to monomeric and dimeric myoglobins, respectively.Liolophura myoglobin was placed on the branch of monomeric myoglobin lineage, showing that it diverged earlier from other monomeric myoglobins. The hemoglobin cluster is also divided into two subclusters. One cluster contains homodimeric, heterodimeric, tetrameric, and didomain chains of erythrocyte hemoglobins of the blood clamsAnadara, Scapharca, andBarbatia. Of special interest is the other subcluster. It consists of three hemoglobin chains derived from the bacterial symbiont-harboring clamsCalyptogena andLucina, in which hemoglobins are supposed to play an important role in maintaining the symbiosis with sulfide bacteria.  相似文献

The complete amino acid sequence of myoglobin from the triturative stomach of gastropodic molluscBursatella leachii has been determined. It is composed of 146 amino acid residues, is acetylated at the N-terminus, and contains a single histidine residue at position 95 which corresponds to the heme-binding proximal histidine. The E7 distal histidine, which is conserved widely in myoglobins and hemoglobins, is replaced by valine inBursatella myoglobin. The amino acid sequence ofBursatella myoglobin shows strong homology (73–84%) with those ofAplysia andDolabella myoglobins.  相似文献

The kinetic data obtained from the action of a cathepsin D-like enzyme from Biomphalaria glabrata hepatopancreas (digestive gland) on MOCAc-Gly-Lys-Pro-Ile-Leu-Phe-Phe-Arg-Leu-Lys(DNp)-D–Arg-NH2, was studied as a data prototype, generated by means of a fluorogenic substrate. An initial fluorescence, due to incomplete energy transfer, of about 8% of the values attained after complete substrate hydrolysis; a non-linear standard curve even at μM concentrations and an exponential decay of the steady state fluorescence of reaction product of the order of 10 4 × s 1 were the main analytical problems encountered. The standard curves for fluorescence of the substrate reaction product after 48 h of hydrolysis, and the reference compound MOCAc-Pro-Leu-Gly-NH2, were fitted by polynomial approximation and the point derivates used as calibration factors. Time dependence of the calibration factor for the reaction product was − 2.96 × 10 4 a.u μM 1 × s 1 that is, in the same order of observed enzymic reaction rates. A mathematical treatment was devised for obtaining rates corrected for errors derived from the three analytical problems indicated. The method is of general application in continuous fluorometric assays, irrespective of the particular enzyme used, but of special value for substrates that present significant initial fluorescence. The reaction rates were 11% higher; as calculated by means of the calibration factor [substrate] ÷ (final  initial fluorescence intensities), which is the prevalent procedure in the literature; leading to underestimation of Km and overestimation of Vmax.  相似文献

The African species of Biomphalaria appeared as a result of the relatively recent west-to-east trans-Atlantic dispersal of the Biomphalaria glabrata-like taxon. In Egypt, Biomphalaria alexandrina is the intermediate host for Schistosoma mansoni. Biomphalaria alexandrina originated in the area between Alexandria and Rosetta and has historically been confined to the Nile Delta. Schistosoma mansoni reached Egypt via infected slaves and baboons from the Land of Punt through migrations that occurred as early as the Vth Dynasty. The suggestion of the presence of Schistosoma mansoni infection in Lower Egypt during Pharaonic times is discussed despite the fact that that there is no evidence of such infection in Egyptian mummies. It is only recently that Biomphalaria alexandrina colonized the Egyptian Nile from the Delta to Lake Nasser. This change was likely due to the construction of huge water projects, the development of new water resources essential for land reclamation projects and the movement of refugees from the Suez Canal zone to the Delta and vice versa. The situation with respect to Biomphalaria in Egypt has become complicated in recent years by the detection of Biomphalaria glabrata and a hybrid between both species; however, follow-up studies have demonstrated the disappearance of such species within Egypt. The National Schistosoma Control Program has made great strides with respect to the eradication of schistosoma; however, there has unfortunately been a reemergence of Schistosoma mansoni resistant to praziquantel. There are numerous factors that may influence the prevalence of snails in Egypt, including the construction of water projects, the increase in reclaimed areas, global climate change and pollution. Thus, continued field studies in addition to the cooperation of several scientists are needed to obtain an accurate representation of the status of this species. In addition, the determination of the genome sequence for Biomphalaria alexandrina and the use of modern technology will allow for the study of the host–parasite relationship at a molecular level.  相似文献

Polyhydroxyalkanoates (PHAs) are a replacement of conventional single-use plastics. Bioprocess conditions of the extreme halophilic archaeon Halogeometricum borinquense strain RM-G1 were selected resulting in the synthesis of 66.80 ± 1.69 % PHA (of cell dry mass) in 72 h using glycerol and tryptone as carbon and nitrogen sources respectively, yielding volumetric productivity of 0.206 ± 0.006 gL−1 h−1 in a repeated batch process in a small-scale bioreactor where 20 % of the production medium was used as the inoculum for the subsequent batch. The purified PHA was characterized as poly(3-hydroxybutyrate-co-3-hydroxyvalerate) with 10.21 mol% 3-hydroxyvalerate content possessing glass transition temperature -12.6 °C, degradation temperature 285 °C, number average molecular weight 156,899 Da, weight average molecular weight 288,723 Da, polydispersity index 1.8 and melting temperatures 139.1 °C and 152.5 °C. Maximum (21.7 ± 0.6 L m-2 h−1) and average (17.2 ± 0.6 L m-2 h−1) flux values were their respective highest and crystallization time was its least (3.0 ± 0.16 h) when ΔT was 90 °C and polytetrafluoroethylene membrane was applied for desalination of the bioreactor effluent by Direct Contact Membrane Distillation. While using polyvinylidene fluoride membrane, maximum 25.5 ± 0.5 L m-2 h−1 and average 18.6 ± 0.2 L m-2 h−1 fluxes were obtained and crystallization time decreased (3.25 ± 0.16 h) even when ΔT was lowered by 20 °C.  相似文献

The karyotypes of Biomphalaria tenagophila collected from Rio de Janeiro, Brazil were studied using the air-drying method. Somatic cells of this species had 2n=36. The 18 chromosome pairs were identified and classified into 3 groups. The diploid cell has 7 pairs of metacentric, 8 pairs of submetacentric, and 3 pairs of subtelocentric chromosomes. Observed chromosomes ranged from 2.4 to 6.4 µm, and the total length was 122.3 µm. This is the first report on the chromosome of B. tenagophila.  相似文献

Despite ongoing preventive chemotherapy campaigns, intestinal schistosomiasis is hyper-endemic in shoreline communities living along Lake Albert, Uganda. To provide a deeper insight into the local epidemiology of Schistosoma mansoni, a variety of field-based studies were undertaken focusing upon schistosome–snail interactions and confirmation of transmission foci. Cercarial shedding patterns of field-caught Biomphalaria spp., as identified by morphology, were hourly observed over a ten day period and showed that Biomphalaria stanleyi produced significantly more cercariae than Biomphalaria sudanica. Peak production times in both species were between 12.00 and 14.00 h indicating greatest infection risk from lake water exposure is during the early afternoon. Laboratory-bred snails were exposed to locally hatched miracidia and susceptibility of Biomphalaria spp. was confirmed experimentally. Biomphalaria stanleyi was a more permissive host. After ascertaining appropriate conditions for infection of laboratory mice, 28 groups of between 5 and 6 naïve mice were placed in floatation cages at four suspected shoreline transmission sites for a 30 minute period of exposure. Eight weeks later, mice (n = 142) were culled and S. mansoni adult worms were retrieved from 10 animals. Taken as a whole, these observations highlight the local importance of B. stanleyi in transmission of intestinal schistosomiasis and clearly demonstrate the risk of infection on the Lake Albert shoreline. To mitigate this risk local environmental modification(s), i.e. improvement in sanitation and hygiene and control of snail populations, is needed to bolster the impact of chemotherapy-based interventions.  相似文献

The single cell gel eletrophoresis or the comet assay was established in the freshwater snail Biomphalaria glabrata. For detecting DNA damage in circulating hemocytes, adult snails were irradiated with single doses of 2.5, 5, 10 and 20 Gy of 60Co gamma radiation. Genotoxic effect of ionizing radiation was detected at all doses as a dose-related increase in DNA migration. Comet assay in B. glabrata demonstrated to be a simple, fast and reliable tool in the evaluation of genotoxic effects of environmental mutagens.  相似文献

The mechanisms underlying cigarette smoke-induced mitochondrial dysfunction in skeletal muscle are still poorly understood. Accordingly, this study aimed to examine the effects of cigarette smoke on mitochondrial energy transfer in permeabilized muscle fibers from skeletal muscles with differing metabolic characteristics. The electron transport chain (ETC) capacity, ADP transport, and respiratory control by ADP were assessed in fast- and slow-twitch muscle fibers from C57BL/6 mice (n = 11) acutely exposed to cigarette smoke concentrate (CSC) using high-resolution respirometry. CSC decreased complex I-driven respiration in the white gastrocnemius (CONTROL:45.4 ± 11.2 pmolO2.s−1.mg−1 and CSC:27.5 ± 12.0 pmolO2.s−1.mg−1; p = 0.01) and soleus (CONTROL:63.0 ± 23.8 pmolO2.s−1.mg−1 and CSC:44.6 ± 11.1 pmolO2.s−1.mg−1; p = 0.04). In contrast, the effect of CSC on Complex II-linked respiration increased its relative contribution to muscle respiratory capacity in the white gastrocnemius muscle. The maximal respiratory activity of the ETC was significantly inhibited by CSC in both muscles. Furthermore, the respiration rate dependent on the ADP/ATP transport across the mitochondrial membrane was significantly impaired by CSC in the white gastrocnemius (CONTROL:-70 ± 18 %; CSC:-28 ± 10 %; p < 0.001), but not the soleus (CONTROL:47 ± 16 %; CSC:31 ± 7 %; p = 0.08). CSC also significantly impaired mitochondrial thermodynamic coupling in both muscles. Our findings underscore that acute CSC exposure directly inhibits oxidative phosphorylation in permeabilized muscle fibers. This effect was mediated by significant perturbations of the electron transfer in the respiratory complexes, especially at complex I, in both fast and slow twitch muscles. In contrast, CSC-induced inhibition of the exchange of ADP/ATP across the mitochondrial membrane was fiber-type specific, with a large effect on fast-twitch muscles.  相似文献

The main photosynthesis and respiration parameters (dark respiration rate, light saturated production rate, saturation irradiance, photosynthetic efficiency) were measured on a total of 23 macrophytes of the Thau lagoon (2 Phanerogams, 5 Chlorophyceae, 10 Rhodophyceae and 6 Phaeophyceae). Those measurements were performed in vitro under controlled conditions, close to the natural ones, and at several seasons. Concomitantly, measurements of pigment concentrations, carbon, phosphorous and nitrogen contents in tissues were performed. Seasonal intra-specific variability of photosynthetic parameters was found very high, enlightening an important acclimatation capacity. The highest photosynthetic capacities were found for Chlorophyceae (e.g. Monostroma obscurum thalli at 17 °C, 982 μmol O2 g−1 dw h−1 and 9.1 μmol O2 g−1 dw h−1/μmol photons m−2 s−1, respectively for light saturated net production rate and photosynthetic efficiency) and Phanerogams (e.g. Nanozostera noltii leaves at 25 °C, 583 μmol O2 g−1 dw h−1 and 2.6 μmol O2 g−1 dw h−1/μmol photons m−2 s−1 respectively for light saturated net production rate and photosynthetic efficiency). As expected, species with a high surface/volume ratio were found to be more productive than coarsely branched thalli and thick blades shaped species. Contrary to Rd (ranging 6.7–794 μmol O2 g−1 dw h−1, respectively for Rytiphlaea tinctoria at 7 °C and for Dasya sessilis at 25 °C) for which a positive relationship with water temperature was found whatever the species studied, the evolution of P/I curves with temperature exhibited different responses amongst the species. The results allowed to show summer nitrogen limitation for some species (Gracilaria bursa-pastoris and Ulva spp.) and to propose temperature preferences based on the photosynthetic parameters for some others (N. noltii, Zostera marina, Chaetomorpha linum).  相似文献

Protein tyrosine phosphatase 1B (PTP1B) is a validated target for developing antiobesity, antidiabetic and anticancer drugs. Over the past years, several inhibitors of PTP1B have been discovered; however, none has been approved by the drug regulatory agencies. Interestingly, the research programs focused on discovering PTP1B inhibitors typically use truncated structures of the protein (PTP1B1-300, 1–300 amino acids), leading to the loss of valuable information about the inhibition and selectivity of ligands and repeatedly misleading the optimization of putative drug leads. Up to date, only six inhibitors of the full-length protein (hPTP1B1-400), with affinity constants ranging from 1.3 × 104 to 3.3 × 106 M−1, have been reported. Towards the discovery of new ligands of the full-length human PTP1B (hPTP1B1-400) from natural sources, herein we describe the isolation of a γ-lactone (1, butyrolactone I) from the fungus Aspergillus terreus, as well as the semisynthesis, inhibitory properties (in vitro and in silico), and the structure-activity relationship of a set of butyrolactone derivatives (1 and 2, and 6–12) as hPTP1B1-400 inhibitors, as well as the affinity constant (ka = 2.2 × 105 M−1) of the 1-hPTP1B1400 complex, which was determined by fluorescence quenching experiments, after the inner filter effect correction.  相似文献

Carbon limited continuous cultures of Lactobacillus rhamnosus ATCC 7469 were grown at dilution rates between 0.1 h−1 and 0.6 h−1. At 0.45 h−1, oxygen uptake decreases producing a deficiency in the production of cell energy, lowering the concentration of biomass and finally accumulating glucose in the broth. Under the lack of energy pressure, L. rhamnosus ATCC 7469 triggers the production of lactic acid from pyruvate freeing NAD+ and stimulates glycolysis to continue, producing extra ATP from substrate-level phosphorylation. The 12-fold growing concentration of lactic acid and the 2-fold increase of succinic acid are in parallel with the steep 4-fold decrease of acetic acid production and small concentration changes of formic and propionic acids.The way the cells balance the available energy between the growing dilution rate and detoxification produces a stress within the culture, detected and described by flow cytometry. As the dilution rate increased, the proportion of L. rhamnosus ATCC 7469 cells with depolarized membrane steadily increased (1% at D = 0.20 h−1, 8% at D = 0.30 h−1, 14% at D = 0.45 h−1 and 26% for D = 0.62 h−1, respectively). Only a low level of 3.7% of the population did not recover from the demanding growth rates in the acidic environment.  相似文献

Isolated closed basins provide a natural laboratory to study the differentiation among wild populations. Here we examined the phylogenetic relationships of the Southern Altiplano populations of Biomphalaria, a genus with medical importance, using nuclear (ITS1, ITS2) and mitochondrial (16S) ribosomal gene markers and a species of Helisoma as outgroup. Phylogenetic trees based on separate and combined analyses show that these populations form a particular lineage within Biomphalaria along with Biomphalaria peregrina (d'Orbigny, 1835) and Biomphalaria oligoza Paraense, 1974. The origin of this clade was estimated to have occurred in the middle Pleistocene. Molecular analyses showed that the nominal species Biomphalaria crequii ( Courty, 1907 ) from the Salar de Ascotán and Biomphalaria costata (Biese, 1951) from the Salar de Carcote, previously synonymized with Biomphalaria andecola (d'Orbigny, 1835) and B. peregrina, respectively, are distinct taxa. Molecular data did not resolve the relationship of Biomphalaria aymara Valdovinos & Stuardo, 1991 from the Isluga swamps to other Biomphalaria species, but confirm that the populations from the Lauca and Huasco basins may represent a distinct undescribed species of Biomphalaria from the Southern Altiplano. Low-molecular-weight hyaluronan (LMW-HA) has attracted much attention because of its many potential applications. Here, we efficiently produced specific LMW-HAs from sucrose in Bacillus subtilis. By coexpressing the identified committed genes (tuaD, gtaB, glmU, glmM, and glmS) and downregulating the glycolytic pathway, HA production was significantly increased from 1.01 g L−1 to 3.16 g L−1, with a molecular weight range of 1.40×106–1.83×106 Da. When leech hyaluronidase was actively expressed after N-terminal engineering (1.62×106 U mL−1), the production of HA was substantially increased from 5.96 g L−1 to 19.38 g L−1. The level of hyaluronidase was rationally regulated with a ribosome-binding site engineering strategy, allowing the production of LMW-HAs with a molecular weight range of 2.20×103–1.42×106 Da. Our results confirm that this strategy for the controllable expression of hyaluronidase, together with the optimization of the HA synthetic pathway, effectively produces specific LMW-HAs, and could also be used to produce other LMW polysaccharides.  相似文献
