该研究选用7条ISSR标记引物对百合属21种野生百合的亲缘关系进行分析,使用POPGEN1.32和MEGA5.1数据处理软件分析数据。结果表明:7条引物扩增出149条条带,其中136条为多态性条带,多态性条带比率为91.06%,平均有效等位点数为1.7624,平均Nei’ s基因多样度为0.4214,平均Shanon信息指数0.6085。遗传距离变化范围为0.3075~0.8873,紫脊百合和尖被百合的遗传距离最大,均值为0.8873,表明21份材料中二者亲缘关系最远;紫脊百合和宜昌百合遗传距离最小,均值为0.3075,表明二者亲缘关系最近。聚类结果与形态学分类大致吻合,21个供试材料可被分成5个类群,大理百合、宝兴百合、卷丹百合、紫斑百合、乳头百合、川百合、兰州百合、山丹百合、绿花百合和毛百合为第Ⅰ类群;紫脊百合、宜昌百合、岷江百合、通江百合和淡黄花百合为第Ⅱ类群;野百合和百合为第Ⅲ类群;青岛百合为第Ⅳ类群;第Ⅴ类群包括玫红百合、有斑百合和尖被百合。毛百合与卷瓣组百合亲缘关系较近,钟花组百合与卷瓣组百合存在基因交流,说明不能仅仅依靠形态学对野生百合进行分类。聚类分析结果中野百合与百合单独聚为一类,这说明是否具叶柄对野生百合的分类是一个重要的形态学特性。 ISSR分子标记适合用于百合属植物的亲缘关系分析。 相似文献

中国和美国大豆疫霉群体遗传结构的ISSR分析 总被引:12,自引:0,他引:12
为探究中国和美国大豆疫霉的遗传关系, 采用简单序列重复区间扩增多态性(ISSR)技术, 对来自中国黑龙江省、福建省和美国的3个大豆疫霉地理群体的遗传多样性进行了分析。通过13个ISSR引物对供试的111株大豆疫霉菌株进行扩增, 共得到102个ISSR条带, 其中多态性条带为88个, 占86%。遗传变异分析表明, 美国群体具有更高的遗传变异度; Nei’s 遗传相似性和主成分分析均显示, 中国福建群体与美国群体间的遗传相似性最高, 而福建群体与黑龙江群体间遗传相似性最低; 聚类分析显示, 供试菌株在88%的相似性水平上可区分为7个聚类组, 且美国群体分布于更多的聚类组中; Shannon-Wiener多样性指数也表明美国群体的遗传多样性最为丰富。综合分析表明, 本研究的结果不支持关于美国的大豆疫霉可能来源于中国的推测。 相似文献

Sequence-related amplified polymorphism (SRAP) markers were used to assess genetic relationships among 76 grape genotypes including Chinese indigenous and newly bred varieties, representatives of foreign grape varieties, and wild Vitis species. Nineteen informative primers were selected from 100 SRAP primer pairs due to their ability to produce clearly and repeatedly polymorphic and unambiguous bands among the varieties. A total of 228 bands were produced; 78.63% of them were polymorphic; the average polymorphism information content (PIC) is 0.76. Genetic relationships were obtained using Nei and Li similarity coefficients. Cluster analysis of SRAP markers through the unweighted pair-group method of arithmetic averages (UPGMA) analysis and principal coordinate analysis (PCoA) were largely consistent. The definition of clusters in the dendrogram and PCoA plot is the same and some degree of grouping by types of grape, ecogeographical origin, and taxonomic status of the varieties was revealed. Three main groups were found after cluster analysis, i.e., table grape of Vitis vinifera; table grape of Euro-America hybrid and wine grape of V. vinifera; wild Vitis species. Groupings indicated a divergence between the table and wine-type varieties of V. vinifera. The results showed that the wild Vitis species that originated from America and China could be clearly differentiated and Vitis hancockii is the most distant from the others of Asian Vitis species. The results also indicated that SRAP markers are informative and could distinguish bud sports of grape. The present analysis revealed that Chinese cultivated and wild grape germplasm are highly variable and have abundant genetic diversity. 相似文献

Genetic interactions between marine finfish species in European aquaculture and wild conspecifics 总被引:2,自引:0,他引:2
The principal species of marine aquaculture in Europe are Atlantic salmon (Salmo salar), sea bass (Dicentrarchus labrax) and sea bream (Sparus auratus). For Atlantic salmon and sea bass, a substantial part of total genetic variation is partitioned at the geographical population level. In the case of sea bream, gene flow across the Azores/Mediterranean scale appears to be extensive and population structuring is not detected. For Atlantic salmon and sea bass, natural population structure is at risk from genetic interaction with escaped aquaculture conspecifics. The locally adaptive features of populations are at risk from interbreeding with non‐local aquaculture fish. Wild populations, generally, are at risk from interactions with aquaculture fish that have been subject to artificial selection or domestication. Atlantic salmon is the main European aquaculture species and its population genetics and ecology have been well‐studied. A general case regarding genetic interactions can be based on the information available for salmon and extended to cover other species, in the appropriate context. A generalized flow chart for interactions is presented. Salmon escape from aquaculture at all life stages, and some survive to breed among wild salmon. Reproductive fitness in the escaped fish is lower than in native, wild fish because of behavioural deficiencies at spawning. However, as the number of salmon in aquaculture greatly exceeds the number of wild fish, even small fractional rates of escape may result in the local presence of large numbers, and high frequencies, of escaped fish. At present, policy and legislation in relation to minimizing genetic interactions between wild and aquaculture fish is best developed for Atlantic salmon, through the recommendations of the Oslo Agreement developed by the North Atlantic Salmon Conservation Organization and subsequent agreements on their implementation. In future, the potential use of genetically modified fish in aquaculture will make additional policy development necessary. Improved containment is recommended as the key to minimizing the numbers and therefore the effects of escaped fish. Emergency recovery procedures are recommended as a back‐up measure in the case of containment failure. Reproductive sterility is recommended as a future key to eliminating the genetic potential of escaped fish. The maintenance of robust populations of wild fish is recommended as a key to minimizing the effects of escaped fish on wild populations. 相似文献

Marmots are large ground squirrels, and 14 species have been reported in the world, including four species of marmots (Himalayan marmot, Tarbagan marmot, gray marmot and long-tailed marmot) living in China. Although these biological resources are abundant in China, information regarding their genetic features is lacking, hampering further study regarding them. The aims of this research were to evaluate genetic variations of four species of Chinese wild marmots, and analyzed kinship of these marmot populations. In the current study, we collected samples of four species of Chinese wild marmot and analyzed the effective allele number, gene diversity, the Shannon index, and polymorphism information to evaluate genetic variations using 13 microsatellite loci. Based on Nei’s genetic distance using the unweighted pair group method, we constructed a dendrogram to analyze the population kinship. We determined that all four Chinese marmot species had high genetic polymorphisms and departure from Hardy-Weinberg equilibrium. The Chinese marmots to be divided into two large groups: Himalayan marmot was independent group. Tarbagan marmot, gray marmot and long-tailed marmot were others; Tarbagan marmot and gray marmot showed a close kinship with each other, but long-tailed marmot did not have a close relationship with the other species. The high polymorphisms and the kinship of Chinese marmot populations were correlated with geographical terrain of their habitat. Himalayan marmot was characterized as living in unique alpine meadows in Qinghai-Tibet plateau and was affected by terrain; however, Tarbagan marmot, gray marmot and long-tailed marmot were characterized as living in grassland or alpine grassland and were not affected by terrain. Genetic features of Chinese wild marmots were investigated in this study. This may give using information regarding protection of Chinese wild marmot resource and further application of biomedical research. 相似文献

The objective of this study was to obtain an overview of the genetic relationships within Perinereis aibuhitensis using Inter-Simple Sequence Repeat (ISSR) and Sequence-Related Amplified Polymorphism (SRAP) markers that were derived from related populations residing in the Chinese coasts. The percentage of polymorphic bands, Nei's gene diversity and Shannon's information index revealed a high level of genetic diversity at the species level. The analysis of molecular variance revealed that 81.22% (ISSR) and 76.29% (SRAP) of variability were partitioned among individuals within populations, which indicated the coherent trend by Nei's genetic differentiation (Gst) (0.2568/0.2876). The gene flow number (Nm) was 1.4470/1.2385, which indicated that there was limited gene exchange between populations. The phylogenetic tree of the ten P. aibuhitensis populations was separated into four major clusters using the neighbor-joining (NJ) method. These results provide a simple and useful basis for P. aibuhitensis germplasm research and aquaculture breeding. 相似文献

Cunila species are among the commonly used South Brazilian plants in popular medicine. This genus presents two centers of distribution in North and South America; the latter being classified into three sections: Incanae, Incisae and Spicatae. Based on the prospective utility as aromatic and medicinal plants, the aim of this work was to examine the genetic diversity among the South American species to contribute to the knowledge of their botanical sections. In this context, 11 Cunila species were analyzed by ISSR using seven primers that generated a total of 107 bands. The relationship was evaluated by constructing dendrograms using the UPGMA algorithm and analysis of principal components. The cluster analysis places the species C. origanoides with the South American species, but forming an independent cluster. Remarkably, among South American species two clusters emerge, one consisting of shrubs and the second by subshrubs species, which refines the botanical taxonomy for several species of the genus. 相似文献

In spite of the importance of and the considerable variability observed in Passiflora (Passifloraceae), little is known about the genetic diversity of most of the species of this genus. We evaluated the genetic diversity by RAPD markers in 18 genotypes of Passiflora trintae. The 15 primers generated 112 markers, 84% of which were polymorphic. The genetic distance estimated by the complement of the Dice index (average dissimilarity = 0.30) and genotype grouping based on the UPGMA algorithm showed low variability among genotypes. More attention should be given to the study and conservation of the biodiversity of this economically important genus. 相似文献

为了调查中国绒山羊遗传资源现状, 作者应用联合国粮农组织和国际家畜研究所推荐的19对微卫星引物并结合荧光PCR技术, 对9个中国地方产绒山羊群体和1个西非山羊品种进行了遗传多样性检测。14个微卫星座位在10个山羊群体中显示为高度多态, 可作为山羊遗传多样性分析的有效标记。多态信息含量和遗传杂合度等数据表明: 目前中国地方产绒山羊群体的遗传多样性较为丰富, 并且大部分保种场较好地保存了这些地方资源。采用非加权配对算术平均法构建的聚类图和采用主成分分析法得到的散点图均显示, 中国山羊与西非山羊为不同的2类; 中国产绒山羊中河谷山羊、河西绒山羊与其他山羊的遗传距离较远; 其他山羊又大致分为2类: 一类由辽宁绒山羊、新疆山羊、柴达木山羊、陕北山羊组成, 另一类由内蒙古绒山羊组成。此研究结果为开展我国地方绒山羊种质特性研究及资源保护和利用提供了科学依据。 相似文献

基于ISSR标记的扁玉螺(Neverita didyma)自然居群遗传结构 总被引:3,自引:0,他引:3
利用ISSR分子标记技术,对采自大连(DL)、烟台(YT)及青岛(QD)近海的扁玉螺3个自然居群的遗传结构和遗传多样性进行了分析。用13个引物对90只个体进行了PCR扩增,共检测到161个位点,3个居群的多态位点比例为74.53%~85.09%,各居群遗传多样性水平的高低依次为YT〉QD〉DL。扁玉螺在物种水平上的Nei’s基因多样性指数和Shannon’s信息指数分别为0.3395和0.5113,在居群水平上分别为0.2811和0.4189,显示出扁玉螺有着较高的遗传多样性。AMOVA分子变异分析表明,扁玉螺的遗传变异有27.16%发生在居群间,72.84%发生在居群内,居群内的遗传变异大于居群间的遗传变异。扁玉螺3个居群间的遗传分化系数(Gst)为0.1720,基因流(Nm)为2.4063,Nei’s遗传距离平均值为0.1228,表明扁玉螺居群间虽然存在着一定程度的遗传分化,但仍属于种内正常分化的范畴。上述结果为保护和利用扁玉螺资源提供了科学依据。 相似文献

Genetic diversity among 83 lentil genotypes including 23 wild types, 19 indigenous varieties, 5 exotic lines and 36 advanced breeding lines was studied using molecular markers. A total of 112 amplicons were produced using 15 RAPD and 8 SSR markers. Dendrogram based on Jaccard similarity coefficient and UPGMA analysis revealed two major clusters and one minor cluster. Cluster I comprised 21 wild accessions of L. orientalis and 1 L. ervoides subspecies. Nineteen Indian varieties grouped together in subcluster IIA indicating their narrow genetic base. Subcluster IIB consisted of 41 genotypes including 5 exotic and 36 advanced breeding lines mainly derived from exotic genotypes. The narrow genetic base of released cultivars and germplasm lines emphasized the need for broadening of genetic base of breeding material using exotic collections and wild species to ascertain genetic improvement upon existing cultivars. 相似文献

The genus Corylus, a member of the birch family Betulaceae, includes several species that are widely distributed throughout temperate regions of the Northern Hemisphere. This study assesses the genetic diversity in 26 international cultivars and 32 accessions of Corylus avellana L. from Portugal: 13 wild genotypes and 19 landraces. The genetic relationships among the 58 hazelnuts (Corylus avellana L.) were analyzed using inter simple sequence repeat (ISSR) and amplified fragment length polymorphism (AFLP) markers. Eighteen ISSR primers and seven AFLP primer pairs generated a total of 570 unambiguous and repeatable bands, respectively, from which 541 (95.03 %) were polymorphic for both markers. Genetic similarity index values ranged from 0.239 for wild types and cultivars to 0.143 for landraces and wild types. The genetic relationships were presented as a Neighbor-Joining method dendrogram and a two-dimensional principal coordinate analysis (PCoA) plot. The Neighbor-Joining dendrogram showed three main clusters, and the PCoA analysis has shown to be congruent with the hierarchical analysis. Bayesian analysis clustered all individuals into three groups showing a good separation among wild genotypes, landraces and cultivars. The genetic diversity found on wild genotypes and Portuguese landraces may provide relevant information for the diversity conservation and it will be useful in breeding programs and to identify local selections for preservation. 相似文献

Chinese wild grapes are almost exclusively dioecious and black-fruited, with rare reports of white and hermaphrodite types in V. davidii. To reveal the molecular mechanisms of these phenotypic variations, specific primers were designed to detect the genotypes of mybA-related genes in Vitis species, including the Chinese wild Vitis species, V. riparia, V. rupestris, cultivars of Vitis vinifera and its hybrids. We report here that three mybA-related genes, VvmybA1a, VvmybA2 and VvmybA3, were only detected in cultivars of V. vinifera and its hybrids, but not in V. riparia, V. rupestris or Chinese wild Vitis species, indicating that these genes could be used to test the genetic relationship to V. vinifera. On the other hand, the genes were not detected in the dioecious varieties of V. davidii, but were in the hermaphrodites. In particular, the white-fruited varieties were homozygous for VvmybA1a and showed a low expression of mybA-related genes and UFGT during the entire maturation period. Simple sequence repeat analysis showed that the hermaphrodite varieties of V. davidii, including the white-fruited varieties, were more closely related to V. vinifera cv. Pinot Noir and V. labruscana cv. Kyoho. These results suggested that the white-fruited and hermaphrodite varieties of V. davidii could be the result of its crossing with V. vinifera. It provides a new approach to identify truly Chinese wild varieties and to search for possible hybridization events. 相似文献

The results of attempted hybridization between European species of large-grained species of Lupinus, L. albus, L. angustifolius and L. luteus and between these and the South American species L. mutabilis are described. Pollen-tube growth in interspecific combinations differed significantly from those after self-pollination only when L. luteus , was the female parent. All combinations Involving L. mutabilis pollen exhibited unilateral compatible pollen-tube growth. Substantial evidence of ovule stimulation and enhanced pod retention was recorded in crosses between the European species and L. mutabilis , but viable seeds were not obtained. The indications, from pollen-tube growth and pod retention in certain crosses, are that genetical isolation between some of the species could be overcome. 相似文献

为了明确广西野生茶树种质资源的遗传背景,该研究从广西的宁明县、金秀县、苍梧县收集到14份地方野生茶树种质资源,以17个国家级茶树良种作为参照,采用EST-SSR分子标记技术,探讨了广西这三个地方野生茶树与国家级茶树良种间的亲缘关系以及广西地方茶树自身的遗传多样性。结果表明:15对EST-SSR引物共检测到68个等位基因,平均每个引物可扩增出4.53个,其中多态性位点为60个,多态性比率达88.2%。平均观测杂合度、平均期望杂合度、平均Shannon信息指数分别为0.42、0.55和0.97。PIC值在0.23~0.74之间,平均为0.52,多态性较好。遗传相似系数在0.53~0.9之间,平均值为0.71,31份供试材料在遗传相似系数为0.71分为5组群,76%参照品种聚在A组群,而广西本地的野生茶树资源则主要分布在B、C、D、E组群。利用该研究中的4对核心引物即可将31份供试材料全部区分开,挑选其中10个多态性较好的等位位点进行编码,构建31份供试种质的DNA分子指纹图谱。这表明广西野生茶树资源与国家级茶树良种间遗传差异较大、亲缘关系较远、遗传基础宽、多样性非常丰富,可作为茶树育种的亲本或开展茶树功能基因研究的材料。 相似文献

利用ISSR分子标记对雄性猕猴桃16个材料进行遗传多样性分析。从100条引物中筛选出10条引物用于ISSR扩增,共扩增出172条带,其中多态性条带140条,多态性百分率为81.4%;经POPGENE 1.32软件分析结果显示,16个雄性猕猴桃材料的遗传距离在0.1503~0.5128之间,平均Nei's基因多样性指数(H)为0.2416,平均Shannon信息指数(I)为0.4048;聚类分析结果显示,在遗传相似系数为0.64处可将供试材料分成4类,第Ⅰ类为中华和美味猕猴桃,第Ⅱ类为阔叶、毛花猕猴桃,第Ⅲ类为魁绿猕猴桃,第Ⅳ类为四萼猕猴桃。结果表明ISSR可用于雄性猕猴桃遗传多样性研究,该研究结果可为猕猴桃种质资源的进一步开发利用提供重要信息。 相似文献

苎麻与近缘种的系统学关系一直存在争议。利用26对多态性好的随机引物,对中国苎麻属的17种9变种的共27份材料进行SRAP分子标记分析。扩增结果是26对引物共扩出368条带,其中363条为多态性带,占98.64%。聚类分析表明:(1)腋球苎麻组的腋球苎麻和苎麻组的苎麻、贴毛苎麻聚在一支并靠近系统树的根部,支持王文采苎麻组起源于原始的腋球苎麻组观点。(2)苎麻和贴毛苎麻聚在一支,支持郭安平苎麻直接起源于贴毛苎麻的观点。(3)苎麻组的青叶苎麻、微绿苎麻和大叶苎麻组的多倍体类群聚在一支,可能是由于在种间杂交导致。 相似文献

DNA polymorphism among 34 Chinese Auricularia auricula cultivars was analyzed using inter-simple sequence repeat (ISSR) and sequence-related amplified polymorphism (SRAP) markers.
Thirty ISSR primers amplified a total of 129 DNA fragments of which 125 (96.9%) were polymorphic, whereas 11 SRAP primer combinations
amplified 154 fragments of which 148 (96.1%) were polymorphic. Both methods were highly effective in discriminating among
the test strains. Phylogenetic trees constructed on the basis of ISSR, SRAP, and combined ISSR/SRAP analyses using the Unweighted
Pair-group Method with Arithmetic Averages (UPGMA) method distributed the 34 strains into four or five major groups. Clustering
analysis based on all the three data sets indicated a high level of genetic diversity among A. auricula, although the combined ISSR/SRAP data were more concordant with the main agronomic characters of strains and their geographical
centers of cultivation. Our findings will facilitate future A. auricula breeding programs and the development of bioactive products from this commercially important medicinal mushroom. 相似文献
