高酶活菌株的筛选及漆酶特性   总被引:5,自引:0,他引:5  
通过Bavendamn氏反应和液体发酵实验筛选出漆酶高产菌株 ,并对其产酶条件和酶活性进行了研究。结果表明 71株实验真菌中有 64株Bavendamn氏反应呈阳性 ,且阳性菌株都具有漆酶活性 ;不同菌株产酶培养基最适碳源、氮源不同 ,采绒革盖菌以淀粉为碳源、干酪素为氮源 ,毛栓菌以麦草粉为碳源、硫酸铵为氮源 ,有利于酶的分泌 ;不同来源漆酶性质不尽相同 ,采绒革盖菌漆酶最适酶解温度为 2 5℃ ,最适酶解pH值为4.6,毛栓菌则分别为 3 0℃和 pH 4.0 ;K+ ,Zn2 + 等对 2种漆酶均有激活作用 ,Ag+ 则能明显抑制漆酶活性。  相似文献

彩绒革盖菌(Corilusversicolor)是一种具有木质素降解能力的白腐菌,其降解木素的能力主要取决于木质素过氧化物酶、漆酶、多酚氧化酶和愈创木酚氧化酶的活力高低,所以研究其活力和分泌情况对木质素的生物降解、生物制浆和环境保护具有十分重要的意义。国外已有彩绒革盖菌漆酶的报道[1,2],但主要集中在漆酶对木质素作用后所造成的木质素结构上的变化,有关漆酶和多酚氧化酶的活性分析报道较少[3];国内有关的研究报道更少[4,5],至今未见彩绒革盖菌漆酶和多酚氧化酶的研究报道。本文主要测定了彩绒革盖菌…  相似文献

为了更好开发利用漆酶,用邻联甲苯胺法比较分析了彩绒革盖菌、毛栓菌和多孔菌在液体培养时的产酶曲线、酶作用的最适pH值、最适酶解温度及无机离子对酶活的影响。结果表明,不同漆酶产酶曲线不同,彩绒革盖菌和多孔菌,第9d达产酶高峰,峰值活力分别达395.6u/ml和412.2u/ml;毛栓菌,第11d达到产酶高峰,峰值本科活较不同真菌漆酶的性质研究高达554.6u/ml。漆酶性质有明显差别,最适酶解温度不同,彩绀革盖菌和多孔菌漆酶最适酶解温度为25℃;毛栓菌为30℃;最适酶解pH值有差异,彩绒革盖菌漆酶最适酶解,pH值为4.5,毛栓菌为4.0,多孔菌为4.2;不同离子对酶活的影响不同;K^、Zn^2 、对彩绒革盖菌所产漆酶有激活作用;K^ 、Zn^2 、Cu^2 对毛栓菌所产漆酶有激活作用;Mn^2 、Mg^2 对多孔菌所产漆酶有激活作用;Ag^ 、Fe^3 对三种菌所产漆本科均有明显抑制作用。  相似文献

研究了彩绒革盖菌在不同硬源和氮源培养基中生长时,对纤维素酶、革纤雄素酶、本质素酶(漆酶、多酚氧化酶、愈创木酚氧化酶)分泌的影响。表明不同碳源和氮源对酶类失发泌影响很大,富含淀粉的物质能明显促进本质素酶的分泌,而专一性底物(纤维素和半纤维素)对纤维素酶和半纤维素酶有诱导作用,麸皮也能话导半纤维素酶的产生。  相似文献

从中国分离的149株木腐菌中,有30株具有漆酶活性。在添加了阿魏酸的液体培养基中,漆酶产量高(每升培养液中漆酶含量在10000单位以上)的菌株有9个:游离细胞培养的彩绒革盖菌Coriolus versicolor G30,相邻小孔菌Microporus affinis G07,血红密孔菌Pycnoporus sanguineus W006,W006-2,W3008,G05和香菇Lentinus edodes G18,以及固定细胞培养的鲑贝革盖菌Coriolus consors 98563和干酪菌Tyromyces sp. 98420。彩绒革盖菌、相邻小孔菌和血红密孔菌具有重要的生物工程研究价值。  相似文献

在以猪粪为原料的静态条垛堆肥的堆体试验中添加了彩绒革盖菌,研究其对堆肥发酵的影响。研究表明彩绒革盖菌在堆肥二次发酵时期有利于堆体温度的提升和保温,说明了在进入堆肥后期彩绒革盖菌对其中剩余的木质素等成分有很好的分解能力,有利于堆肥的腐熟和养分的释放,初步表明彩绒革盖菌是一株理想的堆肥发酵菌株。  相似文献

彩绒革盖菌CV-8胞外漆酶的诱导、纯化及部分性质研究   总被引:2,自引:0,他引:2  
为探讨彩绒革盖菌(Coriolus versicolor)漆酶的生产条件和性质,对其进行了硫酸铵盐析、半透膜透析、DEAE-纤维素离子交换、Sephadex G100柱层析纯化,粗酶液被纯化36.1倍,比活力10180u/mg,回收率44.6%。凝胶过滤法测定酶的分子量为62kDa。研究了漆酶的产酶曲线及酶作用最适条件。结果表明在该培养条件下,彩绒革盖菌第11d达产酶高峰,峰值酶活为398u/ml,酶作用的最适pH值为4.6,最适温度为25℃,Mg2 、Mn2 、Cu2 对漆酶有激活作用,而Ag 、Fe3 和Cl-则抑制漆酶活性。以邻联甲苯胺为底物的表观Km值为892×10-3mol/L。该菌可作为木质素降解和环保工业漆酶生产菌种。  相似文献

以花脸香蘑Lepista sordida为材料,研究其分别在9种碳源和11种氮源液体培养条件下不同阶段pH值和葡萄糖浓度的变化,以及不同碳氮源对其所分泌的木质素过氧化物酶、羧甲基纤维素酶、锰过氧化物酶和漆酶活性的影响。结果表明,pH值在不同碳源培养后期变化显著(P<0.05),而在不同氮源培养阶段无明显变化(P>0.05),葡萄糖浓度和菌丝量在不同碳氮源中也无显著差异(P<0.05)。不同碳源和氮源培养基对花脸香蘑木质素过氧化物酶、羧甲基纤维素酶、锰过氧化物酶和漆酶活性均具有影响(P<0.05)。木糖和米糠有利于花脸香蘑分泌羧甲基纤维素酶(P<0.05),红糖和牛肉浸膏有利于其分泌漆酶(P<0.05),白砂糖和豆粉有利于其分泌锰过氧化物酶(P<0.05),木糖和尿素有利于其分泌木质素过氧化物酶(P<0.05)。本研究为选择合适培养基以提高花脸香蘑生物转化效率提供了理论基础。  相似文献

以糙皮侧耳Pleurotus ostreatus为材料,研究简单碳氮源及木质素纯品诱导条件对其木质纤维素酶活性的影响。结果表明,不同的碳源培养基和氮源培养基对糙皮侧耳漆酶活性、羧甲基纤维素酶活性和木聚糖酶活性均具有极显著的影响(P<0.001),且对糙皮侧耳菌丝生物量也有极显著的影响(P<0.001)。以蔗糖作主要碳源诱导物时,有利于提高糙皮侧耳漆酶活性;以果糖作主要碳源诱导物时,有利于提高糙皮侧耳羧甲基纤维素酶活性和菌丝生物量的积累;以葡萄糖作主要碳源诱导物时,有利于提高糙皮侧耳木聚糖酶活性。以酵母浸粉作主要氮源诱导物时,有利于提高糙皮侧耳漆酶活性和菌丝生物量的积累;以硝酸钾作为主要氮源诱导物时,有利于提高糙皮侧耳羧甲基纤维素酶活性;以硫酸铵作为主要氮源诱导物时,有利于提高糙皮侧耳木聚糖酶活性。碱性木素的存在,有利于提高糙皮侧耳漆酶活性,但不利于菌丝生物量的积累。与此同时,碱性木素的存在对糙皮侧耳羧甲基纤维素酶和木聚糖酶活性并没有促进作用。  相似文献

实验研究了碳源、氮源和综合因素对毛栓菌(Tramets trogii)液体培养产漆酶和生长情况的影响。碳源为玉米面、氮源为豆饼粉时,菌株的产漆酶酶活最高;碳源为可溶性淀粉,氮源为麦麸时,对菌株的生长有利;在液体条件下,菌株产漆酶的最佳培养基组成为玉米面3.5%,豆饼粉4.0%,KH2PO40.3%,MgSO40.15%,VB10.04%。  相似文献

Laccases have great biotechnological potential in diverse industries as they catalyze the oxidation of a broad variety of chemical compounds. Production of laccases by basidiomycetes has been broadly studied as they secrete the enzymes, grow on cheap substrates, and they generally produce more than one isoenzyme (constitutive and/or inducible). Laccase production and isoenzyme profile can be modified through medium composition and the use of inducers. The objective of this work was to increase laccase production by Pleurotus ostreatus CP-50 through culture medium optimization and the simultaneous use of copper and lignin as inducers. Increased fungal growth was obtained through the use of a factorial fractional experimental design 26–2 where the influence of the nature and concentration of carbon and nitrogen sources was assessed. Although specific laccase production (U/mg biomass) decreased when malt extract medium was supplemented with carbon and nitrogen sources, fungal growth and laccase volumetric activity increased four and sixfold, respectively. The effect of media supplementation with copper and/or lignin on laccase production by P. ostreatus CP-50 was studied. A positive synergistic effect between copper and lignin was observed on laccase production. Overall, the use of an optimized medium and the simultaneous addition of copper and lignin improved growth, laccase volumetric activity, and process productivity by 4-, 60-, and 10-fold, respectively.  相似文献

It has been shown that the wood-rotting mushroom Pleurotus dryinus IBB 903 is able to effectively produce cellulases, xylanase, laccase, and manganese peroxidase in submerged fermentation of mandarin peels and tree leaves. Gradual increasing of lignocellulosic substrates concentration from 1 to 4–6% enhanced enzyme accumulation in culture liquid. A simple and inexpensive medium containing mandarin peels and yeast extract as sole carbon and nitrogen sources allowed simultaneous production of high levels of both hydrolases and oxidases by P. dryinus IBB 903. Supplementation of this medium by copper and manganese caused earlier and faster accumulation of laccase and manganese peroxidase increasing their yield by 1.5 and 7.5 times, respectively. In addition, by adding manganese to the medium it is possible to regulate the ratio of laccase and MnP in enzyme preparation. The presence of lignocellulosic substrate is the requisite for MnP production by P. dryinus IBB 903 since there was no production of MnP when mushroom has been cultivated in the synthetic medium with different carbon source. Among carbon source tested only utilization of glucose resulted to 21-fold increase of fungus laccase specific activity compared to control medium without carbon source. Carboxymethyl cellulase and xylanase appeared to be inducible enzymes.  相似文献

The effects of the carbon and nitrogen sources, initial pH and incubation temperature on laccase production by the endophytic fungus Monotospora sp. were evaluated. The optimal temperature and initial pH for laccase production by Monotospora sp. in submerged culture were found to be 30 degrees C and 8.5, respectively. Maltose (2 g l(-1)) and ammonium tartrate (10 g l(-1)) were the most suitable carbon and nitrogen source for laccase production. Under optimal culture medium, the maximum laccase activity was determined to be 13.55 U ml(-1), which was approximately four times higher than that in basal medium. This is the first report on laccase production by an endophytic fungus.  相似文献

漆酶高产菌株的诱变选育及其产酶条件   总被引:10,自引:5,他引:10  
以粗毛栓菌Trametesgallica为出发菌,通过紫外诱变处理其担孢子、PDA-RBBR平板变色法初筛、ABTS法测定培养液漆酶酶活力复筛,获得1株漆酶高产诱变菌株SAH-12。用高氮低碳无机盐培养液(LM3)培养时,其峰值酶活力比出发菌株高出4倍,达到5002.6U/L,且产酶稳定。对SAH-12液体培养产酶条件的研究表明:以纤维二糖和蔗糖为碳源明显优于麦麸、淀粉和葡萄糖,其最高酶活分别达18526U/L和13436U/L;有机氮源较无机氮源更有利于SAH-12漆酶的分泌,以蛋白胨、大豆粕和胰化蛋白胨为氮源时其峰值酶活分别达到20544U/L、19671U/L和16180U/L;适宜初始培养pH为4.0;ABTS、单宁酸、没食子酸对产酶均有明显的诱导作用,其中ABTS和单宁酸的诱导效果相对更好,愈创木酚和吐温80对产酶有一定的抑制作用。  相似文献

Summary The effect of various carbon and nitrogen sources on laccase, manganese-dependent peroxidase (MnP), and peroxidase production by two strains of Pleurotus ostreatus was investigated. The maximal laccase yield of P. ostreatus 98 and P. ostreatus 108 varied depending upon the carbon source from 5 to 62 U l−1 and from 55 to 390 U l−1, respectively. The highest MnP and peroxidase activities were revealed in medium supplemented by xylan. Laccase, MnP, and peroxidase activities of mushrooms decreased with supplementation of defined medium by inorganic nitrogen sources. Peptone followed by casein hydrolysate appeared to be the best nitrogen sources for laccase accumulation by both fungi. However, their positive effects on enzyme accumulation were due to a higher biomass production. The secretion of MnP and peroxidase by P. ostreatus 108 was stimulated with supplementation of casein hydrolysate to the control medium since the specific MnP and peroxidase activities increased 15-fold and 3.5-fold, respectively.  相似文献

研究了碳源、氮源、愈创木酚、香兰素及培养条件对漆酶分泌的影响;结果表明,麦草粉作碳源、(NH_4)_2SO_4作氮源有利于漆酶的分泌,适宜浓度的愈创木酚和香兰素等对漆酶的产生有一定的作用;pH在3.0~8.0的范围内对漆酶的分泌影响差别不大,培养温度、接种量、通气量对漆酶的分泌有较大影响。  相似文献
