The fumigant methyl bromide was evaluated for genotoxicity in the somatic wing-spot assay of Drosophila melanogaster. Third instar larvae trans-dihybrid for mwh and flr3 were exposed to varying concentrations (0-16 mg/l) of the gas for 1 h. Following this exposure via inhalation, the larvae were placed into vials containing Instant Medium. 7 days after the exposure, the adult flies in the vials were collected, and their wings were scored under 400X magnification for the presence of clones of cells possessing malformed wing-hairs. Such clones appeared as mwh-flr3 twin spots and single spots of either mwh or flr3 phenotype. Exposure to methyl bromide was found to result in the positive induction of both twin spots and large (greater than 2 cells) single spots. For each endpoint, a significant exponential association was obtained between concentration and frequency of spots per wing. Methyl bromide was found to be a negative inducer of small (1-2 cells) single spots at all concentrations except 16 mg/l where a positive effect was observed. Because twin spots arise exclusively from mitotic recombination, methyl bromide was identified as having recombinogenic activity in the somatic tissue of Drosophila larvae.  相似文献

Grell's (1964) “non-competitive” data describing segregation of markers on two non-homologous, non-crossover chromosomes inDrosophila melanogaster, called Dp and 4, are explained on the basis of the “elastica” but also on the chance of the two becoming associated with each other. According to the model,early in meiosis the chance that two chromosomes pair with each other at the (terminal) centromeres and telomeres, p1, is an inverse function of difference in their lengths (the Dp chromosome's length, called LDp, was different in different flies, whereas that of the non-exchange 4 was the same throughout). If these two chromosomes, once paired, are not approximately equal in length, thenlate in meiosis as they both spiralize and stiffen, their ends tend to separate as predicted by the elastica, thus preventing co-orientation and bringing about elevation of the % Dp, 4 non-disjunction. Such paired chromosomes are compared with the form of the elastica describing a strung (arrow) bow. That is, beginning with a string and bow of equal lengths and allowing the string to become shorter (Dp shorter than 4), moments at the paired centromeres and/or telomeres would increase much more rapidly than if the bow becomes longer (Dp longer than 4), the model predicts that the former rate of increase is the square of the latter, in good agreement with the data. Other experimental results are also consistent with those predicted by the elastica. In “competitive” data with three non-crossover, non-homologous chromosomes, T (also constant in length) Dp and 4, the following T, Dp: T, 4: Dp, 4 non-disjunction ratios are easily explained on the basis of a random Markov (stochastic) process hypothesizing three steps between pairing and M1: 2/3:: 1/6 : 1/6 at LDp=0.3, 2/5 : 1/5 : 0 at LDp=0.9, 1/6 : 1/2 : 0 at LDp=1.4 and 1/6 : 2/3 : 1/6 at LDp=3.0.  相似文献

The possibility of somatic mobilisation of P elements in Drosophila melanogaster was investigated. Flies, trans-heterozygous for the genetic markers mwh and flr3, were obtained by crossing males containing transposition-competent P elements with females having M cytotype. The hybrid dysgenic flies were reared at 29 degrees C and their wings examined for mutant clones. The frequency of mutant spots found on the wings of the female flies was significantly higher than that of female control flies. We postulate that this increase in frequency may be due to P element mobilisation at high temperature in the wing cells of dysgenic hybrids. This is in direct contrast to the large body of research which indicates that P-transposition-mediated mutation is restricted to the germline.  相似文献

The effects of 13 mutagen-sensitive (mus) mutants (representing seven loci) on mitotic chromosome stability in nonmutagenized cells have been examined genetically. To do this, mus-bearing flies heterozygous for the recessive somatic-cell marker, multiple wing hairs (mwh), were examined for increased frequencies of mwh clones in the wing blade. Mutants at the mus-103, mus-104 and mus-106 loci do not affect the frequency of mwh clones, while mus-101, mus-102, mus-105 and mus-109 alleles cause increases in the frequency of mwh clones. These data show that the wild-type alleles of latter four loci specify functions that are required for chromosome stability in nonmutagenized cells. Analysis of the size distribution of mwh clones produced by these mutants suggests that most chromosome instability caused by these mutants is the consequence of chromosome breakage; in the presence of mus-105 and mus-109 alleles a small fraction of the mwh clones are produced by an event (mitotic recombination, mutation, nondisjunction) that produces euploid clones. To inquire whether any of the extant alleles of the mus-101, mus-102, mus-105 and mus-109 loci might be leaky alleles of loci that carry out essential mitotic functions, chromosome stability in females homozygous for alleles of these loci has been compared to that of females carrying one dose of a mutant over a deficiency for that mus locus. These comparisons show that the extant alleles at the mus-101, mus-109 and mus-105 loci are all leaky mutants. It is suggested that all three of these loci may specify essential mitotic functions.  相似文献

Hydroxyl radical (OH) is a radical species highly destructive for lignin during solid-state fermentation (SSF) of straw with Phanerochaete chrysosporium (Pc). The production of OH at different initial Pb2+ concentrations during SSF of straw with Pc was investigated. The results showed that a modest amount (under 200 mg kg−1) of Pb2+ could enhance the production of OH, while a higher Pb2+ concentration resulted in inhibition. The content of OH reached the peak value at day 12 in the whole tested samples, and the maximal content of OH was obtained at initial Pb2+ concentration of 100 mg kg−1. It was also found that the production of OH was connected to enzymatic activity and oxalate content in some degree, in particular, a significant positive correlation was found between oxalate concentration and production of OH.We found that low concentration of Pb2+ can promote the degradation of lignin, and the higher initial Pb2+ concentration (400 mg kg−1) resulted in inhibition. In addition, it appeared that there was no significant correlation between lignin degradation rate and the production of OH when Pb2+ concentration was taken into account.  相似文献

Summary We tested, through field experiments and simulation models, the hypothesis that fruit-searching tephritid fruit flies adjust their within-tree search persistence according to the sequence and timing of encounters with parasitized (i.e. egg-infested) and unparasitized Crataegus sp. host fruit. In the field, we presented flies with 4 different sequences of unparasitized [=C] and parasitized [=M] fruit: 5M+0C; 1C+5M; 5M+1C, 1C+0M. Following fruit presentation, flies were permitted to forage freely within trees, which harboured no fruit, until emigration occurred. Under these conditions, flies that encountered the aforementioned different sequences of hosts, displayed differences in Giving Up Time, measured as active foraging time and number of leaf visits, in a manner predicted prior to testing. These differences were, however, not statistically significant. Based upon the results described above, we then built 3 simulation models that predicted within-tree Giving Up Times for individual flies: Model 1-Giving Up Time is incremented and decremented by fixed amounts following encounters by the fly with suitable (i.e. for oviposition) and unsuitable hosts, respectively; Model 2-similar to Model 1, but increment and decrement values are variable and are dependent upon the time since previous encounters by the fly with suitable and unsuitable hosts; Model 3-Giving Up Time is fixed. Comparison with previously reported field data for tephritid flies showed that Model 2 predicted rather well, and significantly better than Models 1 and 3, Giving Up Time for wild type tephritid flies under seminatural field conditions. We discuss our results in light of contemporary foraging theory.  相似文献

The newly developed somatic mutation and recombination test, utilizing the wing-hair mutations mwh and flr3, was used to evaluate the genotoxicity of the base analog 5-azacytidine in larvae of Drosophila melanogaster. Third instar larvae were fed media wetted with various concentrations of the compound, and wings of surviving adults were removed and scored for the presence of clones of cells possessing malformed hairs. Wings of exposed flies trans-dihybrid for mwh and flr3 had significantly increased frequencies of twin spots, small single spots and large single spots. Significant linear regression of twin-spot frequencies upon concentration was also obtained. Induction of twin spots by 5-azacytidine unambiguously demonstrates its recombinogenic activity in somatic cells of Drosophila. Significantly increased frequencies of large single spots on wings of inversion-heterozygous flies were also observed and suggest that 5-azacytidine may also be inducing somatic gene mutations (or deletions).  相似文献

We determined the effects of paraquat (PQ) concentrations ranging from 10(-3) to 10(-2) M and three levels of venous PO2 [hypoxia (41 +/- 3 Torr), normoxia (147 +/- 8 Torr), and hyperoxia (444 +/- 17 Torr)] in the presence of 4 x 10(-3) M PQ on microvascular permeability in isolated blood-perfused dog lungs. Capillary filtration coefficient (Kf,c) increased and isogravimetric capillary pressure (Pc,i) decreased 3 h after perfusion with 10(-2) M PQ (n = 7) and 5 h after perfusion with 4 x 10(-3) M PQ (n = 6) but not with 10(-3) M PQ (n = 4). In hyperoxic lungs perfused with 4 x 10(-3) M PQ, Kf,c increased to nine times the base-line value 5 h after PQ [0.15 +/- 0.01 to 1.35 +/- 0.25 (SE) ml.min-1.cmH2O-1.100 g-1]. Pc,i significantly decreased from a base-line value of 9.4 +/- 0.2 to 7.1 +/- 0.4 cmH2O at 3 h. In hypoxic lungs perfused with 4 x 10(-3) M PQ (n = 5), Pc,i and Kf,c changes were not significantly different from those in normoxic lungs treated with PQ. Thus both hyperoxia and an increased dose of PQ shortened the latent period and increased the severity of the PQ-induced microvascular permeability lesion, but hypoxia failed to prevent the PQ damage.  相似文献

Human atopen-specific types 1 and 2 T helper cell clones.   总被引:11,自引:0,他引:11  
Eight representative T lymphocyte clones (TLC) randomly selected from previously described panels of CD4+ housedust mite Dermatophagoides pteronyssinus (Dp)-specific TLC from atopic and nonatopic donors were studied in more detail in a comparative investigation. The TLC from the atopic donors closely resembled murine type 2 Th (Th2) cells by secreting substantial IL-4, IL-5, IL-6, TNF-alpha, and granulocyte-macrophage (GM)-CSF, minimal IFN-gamma, and relatively little IL-2. In contrast, the nonatopic's TLC resembled murine type 1 Th (TH1) cells by secreting substantial IFN-gamma, IL-2, TNF-alpha, and GM-CSF, no IL-4, and little IL-5. A difference with murine Th1 cells was their additional secretion of IL-6. These cytokine profiles were consistent upon stimulation via different activation pathways including stimulation with specific Dp Ag, mitogenic lectins, and antibodies to CD2, CD3, or CD28. The observed differences in IL-2 secretion, however, were most evident upon stimulation with anti-CD28. If TLC cells were cultured with highly purified B cells and stimulated with anti-CD3 in the absence of exogenous IL-4, IgE synthesis was induced only in cultures with the atopics' Th2 clones, which could be completely abrogated by anti-IL-4. The mere presence of exogenous rIL-4, however, did not result in IgE synthesis, nor did unstimulated TLC cells alone. But if unstimulated TLC cells (that proved not to secrete detectable amounts of cytokines) were added together with rIL-4, again IgE synthesis was induced only in cultures with the atopics' Th2 clones, suggesting the involvement of an additional, as yet unidentified accessory helper function of the atopics' Th2 clones for IgE induction. Unstimulated Th2 clones showed a significantly higher expression of CD28 than the Th1 clones, but three days after stimulation, CD28 expression was elevated to comparable levels on both subsets. When added to B cells at this time point, together with rIL-4 and anti-IFN-gamma, still only the atopics' Th2 clones supported IgE synthesis, arguing against a role for CD28 in this accessory helper function. Whereas the atopics' Th2 clones were excellent helper cells for IgE induction, a unique property of the nonatopic's Th1 clones was their cytolytic activity toward autologous APC which could be induced by specific Dp Ag and by anti-CD3. The present data provide clear evidence for the existence of Th1 and Th2 cells in man.  相似文献

Four complexes - [(15C5)4Pc]M(Pc)M(Pc), (Pc2− - phthalocyaninato-dianion, [(15C5)4Pc]2− - 2,3,9,10,16,17,24,25-tetrakis(15-crown-5)phthalocyaninato-dianion, M = Sm, Dy, Tm, Y) were obtained via the reaction of M(Pc)2, H2[(15C5)4Pc] and M(acac)3. The influence of the stability of starting M(Pc)2 on the yields of target compounds was investigated. Increasing the stability of M(Pc)2 leads to higher yields of [(15C5)4Pc]M(Pc)M(Pc) and lower yields of scrambling products. All complexes were characterized by 1H NMR, UV-Vis and FT-IR spectroscopy as well as MALDI TOF mass-spectrometry. The analysis of 1H NMR spectra was performed in terms of lanthanide-induced shifts. Cation-induced dimerisation was studied by means of spectrophotometric titration. Supramolecular dimers {2[(15C5)4Pc]M(Pc)M(Pc) · 4K+} are the largest discrete cofacial supramolecular assemblies built of phthalocyanine building blocks reported up-to-date. The observed increase of the intermolecular excitonic interaction between building blocks with the increase of REE(III) size is tentatively explained in terms of metal-size dependent deformation of phthalocyanine ligands in sandwich complexes.  相似文献

We previously proposed specific interaction of Lex (Gal1 4[Fuc1 3]-GlcNAc1 3Gal) with Lex as a basis of cell adhesion in pre-implantation embryos and in aggregation of F9 teratocarcinoma cells, based on several lines of evidence (Eggenset al., J Biol Chem (1989)264:9476–9484). We now present additional evidence for this concept, based on autoaggregation studies of plastic beads coated with glycosphingolipids (GSLs) bearing Lex or other epitopes, and affinity chromatography on Lex-columns of multivalent lactofucopentaose III (Lex oligosaccharide) conjugated with lysyllysine. Comparative adhesion studies of Lex-expressing tumour cellsvs their Lex-non-expressing variants showed that only Lex-expressing cells adhere to Lex-coated plates and are involved in tumour cell aggregation, in analogy to F9 cell aggregation. The major carrier of Lex determinant in F9 cells is not GSL but rather polylactosaminoglycan (embryoglycan), and we demonstrated autoaggregation of purified embryoglycan in the presence of Ca2+, and reversible dissociation in the absence of Ca2+ (addition of EDTA). Defucosylated embryoglycan did not show autoaggregation under the same conditions. Thus, Lex-Lex interaction has been demonstrated on a lactosaminoglycan basis as well as a GSL basis. We have measured UV-induced mutation frequencies in yeast in a forward, nonselective assay system by scoring white adex ade2 double auxotrophs among parental red-pigmented ade2 clones. The frequencies of sectored and pure mutant clones were determined separately. In excision-defective strains carrying the genes rad1–1, rad3–2 and rad4–4, as well as in the double mutants, rad 1–1 rad 3–2 and rad 1–1 rad 4–4, considerably more sectored than pure clones are induced in the low-dose range; in repair-competent strains, pure mutant clones substantially outnumber the sectored clones. These results can be explained on the basis of known differences in the timing of error-prone repair during the cell division cycle; that is, we assume that error-prone repair occurs primarily before replication in RAD wild-type strains but after replication in excision-deficient mutants. It has been suggested that excision deficiency has a pleiotropic effect on heteroduplex repair and nucleotide excision repair; however, the high percentage (36.6%) of half-sectored clones found in the rad1–1 strain is hard to reconcile with this hypothesis. We propose that heteroduplex repair occurs subsequent to error-prone repair in both excision-proficient and excision-deficient strains.  相似文献

Mitochondria generated nitric oxide (NO) regulates several cell functions including energy metabolism, cell cycling, and cell death. Here we report that the NO synthase inhibitors (L-NAME, L-NNA and L-NMMA) administered either in vitro or in vivo induce Ca2+-dependent mitochondrial permeability transition (MPT) in rat liver mitochondria via a mechanism independent on changes in the energy state of the organelle. MPT was determined by the occurrence of cyclosporin A sensitive mitochondrial membrane potential disruption followed by mitochondrial swelling and Ca2+ release. In in vitro experiments, the effect of NOS inhibitors was dose-dependent (1 to 50 µM). In addition to cyclosporin A, L-NAME-induced MPT was sensitive to Mg2+ plus ATP, EGTA, and to a lower degree, to catalase and dithiothreitol. In contrast to L-NAME, its isomer D-NAME did not induce MPT. L-NAME-induced MPT was associated with a significant decrease in both the rate of NO generation and the content of mitochondrial S-nitrosothiol. Acute and chronic in vivo treatment with L-NAME also promoted MPT and decreased the content of mitochondrial S-nitrosothiol. SNAP (a NO donor) prevented L-NAME mediated MPT and reversed the decrease in the rate of NO generation and in the content of S-nitrosothiol. We propose that S-nitrosylation of critical membrane protein thiols by NO protects against MPT.  相似文献

Spin-coated films of 29H,31H-tetra-4-(2,4-di-tert-amylphenoxy)phthalocyanine (H(2)Pc) and [kappaP,mu-kappaS-(dppeS)Pt(CH(3))](2)[BF(4)](2) have been used as sensing layers deposited in thin film form for the detection of VOCs. The sensing behaviour of the blend was predicted on the basis of mass spectrometric determinations performed on H(2)Pc/Pt-complex solutions, by monitoring the formation of gas-phase ions at the electrospray interface. The addition of small amounts of acetonitrile produced a [M+41](+) peak whereas the addition of similar amounts of methanol, ethanol and isopropyl alcohol did not give the corresponding [M+ROH](+) species. These results were confirmed by sensing tests. A pure phthalocyanine optosensing element did not show relevant selectivity. Conversely, the heterogeneous sensing layer obtained by spin-coating deposition of a Pt-complex/H(2)Pc blend allowed the sensing of acetonitrile vapours with respect to the above mentioned alcohols.  相似文献

Summary We have studied the influence of various factors on the V-type position effect of the white gene transposed to heterochromatin as a result of different chromosome rearraugements in D. melanogaster. Variegation due to the white gene position effect is much weaker if the flies have received Dp(1;3)wvco from parental males, and not females. The origin of the chromosome rearrangement does not have this influence in the case of T(1;4)wm5 or has it to insignificant extent in the case of In(1)wm4. The Y-chromosome in maternal genome strongly suppresses Dp(1;3)wvco-induced variegation even in the progeny which has not received an extra Y-chromosome but only if this progeny gets Dp(1;3)wvco from the same female. The low temperature (16° C) at which parental females have developed, considerably affects the position effect in the progeny with Dp(1;3)wvco, whereas the temperature of males' development has no influence at all. The maternal temperature effect occurs also when Dp(1;3)wvco has come down from the father, though it is stronger if the mother subjected to low temperature treatment bore the rearrangement. The influence of temperature seems to take effect at the final stages of oogenesis.The data obtained lead one to suppose that the influence of genotypic and external factors on variegation is passed to the next generation of flies in different ways. The direction of crosses and additional Y-chromosome heterochromatin in the maternal genome seem to affect variegation in the progeny through changes in the properties (structure) of the chromosome rearrangement expressing the position effect. As to the temperature of the mothers development, only a small part of its influence may be accounted for by the same mechanism, whereas most of the temperature influence seems to be passed on through other components of the nucleus or through the cytoplasm.  相似文献

A novel reductive compound with molecular weight of about 1000 Da, named Pc reducer, was purified from the liquid culture of a white-rot basidiomycete Phanerochaete chrysosporium. It was likely to have an alkene-ester structure according to Fourier-transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance (NMR) spectra. Pc reducer reduced the hydroxyl radical HO and the stable nitroxide radical under certain conditions. It inhibited the repolymerization of the products from the oxidation of phenolic lignin-model compounds by reducing certain intermediate radicals. The activity of manganese peroxidases was promoted by Pc reducer at certain concentrations. Pc reducer could also weaken the repolymerization of fragments from the oxidation of Na-lignosulfonate by lignin peroxidases and manganese peroxidases. It has potential ability to improve the ligninolytic efficiency of peroxidases in P. chrysosporium.  相似文献

Thiosemicarbazones such as Triapine (Tp) and Dp44mT are tridentate iron (Fe) chelators that have well-documented antineoplastic activity. Although Fe–thiosemicarbazones can undergo redox cycling to generate reactive species that may have important roles in their cytotoxicity, there is only limited insight into specific cellular agents that can rapidly reduce Fe(III)–thiosemicarbazones and thereby promote their redox activity. Here we report that thioredoxin reductase-1 (TrxR1) and glutathione reductase (GR) have this activity and that there is considerable specificity to the interactions between specific redox centers in these enzymes and various Fe(III) complexes. Site-directed variants of TrxR1 demonstrate that the selenocysteine (Sec) of the enzyme is not required, whereas the C59 residue and the flavin have important roles. Although TrxR1 and GR have analogous C59/flavin motifs, TrxR is considerably faster than GR. For both enzymes, Fe(III)(Tp)2 is reduced faster than Fe(III)(Dp44mT)2. This reduction promotes redox cycling and the generation of hydroxyl radical (HO) in a peroxide-dependent manner, even with low-micromolar levels of Fe(Tp)2. TrxR also reduces Fe(III)–bleomycin and this activity is Sec-dependent. TrxR cannot reduce Fe(III)–EDTA at significant rates. Our findings are the first to demonstrate pro-oxidant reductive activation of Fe(III)-based antitumor thiosemicarbazones by interactions with specific enzyme species. The marked elevation of TrxR1 in many tumors could contribute to the selective tumor toxicity of these drugs by enhancing the redox activation of Fe(III)–thiosemicarbazones and the generation of reactive oxygen species such as HO.  相似文献

Segregating F(3) families, derived from a cross between oat cultivar Swan and the putative single gene line PC68, were used to determine the association of seed storage protein loci and resistance gene analogues (RGAs) with the crown rust resistance gene Pc68. SDS-PAGE analysis detected three avenin loci, AveX, AveY, and AveZ, closely linked to Pc68. Their diagnostic alleles are linked in coupling to Pc68 and were also detected in three additional lines carrying Pc68. Another protein locus was linked in repulsion to Pc68. In complementary studies, three wheat RGA clones (W2, W4, and W10) detected restriction fragment length polymorphisms (RFLPs) between homozygous resistant and homozygous susceptible F(3) DNA bulks. Four oat homologues of W2 were cloned and sequenced. RFLPs detected with two of them were mapped using F(3) and F(4) populations. Clone 18 detected a locus, Orga2, linked in repulsion to Pc68. Clone 22 detected several RFLPs including Orga1 (the closest locus to Pc68) and three RGA loci (Orga22-2, Orga22-3, and Orga22-4) loosely linked to Pc68. The diagnostic RFLPs linked in coupling to Pc68 were detected by clone 22 in three additional oat lines carrying Pc68 and have potential utility in investigating and improving crown rust resistance of oat.  相似文献

A series of metal chlorides were subjected to the wing spot test of Drosophila melanogaster. In the test, larvae trans-heterozygous for the wing-hair mutations mwh and flr were orally treated at the third instar stage with a test compound and the wings were inspected at the adult stage for spots expressing phenotypes of the markers. CoCl2, MnCl2, MoCl3, NiCl2 and ZnCl2 were clearly effective in inducing spots with one or two mutant hairs (small spots). CoCl2 was clearly effective in inducing spots with three or more mutant hairs (large spots) as well. CrCl3, FeCl2 and FeCl3 were negative under the conditions used. Based on estimated frequencies of small spots induced at the LD50, the genotoxic effectiveness of the positive metal salts were ranked in a sequence of CoCl2 > ZnCl2 > MoCl3 > (MnCl2, NiCl2). Since CoCl2 did not induce large spots in the wings of the mwh/TM3 flies with a suppressed ability of mitotic crossing-over, the large spots induced by this compound in the mwh/flr system were ascertained as mutant clones due to mitotic crossing-overs.  相似文献
