Propagation and activity level of 18 enzymes catalyzing deamination reactions of dicarboxylic and oxyamino acids and enzymes of amino acid reamination and amino acid N-acyl-derivatives' deacylation have been studied in Klebsiella bacteria. Klebsiella the most actively utilizes serin, threonine, aspartic and glutamic acids and aromatic amino acids. The first three amino acids are utilized by deamination, aromatic acids- in aminotransferase reaction with alpha-ketoglutaric acid, glutamic acid--by deamination and decarboxylation. Besides, Klebsiella actively deacylates N-acyl-derivatives of amino acids.  相似文献

This study was conducted using the piglet model to test the hypothesis that mucosal cells of the neonatal small intestine can degrade nutritionally essential amino acids (EAA). Enterocytes were isolated from the jejunum of 0-, 7-, 14-, and 21-day-old pigs, and incubated for 45 min in Krebs buffer containing plasma concentrations of amino acids and one of the following L-[1-14C]- or L-[U-14C]-amino acids plus unlabeled tracees at 0.5, 2, or 5 mM: histidine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophan and valine. In these cells, branched-chain amino acids (BCAA) were extensively transaminated and 15–50% of decarboxylated branched-chain α-ketoacids (BCKA) were oxidized to CO2 depending on the age of piglets. BCAA transamination increased but their decarboxylation decreased between 0 and 14 days of age. Addition of 1 and 2 mM α-ketoglutarate to incubation medium dose-dependently stimulated BCAA transamination without affecting their decarboxylation. Western blot analysis revealed that the abundance of mitochondrial BCAA aminotransferase declined but cytosolic BCAA aminotransferase increased between 0 and 14 days of age, with the cytosolic protein being the major isoform in 7- to 21-day-old pigs. BCKA dehydrogenase protein existed primarily as the phosphorylated (inactive) form in enterocytes of newborn pigs and its levels were markedly reduced in older pigs. All measured parameters of BCAA metabolism did not differ between 14- and 21-day-old pigs. In contrast to BCAA, catabolism of methionine and phenylalanine was negligible and that of other EAA was absent in enterocytes from all ages of piglets due to the lack of key enzymes. These results indicate that enterocytes are an important site for substantial degradation of BCAA but not other EAA in the neonatal gut.  相似文献

1. The concentrations of alanine, aspartate, glutamate, glutamine and serine plus threonine have been measured by enzymic methods in ;quick-frozen' livers from normal, starved, alloxan-diabetic and phlorrhizin-treated rats. 2. The hepatic concentrations of alanine and serine plus threonine were decreased in rats starved for 48hr. Treatment of these rats with phlorrhizin resulted in a rapid fall (within 2(1/2)hr.) in the concentrations of all the glucogenic amino acids except serine plus threonine, which increased. The pattern for alloxan-diabetic rats was similar to that for phlorrhizin-treated animals, except that here serine plus threonine also decreased in concentration. 3. The effects of anoxia on the hepatic concentrations of the glucogenic amino acids are reported. 4. Inhibition of glutamate-pyruvate transaminase in vivo by l-cycloserine resulted in the accumulation of alanine in situations involving high rates of gluconeogenesis from endogenous amino acids. 5. Measurements of the concentrations of the reactants of the glutamate-pyruvate transaminase and glutamate-oxoglutarate transaminase systems in various metabolic states suggest that they are both at or near equilibrium in rat liver. 6. New enzymic methods are described for the determination of serine plus threonine and alanine.  相似文献

The release of radioactive amino acid from the particulate fractions separated from the prelabelled livers of rats by centrifugation has been studied under various conditions. Although pure fractions may not have been obtained, great differences in behavior were observed. In the mitochondria and nuclei dinitrophenol (10(-4)M) causes an inhibition of release, but in microsomes the opposite effect is observed. When the incubation medium is fortified with ATP and phosphocreatine, release is inhibited. In microsomes and nuclei the inhibition proceeds to the extent that the incorporation of preformed radioactive amino acid occurs. Protein is synthesized at a rapid rate. In incubations longer than 1 hour there is always a release of radioactive amino acid. It is concluded from these results that the interpretation of release data from slices or systems such as those studied is impossible without further information concerning some of the unknown variables. The most important unknown is the specific activity of the "free" amino acid in the particulates and the effect of carrier amino acid in the medium of this specific activity.  相似文献

Fatty-acid synthesis has been measured in vivo with3H2O in cafeteria-fed rats exhibiting diet-induced thermogenesis. Synthesis was decreased in brown adipose tissue, the liver, white adipose tissue, and the carcass of the cafeteria-fed animals compared to rats fed the normal stock diet. Whole-body synthesis was also decreased in the cafeteria-fed group. Diet-induced thermogenesis, in contrast to cold-induced non-shivering thermogenesis does not lead to increased fatty-acid synthesis and this is presumably due to the inhibitory effects on lipogenesis of the high dietary fat intake characteristic of cafeteria diets. The results also indicate that the energy cost of body fat deposition in cafeteria-fed rats is lower than in animals fed a low-fat/high-carbohydrate stock diet.  相似文献

The overall oxidative degradation of leucine and phenylalanine, measured in vivo in rats fed on a 2%-casein diet for 8 weeks, is markedly decreased as compared with controls, whereas that of glutamate and alanine is apparently unaffected. The decrease in leucine degradation is due, at least in part, to a block before the formation of 3-methylbutyryl-CoA (isovaleryl-CoA) in the catabolic pathway. This phenomenon is accompanied by increased incorporation of [14C]leucine into liver proteins, decreased urinary excretion of leucine and increased urinary excretion of 4-methyl-2-oxopentanoate (α-oxoisocaproate) by protein-depleted animals. The results suggest the existence of adaptive mechanisms that function to conserve an indispensable carbon skeleton.  相似文献

The effects of the sequential addition of glucose, noradrenaline, propranolol and oleic acid on the rates of O2 consumption and heat production by isolated interscapular brown adipocytes from control and cafeteria-fed rats were compared. Although the chemical agents produced very similar changes in oxidative metabolism, the actual rates of O2 uptake and heat output in adipocytes from the cafeteria-fed rats, when expressed per g dry wt. of cells, were approx. 65% less than those obtained with cells from the control rats. However, when the same results were expressed per 10(8) multiloccular brown adipocytes, rather than gravimetrically, rates of O2 consumption and heat production were equivalent. Further interpretation of these data is complicated, because the average volume of multiloccular brown adipocytes from cafeteria-fed rats was 2.5 times that for multiloccular cells from control animals.  相似文献

Trichosporon cutaneum readily metabolized protocatechuate, homoprotocatechuate, and gentisate, but lacked ring fission dioxygenases for these compounds. Benzoic, salicylic, 2,3-dihydroxybenzoic, and gentisic acids were converted into beta-ketoadipic acid before entry into the Krebs cycle. Benzoic acid gave rise successively to 4-hydroxybenzoic acid, protocatechuic acid, and hydroxyquinol (1,3,4-trihydroxybenzene), which underwent ring fission to maleylacetic acid. Salicylate and 2,3-dihydroxybenzoate were both initially metabolized to give catechol. 2,3-Dihydroxybenzoate was the substrate for a specific nonoxidative decarboxylase induced by salicylate, although 2,3-dihydroxybenzoate was not a catabolite of salicylate. Gentisate was metabolized to maleylacetic acid and was also readily attacked by salicylate hydroxylase at each stage of a partial purification procedure. Phenylacetic acid was degraded through 3-hydroxyphenylacetic, homogentisic, and maleylacetoacetic acids to acetoacetic and fumaric acids. All the reactions of these catabolic sequences were catalyzed by cell extracts, supplemented with reduced pyridine nucleotide coenzymes where necessary, except for the hydroxylations of benzoic and phenylacetic acids which were demonstrated with cell suspensions and isotopically labeled substrates.  相似文献

Optimum conditions for the growth ofPseudomonas arvilla, a hydrocarbon utiliser, have been studied. The microorganism produced economic cell yield at pH 5.7 and 4% kerosene concentration. C10-C16 hydrocarbons were utilised by the strain. The growth was maximum on n-decane. Supplementation of the hydrocarbon medium with 0.5% glucose stimulated the growth. Glutamic acid 16.0 mg; leucine 9.0 mg; valine 10.0 mg; methionine 2.5 mg; arginine 2.5 mg; histidine 1.0 mg were present in 100 ml of the broth. Cell protein contained leucine 13.69%, isoleucine 4.9%, histidine 4.37%, tryptophan 2.33%, methionine 1.8% and arginine 2.70%.  相似文献
