Karyotype, location of the nucleolar organiser region (NOR) and heterochromatin presence and composition were studied in the Antarctic scallop Adamussium colbecki Smith, 1902. The karyotype exhibits 2n = 38 chromosomes with 11 pairs of metacentrics, 5 of submetacentrics, one subtelocentric and two telocentrics. Ag–NOR, CMA3, DA/MM and NOR–FISH evidenced paracentromeric NORs on the short arm of 2nd pair chromosomes. Digestion with three restriction endonucleases followed by sequential staining with Giemsa, CMA3 and DAPI evidenced on all chromosomes centromeric heterochromatin positive for both DAPI and CMA3. In situ hybridisation analysis showed the presence of an AT-rich satellite DNA in the centromeric heterochromatin of several chromosomes. A mosaicism was detected in the germinal cell lines of one specimen, as in six of the 20 plates examined the set had 37 chromosomes with a missing pair of telocentrics and an unpaired metacentric. Comparison of the chromosome sets of all the pectinids studied to date and comparison with a phyletic tree obtained from molecular mitochondrial genes studies yielded good agreement between karyotype morphology and taxonomic classification.  相似文献

The chromosomes of Steindachneridion sp. (2n = 56) and Rhamdia quelen (2n = 58) were analyzed by C-banding (CB) and Chromomycin A3 (CMA3) and 4,6-diamidino-2-phenylindole (DAPI) staining, separately and consecutively, in order to understand the role of base-specific fluorochrome treatment after CB. Both species' chromosomes shared common staining profiles as follows. CB with Giemsa (CBG) revealed weak heterochromatic blocks in the telomeric regions of some chromosomes and conspicuous bands on the short arms of one chromosome pair, where nucleolar organizer regions (NORs) were evidenced by silver-staining. Without CB pretreatment, the NORs were stained conspicuously with CMA3, but not with DAPI. The latter uniformly stained all chromosomes, but leaving the NORs pale. Combination of CMA3 or DAPI staining with CB showed distinctive fluorescent blocks in the NOR-bearing short arms of the single chromosome pair along with several bright fluorescent signals on other chromosomes, which were not evidenced by single CMA3 or DAPI staining. These results suggest a modification of chromatin structure by CB treatment, which may increase the stainability of CMA3 and DAPI.  相似文献

Summary The karyotypes of the rainbow trout (Salmo gairdneri R.) and the brown trout (Salmo trutta L.) were analyzed by means of silver staining and the chromomycin A3/distamycin A/DAPI fluorescence banding technique. The nucleolus organizer regions (NORs) were localized at the secondary constrictions of chromosome no. 14 in S. gairdneri and of chromosome no. 10 in S. trutta. Additional silver positive dots were observed at or close to several centromeres in S. gairdneri. Brilliant chromomycin A3 (CMA3) fluorescence heterochromatin blocks were localized on both sides of the nucleolar constrictions in S. gairdneri. A polymorphic CMA3 positive band was detected close to the NORs of S. trutta. No distamycin A/DAPI intense heterochromatin blocks were detected in the genomes of the two Salmo species investigated.  相似文献

Karyotype and cytogenetic markers of Oligosarcus hepsetus from two Brazilian locations in the Paraíba do Sul River Basin (Brazil) were investigated using differential staining techniques (C-banding, silver (Ag)- and chromomycin A3 (CMA3)-staining) and fluorescent in situ hybridization (FISH) using 18 S rDNA and 5 S rDNA probes. The diploid chromosome number was invariably 2n = 50 with 3 pairs of metacentric, 5 pairs of submetacentric, 8 pairs of subtelocentric and 9 pairs of acrocentric chromosomes. No heteromorphic sex chromosomes were observed. The nucleolar organizer regions (NORs) were detected in the short arms of the largest acrocentric pair using Ag-, CMA3- stainings and FISH with 18 S rDNA probe, the latter showing also positive labeling in the short arms of a small acrocentric pair, not visualized by the former methods. FISH with 5 S rDNA probe showed positive labeling in the two chromosome pairs. While the CMA3-staining exhibited GC-rich heterochromatin segments in two pairs of chromosomes, including those coincided with Ag-NORs, the DAPI staining did not reveal any signal, indicating the absence of AT-rich heterochromatin. FISH with an As-51 satellite DNA probe derived from the closely related Astyanax scabripinnis did not reveal any positive signal, demonstrating the absence of this class of DNA in the genome of the specimens under study.  相似文献

The karyotype of European catfish (Silurus glanis L.) was analyzed sequentially by means of silver staining and the chromomycin A3 (CMA3)/distamycin A (DA)/DAPI fluorescence technique and by C-banding, respectively. The nucleolus organizer regions (NORs) were localized on the submetacentric pair No. 14. Brilliant CMA3 fluorescent heterochromatin blocks corresponded to the NORs visualized by silver staining. No DA/DAPI-bright positive fluorescent patterns were detected while C-banding led to the detection of specific banding patterns on several chromosome pairs.—Using these banding data, the karyotype of S. glanis was redescribed.  相似文献

The chromosomes of the diploid and tetraploid loach Misgurnus anguillicaudatus were analyzed by staining with Ag, chromomycin A3 (CMA3)/distamycin A (DA), and DA/4′,6-diamidino-2-phenylindole (DAPI), and using fluorescence in situ hybridization (FISH) with 5.8S + 28S rDNA as a probe. Nucleolus organizer regions (NORs) were mapped to the telomeric region of the short arms of the largest (first) metacentric chromosome pair in the diploid loach with 2n = 50 and the homologous quartet in the tetraploid loach with 4n = 100. The NORs were positive at the same region of the first metacentric chromosome for Ag and CMA3/DA stainings, but negative for DA/DAPI staining. Four signals at the homologs within the same quartet suggest the duplication of the entire genome from diploid to tetraploid status. However, a size difference was detected between the rDNA signals by FISH and CMA3 banding.  相似文献

Karyotype and cytogenetic characteristics of European smelt Osmerus eperlanus were investigated using different staining techniques (sequential Ag-, CMA3 and DAPI banding) and PRINS to detect 5S rDNA and telomeric sites. The diploid chromosome number was invariably 2n = 56 and karyotype composed of 5 pairs of metacentrics, 9 pairs of subtelocentrics and 14 pairs of subtelo- to acrocentrics. The DAPI-positive heterochromatic regions were found in centromeric positions on bi-armed chromosomes and few acrocentrics. Additionally, some interstitial DAPI-positive bands were identified on three pairs of submetacentric chromosomes. The nucleolar organizer regions (NORs) were detected in the short (p) arms of the largest metacentric pair of chromosomes No. 1. Sequential banding (Giemsa-, AgNO3 and CMA3 stainings) revealed NOR sites corresponding to achromatic regions but not associated with CMA3-positive blocks of heterochromatin located on either side of NORs. Individuals from the analyzed population had this conspicuous pair of chromosomes always in heterozygous combination. A complex inversion system was hypothesized to be involved in the origin of the observed variation but analysis with telomeric PRINS and PNA-FISH did not reveal any Interstitial Telomeric Sites (ITS). Hybridization signals were confined exclusively to terminal chromosomal regions. The 5S ribosomal sites as revealed by PRINS were found to be invariably located in the short (p) arms of four pairs of subtelocentric chromosomes. Cytotaxonomic comparisons of the present results with the voluminous available cytogenetic data-set from salmoniform and esociformes fishes appear to support the recent view, based on robust molecular-based phylogeny, that salmoniform and osmeriform fishes are not as closely related as previously assumed.  相似文献

An effect of C-band pattern and polymorphism on chiasma distribution in pollen meiosis was recently demonstrated inScilla siberica. A further meiotic banding study has been performed in the alliesS. amoena, S. ingridae, andS. mischtschenkoana in order to analyze the effect, if any, of their specific C-band patterns and cytochemically different heterochromatin types on recombination. No clear evidence for a preferential formation of chiasmata adjacent to homozygous intercalary heterochromatin and no consistent reduction of chiasma frequency near strongly heterozygous intercalary heterochromatin blocks, as observed inS. siberica, could be found. Terminal C-band heteromorphism is suspected to cause distal chiasma defaults. The results suggest once more that there is no uniform effect of heterochromatin on crossover distribution.  相似文献

Gymnotiformes are an important component of the Neotropical ichthyofauna and they are known for their ability to generate and detect electrical discharges. Phylogenetic relationships of Gymnotiformes are still not well understood. However, the monophyly of the superfamily Rhamphichthyoidea is well accepted, despite the position of tribe Steatogenini (Steatogenys, Hypopygus and Stegostenopos) within this superfamily is unclear. The genus Steatogenys includes three species that, together with Hypopygus and Stegostenopos, form tribe Steatogenini. Cytogenetic information is currently only available for Hypopygus lepturus. Here, we describe the karyotypes of Steatogenys elegans from four localities and S. duidae from two localities. S. elegans was found to have 2n = 50, ZZ/ZW (12m-sm/38st-a), while S. duidae had 2n = 50 (50m-sm). In S. elegans, constitutive heterochromatin (CH) was observed in the centromeric regions of all chromosomes, in the interstitial region of 1q, and in two blocks of Wq. In S. duidae, CH was observed in the centromeric and pericentromeric regions of all chromosomes, and in the interstitial regions of 2q, 3q, 5q, and 7q. Nucleolar organizer regions (NORs) were identified in the distal regions of one chromosome pair in each species. The CMA3 fluorochrome (specific to G-C rich regions) coincided with the NORs in both species, and with the HC of S. elegans except on chromosome pair 5 and the W. The DAPI fluorochrome (specific to A-T rich regions) coincided with the CH of both species, and was very intense for chromosome pair 5 and the W of S. elegans. Our observations suggest that the ZZ/ZW system observed in S. elegans likely evolved through CH addition followed by a paracentric inversion. The chromosomal data described herein are consistent with the phylogenetic hypothesis that tribe Steatogenini should be positioned within family Ramphychthyidae.  相似文献

In spite of various cytogenetic works on suborder Heteroptera, the chromosome organization, function and its evolution in this group is far from being fully understood. Cytologically, the family Rhyparochromidae constitutes a heterogeneous group differing in chromosome numbers. This family possesses XY sex mechanism in the majority of the species with few exceptions. In the present work, multiple banding techniques viz., C-banding, base-specific fluorochromes (DAPI/CMA3) and silver nitrate staining have been used to cytologically characterize the chromosomes of the seed plant pest Elasmolomus (Aphanus) sordidus Fabricius, 1787 having 2n=12=8A+2m+XY. One pair of the autosomes was large while three others were of almost equal size. At diplotene, C-banding technique revealed, that three autosomal bivalents show terminal constitutive heterochromatic bands while one medium sized bivalent was euchromatic. Microchromosomes (m-chromosomes) were positively heteropycnotic. After DAPI and CMA3 staining, all the autosomal bivalents showed equal fluorescence, except CMA3 positive signals, observed at both telomeric heterochromatic regions of one medium sized autosomal bivalent. Silver nitrate staining further revealed that this chromosome pair carries Nucleolar Organizer Regions (NORs) at the location of CMA3 positive signals. The X chromosome showed a thick C-band, positive to both DAPI /CMA3 while Y, otherwise C-negative, was weakly positive to DAPI and negative to CMA3, m-chromosomes were DAPI bright and CMA3 dull.  相似文献

Chromosomal polymorphism regarding number of NOR sites in the cyprinid fish Chondrostoma lusitanicum was examined using C-banding, silver-staining (Ag), and fluorescent staining with chromomycin A3 (CMA3). The analysis of heterochromatic regions allowed a more precise identification of the centromeric regions and the proposal of a revised haploid chromosome formula (7M: 15S: 3A). We describe variability in the number of NOR regions per genome, number of active NOR sites per cell, and relative size of individual NORs. Individuals expressed two or four NOR-bearing chromosomes. Polymorphism was detected in all the populations studied and sex-related differences were not found. The observed chromosomal NOR phenotypes suggest the occurrence of structural rearrangements during the evolutionary process of this diploid leuciscine cyprinid.  相似文献

Using fluorescence in situ hybridization (FISH), chromomycin (CMA3) staining and silver staining, we studied the nucleolar organizer regions in the spiny lobster Palinurus elephas in order to extend our knowledge on the karyology of this commercially important species. Multiple NORs have been detected by FISH, and CMA3 showed a good correspondence between the localization of GC-rich heterochromatin and the ribosomal genes mapped by FISH. In contrast, the number of Ag-positive regions was higher than the number of FISH and CMA3 signals, which may be explained by silver staining of the kinetochores. A variability in the number of FISH and CMA3 signals has been detected in metaphases I and II which is probably due to the occurrence of rDNA cistrons on B chromosomes.  相似文献

Quantified C-band karyograms are presented for the related speciesScilla siberica, S. mordakiae, S. ingridae, S. amoena, andS. mischtschenkoana. Chromosome structure, banding style, and heterochromatin characters suggest a systematic grouping of two more closely related species pairs:S. siberica andS. mordakiae, S. ingridae andS. amoena; they are part of a larger aggregate, well separated fromS. mischtschenkoana. Four different heterochromatin fractions can be recognized inS. siberica and its relatives, but only two inS. mischtschenkoana. C-bands do not replace, but they are added to euchromatin. The particular origin and history of the cultivatedS. amoena and the triploidS. siberica cv. Spring Beauty appear to be responsible for their karyotype constancy, but chromosome conservatism obviously is genuine inS. mischtschenkoana. The chromosomal characteristics of Clarias magur were examined using conventional (Giemsa-staining, Ag-impregnation and CMA3 + DAPI fluorescence) and molecular/ FISH (18S & 5S rDNA probes + one BAC DNA probe) cytogenetic tools. The diploid chromosome number was 50 and the karyotype consisted of 14 metacentric, 20 sub-metacentric, 8 sub-telocentric, 8 acrocentric chromosomes with 84 chromosome arms without any heteromorphic pair. The C-heterochromatic blocks were located on centromeric position of 13 pairs of chromosomes. The NOR sites, visualized by AgNO3- and CMA3- staining, were situated at p arms of chromosome pair No. 21, which also corresponded to 18S rDNA site visualized by FISH. The FISH signal of ICF_001_D19 clone probe was observed on 18th chromosome pair. The findings of the present study on C. magur provided valuable markers for the chromosome identification and locations of genes of the BAC clone on the chromosome will lead to the construction of physical map of genome of this species.  相似文献

We studied cacti species of the subfamilies Pereskioideae (five species of the southern clade) and both species of Maihuenioideae using molecular cytogenetic techniques and DNA content. Mitotic chromosomes were analyzed for Pereskia aculeata, P. bahiensis, P. grandifolia, P. nemorosa, P. sacharosa, Maihuenia poeppigii, and M. patagonica, using the Feulgen stain, CMA/DAPI fluorescent chromosome banding, fluorescence in situ hybridization (FISH, probes of 5S rDNA and pTa71 for 18-5.8-26S rDNA), and DNA content by flow cytometry technique. The karyotypes were highly symmetrical, most of the pairs being metacentric (m). CMA/DAPI banding revealed the presence of CMA+/DAPI? bands associated with NORs in the first m pair of all species. The co-localization of 18-5.8-26S rDNA loci with CMA+/DAPI?/NORs blocks allowed the identification of homeologous chromosome pairs between species of both subfamilies. FISH using probe 5S rDNA was applied for the first time in both subfamilies. Diploid species had always one m pair carrying 5S rDNA genes, with pericentromeric location in different chromosome pairs. In the tetraploid cytotype of M. patagonica, the 5S rDNA probe hybridized to two pairs. The 2C DNA content obtained by FC varied twofold (from 1.85 to 2.52 pg), with significant differences between species. Mean chromosome length, karyotype formula, percentage of heterochromatin position of 5S rDNA locus, and nuclear Cx DNA content vary among Maihuenia and Pereskia species and allowed to differentiate them. Both genera are closely related and that the differences found are not strong enough to separate Maihuenioideae from Pereskioideae.  相似文献

A comparative chromosomal evaluation was carried out between Vigna unguiculata (cowpea) and V. radiata (mung bean) with chromomycin A3 (CMA3)/4’,6-diamidino-2-phenylindole (DAPI) banding and fluorescent in situ hybridization (FISH) using 5S/45S ribosomal DNA (rDNA) probes. Both species had symmetric karyotypes (2n = 22), with prevalence of centromeres in chromosomes at median (m) and submedian (sm) regions and chromosomes ranging in size from 2.1 to 1.25 μm (V. unguiculata) and 2.18 to 0.93 μm (V. radiata). Three different banding patterns were identified for V. unguiculata: CMA3+/DAPI0, CMA3++/DAPI, and CMA3+/DAPI. The CMA3+/DAPI0 bands were observed in the pericentromeric regions of all chromosomes, while the CMA3++/DAPI and CMA3+/DAPI bands were co-localized with the 45S rDNA in the subtelomeric position (chromosomes B, G, and D, J, respectively) and in the proximal position in chromosome F. Two pairs of chromosomes (D and I) bearing interstitial 5S rDNA have been also identified. Vigna radiata displayed CMA30/DAPI+ bands distributed in the centromeric region of chromosomes B, C, and F, while CMA3++/DAPI bands were co-localized with the 45S rDNA sites in the subtelomeric position of the short arm in the F and K chromosome pairs. Three pairs of 5S rDNA sites were identified, the first in the proximal region of the long arm in chromosome E and the two others in the proximal and subterminal positions in the long arm of chromosome J. These data highlight some divergences regarding the amount and composition of the heterochromatin in both species, allowing the identification of individual chromosomes in V. unguiculata and V. radiata, and a comparison with other members of the Phaseoloid clade.  相似文献

The chromosomes of the European Percidae (Lucioperca lucioperca L., Gymnocephalus cernuus L., Gymnocephalus schraetser L. and Perca fluviatilis L.) were analyzed by means of silver staining chromomycin A3/distamycin A/DAPI and DAPI/actinomycin D fluorescence banding techniques. The nucleolus organizer regions (NORs) were localized at the satellite stalks of chromosome no. 16 in Lucioperca lucioperca and Perca fluviatilis, and of chromosome no. 18 in both Gymnocephalus species. When surveying the karyotype diversity of European loaches of the genus Cobitis to identify species involved in hybrid polyploid complexes, an extensive polymorphism in number and location of NORs was discovered in C. vardarensis using Ag-staining, C-banding, CMA3-fluorescence and fluorescence in situ hybridization (FISH). This species had 2n=50, the karyotype contained 13 pairs of metacentric, 10 pairs of submetacentric and two pairs of subtelocentric chromosomes. The NOR-bearing chromosomes included one medium-sized metacentric pair with a large CMA3-positive heterochromatic pericentromeric block, one small metacentric as well as one large submetacentric pairs. Ribosomal sites were always located in telomeres of these chromosomes. Each of the pair of NOR-bearing chromosomes occurred in three variants – (1) presence and/or (2) absence of NORs on both homologues and (3) heterozygous combination where only one of the homologues bears NORs. Altogether, 10 different NOR cytotypes from 27 theoretically possible ones were discovered among 20 indviduals examined. The number of NORs ranged from two to five per specimen. The results regarding the number and locations of NORs as revealed by banding techniques were confirmed using FISH with rDNA probe. NOR sites were of CMA3-positive, suggesting that ribosomal sites are associated with GC-rich DNA. Very similar structural polymorphism with multiple NORs is expressed in the Danubian loach C. elongatoides indicating a close relationship between both species.  相似文献

Some aspects of both the nucleolar organizer activity and meiosis were studied in the testes of Triatoma vitticeps (Heteroptera, Triatominae). The techniques used included squashing followed by lacto-acetic orcein staining, silver-ion impregnation, fluorescent banding (CMA3, Quinacrine mustard and DAPI) and fluorescent in situ hybridization (FISH). A close relationship between heterochromatin and nucleolus in testicular cells was observed. During meiosis, the silver-ion impregnation pattern varied. At metaphase plate, a small body appeared apart from the chromosomes. In the spermatids this small body was seen in preparations stained with orcein and silver- ion impregnation but not with fluorochromes or FISH. These characteristics combined suggest that these corpuscles represent a source of ribonucleoproteins (RNP) – RNA and specific nucleolar proteins. Silver-ion impregnation and (FISH) revealed nucleolar organizer activity in two metaphase sex chromosomes (X). These results indicate that, in these species, nucleolar organizer regions (NORs) are located in the sex chromosomes, X chromosomes were CMA3+ and Y chromosome was DAPI+.  相似文献

The chromosomal localizations of repetitive DNA clusters (ribosomal DNA and centromere satellites) were analyzed by fluorescent in situ hybridization in five strains of Arabidopsis halleri ssp. gemmifera. All five A. gemmifera strains have three chromosome pairs with 45S (5.8S-16S-26S) rDNA loci, and one pair with both 5S and 45S rDNA loci. These localizations are different from that of A. thaliana. Very unusually, there are three families of centromeric satellite DNAs (pAa, pAge1, and pAge2), and they showed polymorphism among the five strains studied. Overall, we found four different centromere satellite compositions. A plant from Fumuro was heterozygous for the chromosome specificities of centromere satellite families, possibly due to a reciprocal translocation involving centromere regions. Changes of centromeric satellite repeats appear to be rapid and frequent events in the history of A. gemmifera, and seem to occur by exchanging clusters as units.  相似文献
