Three field experiments were conducted in order to evaluate the effect of application of the plant growth regulator chlormequat on growth and yield of the cereal triticale. Application of chlormequat resulted in shorter triticale plants as a result of restricted elongation of the upper internodes of the stem. These shorter plants produced a lower weight of straw and were more resistant to lodging. In two experiments chlormequat application led to no statistically significant increase in triticale grain yield; in a third yield was increased. Chlormequat consistently increased the number of grains per ear but this was offset to some extent by a lower mean grain weight. Yield was closely correlated with the number of grains per m2. Leaf senescence was delayed by chlormequat. The results are consistent with the hypothesis that chlormequat can increase yield potential.  相似文献

The addition of the organophosphorous plant growth regulator Melaphen (4 × 10?12 M) to the incubation medium increases the maximum rate of oxidation of NAD-dependent substrates in rat liver and sugar beet root mitochondria. In addition, Melaphen stimulates electron transport during oxidation of succinate by rat liver mitochondria, but has no effect on the rate of this substrate oxidation in sugar beet root mitochondria. In storage organs of plants, the rate of oxidation of NAD-dependent substrates by mitochondria is relatively low. By stimulating the activity of NAD-dependent dehydrogenases, Melaphen stimulates energy metabolism in the cells and manifests adaptogenic activity by accelerating the germination of seeds. Melaphen does not influence the fluorescence of lipid peroxidation (LPO) products in mitochondria non-exposed to stress, but decreases 1.5–2 fold the LPO fluorescence in rat liver mitochondria exposed to cold stress and artificially “aged” sugar beet root mitochondria. Besides, Melaphen increases the rate of electron transport in a terminal site of respiratory chains of plant and animal mitochondria and decreases LPO. The data obtained testify to antistress activity of Melaphen.  相似文献

Early foliar spray treatments containing gibberellic acid(GA3) significantly lower the concentration ofthe bitter flavonoid naringin in fruit tissues. Sprayscontaining a surfactant and different levels ofGA3 (5, 50, 100, and 500 ppm) or abscisic acid(ABA) (5, 25, and 50 ppm) were applied to young,developing fruit on mature grapefruit (Citrusparadisi) trees during the period from April to June,beginning just after fruit set. The fruit were allowedto mature and were harvested early the following year.Harvested fruit were evaluated for weight, juicecharacteristics, and flavonoid concentrations.GA3 application resulted in larger mature fruit,which yielded juice with the same soluble solids valueas juice from control fruit, but with slightly loweracid percentages and lower concentrations of naringin.ABA treatment had little effect on juice solublesolids, acid content and naringin content except atthe highest concentration of 50 ppm, which lowerednaringin levels slightly in juice.  相似文献

Osmotic priming of carrot seeds for 2 wk in polyethylene glycol (PEG, — 10 MPa) at 15 °C led to more rapid and synchronous germination at 20 °C compared to untreated seeds. These responses were enhanced by a 24 h pre-priming soak in water or a change of solution after the first 24 h of priming to remove leachate. The inclusion of 200 mg litre-1N-substituted phthalimide in the pre-priming soak and/or in the PEG further enhanced the results of priming. Leachate removal combined with phthalimide inclusion gave 79% and 86% germination from seeds of two carrot cultivars during the first day in 20°C water following priming. In contrast, cumulative germination of untreated seeds of the same cultivars was 18% and 61% respectively after 3 days in 20°C water. Seeds primed in PEG containing 200 mg phthalimide litre-1with the solution replaced after the first 24 h germinated earlier and more synchronously than untreated seeds over a range of germination temperatures (5, 10, 15, or 20°C), but the effects of priming were most marked at 5°C.  相似文献

Nodal stem segments of flowering dogwood (Cornus florida L.) were cultured on media containing seven different sources of macronutrients including full- and half-strength Murashige & Skoog (MS) macrosalts, N6, Anderson's (AND), Quoirin & Lepoivre's (LP), Nitsch & Nitsch (NIT), and Woody Plant Medium (WPM). All media contained MS micronutrients, Staba vitamins, 20 g l-1 sucrose, and 6.5 g l-1 Difco Bacto-agar, and were supplemented with 2.2 M 6-benzyladenine (BA) and 0.49 M indole-3-butyric acid (IBA). Following three subcultures, the best shoot proliferation was supported on media containing WPM macronutrients. To optimize the proliferation rate, shoots were cultured on WPM macronutrients supplemented with eight combinations of BA and IBA, and 3.3 M BA without IBA was determined to be the best.Abbreviations BA 6-benzyladenine - IBA indole-3-butyric acid  相似文献

By means of weak-bound spin probes--stable iminoxyl free radicals differing in the level of hydrophobity there were studied thermoinduced structural transitions in the membranes of cell organells of animal liver after intraperitoneal injection of antioxidants and in the course of malignant growth, and in the nuclear membranes of Ehrlich ascite carcinoma as well. It has been found that during the growth of Ehrlich ascite carcinoma changes in rotationary mobility of probes in cell nuclei isolated from the liver of tumour-carrying animal are similar to the changes observed after antiocidants injection. A different pattern is observed in tumour cells. The membranes of ascite cell nuclei are characterized by a weak dependence of tau c on temperature for both probes. Within the temperature range studied no characteristic structural transitions proceeding in the nuclei of intact animals are observed.  相似文献

In plants, the stacking of part of the photosynthetic thylakoid membrane generates two main subcompartments: the stacked grana core and unstacked stroma lamellae. However, a third distinct domain, the grana margin, has been postulated but its structural and functional identity remains elusive. Here, an optimized thylakoid fragmentation procedure combined with detailed ultrastructural, biochemical, and functional analyses reveals the distinct composition of grana margins. It is enriched with lipids, cytochrome b6f complex, and ATPase while depleted in photosystems and light‐harvesting complexes. A quantitative method is introduced that is based on Blue Native Polyacrylamide Gel Electrophoresis (BN‐PAGE) and dot immunoblotting for quantifying various photosystem II (PSII) assembly forms in different thylakoid subcompartments. The results indicate that the grana margin functions as a degradation and disassembly zone for photodamaged PSII. In contrast, the stacked grana core region contains fully assembled and functional PSII holocomplexes. The stroma lamellae, finally, contain monomeric PSII as well as a significant fraction of dimeric holocomplexes that identify this membrane area as the PSII repair zone. This structural organization and the heterogeneous PSII distribution support the idea that the stacking of thylakoid membranes leads to a division of labor that establishes distinct membrane areas with specific functions.  相似文献

Membrane preparations from growing regions of pea stems and activelydividing mouse L-cells form lipid-linked saccharides from GDP-mannose and UDP-N-acetylglucosamine. These lipids have properties which are consistent with those of mono-and di-phosphoryl polyisoprenyl derivatives. In experiments using plant membranes, the monophosphoryl derivative labeled with GDP-(14C) mannose contains mannose only, while the diphosphoryl derivative labeled with the same nucleotide sugar is heterogeneous, containing oligosaccharides corresponding to mannosaccharides of 5, 7, and 9-12 residues. Only the diphosphoryl polyisoprenyl derivatives are labeled with UDP-(14C)glucosamine and these contain predominantly chitobiose and N-acetylglucosamine itself. Unlabeled GDP-mannose added after UDP-N-acetyl (14C)glucosamine results in the formation of higher lipid-linked oligosaccharides which are apparently the same as those which are labeled with GDP-(14C)mannose alone. Incubation of the membranes with GDP-(14C)mannose in the presence of Mn2+, unlabeled UDP-glucose or unlabeled UDP-N-acetylglucosamine results in marked changes in the accumulation of both the polyisoprenyl monophosphoryl mannose and polyisoprenyl diphosphoryl oligosaccharides. Animal cell membranes synthesise lipid-linked oligosaccharides when incubated with UDP-N-acetylglucosamine and GDP-mannose. These oligosaccharides are similar in size to those synthesised by the plant membranes but their formation is more efficient. The potential roles of these compounds in glycoprotein biosynthesis in both plant and animal tissues is discussed.  相似文献

研究了人工抚育年后油松林的生长状况、群落组织和结构.结果表明,不同人工抚育措施的油松林平均胸径、树高、冠幅均有极显著差异(P<0.01),采取抚育措施的两个样地间的油松胸径和冠幅差异均达到显著水平,其中既打枝又间伐的油松林的胸径(7.8±0.29cm)、树高(5.5±0.09m)和平均冠幅(249±7.24cm)最大,未打枝和间伐的油松林最大;随着人工抚育措施加强,单株油松的平均新叶、老叶和枝的生物量逐渐增加,达到极显著水平(P<0.01);在不同抚育措施下,既打枝又间伐的油松林地上生物量最大(44.0t·hm-2)、未打枝和间伐油松林地上生物量最大(14.9t·hm-2);油松新叶和老叶长度的差异性显著;油松的比叶面积在生长初期(5月和月)和末期(9月)差异性显著;人工抚育措施增加了林下植被的种类数量和盖度.  相似文献

The natural occurrence of jasmonic acid and its methyl ester in plants has been studied using different methods such as GC, GC-MS, HPLC, radioimmunoassay, and bioassay. Jasmonic acid was detected in several Leguminosae plants and a number of species belonging to nine other Angiospermae families. Highest amounts occurred in fruit parts, especially the immature pericarp, but it was found also in flowers and vegetative plant parts, e.g. leaves, stems, and germs. Young apple fruits contain both jasmonic acid and methyl jasmonate, and in Douglas fir, the only Gymnospermae species studied, only the methyl ester could be detected. Jasmonic acid is discussed as an endogenous plant growth regulator widely distributed in higher plants.  相似文献

Fluorescent probes located in different membrane regions were used to evaluate the effects of dimyristoylphosphatidylethanol (DMPEt) on the structural parameters (transbilayer rotational and lateral mobility, annular lipid fluidity and protein distribution) of synaptosomal plasma membrane vesicles (SPMV) from the bovine cerebral cortex. An experimental procedure was used based on selective quenching of 1,6-diphenyl-1,3,5-hexatriene (DPH) and 1,3-di(1-pyrenyl)propane (Py-3-Py) by trinitrophenyl groups, and radiationless energy transfer from the tryptophans of membrane proteins to Py-3-Py. DMPEt increased the bulk lateral and rotational mobility, and annular lipid fluidity of SPMV lipid bilayers, and had a greater fluidizing effect on the outer monolayer than the inner monolayer. It also caused membrane proteins to cluster. These effects of DMPEt on neuronal membranes may be responsible for some, though not all, of the general anesthetic actions of ethanol.  相似文献

Summary Turfgrass, like other major crop species, is recalcitrant to manipulation in vitro. To perform efficient genetic transformation of turfgrass, it is necessary to optimize tissue culture conditions. In most reports, plant tissue culture techniques have been applied to propagate a single cultivar or several cultivars in one species of turfgrass. In this experiment, four turfgrass genera were used, namely common bermudagrass, Cynodon dactylon [L.] Pers. (California origin); red fescue, Festuca rubra L. var. rubra ‘Shadow’; perennial ryegrass, Lolium perenne L. ‘Barbal’; and Kentucky bluegrass, Poa pratensis L. ‘Merion.’ Mature seeds were surface-sterilized and cultured on basal Murashige and Skoog (MS) media supplemented with 30–250 μM 2,4-dichlorophenoxyacetic acid (2,4-D) for callus induction. Regeneration media consisted of MS supplemented with 5–10 μM 6-benzyladenine (BA). Among the genera, Poa had the higest callus induction percentage (CIP) regardless of 2,4-D concentration, followed by Cynodon, Lolium, and Festuca, respectively. Cynodon and Lolium had the highest callus regeneration percentage (CRP) and overall regeneration rate (ORR). Festuca had a poor CIP, CRP, and ORR compared to other studied genera. Cynodon produced the highest shoot number per explant. Based on the results of the present study, MS medium supplemented with 60 μM 2,4-D (for callus induction) and 7.5 μM BA (for regeneration) can be used in multi-generic transformation studies with the genera used.  相似文献

The organophosphorus insecticide Selecron [O-(4-bromo-2-chlorophenyl) O-ethyl S-n-propyl-phosphorotioate] at 10 and 50 ppm significantly decreased respiration, mycelial protein, extracellular protein and mycelial dry weight of Aspergillus fumigatus, A. terreus and Myceliophthora thermophila when grown at 45°C. Like the outer membranes of liver mitochondria, those of plant mitochondria are impermeable to cytochrome c when intact and can be ruptured by osmotic shock. Isolated plant outer mitochondrial membranes are also similar to the corresponding liver membranes in terms of phospholipid and sterol content. Sodium dodecyl sulfate-polyacrylamide gradient gel electrophoresis experiments indicate that a single class of proteins (apparent molecular weight 30 000) comprises the bulk of the plant outer membrane protein. There are also considerable amounts of polysaccharide associated with these membranes, which may contribute to their osmotic stability.  相似文献
