Soil salinity affects the growth and yield of crops. The stress of soil salinity on plants can be mitigated by inoculation of plant growth promoting bacteria (PGPR). The influence of PGPR inoculation on wheat (Triticum aestivum L.) crop productivity under salinity stress has not been properly addressed so far. Therefore, the present study was conducted to investigate the effects of various PGPR strains (W14, W10 and 6K; alone and combined) at several growth attributes of wheat plant under different soil salinity gradients (3, 6 and 9 dS m-1). The growth attributes of wheat (height, roots, shoots, spikes, grains quality, biological and economical yield, nutrients nitrogen, phosphorus and potassium in grains) were highly affected by salinity and decreased with increasing salinity level. The PGPR inoculation substantially promoted growth attributes of wheat and prominent results were observed in W14 × W10 × 6K treatment at all salinity levels. The results suggest that inoculation of PGPR is a potential strategy to mitigate salinity stress for improving wheat growth and yield.  相似文献

Bacterial isolates with the ability to tolerate salinity and plant growth-promoting features were isolated from the saline areas of Gujarat, India, that is, Bhavnagar and Khambat. A total of 176 strains of rhizobacteria were isolated out of which 62 bacterial strains were able to tolerate 1 M NaCl. These were then further studied for their potential plant growth-promoting rhizobacteria characteristics like phosphate solubilization, siderophore production, and IAA production. Twenty-eight isolates of the 62 strains showed good tricalcium phosphate solubilization in solid medium in the range of 9–22 mm and 15 isolates showed good phosphate solubilization in liquid medium in the range of 9–45 μg/ml. Siderophore production was checked in all 15 isolates, and 13 were screened out that produced the hydroxamate type of siderophore in the range of 11–50 mM. Among the 13 isolates, 10 were able to produce indole acetic acid in the range of 10–26 μg/ml after 72 h of incubation. Pot trials were carried out on chickpea under 300 mM NaCl stress using the best five isolates. Plants inoculated with MSC1 or MSC4 isolates showed an increase in the parameters that evaluate plant growth when compared to uninoculated controls. Strains MSC1 and MSC4 were identified as Pseudomonas putida and Pseudomonas pseudoalcaligens, respectively, according to sequence analysis of the 16S rRNA gene.  相似文献

The heavy metal-resistant bacteria from rhizospheric soils of wild Populus euphratica forest growing in arid and saline area of northwestern China were investigated by cultivation-dependent methods. After screening on medium sparked with zinc, copper, nickel and lead, 146 bacteria strains with different morphology were isolated and most of them were found to be resistant to at least two kinds of heavy metals. Significant increase in fresh weight and leaf surface area of Arabidopsis thaliana seedlings under metal stress were noticed after inoculated with strains especially those having multiple-resistance to heavy metals such as Phyllobacterium sp. strain C65. Investigation on relationship between auxin production and exogenous zinc concentration revealed that Phyllobacterium sp. strain C65 produced auxin, and production decreased as the concentration of zinc in medium increased. For wheat seedlings treated with zinc of 2 mM, zinc contents in roots of inoculated plants decreased by 27% (P < 0.05) compared to the uninoculated control. Meanwhile, zinc accumulation in the above-ground tissues increased by 22% (P < 0.05). The translocation of zinc from root to above-ground tissues induced by Phyllobacterium sp. strain C65 helped host plants extract zinc from contaminated environments more efficiently thus alleviated the growth inhibition caused by heavy metals.  相似文献

The enzyme 1-aminocyclopropane-1-carboxylate deaminase converts ACC, the precursor of the plant hormone ethylene to α-ketobutyrate and ammonium. The enzyme has been identified in few soil bacteria, and is proposed to play a key role in plant growth promotion. In this study, the isolates of plant growth promoting rhizobacteria were screened for ACC deaminase activity based on their ability to grow on ACC as a sole nitrogen source. The selected isolates showed the presence of other plant growth promoting characteristics such as IAA production, phosphate solubilization and siderophore production. The role of ACC deaminase in lowering ethylene production under cadmium stress condition was also studied by measuring in vitro ethylene evolution by wheat seedlings treated with ACC deaminase positive isolates. Nucleic acid hybridization confirmed the presence of ACC deaminase gene (acdS) in the bacterial isolates.  相似文献

It has been well known that the bacteria of the genus Azotobacter, in addition to the beneficial N2-fixing activity, are able to improve plant growth by a number of direct and indirect mechanisms. To identify this potential in indigenous azotobacteria, the efficiency of 17 isolates of Azotobacter from the rhizosphere of wheat and barley plants cultivated in salt- and/or drought-affected soils in Iran were evaluated for their ability to dissolve inorganic and organic phosphates, siderophore secretion, indole acetic acid (IAA) production; and protease, chitinase, and ACC deaminase (ACCD) activities. First, they were biochemically characterized and one isolate (strain) was identified by 16S rDNA sequencing. Eight isolates were designated as Azotobacter vinelandii and the remaining isolates were identified as A. chroococcum. All isolates hydrolyzed the organic and inorganic phosphate compounds and effectively produced IAA. Fifteen isolates produced siderophore, but only one isolate showed protease activity which is being reported for the first time in relation to Azotobacter. None of the 17 isolates was capable of producing ACCD or chitinase. However, polymerase chain reaction amplification of the ACCD coding genes, by the use of the gene-specific primers, indicated that not all contain the ACCD gene. The standard screening methods with slight modifications, especially in the case of ACCD assay, were applied. The results showed that the use of specific screening methods, modified according to bacterial nutritional requirements, are the efficient methods for precise evaluation of the plant growth promoting rhizobacteria activity.  相似文献

To study the effect of plant species on the abundance and diversity of bacterial antagonists, the abundance, the phenotypic diversity, and the genotypic diversity of rhizobacteria isolated from potato, oilseed rape, and strawberry and from bulk soil which showed antagonistic activity towards the soilborne pathogen Verticillium dahliae Kleb. were analyzed. Rhizosphere and soil samples were taken five times over two growing seasons in 1998 and 1999 from a randomized field trial. Bacterial isolates were obtained after plating on R2A (Difco, Detroit, Mich.) or enrichment in microtiter plates containing high-molecular-weight substrates followed by plating on R2A. A total of 5,854 bacteria isolated from the rhizosphere of strawberry, potato, or oilseed rape or bulk soil from fallow were screened by dual testing for in vitro antagonism towards Verticillium. The proportion of isolates with antagonistic activity was highest for strawberry rhizosphere (9.5%), followed by oilseed rape (6.3%), potato (3.7%), and soil (3.3%). The 331 Verticillium antagonists were identified by their fatty acid methyl ester profiles. They were characterized by testing their in vitro antagonism against other pathogenic fungi; their glucanolytic, chitinolytic, and proteolytic activities; and their BOX-PCR fingerprints. The abundance and composition of Verticillium antagonists was plant species dependent. A rather high proportion of antagonists from the strawberry rhizosphere was identified as Pseudomonas putida B (69%), while antagonists belonging to the Enterobacteriaceae (Serratia spp., Pantoea agglomerans) were mainly isolated from the rhizosphere of oilseed rape. For P. putida A and B plant-specific genotypes were observed, suggesting that these bacteria were specifically enriched in each rhizosphere.  相似文献

从辐射污染区采集42份土样, 分别采用6种分离培养基进行放线菌的分离, 共获得152株放线菌。经形态、核糖体DNA扩增片段限制性内切酶(Amplifed ribosomal DNA restriction analysis, ARDRA)分析比较, 选取其中的60株进行16S rRNA基因测序。通过序列比对、聚类分析, 60株菌分布在放线菌纲中的12个属, 链霉菌属占大多数, 有大量稀有放线菌, 这表明辐射污染区具有较丰富的放线菌属种多样性。  相似文献

Abstract

Phosphate solubilizing bacteria (PSB) can convert insoluble form of phosphorous (P) to an available form which is a major concern in Indian agriculture. In this study, 21 isolates having phosphate solubilizing capability were isolated from different regions of Lucknow, India. Among all, six efficient PSB were confirmed by using in vitro P estimation and 16S rRNA universal primers. The similarity detection was done using random amplified polymorphic DNA (RAPD) finger printing for genotyping the PSB isolates and to determine genetic relatedness between them. Twenty different OPA primers were tested among which four primers produced prominent, highly reproducible, and polymorphic bands. An average of 10.5 polymorphic bands per primer with the amplified DNA fragments ranging from 200 to 2000?bp in size. A dendrogram constructed from these data indicated 25–76% homology. Highest similarity was found in between Bacillus anthracis and Bacillus cereus with 33.8% similarity while least dissimilarity was found in B. anthracis and Pseudomonas fragi with 12% of similarity. These findings provide that there is a great genetic diversity between bacterial isolates from different geographical regions and RAPD can be used as a specific, time consuming and also proves as a reliable molecular tool which helps in strain level discrimination.  相似文献

Bioleaching of potassium from feldspar was optimized for five fungal and two bacterial isolates for carbon, nitrogen, agitation, pH and incubation time. Fungal isolates SDSR and SDS3 and bacterial isolate SDS1 released 34, 42 and 28 ppm K in sucrose-amended medium, respectively. Isolate SDS3 took 10 days for the highest K solubilization, whereas SDSR required 15 days under 150 rpm shaking condition. The pH of medium was found to decrease from 7.0 to 2.05 after the growth of the organisms. High-performance liquid chromatography analysis of the filtrate showed the presence of citric, tartaric and maleic acid, which could be responsible for the solubilization of potassium.  相似文献

405份CIMMYT引进小麦种质的遗传多样性分析   总被引:3,自引:1,他引:3  
为了明确国际玉米改良中心(CIMMYT)引进普通小麦种质材料的遗传多样性特点,为其利用提供参考依据,本研究从均匀分布于小麦基因组的420对SSR引物中选择出条带清晰、多态性较好的62对引物对引自CIMMYT的405份普通小麦种质系进行遗传多样性检测。结果表明,62对SSR引物在405份CIMMYT材料中共检测到198个等位变异,每对引物检测到等位变异的数目为2~8个,平均每对SSR引物能够检测到3.19个等位变异。单个SSR引物的PIC值介于0.03~0.79之间,平均值0.48。405份CIMMYT材料A、B、D基因组之间多态性位点数和等位变异数相差不大,PIC平均值B基因组(0.53)A基因组(0.52)D基因组(0.39)。聚类分析结果显示,62对SSR引物能够将405份CIMMYT材料区分开来,在0.1285遗传距离处将供试材料分为24个类群,类型较为丰富,不同类群的材料在农艺性状和品质性状上存在差异。  相似文献

In order to explore salt tolerant plant growth promoting rhizobacteria (PGPR), 121 isolates were recovered from Tsunami affected area of Andaman and Nicobar Islands, India, out of which 23 were grown at 10% NaCl. Out of total isolates grown at 10% NaCl concentration, 14 solubilized phosphate, 13 showed siderophore production, five showed Indole acetic acid (IAA) production and 16 produced one or more extra cellular enzymes. Isolates SJ10 and CR5 showed the highest enzyme production and PGP properties and five isolates showed antagonistic activity against Sclerotium rolfsii. Screening of potential isolates for nitrogen fixing gene (nifH) showed only three isolates (MM3, MM6 and MRC11) possessed nifH gene. Salt tolerant Bacillus PGPR could be used as a bio-inoculant to enhance the crop growth under Tsunami affected soils.  相似文献

Two Gram-negative, non-motile, short-rod-shaped bacterial isolates, designated 110399T and 110248, were isolated from an oil-polluted saline soil in Shengli Oilfield, Eastern China. The two strains shared 99.9 % 16S rRNA gene sequence similarity with the DNA–DNA relatedness value being 80.0 %. They were both capable to grow at 20–40 °C, pH 7–9, and 1–9 % (w/v) NaCl with the optimum growth happened at 30 °C, pH 8, and 2–6 % (w/v) NaCl. The phylogenetic analysis based on 16S rRNA gene sequences revealed that the two strains were members of Nitratireductor and most closely related to Nitratireductor pacificus pht-3BT and N. basaltis J3T with the 16S rRNA gene sequence similarities being 97.1 and 97.0 %. The DNA–DNA relatedness between the novel strains and two type strains were below 27 ± 7 %. The strains 110399T and 110248 also differed from N. pacificus and N. basaltis in nitrate reduction, salt tolerance, enzyme activities, and utilization of carbon sources. The major cellular fatty acids of strain 110399T were C19:0ω8c cyclo (10.5 %) and Summed Feature 8 (C18:1ω7c and/or C18:1ω6c, 41.5 %) which are typical in the genus Nitratireductor. The predominant ubiquinone was Q-10. The genome DNA G+C content of strain 110399T and 110248 was 61.1 and 61.7 mol%. On the basis of genetic, phenotypic, and chemotaxonomic analyses, strains 110399T and 110248 represent a novel species within the genus Nitratireductor, for which the name Nitratireductor shengliensis sp. nov. is proposed. 研究了22株代表性斜茎黄芪根瘤菌的谷氨酰胺合成酶基因多样性。首先对供试菌株进行了谷氨酰胺合成酶glnA和glnII基因扩增,结果显示从来自Mesorhizobium和Rhizobium属的多数代表菌株都可以扩增到约1kp、大小一致的glnA基因产物,而从Agrobacterium sp.的4株代表菌株未能得到glnA PCR扩增产物。基因glnII的扩增结果显示几乎从所有测试菌株都能够得到基因产物,来自Mesorhizobium septentrionale、M.temperatum和Mesorhizobium spp.的代表菌株都得到了单一的、大小约400bp~500bp的glnII PCR扩增产物,而从Agrobacterium sp.的4株代表菌株扩增得到的glnII PCR扩增产物明显不同于其它斜茎黄芪根瘤菌代表菌,它们都有一条约1 kb的特征PCR扩增产物条带,SDW052和R084还出现了另外2~3个扩增产物条带。此外,基因glnA的RFLP分析结果与我们先前的16S rRNA基因分析结果具有很好的一致性,这些结果都进一步证实了这些根瘤菌的染色体基因多样性。  相似文献

西南冬麦区地方品种HMW-GS组成遗传多样性研究   总被引:2,自引:0,他引:2  
采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)对西南冬麦区(云南、贵州、四川)3个省份共计560份小麦地方品种的高分子量谷蛋白亚基(HMW-GS)组成进行了研究。结果表明:Glu-1位点共有22种等位基因,其中Glu-A1位点4种、Glu-B1位点11种、Glu-D1位点7种;亚基null、7 8和2 12在各自位点的频率最高,分别为89.64%、68.21%和96.43%。亚基组成类型共有46种,以null/7 8/2 12和null/7 9/2 12为主,频率分别为50.89%和11.79%。在这些材料中筛选出一些含有1、2*、17 18、14 15、5 10等优质亚基的材料,其中有52份材料含有优质亚基组合。  相似文献
