To analyze the physiological mechanisms underlying the increased tolerance to drought and high temperature stress combination by overproduction of glycinebetaine (GB) in wheat, a transgenic wheat line T6 and its wild-type (WT) Shi4185 were used. The transgenic line was generated by introducing a gene encoding betaine aldehyde dehydrogenase (BADH) into a wheat line Shi4185. The gene was cloned from Garden Orache (Atriplex hortensis L.). Wheat plants were exposed to drought (withholding irrigation), high temperature stress (40 °C), and their combination at the flowering stage. Analyses of oxygen-evolving activity and photosystem II (PSII) photochemistry, modulated chlorophyll fluorescence, rapid fluorescence induction kinetics, and the polyphasic fluorescence transients (OJIP) were used to evaluate PSII photochemistry in wheat plants. The results suggest that the PSII in transgenic plants showed higher resistance than that in wild-type plants under the stresses studied here, this increased tolerance was associated with an improvement in stability of the oxygen-evolving complex and the reaction center of PSII; streptomycin treatment can impair the protective effect of overaccumulated GB on PSII. The overaccumulated GB may protect the PSII complex from damage through accelerating D1 protein turnover to alleviate photodamage. The results also suggest that the PSII under combined high temperature and drought stress shows higher tolerance than under high temperature stress alone in both transgenic and wild-type plants.  相似文献

A betA gene encoding choline dehydrogenase from Escherichia coli was transformed into cotton (Gossypium hirsutum L.) via Agrobacterium-mediated transformation. Transgenic cotton plants exhibited improved tolerance to chilling due to accumulation of glycinebetaine (GB). The results of our experiment showed that GB contents of leaves of transgenic lines 1, 3, 4, and 5, both before and after chilling stress, were significantly higher than those of wild-type (WT) plants. At 15°C, transgenic lines 1, 3, 4, and 5 exhibited higher germination capacity as determined by the germination speed and final germination percentage and, displayed less inhibition in seedling shoot growth rate than WT plants. Under chilling stress, transgenic lines 4 and 5 maintained higher relative water content, upper carbon dioxide (CO2) fixation capacity and PSII electron transfer rate, better osmotic adjustment (OA), a lower percentage of ion leakage, and less lipid membrane peroxidation when compared with WT plants. Chilling resistance of the transgenic lines was demonstrated to be positively correlated with GB content under chilling stress. The high levels of GB in transgenic cotton plants might not only protect the integrity of cell membrane from chilling damage, but also be involved in OA which alleviated chilling induced water stress. Moreover, under chilling-stressed conditions, transgenic cotton plants enhanced stomatal conductance, PSII electron transport rate, and further leaf photosynthesis through accumulating high levels of GB.  相似文献

Within their natural habitat, crops are often subjected to drought and heat stress, which suppress crop growth and decrease crop production. Causing overaccumulation of glycinebetaine (GB) has been used to enhance the crop yield under stress. Here, we investigated the response of wheat (Triticum aestivum L.) photosynthesis to drought, heat stress and their combination with a transgenic wheat line (T6) overaccumulating GB and its wild-type (WT) Shi4185. Drought stress (DS) was imposed by controlling irrigation until the relative water content (RWC) of the flag leaves decreased to between 78 and 82%. Heat stress (HS) was applied by exposing wheat plants to 40°C for 4 h. A combination of drought and heat stress was applied by subjecting the drought-stressed plants to a heat stress as above. The results indicated that all stresses decreased photosynthesis, but the combination of drought and heat stress exacerbated the negative effects on photosynthesis more than exposure to drought or heat stress alone. Drought stress decreased the transpiration rate (Tr), stomatal conductance (Gs) and intercellular CO2 concentration (Ci), while heat stress increased all of these; the deprivation of water was greater under drought stress than heat stress, but heat stress decreased the antioxidant enzyme activity to a greater extent. Overaccumulated GB could alleviate the decrease of photosynthesis caused by all stresses tested. These suggest that GB induces an increase of osmotic adjustments for drought tolerance, while its improvement of the antioxidative defense system including antioxidative enzymes and antioxidants may be more important for heat tolerance.  相似文献

We investigated the different responses of wheat (Triticum aestivum L.) plants to drought- (DS) and heat stress (HS), and analyzed the physiological mechanisms of glycine betaine (GB) involved in the improvement of wheat tolerance to the combination of these stresses. The transgenic wheat T6 line was generated by introducing a gene encoding betaine aldehyde dehydrogenase (BADH) into the wild-type (WT) Shi4185 line. The gene was cloned from the Garden Orache plant (Atriplex hortensis L.). Wheat seedlings were subjected to drought stress (30%, PEG-6000), heat stress (40°C), and their combination. Photosynthetic gas exchange, water status and lipid peroxidation of wheat leaves were examined under different stresses. When subjected to a combination of drought and heat, the inhibition of photosynthesis was significantly increased compared to that under DS or HS alone. The increased inhibition of photosynthesis by the combined stresses was not simply the additive stress effect of separate heat- and drought treatments; different responses in plant physiology to DS and HS were also found. HS decreased the chlorophyll (Chl) content, net photosynthetic rate (P N), carboxylation efficiency (CE) and apparent quantum yield (AQY) more than DS but DS decreased the transpiration rate (E), stomata conductance (g s) and intercellular CO2 concentration (C i) more than HS. GB over-accumulation led to increased photosynthesis not only under individual DS or HS but also under their combination. The enhancement of antioxidant activity and the improvement of water status may be the mechanisms underlying the improvement of photosynthesis by GB in wheat plants.  相似文献

Effects of foliar application of 100 mmol/L glycinebetaine (GB) on PS II photochemistry in wheat (Triticum aestivum) flag leaves under drought stress combined with high irradiance were investigated. The results show that GB-treated plants maintained a higher net photosynthetic rate during drought stress than non-GB treated plants. Exogenous GB can preserve the photochemical activity of PSII, for GB-treated plants maintain higher maximal photochemistry efficiency of PSII (F(v)/F(m)) and recover more rapidly from photoinhibition. In addition, GB-treated plants can maintain higher anti-oxidative enzyme activities and suffer less oxidative stress. Our data suggest that GB may protect the PSII complex from damage through accelerating D1 protein turnover and maintaining anti-oxidative enzyme activities at higher level to alleviate photodamage. Diethyldithiocarbamate as well as streptomycin treatment can impair the protective effect of GB on PSII. In summary, GB can enhance the photoinhibition tolerance of PSII.  相似文献

Two tobacco (Nicotiana tabacum L.) cultivars differing in drought tolerance were used to study the effects of foliar-applied glycinebetaine (GB, 80 mM) under well-watered and water-deficit conditions. The latter affected shoot biomass and height, with a more significant decrease observed in drought-sensitive cultivar than in drought-resistant cultivar. Foliar-applied GB was absorbed, accumulated by tobacco leaves and improved growth of plants subjected to water deficit. GB-treated plants maintained leaf water status apparently due to the improved osmotic adjustment. GB application enhanced the photosynthesis in water-deficit experiencing plants, mostly due to a greater stomatal conductance and carboxylation efficiency of CO2 assimilation. photosystem II (PSII) activity in GB-treated plants was higher, as suggested by higher actual efficiency of PSII (ΦPSII). GB increased anti-oxidative enzyme activities under water deficit. All these effects resulted in an improved shoot biomass and height. Therefore, foliar GB application at the rapid growth stage favors plant growth in drought-stressed plants, mainly by improving water status and increasing PSII activity. Glycine betaine (GB) is a compatible solute that accumulates rapidly to enhance heat tolerance in many plants grown under heat stress. In this study, a BADH gene (betaine aldehyde dehydrogenase) from spinach was introduced into tomato (Lycopersicon esculentum cv. ‘Moneymaker’) via Agrobacterium-mediated transformation. Transgenic tomato lines expressing BADH exhibited higher capabilities for GB accumulation. Chlorophyll fluorescence analysis of wild type (WT) and transgenic plants exposed to heat treatment (42 °C) showed that transgenic plants exhibited higher photosynthetic capacities than WT plants. This finding suggests that GB accumulation increases tolerance to heat-enhanced photoinhibition. This increased tolerance was associated with an improvement in D1 protein content, which accelerated the repair of photosystem II (PSII) following heat-enhanced photoinhibition. Significant accumulations of hydrogen peroxide (H2O2) and superoxide radical (O2 ?) were observed in WT plants under heat stress. However, these accumulations were much less for the transgenic plants. An important finding reported herein is that exogenous GB cannot directly reduce the content of reactive oxygen species (ROS). In accordance with a lower relative electrolyte conductivity and malondialdehyde content, the activities of antioxidant enzymes were higher in transgenic lines than in WT plants, indicating that the degree of membrane injury in the transgenic plants was lower compared to the WT plants. These results suggest that GB accumulation in vivo cannot directly eliminate ROS. Rather, higher antioxidant enzyme activities must be maintained to lessen the accumulation of ROS in transgenic plants and to decrease the degree of membrane injury.  相似文献

This experiment was conducted to test the effects of foliar application of progesterone on the photochemical efficiency of photosystem II (PSII) and photosynthetic rate in wheat flag leaves subjected to cross-stress of heat and high light during grain-filling stage. The results showed that progesterone pretreatment increased the activities of superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase, and the contents of ascorbic acid and glutathione under the cross-stress. Meanwhile, the rate of O2 ? production, hydrogen peroxide (H2O2) and malondialdehyde contents in progesterone pretreated leaves were significantly lower under heat and high light stress. In parallel with the alleviation of oxidative stress, higher content of D1 protein in PSII reactive center was observed in progesterone pretreated leaves, resulting in a significant increase in the potential (Fv/Fm) and actual (ΦPS II) photochemical efficiency of PSII, and the net photosynthetic rate. In summary, this study suggested that foliar application of progesterone might protect the PSII complex from heat and high light stress-induced damage through enhancing antioxidant defense system and further facilitating D1 protein stability in the wheat leaves.  相似文献

Brassinosteroids (BRs), an important class of plant steroidal hormones, play a significant role in the amelioration of various biotic and abiotic stresses. 24-epibrassinolide (EBR), an active brassinosteroid, was applied exogenously in different concentrations to characterize a role of BRs in tolerance of melon (Cucumis melo L.) to high temperature (HT) stress and to investigate photosynthetic performance of HT-stressed, Honglvzaocui (HT-tolerant) and Baiyuxiang (HTsensitive), melon variety. Under HT, Honglvzaocui showed higher biomass accumulation and a lower index of heat injury compared with the Baiyuxiang. The exogenous application of 1.0 mg L?1 EBR, the most effective concentration, alleviated dramatically the growth suppression caused by HT in both ecotypes. Similarly, EBR pretreatment of HTstressed plants attenuated the decrease in relative chlorophyll content, net photosynthetic rate, stomatal conductance, stomatal limitation, and water-use efficiency (WUE), as well as the maximal quantum yield of PSII photochemistry (Fv/Fm), the efficiency of excitation capture of open PSII center, the effective quantum yield of PSII photochemistry (ΦPSII), photochemical quenching coefficient, and the photon activity distribution coefficients of PSI (α). EBR pretreatment further inhibited the increase in intracellular CO2 concentration, leaf transpiration rate, minimal fluorescence of dark-adapted state, nonphotochemical quenching, thermal dissipation, and photon activity distribution coefficients of PSII. Results obtained here demonstrated that EBR could alleviate the detrimental effects of HT on the plant growth by improving photosynthesis in leaves, mainly reflected as up-regulation of photosynthetic pigment contents and photochemical activity associated with PSI.  相似文献

Gas exchange and chlorophyll fluorescence techniques were used to evaluate the acclimation capacity of the schlerophyll shrub Heteromeles arbutifolia M. Roem. to the multiple co-occurring summer stresses of the California chaparral. We examined the influence of water, heat and high light stresses on the carbon gain and survival of sun and shade seedlings via a factorial experiment involving a slow drying cycle applied to plants grown outdoors during the summer. The photochemical efficiency of PSII exhibited a diurnal, transient decrease (δF/Fm′) and a chronic decrease or photoinhibition (Fv/Fm) in plants exposed to full sunlight. Water stress enhanced both transient decreases of δF/Fm’and photoinhibition. Effects of decreased δF/Fm’and Fv/Fm on carbon gain were observed only in well-watered plants since in water-stressed plants they were overidden by stomatal closure. Reductions in photochemical efficiency and stomatal conductance were observed in all plants exposed to full sunlight, even in those that were well-watered. This suggested that H. arbutifolia sacrificed carbon gain for water conservation and photoprotection (both structurally via shoot architecture and physiologically via down-regulation) and that this response was triggered by a hot and dry atmosphere together with high PFD, before severe water, heat or high PFD stresses occur. We found fast adaptive adjustments of the thermal stability of PSII (diurnal changes) and a superimposed long-term acclimation (days to weeks) to high leaf temperatures. Water stress enhanced resistance of PSII to high temperatures both in the dark and over a wide range of PFD. Low PFD protected photochemical activity against inactivation by heat while high PFD exacerbated damage of PSII by heat. The greater interception of radiation by horizontally restrained leaves relative to the steep leaves of sun-acclimated plants caused photoinhibition and increased leaf temperature. When transpirational cooling was decreased by water stress, leaf temperature surpassed the limits of chloroplast thermostability. The remarkable acclimation of water-stressed plants to high leaf temperatures proved insufficient for the semi-natural environmental conditions of the experiment. Summer stresses characteristic of Mediterranean-type climates (high leaf temperatures in particular) are a potential limiting factor for seedling survival in H. arbutifolia, especially for shade seedlings lacking the crucial structural photoprotection provided by steep leaf angles.  相似文献

The effects of NaCl stress on the growth and photosynthetic characters of Ulmus pumila L. seedlings were investigated under sand culture condition. With increasing NaCl concentration, main stem height, branch number, leaf number, and leaf area declined, while Na+ content and the Na+/K+ ratio in both expanded and expanding leaves increased. Na+ content was significantly higher in expanded leaves than in those just expanding. Chlorophyll (Chl) a and Chl b contents declined as NaCl concentration increased. The net photosynthetic rate, intercellular CO2 concentration, stomatal conductance, and transpiration rate also declined, but stomatal limitation value increased as NaCl concentration increased. Both the maximal quantum yield of PSII photochemistry and the effective quantum yield of PSII photochemistry declined as NaCl concentration rose. These results suggest that the accumulation of Na+ in already expanded leaves might reduce damage to the expanding leaves and help U. pumila endure high salinity. The reduced photosynthesis in response to salt stress was mainly caused by stomatal limitation.  相似文献

Effects of exogenous glycinebetaine (GB, 2–50 mM) on growth, photosynthetic gas exchange, PSII photochemistry, and the activities of key enzymes involved in CO2 fixation in maize plants were investigated. Growth, CO2 assimilation rate, and stomatal conductance increased at low GB concentrations (2–20 mM) but decreased significantly at high GB concentrations (30–50 mM). Leaf relative water content and water potential remained unchanged at low GB concentrations but decreased at high GB concentrations. The maximal efficiency of PSII photochemistry was unchanged either at low or high GB concentrations. The actual PSII efficiency ( Φ PSII) and photochemical quenching (qP) increased at low GB concentrations but decreased at high GB concentrations. At low GB concentrations, there were no significant changes in the efficiency of excitation energy capture by open PSII reaction centres (Fv′/Fm′) and non‐photochemical quenching (qN). At high GB concentrations, Fv′/Fm′ decreased while qN increased significantly. There were no changes in the activities of phosphoenolpyruvate carboxylase, pyruvate phosphate dikinase, and ribulose‐1,5‐bisphosphate carboxylase in control and GB‐fed plants. However, there was a linear correlation between CO2 assimilation rate and stomatal conductance in control and GB‐fed plants. Moreover, there were no significant differences in O2 evolution rate between control and GB fed‐plants under saturated CO2 conditions. The results suggest that exogenous GB application at certain concentrations can enhance CO2 assimilation rate, which can be explained by an increased stomatal conductance.  相似文献

Green house study was aimed to investigate the effect of seed biopriming with drought tolerant isolates of Trichoderma harzianum, viz. Th 56, 69, 75, 82 and 89 on growth of wheat under drought stress and to explore the mechanism underlying plant water stress resilience in response to Trichoderma inoculation. Measurements of relative water content, osmotic potential, osmotic adjustment, leaf gas exchange, chlorophyll fluorescence and membrane stability index were performed. In addition, analysis of the phenolics, proline, lipid peroxidation and measurements of phenylalanine ammonia‐lyase activity were carried out. Seed biopriming enhanced drought tolerance of wheat as drought induced changes like stomatal conductance, net photosynthesis and chlorophyll fluorescence were delayed. Drought stress from 4 to 13 days of withholding water induced an increase in the concentration of stress induced metabolites in leaves, while Trichoderma colonisation caused decrease in proline, malondialdehyde (MDA) and hydrogen peroxide (H2O2), and an increase in total phenolics. A common factor that negatively affects plants under drought stress conditions is accumulation of toxic reactive oxygen species (ROS), and we tested the hypothesis that seed biopriming reduced damages resulting from accumulation of ROS in stressed plants. The enhanced redox state of colonised plants could be explained by higher l ‐phenylalanine ammonia‐lyase (PAL) activity in leaves after 13 days of drought stress in Trichoderma treated plants. Similar activity was induced in untreated plants in response to drought stress but to a lower extent in comparison to treated plants. Our results support the hypothesis that seed biopriming in wheat with drought tolerant T. harzianum strains increased root vigour besides performing the process of osmoregulation. It ameliorates drought stress by inducing physiological protection in plants against oxidative damage, due to enhanced capacity to scavenge ROS and increased level of PAL, a mechanism that is expected to augment tolerance to abiotic stresses.  相似文献

Benzoxazolin-2-(3H)-one (BOA) has been tested in many plants species, but not in soybean (Glycine max). Thus, a hydroponic experiment was conducted to assess the effects of BOA on soybean photosynthesis. BOA reduced net photosynthetic rate, stomatal conductance, and effective quantum yield of PSII photochemistry without affecting intercellular CO2 concentration or maximal quantum yield of PSII photochemistry. Results revealed that the reduced stomatal conductance restricted entry of CO2 into substomatal spaces, thus limiting CO2 assimilation. No change found in intercellular CO2 concentration and reduced effective quantum yield of PSII photochemistry revealed that CO2 was not efficiently consumed by the plants. Our data indicated that the effects of BOA on soybean photosynthesis occurred due to the reduced stomatal conductance and decreased efficiency of carbon assimilation. The accumulation of BOA in soybean leaves reinforced these findings.  相似文献

We have recently isolated the AlSAP (stress-associated protein) gene from the halophyte grass Aeluropus littoralis and demonstrated that AlSAP expression improves tolerance to continuous salt and drought stresses in transgenic tobacco. To extend these findings to an important crop, we generated marker-free transgenic durum wheat plants of the commercial cv. Karim expressing the AlSAP gene. The integration and expression of AlSAP in T3 homozygous plants were ascertained by Southern, Northern and Western blotting respectively. AlSAP wheat lines exhibited improved germination rates and biomass production under severe salinity and osmotic stress conditions. Following a long-term salt or drought stress greenhouse trial, AlSAP lines produced normally filled grains whereas wild-type (WT) plants either died at the vegetative stage under salt stress or showed markedly reduced grain filling under drought stress. Measurements of the RWC (relative water content) and endogenous Na+ and K+ levels in leaves of AlSAP plants, showed a lower water loss rate and a higher Na+ accumulation in senescent-basal leaves, respectively, compared to those of WT plants. Taken together, these results extend to cereals the high potential of the AlSAP gene for engineering effective drought and salt tolerance.  相似文献

Background

The electron transport chain, Rubisco and stomatal conductance are important in photosynthesis. Little is known about their combined responses to heat treatment at different temperatures and following recovery in grapevines (Vitis spp.) which are often grown in climates with high temperatures.

Methodology/Findings

The electron transport function of photosystem II, the activation state of Rubisco and the influence of stomatal behavior were investigated in grapevine leaves during heat treatments and following recovery. High temperature treatments included 35, 40 and 45°C, with 25°C as the control and recovery temperature. Heat treatment at 35°C did not significantly (P>0.05) inhibit net photosynthetic rate (P n). However, with treatments at 40 and 45°C, P n was decreased, accompanied by an increase in substomatal CO2 concentration (C i), decreases in stomatal conductance (g s) and the activation state of Rubisco, and inhibition of the donor side and the reaction center of PSII. The acceptor side of PSII was inhibited at 45°C but not at 40°C. When grape leaves recovered following heat treatment, P n, gs and the activation state of Rubisco also increased, and the donor side and the reaction center of PSII recovered. The increase in P n during the recovery period following the second 45°C stress was slower than that following the 40°C stress, and these increases corresponded to the donor side of PSII and the activation state of Rubisco.

Conclusions

Heat treatment at 35°C did not significantly (P>0.05) influence photosynthesis. The decrease of P n in grape leaves exposed to more severe heat stress (40 or 45°C) was mainly attributed to three factors: the activation state of Rubisco, the donor side and the reaction center of PSII. However, the increase of P n in grape leaves following heat stress was also associated with a stomatal response. The acceptor side of PSII in grape leaves was responsive but less sensitive to heat stress.  相似文献
