Artificial self-purification (depuration) of mussels ( Mytilus edulis ) was undertaken at three temperatures, under conditions similar to those likely to be experienced in the commercial shellfish industry of the UK. During a 72 h depuration period, samples of mussel flesh were examined for three faecal indicator bacteria, Escherichia coli , Group D faecal streptococci and sulphite-reducing Clostridium spores, and two types of bacteriophage. There was a statistically significant difference in the elimination rate of faecal indicator bacteria compared with the slower rate for both bacteriophages.  相似文献

The effect of cadmium (Cd) on the number of different faecal indicator bacteria in sewage, and on species composition of different indicator bacteria, was studied. Different amounts of Cd were added to aliquots of a sewage sample, and after 0, 4% and 24 h of Cd exposure at 20°C conforms, faecal coliforms, faecal streptococci and Clostridium perfringens were enumerated by the membrane filter method. The Cd-induced reduction in the number of coliforms and faecal coliforms during exposure was found to be greater than the decrease in the number of faecal streptococci. In the case of C. perfringens the Cd concentrations used produced no observable effect on the cell number. The addition of Cd changed the faecal coliforms and faecal streptococci density relationship. Escherichia coli seems to be more resistant to Cd than other coliforms and Streptococcus faecalis var. liquefaciens and Streptococcus durans more resistant to Cd than other faecal streptococci. No influence of Cd on gas production by faecal coliforms was observed. Faecal streptococci and C. perfringens seem to be better indicator bacteria than coliforms and faecal coliforms in evaluating the hygienic quality of Cd polluted sewage.  相似文献

We have previously documented that faecal indicator bacteria (Escherichia coli, faecal coliform, enterococci) recommended by the U.S. Environmental Protection Agency (USEPA) to establish recreational water quality standards are naturally found in high concentrations in the surface and subsurface of soils in Hawaii. Rain, the source of all streams in Hawaii, washes the soil sources of faecal bacteria into all the streams of Hawaii, at concentrations which consistently exceed the USEPA recreational water quality standards. The objective of this study was to test the hypothesis that faecal bacteria are able to establish themselves in the soil environments of tropical islands by conducting the same study in Guam, a tropical pacific island with warmer temperatures and higher humidity than Hawaii. The same methods and study design used in Hawaii was used in Guam. The results of the study conducted in Guam revealed that all streams contain consistently high concentrations of faecal coliform, E. coli, and enterocci which exceeded the old USEPA recreational water quality standard of 200 faecal coliform/100 ml as well as the new water quality standards of 126 E. coli/100 ml or 33 enterococci/100 ml. These same faecal indicator bacteria were recovered in high concentrations in surface and subsurface (18-36 cm depth) soil samples in Guam. Limited coastal water analysis showed that most coastal marine waters contain low concentrations of faecal bacteria but coastal waters impacted by stream run-off showed elevated levels of faecal bacteria. The results of this study support the hypothesis that environmental conditions in the tropical areas of the world can support the growth and establishment of populations of faecal bacteria in the soil. Thus, soil becomes an environmental, non-faecal source of faecal indicator bacteria. These results indicate that USEPA water quality standards may not be directly applicable to tropical island environments.  相似文献

The freshwater mussel Anodonta cygnea and four marine shellfish (mussels, Mytilus edulis; cockles, Cerastoderma edule; clams, Mya arenaria; Scrobicularia plana) from a total of six sites were surveyed for Escherichia coli, Clostridium perfringens, faecal streptococci, 25 and 37 °C coliforms, 25 °C and 37 °C total viable numbers and fluorescent pseudomonads. The A. cygnea from an urban lake contained greater numbers of the faecal indicator bacteria than animals from a rural lake. There were also differences in the other bacterial counts and these were discussed with respect to bacterial parameter and animal characteristics. When freshwater mussels were transferred from the city site to the rural site for 24 h the load of faecal indicator bacteria was eliminated or significantly reduced. Other bacterial types took longer to become stabilised. Loss of indicator bacteria from Anodonta was also demonstrated using cleansing in the laboratory. Very high bacterial numbers were found in some marine molluscs notably Scrobicularia plana and most shellfish contained significant numbers of the three faecal indicator bacteria at every sampling occasion. The relationship between bacterial types was discussed and it was concluded that in both freshwater and marine animals the bacterial numbers were determined more by sampling site than by species of shellfish.  相似文献

AIMS: To provide evidence of septic system failure by comparing two faecal indicator bacteria, enterococci and Escherichia coli, from defective septic tanks and adjacent creeks. METHODS AND RESULTS: A biochemical fingerprinting method was used to type and compare enterococci and E. coli strains from 39 septic tanks with creek water samples. Phenotypic diversity of enterococci (0.5 +/- 0.3) and E. coli (0.5 +/- 0.3) in septic tanks were significantly lower than those found in water samples (0.8 +/- 0.1, P < 0.0001 for enterococci and 0.9 +/- 0.1, P < 0.0001 for E. coli). Among 1072 enterococci isolates tested from septic tanks, 203 biochemical phenotypes (BPTs) were found of which 98 BPTs from 33 septic tanks were identical to several water samples. Similarly, among 621 E. coli isolates tested from septic tanks, 159 BPTs were found of which 53 BPTs from 26 septic tanks were also identical to water samples. The number of the latter bacteria was significantly (P = 0.01) higher in water samples collected from downstream compared with that of upstream in the study area. A high similarity between the populations of both indicator bacteria was also found between defective septic tanks and downstream water samples further indicating the contamination of both creeks by defective septic systems. CONCLUSIONS: Biochemical fingerprinting of faecal indicator bacteria is a useful and rapid method to provide direct evidence for septic system failure. Combination of both faecal indicator bacteria (enterococci and E. coli) provides a better judgement of the performance of a septic system. SIGNIFICANCE AND IMPACT OF THE STUDY: This study is the first to provide direct evidence of septic system failure by identifying the presence of specific bacterial types in septic tanks and surface waters. Based on our findings, we suggest that the performance evaluation of a septic system should be accompanied by direct analysis of faecal indicator bacteria.  相似文献

Suspensions of the faecal indicator bacteria Escherichia coli and Enterococcus faecalis were incubated in full sunlight in plastic bottles containing either (i) air-equilibrated (oxygenated) water or (ii) anaerobic (deoxygenated) water. A rapid decrease in cfu ml−1 was observed for actively growing and stationary phase cells of both types of faecal bacteria when illuminated under aerobic conditions, with Ent. faecalis showing the greater enhancement in the rate of inactivation in air-equilibrated water. The demonstration of an oxygen requirement for the inactivation of faecal bacteria in sunlight indicates that solar-based water disinfection systems are likely to require fully aerobic conditions in order to function effectively.  相似文献

AIM: The survival of indicator micro-organisms in aquatic systems is affected by both biotic and abiotic factors. Much of the past research on this topic has been conducted using laboratory-generated cultures of indicator bacteria. For this study, we used natural sources of faecal contamination as inoculants into environmental water samples, thereby representing the wide diversity of organisms likely to be found in faecal contamination. METHODS AND RESULTS: Rates of inactivation of water quality indicators, total coliforms (TC), Escherichia coli, enterococci (EC) and F+-specific coliphage were studied in three experiments using inoculants of sewage influent, sewage effluent and urban storm drain run-off. Effects of temperature, nutrients, total suspended solids, bacterial load and solar irradiation were studied in fresh and seawater matrices. Results demonstrated that temperature and solar irradiation had significant effects upon rates of inactivation (anova, P < 0.001). Inactivation rates were similar, regardless of the inoculant type. EC degraded the slowest in the dark with T90s of 115-121 and 144-177 h at 20 and 14 degrees C, respectively. When incubated in sunlight, EC was inactivated significantly more rapidly than either E. coli or F+-specific coliphage (P < 0.001). CONCLUSIONS: Inactivation of indicator bacteria is not dependent upon the original source of contamination. Inactivation rates of indicator bacteria were similar in fresh and seawater matrices. However, EC degraded more rapidly in sunlight than E. coli. SIGNIFICANCE AND IMPACT OF THE STUDY: This study suggests that the source of faecal contamination is not an important factor to inactivation rates of indicator bacteria. However, rates of inactivation of indicator bacteria are likely system specific.  相似文献

AIMS: To investigate the survival of cultivable bacteria in the tropical Bangpakong estuary (Eastern Thailand) under different salinities and light conditions. METHODS AND RESULTS: Dark and light microcosm experiments using membrane diffusion chambers were carried out under three different experimental conditions, namely (i) low salinity, (ii) progressive mixing with brackish water and (iii) fast mixing with high salinity water spiked with raw urban sewage. Faecal coliforms declined faster than faecal enterococci, as shown by survival T90 values ranging from 82.2 +/- 4.2 to 14.5 +/- 0.8 h and 97.5 +/- 0.4-20.6 +/- 1.2 h, respectively. The survival of freshwater heterotrophic bacteria was higher but variable (121.2 +/- 5.0-30.1 +/- 14.3 h), whereas that of heterotrophic marine bacteria was rather stable (81.5 +/- 4.2-44.6 +/- 2.5 h). CONCLUSIONS: Overall survival was higher in low salinities. Light had a further deleterious effect, since it accelerated the decay of faecal indicators, particularly in high salinities. Faecal enterococci had a higher resistance to environmental conditions compared with faecal coliforms. SIGNIFICANCE AND IMPACT OF THE STUDY: This study is relevant to the understanding of the behaviour of different faecal indicator bacteria and the optimization of sewage treatment plants aimed at the reduction and/or elimination of faecal load discharged into estuarine waters submitted to salinity variations.  相似文献

AIMS: Investigations were carried out to observe the influence of winter/spring and summer periods on the survival of Salmonella typhimurium and indicator bacteria (psychrophilic, mesophilic, coliform and faecal coliform bacteria and faecal streptococci) in the solid fraction of pig slurry from agricultural wastewater treatment plant. METHODS AND RESULTS: Leather squares and PVC bottles with openings served as test carriers. They were inoculated with broth culture of Salm. typhimurium and introduced directly into the solid fraction. During the experiment, quantitative and qualitative examinations were carried out to determine the presence of Salm. typhimurium and observe the dynamics of indicator bacteria in the solid fraction. CONCLUSIONS: Salmonella typhimurium survived for 26 d in summer and for 85 d in winter/spring. The T90 values of indicator bacteria in summer ranged from 35.44 d (coliform bacteria) up to 100.29 d (mesophilic bacteria). The winter T90 values of indicator bacteria ranged from 74.58 d (faecal coliform bacteria) to 233.07 d (coliform bacteria). SIGNIFICANCE AND IMPACT OF THE STUDY: The present study demonstrated that it is necessary to pay increased attention to the manipulation of slurry solid fraction.  相似文献

To close the loop of nutrients in a safe way, the loop of pathogens has to be broken. By sanitising organic fertilisers derived from faeces and manure, the first step in the disease transmission chain is broken. Two alternatives for this treatment, thermophilic composting and ammonia-based treatment, were evaluated and compared to storage treatment. Thermal composting of faecal matter and food waste resulted in a treatment temperature of over 65 degrees C in a 90 L reactor. By using insulation and turning the compost three times during the high temperature period, it is possible to ensure a 5 log(10) reduction of pathogens. Small scale composting of the same material indicated less efficient reduction of faecal bacteria at temperatures around 50 degrees C. In the chemical treatment tested, an addition of 3% N-NH(3) increased the pH to above 9 within 1h and resulted in a good reduction in the indicator organisms for bacteria (Salmonella spp. and faecal coliforms D(r)<0.7 days, Enterococcus spp. D(r)<3 days). Lower addition rates resulted in a longer treatment period needed. The storage period tested resulted in slow reduction of faecal coliforms, Salmonella spp. and Enterococcus spp.  相似文献

The Bacteroides fragilis group has been evaluated as a prospective rapid indicator of faecal contamination of water. Fluorescent antibody (FA) stained B. fragilis group bacteria were enumerated microscopically and compared with faecal coliform or Escherichia coli counts as indicators of faecal contamination. Environmental samples included surface waters (raw drinking water and known contaminated water). Laboratory disinfection experiments with ozone, chlorine and u. v. radiation were also performed. Bacteroides FA counts specifically detected recent human faecal contamination in field samples in 2–3 h. Samples with a high content of particulates or debris limited the sensitivity to about 10 FA counts/ml. Viable counts showed that the sensitivity to all three disinfection agents was essentially the same for Bacteroides and E. coli. Fluorescent antibody counts of Bacteroides , conversely, were not altered by any of the agents. Therefore, the Bacteroides FA method is not recommended for routine monitoring but may be useful for cases where extensive human faecal contamination is suspected (e.g. pipeline rupture or pollution of recreational water) and where rapid remedial action must be taken to protect the public health.  相似文献

Population dynamics of Aeromonas spp. in an urban river watershed   总被引:1,自引:1,他引:0  
Density of Aeromonas spp. at one site in the Buffalo River and at four sites on its upstream tributaries was followed from June 1992–June 1993. Membrane filtration counts of Aeromonas during the summer ranged between 18 and 4000 ml−1, which were one to two logs higher than faecal coliform and faecal streptococci densities. Aeromonas spp. in the Buffalo River, and faecal coliforms, faecal streptococci, and the heterotrophic plate count throughout the watershed, increased by approximately one log during summer rainstorms. However, Aeromonas spp. increased only by a factor of two during rainstorms at the upstream sites. Aeromonas spp. showed a strong positive correlation with both indicator bacteria and total suspended solids at the upstream sites during the summer but not the winter. Correlations between Aeromonas and indicator bacteria remained strong in the Buffalo River during the winter, signifying that different conditions exist in the Buffalo River and its upstream tributaries. The strong correlation between Aeromonas spp. and indicator bacteria in the Buffalo River suggest that, in the absence of media capable of the quantitative recovery of potentially pathogenic aeromonads, standard faecal coliform analyses may adequately assess public health risks from Aeromonas spp. in an urban river used for recreational purposes.  相似文献

The elimination of sewage effluent-associated poliovirus, Escherichia coli, and a 22-nm icosahedral coliphage by the common mussel, Mytilus edulis, was studied. Both laboratory-and commercial-scale recirculating, UV depuration systems were used in this study. In the laboratory system, the logarithms of the poliovirus, E. coli, and coliphage levels were reduced by 1.86, 2.9, and 2.16, respectively, within 52 h of depuration. The relative patterns and rates of elimination of the three organisms suggest that they are eliminated from mussels by different mechanisms during depuration under suitable conditions. Poliovirus was not included in experiments undertaken in the commercial-scale depuration system. The differences in the relative rates and patterns of elimination were maintained for E. coli and coliphage in this system, with the logarithm of the E. coli levels being reduced by 3.18 and the logarithm of the coliphage levels being reduced by 0.87. The results from both depuration systems suggest that E. coli is an inappropriate indicator of the efficiency of virus elimination during depuration. The coliphage used appears to be a more representative indicator. Depuration under stressful conditions appeared to have a negligible affect on poliovirus and coliphage elimination rates from mussels. However, the rate and pattern of E. coli elimination were dramatically affected by these conditions. Therefore, monitoring E. coli counts might prove useful in ensuring that mussels are functioning well during depuration.  相似文献

AIMS: This study evaluates the behaviour in spiked sludge of a pathogenic bacteria, Listeria monocytogenes, by cultural and molecular techniques, and compares its survival with the one of a faecal indicator, Enterococcus faecium. METHODS AND RESULTS: Listeria monocytogenes strain Scott A and E. faecium(T) were followed for 17 days after inoculation in sludge. Kinetics of survival depended on the bacteria and on the technique used [most probable number method, direct plate count or real-time quantitative PCR (qPCR)]. The concentration of L. monocytogenes decreased rapidly regardless of the technique, but the decrease was much more dramatic with culture techniques than with qPCR. On the contrary, the concentrations of culturable E. faecium(T) were stable. CONCLUSIONS: The results suggest that the cells of L. monocytogenes strain Scott A might have entered a viable, but nonculturable (VBNC) status, whereas cells of the indicator bacteria, E. faecium(T), maintained themselves better and stayed culturable. SIGNIFICANCE AND IMPACT OF THE STUDY: The difference of survival kinetics in the sludge of a faecal indicator (E. faecium) and a pathogenic bacterium (L. monocytogenes) may be linked to the fact that they either enter or do not enter into a VBNC status.  相似文献

To address the issue of human sewage reaching corals along the main reef of the Florida Keys, samples were collected from surface water, groundwater and coral [surface mucopolysaccharide layers (SML)] along a 10 km transect near Key Largo, FL. Samples were collected semi‐annually between July 2003 and September 2005 and processed for faecal indicator bacteria (faecal coliform bacteria, enterococci and Clostridium perfringens) and human‐specific enteric viruses (enterovirus RNA and adenovirus DNA) by (RT)‐nested polymerase chain reaction. Faecal indicator bacteria concentrations were generally higher nearshore and in the coral SML. Enteric viruses were evenly distributed across the transect stations. Adenoviruses were detected in 37 of 75 samples collected (49.3%) whereas enteroviruses were only found in 8 of 75 samples (10.7%). Both viruses were detected twice as frequently in coral compared with surface water or groundwater. Offshore, viruses were most likely to be found in groundwater, especially during the wet summer season. These data suggest that polluted groundwater may be moving to the outer reef environment in the Florida Keys.  相似文献

SUMMARY: The suitability, as indicators of the faecal contamination of foods, of two types of obligately anaerobic nonsporeforming bacteria which are very numerous in faeces, viz. members of the genera Bacteroides and Bifidibacterium , was tested. For the counting of Bacteroides , Beerens'bile-azide agar (1957) enriched with 10% (v/v) of blood, and incubated in oval cross section tubes, was found useful. Bifidibacterium could be counted very well in Frisell's tomato-azide agar (1951), incubated in the same way . Both media being not too selective, confirmation of representative numbers of colonies was necessary. After microscopical examination, the biochemical study of properties such as catalase reaction, oxygen tolerance, carbohydrate fermentation and nitrate reduction may be required.
The 3 Bacteroides strains tested died off rather rapidly when present in sterile ice cream under conditions where Enterobacteriaceae and faecal streptococci increased in numbers. Accordingly they were never found in considerable numbers in some 25 samples of food which appeared to contain very high numbers of other bacteria of faecal origin. The 8 Bifidibacterium strains investigated appeared more resistant. However, under conditions permitting growth, they increased less than the Enterobacteriaceae and faecal streptococci; and where the latter did not increase but did survive, the Bifidibacterium decreased in numbers. In line with these findings Bifidibacterium was not even found in numbers of the order of 10/g in the 25 samples of food mentioned above. Neither Bacteroides nor Bifidibacterium can therefore be considered suitable as an indicator of the contamination of foods with bacteria of faecal origin and of the subsequent growth of these bacteria therein.  相似文献

Aims: The development of a microbial source tracking (MST) method is strongly desired to ensure public health and bacteriological safety in coastal recreation areas. We try to specify the source of faecal pollution by applying pulsed‐field gel electrophoresis (PFGE) to the study of the aquatic environment on Aoshima Beach, Japan. Methods and Results: Enterococcus faecium, an enterococcus, was used as a faecal indicator bacterium in this study. Enterococcus faecium strains were isolated and identified from each water sample collected from Aoshima Beach and five rivers (Oyodo, Kiyotake, Kaeda, Chifuku and Tsukunami Rivers) that might be potential sources of faecal pollution. Enterococcus faecium strains collected from water samples were analysed using PFGE. The similarities of all the PFGE types of the Ent. faecium strains were compared using dendrogram analysis. The PFGE types of the strains isolated from Aoshima Beach showed a high similarity to those of the strains isolated from the Oyodo River at a 0·9 similarity level. It was suggested that the Oyodo River is the source of faecal pollution on Aoshima Beach. Conclusions: The PFGE analysis using enterococci is a potential tool for the MST of faecal indicator bacteria that can be applied to the study of the coastal environment. Significance and Impact of the Study: This is one of the studies that PFGE was applied to the coastal environment. The approach using PFGE could estimate the river that is source of faecal pollution in Aoshima Beach. By applying PFGE as a tool of MST method, detailed information of faecal pollution in coastal area can be provided.  相似文献

Alternaria species have been recognized as important cause of allergic diseases and are considered opportunistic pathogens for immunocompromised individuals. In order to assess the contribution of waters to the spread of Alternaria, we examined 390 water samples of various origin for the presence of these microorganisms in parallel with the standard pollution indicator bacteria, i.e. total coliforms, faecal coliforms and enterococci. Alternaria spp. were isolated from 42 out of 196 (21.4%) marine water samples, from 13 out of 68 (19.1%) river samples, whereas out of 126 potable waters only two (1.6%) were found positive. The incidence of Alternaria was significantly higher (p < 0.001) in marine and river samples than in potable waters. The mean values of the colony-forming units of the pollution indicator bacteria did not significantly differ between Alternaria-positive and Alternaria-negative samples.  相似文献
