Soil cores (35 cm long, 7 cm diameter) from the Macaulay Land Use Research Institute's Sourhope Research Station in the Scottish Borders were kept and monitored at constant temperature (18± 1°C) for gas production using a 1.6 mm diameter stainless steel probe fitted with a membrane inlet and connected to a quadrupole mass spectrometer. This provided a novel method for on-line, real time monitoring of soil gas dynamics. In closed-system headspace experiments, O2 and CO2 (measured at m/z values 32 and 44, respectively) showed anti-phase diurnal fluctuations in low-intensity simulated daylight and under a light-dark (LD, 12:12 h) regime. O2 increased during periods of illumination and decreased in the dark. The inverse was true for CO2 production. Ar (m/z = 40) concentration and temperature (°C) remained constant throughout the experiments. The same phase-related oscillations, in CO2 and O2 concentrations, were observed at 2 and 5 cm depth in soil cores. The O2 concentration did not oscillate diurnally at 10 cm depth. In below-ground experiments, CH4 (m/z = 15) concentration showed diurnal cycles at 2, 5 and 10 cm depth. The CH4 production had the same diurnal phase cycle as CO2 but with lower amplitude. Evidence of below-ground diurnal oscillations in N2 (m/z = 28) concentration was provided at 5 cm depth. The scale of production and consumption of gases associated with soil-atmosphere interactions and below-ground processes, are shown to be a multifaceted output of several variables. These include light, circadian-controlled physiological rhythms of plants and microbes, and the interactions between these organisms.  相似文献

To ensure food security in Africa and Asia, developing sorghum varieties with grain quality that matches consumer demand is a major breeding objective that requires a better understanding of the genetic control of grain quality traits. The objective of this targeted association study was to assess whether the polymorphism detected in six genes involved in synthesis pathways of starch (Sh2, Bt2, SssI, Ae1, and Wx) or grain storage proteins (O2) could explain the phenotypic variability of six grain quality traits [amylose content (AM), protein content (PR), lipid content (LI), hardness (HD), endosperm texture (ET), peak gelatinization temperature (PGT)], two yield component traits [thousand grain weight (TGW) and number of grains per panicle (NBG)], and yield itself (YLD). We used a core collection of 195 accessions which had been previously phenotyped and for which polymorphic sites had been identified in sequenced segments of the six genes. The associations between gene polymorphism and phenotypic traits were analyzed with Tassel. The percentages of admixture of each accession, estimated using 60 RFLP probes, were used as cofactors in the analyses, decreasing the proportion of false-positive tests (70%) due to population structure. The significant associations observed matched generally well the role of the enzymes encoded by the genes known to determine starch amount or type. Sh2, Bt2, Ae1, and Wx were associated with TGW. SssI and Ae1 were associated with PGT, a trait influenced by amylopectin amount. Sh2 was associated with AM while Wx was not, possibly because of the absence of waxy accessions in our collection. O2 and Wx were associated with HD and ET. No association was found between O2 and PR. These results were consistent with QTL or association data in sorghum and in orthologous zones of maize. This study represents the first targeted association mapping study for grain quality in sorghum and paves the way for marker-aided selection.  相似文献

Different authors have investigated the immunohistochemical expression of some proteins in the adenocarcinoma of the stomach, including cell cycle regulators proteins like p53 and Bcl-2; growth factors (c-erb-B2 and EPO-R); angiogenesis-related markers such as COX-2 and cellular adhesion molecules (beta-catenin and E-cadherin). While these proteins have been studied in gastric adenocarcinoma, their immunophenotyping in non tumoral gastric mucous membrane remains unexplored. In the present study, we investigated the expression, function and behavior of these proteins in normal gastric mucous membrane to contribute to gain further knowledge on the significance of their loss or overexpression in malignant gastric tumors.  相似文献

在好氧和厌氧两种条件下研究了1,2,4-三氯苯的降解,结果表明,1,2,4-三氯苯的好氧降解和厌氧降解均遵循一级反应动力学在同样水分、温度及初始浓度条件下,1,2,4-三氯苯的好氧降解比厌氧降解迅速,其半衰期分别为1.89~5.86和5.07~19.08d土壤中1,2,4-三氯苯的初始浓度对于其降解也有显著影响,在0~100μg·g-1的范围内,浓度增高时,其降解加快,说明污染物浓度对降解的影响;在10~30℃范围内,温度增高导致降解过程加快,归因于温度升高对微生物酶活性的激活作用.  相似文献

Isocitrate dehydrogenases (IDHs) catalyse oxidative decarboxylation of isocitrate to α-ketoglutarate (α-KG). IDH1 functions in the cytosol and peroxisomes, whereas IDH2 and IDH3 are both localized in the mitochondria. Heterozygous somatic mutations in IDH1 occur at codon 132 in 70% of grade II-III gliomas and secondary glioblastomas (GBMs), and in 5% of primary GBMs. Mutations in IDH2 at codon 172 are present in grade II-III gliomas at a low frequency. IDH1 and IDH2 mutations cause both loss of normal enzyme function and gain-of-function, causing reduction of α-KG to D-2-hydroxyglutarate (D-2HG) which accumulates. Excess hydroxyglutarate (2HG) can also be caused by germline mutations in D- and L-2-hydroxyglutarate dehydrogenases (D2HGDH and L2HGDH). If loss of IDH function is critical for tumourigenesis, we might expect some tumours to acquire somatic IDH3 mutations. Alternatively, if 2HG accumulation is critical, some tumours might acquire somatic D2HGDH or L2HGDH mutations. We therefore screened 47 glioblastoma samples looking for changes in these genes. Although IDH1 R132H was identified in 12% of samples, no mutations were identified in any of the other genes. This suggests that mutations in IDH3, D2HGDH and L2HGDH do not occur at an appreciable frequency in GBM. One explanation is simply that mono-allelic IDH1 and IDH2 mutations occur more frequently by chance than the bi-allelic mutations expected at IDH3, D2HGDH and L2HGDH. Alternatively, both loss of IDH function and 2HG accumulation might be required for tumourigenesis, and only IDH1 and IDH2 mutations have these dual effects.  相似文献

The levels of fructose 2,6-P2 and 6-phosphofructo 2-kinase have been found to be decreased in the liver of both ketotic and non-ketotic diabetic rats, a good correlation between fall of hepatic fructose 2,6-P2, ketonemia and glycemia being observed. The "total" 6-phosphofructo 2-kinase activity and the "active" (non-phosphorylated) from of the enzyme were decreased to a different extent, resulting in a fall of the "active"/"total" activity ratio. Hepatic levels of glucose 1,6-P2 were lowered only in ketotic diabetes. Insulin treatment normalized all the values studied. Insulin administration to control rats decreased the hepatic levels of fructose 2,6-P2 and did not affect glucose 1,6-P2 levels. It also decreased the "active" form of 6-phosphofructo 2-kinase, without significantly altering the "total" activity.  相似文献

We studied the distribution of dissolved O2, CO2, CH4, and N2O in a coastal swamp system in Thailand with the goal to characterize the dynamics of these gases within the system. The gas concentrations varied spatially and seasonally in both surface and ground waters. The entire system was a strong sourcefor CO2 and CH4, and a possible sink for atmospheric N2O. Seasonal variation in precipitation primarily regulated the redox conditions in the system. However, distributions of CO2, CH4, and N2O in the river that received swamp waters were not always in agreement with redox conditions indicated by dissolvedO2 concentrations. Sulfate production through pyriteoxidation occurred in the swamp with thin peat layerunder aerobic conditions and was reflected by elevatedSO 4 2– /Cl in the river water. When SO 4 2– /Cl was high, CO2 and CH4 concentrations decreased, whereas the N2O concentration increased. The excess SO 4 2– in the river water was thus identified as a potential indicator for gas dynamics in this coastal swamp system.  相似文献

We have synthesized several ganglio-oligosaccharide structures using glycosyltransferases from Campylobacter jejuni. The enzymes, alpha-(2-->3/8)-sialyltransferase (Cst-II), beta-(1-->4)-N-acetylgalactosaminyltransferase (CgtA), and beta-(1-->3)-galactosyltransferase (CgtB), were produced in large-scale fermentation from Escherichia coli and further characterized based on their acceptor specificities. 2-Azidoethyl-glycosides corresponding to the oligosaccharides of GD3 (alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->3)-beta-D-Galp-(1-->4)-beta-D-Glcp-), GT3 (alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->3)-beta-D-Galp-(1-->4)-beta-D-Glcp-), GM2 (beta-D-GalpNAc-(1-->4)-[alpha-D-Neup5Ac-(2-->3)]-beta-D-Galp-(1-->4)-beta-D-Glcp-), GD2 (beta-D-GalpNAc-(1-->4)-[alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->3)]-beta-D-Galp-(1-->4)-beta-D-Glcp-), GT2 (beta-D-GalpNAc-(1-->4)-[alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->8)-alpha-D-Neup5Ac-(2-->3)]-beta-D-Galp-(1-->4)-beta-D-Glcp-), and GM1 (beta-D-Galp-(1-->3)-beta-D-GalpNAc-(1-->4)-[alpha-D-Neup5Ac-(2-->3)]-beta-D-Galp-(1-->4)-beta-D-Glcp-) were synthesized in high yields (gram-scale). In addition, a mammalian alpha-(2-->3)-sialyltransferase (ST3Gal I) was used to sialylate GM1 and generate GD1a (alpha-D-Neup5Ac-(2-->3)-beta-D-Galp-(1-->3)-beta-D-GalpNAc-(1-->4)-[alpha-D-Neup5Ac-(2-->3)]-beta-D-Galp-(1-->4)-beta-D-Glcp-) oligosaccharide. We also cloned and expressed a rat UDP-N-acetylglucosamine-4'epimerase (GalNAcE) in E. coli AD202 cells for cost saving in situ conversion of less expensive UDP-GlcNAc to UDP-GalNAc.  相似文献

The aim of the study was to investigate the effect of resistance exercise alone or in combination with oral intake of branched-chain amino acids (BCAA) on phosphorylation of the 70-kDa S6 protein kinase (p70(S6k)) and mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinase (ERK1/2), and p38 MAPK in skeletal muscle. Seven male subjects performed one session of quadriceps muscle resistance training (4 x 10 repetitions at 80% of one repetition maximum) on two occasions. In a randomized order, double-blind, crossover test, subjects ingested a solution of BCAA or placebo during and after exercise. Ingestion of BCAA increased plasma concentrations of isoleucine, leucine, and valine during exercise and throughout recovery after exercise (2 h postexercise), whereas no change was noted after the placebo trial. Resistance exercise led to a robust increase in p70(S6k) phosphorylation at Ser(424) and/or Thr(421), which persisted 1 and 2 h after exercise. BCAA ingestion further enhanced p70(S6k) phosphorylation 3.5-fold during recovery. p70(S6k) phosphorylation at Thr(389) was unaltered directly after resistance exercise. However, during recovery, Thr(389) phosphorylation was profoundly increased, but only during the BCAA trial. Furthermore, phosphorylation of the ribosomal protein S6 was also increased in the recovery period only during the BCAA trial. Exercise led to a marked increase in ERK1/2 and p38 MAPK phosphorylation, which was completely suppressed upon recovery and unaltered by BCAA. In conclusion, BCAA, ingested during and after resistance exercise, mediate signal transduction through p70(S6k) in skeletal muscle.  相似文献

目的:研究补肾活血汤对去卵巢大鼠冠脉微血管内皮细胞形态及功能的影响。方法:3月龄成年雌性大鼠随机分为8组,共计46只,分别为:正常组(NORM)、假手术组(SHAM),去卵巢组(OVX),补肾活血汤高、低剂量组(OVX/BHT-H、OVX/BHT-L)、17β-雌二醇干预组(OVX/ERT)。以透射电镜观察微血管内皮细胞细胞器变化;放射免疫法测定实验大鼠血清E2、ET、PGI2、TXA2的含量;采用硝酸还原酶法测定血浆NO含量。结果:与正常组相比,去卵巢组心肌及微血管内皮细胞细胞器破坏严重,中药组及17β-雌二醇干预组有所改善。与正常组相比,去卵巢组大鼠血浆中E2下降明显,有显著性差异;与去卵巢组相比,中药组大鼠E2明显上升,有显著性差异;与雌激素组相比无明显差异;与去卵巢组相比,中药组大鼠血浆ET-1值有所降低,其中以高剂量组最明显,中药组组大鼠的NO值稍有升高,但与正常组相比,没有显著性差异;中药组大鼠的TXA2值明显降低,其中以高剂量组最明显,中药组PGI2值升高,与正常组相比,有显著性差异,与17β-雌二醇干预组相比无显著性差异。结论:补肾活血汤能预防去卵巢后心肌微血管内皮细胞的形态和功能的破坏,其机制可能与其增加E2含量有关。  相似文献
