(22R,23R)-22,23-dihydroxystigmast-4-en-3-one, (22R,23R)-22,23-dihydroxystigmast-4-en-3,6-dione, (22R,23R)-3beta,5alpha,6beta,22,23-pentahydroxystigmastane, (22R,23R)-5alpha,6alpha-oxido-3beta,22,23-trihydroxystigmastane, (22R,23R)-5beta,6beta-oxido-3beta,22,23-trihydroxystigmastane, and (22R,23R)-3beta,6beta,22,23-tetrahydroxystigmast-4-ene were synthesized. Their cytotoxicities were comparatively studied using the MCF-7 line of carcinoma cells of human mammary gland and cells of human hepatoma of the Hep G2 line.  相似文献

Gamma-radiation-induced chromosomal interchanges in two inbred lines of rye were analyzed by using the C-banding technique. Neither the doses of radiation nor the time elapsed between radiation and cytological observation influenced the contribution of individual chromosomes to the observed interchanges. Dicentric are more frequent than monocentric interchanges. Neither the individual chromosome contribution to interchanges nor the chromosome combinations fit a random distribution. Chromosomes 6(5R), 4(4R) and 7(1R) of the inbred line Pool and 6(5R), 5(6R) and 4(4R) of line Riodeva, which are carriers of large amounts of heterochromatin, are much more frequently involved in interchanges than expected. Likewise, they are involved in the most frequent combinations observed (6-4, 6-7, 4-5, 4-7 and 6-3 (2R) in line Pool and 6-5, 6-4, 4-5 and 6-7 in line Riodeva). The role of C-heterochromatin in radiation-induced chromosomal interchanges is discussed.  相似文献

From the roots of Leontopotium longifolium, three new bisabolane sesquiterpenes, rel-(1S,4R,5S,6R)-4,5-diacetoxy-6-[(R)-1,5-dimethylhexa-3,5-dienyl]-3-methylcyclohex-2-enyl (Z)-2-methylbut-2-enoate (1), rel-(1S,4R,5S,6R)-4,5-diacetoxy-6-[(R)-5-hydroxy-1,5-dimethylhex-3-enyl]-3-methylcyclohex-2-enyl (Z)-2-methylbut-2-enoate (2), rel-(1R,2S,4R,5S)-4-acetoxy-2-[(R)-5-hydroxy-1,5-dimethylhex-3-enyl]-5-methylcyclohexyl (Z)-2-methylbut-2-enoate (3), and a new coumarin, 2,3-dihydro-5-hydroxy-2-(1-methylethenyl)-7H-pyrano[2,3-g][1,4]benzodioxin-7-one (4) together with nine known compounds have been isolated. The structures of these compounds were established by spectroscopic methods. Compounds 1 and 2 exhibited moderate cytotoxic activities against human promyelocytic leukemia (HL-60) cells.  相似文献

Commercially available 5-formyltetrahydrofolate (5-CHO-H4PteGlu) is chemically prepared in a reaction that introduces an asymmetric center at the 6 carbon, and hence is the mixture of diastereomers differing in chirality about this position. (6R)-5-CHO-H4PteGlu, the diastereomer that is not normally found in vivo, was prepared from folic acid. Folic acid was chemically reduced and (6R)-tetrahydrofolate (H4PteGlu) was obtained from the resultant (6R,S)-H4PteGlu by enzymatic consumption of the natural diastereomer of (6R,S)-5,10-CH2-H4PteGlu (reversibly formed from (6R,S)-H4PteGlu in the presence of formaldehyde) with Lactobacillus casei thymidylate synthase. The 5 position of purified (6R)-H4PteGlu was directly formylated in a carbodiimide-catalyzed reaction. The level of contamination of these preparations with the corresponding 6S diastereomers was estimated using the binding of fluorodeoxyuridylate to thymidylate synthase promoted by folate cofactor (for H4PteGlu) and by the growth of folate requiring bacteria (for 5-CHO-H4PteGlu). Purified preparations of (6R)-H4PteGlu promoted the binding of fluorodeoxyuridylate to L. casei thymidylate synthase (in the presence of formaldehyde) only at concentrations greater than 1000-fold higher than equiactive levels of (6S)-H4PteGlu. Likewise, the (6R)-5-CHO-H4PteGlu made by this method was 600 times less active as a growth factor for Pediococcus cerevisiae than was authentic (6S)-5-CHO-H4PteGlu. Hence, the minimum stereochemical purity of these preparations was 99.9% for (6R)-H4PteGlu and 99.8% for (6R)-5-CHO-H4PteGlu.  相似文献

A methanol extract of avocado fruits showed potent inhibitory activity against acetyl-CoA carboxylase, a key enzyme in fatty acid biosynthesis. The active principles were isolated and identified as (5E,12Z,15Z)-2-hydroxy-4-oxoheneicosa-5,12,15-trienyl (1), (2R,12Z,15Z)-2-hydroxy-4-oxoheneicosa-12,15-dienyl (2), (2R*,4R*)-2,4-dihydroxyheptadec-16-enyl (3) and (2R*,4R*)-2,4-dihydroxyheptadec-16-ynyl (4) acetates by instrumental analyses. The IC50 of the compounds were 4.0 x 10(-6), 4.9 x 10(-6), 9.4 x 10(-6), and 5.1 x 10(-6) M, respectively.  相似文献

This report is devoted to study of the hydrocarbon composition of the extract of buds of birch (family Betulaceae). We identified 3,5-dihydroxy-7,4??-dimethoxyflavone, 5,7-dihydroxy-3,4??-dimethoxyflavone, 7-methoxy-4??,5-dihydroxyflavanone, 4??-methoxy-5,7-hydroxyflavanone, 5??,6??-dihydroxycaryophyllen-4(14),8(15)-diene ((1R,5R,6R,9S)-5,6-dihydroxy-11,11-dimethyl-4,8-methylenebicyclo[7.2.0]-undecane), 5??,6??-dihydroxycaryophyllen-3,8(15)-diene ((1R,5R,6R,9S)-5,6-dihydroxy-11,11-dimethyl-8-methylenebicyclo[7.2.0]undec-3-ene), 6??,9??-dihydroxy-??-humulene((1E,4E,6R,9S)-6,9-dihydroxy-4,11,11-trimethyl-8-methylene-1,4-cycloundecanediene), 5??,6??,8??-trihydroxycariolan, and (1R,5R,6R,8R,9S)-trihydroxycariolan. The gas chromatographic retention indices of all identified compounds were determined.  相似文献

Three hydroxylasiodiplodins were isolated from the mycelium extracts of Lasiodiplodia theobromae IFO 31059; and their structures were identified as (3R),(4S)-4-hydroxylasiodiplodin, (3R),(6R)-6-hydroxy-de-O-methyllasiodiplodin and (3R),(5R)-5-hydroxy-de-O-methyllasiodiplodin. Perhexyloligosilanes (R,R)-(+)-MeNpPhSi*(Hex(2)Si)(n)Si*PhNpMe (n = 2; (R,R)-(+)-4a, n = 4; (R,R)-(+)-6a, n = 6; (R,R)-(+)-8a) with chiral methyl(1-naphthyl)phenylsilyl terminals were synthesized and characterized. The absorption wavelengths lambda(max) by (1)L(a,Ph) transition of phenyl chromophore conjugated with oligosilane units in (R,R)-(+)-4a - (R,R)-(+)-8a show bathochromic shift of about 3-4 nm compared with those of the alpha,omega-phenyl substituted perhexyloligosilanes Ph(Hex(2)Si)(m)Ph (m = 4; 4b, m = 6; 6b, m = 8; 8b) having the same silicon chain length. Longer chain length induces the separated lambda(max) of (1)L(a,Ph) from (1)B(b,Np) of naphthyl chromophore with positive exciton chiralities. In (R,R)-(+)-8a, although the extremum wavelengths lambda(ext) of exciton coupling between (1)B(b,Np) and (1)L(a,Ph) are separated by about 80 nm, the compound retains the positive exciton chirality, which provides definite information on the absolute configuration of terminal chiral silicon atoms. Bulky terminal substituents and lowering the temperature affect the conformation of the main chain, inducing extended silicon backbone structure.  相似文献

Reduction of bicyclo[2.2.2]octane-2,6-dione with baker's yeast gave (lR,4S,6S)-6-hydroxybi-cyclo[2.2.2]octan-2-one (95% e.e.) contaminated with 8% of its (1S,4R,6S)-isomer. Similarly, the yeast reduction of 1-methylbicyclo[2.2.2]octane-2,6-dione furnished (1R,4S,6S)-6-hydroxy-1-methylbi-cyclo[2.2.2.]octan-2-one (99.5% e.e.) in 59% yield. The yeast reduction of 4-methylbi-cyclo[2.2.2.]octane-2,6-dione afforded (1R,4S,6S)-6-hydroxy-4-methylbicyclo[2.2.2.]octan-2-one (98% e.e.) contaminated with 3% of its (1S,4R,6S)-isomer in 58% yield.  相似文献

In a continuing research for neolignans from Piper kadsura (Choisy) Ohwi, six benzofuranoid neolignans were isolated from the aerial part of the plant. Their structure determination were based on the spectroscopic analysis (UV, IR, MS, NMR and CD) and derivative synthesis. Three of the isolated compounds were identified as new structures: 7R, 8R, 1′S-△8′-3, 4-methylenedioxy-5′-methoxy-l′, 4′-dihydro-4′-oxo-7, 0, 2′, 8. l′-neolignan ( Ⅰ ), 7 R, 8 R, 1 ′ R- △8′ - 3,4- methylenedioxy- 1 ′- methoxy - 1′,6′- dihydro- 6′- oxo- 7.0.4′,8. 3′-neolignan (Ⅳ) and 7R, 8R, 1′S-△8′-3, 4-methylenedioxy-l′-methoxy-1′,6′-dihydro-6′-oxo-7.0.4′,8.3′-neolignan (Ⅴ). Known compounds among them are 7R, 8S,1′S-△8′-3, 4-methylenedioxy-5′-methoxy-1′, 4′-dihydro-4′-oxo-7. 0. 2′, 8. 1′-neolignan(Ⅱ), 7S, 8S, 1′R-△8′-3, 4, 5′-trimethoxy-1′, 4′-dihydro-4′-oxo-7.0. 2′, 8. 1′-neolignan (Ⅲ) and 75, 85, 1′S-△8′-3, 4, l′-trimethoxy-l′, 6′-dihydro-6′-oxo-7. 0. 4′, 8. 3′-neolignans (Ⅵ). If RNA is extracted from the ribosomes which had been isolated from frozen-thawed tissue of Galleria mellonella, the 28 S RNA, when heated or treated with urea, dissociates into seven different species of polynucleotide fragments. They were designated as R1, R2, R3, R4, R5, R6, and R7, whose molecular weights were estimated to be 1.15x10-6, 0.75x10-6, 0.55x10-6, 0.40x10-6, 0.30x10-6, 0.25x10-6, 0.20x10-6 daltons, respectively. It is likely that R1 and R5 arise from a single nick in original 38 S rRNA. Experiments with isolated R1 suggest that it is made up of a hydrogen-bonded complex of R2 and R4. R5 is a complex of R6 and an unidentified species, X. It is suggested that these fragments result from nicks which are introduced, secondarily, in the phosphodiester bonds by an endogenous endonuclease(s). Since the secondary nicks are limited in number and located in specific points of the molecule, it appears that the reaction is quite specific. It was also shown that the 28 S aphid RNA, which apparently lacks the primary nick, is susceptible to nicking.  相似文献

Four new steroidal saponins, polypunctosides A-D (1-4, resp.), were isolated from the rhizomes of Polygonatum punctatum, together with five known steroidal saponins. On the basis of chemical and spectral evidence, the structures of the new saponins were established as (3beta,23S,25R)-23-(alpha-L-arabinopyranosyloxy)spirost-5-en-3-yl 4-O-(6-deoxy-alpha-L-mannopyranosyl)-D-glucopyranoside (1), (3beta,23S,25R)-23-[(2-O-acetyl-alpha-L-arabinopyranosyl)oxy]spirost-5-en-3-yl 4-O-(6-deoxy-alpha-L-mannopyranosyl)-D-glucopyranoside (2), (3beta,23S,25R)-23-[(3-O-acetyl-alpha-L-arabinopyranosyl)oxy]spirost-5-en-3-yl 4-O-(6-deoxy-alpha-L-mannopyranosyl)-D-glucopyranoside (3), and (3beta,23S,25R)-23-[(4-O-acetyl-alpha-L-arabinopyranosyl)oxy]spirost-5-en-3-yl 4-O-(6-deoxy-alpha-L-mannopyranosyl)-D-glucopyranoside (4). The cytotoxic activity of the isolated saponins was evaluated towards HeLa cells.  相似文献

The (1S,2R,3S,4R)-N(6)-[1-(1,2,3,4-tetrahydro-2,3, 4-trihydroxybenz[a]anthracenyl)]-2'-deoxyadenosyl adduct at X6 of 5'-d(CGGACXAGAAG)-3'.5'-d(CTTCTTGTCCG)-3', incorporating codons 60, 61 (underlined), and 62 of the human N-ras protooncogene, results from trans opening of (1R,2S,3S,4R)-1,2-epoxy-1,2,3, 4-tetrahydrobenz[a]anthracenyl-3,4-diol by the exocyclic N6 of adenine. Two conformations of this adduct exist, in slow exchange on the NMR time scale. A structure for the major conformation, which represents approximately 80% of the population, is presented. In this conformation, an anti glycosidic torsion angle is observed for all nucleotides, including S,R,S,RA6. The refined structure is a right-handed duplex, with the benz[a]anthracene moiety intercalated on the 3'-face of the modified base pair, from the major groove. It is located between S,R,S,RA6.T17 and A7.T16. Intercalation is on the opposite face of the modified S,R,S,RA6.T17 base pair as compared to the (1R,2S,3R,4S)-N6-[1-(1,2,3,4-tetrahydro-2, 3,4-trihydroxybenz[a]anthracenyl)]-2'-deoxyadenosyl adduct, which intercalated 5' to the modified R,S,R,SA6.T17 base pair [Li, Z. , Mao, H., Kim, H.-Y., Tamura, P. J., Harris, C. M., Harris, T. M., and Stone, M. P. (1999) Biochemistry 38, 2969-2981]. The spectroscopic data do not allow refinement of the minor conformation, but suggest that the adenyl moiety in the modified nucleoti111S,R, S,RA6 adopts a syn glycosidic torsion angle. Thus, the minor conformation may create greater distortion of the DNA duplex. The results are discussed in the context of site-specific mutagenesis studies which reveal that the S,R,S,RA6 lesion is less mutagenic than the R,S,R,SA6 lesion.  相似文献

对来自小黑麦与小麦杂种的3个花粉株系,DH220-4,DH220-5和DH220-14进行了形态性状观察,染色体组成分析和抗白粉病鉴定。经过染色体形态和数目观察、原位杂交、C-分带、同工酶等电聚焦和贮藏蛋白的SDS-聚丙烯酰胺凝胶电泳分析,证明其中两个株系,DH220-4和DH220-5是6R/6D代换系,另一个株系DH220-14是1R/1D代换系。经人工接种鉴定,两个6R/6D代换系高抗白粉病。从而进一步证明黑麦的6R染色体上存在抗白粉病的基因。同时还对小麦遗传背景下异源染色体的识别及6R染色体的利用价值等问题进行了讨论。  相似文献

Effects of prostacyclin and carbacyclins on endothelin-induced DNA synthesis were investigated in vascular smooth muscle cells. DNA synthesis was estimated by [3H]thymidine incorporation. Five carbacyclins used in this report were 5-[(1S, 5S, 6R, 7R)-7-hydroxy-6-[(E)-(S)-3-hydroxy-1-octenyl]bicyclo [3.3.0]oct-2-en-3-yl) pentanoic acid (TEI-7165), methyl 5-[(1S, 5S, 6R, 7R)-7-hydroxy-6-[(E)-(S)-3-hydroxy-1-octenyl]bicyclo[3.3.0]oct-2-en-3- yl]pentanoate (TEI-9090), 5-[(1S, 5S, 6R, 7R)-7-hydroxy-6-[(E)-(3S, 5S)-3-hydroxy-5-methyl-1-nonenyl]bicyclo[3.3.0]oct-2-en-3-yl)penta noic acid (TEI-9063), methyl 5-[(1S, 5S, 6R, 7R)-7-hydroxy-6-[(E)-(3S, 5S)-3-hydroxy-5-methyl-1- nonenyl]bicyclo[3.3.0]oct-2-en-3-yl)pentanoate (TEI-1324), 5-[(1S, 5S, 6R, 7R)-7-hydroxy-6-[(E)-(S)-4-hydroxy-4-methyl-1- octenyl]bicyclo[3.3.0]oct-2-en-3-yl] pentanoic acid (TEI-3356). Prostacyclin and the carbacyclins inhibited the endothelin-induced DNA synthesis within the nanomolar range. These results suggest that prostacyclin and carbacyclins are possibly effective in inhibiting the proliferation of vascular smooth muscle cells under some situations in vivo.  相似文献

A new scaffold consisting of a carbocycle and a substituted imidazoline in an orthogonal arrangement was synthesized as a potential specific inhibitor of glycosidases. The spirobicycloimidazoline, (5R,6R,7R,8R)-8-(hydroxymethyl)-2-phenyl-1,3-diazaspiro[4.4]non-1-ene-6,7-diol, was synthesized from methyl 2-O-p-methoxybenzyl-3,4-di-O-benzyl-alpha/beta-D-gluco-6-enopyranoside via (1R,2S,3S,4R,5S)-3,4-bis(benzyloxy)-2-(4-methoxybenzyloxy)-5-vinyl-cyclopentanol. The ring contraction of the 6-enopyranoside in the presence of zirconocene equivalent ('Cp(2)Zr') reagent gave exclusively the corresponding cyclopentanol without cleavage of the PMB protecting group. In the course of the study, a new alpha-mannosidase inhibitor, (1R,2R,3R,5R)-5-amino-3-hydroxymethyl-cyclopentane-1,2-diol, was also discovered.  相似文献
