采用开顶式气室熏蒸法,设置自然条件下臭氧(O3)浓度(对照,约40 nmol·mol-1)、80、160及200 nmol·mol-14个臭氧浓度,观测了不同浓度臭氧条件下银杏叶片可见伤害、活性氧生成量、抗氧化酶活性及相关基因表达变化情况,分析大气臭氧浓度升高对植物活性氧代谢的影响.结果表明: 160和200 nmol·mol-1 O3熏蒸明显伤害银杏叶片,80 nmol·mol-1与对照无差异,无可见伤害.O3处理20 d后,160和200 nmol·mol-1条件下银杏叶片的超氧自由基(O2)产生速率显著高于80 nmol·mol-1和对照,而80 nmol·mol-1与对照无差异;O3处理40 d后,160和200 nmol·mol-1熏蒸下叶片过氧化氢(H2O2)含量显著高于80 nmol·mol-1和对照,而过氧化氢酶(CAT)活性显著高于80 nmol·mol-1和对照,各臭氧处理抗坏血酸过氧化物酶(APX)活性均低于对照.熏蒸40 d后,CAT、APX基因的转录表达持续加强;防御素(GbD)的表达强度则随着臭氧浓度的增加及熏蒸时间的延长而呈显著加强.高浓度臭氧胁迫可使银杏叶片活性氧生成量增加、抗氧化酶活性下降、相关基因表达水平上调,有明显可见叶片伤害.  相似文献

Photosynthetic oxygen exchange has been measured using 18O2 and the mass-spectrometric technique in two mutant strains of Chlamydomonas reinhardtii deficient in electron transport. In the F15 mutant, deficient in PS I, O2 was evolved in the light at a constant rate of about 145 nmol O2/min per mg chlorophyll. At the same time, O2 uptake was increased in the light by about 28%. O2 evolution and the light-stimulation of O2 uptake were inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Antimycin A and salicylhydroxamic acid, both inhibitors of mitochondrial respiration, when added together, inhibited dark respiration and also the light-dependent O2 evolution by about 80%. Similar properties were observed in a mutant strain of Chlamydomonas (F18) lacking the cytochrome b6-f complex. We conclude from these results that in the absence of active Photosystem I, a permanent electron flow can occur in the light from Photosystem II to molecular O2. This electron transfer pathway would involve the plastoquinone pool and the mitochondrial electron transport chain. Because O2 evolution measured in the F15 mutant was severely inhibited by the uncoupler cyanide m-chlorophenylhydrazone, we propose that an energy-dependent reverse electron transfer similar to that of Rhodospirillaceae might occur in the chloroplast of Chlamydomonas.  相似文献

The rate of in vitro Nicotiana tabacum L. “Bel-W3” pollen tube growth was reduced 62 and 44%, respectively, when pollen tubes were exposed to 120 ppb ozone (O3) for 3 hr or 300 μW/cm2 ultraviolet-B (u.v.-B) radiation for 30 min. Petunia hybrida Vilm. “White Cascade” pollen tube growth was reduced 34 and 59%, respectively, upon exposure to O3 or u.v.-B at the above doses. The combination of u.v.-B at 300 μW/cm2 for 30 min, followed by O3 at 120 ppb for 3 hr, reduced pollen tube growth by 79% for “Bel-W3” and 75% for “White Cascade”. The effect appeared to be additive, implying that different target areas may be affected by the two stressors. In the Northeast, plants are exposed to both u.v.-B and O3 during the normal growing season. This may result in an unexpectedly higher stress on the reproductive system than had been previously suspected based on these two stressors acting individually.  相似文献

A mass spectrometer has been used to measure O2 production and consumption and net CO2 exchange near the CO2 compensation point in Chlorella pyrenoidosa and Chlamydomonas reinhardtii grown in air and in 2% (v/v) and 5% (v/v) CO2 in air. Some experiments were run in presence of -hydroxy-2-pyridinemethanesulfonic acid and, for others, data are presented from parallel assays for enzymic glycolate oxidation. Although the glycolate-oxidising enzyme was present in cells grown with and without CO2 enrichment, the activities measured were not compatible with direct measurements of O2 consumption in the light. This and other evidence argues against glycolate being the primary substrate for light-induced O2 consumption by these organisms.  相似文献

Relationships between ozone (O3) tolerance and leaf ascorbic acid concentrations in 03-susceptible (O3S) 'Hark' and O3-resistant (O3R) 'Hood'soybean, Glycine max (L.) Merr., cultivars were examined with high-performance liquid chromatography (HPLC). Leaf samples were analyzed at 4 h intervals during a 24 h period. Soybean cultivars grown in the greenhouse with charcoal filtered (CF) and nonfiltered (NF) air showed daily oscillations in ascorbic acid production. Highest ascorbic acid levels in leaves during light coincided with highest concentrations of photochemical oxidants in the atmosphere at 2:00 p.m. The resistant genotype produced more ascorbic acid in its trifoliate leaves than did the corresponding susceptible genotype. Under CF air (an O3-reduced environment) O3-S and O3-R cultivars showed rhythms in ascorbic acid production. In NF air (an O3 stress environment) the O3-R cultivar alone showed rhythms in ascorbic acid production. Results indicated that superior O3 tolerance in the Hood soybean cultivar (compared with Hark) was associated with a greater increase in endogenous levels of ascorbic acid. Ascorbic acid may scavenge free radicals and thereby protect cells from injury by O3 or other oxyradical products. Plants defend themselves against photochemical oxidant stress, such as O3, by several mechanisms. Experimental evidence indicates that antioxidant defense systems existing in plant tissues may function to protect cellular components from deleterious effects of photochemical oxidants through endogenous and exogenous controls.  相似文献

随着城市化进程的加快, 臭氧(O3)已经成为中国夏季首要大气污染物。已有研究表明O3通过气孔进入叶片显著抑制光合作用, 影响陆地生态系统碳水循环过程。但是O3浓度升高对植物光合和气孔导度模型关键参数影响的研究仍然缺乏。该研究利用开顶式气室, 设置两个O3处理(CF, 过滤空气; E-O3, 未过滤空气+ 60 nmol·mol-1 O3), 选用4种常见的树木(茶(Camellia sinensis)、复叶槭(Acer negundo)、栾树(Koelreuteria paniculata)和蒙古栎(Quercus mongolica)), 通过测定叶片气体交换参数, 探究O3浓度升高对植物光合和气孔导度模型关键参数的影响。结果表明: O3浓度升高显著降低了4种植物的饱和光合速率和光合生化模型参数叶肉导度, 但是O3对光合生化模型参数最大羧化速率和最大电子传递速率的负效应在不同树种间存在差异。此外, 不同植物气孔导度对O3的响应也存在差异。通过对最优化气孔导度模型进行参数化, 结果表明O3显著提高了蒙古栎和复叶槭的斜率参数(g1), 并显著增加了茶的气孔导度模型截距参数(g0), 但降低了复叶槭的g0。在不同O3处理下4种树木的内源水分利用效率与g1呈显著线性负相关关系。综上所述, O3浓度升高显著影响光合生化和气孔导度模型关键参数。  相似文献

Alkaline p-nitrophenylphosphate phosphatase (pNPPase) from the halophilic archaeon Halobacterium salinarum (previously halobium) was solubilized in reversed micelles of cetyltrimethylammonium bromide (CTAB) in cyclohexane with 1-butanol as cosurfactant. The hydrolysis reaction appears to follow Michaelis–Menten kinetics. The dependency of the maximum reaction rate (Vmax) on the water content θ (% v/v) (or ω0 value: molar ratio of water to surfactant concentrations) showed a bell-shaped curve for 0.3 M CTAB, but not for 0.2 M CTAB. The enzyme activity increased with the surfactant concentration at a constant ω0 value (10.27). When the surfactant concentration was increased at a constant θ, the enzyme activity decreased. The enzyme was more stable in reversed micelles than in aqueous media.  相似文献

In this study the response to photoinhibition of photosynthesis and subsequent recovery was examined in plants of Phaseolus vulgaris L. cultivar ‘Pinto’ exposed to charcoal-filtered air or to ozone (O3) at 150 nL L−1 either for 3 h, or for 5 h. The responses were analysed using chlorophyll fluorescence imaging and by conventional fluorometry. Compared to control plants maintained in charcoal-filtered air, in plants exposed for 3 h to O3 and then subjected to high light treatment, the results show an increased tolerance to photoinhibition. Plants exposed to the same O3 concentration but for the longer 5-h period, were not tolerant to the photoinhibition treatment and, instead showed visible symptoms of damage (chlorosis and necrosis) clearly attributable to the longer O3 exposure. Here the detrimental effects of O3 aggravated the effects of the high light photoinhibitory treatment. The leaves exposed to the shorter O3 treatment (150 nL L−1 for 3 h) developed an ability to counteract the negative effects of a high light exposure probably because the O3 had activated an antioxidant system able to protect the photosynthetic machinery.  相似文献

Photostimulation of N2 fixation in the blue-green alga Anabaena cylindrica was investigated using monochromatic light and by comparing action spectra of C2H2 reduction with that of photosynthetic O2 evolution. Maximum nitrogenase activity per unit energy was measured at a wavelength which corresponds to the maximum light absorption of chlorophyll a. The action spectrum of C2H2 reduction indicates a primary involvement of Photosystem I in N2 fixation by blue-green algae.  相似文献

【目的】为科学合理地浇灌臭氧(O3)水防治韭菜迟眼蕈蚊Bradysia odoriphaga幼虫(俗称韭蛆),明确最佳浇灌时期和使用条件。【方法】在平地覆膜环境中浇灌不同浓度(5, 10, 20和30 mg/L)O3水,调查对韭蛆的防治效果;在平地覆膜环境中不同日光强度、3种不同灌溉环境[平地覆膜、小拱棚和小拱棚+草垫]、2种不同灌溉环境[平地覆膜、小拱棚]的不同浇灌频率等条件下分别浇灌30 mg/L O3水,调查对韭蛆的防治效果;调查平地覆膜环境中浇灌30 mg/L O3水对韭菜产量和根系生长的影响;分析日光强度对不同灌溉环境中土壤温度及浇灌O3水对韭蛆的防治效果的影响。【结果】平地覆膜条件下,O3水防治韭蛆的最佳浓度为20~30 mg/L。在平地覆膜条件下浇灌30 mg/L O3水处理中,对韭蛆的防治效果与浇灌当天的日光强度呈正相关;当日光强度超过60 000 lx时,第1天韭蛆的死亡率高达100%;日光强度低于10 000 lx时,对韭蛆无显著的防治效果;日光强度处于中间水平时,前期对韭蛆的防治效果较差,而后期防治效果逐渐增强。土壤升温效果与日光强度呈正相关,也与灌溉环境显著相关,不同灌溉环境下土壤5 cm深处的温度为:平地覆膜>小拱棚>小拱棚+草垫。小拱棚内浇灌O3水对韭蛆的防治效果显著高于平地覆膜组和小拱棚+草垫组。采取连续多次浇灌O3水有利于提高O3水对韭蛆的防治效果,而且小拱棚内韭蛆的防治效果更显著。浇灌O3水时,进水口离田埂末端的距离不能超过40 m。平地覆膜环境中浇灌30 mg/L O3水后,韭菜的株高、重量和须根数量显著高于对照组(浇灌清水);韭菜植株分蘖数与对照组差异不显著。【结论】若能科学合理地浇灌O3水,既能达到防治韭蛆的理想效果,又能促进韭菜生长。  相似文献

采用各种活性氧诱发剂(0.5g/L百草枯、10mmol/L H2O2和10mmol/L H2O2+10mmol/L FeSO4·7H2O)及相应的清除剂(20mmol/L没食子酸丙酯、10个单位CAT和70mmol/L DMSO)、外源乙烯(750μl/L乙烯利)、乙烯生成先导物(合成前体,1mmol/L ACC)和乙烯合成抑制剂(10mmol/L AOA)处理香蕉果肉切片。结果表明,在不同成熟度的香蕉果实中,只有O2·存在及乙烯合成不受抑制时,淀粉酶活性才能呈现,果实才能软化、成熟;而O2·及OH·可能在乙烯合成中所起的作用有所不同。  相似文献

When dark-adapted (5 min in the dark) Chlorella cells were deposited on a bare platinum electrode, treated with DCMU (3-(3,4-dichlorophenyl)-1,1-dimethylurea) and illuminated, O2 was consumed after a lag time of about 250 ms. The comparison of the O2 consumption kinetics with the fluorescence O-I-D-P-S transition (the fast change in chlorophyll fluorescence which occurs after the onset of illumination of dark-adapted algae and is over within 2 s) observed in untreated algae indicates that no O2 is consumed during the fluorescence rise and that O2 uptake is initiated approximately when the maximum level of fluorescence P is reached. Mass spectrometry measurements of O2 exchange (using 18O2) were performed during dark to light transition with DCMU-untreated Chlorella cells. Under these conditions, O2 reduction began after a lag time (about 200–400 ms) and stopped after about 5 s of illumination. The above experiments clearly show that the reduction of O2 starts nearly at the same time that the fluorescence P-S decline. On the other hand, we show that the reduction of CO2 does not interfere in the fluorescence O-I-D-P-S transient. We found the same apparent affinity for O2 (about 57 μM) for both the fluorescence P-S decline and the reduction of O2. At least three consecutive short (2 μs) saturating flashes were required to affect the fluorescence transient significantly and also to induce a significant uptake of O2. Moreover, parabenzoquinone, an artificial Photosystem I electron acceptor, inhibited both the fluorescence D-P rise and the 250 ms lag time observed in the reduction of O2. We conclude from the above results that in the early stages of the illumination of dark-adapted algae, some Photosystem I electron acceptors are in an inactive form. In this form, the electron transport chain is unable to reduce either O2 or CO2. This would lead to the accumulation of electrons on the Photosystem II acceptors (principally QA and the plastoquinone pool) and therefore explains the fluorescence D-P rise. The light activation, probably achieved through the reduction of at least two electron acceptors, first allows the reduction of O2, and therefore explains the P-S fluorescence decline. By accepting electrons before the site of regulation and mediating rapid O2 reduction, parabenzoquinone avoids the accumulation of electrons and therefore inhibits the D-P fluorescence rise.  相似文献

Influence of oxygen on the electron transfers of photosynthesis. II. Influence of very low oxygen concentration on the NADP+ reduction by isolated chloroplasts

The influence of very low O2 concentration on the NADP+ reduction by isolated spinach chloroplasts has been studied.

The results show that in the presence of very low O2 concentration (< 0.3%) NADP+ reduction is partially inhibited. This inhibition may be partially reversed under some conditions, especially when, in spite of the presence of an O2 trap (glucose plus glucose oxidase (EC 1.1.3.4)) an O2 evolution is observed.  相似文献

The interaction of chlorophyll a with superoxide anion was studied in an alkaline DMSO-water system. It was found that O2, directly or via HO2, produces the chlorophyll enolate-ion (Molish's intermediate) that is oxidized to Mg-chlorine(s). The allomerization reaction was found to be inhibited by super oxide dismutase. A possible participation of oxygen radicals in chlorophyll degradation in plants is discussed.  相似文献

为了给全球变化背景下的竹林经营应对策略提供理论依据,运用开顶式同化箱(OTCs)模拟4个大气O3浓度,分别为环境背景大气(40~45 nl·L-1)、降低1/2(22~25 nl·L-1)、倍增1倍(92~106 nl·L-1)和倍增2倍(142~160 nl·L-1),研究分析四季竹(Oligostachyum lubricum(Wen) King f)对大气O3胁迫的光合生理响应规律.结果表明:随着大气O3浓度的升高,四季竹叶片光合色素含量呈降低趋势,倍增1倍处理的大气O3浓度是显著变化的节点;大气O3浓度变化对四季竹Pn日变化影响复杂,环境背景大气、倍增2倍处理呈"单峰"曲线,而降低1/2、倍增1倍处理呈"双峰"曲线.对Tr日变化无明显影响,各处理均呈"单峰"曲线;大气O3浓度较环境背景大气升高或降低,四季竹的光合生理响应都表现为伤害效应.当大气O3浓度倍增1倍及以上,对四季竹会造成严重的伤害,表现为叶片光合色素降解或合成受阻,水分利用效率降低,光合作用能力明显下降.大气O3胁迫对四季竹光合作用的影响表现为非气孔因素限制.  相似文献

主要探讨冬枣(Ziziphus jujuba Mill.cv Dongzao)在-1℃的动态气调(CA-Ⅰ,70%O2 0%CO2处理7天,然后转入5%O2 0%CO2中)、普通气调(CA-Ⅱ,5%O2 0%CO2;CA-Ⅲ,10%O2 0%CO2)及普通冷藏和常温(25℃)等条件下,果实发病率、色素、可溶性固形物、可滴定酸、乙醇和乙酸乙酯含量等的变化.结果表明:与普通冷藏相比,气调贮藏能减缓果实的腐烂,抑制色素的分解和减少果肉中乙醇、乙酸乙酯的含量.动态高氧处理能有效地保持果实的颜色,抑制色素降解及果皮褐变.CA-Ⅲ(10%O2 0%CO2)能有效地减少果肉乙醇的含量而CA-Ⅱ(5%O2 0%CO2)能有效地减少果肉乙酸乙酯的含量.气调贮藏的果实可滴定酸及可溶性固形物含量与其他处理的果实没有明显的差异.  相似文献

1. The methyl viologen-catalyzed Mehler reaction was investigated in intact cells of five species of blue-green algae and Chlamydomonas reinhardi.

2. In the presence of methyl viologen, all the blue-green algae except Anabaena flos-aquae show a light-dependent O2 consumption as well as a post-illumination O2 evolution. The rate of O2 consumption is stimulated by 1 mM KCN, an inhibitor of catalase, but the dark O2 evolution becomes suppressed.

3. A. flos-aquae shows a light-dependent methyl viologen-catalyzed O2 uptake which is not affected by 1 mM KCN. Furthermore, there is no release of O2 in the dark following illumination.

4. With C. reinhardi, the cells do not show any net O2 exchange during or after illumination. Addition of 1 mM KCN, however, results in an immediate O2 uptake in the light.

5. Based on the mechanism postulated for the Mehler reaction in isolated chloroplasts, it was deduced that the differences in the kinetics of the O2 exchange catalyzed by methyl viologen reflect differences in the endogenous catalase activity in these algae. Cells of A. flos-aquae are deficient in catalase activity whereas those of the other blue-green algae possess catalase, although at low activity. C. reinhardi, on the other hand, has high catalase activity in vivo.

6. These findings are corroborated by results obtained from O2 electrode measurements of catalase activity in cell-free extracts of these algae.

The formation of nitrite reductase and cytochrome c in Micrococcus denitrificans was repressed by O2. The purified nitrite reductase utilized reduced forms of cytochrome c, phenazine methosulphate, benzyl viologen and methyl viologen, respectively, as electron donors. The enzyme was inhibited by KCN, NaN3 and NH2OH each at 1 mM, whereas CO and bathocuproin, diethyl dithiocarbamate, o-phenanthroline and ,'-dipyridyl at 1 mM concentrations were relatively ineffective. The purified enzyme contains cytochromes, probably of the c and a2 types, in one complex. A Km of 46 μM for NO2 and a pH optimum of 6.7 were recorded for the enzyme. The molecular weight of the enzyme was estimated to be around 130000, and its anodic mobility was 6.8·10−6 cm2·sec−1·V−1 at pH 4.55.

The most highly purified nitrite reductase still exhibited cytochrome c oxidase activity with a Km of 27 μM for O2. This activity was also inhibited by KCN, NaN3 and NH2OH and by NO2.

A constitutive cytochrome oxidase associated with membranes was also isolated from cells grown anaerobically with NO2. It was inhibited by smaller amounts of KCN, NaN3 and NH2OH than the cytochrome oxidase activity of the nitrite reductase enzyme and also differed in having a pH optimum of about 8 and a Km for O2 of less than 0.1 μM. Spectroscopically, cytochromes b and c were found to be associated with the constitutive oxidase in the particulate preparation. Its activity was also inhibited by NO2.

The physiological role of the cytochrome oxidase activity associated with the purified nitrite reductase is likely to be of secondary importance for the following reasons: (a) it accounts for less than 10% of total cytochrome c oxidase activity of cell extracts; (b) the constitutive cytochrome c oxidase has a smaller Km for O2 and would therefore be expected to function more efficiently especially at low concentrations of O2.  相似文献

通过开顶箱(OTCs)模拟,以环境臭氧(O3)浓度约40 nmol·mol-1为对照,研究大气O3浓度升高(80和160 nmol·mol-1O3)对冷季型草坪草高羊茅生长、亚细胞结构及其活性氧代谢的影响.结果表明: 14 d的80 nmol·mol-1O3熏蒸使高羊茅株高和叶宽降低,总生物量降低43.7%,老叶变黄,而160 nmol·mol-1O3处理高羊茅叶出现大量枯死褐斑,叶尖坏死,新叶卷曲,总生物量降低46.2%,叶肉细胞膜卷曲,叶绿体和线粒体受损严重.与对照相比,80和160 nmol·mol-1O3熏蒸下高羊茅叶片超氧阴离子(O2)产生速率、过氧化氢(H2O2)和丙二醛(MDA)含量显著增加,抗氧化酶活性显著升高,但叶片总酚含量和抗氧化能力随O3浓度升高而先升高后降低.在明显O3伤害症状出现之前,O3已对高羊茅的生长和抗氧化代谢产生不利影响;高羊茅抗氧化系统虽对O3浓度的升高存在一定的适应性反应,但其不能抵御过高浓度的长期胁迫和伤害.  相似文献

Low-temperature photodissociation spectra of membranes from the thermophile PS3 reveal cytochromes o and a3. The latter reacts with O2 at −103°C to give a light-insensitive compound(s), but the initial stages of O2 binding to cytochrome o could not be studied under these conditions. Photochemical action spectra identify cytochromes a3 and o, but not a CO-binding c-type cytochrome, as functional terminal oxidases in this bacterium.  相似文献
