1. Hibernating C. lateralis were exposed to lowered ambient temperatures in order to investigate the relationship between hibernation stress and the thermoregulatory responses of the animals. 2. The least hibernation-stress squirrels exhibited a passive decline in metabolic rate until their body temperatures stabilized close to microenvironmental temperature. 3. The most stressed individuals aroused from hibernation in response to the declining ambient temperatures. 4. Intermediately stressed animals demonstrated an initial passive decline in temperature; however, at various temperatures (0.3-6.8 degrees C), this group increased their metabolic rate but did not arouse from hibernation.  相似文献

  • 1.1. It is shown that Ca2+-dependent activation of respiration of liver mitochondria from hibernating ground squirrels is accompanied by mitochondrial swelling.
  • 2.2. The swelling of mitochondria from hibernating ground squirrels, as well as the activation of mitochondrial respiration, is precluded by cyclosporin A, p-bromphenacylbromide and oligomycin. Carboxyatractiloside, on the contrary, under these conditions favors the swelling and the acceleration of respiration.
  • 3.3. It was concluded that Ca2+-dependent activation of hibernating ground squirrel liver mitochondrial respiration resulted from the appearance of a non-specific permeability pathway and from swelling of mitochondria.

TNF production has been studied in peritoneal macrophages and splenic T cells of Arctic Yakutian ground squirrel (Citellus Undulatus Pallas) in hibernating and awake animals in winter and in prehibernating autumn as well as in active euthermic spring-summer animals. A high level of TNF production in macrophages of ground squirrel is observed over the active period and during arousals in winter. There are no significant season variations in TNF production in splenic T lymphocytes of ground squirrels. This suggests the major role of activated macrophages in the arousals of hibernating animals. T lymphocyte proliferation in ground squirrels in the active period is higher than in winter, and the most significant seasonal variations are found in T cell mitogenic response, which increases in spring-summer period. Evidence is presented that functional activity of macrophages of squirrel in autumn has much in common with that in winter rather than in spring-summer period.  相似文献

Dolichyl pyrophosphate N-acetyl[14C]glucosamine was synthesized after incubation of liver microsomes from hibernating ground squirrels with UDP-N-acetyl[14C )glucosamine. The radioactivity of glycolipid formed by liver microsomes from hibernating ground squirrels was about 2-fold greater than by liver microsomes from active animals. Addition of exogenous dolichyl phosphate to the incubation mixture increased the formation of dolichyl pyrophosphate N-acetyl[14C]glucosamine by microsomes from both active and hibernating ground squirrels about 6 times. Liver microsomes from hibernating ground squirrels converted dolichyl pyrophosphate N-acetyl[14C]glucosamine into dolichyl pyrophosphate N,N'-diacetyl[14C]chitobiose in the presence of unlabelled UDP-N-acetylglucosamine. This conversion was maximal at 1.0 M concentration of unlabelled UDP-N-acetylglucosamine. The level of dolichyl phosphate assessed by the level of dolichyl pyrophosphate N-acetylglucosamine formation was nearly 2 times greater in liver microsomes from hibernating ground squirrels than from active animals.  相似文献

1. The rates of oxidation of various substrates (beta-hydroxybutyrate, succinate, ascorbate + TMPD) and the rate of ATP synthesis in liver mitochondria from active and hibernating ground squirrels were measured. 2. It was shown that the rate of mitochondrial respiration is significantly lower in hibernating animals than in active animals. 3. The degree of inhibition of mitochondrial respiration in hibernating ground squirrels was found to correlate with the length of the respiratory chain fragment involved in the oxidation of a given substrate. 4. The inhibition of mitochondrial respiration in hibernating animals was accompanied by a decrease in the rate of ATP synthesis. 5. The activity of phospholipase A2 in liver mitochondria from hibernating ground squirrels was found to be decreased. The activation of phospholipase A2 by Ca2+ ions eliminated the inhibition of respiration almost completely. 6. It was assumed that the inhibition of mitochondrial respiration during hibernation is (a) related to the suppression of phospholipase A2 activity and (b) caused by the reduced rates of electron transport through the respiratory chain and/or of substrate transport across the mitochondrial membrane.  相似文献

Summer hibernation induced in ground squirrels (Citellus tridecemlineatus) by urine or plasma from hibernating bats (Myotis lucifugus or Eptesicus fuscus). Summer hibernation in the thirteen-lined ground squirrel can be induced by intravenous injection of urine or blood plasma previously isolated from winter hibernating little brown bats (M. lucifugus) or big brown bats (E. fuscus). Urine- and plasma-injected ground squirrels kept at 8 °C hibernated earlier, longer, and deeper (as indicated by core temperature and respiratory rate measurements) than control ground squirrels injected with saline. This successful cross-order induction of hibernation demonstrates that the hibernation-inducing trigger (HIT) may be present in nonrodent mammals.  相似文献

Thyroidal iodine, protein and thyroglobulin (TG) were investigated in a hibernator, Spermophilus richardsoni, sampled in the field and from laboratory-held winter colonies. In field animals, thyroidal iodine and protein contents were similar at onset of hibernation and at terminal arousal. Iodine content was increased in laboratory-held animals, a function of alimentary supply, but no differences were observed between hibernators and non-hibernators. Density sucrose gradients showed that approx. 20% of the TG was present as the 12S precursor sub-unit. No variations in the iodine content, nor the iodoamino acid composition of the TG occurred as a function of hibernation.  相似文献

Oocytes of primordial, primary, growing, and Graaf follicles of the ovaries of 96 ground squirrels (Citellus citellus L.) were studied during the four seasons of the year. Multilamellar bodies in the oocytes were observed by electron microscopy. The bodies underwent changes during the follicle growth. The vesicles included in the multilamellar bodies were filled with an amorphous substance, while in Graaf follicles trapezoidal, rhomboidal, and hexagonal inclusions were found. The primordial follicle of the oocyte had no Balbiani body. In the hibernating ground squirrel the functions of the Balbiani body were probably performed by the multilamellar bodies, which were considered to be analogous to the Balbiani body.  相似文献

The effect of insulin (0.1-100 nM) on isometric force of contraction in isolated ground squirrel papillary muscle was investigated. In summer, autumn and winter active animals, insulin had a negative inotropic effect on papillary muscles, decreasing the amplitude of contraction by about 30% of the control value. In some cases, predominantly in the summer group of animals, insulin produced different effects on contractility: low doses (0.1-0.5 nM) caused a transient activation of isometric contraction by about 10-15% of control, whereas high doses produced a negative inotropic effect by about 30% of the control level. During deep hibernation (at 5-6 degrees C of heart temperature) and during arousal from hibernation (from 3 to 20 degrees C), insulin had no significant effect on contractility. Opposite inotropic effects of insulin at concentrations of 0.1-50 nM were found during arousal: from 26 to 31 degrees C of heart temperature--a positive inotropic effect by about 20-25% of control, and from 32 to 36 degrees C--a negative one by about 30-40% of the control value.  相似文献

In an endeavor to more clearly define the physiological action of hibernation trigger (previously identified by the authors) an experiment was devised using infant ground squirrels. In one case, pregnant ground squirrel mothers were transfused intravenously with cold dialysate of serum of hibernating woodchuck, and in another case, infant (1 week to 3 week) squirrels were transfused intraperitoneally with the same material. When isolated and placed in a cold room (15 °C) at 6 weeks post partum, both the infant born of the transfused mothers, as well as the infant which has been transfused directly, hibernated in the summer. This was in contrast to controls of several kind, i.e., animals not injected, or receiving active or arousing woodchuck serum or heat-treated hibernation serum. Some details for a theory, which had been advanced in a previous article (4), were essentially substantiated by this experiment. It is our belief that this represents the first time mammalian hibernation has been observed in infant hibernators.  相似文献

The ligand-binding characteristics (B(max) and K(D)) of alpha(1)- and beta(1)/beta(2)-adrenoceptors were investigated in membranes prepared from brown adipose tissue (BAT) of warm-acclimated, cold-acclimated, hibernating and arousing ground squirrels (Spermophillus undulatus) and hamsters (Mesocricetus auratus) by specific binding of [(3)H]prazosin and [(3)H]CGP-12177, respectively. The physiological state did not change the affinity for the adrenoceptors in the BAT of ground squirrels and hamsters. There was a significant decrease in alpha(1)-receptor density in arousing ground squirrels and a significant decrease in beta(1)/beta(2) density in hibernating ground squirrels. The level of alpha(1)-receptors was in all conditions higher than that of beta(1)/beta(2) receptors. The results indicate a possible change in balance of adrenoceptor density in the processes of cold acclimation, hibernation and arousal. The balance between the various adrenoceptor subtypes may be important for the final effect of catecholamines in BAT in different physiological states.  相似文献

Incubation of liver microsomes from hibernating ground squirrel with GDP-[14C]mannose and exogenous dolichyl phosphate resulted in the synthesis of dolichyl phosphate [14C]mannose. The mannosyltransferase activity was about 3-fold higher in microsomes from hibernating ground squirrels than in those from active animals. Incubation for 30 min of liver microsomes from hibernating animals with dolichyl pyrophosphate N,N'-diacetyl-[14C]chitobiose and GDP-[14C]mannose led to the synthesis of lipid-[14C]trisaccharide. When liver microsomes were incubated with lipid-[14C]trisaccharide and unlabelled GDP-mannose, lipid-tetra- to heptasaccharides were discovered in the chloroform-methanol (2:1) extract. Since, under the experimental conditions, negligible synthesis of dolichyl phosphate mannose was observed, it was assumed that GDP-mannose was a donor of mannose in the conversion of lipid-trisaccharide into lipid-oligosaccharides containing 2-5 mannose residues.  相似文献
