The main objective of the present study is to test the effect of short preexposure of lentil (Lens culinaris Medik.) plants to low salt concentration on shoot growth, oxidative stress and activity of antioxidant enzymes under high salt stress. To fulfill this objective, lentil plants were pretreated with 10 mM NaCl for 3 days and then they were exposed to high salt concentration of 300 mM for 7 days. After that, shoot growth was evaluated in terms of shoot length, fresh and dry weight. Biochemical changes in terms of oxidative stress and activity of antioxidant enzymes were also assessed in lentil plants. The shoot growth of lentil plants preexposed to low salt concentration was significantly enhanced under high salt stress, whereas it was severely retarded in lentil plants that were directly exposed to high salt stress. Moreover, lipid peroxidation and the accumulation of hydrogen peroxide were highly reduced in the shoot of lentil plants preexposed to low salt concentration. The activity of antioxidant enzymes (catalase and superoxide dismutase) was significantly higher in the shoot of lentil plants preexposed to low salt concentration than those directly exposed to high salt concentration. Overall, the results revealed an enhanced salt tolerance in lentil plants after short exposure to low salt concentration.  相似文献

Chickpea plants were subjected to salt stress for 48 h with 100 mM NaCl, after 50 days of growth. Other batches of plants were simultaneously treated with 0.2 mM sodium nitroprusside (NO donor) or 0.5 mM putrescine (polyamine) to examine their antioxidant effects. Sodium chloride stress adversely affected the relative water content (RWC), electrolyte leakage and lipid peroxidation in leaves. Sodium nitroprusside and putrescine could completely ameliorate the toxic effects of salt stress on electrolyte leakage and lipid peroxidation and partially on RWC. No significant decline in chlorophyll content under salt stress as well as with other treatments was observed. Sodium chloride stress activated the antioxidant defense system by increasing the activities of peroxidase (POX), catalase (CAT) superoxide dismutase (SOD) and ascorbate peroxidase (APX). However no significant effect was observed on glutathione reductase (GR) and dehydro ascorbate reductase (DHAR) activities. Both putrescine and NO had a positive effect on antioxidant enzymes under salt stress. Putrescine was more effective in scavenging superoxide radical as it increased the SOD activity under salt stress whereas nitric oxide was effective in hydrolyzing H2O2 by increasing the activities of CAT, POX and APX under salt stress.  相似文献

To investigate whether paraquat (PQ) is involved in regulation of antioxidant enzymes and lipid peroxidation under short-term salt stress, and to elucidate the physiological mechanism of salt stress mitigated by PQ, a cucumber cultivar (cv. Chunguang no. 2) was exposed to 100 mM NaCl for 48 h after pre-treatment with 10 μM PQ for 1 h. When compared to the control, salt stress increased the levels of malonaldehyde (MDA), superoxide radical (O2·−) and hydrogen peroxide (H2O2) and the activities of antioxidant enzymes, such as superoxide dismutase (EC 1.15.1.1), ascorbate peroxidase (EC 1.11.1.11) and glutathione reductase (EC 1.6.4.2) in the cucumber leaves. Under salt conditions, PQ pre-treatment prevented oxidative stress as observed by the decreases in MDA, H2O2 and O2·− that correlated with the increase in antioxidant defenses. We propose that, at low concentrations, the PQ pre-treatment can reduce the salt-induced oxidative damage by increasing the antioxidative mechanisms in cucumber plants.  相似文献

Reactive oxygen species play a key role in cancer development by inducing and maintaining the oncogenic phenotypes of cancer cells. In this study, we examined lipid peroxidation and antioxidant enzymes activities in the blood and in the tumor of nasopharyngeal carcinoma patients. Plasma malondialdehyde, conjugated dienes, erythrocytes catalase, and superoxide dismutase activities have been measured in 30 untreated nasopharyngeal carcinoma patients and 30 controls on one hand. On the other hand, tumor malondialdehyde level, catalase, and superoxide dismutase activities have been measured in five nasopharyngeal carcinoma patients and compared with four controls. The lipid peroxidation was confirmed in the plasma by the high levels of malondialdehyde and conjugated dienes (p?<?0.001, respectively). Additionally, significantly higher concentrations of malondialdehyde were found in biopsies compared to the control group (p?<?0.001). In erythrocytes, superoxide dismutase activity was higher in patients than in controls (p?<?0.05), while it was unchanged in the tumor (p?>?0.05). Both erythrocytes and tumor catalase activities were significantly lower in patients than in controls (p?<?0.001, respectively). Statistical studies have shown a positive correlation between malondialdehyde level and IgA antibodies level against Epstein–Barr virus capsid antigen (p?<?0.05). In conclusion, we reported the presence of an oxidative stress in the blood and in the biopsies of nasopharyngeal carcinoma patients where Epstein–Barr virus seems to play a role.  相似文献

An experiment was conducted to evaluate the influence of Glomus intraradices colonization on the activity of antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (PX), ascorbate peroxidase (APX), and glutathione reductase (GR)] and the accumulation of nonenzymatic antioxidants (ascorbic acid, α-tocopherol, glutathione, and carotenoids) in roots and leaves of fenugreek plants subjected to varying degrees of salinity (0, 50, 100, and 200 mM NaCl) at two time intervals (1 and 14 days after saline treatment, DAT). The antioxidative capacity was correlated with oxidative damage in the same tissue. Under salt stress, lipid peroxidation and H2O2 concentration increased with increasing severity and duration of salt stress (DoS). However, the extent of oxidative damage in mycorrhizal plants was less compared to nonmycorrhizal plants. The study reveals that mycorrhiza-mediated attenuation of oxidative stress in fenugreek plants is due to enhanced activity of antioxidant enzymes and higher concentrations of antioxidant molecules. However, the significant effect of G. intraradices colonization on individual antioxidant molecules and enzymes varied with plant tissue, salinity level, and DoS. The significant effect of G. intraradices colonization on antioxidative enzymes was more evident at 1DAT in both leaves and roots, while the concentrations of antioxidant molecules were significantly influenced at 14DAT. It is proposed that AM symbiosis can improve antioxidative defense systems of plants through higher SOD activity in M plants, facilitating rapid dismutation of O2 - to H2O2, and subsequent prevention of H2O2 build-up by higher activities of CAT, APX, and PX. The potential of G. intraradices to ameliorate oxidative stress generated in fenugreek plants by salinity was more evident at higher intensities of salt stress.  相似文献

The role of mannitol as an osmoprotectant, a radical scavenger, a stabilizer of protein and membrane structure, and protector of photosynthesis under abiotic stress has already been well described. In this article we show that mannitol applied exogenously to salt-stressed wheat, which normally cannot synthesize mannitol, improved their salt tolerance by enhancing activities of antioxidant enzymes. Wheat seedlings (3 days old) grown in 100 mM mannitol (corresponding to −0.224 MPa) for 24 h were subjected to 100 mM NaCl treatment for 5 days. The effect of exogenously applied mannitol on the salt tolerance of plants in view of growth, lipid peroxidation levels, and activities of antioxidant enzymes in the roots of salt-sensitive wheat (Triticum aestivum L. cv. Kızıltan-91) plants with or without mannitol was studied. Although root growth decreased under salt stress, this effect could be alleviated by mannitol pretreatment. Peroxidase (POX) and ascorbate peroxidase (APX) activities increased, whereas superoxide dismutase (SOD), catalase (CAT), and glutathione reductase (GR) activities decreased in Kızıltan-91 under salt stress. However, activities of antioxidant enzymes such as SOD, POX, CAT, APX, and GR increased with mannitol pretreatment under salt stress. Although root tissue extracts of salt-stressed wheat plants exhibited only nine different SOD isozyme bands of which two were identified as Cu/Zn-SOD and Mn-SOD, mannitol treatment caused the appearance of 11 different SOD activity bands. On the other hand, five different POX isozyme bands were determined in all treatments. Enhanced peroxidation of lipid membranes under salt stress conditions was reduced by pretreatment with mannitol. We suggest that exogenous application of mannitol could alleviate salt-induced oxidative damage by enhancing antioxidant enzyme activities in the roots of salt-sensitive Kızıltan-91.  相似文献

In the present study, four endophytic fungi (GM-1, GM-2, GM-3, and GM-4) were tested for their ability to improve soybean plant growth under salinity stress conditions. The seed germination and plant growth were higher in seeds pretreated with endophytic fungal cultures than their controls. The positive influence of fungi on plant growth was supported by gibberellins analysis of culture filtrate (CF), which showed wide diversity and various concentrations of GAs. Specifically, GA4, GA7, GA8, GA9, GA12, and GA20 were found in fungal CFs. Under salinity stress conditions, GM-1 significantly enhanced the length and fresh weight of soybean plants relative to other fungal treatments. GM-1 effectively mitigated the adverse effects of salinity by limiting lipid peroxidation and accumulating protein content. GM-2, GM-3, and GM-4 also counteracted the salinity induced oxidative stress in soybean plants through reduction of lipid peroxidation and enhancement of protein content, maintaining the length and fresh weight of shoots. The activities of the antioxidant enzymes catalase, superoxide dismutase and peroxidase were inhibited in salinity exposed plants, while GM-1 significantly enhanced these antioxidant enzyme activities in plants under salt stress. GM-1 treatment also showed lower levels of abscisic acid and elevated levels of salicylic acid in plants under salinity stress. Hence, GM-1 was identified as Fusarium verticillioides (teleomorph Gibberella moniliformis) isolate RK01 based on its DNA sequence homology. These results suggest that endophytic fungal (F. verticillioides) pre-treatment of soybean seeds would be an effective method to promote soybean plant growth under salinity stress conditions.  相似文献

Plant exhibits various patterns of survival under salinity and their growth and development depend on their capacity to overcome the stress. Present investigation was focused on the response and regulation of the antioxidant defense system and the level of lipid peroxidation in Panicum miliacium and Panicum sumatrense under salt treatments. NaCl stress was imposed for 20 days after sowing of two Panicum species. The changes in the antioxidant enzyme activity like superoxide dismutase, catalase, peroxidase, and ascorbate peroxidase and the rate of lipid peroxidation level in terms of malondialdehyde (MDA) were recorded in both Panicum species. A great correlation exists between the antioxidant enzymes and lipid peroxidation. The defense mechanism activated in Panicum species studied was confirmed by the increased antioxidant enzyme activities under progressive NaCl stress. MDA content remained close to control at moderate NaCl concentrations and increased at higher salinities. Although lipid peroxidation increased in both Panicum species under salt stress the percent of increase was low in P. sumatrense indicating its salt-tolerant nature. Another possible conclusion is that improved tolerance to salt stress may be accomplished by increased capacity of antioxidative system.  相似文献

The present investigation evaluates the changes in the levels of antioxidant enzymes, lipid peroxidation (LPO), and protein carbonyl content (PCC) in brain mitochondria following thiamine deficiency (TD). The study was carried out on Mus musculus allocated into three groups, namely control and thiamine-deficient group for 8 (TD 8) and 10 (TD 10) days. The LPO was measured in terms of reduced glutathione (GSH) and thiobarbituric acid reactive substance (TBARS). Antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were measured biochemically. A significant increase in the TBARS (p?<?0.0001) and PCC (p?<?0.001) levels in group II (TD 8) and group III (TD 10) animals was observed in comparison to controls. The GSH levels were found to be reduced in both the treated groups compared to the control. A significant reduction in the activities of SOD was also observed in group II (p?<?0.01) and group III (p?<?0.0001) animals in comparison to the control. Enzymatic activities of CAT (p?<?0.001) and GPx (p?<?0.05) were found to be significantly reduced in group III (TD 10) in comparison to the control. In conclusion, reduction in the activities of antioxidant enzymes as well as an increase in LPO and PCC following TD implies oxidative stress in brain mitochondria that may further leads to neurodegeneration.  相似文献

Abstract

The effects of increasing salt concentrations on the growth, electrolyte leakage, lipid peroxidation, and major antioxidant enzyme activities (superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase) of borage plants were investigated. Plants were grown in half strength of Hoagland nutrient solution added with 0, 25, 50, and 75 mM of NaCl. Most measured parameters were affected by salinity. Increasing salt levels caused a significant reduction in leaf area, stem length, stem diameter, flower number, and dry masses of different organs. Growth of borage plants, in terms of dry weight, was affected. As a consequence of salinity stress, lipid peroxidation and membrane permeability was increased. Antioxidant activity showed an increase in the activity of superoxide dismutase, a non-induced activity of catalase and ascorbate peroxidase, and a slight increase in glutathione reductase activity. The results indicate that borage plants appear to be sensitive to salt stress, since enzymes related to antioxidant enzymatic defense system in treated leaves should be highly active.  相似文献

Researches have reported that reactive oxygen species (ROS)-induced oxidative stress plays an important role in cell cryodamage during cryopreservation. In the current study, pollen from Magnolia denudata and Paeonia lactiflora ‘Zi Feng Chao Yang’ was cryopreserved and incubated with exogenous catalase (CAT) and malate dehydrogenase (MDH) immediately after thawing. The effect of CAT and MDH on the germination of cryopreserved pollen was measured. Based on that, the ROS level, lipid peroxidation and antioxidants activities in fresh pollen, cryopreserved pollen added with or without CAT or MDH were determined to investigate their relationship with oxidative stress. Pollen from Magnolia and Paeonia showed a significant loss of germination, but marked increase of ROS and malondialdehyde (MDA) production after cryostorage. Antioxidant profiles in them were also enhanced. CAT and MDH addition increased the post-LN pollen germination of Magnolia and Paeonia significantly. Their germination rate achieved the highest with 100 IU ml?1 MDH and 400 IU ml?1 CAT application, respectively. Compared to their untreated controls, ROS and MDA accumulation reduced significantly in cryopreserved Magnolia pollen treated with 100 IU ml?1 MDH, while superoxide dismutase (SOD) activity improved markedly. In the case of Paeonia, significantly lower level of ROS and MDA, but higher activity of CAT and SOD were observed in cryopreserved pollen treated with 400 IU ml?1 CAT. In conclusion, pollen deterioration after cryopreservation is associated with ROS-induced oxidative stress. Exogenous CAT and MDH can reduce the oxidative damage through the activity stimulation of antioxidant enzymes, and play a protective role in the pollen during cryopreservation.  相似文献

以大豆种子、幼苗为试验材料,采用砂培的方法,研究了0.2mmol·L-1外源水杨酸(SA)对100mmol·L-1 NaCl胁迫下大豆种子萌发、幼苗形态及生物量、膜脂过氧化和抗氧化酶活性的影响。结果显示:NaCl胁迫下,大豆种子萌发和幼苗生长受到显著抑制,且随着胁迫时间的延长(0~3d),大豆幼苗相对电解质渗漏率、硫代巴比妥酸活性产物(TBARS)含量显著升高,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)活性均明显降低。外源SA促进NaCl胁迫下大豆种子萌发和根茎生长,增加幼苗生物量积累,降低幼苗叶片相对电解质渗漏率和TBARS含量,增强其叶片SOD、CAT、APX活性。研究表明,NaCl胁迫能显著抑制大豆种子萌发和幼苗生长,而一定浓度的外源SA能有效提高NaCl胁迫下大豆种子活力及幼苗抗氧化酶活性,减轻膜脂过氧化程度,缓解NaCl胁迫所造成的伤害,提高大豆幼苗抗盐胁迫的能力。  相似文献

The levels of blood lipid peroxidation, glutathione peroxidase, reduced glutathione, and vitamin C were used to follow the level of oxidative damage caused by 2.45 GHz electromagnetic radiation in rats. The possible protective effects of selenium and L-carnitine were also tested and compared to untreated controls. Thirty male Wistar Albino rats were equally divided into five groups, namely Groups A1 and A2: controls and sham controls, respectively; Group B: EMR; Group C: EMR + selenium, Group D: EMR + L-carnitine. Groups B–D were exposed to 2.45 GHz electromagnetic radiation during 60 min/day for 28 days. The lipid peroxidation levels in plasma and erythrocytes were significantly higher in group B than in groups A1 and A2 (p?<?0.05), although the reduced glutathione and glutathione peroxidase values were slightly lower in erythrocytes of group B compared to groups A1 and A2. The plasma lipid peroxidation level in group A2 was significantly lower than in group B (p?<?0.05). Erythrocyte reduced glutathione levels (p?<?0.01) in group B; erythrocyte glutathione peroxidase activity in group A2 (p?<?0.05), group B (p?<?0.001), and group C (p?<?0.05) were found to be lower than in group D. In conclusion, 2.45 GHz electromagnetic radiation caused oxidative stress in blood of rat. L-carnitine seems to have protective effects on the 2.45-GHz-induced blood toxicity by inhibiting free radical supporting antioxidant redox system although selenium has no effect on the investigated values.  相似文献

The main aim of this work was to provide baseline data on aspects of pro-oxidant and antioxidant processes in different life-stages of the marine amphipod Gammarus locusta. The activities of antioxidant enzymes and levels of lipid peroxidation were determined in whole body juveniles, subadults, and male and female adults of a laboratory population of G. locusta. Fatty acid composition of individuals at these different stages of development was also characterised in order to examine the contribution that polyunsaturated fatty acids (PUFA) might make to the peroxidation status of animals. The antioxidant enzymes, measured in whole body 100,000 supernatants, comprised catalase (CAT, EC 1.11.1.6), superoxide dismutase (SOD; EC 1.15.1.1) and glutathione peroxidase (GPX; EC 1.11.1.9). Fatty acids were analysed as fatty acid methyl esters (FAME). Lipid peroxidation was examined in terms of the levels of lipid peroxides determined as thiobarbituric acid-reactive malondialdehyde equivalents. Age-related changes were seen in antioxidant enzyme status: levels of SOD (p<0.01) and GPX (p<0.001) activities decreased progressively during development from juveniles to adults. Sex-related changes in GPX activity were also seen, the levels being higher in adult males than females (p<0.001). The amount of FAME present in whole body amphipod also changed over the life span. Among PUFA, the eicosapentaenoic (C20:5n-3), arachidonic (C20:4n-6) and docosahexaenoic (C22:6n-3) were the most abundant acids in this species, and both their individual concentrations and total PUFA increased progressively with age (up to 3.3-fold; p<0.001). The latter changes may contribute to the explanation of the observed differences in peroxidation status of the animals with age; thus, levels of lipid peroxides increased up to 40% in adult males compared to other age-classes (p<0.01). Overall, the decline in antioxidant enzyme activities, coupled with increased levels of PUFA, as the individual grows older, may render the older animals more susceptible to lipid peroxidation and oxidative stress.  相似文献

To understand the adaptability of alfalfa (Medicago sativa L.) to environmental stresses, we analyzed the activity of several antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), and catalase (CAT), in alfalfa shoots and roots subjected to salt and drought stresses during germination. The germination rate of six alfalfa cultivars was comparatively studied under 200 mM NaCl or 35% PEG treatment. Alfalfa Xinmu No. 1 and Northstar varieties were selected as stress-tolerant and -sensitive cultivars, respectively, and were used for further characterization. After NaCl or PEG treatment, Xinmu No. 1 showed enhanced seedling growth, compared with Northstar. Xinmu No. 1 also exhibited low levels of hydrogen peroxide (H2O2) production and lipid peroxidation, compared with Northstar. In addition, Xinmu No. 1 showed higher enzymatic activity of SOD, APX, CAT, and POD in its shoots and roots than Northstar. These results seem to indicate that Xinmu No. 1 cultivar's tolerance to salt or drought stresses during germination is associated with enhanced activity of antioxidant enzymes. This study highlights the importance of antioxidant enzymes in the establishment of alfalfa seedlings under drought and salinity conditions typical of desertification.  相似文献

The generation of superoxide radicals, lipid peroxidation (as measured by malone dialdehyde formation) and the activity of selected antioxidant enzymes (superoxide dismutase, catalase, ascorbate peroxidase) were assessed in radish (Raphanus sativus L.), in response to elevated concentrations of copper ions in the culture medium in vitro and in vivo. Experiments were performed on 7-day-old seedlings and 5-week-old calluses grown on media supplemented with CuSO4 in concentrations of 10, 100 and 1000 μМ. The exposure to elevated Cu concentrations in the medium significantly reduced both callogenesis and the proliferation of radish calluses in vitro. Cu treatment resulted in the increased generation of the superoxide radical (O2) in radish seedlings and calluses indicating the occurrence of oxidative stress in radish cells, whereas the level of lipid peroxidation (LPO) remained unchanged. Both in calluses and in radish seedlings in vivo, the relative level of oxidative stress was maximal at micromolar Cu concentrations and became attenuated with increasing Cu concentrations. Stronger oxidative stress occurred in the radish seedlings in vivo, compared with radish calluses in vitro. The observed lower sensitivity of calluses to Cu-induced oxidative stress and their ability to proliferate upon exposure to Cu concentrations of up to 1000 μМ demonstrate the potential of in vitro cell-selection to obtain metal-tolerant radish plant lines.  相似文献

Dystrophin is a protein found at the plasmatic membrane in muscle and postsynaptic membrane of some neurons, where it plays an important role on synaptic transmission and plasticity. Its absence is associated with Duchenne's muscular dystrophy (DMD), in which cognitive impairment is found. Oxidative stress appears to be involved in the physiopathology of DMD and its cognitive dysfunction. In this regard, the present study investigated oxidative parameters (lipid and protein peroxidation) and antioxidant enzymes activities (superoxide dismutase and catalase) in prefrontal cortex, cerebellum, hippocampus, striatum and cortex tissues from male dystrophic mdx and normal C57BL10 mice. We observed (1) reduced lipid peroxidation in striatum and protein peroxidation in cerebellum and prefrontal cortex; (2) increased superoxide dismutase activity in cerebellum, prefrontal cortex, hippocampus and striatum; and (3) reduced catalase activity in striatum. It seems by our results, that the superoxide dismutase antioxidant mechanism is playing a protective role against lipid and protein peroxidation in mdx mouse brain.  相似文献

The role of Δ12-acyl-lipid desaturase in plant resistance to hypothermia-induced oxidative stress was investigated. This study focused on modulation of free-radical processes occurring at low temperature in leaf cells of potato plants (Solanum tuberosum L., cv. Desnitsa) transformed with the gene for Δ12-acyl-lipid desaturase from the cyanobacterium Synechocystis sp. PCC 6803. Nontransformed plants of the same cultivar were used as a control material. The plants were grown in vitro on Murashige and Skoog agarized medium containing 2% sucrose. During hypothermia the rate of superoxide anion generation and hydrogen peroxide concentration decreased significantly. In addition, the content of both primary products (conjugated dienes and trienes) and secondary products (malonic dialdehyde) of lipid peroxidation was lower in the transformed plant leaves than in leaves of wild-type plants. It is supposed that the insertion into the plant genome of Δ12-acyl-lipid desaturase stabilizes the composition and physical properties of biomembranes by promoting polyunsaturation of fatty acids, which averts the accelerated generation of O 2 ·? , — and suppresses lipid peroxidation during hypothermia. These changes improved cold resistance of potato plants, which was evident from the less severe injury of leaf blades in cold-treated transgenic plants, as compared to that in the wild-type line. The activity of superoxide dismutase, a key enzyme of the antioxidant defense system was lower in leaves of transformed plants than in leaves of wild-type plants. A comparatively low activity of superoxide dismutase in transgenic plants implies that these plants experience less severe thermal and oxidative stress upon cooling and can cope with the cold without considerable increase in the enzyme activity. It is concluded that the insertion of the desA gene encoding Δ12-acyl-lipid desaturase into cold-resistant potato plants improves plant resistance to cold-induced oxidative stress by decreasing the rate of intracellular free-radical processes.  相似文献

Three strains of green microalgae, Chlorococcum sp.C53, Chlorella sp. E53, and Chlorella sp.ED53 were studied for their antioxidant activities. Crude extracts of these microalgae in hot water and in ethanol were examined for their total phenolic contents and for their antioxidant capacities. In order to determine their phenolic contents, the Folin–Ciocalteu method was used. As for the determination of their antioxidant capacities, four different assays were used: (1) total antioxidant capacity determination; (2) DPPH radical scavenging assay; (3) ferrous ion chelating ability assay; and (4) inhibition of lipid peroxidation (using thiobarbituric acid reactive substance). For all the strains we have studied, their ethanolic extract showed more antioxidant activities than their hot water extract. Categorically, the ethanolic extract of Chlorella sp.E53 exhibited both the highest total phenolic content of 35.5?±?0.14 mg gallic acid equivalent (GAE) g?1 dry weight and the highest DPPH radical scavenging of 68.18?±?0.38 % at 1.4 mg mL?1 (IC50 0.81 mg mL?1), whereas Chlorella sp.ED53 showed both the highest ferrous ion chelation activity of 42.78?±?1.48 % at 1 mg mL?1 (IC50 1.23 mg mL?1) and the highest inhibition of lipid peroxidation of 87.96?±?0.59 % at 4 mg mL?1. This high level of inhibition is comparable to 94.42?±?1.39 % of butylated hydroxytoluene, a commercial synthetic antioxidant, at the same concentration.  相似文献

Iron saccharate complex ISC is an iron supplement used to optimize erythropoiesis in cases of iron deficiencies. Because of the lack of major mechanisms of iron excretion, excess iron unbound to protective molecules is believed to be involved in catalyzing the generation of reactive oxygen species and induction of oxidative stress. This study employed ISC for the purpose of inducing iron overload and hence investigating the consequent iron toxicity, lipid peroxidation and antioxidant extent in a murine species. Male Wistar rats were given iron as intraperitoneal injections of ISC in subacute (0.2 mg Fe kg?1 for 2 weeks) and subchronic (0.1 mg Fe kg?1 for 4 weeks) doses. In iron-overloaded rats, enhanced hepatic iron accumulation (P > 0.001) attended by increased serum concentrations of malondialdehyde (MDA) (P > 0.001) and activities of antioxidant enzymes (superoxide dismutase SOD, catalase CAT and glutathione peroxidase GPx) (P > 0.001) was pointed out. The demonstrated antioxidant boost is attributed to a sense of equilibrium prompted by the potential of iron-induced oxidative stress to modify antioxidant defense capacity and to modulate susceptibility to oxidative stress. Rats seemed to constantly suffer from oxidative stress based on the consistent rise in MDA that was not overwhelmed by the elevated antioxidant input. The current findings are of informative value in drawing attention to the health hazards of applying higher doses of the commercially used iron supplement ISC. Data are virtually significant in elucidating the higher magnitude of subchronic than subacute iron overload in initiating oxidative stress and antioxidant defense. Both pathways proceeded in a time-dependent rather than dose-dependent manner.  相似文献
