Aspergillus niger, a soil-borne fungus is a causative agent of hypocotyl malformations in infected groundnut (Arachis hypogaea L.) plants, but its effect on yield is unknown. This study sought to determine its effect on growth and yield. Seeds of Chinese and JL45 varieties were sown in soil inoculated with A. niger. Fresh and dry weights of the shoots and roots were taken at 10-day intervals. Nodule count was done at 30 days after emergence and subsequently at 10-day intervals. Pods of 20 plants each from inoculated and uninoculated soils were harvested. Growth was suppressed in plants grown on A. niger inoculated soil. Eight-day old plants grown in inoculated soil developed curvatures on their hypocotyls. Nodulation was suppressed (p < 0.05) in plants grown in inoculated soil. Although growth was suppressed in plants grown on inoculated soil, yield of both varieties of groundnut was not affected.  相似文献

A study was conducted to determine whether colonization of legume roots and nodulation byRhizobium meliloti andBradyrhizobium japonicum could be enhanced by using inocula containing microorganisms that produce antibiotics suppressing soil or rhizosphere inhabitants but not the root-nodule bacteria. An antibiotic-producing strain of Pseudomonas and one of Bacillus were isolated, and mutants ofR. meliloti andB. japonicum sp. resistant to the antibiotics were used. The colonization of the alfalfa rhizosphere and nodulation byR. meliloti were enhanced by inoculation of soil withPseudomonas sp. in soil initially containing 2.7×105 R. meliloti per g. The colonization of soybean roots byB. japonicum was enhanced by inoculating soil with three cell densities ofBacillus sp., and nodulation was stimulated byBacillus sp. added at two cell densities. In some tests, the dry weights of soybeans and seed yield increased as a result of these treatments, and co-inoculation with Bacillus also increased pod formation. Inoculation of seeds withBacillus sp. and the root-nodule bacterium enhanced nodulation of soybeans and alfalfa, but colonization byB. japonicum andR. meliloti was stimulated only during the early period of plant growth. Studies were also conducted withStreptomyces griseus and isolates ofR. meliloti andB. japonicum resistant to products of the actinomycete. Nodulation of alfalfa byR. meliloti was little or not affected by the actinomycete alone; however, both nodulation and colonization were enhanced if the soil was initially amended with chitin andS. griseus was also added. Chitin itself did not affectR. meliloti. Treatments of seeds with chitin orS. griseus alone did not enhance colonization of alfalfa roots byR. meliloti or soybean roots byB. japonicum, but the early colonization of the roots by both bacterial species was promoted if the seeds received both chitin andS. griseus; this treatment also increased nodulation and dry weights of alfalfa and soybeans and the N content of alfalfa. It is suggested that co-inoculation of legumes with antibiotic-producing microorganisms and root-nodule bacteria resistant to those antibiotics is a promising means of promoting nodulation and possibly nitrogen fixation.  相似文献

The antifungal properties of isothiocyanates released from the hydrolysis of the glucosinolate sinigrin were assessed on an Armillaria mellea strain in Petri dishes. The fumigation with sinigrin at 100 µM showed a fungicidal effect, while lower rates determined a temporary fungistatic effect. The application of increasing rates of solid and liquid biofumigant formulations on A. mellea inoculated potted peach plants showed an increase of soil nitrate concentration, basal respiration, microbial biomass, leaf nitrogen and chlorophyll concentration. These effects indicated a stimulating effect of Brassicaceae derivatives on soil biological activity and plant growth. The application of a rate of meal higher than 6.42 g kg?1 soil caused a stunted growth and the death of some plants showing a clear phytotoxic effect of the treatments. The inhibition of A. mellea growth observed in in vitro trials was not evidenced in in vivo experiments due to the lack of infection symptoms in experimentally inoculated potted trees.  相似文献

The presence and growth of Aspergillus fumigatus and Aspergillus niger in the soil of ornamental plants have been demonstrated. The ecological conditions in the soil of such plants as influenced by temperature, humidity, desiccation, fertilization and ventilation obviously influence such fungal growth. The epidemiological significance of these findings is of interest with a view to the present efforts to control aspergillosis in the environment of susceptible persons.Observations of a preferential growth of certain Aspergillus species in the soil of defined plants under defined conditions raise problems of soil microbiology.  相似文献

A system was developed to evaluate the effects of root growth of cotton seedlings on the inoculum dynamics ofGliocladium virens in nonsterile soil. In soil infested withG. virens, inoculum densities of the fungus increased when plants remained alive. After 30 days, shoots were excised and the roots allowed to deteriorate. During this portion of the experiment (30–60 days) soil inoculum densities ofG. virens declined. In infested soil without a seedling, inoculum densities remained constant throughout the duration of the experiments. Colonization of roots byG. virens was found to increase throughout the duration of the experiments. At 60 daysG. virens was recovered from approximately 60% of the root pieces (1-cm) sampled. The percentage of primary, secondary, or tertiary roots colonized was different (P = 0.01), but the total colonization of roots at three depths (0–10, 10–20, and 20–30 cm) was not different (P = 0.64). Pot and field experiments on small plots were carried out to study the effect of isolates ofPseudomonas fluorescens-putida bacteria, applied to potato tubers, on the growth response of potato plants. Inoculation of tuber pieces with the isolates in pot experiments caused a better growth of young potato plant (111 % increase). The effect depended on the viable count in the bacterial suspension and on growth conditions of the plants. Inoculation of potato seed pieces before planting in field experiments caused a 4 –30 % improvement in plant growth and tuber yield. The complex action of inoculation with rhizobacteria apparently caused changes in the microflora colonizing the roots and stolons, which in turn brought about a better growth and yield.  相似文献

The role of harmful soil organisms in the degeneration ofAmmophila arenaria at coastal foredunes was examined by the growing of seedlings ofA. arenaria in soil samples collected from its root zone. Three sites, each representing a successive stage in foredune succession were examined: (1) a highly mobile dune (sand accretion of 80 cm year−1) with vigorousA. arenaria, colonizing only the upper 30-cm of the annually deposited layer of sand, (2) a mobile dune with vigorousA. arenaria (sand accretion of 22 cm year−1) and a 1-metre soil profile completely colonized by roots and (3) a stable dune (no sand accretion) with degeneratedA. arenaria and young roots mainly present in the upper 0–10 cm. In the upper part of the highly mobile site, the presence of harmful soil organisms was confined to the root layers and at the mobile site for all depth layers a significant growth reduction ofA. arenaria was observed due to the activity of harmful soil organisms. At the stable site, however, growth had only been reduced in some of the depth layers. At all sites newly formed roots ofA. arenaria had been colonized by harmful soil organisms within one year. If present in sand prior to root growth harmful soil organisms reduced root length and root hair formation severely and they enhanced branching of the roots. It is concluded that harmful soil organisms initiate degeneration ofA. arenaria in stable dunes by attack of the root system, which makes the plants suffer from abiotic stress.  相似文献

Summary Aspergillus flavus andAspergillus niger produce extracellular amylase into the culture medium when grown on basal medium containing 2% (w/v) soluble starch or cassava peel as the sole carbon source. On soluble tarch the highest amylase activities were 1.6 and 5.2 mg of starch hydrolyzed/min per mg protein forA. flavus andA. niger, respectively. When grown on cassava peel, the highest amylase activity in the culture filtrate ofA. flavus was 170-times higher than that on soluble starch, while that ofA. niger was 16-times higher. The mycelial dry weight for both organisms was not significantly affected by the carbon sources. Maximum enzyme activity was obtained at the growth temperature of 29.0±1°C and pH 7 for both organisms. It is concluded that cassava peel might be a better substrate for the production of amylase byA. flavus andA. niger than commercial soluble starch.  相似文献

Both the volume and concentration of filtrate ofAspergillus niger andTrichoderma viride influenced the development of the radicles of cocoa seedlings. Radicles placed in Petri dishes containing filter paper moistened with 20 and 30 ml of undiluted filtrate and filtrate dilutions of 1∶1 and 1∶2 failed to develop lateral roots and eventually died. The culture filtrate ofA. niger was more repressive. Radicles in the same volume of filtrate (20 and 30 ml) of higher dilutions (1∶5 and 1∶10) developed lateral roots and survived. Radicles placed in less volume (≤10 ml) survived and produced lateral roots irrespective of concentration of filtrates. Development of the radicles and roots in the control was consistently better (P=0.05) than in filtrate solutions of eitherA. niger orT. viride. The hypocotyls of seedlings under the influence of metabolites ofA niger showed greater cambial activity and formed xylem vessels and tracheids with larger lumina.  相似文献

Addition of MnSO4 or MnCl2 to a fine sandy soil from South Australia had a negative effect on shoot growth and root elongation ofSolanum opacum in the absence of significant presence of vesicular-arbuscular mycorrhiza (VAM). VAM ameliorated the reduction of plant growth by Mn, even though mycorrhizal development was decreased. Mn inhibited infection of roots by a fine endophyte less than that by some coarse endophytes. High concentrations of available Mn inhibited growth of hyphae of VAM fungi from dried root pieces, a significant source of infection by mycorrhizal fungi in the soil used.  相似文献

In a survey ofAzospirillum spp. on the roots and associated soil of wheat grown in eastern Australia, azospirilla were isolated from approximately 40% of samples from areas of soil pH between 5.0 and 6.6. However, azospirilla isolates were rare in soil between pH 4.5 and 5.0 and absent below pH 4.5. Of 25 independent isolates, 17 wereA. brasiliense and eight wereA. lipoferum. No selection forA. brasiliense Nir strains by wheat roots was observed. Only one of six endorhizosphere isolates wereA. brasiliense Nir, compared with three of nine from unsterilized roots plus associated soil, and three of eight from soil. With a medium buffered with 0.05 M malate and 0.05 M phosphate, it was found that allAzospirillum isolates had a lower minimum pH for growth when supplied with fixed nitrogen than when grown under nitrogen-fixing conditions. Strains isolated from soils had a minimum pH for growth that was less than the pH of the soil from which they were isolated. However, a significant proportion of strains isolated from roots had a minimum pH for growth that was higher than the pH of the associated soil suggesting that the wheat roots provided an ecological niche protecting against soil acidity.  相似文献

A method, based on the negative adsorption of NO2 -, has been developed to determine surface area of roots. Young roots of 3–4 year old plants ofAcacia catechu, Eucalyptus camaldulensis andLeucaena leucocephala were up-rooted and cut into 18 cylindrical pieces. Each root piece was immersed individually for 10 sec in 0.05M, 0.10M and 0.15M aqueous NaNO2 solution and the excess solution on the root surface allowed to drain off. It was then transferred to conical flask containing distilled water and shaken for 15 min for desorption of nitrite. A known quantity of this aliquot was reacted with 1% acidic sulphanilamide and 0.02% NED HCl. A pink colour developed, and its optical density was read at 540 nm. A positive linear correlation was noted between colour density and root surface area. The respective correlation coefficientvalues for 0.05M, 0.10M and 0.15M NaNO2 solutions were 0.973, 0.963 and 0.964 forAcacia catechu, 0.933, 0.903 and 0.898 forEucalyptus camaldulensis and 0.968, 0.976 and 0.972 forLeucaena leucocephala (significant atp<0.001). The method was successfully adopted to determine the root surface area of seedlings ofAlbizia lebbek, A. procera, Acacia auriculiformis, A. nilotica, Dalbergia latifolia andD. sissoo.  相似文献

Summary Hippophaë rhamnoides seedlings were grown in sterilized and unsterilized soil from a decliningH. rhamnoides scrub, to which different numbers ofLongidorus sp. andTylenchorhynchus microphasmis were added. In sterilized and unsterilized soil, retardation of growth, content of dry matter in the shoots, and incidence of deformed short lateral roots of test plants were positively correlated with counts of both nematode species. Nitrogen content in the shoots, nodulation on the roots of test plants and increase increase in nematodes were negatively correlated with the initial number of both nematode species in sterilized soil. In unsterilized soil, an unknown biotic factor was present that reduces growth ofH. rhamnoides, nodulation and multiplication of the nematodes. Co-inoculation with antibiotic-producing bacteria and rhizobia resistant to those antibiotics has been proposed as a means of promoting colonization and nodulation of legumes by root-nodule bacteria. A study was conducted to establish some of the factors affecting co-inoculation with antibiotic-producing strains of Bacillus and Streptomyces griseus. The stimulation of Rhizobium meliloti and yield and N uptake by alfalfa was enhanced with increasing inoculum size of Bacillus sp. S. griseus and chitin added to soil increased nodulation of soybeans by Bradyrhizobium japonicum and increased nodulation, yield, and number of pods on a second crop grown in the same soil. Bacillus sp. persisted in soil in sufficient numbers for at least 51 days to increase colonization of soybean roots by B. japonicum. The populations of S. griseus, Bacillus sp., and antibiotic-resistant isolates of R. meliloti and B. japonicum fell after their addition to seeds. Nevertheless, a benefical effect by the antibiotic-producing bacteria was evident on R. meliloti colonization of the rhizosphere, nodulation, and yield of alfalfa grown from seeds stored 94 days and on B. japonicum colonization, nodule number, yield, and seed weight of soybeans grown from seeds stored 90 days. Because non-antibiotic-producing derivatives of Bacillus sp. and S. griseus did not promote colonization or nodulation of alfalfa roots by R. meliloti, the benefit of this co-inoculation is a result of antibiotic formation.  相似文献

Toxigenic and non-toxigenic black aspergilli belonging to theAspergillus niger aggregate and toA. carbonarius were compared to each other and to strains of other species by DNA fingerprinting. AFLPs showed a clear separation ofA. niger andA. carbonarius. However, no clear correlation between the genetic similarity of the strains and the ability to produce ochratoxin A (OTA) was detected. Based on AFLP, marker sequences were chosen for the construction of SCAR-PCR primers for the detection ofA. carbonarius. A similar approach was used forA. ochraceus, another fungus of concern regarding ochratoxin A contamination of coffee. Cluster analysis ofA. ochraceus isolates mainly from Brazilian coffee showed a very close genetic similarity. Three species specific primer pairs were developed and one of these was used for the PCR and realtime PCR (RT-PCR) based detection of the mould in green coffee.A. ochraceus was specifically and rapidly detected and quantified in green coffee. A positive correlation between the amount of DNA and OTA content was established.  相似文献

Sparkill (25% Cypermethrin) and Tafaban (48% Chlorpyrifos) insecticides were used in the treatment of maize plants in the field to investigate the effect of these compounds on fungi associated with roots and shoots of maize as well as on soil fungi. Generally, the two insecticides exhibited an inhibitive effect on the total and individual counts of glucophilic fungi after different periods of treatment except in some cases the counts were not affected or promoted compared with the control samples. Fungal genera and species associated with roots and shoots of maize plants were isolated from untreated plants as control and then compared with those isolated from plants treated with different doses of insecticides. The most common fungal species recovered from soil and different parts of plants were Alternaria alternata, Aspergillus flavus, A. fumigatus, A. niger, A. terreus, Cochliobolus spicifer, Emericella nidulans, E. nidulans var. lata, Fusarium oxysporum, Gibberella fujikuroi, Nectria haematococca, Penicillium chrysogenum, P. funiculosum, P. oxalicum and Rhizopus stolonifer.  相似文献

Summary Extracted grape waster material and pressed apple pulp were tested as carbon sources forPenicillium funiculosum 515,Myrothecium verrucaria 9095 andAspergillus niger TMF-15. They were good growth substrates, especially forA. niger. When cultivated on mixed substrate in optimized nutrient medium,A. niger accumulated a product of 35% crude protein with a maximum productivity of 0.117 g protein/1/h and cellulose consumption of 90.92%.A. niger also produced the highest levels of cellulase activity. Bacteria, other than aerobic spore bearing species, are preferentially supported by the roots of both varieties.2. In a greenhouse experiment, five ornamental plants, Osmanthus fragrans (OF), Ligustrum vicaryi L. (LV), Cinnamomum camphora (CC), Loropetalum chinense var. rubrum (LC), and Euonymus japonicas cv. Aureo-mar (EJ), were studied for the ability to phytostabilization for Cd-contaminated soil. The results showed that these five ornamental plants can grow normally when the soil Cd content is less than 24.6 mg·kg?1. Cd was mainly deposited in the roots of OF, LV, LC and EJ which have grown in Cd-contaminated soils, and the maximum Cd contents reached 15.76, 19.09, 20.59 and 32.91 mg·kg?1, respectively. For CC, Cd was mainly distributed in the shoots and the maximum Cd content in stems and leaves were 12.5 and 10.71 mg·kg?1, however, the total amount of Cd in stems and leaves was similar with the other ornamental plants. The enzymatic activities in Cd-contaminated soil were benefited from the five tested ornamental plants remediation. Soil urease and sucrase activities were improved, while dehydrogenase activity was depressed. Meanwhile, the soil microbial community was slightly influenced when soil Cd content is less than 24.6 mg·kg?1 under five ornamental plants remediation. The results further suggested that ornamental plants could be promising candidates for phytostabilization of Cd-contaminated soil.  相似文献

One rape (Brassica napus cv. Wesroona) plant and four cotton (Gossypium hirsutum cv. Sicot 3) plants were grown in plastic cells containing soil labelled with 407 kBq of33P g−1 soil. After 5–8 days of growth, the33P depletion zones of all plants were autoradiographed and33P uptake by plants was measured. The autoradiographs were scanned with a microdensitometer and the optical densities at several places within the33P depletion zones of roots were obtained. The volume of soil explored by root hairs was estimated from measurements of root diameters and lengths of roots and root hairs. About half of the total33P depleted by cotion roots came from outside the root hair cylinder whereas most of33P taken up by rape was from within the root hair cylinder. Plants grown in a macrostructured soil may have roots growing in voids, within aggregates or on the surfaces of aggregates. The results of this study demonstrate that root hairs have a strong influence on the accessibility of phosphorus to roots in such a soil, and thus on the phosphorus nutrition of plants.  相似文献
