A significant challenge to population genetic studies of the dengue vector, Aedes aegypti, has been the lack of polymorphic microsatellite loci. In an effort to develop useful markers, we evaluated the genetic variation at 17 microsatellite loci identified in the A. aegypti genome. Nine loci with at least five alleles were identified in field‐collected specimens from Thailand. An additional two loci carried five alleles if samples from an A. aegypti laboratory colony were included. Our results greatly increase the number of highly variable markers available for the study of the genetics and the population structure of this medically important species.  相似文献

A laboratory strain of Aedes aegypti (L) was subjected repeatedly to larval selection pressure with two bacterial insecticides, spinosad (Saccharopolyspora spinosa) and bacilod (Bacillus thuringiensis israelensis). The results indicated that the mosquito Ae. aegypti acquired low resistance to spinosad and bacilod by about 3.1 and 2.4-fold, respectively, due to selection pressure for fifteen successive generations. The slope values of the selected strains were increased gradually from one generation to the next, indicating moderate homogeneity between individuals in their response to the test bio-insecticide. Moreover, larval selection with current bacterial bioinsecticides prolonged the time required to digest a blood meal. It showed an evident decrease in the reproductive potential of adult mosquitoes surviving selected larvae.  相似文献

In French Guiana, pyrethroids and organophosphates have been used for many years against Aedes aegypti. We aimed to establish both the resistance level of Ae. aegypti and the ultra low volume spray efficacy to provide mosquito control services with practical information to implement vector control and resistance management. Resistance to deltamethrin and fenitrothion was observed. In addition, the profound loss of efficacy of AquaK'othrine? and the moderate loss of efficacy of Paluthion? 500 were recorded. Fenitrothion remained the most effective candidate for spatial application in French Guiana until its removal in December 2010. Further investigation of the mechanism of resistance to deltamethrin demonstrated the involvement of mixed-function oxidases and, to a lesser extent, of carboxylesterases. However, these observations alone cannot explain the level of insecticide resistance we observed during tube and cage tests.  相似文献

This study reports the biochemical characterization and comparative analyses of highly active serine proteases in the larval and pupal developmental stages of Aedes aegypti (Linnaeus) using substrate‐SDS‐PAGE. Zymographic analysis of larval stadia detected proteolytic activity in 6–8 bands with apparent molecular masses ranging from 20 to 250 kDa, with activity observed from pH 5.5 to 10.0. The pupal stage showed a complex proteolytic activity in at least 11 bands with apparent Mr ranging from 25 to 250 kDa, and pH optimum at 10.0. The proteolytic activities of both larval and pupal stages were strongly inhibited by phenyl‐methyl sulfonyl‐fluoride and N‐α‐Tosyl‐L ‐lysine chloromethyl ketone hydrochloride, indicating that the main proteases expressed by these developmental stages are trypsin‐like serine proteases. The enzymes were active at temperatures ranging from 4 to 85°C, with optimal activity between 37 and 60°C, and low activity at 85°C. Comparative analysis between the proteolytic enzymes expressed by larvae and pupae showed that substantial changes in the expression of active trypsin‐like serine proteases occur during the developmental cycle of A. aegypti. © 2011 Wiley Periodicals, Inc.  相似文献

Dengue is an important disease that affects humans, and is transmitted by A. aegypti. During 2006, a total of 477 cases of hemorrhagic dengue, and 1 510 of classic dengue were recorded in Sinaloa. Due to this high impact, a study on insect abundance and distribution, as well as their relationship with dengue dispersion, was carried out from April 2008 to March 2009 in Guasave, Sinaloa. The study included a total of six sectors in the city, considering 16 colonies; besides, 96 traps were distributed in these sectors to monitor the vector population density and female number per trap. The adult density index (ADI) and traps positive index (TPI) were calculated, and data were used to obtain the monthly dengue dispersion maps. The highest abundance of female (137 and 139) was found in July and August. In August and September the sectors 2, 4 and 6 showed the highest ADI values 2.44, 3.35 and 2.290, while TPI values were 56.25, 58.82 and 61.54, with the highest epidemiology dengue dispersion. The Pearson correlation (p < 0.05) showed better values with the precipitation (r = 0.80) than mean temperature (r = 0.76) with zero lag months; while the cases of classic dengue and hemorrhagic dengue (CD/HD) is mostly related with the precipitation (r = 0.98), and minimum temperature (r = 0.79), with two lag months. In conclusion, the ADI and TPI index, as well as the maps obtained, allowed us to know the location of epidemiologic dengue risk areas. This information can be used to develop better control measures biological and chemical for the mosquito, in this location.  相似文献

Differences in biological features of immature and adult Aedes aegypti, as well as variability in vector competence, seem consistent with the existence of genetic variation among subpopulations and adaptation to local conditions. This work aims to compare the bionomics of four Ae. aegypti subpopulations derived from different geographical regions reared under temperate conditions. Life statistics of three Ae. aegypti subpopulations from the provinces of Córdoba, Salta, and Misiones were studied based on horizontal life tables. The Rockefeller strain was used as a control. The development time required to complete the larva and pupa stages varied from 6.91 to 7.95 and 1.87 to 2.41 days, respectively. Significant differences were found in mean larval development time between the Córdoba and Orán subpopulations. The larva‐pupa development time was similar in all the subpopulations. However, survival values varied significantly between the Orán and San Javier subpopulations. The proportion of emergent males did not differ from females within each subpopulation nor among them. Adult longevity was similar among the subpopulations. The average number of eggs laid by each female was significantly different. The Rockefeller strain laid a significantly greater number of eggs (463.99 eggs/female) than the rest of the subpopulations. Moreover, differences in the demographic growth parameter Ro were detected among the four subpopulations. The differences obtained in larval development time, larva‐pupa survival values, and net reproductive rates among the subpopulations might reflect underlying genetic differences as a result of colonization from different regions that probably involve adaptations to local conditions.  相似文献

Investigations were made to evaluate the larvicidal activity and smoke repellent potential of Toddalia asiatica (L.) and Aegle marmelos at different concentrations (20, 40, 60, 80 and 100 p.p.m.) against first to fourth instar larvae and pupae of Aedes aegypti (L.). The LC50 of A. marmelos and T. asiatica against first instars was 50.960 and 47.893 p.p.m., respectively; against second instars, 52.979 and 50.922; third instars, 56.653 and 54.461; and fourth instars, 60.778 and 61.278. The LC50 and LC90 for A. marmelos and T. asiatica against pupae were 56.634 and 112.992, and 53.64 and 116.22 p.p.m., respectively. The smoke toxicity of T. asiatica against Ae. aegypti was greater than that of A. marmelos . Smoke-exposed gravid females oviposited fewer eggs when compared to those that were not exposed to smoke. Smoke-exposed females hatched a lower percentage of eggs compared to unexposed females.  相似文献

Development times and mortality factors were investigated for populations of Aedes scapularis (Rondani) occurring in natural temporary ground pools in southeastern Brazil. Analysis of life tables by the key-factor method showed that mortality caused by desiccation of breeding sites was the principal factor accounting for fluctuations in population sizes throughout the study period. For cohorts that completed development, mortality attributed to predation by aquatic insects ranged from 68 to 96% and was the most important cause of death. In these cohorts, final population size was determined by the high mortality rates that occurred during the fourth larval instar and pupal stages; however, the key-factor best accounting for the population fluctuations was predation of fourth instar larvae. The short mean time of 7.1 days for synchronous development of the larvae and the lack of evidence of density-dependent mortality suggested that the A. scapularis populations were not strongly regulated and can attain relatively high densities.  相似文献

We report here the reappearance of Aedes aegypti in the Rimac district, and summarize the history of this mosquito species in Peru since its first detection in 1852. On March 17 2000 were found Ae. aegypti and Culex quinquefasciatus in Mariscal Castilla town, Flor de Amancaes, San Juan de Amancaes, El Altillo and Santa Rosa in the Rimac district, Lima Province.  相似文献

Insecticide susceptibility of a 1995 Townsville Aedes aegypti (L.) population was investigated in the laboratory according to World Health Organization guidelines. Using baseline data from two Townsville populations (1955 and 1989), larval bioassays detected significant increases in susceptibility to synthetic pyrethroids and malathion, and significant reductions in susceptibility to most organophosphates and propoxur. Adult bioassays detected significant resistance to bendiocarb and DDT. Comparison of larval data with the international reference ROCK strain showed substantial resistance to have developed to malathion and fenthion. Further analysis revealed the presence of distinct substrains in the baseline 1989 population, which displayed varying levels of temephos susceptibility. We concluded that susceptibility investigations should assess mosquito populations collected from many sites within an area rather than taking a single population from one site, and that the 1989 Ae. aegypti colony would be unsuitable for use as a susceptible reference population.  相似文献

Investigations were made to test the larval toxicity and smoke repellent potential of Albizzia amara and Ocimum basilicum at different concentration (2%, 4%, 6%, 8% and 10%) against the different instar (I, II, III and IV) larvae and pupae of Aedes aegypti. The LC50 values of A. amara and O. basilicum for I instar larvae was 5.412 and 3.734, II instar 6.480 and 4.154, III instar 7.106 and 4.664, IV instar 7.515 and 5.124, respectively. The LC50 and LC90 values of pupae were 6.792%, 5.449% and 16.925%, 15.474%. The smoke toxicity of A. amara was more effective against A. aegypti than the O. basilicum.  相似文献

Aedes aegypti is the main insect vector of Dengue fever and dengue hemorrhagic fever/dengue shock syndrome and represents the only vulnerable element in the control of this disease. Therefore, the identification and quantification of this mosquito is an important task; however, the majority of taxonomic keys are based on the 4th larval instar. For that reason, this study describes the four larval instars ofA. aegypti using scanning electron microscopy. Morphological changes during larval development were observed at the pecten, comb scales and the ventral brush of the abdominal segment X; however, the 3rd and 4th instars showed similar structures with only a slight variation. The structures described in this study will be helpful in the identification of the four instars of A. aegypti, a fundamental task for comprehending the natural history of dengue mainly in new territories affected.  相似文献

Dengue is a global health concern. Growing insecticide resistance in the primary mosquito vector, Aedes aegypti, limits the effectiveness of vector control, so alternative tools are urgently needed. One approach is the use of biopesticides comprising entomopathogenic fungi, e.g., Beauveria bassiana and Metarhizium anisopliae. These fungi may decrease disease transmission by reducing mosquito vector longevity and also occur worldwide, although many isolates have not been tested for virulence against mosquitoes. Ninety-three isolates of entomopathogenic fungi representing six species (B. bassiana, M. anisopliae, Isaria fumosorosea, I. farinosa, I. flavovirescens, and Lecanicillium spp.) were screened as potential biological control agents of Aedes aegypti. A hierarchical, multi-criteria experimental design was undertaken to find suitable isolates. Initial screening was performed via in vitro assays measuring radial growth and spore persistence, eliminating isolates with poor growth or viability on nutrient-rich substrate. Subsequent measurements of spore persistence revealed that only nine of 30 strains tested had half-lives exceeding 3 weeks. Ten isolates were chosen for in vivo bioassays against adult Ae. aegypti. From these assays, two Australian isolates of B. bassiana, FI-277 and FI-278, appeared to be most promising. Both isolates were shown to be virulent against Ae. aegypti at 20, 26, and 32°C. Spreading spores manually onto substrate was found to be more efficacious than spraying. Ae. aegypti infected by manually-spread spores on cotton substrate were found to have an LT50 of 3.7±0.3 days. These characteristics suggest that FI-277 has promise as a dengue mosquito biocontrol agent, either alone or combined with conventional chemical insecticides.  相似文献

The chemical characterization of the hydrocarbon fraction of the epicuticular lipids of the vector mosquito Aedes aegypti (Linnaeus) was performed using gas chromatography (GC) and gas chromatography-electron impact mass spectrometry (GC-MS). Seventy eight compounds were detected in purified hexane extracts and of these, 42 hydrocarbons were identified and several of the remaining compounds were partially characterized. The hydrocarbon classes present were n-alkanes, monomethylalkanes, dimethylalkanes and alkenes and the results were similar to those published for other Aedes species. Quantitative comparisons of cuticular hydrocarbon profiles were made between males and females, different age groups and between a standard laboratory strain and a recently colonized strain of A. aegypti. These results provide baseline data for further studies on the possible role of mosquito cuticular hydrocarbons in the modification of mating behaviour.  相似文献

Thirty‐two Aedes aegypti populations collected throughout Thailand and five populations of Aedes albopictus from southern Thailand were subjected to standard WHO contact bioassays to assess susceptibility to three commonly used synthetic pyrethroids: permethrin, deltamethrin, and lambda‐cyhalothrin. A wide degree of physiological response to permethrin was detected in Ae. aegypti, ranging from 56.5% survival (Lampang, northern Thailand) to only 4% (Kalasin in northeastern and Phuket in southern Thailand). All 32 populations of Ae. aegypti were found to have evidence of incipient resistance (62.5%) or levels of survival deemed resistant (37.5%) to permethrin. Four populations of Ae. albopictus were found with incipient resistance (97 – 80% mortality) and one with resistance (< 80%) to permethrin. The majority of Ae. aegypti populations (68.7%) was susceptible (> 98% mortality) to deltamethrin, with incipient resistance (observed 97–82% mortality) in other localities. In contrast, all populations of Ae. aegypti were completely susceptible (100% mortality) to the recommended operational dosage of lambda‐cyhalothrin. All five populations of Ae. albopictus were found completely susceptible to both deltamethrin and lambda‐cyhalothrin. Evidence of defined incipient or resistance to synthetic pyrethroids mandates appropriate response and countermeasures to mitigate further development and spread of resistance. In light of these findings, we conclude that routine and comprehensive susceptibility monitoring of dengue mosquito vectors to synthetic pyrethroids should be a required component of resistance management policies and disease control activities.  相似文献

The larval survival and development times of Aedes caspius (Diptera: Culicidae) were examined in the laboratory. These life history traits were estimated using life tables constructed for two populations, one of which had been subjected to a long-term larvicide control program. Traits were evaluated for eight different population densities. The effects of population, larval stage and larval density were investigated using a general linear model. Density was positively correlated with larval survival but did not affect development time. The fourth instar and pupae had the lowest larval survival rates. First and fourth instar larvae had the longest development times. These traits were not significantly different between the two populations. The effect of larvicide control on these traits is discussed.  相似文献

New larval control strategies for integrated vector management of Aedes aegypti are in high demand, including the use of biological control agents. Exposure of Aedes aegypti to parasites, starvation, and overcrowded conditions during larval development reduces the probability of survival to eclosion, can directly affect fitness parameters such as adult size and fecundity, and can affect the size, provisioning, and viability of eggs produced by females. We compared these parameters after exposing larvae to 1) abundant food at low larval densities, 2) food deprivation and high larval density, and 2) infection with the endoparasite Plagiorchis elegans, an entomopathogenic digenean trematode. Female mosquitoes that eclosed from larval conditions of starvation and overcrowding were smaller and laid fewer and smaller eggs than controls. The proportion of females to complete an oviposition cycle was reduced in the P. elegans-infected treatment group. Parasite load was negatively correlated with wing length and egg size. Infection of Ae. aegypti with P. elegans has sublethal effects and may reduce population-level reproductive output, but one-time low-density P. elegans exposure does not have sufficient effect on Ae. aegypti fitness parameters to be considered a viable biocontrol option.  相似文献
