Abstract There exists a wide diversity of halophilic eubacteria with chemoorganotrophic-aerobic metabolism. Most of them have a more moderate salt response than halophilic archaebacteria, falling into the category of moderately halophilic bacteria. Although mostly isolated from salted food, their natural habitats are hypersaline waters of intermediate levels of salt concentration, and hypersaline soils. In hypersaline waters, the taxonomic groups found are the ones that also predominate in ocean waters, such as representatives of the genera Vibrio, Pseudomonas and Flavobacterium . However, in hypersaline soils, the taxonomic groups present are those typical of normal soils, such as Pseudomonas, Bacillus and Gram-positive cocci. The halophilic bacteria from soils are also more resistant to exposure to low salt concentrations than the organisms isolated from waters. Therefore, it seems that the general characteristics of the hypersaline environments drastically affect the types of halophilic bacteria present, and that the halophilic character has arisen in many phylogenetic groups of eubacteria.  相似文献

Photosynthetic reaction centers from a variety of organisms have been isolated and characterized. The groups of prokaryotic photosynthetic organisms include the purple bacteria, the filamentous green bacteria, the green sulfur bacteria and the heliobacteria as anoxygenic representatives as well as the cyanobacteria and prochlorophytes as oxygenic representatives. A number of the cultural and biochemical criteria of the more resistant enterococci were used collectively to speciate 2,477 enterooocci strains isolated from water, soil, vegetation, sewage and human and animal faeces into 2 major groups: Streptococcus faecalis and S. faecium. Streptococcus faecalis was found to predominate in the human and poultry gut, S. faecium predominated in the livestock gut, while both organisms were about equally demonstrated in water, soil, vegetation and sewage samples.  相似文献

本文首次应用定位、定量方法对23例小腿部慢性湿疹患者皮损、邻近皮肤及鼻孔的菌群,从微生态学的角度进行探讨,并将结果同24例正常人小腿皮肤及鼻孔的菌群做了比较。其结果表明:金黄色葡萄球菌为皮损及邻近皮肤的主要菌种,并且其分离率及密度有按正常皮肤、邻近皮肤、皮损依次增高现象;类白喉杆菌的数量在皮损部位明显低于正常皮肤与邻近皮肤;表皮葡萄球菌分离率皮损及邻近皮肤均明显降低。本组结果同正常对照组相比其皮肤微生物群菌谱虽无不同,但其数量的变异是显著的,这对探讨湿疹的病因及用生态制剂等调整皮肤菌群使其恢复微生态平衡达到防治目的有所裨益。  相似文献

Neisseria catarrhalis produces arylamidase intracellularly and is one of the gram-negative bacteria producing exceptionally large amounts of this enzyme.In general, gram-positive bacteria do not produce this enzyme. Arylamidase from N. catarrhalis was purified by salt fractionation, chromatography, and density gradient ultracentrifugation. Its sedimentation coefficient was 6.6; l-alanine-beta-naphthylamide (betaNA) was the most rapidly hydrolyzed amino acid-betaNA. The enzyme had pK(e) values of 6.1 and 8.7 and pK(es) values of 7.1 and 7.9; only those amino acid-betaNA compounds of the l configuration were susceptible to hydrolysis. Arylamidase catalyzed stepwise hydrolysis of dipeptide-betaNA, beginning with the N-terminal residue. Substrates having amino acid residues with larger R groups, such as leucine, interacted much more effectively with enzyme. The significance of the predominate occurrence of arylamidase activity in gram-negative bacteria and the role of this enzyme in the physiology of these organisms remain unclear. It has been established, however, that arylamidase is distinct from leucine aminopeptidase.  相似文献

Members of a consortium of bacteria, isolated from the rumen of sheep, that degrades pyrrolizidine alkaloids (PAs) found in tansy ragwort (Senecio jacobaea) were characterized. An enrichment of ruminal bacteria was isolated from a sample of ruminal fluid using standard anaerobic techniques. The PA degradative capacity of the enrichment was tested by spiking purified PA extract from tansy ragwort. Length heterogeneity analysis by PCR (LH-PCR) and restriction fragment length polymorphism (RFLP) analysis was used to identify members of the consortium. Phylogenetic analysis of the 16S rDNA gene revealed differing results based on the molecular method used. LH-PCR identified 7 different organisms in 3 groups while RFLP identified 6 organisms with differing banding patterns in 5 groups. After the phylogenetic analyses of both methods were combined, the combined isolates represented 6 groups. The majority of the members of this consortium are <97.0% homologous with known bacteria, indicating this consortium may contain novel organisms able to detoxify PAs found in tansy ragwort. Further understanding of the metabolic pathways used by this consortium to degrade PAs could lead to the use of the consortium as a probiotic therapy for livestock and horses afflicted with tansy ragwort toxicosis.  相似文献

Cellulose degrading bacteria were isolated from brackish Phragmites reed beds near the Humber Estuary. Of 23 strains brought into pure culture, all developed characteristic differentiated colonies on certain media. On the basis of colour of the colonies (pseudosori) the organisms could be allocated to two groups, XMo and XMb. Related strains were isolated from a similar habitat on the Dee Estuary. Numerical taxonomy showed the group to be relatively diverse but, with a single exception, the XMo and XMb organisms appeared in separate clusters. The organisms were weakly motile or non-motile and certain cells had a polar flagellum. Pigments isolated from one strain had u.v. spectral characteristics similar to those of the xanthomonadins; the organisms may therefore be related to Xanthomonas .  相似文献

S ummary : The classification of 1,466 bacteria, isolated on Yeastrel-milk agar incubated at 30° from 68 rinses of steam sterilized dairy equipment taken at five farms, showed that micrococci were dominant, with corynebacteria and aerobic sporeforming rods frequent, representatives of these three groups constituting 73% of the isolates. The microflora was characterized by the dominance of organisms which were relatively inactive in milk. Typical milk souring organisms such as lactic streptococci and coli-aerogenes organisms were rarely isolated, but anaerogenic Gram-negative rods resembling Achromobacter, Pseudomonas and Flavobacterium were not uncommon on milking machine clusters and on a cooling unit. The glass recorder jars contained a relatively high proportion of sporeformers.  相似文献

Summary Differences have been noted in metabolic activity of bacteria isolated from water, mud and from Canadian pondweed (Elodea canadensis).The epiphytic bacteria showed the greatest metabolic activity in every instance, benthic bacteria were least active and the water isolates were intermediate.Non-chromogenic bacteria were metabolically more active than the chromogens.Casamino acids proved to be the most readily oxidized substrate among the compounds studied. Glucose, succinate, fumarate and gluconate were readily used by most epiphytic and planktonic bacteria. Acetate was intermediate and fructose was the least suitable substrate for these organisms. Casamino acids proved to be the best substrate for all three groups, although the benthic bacteria were less active with this substrate than the former two groups of bacteria. Gluconate, succinate, pyruvate and acetate were intermediate, and glucose and fructose were least suitable for most of the benthic bacteria.  相似文献

The ability of some bacteria isolated from environmental sources to grow in commonly administered infusion fluids was investigated. Infusion fluids may become contaminated with bacteria present in the environment in which they are being administered and the ability of such bacteria to proliferate within commonly used infusion fluids is therefore clinically significant. Bacteria present in the air and sink drains in wards at the Ife State Hospital were isolated, identified, and comprised fifteen Gram-positive and five Gram-negative organisms. Fourteen of the organisms obtained were introduced into sterile distilled water and five different infusion fluid preparations and were monitored over a 48 h period. It was found that, in general, Gram-positive organisms did not grow in the fluids used, while Gram-negative organisms were able to proliferate in the same fluids. Some Gram-positive organisms showed remarkable ability to remain viable in infusion fluids for more than 48 h.  相似文献

Bacteria from several groups of marine organisms were isolated and, using direct antibiograms, identified those that produce antibacterial substances, using a human pathogenic strain of Staphylococcus aureus ATCC6538 as revealing microorganism. Bacteria which produce substances that inhibited S. aureus growth were identified through morphological, physiological and biochemical tests. Out of 290 bacteria, 54 (18.6%) inhibited the growth of S. aureus, but only 27 survived for identification. Bivalves, sponges and corals were the most represented from which 41.2, 33.3 and 29.7%, respectively, produced antibacterial substances of the isolated bacteria in each group. The marine species with highest proportions of these bacteria were the hard coral Madracis decactis (62.5%), the sponges Cliona sp. (57.1%) and the octocoral Plexaura flexuosa (50.0%). Out of the 27 strains that produced antibacterial substances, 51.8% were Aeromonas spp. and 14.8% Vibrio spp. Marine bacteria that produce antibacterial substances are abundant, most belong in the Vibrionacea group and were isolated mainly from corals and bivalve mollusks.  相似文献

A total of 3819 bacterial cultures isolated from municipal water samples were identified using a combination of Enterotubules and confirmatory media. Frequency distributions for the different genera or groups of bacteria were similar for raw water and drinking water isolations, except for Escherichia organisms which doubled their frequency in raw water. Differences between the membrane filter (MF) and presence-absence (P-A) test with regard to types of organisms isolated were limited to Klebsiella organisms which were preferentially cultured from MF plates. Members of the genus Enterobacter were isolated more than twice as frequently as any of the other coliform genera dealt with in this study. Aeromonas organisms were detected almost as often as such individual genera as Escherichia, Citrobacter, or Klebsiella. Although non-lactose fermenting colonies (false-negatives) of the coliform genera would not be detected by the MF technique, their lack of detection would likely be offset by the Aeromonas colonies (false-positives). At least 25% of the coliform isolates were either anaerogenic or non-lactose fermenters and would therefore go undetected by the most probably number (MPN) technique.  相似文献

Lactobacillus and Bifidobacterium species were the predominant organisms isolated from small intestinal (jejunal) contents of rats, and lactic acid was the only organic acid detected. The numbers of cellulolytic bacteria in small intestines were low (approximately 10(3)/g). The fermentation in ceca was different from that in intestines, as, in addition to small amounts of lactic acid, high concentrations of volatile fatty acids were detected. The mixed cecal microflora was able to digest cellulose (pebble-milled Whatman no. 1) and cabbage. High numbers of cellulolytic bacteria were found (0.5 X 10(8) to 12.2 X 10(8)/g; 6% of total viable bacteria). The predominant celluloytic organism isolated was Bacteroides succinogenes. Ruminococcus flavifaciens was isolated from a few animals. The kinds and numbers of the predominant non-cellulolytic organisms isolated from rat ceca were similar to those described by previous workers.  相似文献

The serC-aroA operon of Salmonella gallinarum was isolated from a gene library using a labelled oligonucleotide probe and by complementation of an aroA Escherichia coli strain. The nucleotide sequence of a 2.6 kbp fragment was determined. The predicted amino acid sequence of the aroA gene product was compared to the equivalent sequence from ten other organisms. Computer-generated evolutionary trees clearly divide the eleven sequences into four different groups: Gram-negative bacteria, Gram-positive bacteria, fungi and plants. These trees depict a close evolutionary relationship between the sequences from Gram-negative bacteria and higher plants.  相似文献

Two distinct bacteria capable of freezing nucleus activity at temperatures as warm as –2.5°C were repeatedly isolated from citrus and avocado plants in Israel. Phage typing has shown them to be related to each other and to one of the two groups of ice-nucleation-active bacteria already described by other workers. Evidence suggesting that these organisms can cause frost damage to the plants they inhabit at temperatures not far below freezing was also obtained.  相似文献

Samples were taken at random intervals over a 2-year period from urban air and tested for viable bacteria. The number of bacteria in each sample was determined, and each organism isolated was identified by its morphological and biochemical characteristics. The number of bacteria found ranged from 0.013 to 1.88 organisms per liter of air sampled. Representatives of 19 different genera were found in 21 samples. The most frequently isolated organisms and their percent of occurence were Micrococcus (41%), Staphylococcus (11%), and Aerococcus (8%). The bacteria isolated were correlated with various weather and air pollution parameters using the Pearson product-moment correlation coefficient method. Statistically significant correlations were found between the number of viable bacteria isolated and the concentrations of nitric oxide (-0.45), nitrogen dioxide (+0.43), and suspended particulate pollutants (+0.56). Calculated individually, the total number of Micrococcus, Aerococcus, and Staphylococcus, number of rods, and number of cocci isolated showed negative correlations with nitric oxide and positive correlations with nitrogen dioxide and particulates. Statistically significant positive correlations were found between the total number of rods isolated and the concentration of nitrogen dioxide (+0.54) and the percent relative humidity (+0.43). The other parameters tested, sulfur dioxide, hydrocarbons, and temperature, showed no significant correlations.  相似文献

Naturally fermented and raw foods contain a range of organisms that may have benefit as additives in some foods and food processing. In particular, potential antifungal properties of these organisms may be potentially utilised as natural alternatives to chemical additives used to delay and prevent spoilage by fungi and yeast. This study examined 12 novel bacteria previously isolated from food as possible biopreservatives. The bacteria from the lactobacilli and dairy propionibacterium groups were tested by agar overlay method for their ability to inhibit the growth of 10 fungi and one yeast commonly associated with food contamination. Eight among eleven tested lactic acid bacteria demonstrated broad spectrum of antifungal activity. Strong fungi inhibition was also demonstrated by the dairy propionibacterium, but efficacy was growth medium dependant. Only one fungi, Geotrichum candidum was highly resistant to the bacteria. Variation between the inhibition results for different bacteria identifies the importance of careful strain selection, and the benefits of strain combinations when selecting biopreservatives for foods. Our abilities to assess health risks to free-ranging dolphin populations, to treat live-stranded or captive dolphins, and to evaluate the risks of disease transmission between humans and dolphins have suffered from a lack of basic information on microorganisms associated with normal, presumably healthy free-ranging individuals. In order to provide these data, we sampled free-ranging bottlenose dolphins (Tursiops truncatus) off Florida, Texas, and North Carolina during 1990-2002. Blowhole and anal/fecal samples yielded 1,871 bacteria and yeast isolates and included 85 different species or groups of organisms. Vibrios, unidentified pseudomonads, Escherichia coli, Staphylococcus spp., and a large group of nonfermenting gram-negative bacteria represented >50% of isolates. Vibrio alginolyticus and Vibrio damsela were the most commonly recovered bacteria from both anal/fecal and blowhole samples. Many organisms occurred sporadically in dolphins that were sampled repeatedly, but some were consistently isolated from individual animals and may indicate the carrier state. Vibrios were common, but some geographic variability in the presence of these and other organisms was noted. Potential pathogens of significance to humans and other animals were recovered.  相似文献

We investigated the diversity, distribution, and phenotypes of uncultivated Chloroflexaceae-related bacteria in photosynthetic microbial mats of an alkaline hot spring (Mushroom Spring, Yellowstone National Park). By applying a directed PCR approach, molecular cloning, and sequence analysis of 16S rRNA genes, an unexpectedly large phylogenetic diversity among these bacteria was detected. Oligonucleotide probes were designed to target 16S rRNAs from organisms affiliated with the genus Chloroflexus or with the type C cluster, a group of previously discovered Chloroflexaceae relatives of this mat community. The application of peroxidase-labeled probes in conjunction with tyramide signal amplification enabled the identification of these organisms within the microbial mats by fluorescence in situ hybridization (FISH) and the investigation of their morphology, abundance, and small-scale distribution. FISH was combined with oxygen microelectrode measurements, microscope spectrometry, and microautoradiography to examine their microenvironment, pigmentation, and carbon source usage. Abundant type C-related, filamentous bacteria were found to flourish within the cyanobacterium-dominated, highly oxygenated top layers and to predominate numerically in deeper orange-colored zones of the investigated microbial mats, correlating with the distribution of bacteriochlorophyll a. Chloroflexus sp. filaments were rare at 60 degrees C but were more abundant at 70 degrees C, where they were confined to the upper millimeter of the mat. Both type C organisms and Chloroflexus spp. were observed to assimilate radiolabeled acetate under in situ conditions.  相似文献
