A simple and easy transformation strategy was accomplished on field growing plants of Phyllanthus amarus, an anti-hepatitis B drug plant. Infection of Agrobacterium rhizogenes strains A4M70GUS and ATCC 15834 on decapitated shoots of field growing P. amarus induced hairy roots and crown gall, respectively. Infection with A4M70GUS yielded a mean of 23.2 roots from 40% plants in 40-day period. The crown gall induced on 30% plants after infection
with ATCC 15834 grew to 5–10 mm in diameter. The roots and crown galls established in vitro on Murashige and Skoog (MS) basal
medium grew well. The hairy roots yielded fivefold (6.91 g) biomass in half-strength MS liquid medium to that of the adventitious
roots derived from internode explants in MS medium with 8.0 μM α-naphthaleneacetic acid (1.39 g). Histochemical assay and
PCR analysis using the primers of uidA coding region confirmed the hairy roots induced by A4M70GUS. The crown galls induced by ATCC 15834 were confirmed by PCR analysis using rolB gene primers. The protocol enables an easy and early accomplishment of hairy roots. 相似文献

A truncated form of the Ti-plasmid virE2 gene from Agrobacterium tumefaciens strains C58 and A6, and A. vitis strain CG450 was transferred and expressed in somatic embryos of grapevine rootstocks 110 Richter (Vitis rupestris × V. berlandieri), 3309 Couderc (V. rupestris × V. riparia) and Teleki 5C (V. berlandieri × V. riparia) via Agrobacterium-mediated transformation to confer resistance to crown gall disease. Transformation was confirmed in 98% of the 322 lines
by enzyme-linked immunosorbent assay for the neomycin phosphotransferase II protein and 97% of 295 lines by polymerase chain
reaction for the truncated virE2 transgene. Southern blot analysis revealed the insertion of truncated virE2 at one to three loci in a subset of seven transgenic 110 Richter lines. In vitro resistance screening assays based on inoculations
of shoot internode sections showed reduced tumorigenicity and very small galls in 23 of 154 transgenic lines. Non-transformed
controls had a 100% tumorigenicity rate with very large galls. Disease resistance assay at the whole plant level in the greenhouse
revealed seven transgenic lines (3 lines of 110 Richter, 2 lines of 3309 Couderc and 2 lines of Teleki 5C) were resistant
to A. tumefaciens strain C58 and A. vitis strains TM4 and CG450 with a substantially reduced percentage of inoculation sites showing gall as compared to controls.
No association was found between the level of resistance to crown gall disease and the source Agrobacterium strain of virE2. Taken together, our data showed that resistance to crown gall disease can be achieved by expressing a truncated form of

The life cycle and developmental stages of Aylax hypecoi (Trotter, 1913, Hymenoptera: Cynipidae: Aylacini) were studied in detail. Aylax hypecoi is known to induce galls in fruits of two Hypecoum species — H. imberbe and H. geslini (Papaveraceae) and the larva develops in host plant fruits. The morphology and development of egg, larva and pupa were investigated, which
has previously not been done. The shape and size of terminal-instar larvae and associated galls are sex-specific. Overwintering

The yeast genes IXR1 and HMO1 encode proteins belonging to the family of chromatin nonhistone proteins, which are able to recognize and bind to irregular
DNA structures. The full deletion of gene IXR1 leads to an increase in cell resistance to the lethal action of UV light, γ-rays, and MMS, increases spontaneous mutagenesis and significantlly decreases the level of UV-induced mutations. It was earlier
demonstrated in our works that the hmo1 mutation renders cells sensitive to the lethal action of cisplatin and virtually does not affect the sensitivity to UV light.
Characteristically, the rates of spontaneous and UV-induced mutagenesis in the mutant are increased. Epistatic analysis of
the double mutation hmo1 ixr1 demonstrated that the interaction of these genes in relation to the lethal effect of cisplatin and UV light, as well as UV-induced
mutagenesis, is additive. This suggests that the products of genes HMO1 and IXR1 participate in different repair pathways. The ixr1 mutation significantly increases the rate of spontaneous mutagenesis mediated by replication errors, whereas mutation hmo1 increases the rate of repair mutagenesis. In wild-type cells, the level of spontaneous mutagenesis was nearly one order of
magnitude lower than that obtained in cells of the double mutant. Consequently, the combined activity of the Hmo1 and the
Ixr1 proteins provides efficient correction of both repair and replication errors. 相似文献

Experiments carried out on mice demonstrated that administration of a platinum-based drug, cisplatin, extensively used in
anticancer chemotherapy, exerts significant hyperalgesic effects; it intensifies both phases of pain behavioral reactions
induced in the formalin test. When introduction of cisplatin was combined with i.p. injections of 100 mg/kg of an aqueous-alcoholic
extract from the leaves of Salvia officinalis, the second phase of cisplatin-enhanced pain in the formalin test was effectively suppressed; the effect was comparable with

Crown gall, caused by Agrobacterium tumefaciens, causes severe damage to apple saplings resulting in weak growth and loss of commercial value. Developing molecular markers
linked to crown gall resistance genes, and establishing a marker-assisted selection (MAS) for such a trait would be an effective
way to improve rootstock breeding for crown gall resistance. The wild apple Malus sieboldii Sanashi 63 carries the crown gall resistance gene Cg effective against the A. tumefaciens strain Peach CG8331 (biovar 2). Applying the genome scanning approach on the mapping population JM7 (cgcg) × Malus sieboldii Sanashi 63 (Cgcg), Cg was mapped on the linkage group (LG) 2. The constructed linkage map of LG 2 of Sanashi 63 spans 59.8 cM and has an average
marker density of 3.5 cM per marker. The 191 bp allele of the simple sequence repeat (SSR) NZmsEB119405 co-segregated perfectly
with Cg in a segregating population of 119 individuals. Quantitative trait loci, accounting for 75.3% to 84.3% of phenotypic variation
were detected in the same position. Testing eight additional rootstocks with the NZmsEB119405 SSR marker revealed that the
191 bp allele is also present in crown gall-susceptible rootstock accessions. Only the markers CH03b01 and NZmsPal92 mapping
at 0.9 and 4.3 cM from Cg, respectively, showed “private” alleles associated to Cg. 相似文献

Gall induction by arthropods results in a range of morphological and physiological changes in their host plants. We examined changes in gas exchange, nutrients, and energetics related to the presence of stem galls on Parthenium hysterophorus L. (Asteraceae) induced by the moth, Epiblema strenuana Walker (Lepidoptera: Tortricidae). We compared the effects of galls on P. hysterophorus in the rosette (young), pre-flowering (mature), and flowering (old) stages. Gall induction reduced the leaf-water potential, especially in flowering stage plants. In young and mature stage plants, galling reduced photosynthetic rates considerably. Gall induction reduced the transpiration rate mostly in mature plants, and this also diminished stomatal conductance. Energy levels in most galls and in shoot tissue immediately below the galls were significantly higher than the energy levels in stem tissue immediately above the galls, indicating that the gall acts as a mobilizing sink for the moth. Galling had significant effects on concentrations of minerals such as boron, chloride, magnesium, and zinc. In galled plants, reduced leaf-water potential and reduced rates of photosynthesis, transpiration, and stomatal conductance may have altered mineral element levels. These observed effects demonstrate that E. strenuana has the potential to regulate P. hysterophorus. 相似文献

A protocol for in vitro propagation of the wild germander (Teucrium polium L.) was developed. In vitro plants were developed from ex vitro axillary buds. Then, shoot tips were excised and established on Murashige and Skoog medium. Proliferation of shoots was tested
with different levels of 6-furfurylaminopurin, 6-benzyladenine, or thiadiazuron. The highest proliferation of T. polium was obtained when 6-benzyladenine and 6-furfurylaminopurin were used at 2.0 and 1.6 mg l−1, respectively. Thiadiazuron gave the lowest response for shoot proliferation. Rooting was experimented at different levels
of Indol-3-butric acid, Indol-3-acetic acid, or 1-naphthaleneacetic acid. 1-Naphthaleneacetic was the only growth regulator
which promoted root induction. Rooted plants were acclimatized successfully with 75% survival and grown in the greenhouse.
In vitro- and in vivo-grown plants were analyzed for essential oil production. In vitro-grown T. polium on MS medium supplemented with 6-benzyladenine and 1-naphthaleneacetic gave higher oil yield than that grown on hormone-free
Murashige and Skoog medium. In vivo (wild)-grown T. polium produced different oil yield when collected in different months (April and October). β-caryophyllene, used as a marker compound
in the essential oil, was identified and quantified by gas chromatography (GC) analysis. Gas chromatography/mass (GC-MS) spectrometry
analysis was also used to identify other components of in vitro cultures and to compare with in vivo-grown plants. 相似文献

Thermal hysteresis proteins (Thps) known as antifreeze proteins for their antifreeze activity, depress the freezing point
of water below the melting point in many polar marine fishes, terrestrial arthropods and plants. For the purpose of breeding
cold-resistant plants, we designed to introduce the Thp gene into the plants. The physiological and biochemical effect of
high-lever expression of the modified Choristoneura fumiferana
Thp (ThpI) in Arabidopsis thaliana plants was analyzed. Under low temperature stress, the ThpI transgenic plants exhibited stronger growth than wild-type plants. The elevated cold tolerance of the ThpI over-expressing plants was confirmed by the changes of electrolyte leakage activity, malonyldialdehyde and proline contents.
These results preliminarily showed that the Thp possibly be used to enhance the low temperature-tolerant ability of plants. 相似文献

The medicinal plant, Nothapodytes foetida, contains a number of important alkaloids like camptothecin (an anticancer drug molecule), but its concentration is less
to meet the existing demand of this important molecule, in an effort for accessible availability of camptothecin. An endophyte
(designated ZP5SE) was isolated from the seed of Nothapodytes foetida and was examined as a potential source of anticancer drug lead compound, i.e., camptothecin, when grown in Sabouraud liquid
culture media under shake flask conditions. The presence of an anticancer compound (camptothecin) in this fungus was confirmed
by chromatographic and spectroscopic methods in comparison with authentic camptothecin. Isolated endophyte (Neurospora crassa) producing camptothecin may become an easily accessible source for the production of a precursor anticancer drug molecule
in the future at a large scale.

In vitro regeneration of black nightshade (Solanum nigrum L.) plants was achieved through callus-mediated shoot organogenesis followed by 30 d indoor ex vitro adaptation to nutritional stress under environmental ambience and thereafter 6-d outdoor acclimatization in pots prior to
field establishment. Relevant physiological parameters including pigment content, chlorophyll a fluorescence, net photosynthetic rate (P
N), transpiration rate (E), and stomatal conductance (g
s) of in vitro-regenerated plants were investigated during the course of ex vitro adaptation. During the first 4 d of indoor transplantation to potting substrate, there was a marginal reduction in the leaf
chlorophyll and carotenoid contents but P
N and E were strongly reduced. The stomatal conductance and E/P
N ratio were significantly higher in plants up to 20 d of indoor adaptation than those of comparable age grown naturally from
seeds. The shape of the OJIP fluorescence transient varied significantly with acclimatization, and the maximum change was
observed at 2.0 ms. The 2.0 ms variable fluorescence (V
j), 30 ms relative fluorescence (M
0), photon trapping probability (TR0/Abs), and photosystem II (PSII) trapping rate (TR0/RC) showed initial disturbance and subsequent stabilization during 30 d of indoor acclimatization. Energy dissipation (DI0/RC) and electron transport probability (ET0/TR0) showed an initial phase of increase during the 4 d after plants were transplanted outdoors. During the 6-d outdoor acclimatization
after transfer of plants to soil, no significant change in total chlorophylls and carotenoids, E, and g
s were observed, but P
N improved after reduction on the first d. The OJIP-derived parameters experienced change on the first d but were stabilized
quickly thereafter. There was no significant difference between outdoor acclimatized plants and those of the seed-grown plants
of comparable age with respect to photosynthetic and fluorescence parameters. Direct transfer of plants without indoor acclimatization,
however, showed a completely different trend with respect to P

The saprophytic bacterium Burkholderia
cepacia has been shown to play an active role as plant growth promoting bacteria (PGPB). In this study, the ability of cell-free
culture medium (CFCM) of B. cepacia to improve early developmental stages of plants has been assessed on two agronomically important crops, maize (Zea
mays) and rice (Oryza
sativa). Treating maize and rice seeds for 45 min before germination significantly improved seed germination and consequent seedling
growth. The effect of CFCM was confirmed by the increased biomass of the shoot and, mainly, the root systems of treated seedlings.
Chromatographic characterization of the CFCM revealed that the spent culture medium of B. cepacia is a complex mix of different classes of metabolites including, among others, salicylic acid, indole-3-acetic acid (IAA)
and several unidentified phenolic compounds. Fractionation of the CFCM components revealed that the impressive development
of the root system of CFCM-treated seedlings is due to the synergistic action of several groups of components rather than
IAA alone. The data presented here suggest that a CFCM of B. cepacia can be used to improve crop germination. 相似文献

A leaf beetle, Crioceris sp. (Coleoptera: Chrysomelidae), was introduced into Australia as a biological control agent of bridal creeper (Asparagus asparagoides L. Druce) during October 2002. Rearing of Crioceris sp. is labour intensive therefore all releases of Crioceris sp. have been under 1000 individuals, which may be too low to ensure establishment if high mortality and high competition
with other agents occurs. The aim of this study is to understand how the presence of two well-established biocontrol agents,
a rust fungus (Puccinia myrsiphylli (Thuem) Wint [Basidiomycota: Uredinales]) and a leafhopper (Zygina sp. [Hemiptera: Cicadellidae]), might influence Crioceris sp. establishment. Crioceris sp. neonate larvae were placed on bridal creeper plants with or without the leafhopper and/or rust. The number of larvae
that pupated was reduced by 38 and 65% in the presence of the rust fungus and leafhopper, respectively and by 45% in the presence
of both agents. As the area infected by the rust increased the area damaged by the leafhopper decreased. The rust appeared
to be negatively impacted by the presence of the leafhopper. In a second experiment, female Crioceris sp. adults were given a choice between uninfested bridal creeper plants and those infested with the rust or the leafhopper.
The females preferred to lay their eggs on plants without leafhoppers but did not seem to be deterred by the presence of the
rust. Consequently, the performance and impact of Crioceris sp. on bridal creeper may be reduced if populations overlap with the other biocontrol agents in the field. 相似文献

Rice gene Oryza sativa Drought Stress Response-1 (OsDSR-1) was one of the genes identified to be responsive to drought stress in the panicle of rice at booting and heading stages
by both microarray and quantitative real-time PCR analyses. OsDSR-1 encodes a putative calcium-binding protein, and its overexpression in Arabidopsis rendered transgenic plants to produce much shorter lateral roots (LRs) than wild-type (WT) plants in the medium supplemented
with abscisic acid (ABA), suggesting that OsDSR-1 may act as a positive regulator during the process of ABA inhibition of LR development. No significant difference was observed
in the total LR length between WT and transgenic plants in the media with the increase of only osmotic stress caused by NaCl,
LiCl, and mannitol, while transgenic Arabidopsis seedlings appeared to produce larger root systems with longer total LR lengths under high-potassium conditions than WT seedlings.
Further analysis showed that external Ca2+ was required for the production of larger root systems, indicating that the promotion by OsDSR-1 of the LR development of transgenic Arabidopsis seemed to occur in a Ca2+-dependent manner under high-potassium conditions. We propose that OsDSR-1 may function as a calcium sensor of the signal transduction pathway controlling the LR development under high-potassium conditions. 相似文献

Sap flow rate was measured in the crown of a solitary specimen of downy oak (Quercus pubescens) infested by mistletoe (Loranthus europaeus). Five oak branches and two mistletoe plants were selected for analysis. The seasonal sum of transpired water expressed per leaf area unit was five times higher in the mistletoe than in the oak. In addition, the diurnal curves of sap flow were different between the plants. In the morning, the sap flow measured in the mistletoe lagged one hour behind the sap flow measured in an oak branch unencumbered by mistletoe. In contrast, no time lag was observed in the evening. The proportion of water transpired at night relative to the total transpiration was 7% in both species. The stomatal conductances derived from the inverted Penman-Monteith equation and their dependence on global radiation and the vapour pressure deficit (D) revealed that D exerts a different behaviour in stomatal control of transpiration in the mistletoe. We also determined that the concentration of calcium in the leaf mass could serve as a proxy for transpiration rate, however the relationship was not proportional. 相似文献

The research on incidence of mistletoe (Viscum album ssp. abietis) on silver fir (Abies alba) was carried out in natural fir stands in Croatia. In the area of Gorski Kotar 32.8 % of the examined dominant fir trees
were infected with mistletoe. The mistletoe incidence was presented according to the damage degrees of silver fir from 0 (healthy
trees) to 4 (dead trees), rated by the crown defoliation and needle discoloration. With the increase of incidence, mistletoe
spreads more onto more vital, i.e. less damaged trees. In the Dinaric Alps 28.6% of the examined fir trees were infected,
this percentage amounting to 27.1% for the mountainous regions between the Sava and Drava rivers.
The site and stand parameters (exposure, elevation, site quality, forest community and crown closure) were analysed in order
to establish whether there was a correlation between these parameters and mistletoe incidence. There was a negative correlation
between the elevation and mistletoe incidence. Among the compartments with a closed stands there were considerably less compartments
with higher mistletoe incidence than among compartments with a sparsely closed and understocked stands. The other analysed
site and stand parameters individually had no significant influence on mistletoe incidence.
The correlation of silver fir mortality in 2004 and mistletoe incidence in 2002/03 was analysed, and their strong correlation
was established. Mistletoe could be considered as a bioindicator of silver fir decline, and probably a significant contributor
to that decline. In the areas where mistletoe incidence is great it can be presumed that silver fir is significantly damaged. 相似文献

A transfer DNA (T-DNA) carrying the marker gene nptII was detected in the genomes of diploid and haploid maize plants obtained after the treatment of pistil filaments with a
suspension of Agrobacterium during artificial pollination. PCR analysis of total DNA isolated from 155 canamycin-resistant diploid F1 seedlings revealed
T-DNA insertions in the genomes of 111 plants (32.7% of the total number of analyzed seeds). The example of matroclinal haploids
was used to demonstrate that T-DNA may be transported to the egg cell by the growing pollen tube (PT). Twelve out of 16 analyzed
haploid plants contained the T-DNA insertion. The possible mechanism of the transfer of the Agrobacterium T-DNA to the maize genome during pollination is discussed. 相似文献

Mature seed-derived embryogenic calli of indica rice (Oryza sativa L. cv. PAU201) were induced on semisolid Murashige and Skoog medium supplemented with 2.5 mg dm−3 2,4-dichlorophenoxyacetic acid + 0.5 mg dm−3 kinetin + 560 mg dm−3 proline + 30 g dm−3 sucrose + 8 g dm−3 agar. Using OsglyII gene, out of 3180 calli bombarded, 32 plants were regenerated on medium containing hygromycin (30 mg dm−3). Histochemical GUS assay of the hygromycin selected calli revealed GUS expression in 50 % calli. Among the regenerants,
46.87 % were GUS positive. PCR analysis confirmed the presence of the transgene of 1 kb in 60 % of independent plants. Further,
these plants have been grown to maturity in glasshouse. In vitro screening for salt tolerance showed increase in fresh mass of OsglyII putative transgenic calli (185.4 mg) as compared to control calli (84.2 mg) on 90 mM NaCl after 15 d. When exposed to 150
mM NaCl, OsglyII putative transgenic plantlets showed normal growth while the non-transgenic control plantlets turned yellow and finally did

Genes of CLE (CLAVATA3/ESR-related) family encode peptide ligands that regulate plant development in response to external stimuli such as rhizobial
infection and the nitrate application as well as various internal stimuli. To investigate whether LjCLE gene(s) may involve in plant response to inorganic phosphate (Pi), we analyzed Pi responses of 39 LjCLE genes in hydroponically grown Lotus japonicus plants (ecotype Miyakojima ‘MG-20’). Two LjCLE genes, LjCLE19 and 20, were up-regulated specifically and greatly in roots of L. japonicus by Pi addition to the hydroponic solution. When the external Pi level increased, expressions of LjCLE19 and 20 increased before the increase in the Pi content in plants. On the other hand, when the external Pi level decreased, the Pi
content in plants decreased first, then expression levels of LjCLE19 and 20 decreased. Based on our results, we discuss the relationship between LjCLE19 and 20 and the tissue Pi levels in plants. This is the first report showing induction of specific CLE genes by phosphate. 相似文献
