研究了家蝇对三氟氯氰菊酯抗性的遗传,通过对抗性品系和敏感品系杂交后代的抗性遗传分析发现家蝇对三氟氯氰菊酯的抗性受多因子控制,抗性显性率为-0.102,为不完全隐性。其对三氟氯氰菊酯的抗性现实遗传力为0.120。  相似文献

为了了解并掌握马铃薯甲虫Leptinotarsa decemlineata (Say)在新疆的抗性水平及动态, 为科学用药提供理论依据, 利用点滴法测定了新疆北部马铃薯甲虫6个田间种群(乌鲁木齐、 阜康、 昌吉、 阿勒泰、 尼勒克和特克斯) 的4龄幼虫对14种常规药剂的敏感性, 并测试了磷酸三苯酯(TPP)、 顺丁烯二酸二乙酯(DEM)和氧化胡椒基丁醚(PBO)的增效作用。结果表明: 与已报道的基线相比, 特克斯种群对几种药剂都敏感, 因此确定特克斯田间种群为敏感品系。与特克斯敏感品系相比, 昌吉种群对三氟氯氰菊酯、 溴氰菊酯、 克百威及丁硫克百威的抗性倍数分别为108.8, 30, 17.6和24.7, 而尼勒克种群对这4种药剂的抗性倍数分别为29.4, 42.5, 10.1和89.9。此外, 昌吉和阜康种群对高效氯氰菊酯也分别产生8.9和13.0倍的中等抗性。增效测定结果显示: 对于尼勒克种群, PBO对三氟氯氰菊酯有明显增效作用; 对于昌吉种群, PBO, TPP和DEM对克百威有明显增效作用。因此推断解毒酶活性增强可能是新疆马铃薯甲虫对三氟氯氰菊酯和克百威的抗性机制之一。由于增效作用不能完全消除抗性, 因此新疆马铃薯甲虫的抗药性还可能涉及到其他机制。  相似文献

采用浸液生测法研究了斜纹夜蛾Spodoptera litura巴基斯坦抗性种群中酶抑制剂[胡椒基丁醚(PBO)和脱叶膦(DEF)]对丙溴磷、灭多威、硫双灭多威、氯氰菊酯、氯氟氰菊酯、联苯菊酯、茚虫威和多杀菌素等杀虫剂的增效作用。结果表明:PPO和DEF对氨基甲酸酯杀虫剂灭多威和硫双灭多威均具有增效作用,但对有机磷杀虫剂丙溴磷不具有增效作用。两种抑制剂对氯氰菊酯均产生增效作用,但对联苯菊酯没有增效作用。PPO 和DEF增加了氯氟氰菊酯对Multan种群的毒性,但没有增加其对Mailsi种群的毒性。DEF对多杀菌素具有增效作用,但PBO对其没有增效作用。PBO和DEF对氨基甲酸酯杀虫剂、拟除虫菊酯杀虫剂、茚虫威和多杀菌素具有明显的增效作用,这说明细胞色素P450单加氧酶和酯酶的解毒作用至少部分参与了斜纹夜蛾对这些杀虫剂的抗性过程。不过,两种增效剂对杀虫剂增效作用范围有限,暗示对于斜纹夜蛾巴基斯坦种群而言,其他的机制(如靶位点不敏感、表皮穿透作用降低)可能是更重要的抗性机制。  相似文献

化学防治是世界各地防治小菜蛾Plutella xylostella (L.)的主要方法。选择对小菜蛾各发育阶段高效的杀虫剂有助于很好地控制这一害虫。本研究用浸叶法测定了茚虫威和六伏隆对小菜蛾卵、幼虫和成虫的毒性。茚虫威和六伏隆对小菜蛾卵的LC50值分别为201.40 和 37.32 mg/L。茚虫威和六伏隆对3龄幼虫也有很好的毒性(茚虫威的LC50 为 4.82 mg/L; 六伏隆的LC50 为1.48 mg/L)。对成虫的毒性结果表明, 这两种杀虫剂对成虫均没有理想的毒性(茚虫威的LC50 为845.20 mg/L; 六伏隆的LC50 为3 438 mg/L)。根据计算的LC50值, 六伏隆对小菜蛾卵和幼虫的毒性比茚虫威高, 但是茚虫威对成虫的毒性更高。  相似文献

分别采用药膜法和浸叶法测定了菜蛾绒茧蜂Apanteles plutellae和小菜蛾Plutella xylostella对杀虫剂的敏感度。结果显示: 有机磷、氨基甲酸酯、拟除虫菊酯类杀虫剂、阿维菌素和锐劲特对菜蛾绒茧蜂高毒,而抑太保和Bt为低毒,然而,短时间(1 h)接触常规防治剂量的锐劲特、氰戊菊酯、氯氰菊酯和乙酰甲胺磷对菜蛾绒茧蜂低毒。增效剂胡椒基丁醚(PB)、磷酸三苯酯(TPP)和马来酸二乙酯(DEM)对菜蛾绒茧蜂的甲胺磷、克百威、氰戊菊酯、氯氰菊酯、阿维菌素和锐劲特敏感性增效显著,但对抑太保无增效作用。PB的增效作用显著高于TPP 和DEM。PB和TPP对菜蛾绒茧蜂羧酸酯酶(CarE),以及DEM对谷胱甘肽S转移酶(GST)具显著的活体抑制作用,但PB,TPP和 DEM对菜蛾绒茧蜂乙酰胆碱酯酶(AChE)无抑制作用。菜蛾绒茧蜂AChE的米氏常数(Km)、最大反应速度(Vmax)、CarE和GST活性分别为小菜蛾的0.22、2.08、4.60和0.45倍,甲胺磷、敌敌畏和克百威对菜蛾绒茧蜂AChE的双分子速度常数(Ki)分别为对小菜蛾的14.7、10.5 和26.0倍。酶与抑制剂反应温度增高将导致酶抑制率增高,尤其对菜蛾绒茧蜂AChE的抑制作用更为显著。上述结果表明,菜蛾绒茧蜂对有机磷和氨基甲酸酯类杀虫剂的高敏感性与其显著高的AChE敏感性有关,氧化代谢的解毒作用对菜蛾绒茧蜂耐药性的影响大于水解作用。此外,对小菜蛾和菜蛾绒茧蜂杀虫剂敏感性差异的毒理学原因进行了讨论。  相似文献

在伊朗, 麦扁盾蝽Eurygaster integriceps是小麦和大麦上的主要害虫。新烟碱类杀虫剂是一类具有较强活性的杀虫剂, 对刺吸式口器害虫具有良好的效果。本研究应用触杀和熏蒸两种生测方法测定了4种新烟碱类杀虫剂(噻虫嗪、吡虫啉、呋虫胺和啶虫咪)对麦扁盾蝽3龄若虫和成虫的毒效。触杀法生测结果显示, 各杀虫剂对麦扁盾蝽没有强烈的触杀活性, 但是在高浓度下(5 000 mg/L a. i.), 可见较高的死亡率。熏蒸法生测结果显示, 所有杀虫剂对麦扁盾蝽的若虫和成虫均有较好的毒性。结果提示应用熏蒸方法可能是治理这一重要害虫的新方法。  相似文献

以肥熟土垫旱耕人为土为供试土样,用分层土柱试验法研究了不同层次水分、氮、磷组合对冬小麦(Triticum aestivum L.)氮磷养分有效性和产量效应的影响.结果表明,不同土层水肥处理的氮磷养分有效性和产量效应差异显著.氮素养分有效性在4.73%~41.19%之间,磷素养分有效性在4.11%~13.58%之间.对氮素养分有效性,单施氮整体湿润时(0~90cm土层湿润)较上干下湿(0~30cm土层干旱胁迫,30~90cm土层湿润)低4.87%,而氮磷配施在整体湿润时较上干下湿高6.38%,差异均达显著水平;对磷素养分有效性,氮磷配施时,在整体湿润时较上干下湿增加5.01T(p<0.05).从不同施肥土层看,氮素养分有效性均以0~90cm土层施肥处理最高;对氮磷配施处理,在上干下湿时分别比0~30cm、30~60cm和60~90cm土层施肥处理高9.5%、10.1%和20.2个%;对磷素养分有效性,整体湿润处理,以0~30cm土层施肥显著高于其它土层施肥处理.单施氮或磷,上干下湿时氮磷养分的产量效应均高于整体湿润处理,但氮磷配施时均以整体湿润处理较高;从不同土层施肥看,氮素养分的产量效应以0~90cm土层施肥最高;磷素养分的产量效应则表现为0~90cm与0~30cm土层施肥处理显著高于30~60cm和60~90cm土层施肥处理.分析0~90cm土层残留硝态氮和有效磷累积量可以看出,不同处理土壤残留硝态氮含量存在显著差异,上干下湿时CK、单施氮、单施磷和氮磷配施土壤残留硝态氮分别比整体湿润相应施肥处理增加125.8%、20.1%、21.9%和2.1%;不同处理有效磷差异性不及硝态氮明显.整体看,在两种水分状况下,均以0~90cm和0~30cm土层施肥有利于提高氮磷养分对冬小麦的有效性和产量效应,减少硝态氮和有效磷在土壤中的残留累积.考虑到生产上的可操作性,仍以施入0~30cm土层最适,说明即使在上干下湿情况下,保证上层有效养分供应仍具重要作用.  相似文献

为了揭示CO2和O3浓度升高及其复合作用对植物活性氧(ROS)代谢及抗氧化酶活性的影响机理,以玉米(Zea mays L.)为研究材料,利用开顶式气室(OTCs)研究了CO2和O3浓度升高及其复合作用下,玉米叶片活性氧产生速率、含量,膜脂过氧化程度,抗氧化酶活性,净光合速率及玉米籽粒产量的变化.结果表明,在整个生育期内,与对照相比,高浓度CO2((550±20)μmol · mol-1)处理下,玉米叶片净光合速率升高,O- ·2 产生速率、H2O2含量下降, MDA含量、相对电导率减小,SOD、CAT、POD活性增强,玉米百粒重和穗粒数增加;而在O3浓度为(80±10)nmol · mol-1的条件下,玉米叶片净光合速率下降,O- ·2 产生速率、H2O2含量升高,MDA含量、相对电导率增大,SOD、CAT、POD活性减弱,玉米百粒重和穗粒数降低;CO2和O3浓度升高复合((550±20)μmol · mol-1 (80±10)nmol · mol-1)处理下,玉米叶片的净光合速率、H2O2含量、SOD活性先升高后降低, MDA含量、相对电导率、CAT活性增加,POD活性减弱,而O- ·2 产生速率几乎不变化,且玉米的百粒重和穗粒数略低于对照.以上结果说明,CO2浓度升高抑制了玉米叶片活性氧的代谢速率,提高了抗氧化酶的活性,从而增强了光合作用,使玉米籽粒产量增加,对玉米表现为保护效应,而O3浓度升高促进了玉米叶片活性氧的代谢速率,降低了抗氧化酶的活性,抑制了光合作用,使玉米籽粒产量下降,对玉米表现为伤害效应.在CO2和O3浓度升高复合处理下,CO2浓度升高在一定程度上缓解了O3浓度升高对玉米的伤害效应,而O3浓度升高亦在一定程度上削弱了CO2浓度升高对玉米的保护效应.  相似文献

Abstract  In this paper, we reported the differences in susceptibility to insecticides between adults and larvae of housefly, Musca domestica (L.), and the mechanisms for the differences. The larvae of housefly were much more tolerant to insecticides than the adults, and the tolerance ratio to cyhalothrin was as high as 205.5 for susceptible strain. Mechanism studies showed that higher GST activity was associated with higher insecticide tolerance in the larvae. The co-toxicity coefficient of the mixture of cyhalothrin and methylene dithiocyanate(4:1) on pyrehid-resistant houseflies was 188.  相似文献

Abstract  The induction of metallothionein (MT) was studied in the housefly larvae ( Musca domestica ). Upon dietary exposure to Cd, two Cd-binding proteins were isolated from the whole body homogenates, using gel filtration and ion exchange chromatography. Mass spectrometric measurement revealed that they have high purity and the molecular weight are 9045.9Da and 11560.2Da, respectively. Amino acid analysis showed that the content of cysteine is the highest, attaining to 18.2%. However, aromatic amino acid residues such as tyrosine (2.5%) and phenylalanine (3.1%) were also detected. In addition, MT from housefly larvae has strong heat stability. All these results suggest that the properties of MT isolated from housefly are very similar to that of mammalian MT, but some differences still exist.  相似文献

Abstract Acetylcholinesterase (AChE) in the susceptible (S) and the resistant (R) strains of housefly (Musca domestica) was investigated using kinetic analysis. The Vmax values of AChE for hydrolyzing acetylthiocholine (ATCh) and butyrylthiocholine (BTCh) were 4578.50 and 1716.08nmol/min/mg* protein in the R strain, and were 1884.75 and 864.72 nmol/min/mg. protein in the Sstrain, respectively. The Vmax ratios of R to S enzyme were 2.43 for ATCh and 1.98 for BTCh. The Km values of AChE for ATCh and BTCh were 0.069 and 0.034 mmol/L in the S strain, and 0.156, 0.059 mmol/L in the R strain, respectively. The Km ratios of R to S enzyme were 2.26 for ATCh and 1.74 for BTCh. The ki ratios of S to R enzyme for three insecticides propoxur, methomyl and paraoxon were 46.04, 4.17 and 2. 86, respectively. In addition, kcat and kcat/Km for measuring turnover and catalytic efficiency of AChE were determined using eserine as titrant. The kcat values of AChE from the R strain for both ATCh and BTCh were higher than those values from the S strain. But the values of kcat/Km were in contrary to the kcat values with R enzyme compared to S enzyme. The AChE catalytic properties and sensitivity to the inhibition by three insecticides in the R and S strains of housefly were discussed based on contribution of Vmax, Km, ki, kcat and kcat/Km. All these data implied that AChE from the R strain might be qualitatively altered. We also observed an intriguing phenomenon that inhibitors could enhance the activity of AChE from the resistant strain. This “flight reaction” of the powerful enzyme might be correlated with the developing resistance of housefly to organophosphate or carbamate insecticides.  相似文献

本文对家蝇抗性和敏感品系中的乙酰胆碱酯酶 (AChE)的催化特性进行了研究。抗性品系中AChE水解ATCh和BTCh的Vmax分别为 4578.50和 1 71 6.0 8;而敏感品系的Vmax则为 1 884.75和864.72nmol/min/mgprotein ;Vmax的比率 (R/S)对ATCh是 2 .2 6倍 ;对BTCh则是 1 .74倍。AChE的Km 值在敏感品系中分别是 0 .0 69和 0 .0 34;而在抗性品系中则分别是 0 .1 56和 0 .0 59mmol/L ;Km 的比率 (R/S)对ATCh是 2 .43倍 ;对BTCh则是 1 .98倍。此外 ,我们还用eserine作为滴定剂测定了AChE的转换数kcat和酶特异性常数kcat/Km,抗性品系中的AChEkcat值均比敏感品系的要高 ;而kcat/Km 值与此相反。本文着重分析了抗性品系与敏感品系间AChE的催化特性以及对残杀威、灭多威、对氧磷的敏感度差异 ,研究结果表明抗性品系中的AChE性质有可能发生变化。同时还观察到某些杀虫剂能增强抗性品系AChE的活力 ,我们认为这种“增强反应”可能与家蝇对有机磷或氨基甲酸酯类杀虫剂的抗性发展有关  相似文献

本文研究了溴氰菊酯对正常品系雌性家蝇Musca domestica vieina腹部微粒体细胞色素P-450的诱导作用.结果表明溴氰菊酯对P-450有一定的诱导作用,在低于LD50的波度范围内,浓度高的溴氰菊酯作用较明显,浓度低的溴氰菊酯在重复处理家蝇后也显示出诱导作用.这种诱导作用在24小时内有随时间的延长而增强的趋势.另外对照组与处理组的P-450 CO差光谱的特征吸收峰的波长均为451nm,表明诱导作用未改变P-450的光谱性质.  相似文献

本文运用活性炭结合试验测定了家蝇卵巢中保幼激素结合蛋白(JHBP)的含量.卵巢中JHBP对JHⅢ有较高的结合活性,其结合常数为2.1×10~(-8)M,~3H保幼激素Ⅲ(~3H-JH)和JHBP的结合可被未标记的JH Ⅲ抑制,但保幼激素的类似物ZR 512或ZR515均无抑制效应.卵巢中JHBP的含量在家蝇羽化后48小时达最大值,是羽化后60小时、72小时家蝇卵巢中JHBP含量的6.5倍和15.5倍.羽化后24小时、36小时的家蝇卵巢无JHBP的结合活性.若刚羽化的家蝇用JHⅢ处理,36小时后,JHBP则可检测到.本实验的结果表明卵巢中JHBP浓度的变动可能和JH对家蝇卵黄发生的作用有关.  相似文献

Abstract  By using charcocal binding assay, the juvenile hormone binding protein (JHBP) was determined in the ovaries of houseflies. This ovarian JHBP possesses high affinity with juvenile hormone III (JH III) and has a Kd of 2.1 III 10--8 M. The binding of 3H-juvenile hormone III (3H-JH III) to this protein was inhibited by unlablled JH III, but not by juvenile hormone analog ZR 512 or ZR 515. The level of this ovarian JHBP reached the highest in houseflies 48 h after emergence, and was 6. 5-fold and 15. 5-fold higher than that in housefIies 60 h and 72 h after emergence, respectively. No binding activity was detected in the ovaries of houseflies 24 h or 36 h after emergence. The absence of JHBP in the ovaries of houseflies 36 h after emergence could be reversed by applying JH III to newly emerged houseflies. The data suggest that the fluctuation of the JHBP concentration might associate with the action of juvenile hormone (JH) on housefly vitellogenesis.  相似文献

Associations between developmental stability, sexual selection, and viability selection were studied in the domestic fly Musca domestica (Diptera, Muscidae). Developmental stability of the wings and tibia of flies of both sexes, measured in terms of their level of fluctuating asymmetry, was positively associated with mating success in free ranging populations and in sexual selection experiments. Mated individuals may have obtained indirect fitness benefits from sexual selection of two different kinds. First, the entomopathogenic fungus Enthomophthora muscae (Zygomycetes, Entomophthorales) infects and kills adult domestic flies, and flies dead from fungus infections had more asymmetric wings than flies dead for other reasons. Experimental deposition of fungus spores on uninfected flies demonstrated that flies with asymmetric wings were more susceptible to fungus infections than flies with symmetric wings. Second, domestic flies were frequently eaten by insectivorous barn swallows Hirundo rustica, and flies depredated by birds had more asymmetric wings and tibia than surviving flies.  相似文献

在一定时间内,刚特拉津 (Atrazinc)对棉铃虫 Heliothis armigera 幼虫羧酸酯酶以及GSH-S-转移酶(GST)活性有明显的诱导作用,羧酸酯酶活性最高增加146%,GST增加280%。 对羧酸酯酶的诱导高峰时间要落后于GST,不同施药剂量的诱导高峰时间以及导增加的量也不相同。 敌敌畏对家蝇Musca domestica vicina GST活性没有明显的诱导作用,阿特拉津对家蝇GST活性也没有产生诱导作用。  相似文献
