以葡萄糖为底物,以经加热预处理并活化过的厌氧污泥为种泥,研究了初始pH值对产氢产乙酸/耗氢产乙酸两段耦合工艺厌氧发酵定向生产乙酸的影响。实验考察了7个初始pH值(5、6、7、8、9、10、11)条件下的底物降解、产物产生和发酵过程pH值的变化。结果表明:产氢产乙酸段初始pH值的变化不仅影响本阶段产酸,而且影响耗氢产乙酸段产酸。初始pH=5时主要进行乙醇型发酵;pH=6和7时主要进行丁酸型发酵;pH=8时混合酸型发酵类型逐渐占优势,pH=8~11时均以乙酸为主要产物,耦合系统生产乙酸最优初始pH值为10。在初始pH=8~11范围内,产氢产乙酸段初期的乙醇浓度一般较高,但到后期因乙醇被微生物进一步代谢转化成乙酸而使其含量下降。  相似文献

污泥厌氧发酵生产挥发性脂肪酸相较产甲烷,是更具应用价值的污泥稳定途径及资源化利用方式,得到国内外学者的普遍重视。考虑到产酸量低和产酸过程的不稳定性是限制污泥发酵产酸的主要问题,采用生物强化方法实现挥发性脂肪酸的大量积累,与物理和化学方法相比,具有成本低、无二次污染等优点。根据生物强化制剂的类型,归纳了微生物纯培养物、微生物混合培养物及生物酶强化对污泥厌氧发酵产酸的影响,并在此基础上对生物强化技术控制污泥定向产酸、调控奇偶数碳比率等方面的应用进行讨论。此外,分析了影响挥发性脂肪酸产量和组分的因素,如pH、温度、底物、水力停留时间和污泥龄等。最后对生物强化技术的发展方向进行了展望,以期为深入探究污泥资源化利用提供借鉴。  相似文献

[目的]研究污泥厌氧消化产挥发性脂肪酸(VFA)过程中的有机物碳流的转化机制,阐明乙酸累积机理。[方法]研究溴乙烷磺酸盐(BES)和氯仿(CHCl3)抑制模型下中间代谢产物和气体的累积,检测各产乙酸功能菌群数量,推断污泥产酸发酵过程中的有机物碳流方向和乙酸累积机理。[结果]BES模型乙酸浓度达27 mmol/L,fhs基因拷贝数比对照组高2-3倍,产氢产乙酸菌略有下降。CHCl3模型乙酸浓度达22 mmol/L,fhs基因拷贝数比BES组低一个数量级,产氢产乙酸菌下降明显。[结论]BES特异性较高,除产甲烷菌外对其他厌氧产酸细菌没有影响,乙酸浓度增加并且其主要来源于水解发酵产酸以及同型产乙酸过程。氯仿除抑制产甲烷菌外,对同型乙酸菌和产氢产乙酸菌也有强烈的抑制作用。  相似文献

为了提高有机酸的浓度,探讨污泥产酸工业化的可行性,考察了三种产气抑制剂对污泥和食 品废弃物混合发酵的VFA浓度及其组成、底物转化率、产气量及产甲烷势的影响.结果表明 ,采用BES、氯仿和碱(pH10)三种手段可以有效抑制发酵后期产气作用,提高底物产酸转化率,并且对有机酸组成也有一定影响.  相似文献

有机酸是重要的化工原料,从市政污泥厌氧发酵过程提取有机酸、用于生产高附加值的产品可以实现废物资源化,本研究在确定最佳有机溶剂和萃取剂的基础上,考察了有机溶剂对城市污泥厌氧发酵生产有机酸的影响.结果表明,较合适的溶剂和萃取剂分别为磺化煤油和三烷基氧膦.少量磺化煤油对城市污泥发酵产生产有机酸有一定促进作用.  相似文献

不同pH缓冲液对由乙酸产甲烷菌群结构的影响   总被引:1,自引:0,他引:1  
【目的】研究不同p H缓冲液对乙酸产甲烷过程及对细菌和古菌群落结构的影响。【方法】分别添加磷酸盐(PB)、4-羟乙基哌嗪乙磺酸(HEPES)、哌嗪-1,4-二乙磺酸(PIPES)和Na HCO3/CO2缓冲液到乙酸产甲烷菌系中,定期监测甲烷产生趋势,到稳定期后收集菌体,进行16S rRNA基因的末端限制性片段多态性分析(T-RFLP)。【结果】发现PB组的乙酸产甲烷菌系延滞期约为40d,显著高于其他组的20-24 d(P0.05);Na HCO3/CO2组乙酸转化为甲烷的比例为(88.3±0.5)%,显著高于其他组的77%-81%(P0.05);不同缓冲液组的最大甲烷比生长速率为0.46-0.57 d-1(P0.05);Na HCO3/CO2组的细菌群落变化最明显,主要是未培养细菌(unclassified bacteria)、螺旋菌科细菌(Spirochaetaceae)和未培养WWE1类群的丰度较其他组分别增加到(15.5±9.4)%、(7.3±4.6)%和(17.6±6.3)%,而互养菌科(Synergistaceae)的细菌丰度降低到(8.9±8.1)%。AC+PB组中的古菌类群发生了明显变化,以竹节状甲烷鬃毛菌(Methanosaeta harundinacea)相关的产甲烷古菌占主导(97±2%),而在HEPES、PIPES和Na HCO3/CO2组和不加缓冲液组中同时存在两类乙酸营养型产甲烷古菌M.harundinacea和联合鬃毛甲烷菌(Methanosaeta concilii),以及属于甲烷杆菌目(Methanobacteriales)的氢营养型产甲烷古菌。【结论】在乙酸产甲烷菌系中加入PB增加了甲烷产生的延滞期,加入Na HCO3/CO2增加了甲烷产量,但是添加p H缓冲液不会影响到菌系的最大甲烷比生长速率。加入PB和Na HCO3/CO2都会显著改变微生物的菌群结构。这些研究为设计适宜的产甲烷菌系生长条件提供了参考。  相似文献

摘要:【目的】研究不同温度条件下的石油烃降解产甲烷菌系中是否存在乙酸互营氧化产甲烷代谢途径。【方法】以3个不同温度条件的正十六烷烃降解产甲烷菌系Y15(15℃)、M82(35℃)和SK(55℃)作为接种物,通过乙酸喂养实验、并添加乙酸营养型产甲烷古菌的选择性抑制剂NH4Cl和CH3F,结合末端限制性片段长度多态性(terminal restriction fragment length polymorphism,T-RFLP)和克隆文库技术,分析乙酸产甲烷潜力及产甲烷古菌群落的演替趋势,推测产甲烷代谢途径的变化趋势。【结果】无论是否添加NH4Cl和CH3 F,这3个菌系都可以利用乙酸生长并产生甲烷,但是添加NH4Cl和CH3 F后产甲烷延滞期增加,最大比甲烷增长速率降低;只添加乙酸后,3个不同温度的菌系的古菌群落主要由乙酸营养型产甲烷古菌甲烷鬃毛菌属(Methanosaeta)组成,其丰度分别为92.8±1.4%、97.3±2.4%和82.8±9.0%;当添加选择性抑制剂NH4Cl,3 个菌系中的Methanosaeta的丰度分别变为98.5±0.7%、87.4±4.8%和6.1±8.6%,中温菌系M82中氢营养型产甲烷古菌甲烷袋装菌属(Methanoculleus)的相对丰度增加到12. 6±4.0%,高温菌系SK中另一类氢营养型产甲烷古菌甲烷热杆菌属(Methanothermobacter)增至84.3±1.5%;当添加选择性抑制剂CH3 F,Methanosaeta丰度分别降至77.1 ± 14.5%,86.4±6.1%和35.8±7.8%,低温菌系Y15中的甲烷八叠球菌属(Methanosarcina)增高(15.7±21%),这类产甲烷古菌具有多种产甲烷代谢途径,M82中Methanoculleus丰度上升到13.6±13.1%,SK中Methanothermobacter丰度增大到48.5±11.2%。【结论】在低温条件下,菌系Y15可能主要通过乙酸裂解完成产甲烷代谢,在中高温条件下,菌系M82和SK中可能存在乙酸互营氧化产甲烷代谢途径,并且甲烷的产生分别通过不同种群的氢营养型产甲烷古菌来完成。  相似文献

添加厨余垃圾对剩余污泥厌氧消化产沼气过程的影响   总被引:4,自引:0,他引:4  
为提高剩余污泥厌氧消化的沼气产量和甲烷含量,研究了厨余垃圾的不同添加量对剩余污泥厌氧消化性能的影响。结果表明,在35℃下,随着剩余污泥中厨余垃圾添加量的增加,厌氧消化系统中碳氮质量比(C/N)、胞外多聚物(EPS)等生理生化指标均有不同程度的改善。其中当剩余污泥与厨余垃圾质量比为2:1时,混合有机废弃物中沼气产量和甲烷含量均达到最大值,每克挥发性固体(VS)产生了156.56mL沼气,甲烷体积分数为67.52%,分别比剩余污泥单独厌氧消化时的产气量提高了5倍和1.5倍。  相似文献

考察了不同的预处理方式对餐厨垃圾与市政污泥混合厌氧消化产甲烷的影响。结果表明:对市政污泥进行热碱预处理效果最优,与空白组相比,热碱处理组累计单位质量的挥发性固体(VS)的甲烷产量达到了255.00 L,提高了13.03%,溶解性化学需氧量(SCOD)的降解率提高了27.05%,总固体(TS)的降解率提高了16.65%,VS的降解率提高了9.08%。在厌氧消化过程中,利用碳水化合物及蛋白质的热袍菌门(Thermotogae)的比例在所有处理中都有所上升;与产生短链脂肪酸有关的Erysipelotrichaceae菌属在预处理组中的丰度显著高于空白组,热碱处理组中Erysipelotrichaceae在末期的丰度最高,为19.21%;Petrimonas在厌氧消化体系中的作用为产氢产乙酸菌,热碱处理组的Petrimonas的丰度在产气高峰时最高,为6.84%。不同处理系统起主要产甲烷作用的古菌属为Methanosarcina,在产气高峰期热碱预处理组中Methanosarcina的丰度最高。预处理能够破环市政污泥的细胞结构,提高污泥的可生物利用性,提高混合厌氧消化产甲烷的效率。  相似文献

多级逆流工艺促进城市污泥厌氧发酵生产挥发性脂肪酸   总被引:2,自引:0,他引:2  
采用一种新型的厌氧发酵工艺——多级逆流发酵工艺对城市污泥进行厌氧发酵, 实现高效产挥发性脂肪酸的目的。结果表明, 实验条件下应用多级逆流发酵工艺, 挥发性脂肪酸浓度与产率分别达到(10.5±0.5) g/L和0.20 gVFAs/ gVS, 与普通厌氧发酵工艺相比, 分别提高了31%和54%。此外, 在多级逆流工艺中, 底物有机质去除率可达50%, 较普通厌氧发酵提高了37%。进一步分析多级逆流工艺产酸的机制, 发现产酸效率的提高在于降低了发酵产物对厌氧产酸细菌的抑制效应, 并且工艺的VFAs产率以及有机质去除率分别取决于第一级和第三级厌氧发酵过程。因此, 城市污泥采用多级逆流工艺厌氧发酵不仅能够有效促进挥发性脂肪酸的生成, 而且能够较大程度上提高污泥中有机质的去除率。  相似文献

Using volatile fatty acids (VFA) from the anaerobic digestion liquor of sewage sludge, up to 9.2 mm 5-aminolevulinic acid (ALA) could be produced by Rhodobacter sphaeroides under anaerobic-light (5 kLux) conditions with repeated addition of levulinic acid (LA) and glycine and using a large inoculum (approx. 2 g/l of cells, initially from glutamate/malate medium). As the VFA medium also contained organic nitrogen sources such as glutamic acid, the cells were later grown up to about 2 g/l in the VFA medium instead of the glutamate/malate medium. ALA production was then again promoted by adding LA and glycine. Using this improved method, up to 9.3 mm ALA was produced by feeding propionate and acetate together with LA and glycine, indicating that VFA medium formed from sewage sludge could be useful for ALA production.  相似文献

Volatile fatty acids (VFAs) are the most suitable and biodegradable carbon substrates for many bioprocesses. This study explored a new approach to improve the VFAs production from anaerobic co-digesting waste activated sludge (WAS) with corn straw (CS). The effect of feedstock proportion on the acidification efficiency was investigated. The maximum VFAs yield (corresponding fermentation time) was substantially increased 69% (96 h), 45% (72 h), 13% (120 h) and 12% (120 h) with 50%, 35%, 25% and 20% CS proportion of feedstock, respectively. HAc (acetic acid) was consistently the most abundant, followed by HPr (propionic acid) and n-HBu (butyric acid) in the co-digesting tests. The increase of CS in feedstock led to more production of HAc and HPr. Moreover, the consumption of protein and carbohydrate were also improved remarkably from 2955 and 249 mg COD/L (individual WAS fermentation) to 6575 and 815 mg COD/L (50%WAS:50%CS co-digestion) from 120 onward, respectively. The highest contribution of CS to additional VFAs production was1113 mg VFAs (as COD)/g CS/L in the 65%WAS:35%CS co-digesting test. Our study indicated a valuable method to improve VFAs production from anaerobic co-digesting WAS and CS.  相似文献

Despite their major contribution to the energy supply of ruminants, the production of volatile fatty acids (VFA) in the rumen is still poorly predicted by rumen models. We have developed an empirical approach, based on the interpretation of large bibliographic databases gathering published in vivo measurements of ruminal VFA production rate (PR), rates of duodenal and faecal digestion and molar percentages of VFA in the rumen. These databases, covering a wide range of intake levels and dietary composition, were studied by meta-analysis using within-experiment models. We established models to quantify response laws of total VFA-PR and individual VFA molar percentages in the rumen to variations in intake level and dietary composition. The rumen fermentable organic matter (RfOM) intake, estimated from detailed knowledge of the chemical composition of diets according to INRA Feed Tables, appears as an accurate explanatory variable of measured total VFA-PR, with an average increment of 8.03 ± 0.64 mol total VFA/kg RfOM intake. Similar results were obtained when total VFA-PR was estimated from measured apparent RfOM (total VFA-PR/RfOM averaging 8.3 ± 1.2 mol/kg). The VFA molar percentages were related to dry matter intake and measured digestible organic matter (OM), digestible NDF and rumen starch digestibility, with root mean square error of 1.23, 1.45, 0.88 and 0.41 mol/100 mol total VFA for acetate, propionate, butyrate and minor VFA, respectively, with no effect of pH on the residuals. Stoichiometry coefficients were calculated from the slopes of the relationships between individual VFA production (estimated from measured apparent RfOM and individual VFA molar percentages) and measured fermented fractions. Coefficients averaged, respectively, 66, 17, 14 and 3 mol/100 mol for NDF; 41, 44, 12 and 4 mol/100 mol for starch; and 46, 35, 13 and 6 mol/100 mol for crude protein. Their use to predict VFA molar percentages appear relevant for most dietary conditions, that is, when the digested NDF/digested OM ratio exceeded 0.12. This study provides a quantitative review on VFA yield in the rumen. It contributes to the development of feed evaluation systems based on nutrient fluxes.  相似文献

Nested PCR is a highly sensitive procedure for monitoring species of arbuscular mycorrhizal (AM) fungi and for determining their abundance in planta and in soil. DNA sequence variability in the D1 and D2 domains of the large ribosomal subunit is sufficient to design primers which discriminate between AM fungi at the species level. The usefulness of this molecular approach is illustrated in the present study on the differential impact of sewage sludges on a community of three AM fungi (Glomus mosseae, Glomus intraradices, Gigaspora rosea). Nested PCR was applied to trypan blue-stained mycorrhizal root fragments and soil mycelium from pot cultures of Medicago truncatula inoculated with the three fungi separately or together, and grown in sand containing sewage sludge that had been enriched or not with metallic or organic pollutants. G. intraradices and Gig. rosea varied in behaviour depending on whether they were inoculated alone or as a mixed community. G. mosseae showed a similar sensitivity towards each sewage sludge whether in community or alone, making it a potential candidate for ecotoxicological tests using M. truncatula to evaluate the quality or potential toxicity of sewage sludges which are widely used as fertilizers in agricultural lands.
