As part of our study of the Acanthaceae of Costa Rica and from a phylogenetic study of Ruellia as a whole, we place R. barbillana and R. metallica in synonymy with R. terminalis. We discuss species limits of R. terminalis and provide a key that distinguishes it from close relatives including R. grantii, R. oaxacana, and R. phyllocalyx. Ruellia puri is put into synonymy with R. jussieuoides, and R. standleyi is synonymized with R. ochroleuca. Disjunct distributions characterize R. terminalis, R. jussieuoides, and R. ochroleuca, although this pattern is most extreme in R. ochroleuca. Phylogenetic analysis of DNA sequence data lends support to our synonymy decisions. New lectotypifications are provided for Arrhostoxylum achimeniflorum, Dipteracanthus puri, Dipteracanthus puri β angustifolius, Dipteracanthus puri γ gymnocladus, Lychniothyrsus ochroleucus, and Otacanthus pearcei .  相似文献

We describe four new species of Ruellia from Costa Rica; three from the Osa Peninsula, Puntarenas province (R. exilis, R. mira, R. odorata), and one from the southwestern corner of San José province (R. norvegigratiosa). Ruellia matagalpae is reported as a new record for the country. New combinations are made in Ruellia for the two Costa Rican species of Blechum (R. blechum and R. costaricensis). Three species are reduced to synonymy, and another species is considered doubtfully worthy of recognition. Notable range extensions are reported for two species. Corollas and fruits of all 22 Costa Rican species of Ruellia are shown and compared as easily used identification aids. An informal guide to species groups and a dichotomous key to the 22 species are presented.  相似文献

Background: Hybridisation between locally common and rare plant species is of conservation concern as it may lead to the extinction of the rare taxon. Rumex aquaticus (2n = ca. 200, Polygonaceae) is a widespread Euro-Siberian species but rare in the British Isles. It hybridises freely with the locally abundant congener R. obtusifolius (2n = 40) producing fertile and viable hybrids.

Aims: We aimed to establish whether hybridisation poses an extinction threat to R. aquaticus in Britain.

Methods: Flow cytometry, AFLPs and morphological characters were used to assess the extent of hybridisation between R. obtusifolius and R. aquaticus.

Results: Flow cytometry was the most useful tool to distinguish between hybrids and parental species and indicated that out of 88 samples from two hybrid populations, 85% were likely to be F1s and only 15% were likely to be back-crosses to R. aquaticus. AFLP markers were only able to identify putative F1 hybrids reliably and 11 morphological traits could not distinguish unambiguously between hybrids and pure R. aquaticus.

Conclusions: Although our data suggest that genetic assimilation of pure R. aquaticus populations is unlikely to be the case in the future, there is a potential threat from demographic swamping by hybrids and targeted protection of R. aquaticus habitat is proposed as a management strategy.  相似文献

【背景】桉树(Eucalyptus)青枯病危害严重,丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)与桉树共生影响桉树对青枯病的抗性,而AMF响应桉树青枯菌侵染的机制仍不清楚。【目的】探索AMF响应桉树茄科雷尔氏菌(Ralstonia solanacearum)的侵染机制。【方法】以非菌根化和异形根孢囊霉(Rhizophagus irregularis)菌根化巨桉(Eucalyptus grandis)分别受茄科雷尔氏菌侵染0、24、48和96 h接种后(hour post-inoculated, hpi)的根系组织为研究对象,基于转录组测序筛选和鉴定菌根化巨桉根系中异形根孢囊霉响应茄科雷尔氏菌侵染的基因信息。【结果】与对应非菌根化桉树受茄科雷尔氏菌侵染的时间点相比,菌根化桉树中异形根孢囊霉响应青枯菌侵染显著差异表达基因为3 382–5 989个,随青枯侵染时间进程的增加,异形根孢囊霉特异性响应茄科雷尔氏菌侵染差异表达基因数量逐渐增多。茄科雷尔氏菌侵染24 hpi时,异形根孢囊霉显著富集共生体生长、孢子形成和凋亡信号通路、铁载体等相关基因;茄科雷尔氏菌侵...  相似文献

【目的】铜绿假单胞菌(Pseudomonas aerugionsa)二鸟苷酸环化酶SiaD调控着铜绿假单胞菌的生物被膜形成等表型。在研究过表达siaD对生物被膜的调控作用时发现,与野生型siaD基因回补菌株相比,一株回补菌株的生物被膜产量显著升高。本文的目的即是探究该菌株生物被膜产量升高的原因,并对该菌株的其他表型进行研究。【方法】通过测序确定突变位点;利用生物被膜定性和定量实验对发生点突变的菌株表型进行分析;利用Western blotting实验检测SiaDR119M蛋白表达水平;利用GST-pulldown实验检测SiaC蛋白与SiaDR119M蛋白在体外的结合能力;针对siaDR119M点突变基因进行融合蛋白表达载体的构建,表达并纯化该蛋白,利用高效液相色谱检测SiaDR119M的酶活;为了进一步研究c-di-GMP与细菌运动能力的关系,对细菌的运动能力进行检测。【结果】测序比对结果显示,序列的第119个氨基酸发生了突变,由精氨酸突变成了甲硫氨酸。生物被膜定性和定量实验显示,与野生型siaD...  相似文献

【背景】鸭疫里默氏杆菌广泛存在于养殖场,引起雏鸭发生传染性浆膜炎,严重危害养鸭业的发展。【目的】提高鸭疫里默氏杆菌发酵培养水平和抗原活性,为鸭疫里默氏杆菌灭活疫苗的研制提供技术指引。【方法】利用单因素及响应面的试验设计方法,针对鸭疫里默氏杆菌进行疫苗培养基的研制,并探究不同发酵时间点该菌的抗原活性,选择抗原活性最高点时制备灭活疫苗,通过动物免疫保护试验评价疫苗免疫效果。【结果】使用研制的疫苗培养基发酵培养鸭疫里默氏杆菌,其活菌数能够达到4.68×1010 CFU/mL,较市面上该菌专用的培养基提高2.29倍。该菌发酵12 h后抗原活性达到最高,在此时制备的灭活疫苗诱导小鼠产生的抗体水平显著高于商品化灭活疫苗,攻毒保护率达到了100%。【结论】本研究研制的培养基具有优异的增菌效果,可作为生产鸭疫里默氏杆菌灭活疫苗抗原的发酵培养基,疫苗生产过程中可选择菌株抗原活性达到最高时收集菌体。  相似文献

Introduction. During a recent explorative field trip to the Western Ghats a new species of Riccia was discovered with possible links to the northern Australian species, Riccia caroliniana Na-Thalang.

Key Results. Morphological similarities between the new species, R. sahyadrica and R. caroliniana include the ventral positioning of the photosynthetic tissue in the ventral section of the thallus in specialised structures known as ventral lamellae. These form a v-shaped patterning along the ventral surface of the thallus and are unique to these two species. Both species also bear similar spores with differences in the micro-ornamentation. Differences between the two species also extend to sexual condition, whereby R. caroliniana is dioicous and R. sahyadrica is monoicous.

Conclusions. A new species, Riccia sahyadrica Manju & Cargill is described and illustrated and compared to the only other morphologically similar species, R. caroliniana endemic to northern Australia. Both species possess the unique characteristic of bearing ventral photosynthetic tissue in contrast to the dorsal position typically seen in all other Riccia species.  相似文献

【目的】研究红球菌R04细胞的分裂方式及联苯对其形态和细胞分裂的影响。【方法】以一株多氯联苯降解菌株(Rhodococcus sp.R04)为研究对象,利用荧光显微镜、扫描电子显微镜及透射电子显微镜分析红球菌R04在不同培养条件下的细胞分裂。【结果】红球菌R04细胞表现出对称分裂(约占30%)和不对称分裂(约占70%)两种分裂方式,且培养条件不影响不对称分裂细胞所占的比例。细胞分裂过程中,隔膜主要分布于细胞长度的30%–50%。在联苯的分解代谢过程中,红球菌R04细胞的生长分裂会受到联苯的抑制,但不影响红球菌R04细胞的分裂方式,在联苯胁迫下,细胞形成丝状化,表现出异常分裂,随着培养时间的延长,在细胞生长指数后期至转换期,细胞能够进行正常分裂。【结论】环境异生型化合物联苯/多氯联苯对其降解菌株——红球菌R04细胞的生长和分裂有较强影响,但是并不影响其分裂方式。  相似文献

Aim

The secretion of allelochemicals from plant roots plays a key role in soil sickness and soil-borne disease. The goal of this study was to investigate the role of allelopathic chemicals in Ralstonia solanacearum-infected tobacco roots.

Methods

The organic acids investigated in the present study are major components of tobacco root exudates. Through a swarming assay, we assessed the chemotaxis and colonization of R. solanacearum in response to organic acids.

Results

Fumaric acid was detected, and the results showed that this acid could serve as a semiochemical for attracting R. solanacearum and inducing the formation of biofilms of this species. The results also revealed that cinnamic and myristic acids play significant roles on swarming motility and chemotaxis. In addition, cinnamic, myristic and fumaric acids could enhance the expression of chemotaxis- and motility-related genes in R. solanacearum cultured in minimal medium. Furthermore, these three acids promote R. solanacearum colonization and accelerate disease progression in tobacco.

Conclusion

Cinnamic, myristic and fumaric acids could serve as semiochemical attractants to induce the colonization and infection of R. solanacearum. The results of the present study enhance our understanding of the ecological effects of plant root exudates in plant-microbe interactions and help to reveal the relationship between tobacco bacterial wilt and the autotoxins and allelochemicals that accumulate from root exudates.

The enzymatic hydrolysis of (R, S)-5-acyloxymethyl-3-alkyl-oxazolidin-2-one I and the behavior of (S)-I for extraction with an organic solvent were examined so as to extend the biological resolution to racemates, and to learn about more appropriate combinations of substrates with lipases on the asymmetric hydrolysis. The combination of (R, S)-5-hexanoyloxymethyl-3-tert-butyl-oxazolidin-2-one 4 with lipoprotein lipase Amano 3 (L. P. L. Amano 3, origin; Pseudomonas aeruginosa) and that of (R, S)-5-octanoyloxymethyl-3-isopropyl-oxazolidin-2-one 14 with L. P. L. Amano 3 efficiently gave (S)-5-hydroxymethyl-3-tert-butyl-oxazolidin-2-one (S)-lla (99% e.e.) and (S)-5-hydroxymethyl-3-isopropyl-oxazolidin-2-one (S)-IIb (99% e.e.),respectively. New combinations are proposed in anticipation of the Polygonaceae treatment in the forthcoming volume of Intermountain Flora: Polygonum kelloggii var. esotericum, P. kelloggii var. watsonii , Rumex densiflorus var. pycnanthus , R. salicifolius var. utahensis, and R. occidentalis var. tomentellus. Thirty different Streptomyces strains were isolated from soil samples collected from different places in Egypt. The strains were isolated using Oatmeal agar and ISP medium 2 agar at pH 7.5. The isolate Streptomyces sp. Eg23 was selected for further working up to yield three new streoisomers of anthracycline metabolites. The structures were elucidated as (7R, 9R, 10S)-e-Rhodomycinone (1), (7R, 9R, 10S) Aklavinone (2), and (9R, 10S) 7-Desoxy-z-Rhodomycinone (3) by interpretation of their 1D and 2D NMR spectra. The absolute stereochemistry of 1 was confirmed by modified Mosher’s method. The (7R, 9R, 10S)-e-Rhodomycinone (1) showed activity against human lung cancer cell H460, murine Lewis lung cancer cell LL/2 and human breast cancer cell MCF-7. Compounds 13 were known synthetically, but until now have not been isolated as natural products from a microorganism. The results showed that the Streptomyces sp. Eg23 seems to be an interesting target for studying the regio-and stereochemistry of the cyclization step catalysed by polyketide cyclases to produce new anthracyclines through combinatorial biosynthesis.
