The objective of this study was to determine if there is an association between facial hair whorl patterns on the bovine forehead and sperm morphology. Breeding soundness exams were conducted on 219 yearling bulls at three Colorado State University facilities. There were 150 Angus bulls and the remaining bulls were of several different breeds. Hair whorl patterns on the forehead were classified as round or nonround epicenters. Angus bulls with round epicenters had a higher percentage of morphologically normal spermatozoa compared to nonround epicenters (P < 0.05). Hair whorls on Angus bulls were sorted into two extreme groups of round spirals, with rotation or long lines that were longer than the width of the eyes. Bulls with round spirals had a greater percentage of normal spermatozoa compared to bulls with long lines (P < 0.05). Hair whorl pattern on Angus bulls also had an effect on the percentage of bulls that had the minimum threshold value of 70% normal spermatozoa. Eighty percent of Angus bulls with round epicenters had > 70% or more normal spermatozoa compared to 59% with a nonround epicenter (P < 0.01). Fifty percent of Angus bulls with long lines failed because they had < 70% normal spermatozoa. There were no significant differences in the remaining non-Angus bulls. Hair whorl patterns may be useful for making early culling decisions.  相似文献

Yearling, grass-fed, beef bulls at the USDA Subtropical Agricultural Research Station, Brooksville, Florida, were assessed for physical and semen traits in January, April, July and October of 1991 (Trial 1) and 1992 (Trial 2). Bulls were given a breeding soundness evaluation (BSE) using revised semen and scrotal circumference (SC) criteria. In Trial 1, the bulls consisted of Angus (n = 15), Brahman (n = 14), Hereford (n = 15) and Senepol (n = 14). In Trial 2, the breeds were Angus (n = 15), Brahman (n = 16), Romosinuano (n = 13) and Nellore x Brahman (n = 9). Trial bulls generally showed delayed growth compared with grain-fed bulls in temperate environments. Breed influenced semen traits (percentage sperm motility, normal spermatozoa and those with primary abnormalities) in both trials. Temperate Bos taurus breeds (Angus, Hereford) were generally superior to Bos indicus breeds (Brahman, Nellore x Brahman). Tropically-adapted Bos taurus breeds (Senepol, Romosinuano) were intermediate for those traits tested. In general, tropically-adapted Bos taurus breeds were more similar in reproductive development to temperate Bos taurus than to Bos indicus breeds. Breed by test period interactions occurred and were mainly influenced by delayed sexual maturity of Bos indicus bulls. Qualitative semen traits increased with bull age, particularly from 12 to 18 mo. Scrotal circumference development was slower in the Bos indicus breeds. Bulls of satisfactory BSE status at 18.1 to 22 mo of age were 73.9% in Trial 1 and 58.5% in Trial 2. Brahman bulls had the least satisfactory BSE scores in both years (Trial 1, 44.4%; Trial 2, 22.2%). Most bulls failed to achieve satisfactory BSE status due to a small SC relative to age (Trial 1, 66%; Trial 2, 72%). The most efficacious use of the BSE was > or = 15 mo in Bos taurus bulls and > 18 mo for Bos indicus bulls. Although the BSE has proven to be useful for the assessment of young, pasture-raised bulls in semi-tropical environments, use of SC thresholds linked more with growth traits than with calendar age would improve comparisons of relative reproductive development in such bulls, particularly those of Bos indicus derivation.  相似文献

In this comparative study, reproductive parameters and semen characteristics of cloned bulls (n = 3) derived from somatic cell nuclear transfer (SCNT) were compared to their original cell donor Holstein-Friesian (n = 2) bulls from the same enterprise to assess the differences in reproductive potential between a donor bull and its clones. The parameters evaluated included motility of fresh, frozen-thawed and Percoll-treated frozen-thawed spermatozoa, as well as in vitro fertilization (IVF) ability, embryo quality, birth and survival of calves following IVF and embryo transfer with frozen-thawed semen. With fresh semen, spermatozoa from one cloned bull had lower motility than its donor. Cloned bulls had higher velocity parameters in fresh semen, but those effects were not obvious in frozen-thawed or frozen-thawed semen selected with a Percoll gradient. Semen collected from cloned bulls had significantly higher IVF rates compared to donors; however, embryo development per cleaved embryo or quality of blastocysts did not differ between donors and cloned bulls. Pregnancy and live offspring rates from one donor and its cloned bull did not differ between fresh (40%, 16/40 versus 46%, 17/37) and vitrified/thawed (13%, 2/16 versus 25%, 4/16) embryo transfer following IVF. A total of 26 calves were obtained from genotypically identical donor and cloned bulls with no signs of phenotypical abnormalities. These preliminary results suggested that the physiology of surviving postpubertal cloned bulls and quality of collected semen had equivalent reproductive potential to their original cell donor, with no evidence of any deleterious effects in their progeny.  相似文献

A total of 17 bulls was used to study the effects of boldenone undecylenate on growth and semen characteristics in beef bulls. In trial 1 nine mature mixed-breed beef bulls with satisfactory semen quality were divided into two groups. Group I (n = 5) received boldenone undecylenate (1.1 mg/kg) at 21 d intervals for a total of seven treatments (147 d). Group II (n = 4) served as untreated controls. Semen was collected from each group by electroejaculation on each treatment day and evaluated according to the standards of the Society for Theriogenology. Although neither the percentage of spermatozoa with primary or secondary morphological abnormalities was different, the ejaculates of Group I bulls contained a higher percentage of abnormal spermatozoa than those in Group II. In trial 2, eight mixed-breed bull calves, average weight 140.4 kg, were maintained under drylot conditions in a single paddock. The bulls were divided into two equal groups. Group I (n = 4) received boldenone undecylenate as in Trial 1. Group II (n = 4) served as untreated controls. The bulls were weighed and the scrotal circumference (SC) was measured every 21 d until it reached 30 cm, at which time semen was collected and evaluated as in Trial 1. Group I bulls had a higher percentage of spermatozoa with primary morphological abnormalities than bulls in Group II. Group I bulls had a higher average daily gain (ADG) than Group II bulls and required 21 d longer for the SC to reach 30 cm. Semen quality for all bulls was satisfactory at each sampling day.  相似文献

Young (9 to 10 mo) Aberdeen-Angus bulls (n = 5) in excellent body condition and pastured with approximately 35 other bulls of similar age on a farm in southeastern Alabama had fever, harsh pulmonary sounds, increased respiratory rate and variable amounts of hindlimb and scrotal edema. Bulls had mild microcytic, normochromic anemia. Numerous eperythrozoon organisms were identified on blood smears. Indirect hemagglutination inhibition test results for Eperythrozoon suis antibodies were negative either because E. suis antigens do not cross react with cattle eperythrozoon organism antibodies or blood was collected before there was sufficient time for seroconversion. Bulls had swelling of the scrotal wall, soft testes, and poor semen quality, characterized by low progressive motility and a high percentage of spermatozoa with primary and secondary abnormalities. Some of these abnormalities may be attributed to the age of the bulls. However, loss of scrotal thermoregulation was a major cause of testicular degeneration leading to poor semen quality. Other bulls in the herd had fever, increased respiratory rate, and swollen hindlimbs or scrotum. Subsequent to administration of oxytetracycline, parasitemia resolved rapidly, and clinical signs gradually abated. Four of five bulls successfully passed breeding soundness examinations six months after initial clinical illness. The remaining bull failed twice due to the presence of testicular inflammation.  相似文献

Detailed semen evaluations were carried out on approximately 363 Santa Gertrudis, 5/8 Brahman and Brahman bulls on 12 different properties across northern Australia, as part of systematic breeding soundness examinations. A subset of bulls (n=245) were subsequently mated in groups, to cows and heifers at bull:female ratios of 2.5-6.0%, with the paternity of resulting calves being determined by microsatellite DNA testing. Motility traits of semen and spermatozoa were moderately repeatable and correlated with each other, but were unrelated to calf output. The percentage of morphologically normal spermatozoa in ejaculates was moderately to highly repeatable (e.g. r=0.10-0.64). The most common morphological abnormalities seen were mid-piece abnormalities, in particular, distal mid-piece reflex associated with a cytoplasmic droplet. Semen quality, particularly percent normal spermatozoa, was consistently related to calf output. In general, bulls with <50% normal spermatozoa sired few calves while bulls with the highest calf outputs had >70% normal spermatozoa. The presence or absence of heparin binding proteins in semen did not influence calf output. Semen from 93% of tested bulls was positive for heparin binding proteins. These results confirm that examination of semen, in particular, evaluation of percent morphologically normal spermatozoa, should be included in the breeding soundness examination of bulls.  相似文献

The objective of this study was to test the efficacy of an inexpensive, reusable internal artificial vagina (IAV) developed for breeding soundness evaluation of range beef bulls. In addition, sexual behavior during semen collection by IAV was compared to behavior during pasture breeding. Breeding soundness exams (BSEs) were conducted on 165 bulls in two consecutive years (96 in Year 1 and 69 in Year 2). In Year 1, BSEs were done twice in all bulls, once by a conventional protocol using electroejaculation (EEJ), followed by the IAV method, one week later. In Year 2, all BSEs were done on one day; 69 bulls by the IAV method followed by EEJ in 21 bulls that failed to serve the IAV. For semen collection using an IAV, mount cows were restrained in breeding crates and an IAV was inserted into the vagina just beyond the depth of the vestibular sphincter. After each copulation, the IAV was replaced for the next bull to be tested. Semen collection by IAV was successful for all bulls that mounted and penetrated cows during the testing period (54.3 and 69.6% of the bulls served the cows with IAVs in Year 1 and 2, respectively). Semen was collected successfully by EEJ from all bulls in both years. Differences were observed between semen collection methods in semen volume and percentage of sperm staining alive; however, the differences were opposite in Year 1 and 2 and, therefore, were probably due to natural variations in time and within bull rather than the method of semen collection. Semen collection by IAV allowed the detection of problems that prevented copulation in 8 bulls (4.8%) that were determined to be satisfactory potential breeders when semen was collected by EEJ. In Year 1, breeding observations were made at pasture for 15 bulls that served, and 15 bulls that did not serve cows with an IAV. Bulls that did not serve the IAV during the test period had fewer mounts, attempts to mount, and completed services at pasture than bulls that had served the IAV, indicating that such bulls are likely to be less efficient breeders. In conclusion, IAVs might be used as an alternative to EEJ for breeding soundness evaluation in about 50-70% of range bulls. Semen collection by IAV also allows observation of sex drive and mating ability thus improves the ability to select sound breeding bulls.  相似文献

Pubertal Angus bulls (n=10, 503 days of age and weighing 366 kg) and Senepol bulls (n=10, 457 days of age and weighing 381 kg) were stratified by age and weight into 2 dietary treatments formulated to provide equal amounts of crude protein and 75% (below) or 150% (above) of the maintenance requirements for metabolizable energy. Measurements to assess body growth and libido were collected at 28-day intervals for 112 days (June through September). Twice during each 28-day interval, the bulls were subjected to breeding soundness examinations. At the end of the experiment, gonadotropin releasing hormone (GnRH) - induced secretion of luteinizing hormone (LH) and testosterone (T) in the serum were determined. At the end of the experiment, bulls fed the above maintenance diet (P<0.0001) were 91 kg heavier, had 1.7 mm more backfat thickness and 12.6 cm(2) larger ribeye area than bulls on a below maintenance diet. Diet affected (P<0.003) the average daily change in scrotal circumference, but not the libido score (P>0.1) or semen quality. In general, Angus bulls had superior initial semen quality (P<0.06); however, during summer, semen quality tended to decrease in Angus but not in Senepol bulls. The final rectal temperature was 0.5 degrees C lower (P<0.003) in Senepol than in Angus bulls. Basal T concentrations and area under the GnRH-induced T curve were greater (P<0.07) for bulls fed the above rather than the below maintenance diet. Angus bulls had a higher (P<0.03) maximal LH response to GnRH and larger area under the GnRH-induced LH curve than Senepol bulls.  相似文献

The study was conducted to evaluate the effects of scrotal insulation on semen samples collected from bulls on embryonic development after IVF. Semen samples were obtained and cryopreserved from four Holstein bulls before and after a scrotal insulation period of 48 h (Day 0). Three types of samples were used for IVF: (1) semen from the test bulls collected 5 d prior to scrotal insulation (pre-insult); (2) semen from Day 13 (2-week post-insult; 2-week PI); and (3) semen from Day 20 (3-week PI). After 18 h of sperm-oocyte co-incubation, the zygotes were cultured for 8 d when a developmental score (0=degenerate, 1=2-cell embryo through 5=blastocyst) was assigned to each embryo. The post-thaw morphological evaluation of sperm samples revealed a decrease (P<0.01) in the percentages of normal spermatozoa in the 3-week PI samples in comparison with the pre-insult samples for Bulls I and III (74-22.3% and 67.7-0.5 %, respectively). The percentage of vacuolated spermatozoa increased significantly for Bull II. The cleavage and blastocyst formation rates and embryo development scores were affected (P<0.01) by the interaction of bull by sample collection time. For Bulls I and III (severe responders) the scrotal insulation effects persisted from the time of cleavage through blastocyst formation. In contrast, the cleavage and blastocyst formation rates for Bulls II and IV were unaffected, despite high percentages of vacuolated spermatozoa present in the post-insult samples for Bull II. In conclusion, the use of scrotal insulation to elevate scrotal temperature was an effective method to obtain semen samples with high percentages of abnormal spermatozoa. The decrease in embryonic development after IVF when using spermatozoa with morphological abnormalities seemed to be multifaceted and related to changes in head morphology.  相似文献

Ninety-three beef bulls and 2316 females were used to determine the relationships between breeding assessments of bulls and subsequent mating performance. Each bull was given a breeding soundness examination (BSE) and two 10-min libido/serving capacity (L/SC) tests. Breeding potential of each bull was classified as satisfactory (BSE score = 60 to 100) or questionable (BSE score = 30 to 59); libido was classified as either high (mean score = 9.0 to 10) or medium (mean score = 6.0 to 8.5). Bulls were then joined, single-sire, with groups of females which had received one of two treatments to synchronize estrus. Bull-to-female ratios ranged from 1:7 to 1:51. Continuous observations were conducted on the mating activity of each group. One bull was removed from the experiment due to a severe breeding problem. Bulls of satisfactory breeding potential (n = 80) did not differ (P > 0.10) from bulls of questionable breeding potential (n = 12) in measurements of mating activity. However, by the end of the synchronized breeding period, bulls classified as satisfactory breeders achieved approximately a 9% higher (P < 0.10) pregnancy rate than did bulls of questionable breeding status (45.6 +/- 2.1% vx 36.5 +/- 5.3%). Bulls with a high libido (n = 69) serviced more (P < 0.01) estrous females (81.3 +/- 3.1% vs 63.5 +/- 4.2%) than bulls with a medium libido (n = 23). However, pregnancy rates achieved by bulls of either libido classification did not differ significantly. Individual components of the BSE as well as mean libido score were poorly correlated with pregnancy rates (r = -0.22 to 0.18). It was concluded that classification of bulls by mean libido score can aid in identifying groups of bulls that service more estrus synchronized females, whereas classification by BSE score aids in identifying groups of bulls that impregnate more females.  相似文献

Two experiments were designed to determine the efficacy of intraglandular antibiotic treatment in beef bulls. Experiment 1 was designed to evaluate the glandular tissue reaction to intraglandular antibiotic treatment. Experiment 2 was conducted to determine the efficacy intraglandular injection of antibiotics for the treatment of naturally occurring cases of vesicular adenitis. Healthy beef bulls (n=15), 2 and 3 years of age, were randomly allocated to three equal treatment groups to receive 10% of the daily recommended parenteral dose of penicillin, ceftiofur, or oxytetracycline in a volume of 6 mL injected directly into one of the vesicular glands. Ultrasonography was performed before, immediately after, and at 24, 48 and 168 h after intraglandular injection. The size and hardness of vesicular glands injected with oxytetracycline was greater (P<0.01) than those injected with ceftiofur. Ultrasonographic pixel intensity increased (P<0.01) after treatment with antibiotics, especially after treatment with oxytetracycline or penicillin. In Experiment 2, yearling beef bulls with clinical vesicular adenitis (n=14) were referred to the Western College of Veterinary Medicine for treatment. Eight bulls had unilateral and six had bilateral vesicular adenitis. The most common isolate was Arcanobacterium pyogenes. Corynebacterium pseudotuberculosis was isolated from one bull. Bulls were subjected to rectal palpation and ultrasonography of the vesicular glands, semen collection by electroejaculation, and intraglandular treatment with ceftiofur (n=13) and if necessary, a second intraglandular treatment of penicillin (n=6). One bull was treated only with an initial intraglandular injection of penicillin. Bulls were evaluated once a week over 6 weeks by palpation of the glands, and evaluation of semen. All bulls recovered from vesicular adenitis after 3-6 weeks. There was a difference in the amount of pus (P=0.042), leukocytes (P<0.001) and blood (P=0.003) present in ejaculates from before treatment to 3 weeks after treatment. Pixel intensities in ultrasonographic images of healthy or affected vesicular glands, whether treated or untreated, did not change over time. Intraglandular injection of ceftiofur in yearling bulls via the ischiorectal fossa was effective for treating vesicular adenitis.  相似文献

In prepubertal bull calves there is an early transient rise in gonadotrophin secretion between 10 and 20 wk of age, and it has been suggested that this plays a role in the attainment of sexual maturation. To test this, we looked for differences in the gonadotrophin secretory pattern from birth to puberty between early and late maturing bulls. We also characterized the changes in semen morphology that occur about the time of puberty. Blood samples were collected (n=28) every wk from 2 to 20 wk of age and then every 2 wk until 50 wk of age. Semen was collected by electroejaculation at approximately 4-wk intervals from 36 to 49 wk of age. Puberty was defined as the first age at which an ejaculate contained 50 million spermatozoa with a minimum of 10 % motility Bulls were divided into early (n = 14) and late (n = 14) maturing groups based on the age at puberty (41.9 +/- 0.3 and 48.3 +/- 0.7 wk of age, respectively). There was a transient increase in serum concentrations of LH and FSH between 2 and 24 wk of age; LH concentrations were greater in early maturing bulls than in late maturing bulls at 12, 13, 15, 17 and 48 wk of age (P < 0.05). Serum concentrations of testosterone and FSH did not differ between groups (P > 0.05). As the bulls matured there was an increase in the percentage of normal and live sperm cells, cell motility and the number of cells per ejaculate (P < 0.05), and a decrease in the percentage of proximal droplets and knobbed acrosomes (P < 0.05). We concluded that, during the early rise in LH secretion, early maturing bulls had higher circulating LH concentrations than late maturing bulls. During the weeks preceding and following puberty there was an increase in the quality of semen collected by electroejaculation.  相似文献

In the Canadian Animal Genetic Resource Program, bull semen is donated in frozen or fresh (diluted) states. This study was designed to assess the cryopreservation of diluted bull semen shipped at 4°C overnight, and to determine the post-thaw quality of shipped semen using different straw volumes and freezing rates. Semen was collected from four breeding bulls (three ejaculates per bull). Semen was diluted in Tris-citric acid-egg yolk-glycerol (TEYG) extender, cooled to 4°C and frozen as per routine (control semen). After cooling to 4°C, a part of semen was removed and shipped overnight to the research laboratory via express courier (shipped semen). Semen was packaged in 0.25 or 0.5 ml straws and frozen in a programmable freezer using three freezing rates, i.e., -10, -25 or -40°C/min. Control semen was also shipped to the research laboratory. Post-thaw sperm motility characteristics were assessed using CASA, and post-thaw sperm plasma membrane, mitochondrial membrane potential and normal acrosomes were assessed using flow cytometry. Post-thaw sperm quality was greater in shipped semen as compared to control (P<0.001). The shipped semen packaged in 0.25 ml straws had better post-thaw sperm quality than in 0.5 ml straws (P<0.001). Freezing rate had no effect on post-thaw sperm quality. In conclusion, bull semen can be shipped overnight for subsequent cryopreservation and gene banking. Overnight shipping of semen was found advantageous for bull semen cryopreservation. Semen packaging in 0.25 ml straws yielded better post-thaw quality than 0.5 ml straws.  相似文献

Pen-O-Blocks were easily installed in 15 bulls. Libido appeared to be unaffected. However, with the exception of one bull, infections occurred, or the penis bypassed the block, or the pin broke, and in 14 tests the Pen-O-Blocks were removed within 5 weeks of installation. Recovery from infection following removal of the device was rapid and uneventful.Eight bulls had their penes deviated between 2 weeks and 4 months of age. Four of these also were castrated and implanted with androgens at 12 months of age. Semen quality and libido in the deviated bulls were normal.Bulls and steers were equipped with Chin Ball mating devices. Bulls generally were more effective than steers in detecting estrus. When placed in pens 24 hours per day with 20 heifers each, one bull detected 97% and another bull detected 100% of the potential estruses during a period of two months. An observer watching for .5 hours each morning and each late afternoon detected 74%.While steers were less effective, they were more docile than the bulls. Thus, with selection of active animals and adjustment of hormone therapy, implanted steers may be successful detectors of estrus with minimal danger to the owner. Reproductive development in the androgen-treated steers was delayed but maturation of the penis had occurred in steers slaughtered at 31 months of age. Body weight gains of steers tended to be accelerated during periods when they had implants.  相似文献

Crossbred (Bos taurus) yearling beef bulls were assessed for breeding soundness and physical traits prior to multi-sire natural mating at pasture. Bulls (n = 60) were assigned to six groups of nine or 10 bulls and two bull-groups were rotated on 14-day intervals during a 63-day mating season in each breeding herd (n = 3) of 191-196 cows. The remaining bulls (n = 14) were maintained under similar environmental conditions without mating exposure. Bulls were observed during mating and assessed for breeding soundness and changes following mating. Bulls used for breeding (UFB) lost 77 kg of body weight and declined from body condition scores of 6 to 4.5, whereas bulls not used for breeding (NUB) maintained body condition scores of 6 and gained 27 kg. The UFB bulls incurred a 75% total injury rate with 63% incidence of lameness and 12% incidence of reproductive injuries, resulting in a 22% attrition rate. Only 45% were physically sound at the end of mating. Scrotal circumference declined in UFB bulls (-4.58%) and increased in NUB bulls (2.49%). From the 98% BSE-satisfactory rate (UFB) prior to breeding, only 61% were BSE-satisfactory post-breeding. The NUB bulls declined from 57 to 36% satisfactory. The BSE classification was influenced by significant increases in abnormal spermatozoa (primary and secondary), which was significantly associated with injuries incurred during mating. Group and breed differences in injury rates and BSE-status following mating were evident. Environmental conditions and mating activity influenced bull seminal quality and physical condition. Pregnancy rates in all three breeding herds (91-96%) were similar, with insignificant differences between bull-groups; the effects of physical and reproductive changes on individual bull fertility were immeasurable.  相似文献

Sixteen yearling bulls were utilized to investigate administration of ergotamine tartrate on semen parameters, fertilization, and endocrinology. Bulls were allotted to a control diet of cracked corn, corn silage, and soybean meal (CON, n = 8) or a diet supplemented daily with 40 microg/kg body weight of ergotamine tartrate (ET, n = 8). Blood samples, average daily gain, scrotal circumference and rectal temperatures were collected every 14 day. Semen samples were obtained every 60 day and evaluated for motility and morphology. Scrotal temperatures were obtained by thermography immediately before electroejaculation. Semen from a subset of bulls from each treatment was also evaluated for in vitro fertilization. Administration of ET increased rectal temperature and resulted in lower scrotal temperatures compared to CON bulls (P < 0.05). However, prolactin, scrotal circumference, testosterone, and semen motility and morphology did not differ between groups throughout the experimental period (224 day). Cleavage rates of embryos derived from in vitro fertilization (IVF) with semen of bulls, fed with ET, were reduced compared to CON (P < 0.05); however, development of cleaved embryos to blastocyst did not differ between treatments. In conclusion, extended exposure of bulls to ET appeared to reduce fertilization potential of sperm.  相似文献

A total of 92 range beef bulls (Hereford = 60; Angus = 32) were given a breeding soundness examination (BSE) and two assessments for sex drive prior to their use in 23 breeding trials employing estrous synchronized females. Bulls were in three age groups: yearlings (n=29), two year olds (n=36), and three year olds and older (n=27). All yearling bulls were virgins, but the majority of the older bulls had previous mating experience. Angus bulls were superior (P<0.01) to Herefords in spermatozoal morphology and BSE score. Scrotal circumference increased with age beyond two years in Angus bulls but not in Herefords. Spermatozoal abnormalities generally decreased with age. BSE scores did not differ significantly among age groups. Apart from number of mounts, measures of sex drive did not differ with age or breed of bulls. Semen production data from 792 Norwegian Red AI bulls, of 46 sires, were analyzed. Semen production was affected by AI center, year and season of collection, number of months the bull had been collected, and sire (each P < 0.01). Heritability for produced accepted doses was estimated to be 0.34 (+/-0.13). This should allow a successful phenotypic selection for this trait. A negative correlation between number of accepted and number of discarded doses (-0.82+/-0.11) was found. Selection based on number of accepted doses per bull will therefore result in improvement of semen quality.  相似文献

Semen from 200 Holstein bulls in an artificial insemination center was examined for the frequency of craters on the surface of sperm heads, as visualized with the aid of differential interference contrast microscopy. Semen from 100 of these bulls was examined in more detail in 2 experiments by staining with eosin-aniline blue to determine the relationship of unstained spermatozoa, and spermatozoa with normal acrosomes with apical ridges to the incidence of craters and fertility. Only 3 of 100 bulls had a substantial incidence of craters (15 to 23%), whereas the average of the other 97 bulls in 2 experiments was 1 to 3%. The percentage of sperm cells with craters was correlated (P < 0.05) with the percentage of unstained spermatozoa (r = -0.29 and sperm cells with normal acrosomes (r = -0.52) but was not significantly correlated (r = -0.24) with the nonreturn rate. One bull with many sperm cells with craters was slaughtered, and the epididymal spermatozoa were examined. The high incidence of sperm cells with craters was limited to one side, with the testis on that side having 2 Sertoli cell tumors. The remaining 2 bulls as well as one other that produced 16% of sperm cells with craters did so only temporarily. Within a few months crater sperm production had decreased and semen quality increased. The condition usually appears to be transitory, presumably due to temporary stress.  相似文献

The objective was to evaluate the relationship of a competitive index (CI) determined by heterospermic performance and post-thaw semen quality of the same stored ejaculates. Semen from multiple ejaculates collected in succession from each bull (four Holstein and four Jersey) was pooled. Heterospermic doses (20x10(6)/straw) were made to obtain all possible Holstein-Jersey combinations (16 two-bull combinations) and contained 20x10(6) sperm/mL/bull. Cows at two University dairy farms were inseminated on observed or synchronized estrus. The sire of calves (N=460) were determined and a CI was determined for each bull (based on the number of calves sired). Prior to preparation of the heterospermic doses, a sub-sample of semen from each bull was taken, processed, frozen, and stored concurrently with heterospermic samples. Post-thaw semen samples (homospermic) from each bull were assessed for: sperm morphology, acrosome integrity, sperm motility parameters assessed by computer assisted sperm analysis (CASA), flow cytometry analysis of DNA Fragmentation Index (DFI), and Plasma Membrane Integrity (PMI). Heterospermic performance of Holstein bulls was superior to that of Jersey bulls. The DFI was negatively correlated to CI (r=-0.87; P<0.001), whereas the PMI (r=0.87; P<0.001) and total progressive motility (r=0.74; P<0.05) assessed by CASA were positively correlated to CI. In multivariate regression models, the DFI and PMI accounted for 87% variance in competitive index. In conclusion, bulls with less DFI and higher PMI had higher probabilities of siring calves.  相似文献
