I question Hanski's [I. Hanski, A practical model of metapopulation dynamics, J. Animal Ecol. 63 (1994) 151] assumption that incidence functions are relevant approximations of the equilibrium dynamics of stochastic metapopulation models to estimate models' parameters based on snapshot data. Based on ten different metapopulation models, this assumption is found to be at least partly unjustified when referring to the asymptotic behaviour of the models. This leads me to recommend the use of explicit extinction-colonisation transition probabilities and process data (rather than snapshot data) in the estimation process of metapopulation models.  相似文献

Summary .   In a presentation of various methods for assessing the sensitivity of regression results to unmeasured confounding, Lin, Psaty, and Kronmal (1998, Biometrics 54 , 948–963) use a conditional independence assumption to derive algebraic relationships between the true exposure effect and the apparent exposure effect in a reduced model that does not control for the unmeasured confounding variable. However, Hernán and Robins (1999, Biometrics 55 , 1316–1317) have noted that if the measured covariates and the unmeasured confounder both affect the exposure of interest then the principal conditional independence assumption that is used to derive these algebraic relationships cannot hold. One particular result of Lin et al. does not rely on the conditional independence assumption but only on assumptions concerning additivity. It can be shown that this assumption is satisfied for an entire family of distributions even if both the measured covariates and the unmeasured confounder affect the exposure of interest. These considerations clarify the appropriate contexts in which relevant sensitivity analysis techniques can be applied.  相似文献

The discussion about the ethics of xenotransplantation seems to focus upon the benefits for individual patients and the potential risks for human society, in general, to contract a newly emerging retrovirus. In these risk-benefit considerations, the moral concern for the research animals involved appears to be absent. This is remarkable, because the presumed successful xenograft is not expected very soon. A lot of basic problems in pig and primate xenotransplantations still need solving. These new experiments in our own biomedical laboratories raise questions regarding animal welfare and ethical justification in the light of possible alternative strategies. In this article, I discuss some of the moral issues related to preclinical, fundamental xenotransplantation research.  相似文献

Two models have been used to describe indefinite self-association (stacking). In the more popular isodesmic model addition of molecules to the growing stack occurs with the same equilibrium constant, while in the attenuated model successive equilibrium constants decrease in value. In an attempt to choose between the two models application was made of the maximum-entropy method. This paper points out that the conclusions drawn from this method are not proven as the application assumed specific limiting values for experimental values of a molecule in the interior of a stack, and these values are not identical in the two models for the two systems considered.  相似文献

It is becoming increasingly apparent that the species abundance distribution of many ecological communities contains multiple modes, a phenomenon that has been largely overlooked. Here, we test for multiple modes in the species abundance distribution using a combination of one, two and three mode Poisson lognormal distributions and an extensive arthropod dataset from the Azores. We consider the abundance distribution of twelve native laurisilva forest fragments and the combination of fragments within five islands, allowing us to detect whether patterns are consistent across scales. An information theoretic approach is employed to determine the best model in each case. To explore the processes driving multimodal abundance distributions we tested various potential mechanisms. We classified species as core if they are present in over half the fragments in our study, and as satellite if they are sampled in fewer than half the fragments. Furthermore, species are classified based on body size, whether they are indigenous (i.e. endemic or native non‐endemic) or introduced to the Azores, abundance in land uses other than native forest, and dispersal ability. We find that models incorporating multiple modes perform best for most fragments and islands. A large number of communities are bimodal, comprising a mode of very rare species and a mode of relatively common species. Deconstructing the full assemblages into their constituent subsets reveals that the combination of ecologically different groups of species into a single sample underpins the multimodal pattern. Specifically, the rarer mode prevailingly contains a higher proportion of satellite taxa, introduced species and species that are more adapted to anthropogenic land uses that surround the native forest.  相似文献

The discovery of RNA interference (RNAi) and the development of technologies exploiting its biology have enabled scientists to rapidly examine the consequences of depleting a particular gene product in a cell or an animal. The availability of genome-wide RNAi libraries targeting the mouse and human genomes has made it possible to carry out large scale, phenotype-based screens, which have yielded seminal information on diverse cellular processes ranging from virology to cancer biology. Today, several strategies are available to perform RNAi screens, each with their own technical and monetary considerations. Special care and budgeting must be taken into account during the design of these screens in order to obtain reliable results. In this review, we discuss a number of critical aspects to consider when planning an effective RNAi screening strategy, including selecting the right biological system, designing an appropriate selection scheme, optimizing technical aspects of the screen, and validating and verifying the hits. Similar to an artistic production, what happens behind the screen has a direct impact on its success.  相似文献

The availability of tools for probing the genome and proteome more efficiently has allowed for the rapid discovery of novel genes and peptides that play important, previously uncharacterized roles in neuroendocrine regulation. In this review, the role of a class of neuropeptides containing the C-terminal Arg-Phe-NH(2) (RFamide) in regulating the reproductive axis will be highlighted. Neuropeptides containing the C-terminal Phe-Met-Arg-Phe-NH(2) (FMRFamide) were first identified as cardioregulatory elements in the bi-valve mollusk Macrocallista nimbosa. During the past two decades, numerous studies have shown the presence of structurally similar peptides sharing the RFamide motif across taxa. In vertebrates, RFamide peptides have pronounced influences on opiatergic regulation and neuroendocrine function. Two key peptides in this family are emerging as important regulators of the reproductive axis, kisspeptin and gonadotropin-inhibitory hormone (GnIH). Kisspeptin acts as the accelerator, directly driving gonadotropin-releasing hormone (GnRH) neurons, whereas GnIH acts as the restraint. Recent evidence suggests that both peptides play a role in mediating the negative feedback effects of sex steroids. This review presents the hypothesis that these peptides share complementary roles by responding to internal and external stimuli with opposing actions to precisely regulate the reproductive axis.  相似文献

There is much controversy in the literature concerning prevalence, cause, diagnosis and treatment of hyperkinesis. The subject is complex; there is much ambiguity, and many research studies and reports of successful treatment appear to be overly simplistic in their approach to the problem. The use of the term "hyperkinesis" as synonymous with "hyperactivity" and often also with "minimal cerebral dysfunction" causes much confusion. There appear to be some common underlying beliefs about hyperkinetic children; these are critically examined. From integration of reports in the literature with clinical experience, it is contended that the term hyperkinesis should be restricted to the definition used in the British studies, and hyperactivity should be considered only a symptom. Hyperactivity has a large number of underlying causes, and management plans need to be individualized.  相似文献
