The effects of plant growth regulators were investigated onanthocyanin synthesis induced by removing auxin from carrotsuspension cultures. Of the auxins tested, 2,4-D showed thestrongest inhibiting effect on anthocyanin synthesis and hadthe strongest promoting effect on undifferentiated growth. When2,4-D was added to anthocyanin synthesizing cells, in whichcell division had ceased, anthocyanin synthesis was repressedimmediately, accumulated anthocyanin disappeared and cell divisionresumed. All cytokinins examined promoted anthocyanin synthesisin the absence of auxin. Both gibberellic acid (GA3) and abscisicacid inhibited anthocyanin synthesis in media lacking 2,4-D,though GA3 showed no effect on cell division. These effectsof growth regulators on anthocyanin synthesis are similar tothose reported for their effects on embryogenesis [Fujimuraand Komamine (1975) Plant Sci. Lett. 5: 359, (1979) Z. Leaflet pairs from detached pinnae of Mimosa pudica opened afterthe pinnae had been irradiated with light (2 W·m–2)of 726 or 403 nm, whereas they remained almost closed with lightof 585 or 656 nm. Light-induced leaflet opening was observedonly in the daytime, from 6:00 to 16:00. Application of IAAat more than 30 µg/ml to the cut end of the pinna rachisesmade the leaflets open even in darkness with almost constantlag times of about 100 min which were independent of the concentration.NAA and 2,4-D also made the leaflet open at lower concentrationsthan IAA. Auxin-induced leaflet opening showed diurnal variation.Application of IAA for 2 to 6 min, depending on the concentration,was enough to open the leaflets. Tentoxin strongly inhibited the ATPase activity of isolatedcoupling factor 1 (AF1) from the cyanobacterium Anacystis nidulans,with 50% inhibition occurring at 0.3 µM. When thylakoidsfrom A. nidulans were preincubated with 0.3 µM tentoxinfor 30 min, photophosphorylation was inhibited by 50%. Measurementsof fluorescence from 9-aminoacridine indicated that tentoxininhibited the utilization of the proton gradient by ATP formationin thylakoids. These results indicate that tentoxin is a strongenergy-transfer inhibitor of photophosphorylation in A. nidulans.Tentoxin decreased the level of ATP in intact cells both inthe light and in darkness, its effects being much stronger inthe dark. The activities of enzymes involved in general phenylpropanoidmetabolism were followed in a carrot suspension culture duringthe induction and reduction of anthocyanin synthesis regulatedby 2,4-D. When no anthocyanin synthesis occurred in a mediumcontaining 2,4-D (+2,4-D medium), the activities of phenylalanineammonia-lyase (PAL) and 4-coumarate:CoA ligase (4CL) increased1 day after transfer due to the transfer effect, but subsequentlydecreased and remained at a low level. Cinnamate-4-hydroxylase(C4H) activity showed a low level throughout culture. When cellswere transferred to a medium lacking 2,4-D (–2,4-D medium),the activities of PAL, C4H and 4CL increased and maximum activitiesof these enzymes were observed 6–7 days after transfer,when anthocyanin was most rapidly synthesized. When cells were cultured in the –2,4-D medium, the additionof 2,4-D immediately reduced the induced activity of PAL. PALactivity was super-induced by the transfer effect, while anthocyaninsynthesis decreased. The addition of intermediates of generalphenylpropanoid metabolism, with 2,4-D, to the medium 6 daysafter transfer to the –2,4-D medium did not promote anthocyaninsynthesis, whereas dihydroquercetin did promote it. Trichosporon cutaneum WY2-2 was shown to metabolize p-hydroxybenzoatevia protocatechuate and hydroxyquinol. Using superoxide dismutaseas a stabilizer of hydroxyquinol, the conversion of protocatechuateto hydroxyquinol and the ring fission process of hydroxyquinolwere confirmed. Hydroxyquinol was chemically identified as theproduct of protocatechuate hydroxylase reaction. Partially purifiedprotocatechuate hydroxylase was highly specific for protocatechuate;its Km values for protocatechuate and NADH were 17.6 and 12.4µM, respectively. It catalyzed equimolar CO2 formation,NADH oxidation and O2 consumption from protocatechuate. A cDNA for the phytochrome of the fern Adiantum capillus-venerisL. was cloned and sequenced. The deduced phytochrome is 50{smalltilde}55% identical to phytochromes of seed plants, and 68%identical to Selaginella phytochrome. cDNA for an acid invertase (EC 3.2.1.26 [EC] ) of tomato (Lycopersiconesculentum Mill.) fruit was introduced into tomato plants underthe control of the cauliflower mosaic virus 35S promoter inthe antisense orientation. The antisense gene effectively suppressedthe invertase activity in soluble and cell wall fractions fromripening fruits. The sucrose content of fruits of the transformantswas markedly increased, while the hexose content was reduced.These results indicate that acid invertase is one of main determinantsof the sugar composition of tomato fruit. The invertase activityin the cell wall fraction of the leaf tissues of the transformantswas not suppressed to the same extent as that in the solublefraction. Wounding of the control leaf tissues induced invertaseactivity in both soluble and cell wall fractions. The inductionof activity in the soluble fraction was suppressed by the antisensegene, while that in the cell wall fraction was unaffected. The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. Suspension cultures of Oryza sativa L. var IR 20 grew in Murashigeand Skoog medium (MS) supplemented with 2,4-D and kinetin ina density-dependant manner with a critical minimum inoculation-densityof 10,000 cells ml–1. Conditioned medium obtained fromthese cultures and added to MS+2,4-D+kinetin induced the growthof cultures at 1,000 cells ml–1. Growth stimulation byconditioned medium was mimicked by spermidine but not by otherpolyamines viz. putrescine and spermine. The thiamine requirements of cells of several plant speciesin suspension culture were investigated. Omission of thiaminefrom the medium leads to a rapid and complete cessation of growthin subcultures of soybean, tobacco and rice cells. The criticallevels of thiamine content in these cells are considered tobe in the vicinity of 0.6, 0.5 and 0.2 µg per g dry weight,respectively. Soybean cells can grow satisfactorily when suppliedwith an equimolar mixture of two thiamine precursors, pyrimidineand thiazole moieties. Neither moiety alone can substitute forthiamine. On the other hand, Rula and peanut cells can be successfullysubcultured for ten passages in the absence of externally suppliedthiamine. When grown without thiamine, Rula cells had an averagethiamine content of 0.5-0.6 µg in darkness and 3.5 µgper g dry weight in the light. Etiolated oat protoplasts were treated with dibutyryl cAMP tostudy possible function of cAMP in the development by measuringthe protoplast swelling. The mean diameter of protoplasts inthe absence of any chemical treatment was 33.58±1.26(SE) µm, which increased to 36.96±0.86 µmin the presence of 100 µM dibutyryl cAMP. Prostacyclin,a potent activator of adenyl cyclase, also showed a significantswelling effect (diameter 38.01±0.98 µm). Sulphite-cytochrome c reductase (sulphite: ferricytochrome coxidoreductase, EC 1.8.2.1 [EC] ) derived from Thiobacillus novelluswas purified by chromatography on a DEAE-cellulose column andby gel filtration with a Sephadex G-100 column. Although thereductase thus purified moved as a single band both in gel filtrationand in isoelectric focusing it was always split into two bandsby polyacrylamide gel electrophoresis; the one had the enzymaticactivity and showed absorption spectrum of cytochrome, whilethe other had no activity and was colourless, in contrast withthe results reported by Charles and Suzuki [(1966) Biochim.Biophys. Acta 128: 522]. The enzymatic properties of the purifiedreductase were almost the same as those of the enzyme obtainedby Charles and Suzuki. Cytochrome c-551 free of the reductase activity was obtained.Its molecular weight was determined to be 23,000 by polyacrylamidegel electrophoresis in the presence of sodium dodecyl sulphate.The cytochrome seemed to exist in the organism as a complexwith the reductase or a subunit of the enzyme. In the stateof the complex with the enzyme, the cytochrome was reduced veryquickly on addition of sulphite, while the cytochrome free ofthe reductase activity was hardly reduced by the enzyme withsulphite. Intravacuolar pigmented structures occurred in anthocyanin-producingcultured cells of sweet potato (Ipomoea batatas) were characterized.Formation of the pigmented structures in sweet potato cellswas induced by transfer of callus cultured in 2,4-D containingagar medium into 2,4-D free liquid medium under continuous illumination.These structures were found in the vacuoles. The pigmented structureswere isolated from the protoplasts by precipitation in 60% (w/w)sucrose after centrifugation. Electron microscopic observationsof the anthocyanin-containing cultured cells showed these structureshad neither membrane boundary nor internal structures, and werefound as strongly osmiophilic globules in vacuoles. Numeroussmall osmiophilic globules were observed in central vacuolesat the early stage of anthocyanin accumulation, but not foundin cytoplasm. Similar pigmented structures in vacuoles werealso formed by treatment with neutral red. A high-affinity binding site for N-acetylchitooligosac-chlarideelicitor was found to localize in the plasma membrane from suspension-culturedrice cells. Binding kinetics as well as the specificity of thisbinding site corresponded well with the behavior of the ricecells to the editor. Regulatory effects of light on senescence of rice leaves wereinvestigated by measuring degradation of chlorophyll and proteinsin leaf segments which had been kept in the dark or under illuminationwith light of different intensities and colors. When leaveshad been left in total darkness for three days at 30°C,there was an initial long lag that lasted for one whole dayand then chlorophyll was rapidly degraded in the second andthird days. Breakdown of chlorophyll was strongly retarded bycontinuous illumination with white light of intensity as lowas 0.5 µmol photons m–2 s–1 but the effectof light decreased at intensities above 10 µmol photonsm–2 s–2. The initial lag and subsequent degradationof chlorophyll in the dark were little affected by illuminationwith red or far red light at the beginning of dark treatment.However, a brief illumination with red light at the end of thefirst and/or second day significantly suppressed degradationof chlorophyll during subsequent dark periods and the effectof red light was nullified by a short irradiation with far redlight. Thus, degradation of chlorophyll is regulated by phytochrome.Thylakoid membrane proteins and soluble proteins were also largelydegraded during three days in the dark. Degradation of membraneproteins such as the apoproteins of light-harvesting chlorophylla/b proteins of photosystem II and chlorophyll a-binding proteinsof reaction center complexes showed a long lag and was stronglysuppressed by illumination with weak white light. Thus, theloss of chlorophyll can be correlated with degradation of chlorophyll-carryingmembrane proteins. By contrast, light had only a weak protectingeffect on soluble proteins and ribulose-1,5-bisphosphate carboxylase/oxygenaserapidly disappeared under illumination with weak white light.Thus, breakdown of thylakoid membrane and soluble proteins aredifferently regulated by light. Dark-grown cells of a mutant strain of Chlorella regularis containedchlorophyll a and protochlorophyll, phytyl ester of protochlorophyllide.Under illumination, protochlorophyll was quantitatively anddirectly converted into chlorophyll a. The photoconversion wasdependent on light intensity and temperature and proceeded ina cell-free preparation. The effect of azide, 2,4-dinitrophenol, malonate and 2-deoxyglucoseon the physiology and morphology of normal and mutant strainsof Schizophyllum commune was studied. Normal, thin and puffstrains showed a decrease in dry weight and an increase in glucoseutilization in response to azide, dinitrophenol and 2-deoxyglucose.Streak was less sensitive to azide and dinitrophenol and almostcompletely resistant to 2-deoxyglucose. Numerous morphogeneticeffects were caused by the inhibitors. These included the inductionof certain aspects of the morphology of the common-A sexualreaction in normal and thin homokaryons by 2-deoxyglucose, areversion to normal morphology by thin strains in response toazide and dinitrophenol, and a morphogenetic insensitivity tothe inhibitors by the streak mutants. A novel NAD(P)H-quinone oxidoreductase (NQR) was isolated fromthe cyanobacterium Synechocystis PCC6803 by ion-exchange, affinityand gel-filtration chro-matographies. Isolated NQR was foundto be a drgA gene product that was a homodimer composed of 23-kDasub-units. It showed NAD(P)H-plastoquinone oxidoreductase activitywith Km values for NADPH and NADH of 12 and 48 µM respectively.The activity was inhibited by thiol-modifying reagents, butnot by rotenone, amobarbital, salicylhydroxamic acid, dicumarol,flavone, or diphenylene-iodonium chloride. Therefore, the Cys-147residue is probably involved in the catalytic reaction. Protoplasts prepared from suspension-cultured Vinca rosea cellswere grown in a liquid medium, and the effects of osmolarityand growth regulators on cell division and on the compositionof regenerated cell walls were investigated. The concentrationof mannitol optimal for cell division was 0.3–0.4 M. Thepresence of 2,4-D was essential for cell division, and 6-benzylaminopurine(BAP) enhanced cell division at concentrations of 0.03–0.1ppm. However, the composition of regenerated cell walls wasabnormal under suspension culture; the predominant sugar wasglucose, indicating that the regenerated cell walls consistedmostly of glucans, and that the other cell-wall components werereleased into the medium. Mannitol, 2,4-D, and BAP at variousconcentrations did not significantly affect the sugar compositionof the regenerated cell walls. Compared with liquid culture,cell division was stimulated when protoplasts were culturedon agar, and their regenerated cell walls had a compositionsimilar to that of the original culture.
