The performance of first‐generation hybrids determines to a large extent the long‐term outcome of hybridization in natural populations. F1 hybrids can facilitate further gene flow between the two parental species, especially in animal‐pollinated flowering plants. We studied the performance of reciprocal F1 hybrids between Rhinanthus minor and R. major, two hemiparasitic, annual, self‐compatible plant species, from seed germination to seed production under controlled conditions and in the field. We sowed seeds with known ancestry outdoors before winter and followed the complete life cycle until plant death in July the following season. Germination under laboratory conditions was much lower for the F1 hybrid formed on R. major compared with the reciprocal hybrid formed on R. minor, and this confirmed previous results from similar experiments. However, this difference was not found under field conditions, which seems to indicate that the experimental conditions used for germination in the laboratory are not representative for the germination behaviour of the hybrids under more natural conditions. The earlier interpretation that F1 hybrid seeds formed on R. major face intrinsic genetic incompatibilities therefore appears to be incorrect. Both F1 hybrids performed at least as well as and sometimes better than R. minor, which had a higher fitness than R. major in one of the two years in the greenhouse and in the field transplant experiment. The high fitness of the F1 hybrids confirms findings from naturally mixed populations, where F1 hybrids appear in the first year after the two species meet, which leads to extensive advanced‐hybrid formation and introgression in subsequent generations.  相似文献

The aim of this study was to develop a new fungal strain that simultaneously amplifies the carbon source spectrum and increases arachidonic acid (AA) productivity using genome shuffling between Diasporangium sp. and inactive Aspergillus niger. Through three rounds of genome shuffling, one of the stable daughter strains (F1) acquired the ability to produce arachidonic acid and utilize various carbon sources. Compared to the parental Diasporangium sp., which could only use four out of eight carbon sources tested, F1 could utilize all eight carbon sources. During fermentation with CMC-Na as the carbon source, F1 was able to obtain 30.16% of lipid effectively whereas the parental Diasporangium sp. was not able to grow at all. When glucose was used as the carbon source, the CMCase activity of F1 was 879.36 U, 298.23% higher than that of the parental Diasporangium sp. Under optimized fermentation conditions in a 5-L fermentation container, the AA yield of F1 reached 0.81 g/l, 94.78% higher than that of the parental generation. These results indicate that inter-kingdom genome shuffling can be used successfully in eukaryotic microorganisms and that it can effectively improve the production of desired metabolites within a short period of time. The findings of this study may be useful for extending the application of genome shuffling in eukaryotic microbial breeding.  相似文献

Quercus crassifolia and Q. crassipes are dominant species in temperate forests of central Mexico and hybridize between each other when they occur in sympatry. Oak canopies contain a considerable portion of arthropod diversity and the hybrid zones can provide new habitats to epiphyte fauna. We tested if the establishment of hybrids in contact zones with their parental hosts increases the species diversity of canopy arthropods assuming that hybrid trees constitute new genotypes of potential new habitats to small organisms. We examined the effect of hybridization on some community structure parameters (diversity, composition, similarity and density of arthropod fauna) of canopy arthropods compared to their parental species in a hybrid zone located in central Mexico. We employed 17 leaf morphological traits and six diagnostic RAPD primers to identify parental and hybrid plants. The RAPDs marker showed unidirectional introgression towards Q. crassifolia, and were detected hybrid (F1), backcrosses and introgression individual trees. In total, 30 oak canopies were fogged during rainy and dry season. We recognized 532 taxa of arthropods belonging to 22 orders associated with tree canopies. The taxonomic status of host‐trees may be an important factor in the arthropod community structure and that seasonality (dry and rainy) is not a factor that could modify their organization. Trees of Q. crassipes registered the highest densities of arthropod fauna followed by hybrid hosts (F1); trees originated by backcrosses towards Q. crassifolia registered a significant less arthropod density than F1 hybrids; and trees of Q. crassifolia had the lowest density. Hybrid plants and Q. crassipes individuals had higher diversity (H′) of arthropods than Q. crassifolia plants. Hybrid plants had also more rare species in both seasons in comparison with parental species. This study suggests that hybrid oaks act as a center of biodiversity by accumulating arthropods of both parental and different species including a considerable number of rare species.  相似文献

Recognizing the predominant mode of selection in hybrid systems is important in predicting the evolutionary fate of recombinant genotypes. Natural selection is endogenous if hybrid genotypes are at a disadvantage relative to parental species independent of environment. Alternatively, relative fitness can vary in response to environmental variation (exogenous selection), and hybrid genotypes can possess fitness values equal to or greater than that of parental species. I investigated the nature of natural selection in a leopard frog hybrid system by rearing larvae of hybrid and parental genotypes between Rana blairi and R. sphenocephala in 1000-L outdoor experimental ponds. Three hybrid (F1, backcrossj [B1], backcross2 [B2]) and two parental (R. blairi [BB] and R. sphenocephala [SS]) larval genotypes were produced by artificial fertilzations using adult frogs from a natural population in central Missouri. Resultant larvae were reared in single-genotype populations and two-way mixtures at equal total numbers from hatching to metamorphosis. In single-genotype ponds, F1 hybrid larvae had highest survival and BB were largest at metamorphosis. When F1 and SS larvae were mixed together, F1 hybrids had reduced survival and both F1 and SS larvae metamorphosed at larger body masses than when reared separately. When mixed, both B1 and SS larvae had shorter larval period lengths than when reared alone. Higher proportion of B1 metamorphs were produced when larvae were mixed with either parental species than when reared alone. Larval fitness components as measured by survival, body mass at metamorphosis, proportion of survivors metamorphosing, and larval period length for B2 hybrid and BB larvae were similar in single-genotype populations and mixtures. Comparison of composite fitness component estimates indicated hybrid genotypes possess equivalent or higher larval fitness relative to both parental species for the life-history fitness components measured. Despite reduced survival of F1 hybrids in mixtures, backcross-generation hybrid genotypes demonstrated high levels of larval growth, survival, and metamorphosis in mixtures with parental species. Consequently, this study suggests natural hybridization and subsequent backcrossing between R. blairi and R. sphenocephala can produce novel and relatively fit hybrid genotypes capable of successful existence with parental species larvae. Thus, the evolutionary fate of hybrid and parental genotypes in this system may be influenced by exogenous selection mediated by genotypic composition of larval assemblages.  相似文献

Hybrid fitness is an important parameter to predict the evolutionary consequences of a hybridization event and to characterize hybrid zones. We studied fitness parameters of F1 and later‐generation hybrids between the lowland species Salix purpurea and the alpine S. helvetica that have recently emerged during colonization of an alpine glacier forefield. Fruit production (number of capsules per catkin and fruit set) did not differ between hybrids and parents, but the number of seeds per capsule of F1 hybrids was slightly lower than that of later‐generation hybrids and of the parents. Germination rates and seedling growth were tested on three substrates (pH 4.5, 7.0, and 8.0). Germination rates of seeds collected from F1 hybrids were lower on acid and neutral substrates, but equal at pH 8.0 compared to all other groups, while the seeds from later‐generation hybrids performed as well as the parents on all three substrates. In seedling growth, the colonizer S. purpurea performed better than all other taxa on all three substrates, while hybrids resembled the subalpine species S. helvetica. Results suggest that endogenous selection acts against F1 hybrids, but favors fitter genotypes in later‐generation hybrids. Exogenous selection via soil pH appears to be weak during seedling establishment. The pioneer vegetation on the glacier forefield may offer sufficient niche space for hybrid seedlings. Owing to the relatively high fitness of the hybrids and the scattered distribution of hybrids and parental individuals on the glacier forefield, this hybrid zone can be assigned to a mosaic model, probably facilitating gene flow and introgression between the parental species. As establishment of the hybrid zone appears to be linked to a colonization process, we propose to call it a pioneer mosaic hybrid zone.  相似文献

The possible involvement of gibberellins and amylase in heterosisof maize seedlings was investigated in two parental inbreds,CM7 and CM49, and their single cross F1 hybrid, CM7xCM49. Germinationof all three genotypes was complete within 36 h after the onsetof imbibition. By 48 h, heterosis (hybrid vigour) for increasedshoot and root length was consistently observed. The endogenousconcentration of gibberellin A1 (GA1) was measured in 48 h seedlingsby gas chromatography-mass spectrometry with selected ion monitoring(GC-SIM) using [2H2]-GA1 as an internal standard. The GA1 concentrationwas highest in the hybrid (59 ng g–1 dry wt.), intermediatein CM49 (9.0 ng g–1), and lowest in CM7 (<5.0 ng g–1).Amylase activities in all three genotypes were very low at 24h, but increased during the next 24 h, after which time amylaseactivity in the hybrid was significantly higher than that ofeither parental inbred. Inhibitors of gibberellin (GA) biosynthesis,AMO-1618 or CCC, inhibited germination, shoot and root growth,and amylase activity in all three genotypes. Conversely, exogenousgibberellic acid (GA3) increased amylase activity, particularlyin the inbred CM7. Amylase isozymes were separated through polyacrylamidegel electrophoresis and generally similar profiles of starchdegrading enzymes were observed in the three genotypes. SinceGA is known to control a-amylase biosynthesis in some cereals,these results are consistent with the hypothesis that GAs areinvolved in the regulation of heterosis in maize. A higher endogenousGA1 concentration in the hybrid could result in increased amylaseactivity in the hybrid seedlings and consequently, more rapidstarch hydrolysis which fuels heterosis for early growth. Genetic control of alcohol dehydrogenase isozymes in maize   总被引:2,自引:0,他引:2  
By means of horizontal gel electrophoresis and the zymogram technique, genetic variants and the formation of a hybrid molecule of the enzyme alcohol dehydrogenase (ADH) have been found in Zea mays. Each inbred homozygous stock examined showed two types of ADH isozyme patterns: a fast faint zone and a slower deeply staining zone, both anode-migrating at pH 8.5. The variants found differed in that each of the ADH zones varied in its electrophoretic mobility when compared to its counterpart in the other strain. When appropriate genetic crosses were made, the resulting heterozygotes showed the parental ADH zones, and, in addition, a band of intermediate mobility was formed between the deep-staining ADH bands. However, in the fast-moving zone only the parental isozymes were represented in the heterozygote. The formation of the hybrid molecule and the apparent gene dosage effects support the hypothesis that ADH-2 in maize exists as a dimer, whereas ADH-1 may exist as a monomer.This work was supported by the U.S. Atomic Energy Commission, under contract No. AT(11-1)-1338.  相似文献

The genetic composition of a hybrid zone can provide insight into the evolution of diversification in plants. We carried out morphological and amplified fragment length polymorphism analyses to investigate the genetic composition of a hybrid zone between two violets, Viola bissetii Hemsl. and Viola rossii Maxim. Our aim was to clarify the formation and maintenance of hybrids between these Viola species. We found that most hybrid individuals (V. bissetii × V. rossii) were of the F1 generation, with a few of the F2 generation. We found no backcrosses. The scarcity of post‐F1 hybrids indicates that a species barrier is established between the parental species. The F1‐dominated hybrid zone occupied only a narrow, intermediate ecotone between the parental habitats, suggesting that selection by environmental factors against hybrids may help to maintain the current conditions in this hybrid zone.  相似文献

Comparative genetic mapping in interspecific pedigrees presents a powerful approach to study genetic differentiation, genome evolution and reproductive isolation in diverging species. We used this approach for genetic analysis of an F1 hybrid of two Eucalyptus tree species, Eucalyptus grandis (W. Hill ex Maiden.) and Eucalyptus globulus (Labill.). This wide interspecific cross is characterized by hybrid inviability and hybrid abnormality. Approximately 20% of loci in the genome of the F1 hybrid are expected to be hemizygous due to a difference in genome size between E. grandis (640 Mbp) and E. globulus (530 Mbp). We investigated the extent of colinearity between the two genomes and the distribution of hemizygous loci in the F1 hybrid using high-throughput, semi-automated AFLP marker analysis. Two pseudo-backcross families (backcrosses of an F1 individual to non-parental individuals of the parental species) were each genotyped with more than 800 AFLP markers. This allowed construction of de novo comparative genetic linkage maps of the F1 hybrid and the two backcross parents. All shared AFLP marker loci in the three single-tree parental maps were found to be colinear and little evidence was found for gross chromosomal rearrangements. Our results suggest that hemizygous AFLP loci are dispersed throughout the E. grandis chromosomes of the F1 hybrid.Communicated by O. Savolainen  相似文献

Hybridization phenomena in anurans have traditionally been studied through morphological comparisons, under the assumption that various hybrids (e.g., F1‘s, backcrosses) are predictably intermediate to parental species. We critically evaluate this assumption by examining morphology in genetically categorized hybrids between the treefrogs Hyla cinerea and H. gratiosa. A total of 202 frogs from a hybridizing population in Alabama were assayed for allozyme and mitochondrial DNA genotype and for a large suite of osteological characters. Discriminant analyses demonstrated distinct morphological separation between the genetically “pure” parental species. Morphometric analyses of genetically identified hybrids showed: 1) an extreme range of phenotypic expression within F1 and backcross classes, and 2) no apparent directional parental bias on the F1 phenotype. Had morphology alone been used as a guide, over 40 percent of the individuals with known hybrid ancestry would have been misclassified as “pure” parental species, and about 25 percent of the backcross individuals would not have been distinguished from F1‘s. These results exemplify the utility of joint comparisons of morphology and genotypic constitution in studies of natural hybridization, and they emphasize the limitations inherent in describing hybrid classes solely by morphological criteria.  相似文献

Effects of H2S on seed germination under chromium (Cr) stress were investigated in wheat (Triticum aestivum L.). Under Cr stress, the percentage of germination of wheat seeds decreased, but this decrease could be alleviated by pretreatment with NaHS, an H2S donor, in a dose-dependent manner. Furthermore, NaHS significantly enhanced the activities of amylase, esterase, superoxide dismutase, catalase, ascorbate peroxidase, and guaiacol peroxidase in Cr-stressed germinating seeds, whereas reduced the Cr-induced increase in lipoxygenase activity and over-production of malondialdehyde (MDA) and H2O2, and sustained slightly higher content of endogenous H2S.  相似文献

Summary Crosses were made, in all combinations, between 2 parental genotypes of Linum and their reciprocal F 1 hybrids. The parents and progeny obtained were grown in controlled environmental conditions and sampled at 35 and 70 days after germination to determine, on an individual plant basis, total plant fresh weight and peroxidase activity of main stem tissue. Peroxidase activity required transformation to a log10 scale, whereas the original linear scale was satisfactory for plant weight. There was no correlation between plant weight and corresponding peroxidase activity. Pronounced heterosis appeared in the F 1's for both characters at sample 1, but this heterosis had declined at sample 2 and in the F 2's. Heterosis operated in a positive direction for plant weight and in a negative direction for peroxidase activity. No consistent differences were found amongst the variances of segregating or non-segregating generations for either character.  相似文献

Two grasshopper species, Stenobothrus rubicundus and Stenobothrus clavatus, were previously shown to hybridize in a narrow contact zone on Mount Tomaros in northern Greece. The species are characterized by complex and completely different courtship songs. In the present study, we investigated female preferences for the courtship songs of S. rubicundus, S. clavatus and hybrids in playback experiments. Playback of the courtship songs revealed assortative preferences in females of the parental species: they significantly more often preferred the songs of conspecific males. Hybrid females showed a lower selectivity than parental females, responding somewhat equally eager to playback of the songs of S. clavatus, S. rubicundus, and natural hybrid song, although less actively to the F1 hybrid song. The results suggest that hybrid males may lose to males of parental species, whereas hybrid females would even have an advantage over parental females. Comparison of responses of females from allopatric populations and Mount Tomaros to different song types shows no evidence for reinforcement. Asymmetry found in female preferences may have implications for the structure of the hybrid zone. © 2013 The Linnean Society of London  相似文献

In surveys of hybrid zones, dominant genetic markers are often used to identify individuals of hybrid origin and assign these individuals to one of several potential hybrid classes. Quantitative analyses that address the statistical power of dominant markers in such inference are scarce. In this study, dominant genotype data were simulated to evaluate the effects of, first, the number of loci analyzed, second, the magnitude of differentiation between the markers scored in the groups that are hybridizing, and third, the level of genotyping error associated with the data when assigning individuals to various parental and hybrid categories. The overall performance of the assignment methods was relatively modest at the lowest level of divergence examined (Fst ~ 0.4), but improved substantially at higher levels of differentiation (Fst ~ 0.67 or 0.8). The effect of genotyping error was dependent on the level of divergence between parental taxa, with larger divergences tempering the effects of genotyping error. These results highlight the importance of considering the effects of each of the variables when assigning individuals to various parental and hybrid categories, and can help guide decisions regarding the number of loci employed in future hybridization studies to achieve the power and level of resolution desired.  相似文献

Within a broad (>200 km wide) hybrid zone involving three parapatric species of Aesculus, we observed coincident clines in allele frequency for 6 of 14 electrophoretic loci. The cooccurrence of alleles characteristic of A. pavia, A. sylvatica, and A. flava was used to estimate genetic admixtures in 48 populations involving various hybrids between these taxa in the southeastern United States. High levels of allelic polymorphism (up to 40% greater than the parental taxa) were observed in hybrid populations and also in some populations bordering the hybrid zone. A detailed analysis of a portion of the hybrid zone involving A. pavia and A. sylvatica revealed a highly asymmetrical pattern of gene flow, predominantly from Coastal Plain populations of A. pavia into Piedmont populations of A. sylvatica. Computer simulations were used to generate expected genotypic arrays for parental, F1; and backcross individuals, which were compared with natural populations using a character index scoring system. In these comparisons, hybrid individuals could be distinguished from either parent, but F1 and backcross progeny could not be distinguished from each other. Most hybrid populations were found to include hybrids and one of the parental taxa, but never both parents. Three populations appeared to be predominantly hybrids with no identifiable parental individuals. Hybrids occurred commonly at least 150 km beyond the range of A. pavia, but usually not more than 25 km beyond the range of A. sylvatica. Introgression, suggested by genetically hybrid individuals and significant gene admixtures of two or more species in populations lacking morphological evidence of hybridization, may extend the hybrid zone further in both directions. The absence of one or both parental species from hybrid populations implies a selective disadvantage to parentals in the hybrid zone and/or that hybridization has occurred through long-distance gene flow via pollen, primarily from A. pavia into A. sylvatica. Long-distance pollen movement in plants may generate hybrid zones of qualitatively different structure than those observed in animals, where gene flow involves dispersal of individuals.  相似文献

Genetic variation in the synthesis of halogenated secondary metabolites in the Japanese marine red alga Laurencia nipponica Yamada (Rhodomelaceae, Ceramiales) has been investigated in laboratory crossing experiments and chemical analyses, F1 tetrasporophytes and F1 gametophytes resulting from crosses within chemical races produced major metabolites characteristic of these races. F1 tetrasporophytes derived from reciprocal interracial crosses produced: (i) both parental types of secondary metabolites; (ii) either of the parental types; or (iii) a further major compound in addition to both parental types or in addition to either of the parental types. The latter cases suggest that hybrid-specific products were formed by the combined enzymatic complements of the parents, as F1 gametophytes derived from these interracial F1 tetrasporophytes yielded one or other of their parental products in an approximate 1:1 ratio. The population structure was analyzed at localities in Hokkaido, where two of the chemical races occur sympatrically. At Usujiri (Minami-kayabe), where the prepacifenol race and the laureatin race were sym-patric, hybrid gametophytes (recombination type) were found in high frequency in addition to hybrid tetra sporophytes, which strongly suggests that a new, pre-pacifenol/laureatm race is beginning to be produced by natural hybridization and recombination. By contrast, at Oshoro Bay, where the laurencin race and the epi-lauraliene race grew together, the interracial hybrids were rare: only a few tetrasporophytes (probably F1 generation) were found, suggesting that racial integrity may be retained by habitat segregation and/or the absence of recombination-type gametophytes.  相似文献

The heat-resistant subline 147S was obtained in Drosophila virilis by selecting for viability individuals of heat-sensitive stock 147. It was shown that in the heat-treated 147S pupae the activity of juvenile hormone (JH)-esterase is decreased and, consequently, the titer of juvenile hormone is increased compared with those in the control pupae. These changes are consistent with those observed earlier for resistant stock 101. Heat-resistant stocks 101 and 147S were crossed with heat-sensitive stock 147, whose heat-treated larvae show earlier activation and higher activity of JH-esterase than control larvae. The viability and electrophoretic esterase patterns were analyzed in the F1 and F2 hybrids at different temperatures. It was found that the F1 hybrid is resistant to the effect of high temperature and its activity level of JH-esterase is lower compared with controls. In the F2 hybrid, there was a 3:1 segregation of viability and a 1:2:1 segregation of the activity level of JH-esterase at high temperatures. It is concluded that the activity level of JH-esterase and heat resistance in D. virilis are monogenically controlled at high temperatures.  相似文献

Five F1 hybrid strains were established from rare survivors in intergroup crosses between three closely related mating groups (A, B and H) of the Closterium ehrenbergii Meneghini ex Ralfs species complex. Cell sizes of these five strains studied under our standard culture conditions were compared to those of their parental stains and also to the total range of cell-size variation in each mating group. All five F1 strains were larger in mean cell width than their parental strains. In cell length, three of them were larger than, one was the same as, and the other was intermediate between their parental strains. Their cell sizes were always larger than the range of their respective smaller parental mating group and three of them were larger than the range of their respective larger parental mating group.  相似文献
