以小麦品种'生抗1号'(对照)及其转反义trxs基因纯合株系为材料,测定了它们在种子后熟过程中硫氧还蛋白h(Trxh)、淀粉酶和蛋白酶活性以及呼吸速率等生理指标,以探讨转反义trxs基因小麦籽粒在后熟过程中的生理生化变化规律,为提高小麦耐储性探索有效途径.结果显示,在小麦后熟过程中,转基因小麦籽粒的Trxh活性、淀粉酶活性、蛋白水解酶活性和种子呼吸速率均比对照明显下降;后熟0~30 d,转基因小麦种子的Trxh活性、α-淀粉酶、β-淀粉酶、脱支淀粉酶、蛋白酶活性和种子呼吸速率平均比对照分别降低16.07%、31.64%、6.44%、21.63%、27.67%和52.90%.研究表明,反义trxs基因导入干扰或部分抑制了小麦内源同源基因的表达,致使小麦种子Trxh活性、呼吸速率和水解酶活性降低,从而延缓了籽粒储藏物质的转化速率,提高了小麦种子的耐贮藏性.  相似文献

采用考马斯亮蓝G-250染色法和SDS-PAGE电泳技术对翅果油树3种生态类型种子贮藏蛋白含量进行测定和图谱分析.结果显示:大宫灯、长果型、小宫灯3种生态类型种子中总贮藏蛋白含量分别为33.753%、32.075%和26.633%;3种生态类型种子贮藏蛋白均以谷蛋白含量最高(65.971%~68.267%),其次为球蛋白和清蛋白,醇溶蛋白的含量最低;电泳图谱显示,3种生态类型之间清蛋白、球蛋白、醇溶蛋白和谷蛋白的蛋白质条带信息均存在差异.研究表明,翅果油树3种生态类型间种子贮藏蛋白具有多态性.  相似文献

以羧甲基壳聚糖(NCMC) 处理玉米开花期果穗,发育中籽粒的谷氨酰胺合成酶(GS) 、谷氨酸脱氢酶(GDH) 及谷丙转氨酶(GPT) 活性均明显增强,而蛋白水解酶活性下降。羧甲基壳聚糖处理离体玉米籽粒酶提取液,酶液中GDH 活性明显提高。羧甲基壳聚糖处理玉米果穗,其发育籽粒中可溶性蛋白和成熟种子中贮藏蛋白含量明显提高。这都表明,羧甲基壳聚糖对玉米氮代谢、蛋白质合成与积累具有明显的生理调节作用。  相似文献

采用盆栽试验,研究了花后渍水、盐胁迫和盐渍处理对2个小麦品种(扬麦12和淮麦17)籽粒产量及蛋白质和淀粉积累与组分的影响.结果表明:与对照相比,花后渍水、盐胁迫和盐渍处理显著降低了小麦花前贮藏氮素(花前贮藏干物质)转运量和花后同化氮素(花后同化物)输入籽粒量,从而导致小麦籽粒产量、蛋白质和淀粉产量显著降低,其中盐胁迫和盐渍处理表现更为明显.与对照和渍水处理相比,盐胁迫和盐渍处理显著降低了小麦籽粒蛋白质积累量及谷/醇蛋白比,显著提高了蛋白质组分含量;同时降低了小麦籽粒淀粉积累量、淀粉组分含量及直/支链淀粉比;盐胁迫处理对扬麦12的影响较盐渍处理明显,而盐渍处理对淮麦17的影响较盐胁迫处理明显.渍水条件下小麦籽粒蛋白质和淀粉积累量均下降,除淮麦17谷蛋白和清蛋白含量有所提高外,淮麦17其他蛋白组分含量和扬麦12各蛋白组分含量均下降.  相似文献

系统比较了转多聚半乳糖醛酸酶(PG)反义基因和对照番茄果实成熟过程中绿熟、转色、粉顶、粉红、全红5个时期的PG活性和与其相关的生理、生化组分的动态变化。实验表明,转基因果与对照果相比,PG活性始终处于较低水平,PG活性强烈被抑制是在全红期;果实的硬度、贮藏寿命指数都高于对照果;番茄红素合成积累进程被延缓;可溶性果胶含量、电解质外渗百分率均低于对照果。外源乙烯刺激引起对照果PG活性剧增,而转基因果表现钝化。讨论了PG活性与果实成熟、耐贮性及软化的关系,及对外源乙烯刺激的敏感性。首次明确提出PG活性在对照果中极大地表达,在转基因果中强烈被抑制是在全红期 ,而不是在整个成熟期;PG活性在果实软化中起直接和重要作用;PG活性的高低与番茄红素的合成与积累有关。  相似文献

以半蔓生型奶花芸豆(Y09)和直立型奶花芸豆(Y06)为材料,通过SDS-PAGE分析研究了开花后种子形成过程中贮藏蛋白(SP)的积累规律.结果表明:2个芸豆品种开花后种子形成过程中,子叶、胚蛋白积累变化趋势基本一致,但其变化幅度存在显著差异,其中,在开花后20d内,半蔓生型芸豆Y09的籽粒蛋白积累量大于直立型芸豆Y06,但在开花后25~40d,Y06籽粒蛋白积累量显著高于Y09;两芸豆品种的子叶和胚均含有丰富的贮藏蛋白(SP)(14.4~97.4kD),其中子叶的41.0、43.9、39.4kD3种蛋白含量较高,约占子叶总蛋白含量(光密度)50%以上,而胚蛋白亚基分布相对均匀;两品种的子叶、胚中蛋白亚基差异较小,只有个别亚基存在差异.  相似文献

以3种不同转基因抗虫棉及其亲本对照为材料,研究了盆栽条件下不同品种棉花苗期氮代谢特征。结果表明与非转基因棉花比较,转Bt基因棉Z30叶片内的硝酸还原酶活性、肽酶活性、游离氨基酸含量均没有明显变化,但转氨酶活性显著增加(增幅为14.03%),可溶性蛋白含量显著减少(降幅为26.29%)。双价转基因棉花CCRI41叶片内除可溶性蛋白含量没有明显变化,其它所测指标均发生了显著的改变。双价转基因棉花SGK321叶片内硝酸还原酶活性、转氨酶活性、氨基酸含量没有明显变化,但肽酶活性和可溶性蛋白含量均显著增加(增幅分别为7.01%和121.32%)。随着转入基因的多样化,其可能引发转基因棉花产生的非预期效应更加不确定与复杂。  相似文献

测定了水稻、玉米和狼尾草花粉在低温(4℃),低湿(RH45%),低温(4℃)高湿(RH90%),高温(35℃),低湿(RH45%)和高温(35℃),高湿(RH90%)4种贮藏条件下的蛋白酶活性,蛋白质含量,游离氨基酸含量的变化,结果表明:在低温高湿条件下,蛋白质降解和游离氨基酸含量上升慢;在高温条件下蛋白质降解快,游离氨基酸含量迅速上升。3种植物花粉中,水稻花粉蛋白酶活性强,在贮藏过程中蛋白质降解速率和游离氨基酸含量上升快:狼尾草花粉蛋白酶活性低,在贮藏过程中蛋白质降解速率和游离氨基酸含量上升慢;玉米花粉的蛋白酶活性、蛋白质降解速率和游离氨基酸含量上升速率居两者之间。  相似文献

以‘翠冠’梨为材料,研究了套双层遮光纸袋梨果实贮藏过程中蔗糖、果糖、葡萄糖、山梨醇及糖代谢中酶活性的变化规律。结果表明,贮藏套袋梨果实中果糖、葡萄糖、山梨醇和蔗糖含量都低于未套袋对照;套袋梨果实中山梨醇脱氢酶活性在贮藏的前5d都低于对照,贮藏10d后活性均高于对照,且与山梨醇含量呈现极显著正相关;贮藏套袋梨果实中蔗糖磷酸合酶(SPS)及蔗糖合酶(SS)分解和合成方向活性都是前期低于对照,贮藏后期都高于对照,且蔗糖含量与蔗糖磷酸合酶和蔗糖合酶(分解方向)活性都呈显著正相关;贮藏的套袋梨果实和对照中的山梨醇含量与果糖含量均呈极显著负相关,蔗糖含量与葡萄糖含量呈极显著负相关,即在贮藏过程中山梨醇可能转化为果糖,而蔗糖则转化成葡萄糖。  相似文献

利用转基因技术是改良大麦品种品质的有效途径.研究了转trxS基因对大麦种子发芽过程中水解酶活性的影响,结果表明转基因种子中α-淀粉酶、自由态β-淀粉酶和极限糊精酶的活性比未转基因种子高;转基因种子醇溶蛋白和谷蛋白中巯基的含量提高,说明该基因能够表达,为大麦育种和品质改良提供新的途径.  相似文献

The genetic associations and differences of four protein fractions were investigated in Tibetan wild barley. Albumin, globulin and hordein contents were under genetic control probably via multiple genes/quantitative trait loci. A correlation analysis showed that globulin was significantly associated with albumin, glutelin and hordein, while hordein was closely correlated with glutelin. Forty-nine diversity array technology (DArT) markers, which were distributed over seven chromosomes, were associated with the protein fraction contents. Those DArT markers associated with hordein were the same as those associated with globulin and glutelin. Only five markers associated with hordein, globulin and glutelin were also associated with albumin. Most of the protein fraction contents are therefore controlled by same genes which may contribute to total protein content. The discovery of new markers associated with specific protein fractions could be used to detect genes controlling protein content in the barley germplasm.  相似文献

Glutelin is a major seed storage protein, accounting for 60?C80?% of the total endosperm protein content in rice. To test whether we could augment the expression of an introduced recombinant protein in rice by suppressing the glutelin gene, we generated transgenic glutelin RNAi (glu RNAi) rice seeds. RNA gel blot analyses confirmed that the endogenous glutelin gene was severely suppressed in these transgenic rice lines. RT-PCR analysis further revealed that all the members of glutelin multigene family were downregulated. Transgenic glu RNAi rice seeds expressing a recombinant red fluorescent protein (RFP) showed stronger fluorescence than seeds transformed with the RFP gene only. Western blot analysis further revealed that the relative accumulation of RFP in glu RNAi seeds was twofold higher than that in the RFP-only transgenic seeds. These results suggest that RNAi targeting of an endogenous storage protein could be of great utility in obtaining higher transgene expression in genetically engineered rice and other plant lines.  相似文献

The feasibility of producing plant cell wall polysaccharide-hydrolysing feed enzymes in the endosperm of barley grain was investigated. The coding region of a modified xylanase gene (xynA) from the rumen fungus, Neocallimastix patriciarum, linked with an endosperm-specific promoter from cereal storage protein genes was introduced into barley by Agrobacterium-mediated transformation. Twenty-four independently transformed barley lines with the xylanase gene were produced and analysed. The fungal xylanase was produced in the developing endosperm under the control of either the rice glutelin B-1 (GluB-1) or barley B1 hordein (Hor2-4) promoter. The rice GluB-1 promoter provided an apparently higher expression level of recombinant proteins in barley grain than the barley Hor2-4 promoter in both transient and stable expression experiments. In particular, the mean value for the fungal xylanase activity driven by the GluB-1 promoter in the mature grains of transgenic barley was more than twice that with the Hor2-4 promoter. Expression of the xylanase transgene under these endosperm-specific promoters was not observed in the leaf, stem and root tissues. Accumulation of the fungal xylanase in the developing grains of transgenic barley followed the pattern of storage protein deposition. The xylanase was stably maintained in the grain during grain maturation and desiccation and post-harvest storage. These results indicate that the cereal grain expression system may provide an economic means for large scale production of feed enzymes in the future.  相似文献

C-hordein in barley and ω-gliadins in wheat are members of the prolamins protein families. Prolamins are the major component of cereal storage proteins and composed of non-essential amino acids (AA) such as proline and glutamine therefore have low nutritional value. Using double stranded RNAi silencing technology directed towards C-hordein we obtained transgenic barley lines with up to 94.7 % reduction in the levels of C-hordein protein relative to the parental line. The composition of the prolamin fraction of the barley parental line cv. Golden Promise was resolved using SDS-PAGE electrophoresis, the protein band were excised and the proteins identified by quadrupole-time-of-flight mass spectrometry. Subsequent SDS-PAGE separation and analysis of the prolamin fraction of the transgenic lines revealed a reduction in the amounts of C-hordeins and increases in the content of other hordein family members. Analysis of the AA composition of the transgenic lines showed that the level of essential amino acids increased with a concomitant reduction in proline and glutamine. Both the barley C-hordein and wheat ω-gliadin genes proved successful for RNAi-gene mediated suppression of barley C-hordein level. All transgenic lines that exhibited a reduction for C-hordein showed off-target effects: the lines exhibited increased level of B/γ-hordein while D-hordein level was reduced. Furthermore, the multicopy insertions correlated negatively with silencing.  相似文献

A chimeric gene encoding a precursor polypeptide of sesame 2S albumin, a sulfur-rich seed storage protein, was expressed in transgenic rice plants under the control of the glutelin promoter with the aim of improving the nutritive value of rice. Rice grains harvested from the first generation of ten different transformed lines inherited the transgene, and the accumulated sesame 2S albumin was presumably processed correctly as its mature form in sesame seed. This transgene was specifically expressed in maturing rice seeds with its encoded sesame 2S albumin exclusively accumulated in the seeds. The crude protein content in rice grains from five putative homozygous lines was increased by 0.64-3.54%, and the methionine and cysteine contents of these transgenic rice grains were respectively elevated by 29-76% and 31-75% compared with those of wild-type rice grains.  相似文献

Rice (Oryza sativa var. Nipponbare) was transformed with an artificial avidin gene. The features of this construct are as follows: (1) a signal peptide sequence derived from barley alpha amylase was added at the N-terminal region, (2) codon usage of the gene was optimized for rice, and (3) the gene was driven by rice glutelin GluB-1, an endosperm-specific promoter. Avidin was produced in the grain of the transgenic rice but not in the leaves. The concentration of avidin in the kernels was about 1,800 ppm. All larvae of the confused flour beetle (Tribolium confusum) and Angoumois grain moth (Sitotroga cerealella) died when fed transgenic avidin rice powder or kernels, respectively, whereas most of the test insects developed into adults when they were fed a nontransgenic rice control diet. Avidin extracted from the transgenic rice kernel lost most biotin-binding activity after 5 min heating at 95 degrees C.  相似文献
