In this study, de-juiced Italian ryegrass and timothy grass were successfully fractionated into one chlorophyll rich fraction, two lignin fractions, two hemicellulosic preparations, and one cellulosic fraction by sequential processes with 80% ethanol containing 0.2% NaOH, 2.5% H2O2–0.2% EDTA containing 1.5% NaOH, and 2.5% H2O2–0.2% TAED containing 1.0% NaOH at 75 °C for 3 h, respectively. The yields of chlorophyll rich fraction and total lignin fraction were found to be 6.1 and 11.0% from de-juiced Italian ryegrass, and 12.7 and 13.2% from de-juiced timothy grass. It should be noted that the two chlorophyll rich fractions contained noticeable amounts of contaminated hemicelluloses (17.1–18.8%) and lignin, whereas the four lignin fractions contained only small amounts of bound hemicelluloses (5.6–8.6%). Nitrobenzene oxidation and NMR spectra of the lignin fraction revealed that the presence of both guaiacyl and syringyl units, as well as p-hydroxyphenyl structures. Small amounts of esterified p-coumaric acids and mainly etherified ferulic acids were also identified in the lignin fractions. It was found that the lignin fractions obtained from the two different grasses had very similar structural composition. They are distinguished by rather low amounts of β-O-4 structures, resinol units (β-β), and of condensed units (β-5 and 5-5). This trait is even more pronounced in the production of phenolic compounds for chemical industry from grass.  相似文献

A water-soluble polysaccharide complex (A) composed of galactose (17.9%), 3-O-methyl-galactose (3.0%), glucose (15.5%), mannose (8.3%), arabinose (30.4%), xylose (7.6%), fucose (2.6%), rhamnose (6.7%), and uronic acids (8.0%) has been isolated from the aerial parts of sage (Salvia officinalis L.) by cold water extraction. It showed a broad molecular-mass distribution pattern (Mw approximately 2000-93,000) with a predominance of polymers with Mw< 10,000. Ion-exchange chromatography of A afforded six polymeric fractions (A1-A6) in which arabinogalactans associated with galacturonan and/or rhamnogalacturonan backbones prevail. Sage polysaccharides were examined for their immunomodulatory activity in the comitogenic thymocyte test which is interpreted as being an in vitro correlate of adjuvant activity. The acidic polysaccharide fractions A2, A3 and A4 exhibited the highest mitogenic and comitogenic activities of all fractions tested, and relatively high SI(comit)/SI(mit) ratios approximately 3 indicate potential adjuvant properties of these polysaccharides.  相似文献

Seven residual hemicellulosic preparations (19.6-45.0% of the original hemicelluloses) were extracted from wheat straw pre-treated with various organic solvents using 1.8% H2O2-0.18% cyanamide at 50 degrees C and pH 10.0 for 4 h. Their chemical compositions and physicochemical properties were determined using GC, HPLC, GPC, FT-IR and 13NMR spectroscopy. The results indicated that all the residual hemicellulosic preparations were heteropolysaccharides containing xylose, glucose, arabinose, galactose, mannose, rhamnose and 4-O-methyl-alpha-D-glucopyranosyluronic acid. The predominant monosaccharide was xylose, ranging between 67.7% and 81.9% of the total neutral sugars, composed mainly of L-arabino-(4-O-methyl-D-glucurono)-D-xylan. The content of contaminant lignin in the isolated residual hemicelluloses was 2.89-5.31%. The Mw values of the two residual hemicellulosic preparations H6 and H7 (42,710 and 44,080 g mol-1, respectively) obtained from the aqueous-alcohol pre-treated straw were much higher than those of H1-H5 (12,980-15,950 g mol-1) extracted from the organic acid pre-treated straw.  相似文献

Eight hemicellulosic fractions were obtained by sequential treatment of dewaxed barley straw with 0.1 M NaOH at 45 °C for 3 h, 0.25, 0.5, 1.0, 1.5, 2.0, and 3.0% H2O2 at 45 °C for 3 h at pH 11.5, and 10% KOH–1% Na2B4O7·10H2O at 28 °C for 15 h under continuous agitation. The yields of the fractions were 8.0, 3.1, 3.3, 3.3, 2.2, 2.0, 2.0, and 9.9%, respectively, of the initial amount of barley straw, corresponding to the dissolution of 21.6, 8.4, 8.9, 8.9, 5.9, 5.4, 5.4, and 26.7% of the original hemicelluloses. Meanwhile, the successive treatment also solubilized 29.1, 15.8, 14.6, 10.8, 4.5, 3.2, 2.7, and 3.7% of the original lignin, respectively. This sequential extraction together resulted in dissolution of 91.1% of the original hemicelluloses and 84.8% of the original lignin. The 0.1 M NaOH-soluble hemicellulosic fraction contained mainly xylose, glucose, and arabinose, 44.2, 15.7, and 15.2%, respectively, while the 10% KOH–1% Na2B4O7·10H2O-soluble fraction predominated in xylose, 75.0%. The six alkaline peroxide-soluble fractions were composed of 50.3–54.4% xylose, 14.7–16.9% arabinose, 6.8–10.7% glucose, 6.8–8.5% glucuronic acid or 4-O-methyl- -glucuronic acid, 0.4–1.5% mannose, and 0.3–1.2% rhamnose. All the hemicellulosic fractions contained substantial amounts of glucuronoarabinoxylans and noticeable quantities of β-glucans. In comparison, the six hemicellulosic fractions, isolated with alkaline peroxide, had much higher molecular weights (56,890–63,810 g mol−1) than those of the two hemicellulosic preparations (28,000–29,080 g mol−1), isolated with alkali in the absence of hydrogen peroxide. The thermal stability of the hemicelluloses increased with an increment of their molar mass.  相似文献

Two-stage treatments of sugarcane bagasse with mild alkali and acidic 1,4-dioxane were performed. Pretreatment with 1M NaOH aqueous solution at 20, 25, 30, 35, and 40 degrees C for 18 h released 55.5%, 57.3%, 59.1%, 60.9%, and 62.1% of the original hemicelluloses, respectively. Post-treatment of the corresponding alkali-treated residue with 1,4-dioxane-2M HCl (9:1, v/v) at 87 degrees C for 2h, respectively, degraded 11.6%, 11.9%, 11.4%, 10.9%, and 10.6% of hemicelluloses (% dry starting material). It was found that the five alkali-soluble hemicellulosic preparations contained a much higher amounts of xylose (78.0-82.2%) and slightly higher uronic acids (4.8-5.8%), mainly 4-O-methyl-alpha-d-glucopyranosyluronic acid, but were lower in arabinose (9.3-11.7%) and glucose (2.2-4.1%) than those of the corresponding five acidic dioxane-degraded hemicellulosic fractions in which xylose (44.9-46.8%), arabinose (35.9-38.1%), and glucose (13.0-13.7%) were the major sugar constituents. The studies revealed that the five alkali-soluble hemicellulosic preparations were more linear and acidic, and had a large molecular weight (35,200-37,430 g mol(-1)) than those of the hemicellulosic fractions (12,080-13,320 g mol(-1)) degraded during the acidic dioxane post-treatment. This demonstrated that the post-treatment with acidic dioxane under the condition used resulted in substantial degradation of the hemicellulosic polymers. The 10 hemicellulosic samples were further characterized by FT-IR and 1H and 13C NMR spectroscopy, GPC and thermal analysis, and the results are reported.  相似文献

Abaca fibre polysaccharides were fractionated into water soluble, pectic, 1% NaOH soluble, hemicellulosic and cellulose fractions by extraction with hot water, dilute hydrochloric acid (pH 1.6), aqueous 1% NaOH and 17.5% NaOH, respectively. Cellulose (60.4–63.6%) and hemicelluloses (20.8%) were the major polysaccharides in abaca fibres. The hot water soluble polysaccharides contained noticeable amounts of pectic substances and a large proportion of neutral polysaccharides. The pectic polysaccharide preparation was enriched in both galacturonic acid and neutral sugars, including xylose, glucose, galactose, arabinose, and rhamnose. Extraction of the fibre with aqueous 1% NaOH produced the hemicellulose–lignin complex, which was enriched in xylose and, to a lesser extent, glucose-, arabinose- and galactose-containing polysaccharides, together with 7.6% associated lignin. Further extraction of the delignified fibre residue with aqueous 17.5%. NaOH removed the hemicellulose fractions, which were strongly enriched in xylose-containing polysaccharides. Besides ferulic and p-coumaric acids, six other phenolic monomers were also detected in the mixtures of alkaline nitrobenzene oxidation of associated lignin in all the polysaccharide fractions. The content of bound lignin in water soluble, pectic, and 1% NaOH soluble polysaccharides (Fractions 1, 2, and 3), isolated directly from the lignified fibres, was 12 times that of the hemicellulosic preparations (Fractions 4 and 5) isolated from the delignified fibre residues.  相似文献

The following fractions were obtained from the wall material of Gliocladium viride : F1 (27.5%), a glucan, containing xylose, mannose and galactose, coluble in 1 M NaOH at 20°C; F2 (6.7%), a β-glucan-chitin complex, solubilized with 1 M NaOH at 20°C from the previous residue left overnight at −20°C; F3 (8.1%), a glucan, containing mannose and galactose solubilized with 1 M NaOH at 70°C; and F4, the insoluble residue, a β-glucan-chitin complex similar to F2, amounting to 31.3% of the wall material.
F1 was extracted with distilled water. The soluble material (F1S) was a galactomannoglucan (54.7%) and the inscluble (F1P) a glucan (45.3%). Periodate oxidation revealed the presence of glycerol, erythritol, threitol, ribitol, arabitol, mannose, galactose and glucose in F1S, and glycerol and glucose as the main components in F1P. The fractions obtained when F1S was purified through Sepharose CL6B, were methylated.  相似文献

A new high-viscous polysaccharide polymyxan from Bacillus polymyxa 88A is described. Polymyxan consists of an acid high-viscous polysaccharide (Mw 1-10 MD) and a neutral low-viscous polysaccharide (Mw 100-300 kD), which is a glucomannan containing equal amounts of monosaccharides and traces of uronic acids. The acid high-viscous polysaccharide consists of 36% glucose, 36% mannose, 7% galactose and 21% glucuronic acid. Data are presented on the application of polymyxan in baking industry and for preparation of drilling muds.  相似文献

One organic and three alkaline hemicellulosic fractions were isolated by an ultrasound-assisted extraction which partially delignified the fast-growing poplar wood. Successive treatments were conducted with dimethyl sulfoxide under ultrasonic irradiation at 570 W, 25 °C for 30 min, 70% ethanol containing 1% NaOH, 3% NaOH and 6% NaOH at 75 °C for 3 h, respectively. The four hemicellulosic fractions obtained were comparatively studied by sugar analysis, alkaline nitrobenzene oxidation of bound lignin, GPC, FT-IR, 1D and 2D NMR spectroscopy as well as TGA and DTA. The results showed that the ultrasonic treatment and sequential extractions with three different concentrations of NaOH led to a release of 75.5% of the original hemicelluloses and 96.2% of the lignin. All four purified hemicellulose fractions contained relatively low amounts of associated lignin, ranging between 0.96 and 3.10%. In addition, the hemicellulosic fraction H4 isolated with 6% NaOH is formed by a linear backbone of four (β-1 → 4)-xylopyranosyl residues and at least one of the xylose residues is monosubstituted at C-2 by a 4-O-methylglucuronic acid, giving a typical ratio of 4-O-methyl glucuronic acid to Xyl of 1 to 4.  相似文献

The present study was undertaken to investigate the extractability of the hemicelluloses from bagasse obtained by ultrasound-assisted extraction methods. The results showed that the ultrasonic treatment and sequential extractions with alkali and alkaline peroxide under the conditions given led to a release of over 90% of the original hemicelluloses and lignin. This fact as well as the sugar composition and structural features of the isolated seven hemicellulosic fractions indicated that ultrasonication attacked the integrity of cell walls, cleaved the ether linkages between lignin and hemicelluloses, and increased accessibility and extractability of the hemicelluloses. Increasing alkali concentration from 0.5 to 2M and alkaline peroxide percentage from 0.5% to 3.0% resulted in degradation of hemicellulosic backbone as shown by a decrease in their molecular weights from 43,580 to 14,470 and 30,180 to 18,130gmol(-1), respectively. However, there were no significant differences in the structural features of the seven sequential alkali- or alkaline peroxide-soluble hemicellulosic fractions, which are composed mainly of L-arabino-(4-O-methyl-D-glucurono)-D-xylans. Ferulic and p-coumaric acids were found to be chemically linked with hemicelluloses.  相似文献

Three organosolv and three alkaline hemicellulosic fractions were prepared from lignocellulosic biomass of the fast-growing shrub Tamarix austromongolica (Tamarix Linn.). Sugar analysis revealed that the organosolv-soluble fractions contained a higher content of glucose (33.7-6.5%) and arabinose (14.8-5.6%), and a lower content of xylose (62.2-54.8%) than the hemicellulosic fractions isolated with aqueous alkali solutions. A relatively high concentration of alkali resulted in a decreasing trend of the xylose/4-O-methyl-d-glucuronic acid ratio in the alkali-soluble fractions. The results of NMR analysis supported a major substituted structure based on a linear polymer of β-(1 → 4)-linked d-xylopyranosyl residues, having ramifications of α-l-arabinofuranose and 4-O-methyl-d-glucuronic acid residues monosubstituted at O-3 and O-2, respectively. Thermogravimetric analysis revealed that one step of major mass loss occurred between 200-400 °C, as hemicelluloses devolatilized with total volatile yield of about 55%. It was found that organosolv-soluble fractions are more highly ramified, and showed a higher thermal stability than the alkali-soluble fractions.  相似文献

The gum exudate from Combretum hartmannianum is water-soluble, forms very viscous solutions, and contains galactose (22%), arabinose (43%), mannose (10%), xylose (6%), rhamnose (4%), glucuronic acid (6%), 4-O-methylglucuronic acid (2%), and galacturonic acid (7%). The acidic components produced on hydrolysis of the gum were 6-O-(β-D-glucopyranosyluronic acid)-D-galactose, and two saccharides that had the same chromatographic mobility, and contained mannose and galacturonic acid, and galactose and 4-O-methylglucuronic acid, respectively. Methylation and methanolysis of the gum indicated the presence of terminal uronic acid, rhamnose, xylose, galactose, arabinofuranose, and arabinopyranose. Controlled, acid hydrolysis indicated the presence of (1→3)-linked arabinopyranose side-chains and (1→6)-linked galactose residues. C. hartmannianum gum, when subjected to two Smith-degradations, yielded Polysaccharides I and II, both of which contained galactose, arabinose, and mannose. Insufficient crude gum was available for a complete structural study, but the molecule was shown to contain long, sparsely branched chains of (1→6)-linked galactose residues, to which are attached (1→3)-linked arabinose and (1→3)-linked mannose side-chains.  相似文献

Cell wall material (CWM) isolated from beeswing wheat bran contains 66% carbohydrate, 12% Klason lignin, 6% protein and 4% ash. The relative proportions of sugars in the CWM are arabinose 34%, xylose 26%, galactose 2%, glucose 32% and uronic acid 6%. The uronic acid was shown to consist of glucuronic acid and its 4-O-Me analogue in the ratio 1.8:1. Partial acid hydrolysis of the CWM yielded neutral sugars and a uronic acid fraction. The latter was shown to contain Glc p A-(1→2)-Xyl p and Glc p A-(1→2)-O-Xyl p-(1→4)-Xyl p and their 4-O-Me substituted uronic acid analogues. Methylation analysis of the whole CWM and partially degraded methylated CWM revealed the nature of the constituent glycosidic linkages. From the combined evidence we infer that the major structural features of the non-cellulosic polysaccharides are a linear chain of xylopyranose units joined by (1→4)-linkages, and arabinofuranose, xylose, galactose (and uronic acid) end groups, which in at least some of the polysaccharides, are attached directly by (1→2)- and/or (1→3)-linkages to the xylan chain. The CWM has been fractionated by successive extractions with water at 80°, 0.2 M (NH4)2C2O4 at 80°, Na chlorite/HOAc at 70°, 0.2 M (NH4)2C2O4 at 80°, 1 M and 4 M KOH, and the neutral sugar composition of the fractions determined. It is concluded from these and other experiments that the CWM contains two main types of polysaccharides, the arabinoxylans and cellulosic polymers, and that phenolic ester linkages play a role in holding them together.  相似文献

Highly viscous extracellular proteoglycan (EPG) has been isolated from culture medium of the unicellular red alga Rhodella grisea (Rhodophyceae) by ethanol precipitation. EPG was composed of xylose (29.3%), 3-O-methyl-xylose (26.0%), uronic acids (17.1%), rhamnose (14.4%), galactose (7.5%), glucose (3.9%), arabinose (1.4%) and mannose (0.4%), and traces of fucose, 4-O-methyl-xylose and 2,3-di-O-methyl-rhamnose or fucose. In addition, the polymer contained proteins (13.1%), sulphates and 13C-CP MAS spectra indicated the presence of acetyl and succinyl groups. The molecular mass was estimated to be 136,000. Ion-exchange chromatography afforded five fractions differing in composition of neutral sugars, uronic acids, and protein content indicating thus the complex structure of the EPG.  相似文献

The influence of other hemicellulosic sugars (arabinose, galactose, mannose and glucose), oxygen limitation, and initial xylose concentration on the fermentation of xylose to xylitol was investigated using experimental design methodology. Oxygen limitation and initial xylose concentration had considerable influences on xylitol production by Canadida tropicalis ATCC 96745. Under semiaerobic conditions, the maximum xylitol yield was 0.62 g/g substrate, while under aerobic conditions, the maximum volumetric productivity was 0.90 g/l h. In the presence of glucose, xylose utilization was strongly repressed and sequential sugar utilization was observed. Ethanol produced from the glucose caused 50% reduction in xylitol yield when its concentration exceeded 30 g/l. When complex synthetic hemicellulosic sugars were fermented, glucose was initially consumed followed by a simultaneous uptake of the other sugars. The maximum xylitol yield (0.84 g/g) and volumetric productivity (0.49 g/l h) were obtained for substrates containing high arabinose and low glucose and mannose contents.  相似文献

A fucoidan, obtained from the hot-water extract of the brown seaweed, Hizikia fusiforme, was separated into five fractions by DEAE Sepharose CL-6B and Sepharose CL-6B column chromatography. All five fractions contained predominantly fucose, mannose and galactose and also contained sulfate groups and uronic acid. The fucoidans had MWs from 25 to 950 kDa. The structure of fraction F32 was investigated by desulfation, carboxyl-group reduction, partial hydrolysis, methylation analysis and NMR spectroscopy. The results showed that the sugar composition of F32 was mainly fucose, galactose, mannose, xylose and glucuronic acid; sulfate was 21.8%, and the MW was 92.7 kDa. The core of F32 was mainly composed of alternating units of -->2)-alpha-D-Man(1--> and -->4)-beta-D-GlcA(1-->, with a minor portion of -->4)-beta-D-Gal(1--> units. The branch points were at C-3 of -->2)-Man-(1-->, C-2 of -->4)-Gal-(1--> and C-2 of -->6)-Gal-(1-->. About two-thirds of the fucose units were at the nonreducing ends, and the remainder were (1-->4)-, (1-->3)- and (1-->2)-linked. About two-thirds of xylose units were at the nonreducing ends, and the remainder were (1-->4)-linked. Most of the mannose units were (1-->2)-linked, and two-thirds of them had a branch at C-3. Galactose was mainly (1-->6)-linked. The absolute configurations of the sugar residues were alpha-D-Manp, alpha-L-Fucp, alpha-D-Xylp, beta-D-Galp and beta-D-GlcpA. Sulfate groups in F32 were at C-6 of -->2,3)-Man-(1-->, C-4 and C-6 of -->2)-Man-(1-->, C-3 of -->6)-Gal-(1-->, C-2, C-3 or C-4 of fucose, while some fucose had two sulfate groups. There were no sulfate groups in either the GlcA or xylose residues.  相似文献

To improve 1,3-propanediol (1,3-PD) production by an economic and efficient approach, hemicellulosic hydrolysates (HH) used as cosubstrate resulted in more biomass and higher reducing power for 1,3-PD production. The effects of primary degradation products such as individual sugars (xylose, glucose, mannose, arabinose and galactose) and major inhibitors (furfural, acetate and formate) on the Klebsiella pneumoiae growth and 1,3-PD production were investigated in this study. Xylose and mannose could efficiently promote the 1,3-PD production and cell growth. Furfural (0.28 g/l) and sodium acetate (1.46 g/l) in low concentration were not inhibitory to Klebsiella pneumoniae, rather they have stimulatory effect on the growth and 1,3-PD biosynthesis, especially the acetate. In fed-batch fermentation with HH as cosubstrate, the final 1,3-PD production, conversion from glycerol and productivity were 71.58 g/l, 0.65 mol/mol and 1.93 g/l/h, respectively, which were 17.8%, 25.0% and 17.7% higher than that from glycerol alone.  相似文献

Prolonged growth of cell cultures of sycamore (Acer pseudoplatanus L.) on agar medium containing myo-inositol-2-(3)H resulted in incorporation of label predominately into uronosyl and pentosyl units of cell wall polysaccharides. Procedures normally used to distinguish between pectic substance and hemicellulose yielded carbohydrate-rich fractions with solubility characteristics ranging from pectic substance to hemicellulose yet the uronic acid and pentose composition of these fractions was decidedly pectic. Galacturonic acid was the only uronic acid present in each fraction. Subfractionation of alkali-soluble (hemicellulosic) polysaccharide by neutralization followed by ethanol precipitation gave 3 fractions, a water-insoluble, an ethanol-insoluble, and an ethanol-soluble fraction, each progressively poorer in galacturonic acid units and progressively richer in arabinose units; all relatively poor in xylose units.Apparently, processes involved in biosynthesis of primary cell wall continued to produce pectic substance during cell enlargement while processes leading to biosynthesis of typically secondary cell wall polysaccharide such as 4-0-methyl glucuronoxylan were not activated.  相似文献

Ripening of mango is characterized by a gradual, but natural softening of the fruit, which is due to progressive depolymerization of pectic and hemicellulosic polysaccharides with significant loss of galactose, arabinose and mannose residues at the ripe stage. Structural characterization employing permethylation followed by GC-MS analysis, IR and 13C NMR measurements revealed the major CWS fractions of both unripe and ripe mangoes to be of variable molecular weights and having a 1,4-linked galactan/galacturonan backbone, which is occasionally involved in side chain branches consisting of single residues of galactose and arabinose or oligomeric 1,5-linked arabinofuranose residues linked through 1,3-linkages; whereas the major hemicellulosic fractions of unripe mango to be of xyloglucan-type having 1,4-linked glucan backbone with branching by non-reducing terminal arabinose and xylose residues.  相似文献
