Antarctic fish, such as the Trematomus bernacchii, living at −1.9°C maintain a serum osmolality of around 600 mOsm kg−1, nearly twice that of temperate fish. Upon warm acclimation, Antarctic fish significantly lower their serum osmolality. It has been suggested that this response to warm acclimation is due to stress. The purpose of this study was to determine, whether upon warm acclimation there was a change in the levels of the stress hormone cortisol and hematocrit associated with the decrease in serum osmolality. T. bernacchii were warm acclimated up to 4 weeks and serum osmolality, cortisol and hematocrit were measured. Upon warm acclimation to +1.6 and +3.8°C over the course of 4 weeks, T. bernacchii significantly lowered their serum osmolality (from 547 ± 4 mOsm kg−1 to 494 ± 6 and 489 ± 4 mOsm kg−1, respectively), yet did not alter their serum cortisol (29 ± 6 nl ml−1) or hematocrit (22 ± 1%) levels. These results suggest that warm acclimation does not induce a stress response in T. bernacchii.  相似文献

In 2005 and 2006, adult sockeye salmon (Oncorhynchus nerka) were captured en route to spawning grounds and placed in either a slow (∼ 0.1 m·s−1) or fast (∼0.4 m·s−1) water velocity treatment for 18 days in order to assess how migrational energy depletion during the final stages of maturation affected physiological condition and survival. Fish in the fast treatment utilized more energy than the slow treatment in 2005 (0.91 MJ kg−1 vs. 0.43 MJ kg−1; P = 0.010), and 2006 (0.72 MJ kg−1 vs. 0.37 MJ kg−1; P = 0.021). Non-treatment fish captured upon arrival at spawning grounds showed energy levels intermediate to the two treatments in 2005 and lower than both in 2006, suggesting that energy use during the treatments were within levels normally experienced by this population. No differences in survival were found between treatments (P > 0.05), although females had lower survival than males in both years (both P < 0.01). After 18 days, surviving fish from the fast treatment showed signs of elevated physiological stress relative to fish from the slow treatment. Specifically, plasma osmolality was lower in fast fish in 2005 (P < 0.001), as was plasma chloride in both years (both P < 0.02). In 2006, plasma lactate was higher (P = 0.014) in fast fish. Within the ranges of energetic depletion that were examined here, a more energy-intensive migration can have a substantial influence on the physiological condition and stress of adult sockeye salmon, but not on survival.  相似文献

Drinking in Atlantic salmon (Salmo salar) juveniles was investigated in fresh water and following transfer to sea water. There was a significant effect of fish size on drinking, and smolts (20–30 g) imbibed about ten times less water than alevins of 0.2–0.3 g. Freshwater smolts drank at a rate of 0.15 ± 0.03 ml · kg−1 · h−1 and administration of doses of 10 or 20 mg · kg−1 of papaverine (stimulator of the renin- angiotensin system RAS) or [Asn1, Val5]-Angiotensin II (0.4 μmol · kg−1) resulted in significant increases in drinking, while administration of the angiotensin converting enzyme inhibitor, enalapril (50 mg · kg−1) had no effect on drinking. Transfer of Atlantic salmon smolts to 1/3, 2/3 and full strength sea water resulted in significant increases in drinking to 1.06 ± 0.12, 1.24 ± 0.0.16 and 3.89 ± 0.28 ml · kg−1 · h−1, respectively. In sea water, stimulation of the endogenous RAS by administration of papaverine (20 mg · kg−1) resulted in a 20% increase in drinking, while administration of enalapril to doses of 50 and 200 mg · kg−1 lowered drinking to 1.99 ± 0.48 and 0.32 ± 0.06 ml · kg−1 · h−1, respectively. All treatments were without effect on blood plasma levels of Na+ and Cl in fresh water, while in sea water smolts both stimulation and inhibition of drinking resulted in hemoconcentration of Na+ and Cl. The sperm allocation pattern of a copulating marine cottid fish, Alcichthys alcicornis, was investigated. A total of 86 mating events using six males were conducted in aquarium tanks over 10 days, and in 36 of them, spermatozoa were collected using a false copulation method. Males released 3–8 × 108 spermatozoa in early events, with the number decreasing gradually during subsequent mating events. This sperm allocation was represented as an “early investment and tapering” pattern. It was discussed why males have significantly higher sperm release in early spawning events. The reproductive behavior consists of spawning and subsequent copulation. Spermatozoa have the ability to fertilize eggs from multiple clutches, and in earlier produced clutches the level of sperm competition should be relatively low. In addition, if early spawn happens to be the first spawn with a female, spermatozoa that are released into the water column after spawning are responsible for fertilizing the female’s first clutch. The probability of this occurring should decrease dramatically as the season progresses, due to the highly synchronous seasonal spawning of females. All of these factors should select for high sperm numbers in early ejaculates. Based on such reproductive ecology of A. alcicornis we hypothesize that this sperm allocation pattern is an adaptive reproductive strategy in response to egg availability and sperm competition occurring within the ovarian cavity.  相似文献

The contributions of circulating angiotensin II (Ang II) and catecholamines to cardiovascular control in the spiny dogfish were investigated by monitoring the effects of exogenous and endogenous dogfish [Asn1, Pro3, Ile5]-Ang II (dfAng II) on plasma catecholamine levels and blood pressure regulation. Bolus intravenous injections of dfAng II (30–1200 pmol kg−1) elicited dose-dependent increases in plasma adrenaline and noradrenaline concentrations, caudal artery pressure (P CA), and systemic vascular resistance (R S), and a decrease in cardiac output (Q). Similar injections of Ang II in dogfish pre-treated with the α-adrenoceptor antagonist yohimbine (4 mg kg−1) also elicited dose-dependent increases in plasma catecholamine levels yet the cardiovascular effects were abolished. Dogfish treated with yohimbine were hypotensive and had elevated levels of plasma Ang II and catecholamines. Intravenous injection of the smooth muscle relaxant papaverine (10 mg kg−1) elicited a transient decrease in P CA and R S, and increases in plasma Ang II and catecholamine levels. In dogfish first treated with lisinopril (10−4 mol kg−1), an angiotensin converting enzyme inhibitor, papaverine treatment caused a more prolonged and greater decrease in P CA and R S, an attenuated increase in plasma catecholamines, and no change in plasma Ang II. By itself, lisinopril treatment had little effect on P CA, and no effect on R S, plasma Ang II or catecholamines. In yohimbine-treated dogfish, papaverine treatment elicited marked decreases in P CA, R S, and Q, and increases in plasma Ang II and catecholamines. Among the three papaverine treatments, there was a positive linear relationship between plasma Ang II and catecholamine concentrations, and the cardiovascular and hormonal changes were most pronounced in the yohimbine + papaverine treatment. Therefore, under resting normotensive conditions, while Ang II does not appear to be involved in cardiovascular control, catecholamines play an important role. However, during a hypotensive stress elicited by vascular smooth muscle relaxation, Ang II indirectly contributes to cardiovascular control by dose-dependently stimulating catecholamine release. We used a still-water swim channel in conjunction with open-flow oxygen and carbon dioxide respirometry to examine the energy requirements of river-otters (Lutra lutra L.) swimming voluntarily underwater in Neumünster Zoo (Germany). While at rest on land (5 °C), river-otters had a respiratory quotient of 0.77 and a resting metabolic rate of 4.1 W kg−1. This increased to an estimated 6.4 W kg−1 during rest in water (11–15 °C) and to 12.3 W kg−1 when the animals were feeding in the channel. River-otters swimming under water preferred a mean speed of 0.89 m s−1, and their energy requirements attained 11.6 W kg−1. We used tritium-labeled water to measure total body water, water influx (which approximated oxidative water production) and water efflux in free-flying tippler pigeons (Columba livia) during flights that lasted on average 4.2 h. At experimental air temperatures ranging from 18 to 27 °C, mean water efflux by evaporation and excretion [6.3 ± 1.3 (SD) ml · h−1, n = 14] exceeded water influx from oxidative water and inspired air (1.4 ± 0.7 ml · h−1, n = 14), and the birds dehydrated at 4.9 ± 0.9 ml · h−1. This was not significantly different from gravimetrically measured mass loss of 6.2 ± 2.1 g · h−1 (t = 1.902, n = 14, P>0.05). This flight-induced dehydration resulted in an increase in plasma osmolality of 4.3 ± 3.0 mosmol · kg−1 · h−1 during flights of 3–4 h. At 27 °C, the increase in plasma osmolality above pre-flight levels (ΔP osm = 7.6±4.29 mosmol · kg−1 · h−1, n = 6) was significantly higher than that at 18 °C (ΔP osm = 0.83±2.23 mosmol · kg−1 · h−1, (t = 3.43, n = 6, P < 0.05). Post-flight haematocrit values were on average 1.1% lower than pre-flight levels, suggesting plasma expansion. Water efflux values during free flight were within 9% of those in the one published field study (Gessaman et al. 1991), and within the range of values for net water loss determined from mass balance during wind tunnel experiments (Biesel and Nachtigall 1987). Our net water loss rates were substantially higher than those estimated by a simulation model (Carmi et al. 1992) suggesting some re-evaluation of the model assumptions is required. β-Endorphin (BE) infusion at rest can influence insulin and glucagon levels and thus may affect glucose availability during exercise. To clarify the effect of BE on levels of insulin, glucagon and glucose during exercise, 72 untrained male Sprague-Dawley rats were infused i.v. with either: (1) BE (bolus 0.05 mg · kg−1 +0.05 mg · kg−1 · h−1, n = 24); (2) naloxone (N, bolus 0.8 mg · kg−1 + 0.4 mg · kg−1, n = 24); or (3) volume-matched saline (S, n = 24). Six rats from each group were killed after 0, 60, 90 or 120 min of running at 22 m · min−1, at 0% gradient. BE infusion resulted in higher plasma glucose levels at 60 min [5.93 (0.32) mM] and 90 min [4.16 (0.29) mM] of exercise compared to S [4.62 (0.27) and 3.41 (0.26 mM] and N [4.97 (0.38) and 3.44 (0.25) mM]. Insulin levels decreased to a greater extent with BE [21.5 (0.9) and 18.3 (0.6) uIU · ml−1] at 60 and 90 min compared to S [24.5 (0.5) and 20.6 (0.6) uIU · ml−1] and N [24.5 (0.4) and 21.6 (0.7) uIU · ml−1] groups. Plasma C-peptide declined to a greater extent at 60 and 90 min of exercise with BE infusion compared to both S and N. BE infusion increased glucagon at all times during exercise compared to S and N. Nitrogen (N) and energy (E) requirements of the phyllostomid fruit bat, Artibeus jamaicensis, and the pteropodid fruit bat Rousettus aegyptiacus, were measured in adults that were fed on four experimental diets. Mean daily food intake by A. jamaicensis and R. aegyptiacus ranged from 1.1–1.6 times body mass and 0.8–1.0 times body mass, respectively. Dry matter digestibility and metabolizable E coefficient were high (81.1% and 82.4%, respectively) for A. jamaicensis and (77.5% and 78.0%, respectively) for R. aegyptiacus. Across the four diets, bats maintained constant body mass with mean metabolizable E intakes ranging from 1357.3 kJ · kg−0.75 · day−1 to 1767.3 kJ · kg−0.75 · day−1 for A. jamaicensis and 1282.6–1545.2 kJ · kg−0.75 · day−1 for R. aegyptiacus. Maintenance E costs were high, in the order of 3.6–5.4 times the basal metabolic rate (BMR). It is unlikely that the E intakes that we observed represent a true measure of maintenance E requirements. All evidence seems to indicate that fruit bats are E maximizers, ingesting more E than required and regulating storage by adjusting metabolic output. We suggest that true maintenance E requirements are substantially lower than what we observed. If it follows the eutherian norm of two times the BMR, fruit bats must necessarily over-ingest E on low-N fruit diet. Dietary E content did affect N metabolism of A. jamaicensis. On respective low- and high-E diets, metabolic fecal N were 0.492 mg N · g−1 and 0.756 mg N · g−1 dry matter intake and endogenous urinary N losses were 163.31 mg N · kg−0.75 · day−1 and 71.54 mg N · kg−0.75 · day−1. A. jamaicensis required 332.3 mg · kg−0.75 · day−1 and 885.3 mg · kg−0.75 · day−1 of total N on high- and low-E diets, respectively, and 213.7 mg · kg−0.75 · day−1 of truly digestible N to achieve N balance. True N digestibilities were low (29% and 49%) for low- and high-E diets, respectively. For R. aegyptiacus, metabolic fecal N and endogenous urinary N losses were 1.27 mg N · g−1 dry matter intake and 96.0 mg N · kg−0.75 · day−1, respectively, and bats required 529.8 mg · kg−0.75 · day−1 (total N) or 284.0 mg · kg−0.75 · day−1 (truly digestible N). True N digestibility was relatively low (50%). Based on direct comparison, we found no evidence that R. aegyptiacus exhibits a greater degree of specialization in digestive function and N retention than A. jamaicensis. When combined with results from previous studies, our results indicate that all fruit bats appear to be specialized in their ability to retain N when faced with low N diet. The aim of the present study was to measure zinc (Zn) and iron (Fe) concentration in human semen and superoxide dismutase (SOD) activity in seminal plasma and correlate the results with sperm quality. Semen samples were obtained from men (N = 168) undergoing routine infertility evaluation. The study design included two groups based on the ejaculate parameters. Group I (n = 39) consisted of males with normal ejaculate (normozoospermia), and group II (n = 129) consisted of males with pathological spermiogram. Seminal Zn and Fe were measured in 162 samples (group I, n = 38; group II, n = 124) and SOD activity in 149 samples (group I, n = 37; group II, n = 112). Correlations were found between SOD activity and Fe and Zn concentration, and between Fe and Zn concentration. SOD activity was negatively associated with volume of semen and positively associated with rapid progressive motility, nonprogressive motility, and concentration. Negative correlation was stated between Fe concentration and normal morphology. Mean SOD activity in seminal plasma of semen from men of group I was higher than in seminal plasma of semen from men of group II. Fe concentration was higher in teratozoospermic males than in males with normal morphology of spermatozoa in group II. Our results suggest that Fe may influence spermatozoa morphology.  相似文献

Springhares are large rodents that live in arid and semi-arid regions of Africa. We deprived springhares of water for periods of up to 7 days to determine what physiological adaptations, if any, enable them to survive in arid regions without drinking. During water deprivation, springhares lost up to 30% body weight and produced a mean maximum urine concentration of 2548 mosmol kg−1 with a maximum of 3076 mosmol kg−1 in an individual animal. Haematocrit and plasma sodium and potassium concentrations were well regulated throughout water deprivation at 47.5 ± 3.8% and 132.6 ± 7.4 mmol l−1 and 3.5 ± 0.7 mmol l−1, respectively, while plasma osmolality increased slightly from 293 ± 12.5 mosmol kg−1 to 324 ± 7.3 mosmol kg−1. Springhares thus appeared to be good osmoregulators and were able to maintain plasma volume during 7 days of water deprivation. In addition to the production of a relatively concentrated urine, water loss was limited by the lowered solute load and faecal water loss achieved by a reduction in food consumption and by the production of very dry faeces. We measured the levels of malondialdehyde (MDA), protein carbonyls, glutathione S-transferase (GST) and reducte glutathione (GSH) in seminal plasma and spermatozoa from 95 subjects including 50 infertile patients to evaluate the association between oxidative stress and damage and the components of the anti-oxidant defenses in seminal plasma and spermatozoa of infertile subjects and concentrations of cadmium (Cd) and lead (Pb) in the blood and seminal plasma because of tobacco smoke exposure. The reactive oxygen species (ROS) in spermatozoa were also evaluated by luminol (5-amino-2,3-dihydro-1,4-phthalazinedione)-enhanced chemiluminescence assay. The sperm count, motility, and morphology in the smokers infertile group were found to be lower than those in the fertile male group and nonsmokers infertile group (p < 0.001). Concentrations of Cd, Pb, MDA, protein carbonyls, and ROS levels in the smokers infertile group were significantly higher than those in the fertile male and nonsmokers infertile male groups (p < 0.001). However, GSH levels and GST activities were decreased in the smokers infertile male group than those in the fertile male and nonsmokers infertile male groups (p < 0.001). The results indicate that smoking could affect semen quality and oxidative lipid and protein damage in human spermatozoa. From Pearson correlation analysis, positive correlations were demonstrated between the seminal plasma Cd and seminal plasma protein carbonyls and between seminal plasma Pb and spermatozoa ROS levels in smokers of the subfertile group, while there was a significant positive correlation between blood Cd and ROS levels in smokers of the fertile group. There was also a significant negative correlation of the Cd level of the blood and GSH levels of the sperm and seminal plasma. These findings suggest that cigarette smoking enhances the levels of Cd and Pb in seminal plasma and blood and the extent of oxidative damage associated with a decrease in components of the anti-oxidant defenses in the sperm of infertile males.  相似文献

The relationship between body temperature (T b) and the plasma concentrations of arginine vasotocin (AVT) and angiotensin II (AII) was examined in conscious, adult Pekin ducks. Exposure of birds to an ambient temperature of 40 °C for 3 h increased T b by about 1.5 °C and increased breathing rate five-fold. Plasma osmolality was elevated from the normothermic value of 294.9 ± 1.4 mosmol kg−1 by about 8 mosmol kg−1 Circulating AVT levels increased by about 2 pg ml−1 from a basal concentration of 4.98 ± 0.15 pg ml−1, a rise which could be accounted for by the change in osmotic status. Plasma AII concentrations were unchanged from the pre-heat exposure value of 31.8 ± 3.4 pg ml−1. Time control birds, exposed only to an ambient temperature of 22 °C demonstrated no significant changes in any of the measured variables. The results suggest that an increased T b has no direct effect on the circulating concentrations of AVT or AII in ducks. Accepted: 2 June 1997  相似文献

Sperm motility and composition of the seminal fluid in Lota lota were investigated. Fives after motility initiation, 88.2 ± 12.4% of the spermatozoa were motile, their mean average path swimming velocity was 61.6 ± 16.3 μm s?1 and their principal swimming type the linear motion (77.4 ± 20.9%). In distilled water the rate of motile spermatozoa decreased to 0% in 40s. In 25–50 mosmol kg?1 electrolyte (NaCl) or non-electrolyte (glucose, sucrose) solutions, motility was prolonged for 10s and these solutions can therefore increase the efficiency of artificial fertilization when used for sperm motility activation. When semen was diluted in electrolyte or non-electrolyte solutions with osmolalities higher than 50 mosmol kg?1, sperm motility rates and swimming velocities decreased, and at osmolalities of 400 mosmol kg?1 motility was completely suppressed. In the seminal fluid with an osmolality of 290.08 ± 45.22 mosmol kg?1, sodium levels of 139.86 ± 23.79 mmol × 1?1, potassium levels of 11.59 ± 2.45 mmol × 1?1 and calcium levels of 0.20 ± 0.08 mmol × 1?1, sperm motility was inhibited. Under in vitro conditions, artificial saline solutions resembling the seminal plasma composition and 400 mosmol kg?1 NaCl or glucose solutions were useful as motility inhibiting solutions for predilution of semen. Sperm motility was not affected by pH 7.5–9.0, but at pH 6 the motility rate and the swimming velocity were reduced; seminal fluid pH was 8.47 ± 0.02. Therefore buffering of the artificial saline solutions can provide more stabile conditions for semen during storage and activation. Temperature optimum of semen was between 2 and 5°C. At higher temperatures semen became spontaneously motile. Therefore, controlled temperature conditions are an important factor for handling of semen. The qualitative, organical composition of seminal fluid was similar as in other fresh water teleosts.  相似文献

Myosin associated with the male germ cells of angiosperms interacts with actin, promoting transport of the non-motile generative and later sperm cells in the pollen tube. Myosin localizing on the sperm cell plasma membrane seems negligible in Plumbago, as reflected by the absence of: (i) anti-myosin labeling using immunoelectron microscopy, (ii) sperm motility on actin matrices, and (iii) electrophoretic movement changes after addition of antibody. Sperm cells injected directly into actively streaming Nitella internodal cells, however, follow actin bundles and their movement is sensitive to ATP and Mg2+. This may be based on simple charge binding since negatively charged latex beads also migrate on actin, whereas neutral or positively-charged latex beads do not. Sperm cells are negatively charged according to capillary microelectrophoresis, whereas killed sperm cells, which are positively charged do not migrate. The sperm cell that normally fertilizes the egg has a higher calculated charge (8.277 × 103 esu/cm2) compared with the sperm cell that fuses with the central cell (6.120 × 103 esu/cm2). The spermatozoa of Boops boops, Diplodus sargus, Mullus barbatus, and Trachurus mediterraneus were motile in sea water, and in electrolyte solutions (NaCl) and non-electrolyte solutions (glucose) with an osmolality of 600–1000 mosmol kg?1. Their mean motility rate 10 s after initiation was about 80%, while about 10% of the motile spermatozoa moved non-linearly, 45% linearly, and 45% circularly. The average path swimming velocity was significantly higher in M. barbatus (about 90 μm s?1) than in the other species (70 μm s?1). The number of motile spermatozoa decreased to 0% within 50 s after initiation of motility in T. mediterraneus, within 90 s in M. barbatus . In B. boops and D. sargus about 90% of the spermatozoa stopped movement during the first 90 s of the motility period, while the rest remained motile for 2–3 h. Motility of B. boops and D. sargus spermatozoa was reversibly suppressed in the seminal plasma, and in electrolyte and non-electrolyte solutions of 100–200 mosmol kg?1. The trigger for motility activation was hyperosmolality (700–1000 mosmol kg?1). Motility of M. barbatus and T. mediterraneus sperm was only partly suppressed in the seminal plasma since freshly collected semen contained about 25–50% locally motile spermatozoa. When sperm was activated immediately after collection with electrolyte and non-electrolyte solutions of 700–1000 mosmol kg?1 spermatozoa moved progressively. The motility of those spermatozoa which had not yet been motile after collection was completely and reversibly suppressed in M. barbatus at osmolalities of 1200 mosmol kg?1, and at osmolalities of 100–200 mosmol kg?1 in T. mediterraneus . Therefore two triggers were necessary for initiation of motility. The nature of the first trigger was uncertain, the second trigger was a switch to hypoosmolality in M. barbatus and to hyperosmolality in T. mediterraneus . The sperm organisation of B. boops, D. sargus, M. barbatus and T. mediterraneus revealed species-specific parameters which could not be related with the sperm motility behaviour.  相似文献

 The spawning grounds of a cottid fish, Trachidermus fasciatus, were investigated off the mouth of the Kashima River in Ariake Bay from January to March, 1996–1999. The nests were found about 1.1–2.3 km off the mouth of the river, where the bottom salinity was 8‰–21‰ at low tides. Oyster shells were mainly found to be used as nests. A male, a pair, a male and egg clusters, a pair and egg clusters, or a female were found inside shells. The average number of guarded egg clusters per male in shells was 1.3 ± 0.4. This study compared the effects of supplementing the normal diets of six trained cyclists [maximal oxygen uptake O2max) 4.5 (0.36)l · min−1; values are mean (SD)] with additional carbohydrate (CHO) on muscle glycogen utilisation during a 1-h cycle time-trial (TT). Using a randomised crossover design, subjects consumed either their normal diet (NORM) for 3 days, which consisted of 426 (137) g · day−1 CHO [5.9 (1.4) g · kg−1 body mass (BM)], or additional CHO (SUPP) to increase their intake to 661 (76) g · day−1 [9.3 (0.7) g · kg−1 BM]. The SUPP diet elevated muscle glycogen content from 459 (83) to 565 (62) mmol · kg−1 dry weight (d.w.) (P < 0.05). However, despite the increased pre-exercise muscle glycogen stores, there was no difference in the distance cycled during the TT [40.41 (1.44) vs 40.18 (1.76) km for NORM and SUPP, respectively]. With NORM, muscle glycogen declined from 459 (83) to 175 (64) mmol · kg−1 d.w., whereas with SUPP the corresponding values were 565 (62) and 292 (113) mmol · kg−1 d.w. Accordingly, both muscle glycogen utilisation [277 (64) vs 273 (114) mmol · kg−1 d.w.] and total CHO oxidation [169 (20) vs 165 (30) g · h−1 for NORM and SUPP, respectively] were similar. Neither were there any differences in plasma glucose or lactate concentrations during the two experimental trials. Plasma glucose concentration averaged 5.5 (0.5) and 5.6 (0.6) mmol · l−1, while plasma lactate concentration averaged 4.4 (1.9) and 4.4 (2.3) mmol · l−1 for NORM and SUPP, respectively. The results of this study show that when well-trained subjects increase the CHO content of their diet for 3 days from 6 to 9 g · kg−1 BM there is only a modest increase in muscle glycogen content. Since supplementary CHO did not improve TT performance, we conclude that additional CHO provides no benefit to performance for athletes who compete in intense, continuous events lasting 1 h. Furthermore, the substantial muscle CHO reserves observed at the termination of exercise indicate that whole-muscle glycogen depletion does not determine fatigue at this exercise intensity and duration. The influx of glucose into the brain and plasma glucose disappearance were estimated in rainbow trout (Oncorhynchus mykiss) intravenously injected (1 ml · kg−1 body weight) with a single dose (15 μCi · kg−1 body weight) of 3-O-methyl-D-[U-14C]glucose ([U-14C]-3-OMG) at different times (2–160 min), and after intravenous injection at 15 min of increased doses (10–60 μCi · kg−1 body weight) of [U-14C]-3-OMG. Brain and plasma radiotracer concentrations were measured, and several kinetic parameters were calculated. The apparent brain glucose influx showed a maximum after 15–20 min of injection then decreased to a plateau after 80 min. Brain distribution space of 3-OMG increased from 2 min to 20 min reaching equilibrium from that time onwards at a value of 0.14 ml · g−1. The unidirectional clearance of glucose from blood to brain (k1) and the fractional clearance of glucose from brain to blood (k2) were estimated to be 0.093 ml · min−1 · g−1, and 0.867 min−1, respectively. A linear increase was observed in brain and plasma radiotracer concentrations when increased doses of [U-14C]-3-OMG were used. All these findings support a facilitative transport of glucose through the blood-brain barrier of rainbow trout with characteristics similar to those observed in mammals. The injection of different doses of melatonin (0.25–1.0 mg · kg−1) significantly increased brain glucose influx suggesting a possible role for melatonin in the regulation of glucose transport into the brain. This study examined the thermoregulatory responses of men (group M) and women (group F) to uncompensable heat stress. In total, 13 M [mean (SD) age 31.8 (4.7) years, mass 82.7 (12.5) kg, height␣1.79␣(0.06) m, surface area to mass ratio 2.46␣(0.18) m2 · kg−1 · 10−2, Dubois surface area 2.01 (0.16) m2, %body fatness 14.6 (3.9)%, O2peak 49.0 (4.8) ml · kg−1 · min−1] and 17 F [23.2 (4.2) years, 62.4 (7.7) kg, 1.65 (0.07) m, 2.71 (0.14) m2 · kg−1 · 10−2, 1.68 (0.13) m2, 20.2 (4.8)%, 43.2 (6.6) ml · kg−1 · min−1, respectively] performed light intermittent exercise (repeated intervals of 15 min of walking at 4.0 km · h−1 followed by 15 min of seated rest) in the heat (40°C, 30% relative humidity) while wearing nuclear, biological, and chemical protective clothing (0.29 m2 ·°C · W−1 or 1.88 clo, Woodcock vapour permeability coefficient 0.33 i m). Group F consisted of eight non-users and nine users of oral contraceptives tested during the early follicular phase of their menstrual cycle. Heart rates were higher for F throughout the session reaching 166.7 (15.9) beats · min−1 at 105 min (n = 13) compared with 145.1 (14.4) beats · min−1 for M. Sweat rates and evaporation rates from the clothing were lower and average skin temperature () was higher for F. The increase in rectal temperature (T re) was significantly faster for the F, increasing 1.52 (0.29)°C after 105 min compared with an increase of 1.37 (0.29)°C for M. Tolerance times were significantly longer for M [142.9 (24.5) min] than for F [119.3 (17.3) min]. Partitional calorimetric estimates of heat storage (S) revealed that although the rate of S was similar between genders [42.1 (6.6) and 46.1 (9.7) W · m−2 for F and M, respectively], S expressed per unit of total mass was significantly lower for F [7.76 (1.44) kJ · kg−1] compared with M [9.45 (1.26) kJ · kg−1]. When subjects were matched for body fatness (n = 8 F and 8 M), tolerance times [124.5 (14.7) and 140.3 (27.4) min for F and M, respectively] and S [8.67 (1.44) and 9.39 (1.05) kJ · kg−1 for F and M, respectively] were not different between the genders. It was concluded that females are at a thermoregulatory disadvantage compared with males when wearing protective clothing and exercising in a hot environment. This disadvantage can be attributed to the lower specific heat of adipose versus non-adipose tissue and a higher percentage body fatness. Accepted: 31 October 1997  相似文献
