The isolation and identification of rhubarb viruses occurring in Britain   总被引:1,自引:0,他引:1  
Virus-like symptoms were common in British crops of rhubarb. All plants tested of the three main varieties, ‘Timperley Early’, ‘Prince Albert’ and ‘Victoria’, were virus-infected. Turnip mosaic virus and a severe isolate of arabis mosaic virus (AMV) were obtained from ‘Timperley Early’; and ‘Prince Albert’ contained turnip mosaic virus, cherry leaf roll virus (CLRV), a mild isolate of AMV and, infrequently, cucumber mosaic virus (CMV). The main commercial variety ‘Victoria’ contained turnip mosaic virus, CLRV, a mild isolate of AMV and, infrequently, strawberry latent ringspot virus (SLRV). All the viruses were identified serologically. The rhubarb isolates did not differ markedly from other isolates of these viruses in herbaceous host reactions, properties in vitro or particle size and shape. A rhubarb isolate of CLRV was distinguished serologically from a cherry isolate of the virus. Turnip mosaic virus, CLRV and SLRV, were transmitted with difficulty, but AMV isolates were readily transmitted by mechanical inoculation. Turnip mosaic virus was also transmitted to rhubarb by Myzus persicae and Aphis fabae. CLRV was transmitted in 6–8% of the seed of infected ‘Prince Albert’ and ‘Victoria’ rhubarb and in 72% of the seed of infected Chenopodium amaranticolor. Mild isolates of AMV were also transmitted in 10–24% of the seed of infected ‘Prince Albert’ and ‘Victoria’ plants.  相似文献

Illus. 1961.—Chromosome numbers are given for the trailing blackberry varieties, ‘Young’ (2n = 49), ‘Boysen’ (2n = 49), ‘Nectar’ (2n = 49) and for related forms which include the parents of ‘Young,’ ‘Phenomenal’ (2n = 42) and ‘Mayes’ (2n = 56), and 3 cytologically resynthesized ‘Young’ plants (2n = 49) as a basis for interpreting the postulated origin of ‘Young.’ Cytological evidence substantiated the conclusion that ‘Young’ is a hybrid between ‘Phenomenal’ and ‘Mayes.’ Contributions to the understanding of genomic relationships in Rubus are offered from detailed analyses of meiosis in ‘Phenomenal,’ ‘Mayes,’ ‘Young,’ and ‘Boysen.’ ‘Phenomenal’ and ‘Mayes’ both had a very regular meiosis. ‘Young,’ as well as ‘Boysen,’ showed a greater degree of chromosome association than either parent of ‘Young.’ Meiotic behavior in ‘Boysen’ presented a close parallel to that of ‘Young’ which, correlated with morphological similarities and the same 2n chromosome number, suggests a similar origin. The mode of reproduction in ‘Young’ and ‘Boysen’ was found to be sexual on the basis of morphological variation in the open-pollinated (selfed) progeny, the varying aneuploid somatic chromosome numbers in these progeny (2n = 32–54) and aneuploid chromosome numbers in hybrids having either variety as one parent. The productiveness of ‘Young’ and ‘Boysen’ in commercial plantings and their successful utilization in breeding programs indicate a high fertility regardless of their having an odd multiple of the basic number. It is concluded that the production of balanced euploid gametes is not necessarily a criterion of fertility, at least not at this high level of ploidy.  相似文献

The effects of four rice varieties highly susceptible ( Xiang-Zao-Xian31 and Jia-Yu 164 ), moderately susceptible ( Wei-You402 ), and resistant ( Zhu-Liang-You02 ) to the whitebacked planthopper Sogatellafurcifera on the nymphal development, reproduc- tion and survival of Microvelia horoathi were investigated through a food chain using SogateUafurcifera as herbivore prey. Significant longer nymphal developmental time were observed in the 1 st, 4th, 5th instar and whole nymphal stage of M. horvathi raised on S. furcifera fed ZLY02 and WY402 than those of the other two varieties. M. horvathi raised on S. furcifera fed resistant ZLY02 showed a significant lower survival of whole nymphal stage than those fed other three susceptible varieties. Moreover, M. horvathi raised on S. furcifera fed resistant ZLY02 had shorter female longevity and lower lifetime fecundity than those of on susceptible XZX3l. Our results suggest that the S. furcifera- resistant rice variety ZLY02 had a negative effect on the growth and development of the predator M. horvathi through a food chain.  相似文献

Abstract

Fusarium moniliforme Sheld. var. subglutinans Wr. et Rg. causes malformations in young inflorescences of mango and seriously affects fruit formation. Quantitative studies of total, soluble and protein nitrogen contents of malformed inflorescence of two cultivars, ‘Himsagar’ and ‘Bombai’ were made. Healthy inflorescences of the former cultivar showed slightly higher nitrogen content than that of the latter. After infection, considerable increases in both soluble and protein fractions of nitrogen were observed in both host varieties. The proportion of these two nitrogen fractions, however, showed some changes in ‘Bombai’ but in ‘Himsagar’ it remained more or less unaltered.  相似文献

Enzymic browning of tuber tissue was evaluated quantitatively for 50 genotypes, representing seven accessions, of Solanum hjertingii, a wild potato species from north-east Mexico, together with five commercial varieties of Solanum tuberosum with a known range of enzymic browning. Ninety-four percent of S. hjertingii clones examined showed less browning than the commercial varieties, with 66% exhibiting half the ‘potential browning’ (a measure of the total enzymic browning resulting from disruption of cellular compartmentalisation in that system) of cv. Maris Piper, a low enzymic browning cultivar. Of these clones, 18 showed no visual discoloration when sliced. Results clearly indicate that the lack of enzymic browning in S. hjertingii is a ‘true’ character. The efficiency of one subjective and two objective methods for obtaining different measures of enzymic browning was assessed. Potential browning of a rehydrated freeze-dried powder was adopted as the most efficient technique for screening enzymic browning.  相似文献

Roots of two Chinese cabbage (Brassica campestris L. ssp. pekinensis) varieties, one tolerant and one susceptible, were inoculated with Plasmodiophora brassicae in liquid medium and in soil. Chitinase and peroxidase activities were determined in roots and shoots 1–21 days after inoculation with resting spores of Plasmodiophora and the enzyme activities compared with healthy tissue of the same age. In infected roots of the susceptible variety ‘Granat’ chitinase activity was higher than in the control 10 days after inoculation with spores. In the tolerant variety ‘Parkin’ we detected an increase in chitinase activity at the same time, which was about twice that of ‘Granat’. Chitinase activity in ‘Granat’ was also enhanced on day 13, 14 and 17 after inoculation, whereas chitinase activity in ‘Parkin’ was lower in the infected roots than in the controls during that period. In the shoots no correlation between chitinase activity and infection in the two varieties was observed. Chitinase from Chinese cabbage was further characterized and showed a pH optimum at pH 4.5–5.5 and a temperature optimum at 35–45°C. After isoelectric focusing 7 isoenzymes were discovered, but there were almost no differences between infected and healthy root extracts. Two isoenzymes with pI 8.7 and 8.8 showed cross-reactivity with an antiserum against bean chitinases. The molecular mass of these isoenzymes was determined as 33 kDa. Total peroxidase activity was generally higher in root tissue of both varieties than in the shoots. Peroxidase activity was increased most prominently in infected ‘Granat’ roots on day 13 after inoculation and of both varieties on day 17 compared to the controls. In clubbed tissue of ‘Granat’ a specific peroxidase isoenzyme appeared the first time 21 days after inoculation and was most prominent 28–30 days after inoculation. This isoenzyme had a molecular mass of ca 24 kDa and a pI of ca 8.8. With respect to our results the strategy of the Plasmodiophorales for plant attack is discussed.  相似文献

The effect of arbuscular mycorrhizal fungi (AMF) on olive (Olea europaea) growth and development was followed for 4 years after transplanting in irrigated commercial orchards under arid conditions. Sites I and II were irrigated with saline water (EC?=?4.5 dS/m). In site I, the soil was infested with Verticillium dahliae and olive varieties ‘Picual’ (Verticillium susceptible) and ‘Barnea’ (relatively Verticillium tolerant) were tested. In site II, the soil was virgin soil (previously non-cultivated soil) and olive varieties ‘Souri’ and ‘Barnea’ were tested. Plants for all sites were inoculated in the nursery with Glomus intraradices alone or in a mixture with G. mosseae. Relative to non-inoculated trees, AMF colonization enhanced vegetative growth, expressed as tree height and trunk circumference, at all sites. At first commercial harvest, AMF-treated trees had higher fruit and oil yields than non-mycorrhitic controls. Under saline water irrigation, differences between inoculated and non-inoculated treatments were reduced in the slow-growing ‘Souri’ but remained apparent in the modern fast-growing ‘Barnea’. AMF colonization did not appear to improve tolerance of either ‘Picual’ or ‘Barnea’ to V. dahliae, and both were more susceptible than the non-inoculated controls. Thus inoculation of olive plants with AMF improves transplant growth and adaptation in arid areas during the first 3 years of growth and until the first commercial harvesting season.  相似文献

Summary

Restoration of scrub and woodland in deforested upland sites is an important conservation activity. However, little is known about the mycorrhizal colonisation potential of upland soils or the factors that influence the distribution of mycorrhizal inoculum. We investigated the effect of existing vegetation on mycorrhizal colonisation potential for a sub-arctic willow (Salix lapponum) by planting uninoculated cuttings into plotsrepresenting two upland habitats with either grassand herbs (‘grass’) or Vaccinium myrtillus (‘vaccinium’) and assessing mycorrhizal colonisation after 14 months using morphological and molecular techniques. From 40 willow cuttings (20 in each habitat), DNA sequences of rive ectomycorrhizal (EcM) fungal taxa were recovered: Laccaria proxima, Thelephora terrestris, Hebeloma sp., ‘Thelephoraceae sp.’ and ‘Pezizales sp.’. Cuttings in the ‘grass’ habitat were dominated by Laccaria proxima and ‘Pezizales sp.’ and in the ‘vaccinium’ habitat by Thelephora terrestris which was absent from the ‘grass’ habitat. There were no significant differences between habitats in frequency of EcM inoculum (overall percentage of cuttings colonised = 70%) or colonisation potential (overall mean percentage of root tips colonised per cutting = 20 %). These data suggest that the mycorrhizal colonisation potential and diversity of fungi available to willow in these upland soils are low and planted willow may benefit from inoculum enhancement.  相似文献

Abstract

The palatability to common carp, Cyprinus carpio L. of three newly developed differently flavoured floating pellets made from a high proportion (40%) of brewer's spent grain (BSG) was tested using a multiple-offer feeding experiment. The addition of ‘bold’ flavours, such as vanilla or strawberry essence, may help mask the unpleasant taste of some piscicides; however, their inclusion must not compromise uptake by carp. There were no significant differences between the consumption rates of the three varieties, and all flavours were readily consumed. Therefore, it is suggested that highly flavoured pellets made with BSG have a strong potential to mask the flavour of an unpalatable toxin, and further research is now needed to test this hypothesis.  相似文献

Abstract

Cyprids are the final planktonic stage in the larval dispersal of barnacles and are responsible for surface exploration and attachment to appropriate substrata. The nanomechanical properties of barnacle (Balanus amphitrite) cyprid permanent cement were studied in situ using atomic force microscopy (AFM). Force curves were recorded from the cement disc continually over the course of its curing and these were subsequently analysed using custom software. Results showed a narrowing of the pull-off force distribution with time, as well as a reduction in molecular stretch length over time. In addition, there was a strong correlation between maximum pull-off force and molecular stretch length for the cement, suggesting ‘curing’ of the adhesive; some force curves also contained a ‘fingerprint’ of modular protein unfolding. This study provides the first direct experimental evidence in support of a putative ‘tanning’ mechanism in barnacle cyprid cement.  相似文献

Black‐coloured eggplants (Solanum melongena) represent the commercially most important group of eggplants in Europe and North America. Most of the modern varieties of black eggplants correspond to F1 hybrids, which at the same time constitute an elite gene pool for the development of new varieties. However, there are many black landraces and old varieties, which could be useful as sources of variation for black eggplant breeding programmes as well as for the broadening of the genetic diversity of the breeders’ gene pool. We have studied the morphological and molecular [amplified fragment length polymorphism and simple sequence repeat (SSR)] diversity in a collection of 38 black eggplant accessions, including commercial (modern F1 hybrid and old nonhybrid) varieties and landraces as well as in six nonblack control eggplants, from different origins. The results show that black eggplants contain a considerable morphological and molecular diversity, but commercial varieties, and in particular F1 hybrids, display a reduced morphological and molecular diversity when compared with landraces. The principal components analysis morphological and principal coordinates analysis molecular analyses show that commercial F1 hybrids group together, indicating that they share a common and narrow gene pool. Commercial F1 hybrids present a series of productive advantages, like early production, intense black colour (low L*, a* and b*) values and absence of fruit calyx prickles. However, several of the landraces and old nonhybrid varieties studied present a high yield as well as other traits of interest for eggplant breeding. Furthermore, given the low genetic diversity of F1 hybrids and the moderate level of SSR heterozygosity found in these materials (0.382), introduction of black landraces and old varieties in the present breeding programmes could contribute to broadening the gene pool used by breeders and this could help increase the heterosis for yield of F1 hybrids, which is greatly favoured by high heterozygosity levels.  相似文献

Mechanisms of host plant resistance against insect pests can be manifold. Resistance screenings generally use single target insect pests, but the resistance thus screened may not always be specific to the target insect species. We conducted a test for non‐specific resistance in indica rice varieties with resistance genes against brown planthopper (BPH), by using the Indian meal moth, Plodia interpunctella. The test system was very simple, and only required the non‐pest moth to be reared on rice flour. We compared the survival rate, developmental period and adult weight of the moth on three rice varieties: ‘Nipponbare’, a BPH‐susceptible japonica variety, and ‘Thai Collection 11’ and ‘Pokkali’, two resistant indica varieties. Our results were straightforward and demonstrate that resistance in the two resistant rice varieties is not BPH specific, because development of the moth was retarded and adult body weight was reduced.  相似文献

In controlled laboratory tests, using shallow glass chambers containing a coarse river sand substrate between ‘blank’ and ‘test’ sample cages, 3rd‐instar larvae of Costelytra zealandica (White) were attracted to parsnip and carrot root, freshly ground nutmeg, and ß‐pinene (a constituent common to these materials). The terpenol a‐fenchol was strongly attractive to the larvae. Ryegrass root, α‐pinene, and limonene were not significantly attractive, and limonene may indeed have some repellent effect.  相似文献

Phacidiopycnis washingtonensis was identified by morphology and ITS sequence analysis as the cause of rubbery rot, a new storage disease of apples in northern Germany. Infected fruits had an unusually firm texture and pale appearance after storage in ultra‐low oxygen conditions, but turned dark brown to black in ambient atmosphere. Ultimately, the surface of rotted fruits became covered by black pycnidia producing cream‐coloured conidial exudates. Rubbery rot affected several apple varieties, including the commercially important ‘Jonagold’ and ‘Elstar’. Losses during storage were commonly below 1% but reached 5–10% in a few cases. Fruits of ‘Golden Hornet’ crab apple trees planted as pollinators in commercial orchards became heavily infected by P. washingtonensis in October. Conidia were released throughout the following season from infected fruit mummies, which remained attached to the crab apple tree.  相似文献

Six cabbage (Brassica oleracea var. capitata) varieties with different levels of resistance to Mamestra brassicae (Lepidoptera: Noctuidae) were investigated in order to assess whether antibiosis and antixenosis mechanisms are involved in the resistance to this pest or not. Four experiments were conducted to determine the effect of variety and plant ontogeny on larval behaviour, adult oviposition and leaf damages in non‐choice and choice tests. Larval survival, time to development and larval weights differed depending on the varieties and plant stages that we tested. At the pre‐head stage, larval mortality was higher, larvae died faster, time to pupation was shorter, pupae were lighter and the percentage of viable pupae and growth index (GI) values were lower than larvae reared from plants at the head stage. The commercial hybrid ‘Corazón de buey’ and the local variety named ‘BRS0535’ exhibited antibiosis to M. brassicae as they reduced its survival and growth and delayed its development time. In addition, these varieties were the most resistant after artificial infestation in terms of head foliage consumption and number of larvae per plant. Oviposition tests demonstrated that resistance found in ‘Corazón de buey’ and BRS0535 could be also based on antixenosis mechanisms as they resulted in fewer egg batches on plants, whereas BRS0402 could be classified as resistant because M. brassicae larvae showed less preference for it. Thus, resistance to M. brassicae found in cabbage crops may be due to the joint action of several factors involving antibiosis and antixenosis. We found significant differences in the resistance of BRS0535 depending on the plant ontogeny as it loses its resistance while developing. Further studies are required to identify the mechanism of antibiotic resistance which is present in this variety at the pre‐head stage and the changes that occur in plant defence as it grows.  相似文献

Ditylenchus destructor and Ditylenchus dipsaci are economically important plant‐parasitic nematodes, affecting potato production mostly in temperate climates. Management through crop rotation is not feasible because of their wide host range. These nematodes are listed as quarantine pests in many countries. Limited information exists on the resistance and tolerance of currently cultivated potatoes to D. destructor and D. dipsaci. Two greenhouse experiments were conducted to screen 25 potato varieties for resistance to and tolerance for D. destructor and D. dipsaci infections. Reproduction factor (RF) and relative susceptibility (RS) were used to evaluate resistance, while potato tuber damage and tuber weight reduction was used to evaluate tolerance. Based on the RF, 16 varieties were evaluated as susceptible (S) while 5 varieties were evaluated as resistant (R) to D. destructor; varieties ‘Innovator’, ‘Aveka’ and ‘Spunta’ were resistant to D. dipsaci based on RF. ‘Désirée’ was observed to be highly susceptible to D. destructor and D. dipsaci in both experiments and was used as the standard susceptible control variety for the calculation of RS. A scale of 1–9 was used to classify RS of the potato varieties to D. destructor and D. dipsaci, where 9 indicated the highest level of resistance. All classes of resistance to D. destructor and D. dipsaci were observed in the potato varieties tested in the experiments. Six varieties had significantly lower RS to D. dipsaci than the standard susceptible control variety. Tolerant to highly sensitive potato varieties to both nematodes were also observed. RS and external potato tuber damage were identified as suitable methods for resistance and tolerance determination, respectively. This study provides essential information on the status of resistance and tolerance in potato varieties against D. destructor and D. dipsaci but needs to be confirmed under field conditions.  相似文献

Kiwifruit is a common and popular fruit around the world. However, white peach scale (Pseudaulacaspis pentagona [Targioni-Tozzetti]), a scale insect with a wide range of hosts, seriously affects the yield and quality of kiwifruit. To investigate the differences in resistance of different kiwifruit cultivars to P. pentagona, cellular structure and gene expression assays were used to explain the mechanism. Based on the stability of the infestation rate, we selected four cultivars from 50 kiwifruits for in-depth study, including ‘LC-04285’, ‘CF-3’, ‘DA-7B’ and ‘Hayward’. The anatomical structure of the canes analysis revealed that the resistant cultivar ‘LC-04285’ had thicker cuticle, denser epidermis and cortex. The expression levels of genes related to cuticle synthesis and formation of epidermis and cortex were also higher in ‘LC-04285’. Further investigation revealed that Jasmonic acid (JA) receptors COI1 were highly expressed in ‘LC-04285’. However, the expression levels of AcJAZs were higher in the susceptible cultivar ‘Hayward’. Besides, AcICS, AcPAL4, AcPAL5 and AcNPRs, which were involved in salicylic acid (SA) synthesis and SA response, were also highly expressed in ‘LC-04285’. Our results preliminarily revealed the mechanism of kiwifruit resistance to P. pentagona at the molecular and cellular levels. This study provided useful guidance for breeding insect-resistant kiwifruit in the future.  相似文献

Capsule Based on the 1999 Witherby Memorial Lecture – reviews how developments in molecular and population genetics have led to a reappraisal of species limits in birds.

The taxonomy of birds of the West Palearctic has moved from the comparative stability of the ‘Voous List’ into a period of serious activity, with new data emerging in almost every issue of every evolutionary and avian journal! This activity comes from two directions. Firstly, developments in population genetics, molecular biology, acoustics, behaviour and distributional studies have opened new avenues to measuring differentiation among groups of birds. This, in turn, has led to the recognition that earlier views of what constitutes a ‘species’ are in need of modification (‘improvement’), and the emergence of the ‘lineage concept’ of species. I review some of the species concepts most relevant to avian studies, and attempt to show how and why this change has happened, and its consequences for taxonomy and species limits. Examples are given in the form of ‘case studies’, and include Carrion/Hooded Crows Corvus corone/cornix, Green-winged/Eurasian Teals Anas carolinensis/crecca and Phylloscopus warblers.  相似文献
