Summary 1. The extent of nodulation and nitrogen fixation by a population ofHippophaë rhamnoides bushes occurring on a sand dune system on the east coast of England has been studied.2. Nodules were found on all plants (the latter ranged from 3 to 16 years in age) and using the isotope N15 it was confirmed that these nodules fixed nitrogen. The nodules on the youngest plants were most active in fixation but because of the greater total nodule mass older plants fixed the largest quantities of nitrogen.3. Large increases in total nitrogen (i.e. soil plus plant nitrogen) occurred with increase in age of the plants, such nitrogen increases ranging from 27 kg per hectare per annum in the presence of bushes 0 to 3 years old to 179 kg per hectare per annum in the presence of bushes 13 to 16 years old.4. The N15-technique indicated that a proportion of the nitrogen accumulated was due to fixation byHippophaë but nitrogen contributions from other sources could not be ruled out.5. The levels of nitrate-nitrogen and ammonium-nitrogen in the soil underHippophaë showed a marked seasonal variation, increasing to a maximum in the winter and to a minimum in the summer. These levels were in general inversely related to the level of average monthly rainfall. Because of the low levels of combined nitrogen in the soil during the summer months it seems unlikely that combined nitrogen should markedly inhibit fixation in this situation.  相似文献

In cells of Zea mays (root hairs, coleoptiles) and Riccia fluitans (rhizoids, thalli) intracellular Ca2+ and pH have been measured with double-barrelled microelectrodes. Free Ca2+ activities of 109–187 nM (Riccia rhizoids), 94–160 nM (Riccia thalli), 145–231 nM (Zea root hairs), 84–143 nM (Zea coleoptiles) were found, and therefore identified as cytoplasmic. In a few cases (Riccia rhizoids), free Ca2+ was in the lower millimolar range (2.3±0.8 mM). A change in external Ca2+ from 0.1 to 10 mM caused an initial and short transient increase in cytoplasmic free Ca2+ which finally levelled off at about 0.2 pCa unit below the control, whereas in the presence of cyanide the Ca2+ activity returned to the control level. It is suggested that this behaviour is indicative of active cellular Ca2+ regulation, and since it is energy-dependent, may involve a Ca2+-ATPase. Acidification of the cytoplasmic pH and alkalinization of the vacuolar pH lead to a simultaneous increase in cytoplasmic free Ca2+, while alkalinization of pHc decreased the Ca2+ activity. Since this is true for such remote organisms as Riccia and Zea, it may be concluded that regulation of cytoplasmic pH and free Ca2+ are interrelated. Summary A diallel analysis of combining ability, including maternal effects, genotype X environment interaction and the progress under selection, is reported in three selected crosses of Brassica campestris L. var. yellow sarson, involving 15 types, including 10 four-valved and 5 two-valved types from different parts of India. Twelve characters, including oil content, were studied in the f1 generation.The investigation has revealed only marginal superiority of f1's over the parents for most of the characters related to yield. There was no relation between heterozygosity and stability of performance over environments for yield or its components or for oil content. Substantial maternal effects were observed which also interacted with environments. Creation of variation for primary and secondary branches would be essential for changing yield level in yellow sarson. The presence of limited additive variation available for selection for yield components should be augmented by biparental mating the early segregating generations to break linkages, and was demonstrated by the recombinants obtained when this method was adopted.The magnitude of genotype — environment interactions in this study, as compared with the total genetic components for yield, oil content, number of siliquae on main axis and presence of large reciprocal variances in relation to general and specific combining ability variances for practically all characters, and the large interaction of , narrowed down the expected effectiveness of selection.Biparental mating in the three best crosses yielded three new recombinants outyielding the best check T 10 by the margins of 14%, 39% and 15%, respectively, in the yield trial. These recombinants had more primary branches and secondary branches, larger main axes and more siliquae with an increased number of seeds per siliqua, than any of the F1's in this study.  相似文献

Pelargonium × hortorum ‘Grand Prix’ which is susceptible to leaf yellowing and ‘Bergpalais’ which is not susceptible to leaf yellowing were chosen for the experiments. Ethylene production and action as well as the associated morphological response of Pelargonium shoots grown in the presence of a precursor of ethylene biosynthesis 1-aminocyclopropane-1-carboxylic acid (ACC), ethylene inhibitors: α-aminooxyacetic acid (AOA) and silver nitrate (AgNO3) and different cytokinins: (meta-topolin) (mT) or 6-benzylaminopurine (BAP) were studied. It was found that ‘Grand Prix’ was more sensitive to ethylene than ‘Bergpalais’ and it showed the leaf yellowing in response to 0.1 mg l?1 ACC. Moreover, it was noted that ACC added separately or together with cytokinin influenced Pelargonium morphogenesis. Depending on the concentration of ACC (0.1–2.0 mg l?1), it either stimulated or inhibited shoot and root formation as well as the growth of shoots and leaf blades. ACC-induced leaf yellowing in ‘Grand Prix’ was effectively inhibited by mT. In contrast, BAP did not enhance shoot quality. Simultaneously, the presence of mT in the medium resulted in up to a twofold increase in the ethylene production by ‘Grand Prix’ shoots throughout the culture period compared with the shoots growing on the BAP-medium. The inhibitor of ethylene action (AgNO3) added with cytokinin prevented the yellowing of Pelargonium shoots, but simultaneously influenced the formation of mature shoots with limited long-term multiplication potential. The shoots of P. × hortorum ‘Grand Prix’ treated with AgNO3 and mT emitted two- and sevenfold more ethylene after 11th and 21st day of culture compared with those treated with AgNO3 and BAP. It is suggested that mT inhibits the early senescence of Pelargonium in vitro by decreasing its sensitivity to ethylene.  相似文献

Coniferin specific- and isoflavone 7-glucoside specific -glucosidases have been localized in stem and root sections of chick pea (Cicer arietinum L.) seedlings by the indirect immunofluorometrical method. The coniferin specific -glucosidase has been found in the cell walls of the tracheary elements and of the endo-, epi-, and exodermis. All these tissues are known to contain either lignin or polymers, like suberin and cutin, which consist partially of phenylpropanoid elements. The localization of this -glucosidase is therefore in agreement with its postulated relationship to the phenylpropanoid metabolism. The isoflavone 7-glucoside specific -glucosidase, on the other hand, is predominantly located in the parenchymatic cortex cells, and obviously in the cytoplasm. These cells are known to contain the isoflavone formononetin, which has been shown to undergo turnover in chick pea seedlings. Two novel delphinidin 3-(tri or di)-glycoside-7-glycosides were isolated from the violet–blue flowers of Vinca minor L. and V. major L. (Family: Apocynaceae), and determined to be delphinidin 3-O-[2-O-(β-xylopyranosyl)-6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-(2G-xylosylrobinobioside)-7-rhamnoside] as major floral anthocyanin of V. minor and delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-robinobioside-7-rhamnoside] as major floral anthocyanin of V. major by chemical and spectroscopic methods. In addition, chlorogenic acid and kaempferol 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= kaempferol 3-robinobioside-7-rhamnoside (robinin)] were identified in these flowers. Summary Homozygote plants for the (S) self-incompatibility gene have been produced in Brassica campestris L. var. T 15. Stigmas from plants designated S 1 S 1, S 2 S 2 and S 4 S 4 were extracted and their protein separated on an isoelectric focusing mini-gel. Differences were observed between proteins from stigmas of the three S-homozygous groups: S-genotype specific proteins were determined for S 1 S 1 and S 2 S 2 stigmas that were absent in the self-compatible S 4 S 4 stigmas. Carbon dioxide (CO2), which is known to block the self-incompatibility reaction in Brassica, was applied to [35S]-methionine unpollinated, self- and cross-pollinated stigmas to observe the effect of external CO2 on the synthesis of these S-associated proteins. The results indicate that pollination triggers a dramatic reduction in protein synthesis in general and in the synthesis of S2-associated protein after self-pollination in particular.  相似文献

 The development of the embryo and endosperm has been investigated in an intraspecific Tulipa gesneriana cross and in the incongruent cross T. gesneriana ×T. agenensis at intervals of 10 days, from 12 to 82 days after pollination (DAP). In both tulip crosses, the zygote gives rise to an apparently undifferentiated cell mass, the proembryonal cell mass, on which a suspensor then develops. Subsequently, a globular embryo is formed on top of the suspensor. This embryo finally elongates, giving rise to a spindle-shaped embryo. The cellular endosperm fills the whole embryo sac in mature seeds, except for a region immediately around the embryo. In both crosses, aberrant developments were found. In the intraspecific T. gesneriana cross, the pollen tubes did not open in a number of ovules. In other ovules, the pollen tubes seemed to have opened, but an embryo or endosperm was not found or only endosperm was observed. In the cross T. gesneriana ×T. agenensis, fewer pollen tubes entered the ovules than in the intraspecific T. gesneriana cross. The ovules with embryo and endosperm formation of the incongruent interspecific cross showed, in general, retarded development in comparison with the intraspecific T. gesneriana cross. The first globular embryos and spindle-shaped embryos were found at the later fixation dates and the relatively lower number of spindle-shaped embryos at 82 DAP had a shorter average length. The number of ovules with deformations in embryo and/or endosperm development was also higher in the cross T. gesneriana × T. agenensis in comparison with the intraspecific T. gesneriana cross. Between 87% and 100% of the ovules with embryo and endosperm development showed normal development in the intraspecific T. gesneriana cross, while in the incongruent interspecific cross, from 22 DAP, between 17% and 56% of the ovules showed normal development. Of those ovules with aberrations in embryo and/or endosperm formation, about 80% had a deformed endosperm, of which more than 50% also contained a deformed embryo. The cumulative ozone effect on morphological parameters (visible leaf injury, plant height and leaf growth, number of bean pods, petunia flowers and stalks) was examined in this study. Well-known ozonesensitive (Bel W3) and ozone-resistant (Bel B) tobacco cultivars as well as bean cv. Nerina and petunia cv. White cascade, both recognized as ozone sensitive, were used in the experiment. Investigations were carried out at two exposure sites varying in tropospheric ozone levels. Ozone negatively affected the leaf growth of both tobacco cultivars and bean. A negative relation was also found for ozone concentration and tobacco plant height. Number of petunia flowers and stalks and bean pods was positively correlated with ozone concentration. This could have been connected with earlier plant maturation due to faster generative development of plants in ozone-stress conditions.  相似文献

Intergenomic F1 hybrids between L. auratum x L. henryi and their BC1 progeny were investigated through genomic in situ hybridization technique (GISH) to determine their potential value in lily breeding. We confirmed that F1 intergenomic hybrids possessed a set of chromosomes (x=12) from both parents and that flowers of the F1 auratum × henryi hybrid showed an intermediate morphological phenotype. Pollen size, viability and germination ability were measured through microscopic observations. F1 intergenomic hybrids produced a relevant frequency of 2n-gametes, which were successfully used to perform crosses with Oriental hybrids, resulting in the triploid Oriental Auratum Henryi (OAuH) hybrid. Twenty BC1 plants were generated by crossing between four different Oriental hybrid cultivars and F1 AuH hybrids using an in vitro embryo rescue technique, after which the genome constitution and chromosome composition were analyzed by GISH. All plants were triploid, showing 12 from female parents (diploid Oriental hybrid) and 24 from male parents (diploid F1 AuH hybrid). Overall, 16 out of 20 BC1 progeny possessed recombinant chromosomes with 1-5 crossover sites per plant. Cytological analysis of 20 BC1 plants by GISH verified that the occurrence of 2n pollen formation in all F1 AuH hybrids was derived from the FDR (first division restitution) mechanism, in which the genome composition of all BC1 plants possess 12 Oriental + 12 L. auratum + 12 L. henryi chromosomes. Allotriploids derived from the AuH hybrid were used as female for crossing with the diploid Oriental hybrid cultivar ''Sorbonne'' and considerable numbers of plants (0-6.5 plants per ovary) were only obtained when female OAuH (BC1) triploids were used. Taken together, the results of this study indicate that production and analysis of F1 AuH hybrids and their progeny through sexual polyploidization can be useful for efficient creation of important horticultural traits.  相似文献

There are several wild and cultivated plants that offer excellent opportunities for being used as herbal and therapeutic agents. The identification of medicinal properties of plants and their effectiveness in treating diseases are important components in medicinal plant research and this can pave ways for further improvements in traditional drug use. Climatic conditions and phytogeography in Pakistan are ideal for naturally occurring diverse flora and managed cultivation of hundreds of plants of medicinal significance. Nigella sativa commonly known as the black seed is an important medicinal plant that has been widely used as a multipurpose medicinal agent in different countries since old times. The plant is abundantly cultivated in Pakistan for uses as condiment and medicines. It possesses important classes of bioactive compounds among which thymoquinone has attracted significant attention from the scientific community because of its active role in treating a diverse spectrum of diseases. The black seeds are used for reducing adverse effects of arthritis, asthma, inflammation, liver and gastro disorders besides their potential role in diabetes and cancers. The focus of this review is to highlight the medicinal significance of N. sativa in traditional medicine and opportunities for exploitation in contemporary medicine.  相似文献

Nodal cuttings of D. alata L. Barzo fuerte and Florido, as well as of D. abyssinica Hoch, were cultured in vitro in order to assess the influence of the photoperiod on the production of microtubers. For both species, the highest number of microtubers was obtained under 16 and 24-h photoperiods, whereas larger microtubers were generally produced at 8-h photoperiod. In D. alata, further increase in microtuber size was observed in a culture medium where NH4NO3 was omitted. This effect was less noticeable in D. abyssinica.  相似文献

Summary Nodal cuttings of Dioscorea alata L. Brazo fuerte and D. abyssinica Hoch. were cultured in vitro to assess the influence of NAA on the production of microtubers. In D. alata, high concentrations of NAA (27 and 54 M) favored the production of large microtubers, whereas the highest number of microtubers was obtained with 2.7 M. D. abyssinica was found to be more sensitive to NAA since concentrations higher than 0.27 M promoted the growth of callus on the root system. In this species, the production of the largest microtubers was obtained at 2.7 M whereas the number of microtubers was not affected by any concentration tested. In D. alata, the effects of ABA and BAP were also evaluated. The weight of the microtubers increased with increasing concentrations of ABA. This effect, however, was observed only on expiants cultured under 8 h photoperiod, but not on those cultured under 16 h. Summary Isolated guard cells ofCommelina communis L., in epidermal strips in which all cells other than guard cells have been killed by treatment at low pH, will open to a degree dependent on the K (Rb)/Cl(Br) concentration in the bathing medium. Estimates of the changes with aperture of the ion concentrations in the guard cells were made by measurement of86Rb uptake from RbCl, of82Br uptake from K82Br, and of potassium activity with a potassium-sensitive microelectrode. The osmotic effects of such changes were compared with the previous estimates of the osmotic changes required to change the aperture. The results suggest that a substantial fraction of the osmotic pressure of isolated guard cells is contributed by solutes other than KCl (or other potassium salts), and that, even in stomata opened by incubation on KCl solutions, a substantial fraction of the increase in osmotic pressure associated with opening is contributed by solutes other than KCl.  相似文献

Summary The distribution of two seed proteins, namely conglutin and a legumin-like globulin, in developing and mature seeds ofLupinus albus L. has been examined by immunocytochemistry and the concomitant modifications of their constituent polypeptides followed by SDS-PAGE. Both proteins were found within vacuolar protein bodies in various tissues of the cotyledons, although with some differences in the distribution patterns. The legumin-like protein was found to be deposited within the large storage parenchyma cells of the cotyledons in a manner similar to that reported for other storage proteins; little or no immunolabelling was associated with the cotyledonary epidermal and vascular parenchyma cells. In contrast conglutin was present in all cell types.A precursor of the legumin-like protein accumulated transiently in the developing cotyledon, but was subsequently modified by proteolytic cleavage. The onset of such modification was concomitant with a transition in the predominant vacuolar forms within the storage parenchyma cells.
