The Marble gallwasp Andricus kollari has a native range divided into two geographically separated lifecycles. In Eastern Europe and Turkey, the lifecycle involves a sexual generation on Turkey oak, Quercus cerris, while in Iberia and North Africa the sexual generation host is cork oak, Q. suber. Over the last 500 years, A. kollari has expanded its range into northern Europe, following human planting of Q. cerris from Italy and the Balkans. We ask: (i) what is the genetic relationship between eastern and western distributions of Andricus kollari? Can we determine which lifecycle is ancestral, and how long ago they diverged? (ii) To what extent have eastern and western native ranges contributed to northwards range expansion? (iii) Is there any evidence for hybridization between the two life cycle types? We present analyses of allozyme data for 13 polymorphic loci and of sequence variation for a 433 bp fragment of the mitochondrial cytochrome b gene. These show: (i) that four haplotype lineages (one in Spain, two in Hungary/Italy and one in Turkey) diverged more or less simultaneously between 1 and 2 million years ago, suggesting the existence of at least four refuges through recent ice age cycles. Our data cannot resolve which lifecycle type is ancestral. (ii) Populations north of putative refuges are divided into two sets. Populations in south‐west France are allied to Spain, while all remaining populations in northern Europe have been colonized from Italy and the Balkans. (iii) The transition from one race to another in south‐west France is marked by abrupt transitions in the frequency of refuge‐specific private alleles and corresponds closely to the northern limit of the distribution of cork oak. Although hybrids were detected in north‐west France, none were detected where the two lifecycles meet in south‐western France. The biology of oak gallwasps predicts that any hybrid zone will be narrow, and limited to regions where Q. cerris and Q. suber meet. Our data suggest that eastern and western A. kollari are effectively separate species.  相似文献

ABSTRACT

A phytosociological study of the urban woods of Rome is described. Six syntaxonomical types were identified: 1) Orno-Quercetum ilicis, an edaphic-xerophytic variant of the natural potential vegetation of the city (deciduous Quercus sp. pl. woods); 2) Aquifolio-Fagetum carpinetosum betuli, in the northern slopes of alluvial valleys; 3) Q. suber and Q. pubescens wood, ranked in Quercetalia pubescenti-petraeae order 4) Quercetum ilicis galloprovinciale suberetosum; 5) coppice of Q. cerris and Q. frainetto, which belongs to the Teucrio siculi-Quercion cerridis alliance; 6) mixed mesophile wood of Q. cerris and Ostrya carpinifolia which can be considered a transition between the Doronico-Fagion and the Teucrio siculi-Quercion cerridis alliances. All of these six woody vegetation types are characterised by the large penetration of Mediterranean species belonging to the Quercetea and Quercetalia ilicis and the middle-European ones of the Querco-Fagetea. This mosaic of species exemplifies the bioclimatic characteristics of Rome, which is situated in a Mediterranean transitional region.  相似文献

ABSTRACT

Six species of the Quercus genus (Quercus ilex L., Q. coccifera L., Q. suber L., Q. trojana Webb, Q. macrolepis Kotschy, Q. cerris L.) have been screened for cytosolic ascorbate peroxidase (APX) by means of native polyacrylamide gel electrophoresis (PAGE). A single isozyme was found in five species (Q. trojana, Q. suber, Q. cerris, Q. macrolepis and Q. coccifera), while Q. ilex shows two different APX proteins. The data showed marked similarities among Q. trojana, Q. suber, Q. cerris and Q. macrolepis with respect to the electrophoretic mobility. The validity of APX electrophoretic patterns in systematic studies is discussed.  相似文献

An experimental crossing program was carried out in order to test the hypothesis thatArmeria villosa subsp.carratracensis, a serpentine endemic from Southern Spain, is of hybrid origin. Fruit-set and mainly pollen stainability data in artificial hybrids demonstrate that internal interspecific barriers are weak. Two generations of backcrossing with the two putative parents —A. colorata andA. villosa subsp.longiaristata— can restore pollen stainability to an average of 83.2 and 68.7, respectively. A morphometric analysis of 148 specimens belonging to parental species, different artificial hybrid generations and wild putative hybrid race shows that (1) morphological characters have a strong genetic basis and thus are good markers in identifying hybrids and introgression inArmeria; (2) artificial backcrossing produces in morphometrical characters a variation which conforms in both trend and magnitude to what is commonly attributed in the wild to introgression; (3) the position of the putative hybrid race is intermediate between the two parental species used in the program. The taxonomy of the genusQuercus is still unclear. In order to elucidate the taxonomy of Mediterranean oaks we have analyzed ribosomal RNA genes ofQuercus cerris, Q. coccifera, Q. trojana, Q. ilex, Q. suber, andQ. macrolepis by means of Southern blot hybridization. Oak nuclear DNA was extracted from root tips of 300 acorns and from catkins of single plants. EcoRI and BamHI restriction endonucleases were used. DNA electrophoresis and rRNA/DNA hybridization were performed usingVicia faba rRNA 18 S and 25 S as probes. The rRNA genes of all the species studied have an identical restriction mapping in the 18 S and 25 S regions, while differences in length are present in the intergenic regions.Q. cerris possesses at least four types of genes of 12.1, 11.5, 8.5, and 8.3 kb;Q. coccifera at least three types of 12.4, 10.4, and 10.1 kb;Q. trojana possesses the same rRNA genes asQ. cerris plus another gene type 12.0 kb long, with EcoRI and BamHI restriction sites in the intergenic spacer;Q. ilex at least three types of 12.4, 10.85, and 9.5 kb;Q. suber at least five types of 11.5, 11.0, 8.6, 8.5, and 8.3 kb;Q. macrolepis, finally, at least seven types of 11.5, 11.0, 10.2, 8.6, 8.5, 8.3, and 8.15 kb.Q. coccifera andQ. ilex rDNA appears quite different respect to other species examined, while high similarity seems to exist betweenQ. cerris, Q. trojana, Q. suber, andQ. macrolepis. Tucker, J. M. (U. California, Davis.) Studies in the Quercus undulata complex. III. The contribution of Q. arizonica . Amer. Jour. Bot. 50(7): 699–708. Illus. 1903.—Of the 7 oak species involved in the Quercus undulata complex, Q. arizonica contributes the least of all. The latter has hybridized with Q. gambelii at a few widely separated localities (listed in Table 1) in central Arizona, and northern Sonora and Chihuahua, Mexico. The putative hybrids (identifiable as Quercus undulata) occur as isolated individuals with the parental species. The parents, although regionally sympatric, are usually ecologically isolated. They differ in a number of morphological characters, 6 of which were analyzed in detail. The resulting data, presented as pietorialized scatter diagrams, demonstrate that the putative hybrids are intermediate, in general. This is taken as evidence of their hybrid nature. Factors limiting hybridization in oaks are discussed. It is speculated that hybridization between Q. arizonica and Q. gambelii has occurred since the postglacial hypsithermal interval.  相似文献

In this study foliar sclerophyll indexes, N %, P % and K % concentrations and correlations between sclerophyll index and these parameters were investigated in Quercus cerris L. var. cerris and Phillyrea latifolia L. In addition to this soil samples were collected and some physical and chemical analysis were carried out in these samples. Finally it has been determined that sclerophyll index was correlated with leaf N % concentrations in Q. cerris var. cerris and P. latifolia and leaf K % concentration in Q. cerris var. cerris.  相似文献

New records of Cynipid gall wasps and inquilines for the Italian peninsula and Sicily and their new host plants for the Palaearctic Region are listed and commented on. Among them we find: Cerroneuroterus cerrifloralis (Müllner 1901) as new for Italy and new for the Palaearctic region as host on Quercus suber; Andricus multiplicatus Giraud 1859 on Q. suber, as new host for the Palaearctic region; Aylax papaveris (Perris 1839), reported in Italy over a century ago, but later overlooked; Cerroneuroterus minutulus (Giraud 1859), also reported more than a century ago from Sicily, but later overlooked. Among the inquilines are here listed: Synergus variabilis Mayr 1872, emerged from Janetia cerris (Kollar 1850) galls (Diptera Cecidomyiidae), and found for the first time in the Palaearctic Region as host on Q. suber; Saphonecrus haimi (Mayr 1872) and Saphonecrus barbotini Pujade-Villar & Nieves-Aldrey 1986, are new records for Italy.  相似文献

Healthy and declining English oak (Quercus robur) and Turkey oak (Q. cerris) in north‐western Italy, in a plain oak forest showing decline for oak puzzle disease, were tested to assess possible variations in the composition of their fungal endophytic communities and their relation to the health status of trees. Samples collected in spring (buds) and in autumn (leaves, annual shoots and twigs) were surface‐sterilized, cut into fragments and placed on potato dextrose agar for a month; 26 fungal species were isolated, the most frequent being Tubakia dryina, Dendrodochium sp., Eutypella sp. and a sterile mycelium. Correspondence analysis showed significant qualitative differences between assemblages inhabiting twigs and herbaceous tissues that were due to the low frequency of Tubakia dryina in twigs and its higher frequency in buds, leaves and shoots. Tubakia dryina was isolated more frequently from leaves of declining oaks and from buds of healthy oaks; Monochaetia monochaeta showed a preference for healthy trees, especially leaves and buds. According to the Shannon–Wiener index, endophytic fungal communities of leaves, twigs and buds of declining English oak were poorer than those of declining Turkey oak, but there were no significant differences between healthy hosts.  相似文献

Abstract

By using the protoplast fusion technique, we have obtained 44 regenerated plants, phenotypically different and distinct from their parents, among which we have identified a fertile symmetric somatic hybrid, designated as TG-32, between N. tabacum var. Gexin No.1 and N. glauca. The morphology, fertility, chromosome number and nuclear constitution of the somatic hybrid have been studied in detail. Unlike other asymmetric interspecific somatic hybrids, the chromosome number of the symmetric somatic hybrid is 72, equal to the sum of chromosomes of both parents. The TG-32 plant has flowers similar to those of N. tabacum, but with petals similar to those of N. glauca. Interestingly the offspring of TG-32 vary in seed production ability with temperature, and produce more seeds under a relatively low temperature. Two SCAR markers were used to evaluate genetic variability and structure. The hybrid amplified the expected fragment, but the parents showed only one of two markers. This experimental result supports the hypothesis of the co-existence of two parental genomes in the somatic hybrid.  相似文献

Allozyme variation at 14 loci was studied electrophoretically in 19 population samples of Quercus cerris, Q. pubescens, Q. petraea, and Q. robur. Genetic variation, measured as mean heterozygosity, was found to be high within populations but low among populations. A significant deficit of heterozygotes was observed in most of the populations of Q. pubescens, Q. petraea, and Q. robur. At the species level, Q. cerris (sect. Cerris) appears to be clearly separated from the rest (sect. Quercus). Q. pubescens, Q. petraea, and Q. robur form a cluster of closely-related species. The degree of genetic differentiation among these species is low, i.e. in the range normally found among con-specific populations. Nevertheless, allozyme data allow the elaboration of a dendrogram which is in reasonable accordance with the taxonomic classification, but also supports the importance of hybridization and introgression. This is also underlined by the fact that the allelic differentiation at the Got-2 locus corresponds with the phenotypic classification (petraea-like, hybrids, and robur-like) in samples from mixed populations.  相似文献

Elevated CO2 may affect litter quality of plants, and subsequently C and N cycling in terrestrial ecosystems, but changes in litter quality associated with elevated CO2 are poorly known. Abscised leaf litter of two oak species (Quercus cerris L. and Q. pubescens Willd.) exposed to long-term elevated CO2 around a natural CO2 spring in Tuscany (Italy) was used to study the impact of increasing concentration of atmospheric CO2 on litter quality and C and N turnover rates in a Mediterranean-type ecosystem. Litter samples were collected in an area with elevated CO2 (>500 ppm) and in an area with ambient CO2 concentration (360 ppm). Leaf samples were analysed for concentrations of total C, N, lignin, cellulose, acid detergent residue (ADR) and polyphenol. The decomposition rate of litter was studied using a litter bag experiment (12 months) and laboratory incubations (3 months). In the laboratory incubations, N mineralization in litter samples was measured as well (125 days). Litter quality was expressed in terms of chemical composition and element ratios. None of the litter quality parameters was affected by elevated CO2 for the two Quercus species. Remaining mass in Q. cerris and Q. pubescens litter from elevated CO2 was similar to that from ambient conditions. C mineralization in Q. pubescens litter from elevated CO2 was lower than that from ambient CO2, but the difference was insignificant. This effect was not observed for Q. cerris. N mineralization was higher from litter grown at elevated CO2, but this difference disappeared at the end of the incubation. Litter of Q. pubescens had a higher quality than Q. cerris, and indeed mineralized more rapidly in the laboratory, but not under field conditions.  相似文献

Quercus suber L. is a Mediterranean forest species with ecological, social and economic value. Clonal propagation of Q. suber elite trees has been successfully obtained from in vitro‐derived somatic and gametic embryos. These clonal lines play a main role in breeding and genetic studies of Q. suber. To aid in unravelling diverse genetic and biological unknowns, a proteomic approach is proposed. The proteomic analysis of Q. suber somatic and gametic in vitro culture‐derived embryos, based on DIGE and MALDI‐MS, has produced for the first time proteomic data on this species. Seventeen differentially expressed proteins have been identified which display significantly altered levels between gametic and somatic embryos. These proteins are involved in a variety of cellular processes, most of which had been neither previously associated with embryo development nor identified in the genus Quercus. Some of these proteins are involved in stress and pollen development and others play a role in the metabolism of tannins and phenylpropanoids, which represent two of the major pathways for the synthesis of cork chemical components. Furthermore, the augmented expression levels found for specific proteins are probably related to the homozygous state of a doubled‐haploid sample. Proteins involved in synthesis of cork components can be detected at such early stages of development, showing the potential of the method to be useful in searching for biomarkers related to cork quality.  相似文献

ABSTRACT

The hypothesis that Lathyrus plitmannii may be of hybrid origin was tested on a morphological data set, derived from measurements of 50 herbarium specimens, by employing the computer program HYWIN. Results showed that all the specimens of L. plitmannii were putative hybrids and that the highest ranked pair of specimens that best matches the morphologies of its parents was L. gorgonei and L. pseudocicera. Examination of the distribution patterns of the three species shows that L. plitmannii is confined to regions common to both putative parents and that hybrids could be formed, assuming that genetic barriers were overcome. Additional supporting evidence includes overlaps in the edaphic requirements and the habitats of the three species. Lathyrus plitmannii is recognised as a distinct species that should be regarded as derivative of hybridisation between L. gorgonei and L. pseudocicera.  相似文献

An investigation of randomly amplified polymorphic DNA (RAPD) and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) marker distribution was made for two well-characterised hybrids and their parents,Leucaena leucocephala andL. esculenta andParkinsonia aculeata andCercidium praecox. Three chloroplast DNA (cpDNA) markers identified the maternal parent of eachL. leucocephala ×L. esculenta hybrid. Fifteen species-diagnostic RAPD markers (invariant in one taxon and absent from the other) were always present in theLeucaena hybrid and assumed to be of nuclear origin, whilst three RAPD markers showed expression patterns identical to the cpDNA markers and were assumed to be of organellar origin. No RAPD or PCR-RFLP taxon-diagnostic markers were discovered for eitherP. aculeata orC. praecox. However, 21 RAPD markers were species-specific (polymorphic within one taxon but absent from the other) and Southern analysis indicated that none of the markers were of organellar origin. Only 67% additivity of markers specific toP. aculeata andC. praecox was demonstrated in the hybrids between these two species, whilst inLeucaena 97% additivity was demonstrated. Differences between the two hybridising situations were related to the behaviour of the molecular markers and the biology of the species.  相似文献

Interspecific hybridization occurs with high frequency in the genusQuercus, but few studies have analyzed and compared micromorphological characters in putative parental species and their hybrids.Quercus eduardii andQ. conzattii are two Mexican black oak species that, although distantly related, have formed at least one population of hybrid origin, where individuals with intermediate macromorphology are present. The purpose of this investigation was to analyze the degree of differentiation in micromorphological characters between the two species and to assess the expression of these characters in individuals with intermediate macromorphology. Foliar trichomes, epicuticular waxes, stomata, and pollen grains, were examined using scanning electron microscopy (SEM) in the three types of individuals (Q. conzatti, Q. eduardii, and intermediates). Trichome density was quantified with light microscopy. Types of trichomes present, length of trichome arms, types of epicuticular wax on the leaf surfaces, and the position of stomata with respect to the foliar surface were characters useful to differentiate betweenQ. conzattii andQ. eduardii. Plants with intermediate macromorphology displayed a pattern of micromorphological characters that were identical to one parental species (Q. conzattii), or extreme or novel relative to both species.  相似文献

Intraspecific barriers promote outcrossing while interspecific mechanisms may contribute to the isolation of species; both control the exchange of genes between plants. In this paper we establish that post-pollination mechanisms promote outcross and act at different temporal and spatial levels to control seed set and quality in Quercus species. Controlled pollinations were performed to investigate intraspecific crossing barriers in Q. suber and pollen-pistil interactions following interspecific pollinations with some simpatric Quercus species. Cytological data of intraspecific crosses in Q. suber and Q. ilex have shown different kinetics on pollen tube growth after self and outcross pollination, but pollen tubes were able to reach the base of the styles in both species and seed set was successful. Although some pre-zygotic interaction is occurring at the style, the most important interaction takes place at ovary. The cross between Q. ilex and Q. suber is possible only in one direction, revealing a case of unilateral incongruity. We show that the lack of seed set observed in the cross Q. suber×Q. ilex is due to the inability of pollen tubes to penetrate the transmitting tissue after germination. With Q. suber mainly as female parent, pollinations with other simpatric Quercus species have shown different levels of constraint on pollen tube progression at stigma and style levels. Eventual hybridisation between Quercus species will depend on the compatibility of pollen-pistil interactions, on the competitive ability of pollen genotypes, and, most important, on the overlapping of geographical, phenological and ecological factors. Differences in seed set and seed allocation was evident in all crosses, and was particularly outstanding in interspecific and in self intraspecific crosses, determining the ultimate level of seed production and quality in Quercus species. Hybrid seedlings from the cross Nicotiana tabacum × N. suaveolens express lethality at 28°C. We carried out a cross between monosomic lines of N. tabacum lacking the Q chromosome and N. suaveolens by test-tube pollination and ovule culture at 28°C. To suppress hybrid lethality, hybrid seedlings obtained were transferred to 36°C immediately after germination and cultured. We determined whether Q-chromosome-specific DNA markers were detected among hybrid seedlings. When hybrid seedlings cultured at 36°C were transferred to 28°C, hybrid seedlings in which Q-chromosome-specific DNA markers were detected expressed hybrid lethality, while hybrid seedlings in which Q-chromosome-specific DNA markers were not detected did not express hybrid lethality. From these results, we concluded that the presence of the Q chromosome of N. tabacum is related to hybrid lethality observed in crosses between N. tabacum and N. suaveolens. This is the first report that clearly demonstrates the relationship between a certain chromosome and hybrid lethality in the genus Nicotiana using chromosome-specific DNA markers. Additionally, we confirmed that the Q chromosome belongs to the S subgenome because Q-chromosome-specific DNA markers were detected only in N. sylvestris.  相似文献
