ABSTRACT

Hermodactylus tuberosus Mill. (Iridaceae) is a species with a very short style and a tricarpellar unilocular ovary. For ornamental purposes it is usually reproduced by means of tubers, although both vegetative and seed reproduction occur in nature. However, its capsules do not contain many seeds in comparison to the high number of ovules inside the ovary. To understand this conflicting process a study on H. tuberosus pollen-pistil interaction was carried out. Pollen viability was evaluated by different techniques. Germination rate was tested in vitro and in vivo after stigma, intrastylar and intraovarian pollinations. A high percentage of the pollen grains were found to be viable resulting in a high percentage of in vitro germination and a low rate of germination on the stigma. Indeed, many pollen tubes developed after intrastylar and intraovarian pollinations. Several anomalies characterized pollen germination and pollen tube growth in vitro and in vivo. Observations from field trials indicated that capsules were only produced from open or hand, cross- pollinated flowers, confirming that the plant is self- incompatible and not apomictic. In addition, all the mature capsules contained only a few mature seeds suggesting the existence of pre- and postzygotic seed set inhibition.  相似文献

Crocus cartwrightianus andC. cartwrightianus cv.albus pollen have been studied from structural and ultrastructural points of view and the germination assayed in vivo and in vitro.C. cartwrightianus pollen is regularly shaped and sized, has a low percentage of anomalous grains and has a high germination rate in vitro, whileC. cartwrightianus cv.albus is less regularly shaped with some variation in size and has a high percentage of anomalous grains and a low germination percentage in vitro. Ultrastructural observations have revealed, in the pollen of both the taxa, the presence of a thin elongated vegetative nucleus and a generative cell surrounded by a thin membrane. However,C. cartwrightianus pollen shows a thicker intine, andC. cartwrightianus cv.albus shows numerous pollen germination anomalies which are in common withC. sativus.  相似文献

Pollen of Crocus biflorus Miller subsp. biflorus from natural habitats of Tusculum (Frascati, near Rome, Italy) has been studied in order to compare its structure and physiology to pollen of other Crocus species belonging to the Crocus sativus group. Mature pollen grains are rounded, 60 μm in diameter, in-aperturate (but with surface incisions where exine is lacking). DAPI staining reveals a spindle-shaped generative nucleus which is intensely fluorescent, and vegetative nucleus which is less fluorescent, and is elongated with numerous lobes. At anthesis the pollen is bicellular, but about 2% of tricellular grains occur among the pollen grains released from the anthers as well as on both naturally or handpollinated stigmas. Pollen germination is low in vitro, but higher in vivo. The pollen tubes are of normal shape. An electron-dense surface coat is sometimes visible on the exine, which in many cases, is detached from the exine. The vegetative cytoplasm is very rich in glycolipid bodies surrounded by endoplasmic reticulum. The generative cell has a lobed cell wall and is surrounded by the vegetative nucleus.  相似文献

Summary Pearl millet [Pennisetum glaucum (L.) R. Br.] pollen has been successfully stored for 2,615 and 2,911 days at -18° and -73 °C, respectively, and continues to be viable. Viability of pollen stored at -73 °C appears to be little affected either by pollen storage moisture contents below 7.2% or by storage in glass vial or zip-lock plastic bag containers. Pollen moisture content appears to be more critical for maintaining viability at -18°C than at -73°C. Glass vials appear to be more desirable for longer term (>3 years) storage at -18°C.  相似文献

Anthers of Capsicum annuum L. were cultured on Murashige and Skoog (MS) medium containing 0.1 mg l−1 NAA and 0.1 mg l−1 kinetin. Inoculated anthers were subjected to 31 °C and development of microspores in anthers of varying stages was observed cytologically using 4′-6-diamidino-2-phenylindol-2HCl (DAPI). Pepper was characterized by a strong asynchrony of pollen development within a single anther. Percentage of pollen at different stages changed with the culture period, and the proportion of dead pollen increased drastically from day 2 after culture. Microspores that were cultured at the late-uninucleate stage followed one of two developmental pathways. In the more common route, the first sporophytic division was asymmetric and produced what appeared to be a typical bicellular pollen. Embryogenic pollen was formed by repeated divisions of the vegetative nucleus. In the second pathway, which occurred in fewer microspores, the first division was symmetric and both nuclei divided repeatedly to form embryogenic pollen. In early-bicellular pollen, sporophytic pollen was produced through division of the vegetative nucleus. In mid-bicellular pollen, the generative nucleus may undergo division to produce two or more sperm-like nuclei. However, division of the generative nucleus alone to form the embryo was never observed. The anther stage optimal for embryo production contained a large proportion (>75%) of early-binucleate pollen. Associations were found among the percentage of early-binucleate pollen, the frequency of embryogenic multinucleate pollen, and the yield of pollen embryos.  相似文献

The vegetative nucleus (VN) of Nicotiana tabacum L. has been qualitatively and quantitatively studied in fresh, hydrated, and activated pollen. Techniques included the use of optical sectioning by confocal scanning laser microscopy to obtain volume and surface area measurements, and stereoscopic pairs; and freeze-etch electron microscopy to estimate the frequency of nuclear pores per m2 in the vegetative nucleus. Several morphological changes were observed to occur in pollen grain nuclei during the early processes of tube growth. In freshly dehisced pollen grain, the vegetative and generative nuclei were side by side, but following hydration and activation of the grain, the elongated generative nucleus became partially surrounded by the vegetative nucleus. It was found that during hydration, the surface area of the vegetative nucleus increased and there was a decrease in the frequency of nuclear pores. The calculated total number of pores remained similar. After activation and pollen-tube growth, the vegetative nucleus retained the same surface area as in the hydrated state but the frequency of nuclear pores decreased; therefore, the calculated total number of pores was significantly lowered. In this study, pollens of Burdur dimriti, Sariemin, Tilki kuyrugu, Razaki, Buzgulu, Siyah buzgulu, and Siyah gemre table grape varieties (Vitis vinifera L.) grown in Isparta, Turkey were tested in vitro for viability, germination capability, and production level. It was determined that active pollen levels varied between 23.8 % and 80.8 % in FDA (flourescein diacetat), 31.5 % and 68.8 % in TTC (2,3,5-triphenyl tetrazolium chloride) tests. In respect to pollen germination rates, the best medium in hanging drop method and saturated petri method were 20 % sucrose concentration and 1 % agar+15 % sucrose solution, respectively. The highest and the lowest pollen production levels per flower were determined 9000 in Siyah gemre and 2906 in Siyah dimrit grape varieties, respectively.  相似文献

Pollen collected by honeybees foraging in the region of Bursa, Turkey was analysed for a whole year. Pollen loads were collected from the hives of Apis mellifera anatoliaca once a week and were classified by colour. Forty‐one taxa were identified from the pollen analyses of the loads and 14 of these had percentages higher than 1%. Only 2.05% of the total pollen could not have been identified. Dominant taxa include; Brassicaceae (11.19%), Helianthus annuus L. (10.84%), Cichorioideae (8.93%) Salix spp. (7.99%), Rosaceae (7.37%), Centaurea spp. (7.56%), Papaver spp. (7.41%), Knautia spp. (6.99%), Fabaceae (6.01%), Asteraceae (5.73%), Xanthium spp. (2.65%), Chrozophora spp. (2.45%), Plantago spp. (1.56%) and Acer spp. (1.54%) representing 88.23% of the total. Distinct variations in plant usage are seen through the year with initial use of Rosaceae, Salix, and to a lesser extent Brassicaeae. As these groups finish flowering the bees move onto Helianthus annuus, Centaurea through the summer followed by Asteraceae in the late summer and Fabaceae in the autumn. There is a strong reliance on crop species for pollen forage but a number of indigenous species are also seen within the samples. The most productive period for collecting various pollen types, and the ideal period to determine pollen preferences of honey bees was June‐August.  相似文献

Investigations of mitosis in 10 populations of Anemone nemorosa L. all indicated that this species has a chromosome number of 2 n = 30. No B chromosomes were found. There were no significant differences in chromosome morphology between the populations. Meiosis in the anthers, studied in three populations, was found to be mostly regular, with 15 bivalents, although occasional irregularities such as bridges and pentads were observed. Pollen viability was tested in 12 populations and although fairly low, with micro pollen grains in four of the populations, was not low enough to prevent a high proportion of fertilized achenes.  相似文献

The structure of the massulae composing the pollinium ofLoroglossum hircinum was studied before pollination and 12 and 24 hours afterwards. The grains are grouped in tetrads closely packed in massulae. The exine is only present on the outside of the massulae. The intine consists of two layers: a compact layer surrounding the pollen grain and a looser layer surrounding the pollen grain and a looser layer surrounding the tetrad. Twelve hours after pollination, pollen volume and the space between the tetrads increase due to vacuolization. Twenty-four hours after pollination, pollen volume and tetrad spacing are higher due to vacuolization and some grains have emitted pollen tubes. Pollen growth due to vacuole formation, and the absence of common walls between adjacent tetrads lead to crumbling of the massulae. The mature pollen grain does not have apertures: the site of pollen tube emission is determined after pollination. The first grains to germinate are those in the centre of the massula. The vegetative cell nucleus is the first to enter the pollen tube; the generative cell elongates and undergoes the second haploid mitosis shortly after entering the pollen tube.  相似文献

The aim of this paper is to characterise theairborne behaviour of an allergenic andabundant pollen type, Platanus, in sevensites of the Aerobiological Network ofCatalonia (Xarxa Aerobiològica deCatalunya, XAC). In Catalonia, the mostextended Platanus species is Platanus × hybrida Brot., widely planted inforestry and as ornamental tree in streets andgardens. Naturalised specimens can also befound along river banks.The pollen records analysed are from differentperiods comprised between 1994 and 2000 inBarcelona, Bellaterra, Cambrils, Girona,Lleida, Manresa and Tarragona. All data wereobtained using seven-day volumetric spore Hirsttraps.Platanus pollination is concentrated in afew weeks, between March and April. Platanus pollen is present in all theatmospheric pollen spectra for the sitesmentioned above. In Barcelona, Platanuspollen attained the highest concentrations,with an extreme annual index (sum of the meandaily concentrations) of 25790 in 1997, and amaximum daily concentration of 2567 P/m3on 15th March 2000. The lowest annual index(647) corresponds to Cambrils, 2000, and thelowest maximum daily concentration (74P/m3) was registered on 7th April1996, in Manresa.The interannual variation of Platanuspollen values can be explained not only bymeteorological parameters and the endogenousfactors of the tree, but also by human activitysuch as elimination, pruning and watering.  相似文献

Microsporangium dehiscence, pollen production and dispersal were studied in Himalayan cedar (Cedrus deodara) during 1998 and 1999. Microsporangium dehiscence showed diurnal periodicity and was found to be related to air temperature and relative air humidity, with a strobilus taking 2 d to dehisce completely in warmer conditions and 3 d in cooler ones. The frequency of flowering in C. deodara was highly variable during the two successive years; however, cyclical production of pollen grains was observed in 50% of the trees. The maximum concentration of pollen grains in the air was found between 1200 and 1600 h, and this period was also noted to be the best time for pollination. Studying migration of pollen grains from isolated single trees in three directions showed that migration was not uniform in all directions. Long-distance transport of pollen grains was observed in the downhill direction. However, in the uphill and horizontal directions grains could travel only up to 97.5 and l95.1 m, respectively, and the frequency of pollen grains to the source frequency at these distances was only 1.9 and 2-5%, respectively. The results suggest that an isolation barrier of 190 m may be considered as a minimum for the management of deodar seed orchards.  相似文献

In tristylous Pontederia cordata (Pontederiaceae), conspicuous differences in the size of pollen grains and discrete variation in the length of reproductive organs provide a suitable experimental system for the study of fine-scale pollination events. At a population of P. cordata at Pothole Lake, Ontario, the majority of flowers are visited by bumble bees which remove on average 45% of the pollen during single visits to previously unvisited flowers. The amount and proportion of pollen removed are significantly different among floral morphs and stamen levels. Deposition of the three pollen types on the bodies of Bombus spp., Apis mellifera and Melissodes apicata is non-random: large- and medium-size pollen tends to remain in greatest concentrations where it is initially deposited, whereas small-size pollen is displaced from the proboscis to more posterior body parts, probably as a result of grooming activities. Stigmatic pollen loads of individual flowers following single bumble bee visits indicate that the mid-styled morph captures the largest total pollen load, and the short-styled morph the smallest. The largest proportion of compatible pollen grains is deposited on stigmas of the long-styled morph. Pollen load data from "single visit" flowers is in general agreement with previously published population surveys involving multiply-visited flowers.  相似文献

BACKGROUND AND AIMS: Flowers of Commelina coelestis and C. dianthifolia provide pollen alone as a floral reward, and rely on visual cues to attract pollinators. Three stamen types, all producing pollen, occur in each of these species: two cryptically coloured lateral stamens, a single cryptically coloured central stamen and three bright yellow staminodes that sharply contrast with the blue to purple corolla. The objective was to compare the stamen structure and pollen characteristics of each of the three stamen types, and to test the hypothesis that the staminodes are poor contributors of viable pollen for the siring of seed. The pollination roles of the three stamen types and the breeding systems of both species were also explored. Methods: Light, fluorescence and scanning electron microscopy were utilized to examine stamen morphology and pollen structure and viability. Controlled hand pollinations were used to explore the breeding system of each species. Filament and style lengths were measured to investigate herkogamy and autogamy. KEY RESULTS: Pollen from all stamen morphs is viable, but staminode pollen has significantly lower viability. Pollen polymorphism exists both (a) between the lateral and central stamens and the staminodes, and (b) within each anther. Lateral and central stamens have thicker endothecia with a greater number of secondary cell wall thickenings than the staminodes. CONCLUSIONS: Both species are entomophilous and facultatively autogamous. Lateral stamen pollen is important for cross-pollination, central stamen pollen is utilized by both species as a pollinator reward and for delayed autogamy in C. dianthifolia, and the staminodes mimic, by means of both colour and epidermal features, large amounts of pollen to attract insects to the flowers. Pollen from all three anther morphs is capable of siring seed, although staminode pollen is inferior. The thin staminode endothecium with fewer secondary thickenings retards staminode dehiscence.  相似文献

An electron microscope study of the vitelline follicles of Gorgoderina vitelliloba indicates that they contain vitelline cells in various stages of development. Juvenile cells are small and characterised by a little cytoplasm. During differentiation a large amount of granular endoplasmic reticulum develops. In more mature cells, indistinct Golgi complexes give rise to globules of shell protein which migrate to form clusters at the periphery of the cell. Further maturation results in the appearance of large lipid bodies in the vitelline cell cytoplasm.Developing vitelline cells are ensheathed by nurse cell cytoplasm containing numerous small vacuoles which appear to be derived from smooth endoplasmic reticulum. It is suggested that nurse cells may have a role in selection and transport of nutrient material for vitelline cells and that they manufacture precursors of lipid which is subsequently stored as a food reserve in mature vitelline cells. Possible transport sites between parenchymal cells and nurse cells were identified.  相似文献

Variation in anther and pollen morphology is described and illustrated for species of Leucaena and a selection of species from closely related genera. This is discussed in relation to the variation found in the closely related genera of the informal Leucaena, Dichrostachys and Xylia groups of the tribe Mimoseae. A number of characters for cladistic analysis are identified and putative homologies assessed. Preliminary taxonomic conclusions about the status and relationships of L. multicapitula are discussed. Mode of anther dehiscence is also described and illustrated. The observation of persistent intact tapetal membranes after anther dehiscence in some species of Leucaena is discussed in relation to pollen unit.  相似文献

Summary A procedure for isolating sperm cells of perennial ryegrass (Lolium perenne L.) was developed. The sperm cells were released from the pollen grains by osmotic shock, with the right combination of pH and osmolality being important for optimal release. Various combinations of vitamins E, C and fetal calf serum were tested with the aim of improving yield and long-term viability, and their possible mode of action as important components for improvement of these two parameters is discussed. Under optimized conditions, a yield of 12% was established, and the storage time after which 50% of the sperm cells were still viable was improved to 60 h. Cytological observations demonstrated that sperm cells of perennial ryegrass are true protoplasts, which may allow future fusion experiments to be carried out.  相似文献

Background and Aims

The coastal plain of Israel hosts the last few remaining populations of the endemic Iris atropurpurea (Iridaceae), a Red List species of high conservation priority. The flowers offer no nectar reward. Here the role of night-sheltering male solitary bees, honey-bees and female solitary bees as pollinators of I. atropurpurea is documented.

Methods

Breeding system, floral longevity, stigma receptivity, visitation rates, pollen loads, pollen deposition and removal and fruit- and seed-set were investigated.

Key Results

The main wild pollinators of this plant are male eucerine bees, and to a lesser extent, but with the potential to transfer pollen, female solitary bees. Honey-bees were found to be frequent diurnal visitors; they removed large quantities of pollen and were as effective as male sheltering bees at pollinating this species. The low density of pollen carried by male solitary bees was attributed to grooming activities, pollen displacement when bees aggregated together in flowers and pollen depletion by honey-bees. In the population free of honey-bee hives, male bees carried significantly more pollen grains on their bodies. Results from pollen analysis and pollen deposited on stigmas suggest that inadequate pollination may be an important factor limiting fruit-set. In the presence of honey-bees, eucerine bees were low removal–low deposition pollinators, whereas honey-bees were high removal–low deposition pollinators, because they removed large amounts into corbiculae and deposited relatively little onto receptive stigmas.

Conclusions

Even though overall, both bee taxa were equally effective pollinators, we suggest that honey-bees have the potential to reduce the amount of pollen available for plant reproduction, and to reduce the amount of resources available to solitary bee communities. The results of this study have potential implications for the conservation of this highly endangered plant species if hives are permitted inside reserves, where the bulk of Oncocyclus iris species are protected.  相似文献
