Large single crystals of isocitrate dehydrogenase from Azotobacter vinelandii have been grown by vapor diffusion from ammonium sulfate and phosphate solutions. The crystals are tetragonal, space group P42212 with cell dimensions a = 122.1 A?, c = 163.9 a?. There are two molecules of 80,000 molecular weight per asymmetric unit. Native data to 5.5 Å resolution have been collected on a diffractometer. A rotation function using data between 10 Å and 6 Å resolution indicates three possible orientations of the non-crystallographic 2-fold axis relating the two molecules.  相似文献

An X-ray study of orthorhombic crystals of cytosolic aspartate transaminase from chicken heart has been carried out at 5 Å resolution. The crystals belong to space group P212121, with unit cell dimensions a = 62.7 A?, b = 118.1 A?, c = 124.5 A?. The electron density map has been calculated on the basis of five heavy-atom derivatives. The model of the molecule derived from this map revealed clearly two subunits of similar structure related by a non-crystallographic dyad. The secondary structure of the protein comprises nine helical segments per subunit.The enzyme has been shown to be catalytically active in the crystal form. Removal of the coenzyme from the crystals made it possible to derive from the difference Fourier map the position of the active site in the enzyme molecule.Significant conformational changes have been observed which accompany the interconversion of intermediates of the enzymic reaction.  相似文献

Tyrosyl-transfer RNA synthetase from Bacillus stearothermophilus has been crystallized as hexagonal plates, P3121, a = b = 64.6 A?, c = 238.8 A?, with the dimeric molecule (molecular weight, 90,000) occupying two crystallographic asymmetric units (Reid et al., 1973). Three heavy-atom derivatives have been identified and X-ray diffraction measurements have been made to 2.7 Å resolution, using the oscillation method. The three heavy-atom derivatives were methyl mercury (two sites, half occupied, 3 Å apart), uranyl acetate (single fully occupied site) and chloroplatinite PtCl42? (three sites of differing occupancy). The results were used to compute an electron density map at 2.7 Å resolution, which shows the monomer as a unit of about 60 Å × 60 Å × 40 Å. The maximum dimension of the dimer is about 130 Å. Most of the polypeptide chain has been traced uniquely. It includes five α-helices more than 12 Å long and several shorter helices. A six-stranded pleated-sheet structure lies in the centre of each subunit. The catalytic site of the enzyme is believed to be adjacent to the mercury-binding group.  相似文献

The crystals of ribonuclease St, the extracellular ribonuclease from Streptomyces erythreus, have been obtained from (NH4)2SO4 solution with acetate buffer (pH 4.1). The crystals belong to a monoclinic space group C2 with dimensions a = 88.4 A?, b = 33.0 A?, c = 69.0 A?, β = 98.4 °. There are two protein molecules per asymmetric unit. A pencillin-sensitive enzyme, the exocellular dd-carboxypeptidase-transpeptidase from Streptomyces R61, has been crystallized from polyethylene glycol (Mr = 6000 to 7500) solution at pH 7·6. X-ray examination of the orthorhombic crystals shows the space group is P212121, with unit cell dimensions a = 51·1 A?, b = 67·4 A?, and c = 102·9 A?. With four molecules of molecular weight 38,000, the A?3/dalton ratio for the cell is 2·33. The crystals are stable to irradiation for 75 hours and are suitable for structure analysis to at least 2·4 Å resolution. The radius of gyration of the molecule in solution at pH 6.8 is 20.8 Å.  相似文献

Crystals of crambin, a plant seed protein of molecular weight 5000, diffract X-rays strongly to the interplanar spacing limit of 0.88 Å. These diffraction data should allow a definition of atomic structure that is on a par with that typically obtained from crystals of small organic molecules. The crystals are in space group P21 and have unit cell dimensions a = 41.1 A?, b = 18.7 A?, c = 22·7 A?, and β = 90.6 °. The asymmetric unit contains one protein molecule.  相似文献

The esteroproteolytic enzyme, a serine protease from porcine pancreas, has been crystallized and characterized by X-ray diffraction. Two closely related crystal forms were observed. The unit cell dimensions of the first form are a = 59.2 A?, b = 96.4 A? and c = 47.4 A?, space group P21212 with one molecule (mol. wt 30,000) per asymmetric unit. The unit cell dimensions of the second form are a = 59.2 A?, b = 96.4 A? and c = 94.8 A?, space group P212121. Mercury derivatives of enzyme inhibitors were used to obtain multiple isomorphous heavy-atom substituents suitable for phase determination.  相似文献

The structural protein, δ-crystallin, has been purified and crystallized from adult turkey lens. The crystals are orthorhombic and normally belong to space group P21212 with unit cell dimensions a = 99.9(2) A?, b = 133.4(3) A? and c = 69.1(2) A?. This corresponds to two molecules of molecular weight approximately 200,000 per unit cell. A second crystal form has also been found in which b and c increase to 135.4(3) Å and 140.0(3) Å. respectively, indicating four molecules per unit cell.  相似文献

Tyrocidine A was crystallized from 2-methyl-2,4-pentanediol and water, or methanol, to yield crystals that are large enough for X-ray diffraction studies. Four crystals were examined; three were in equilibrium with mother liquor and the fourth was air-dried. They belong to the rhombohedral space group R32. Parameters of the hexagonal cell of the fully solvated crystals vary slightly and are approximately a = 34 A? and c = 50 A?. The asymmetric unit consists of one molecule of tyrocidine and several molecules of 2-methyl-2,4-pentanediol. Air-dried crystals appear to contain about half the number of solvent molecules. Three-dimensional X-ray diffraction data showing a maximum resolution of s = (1.7 A?)?1 have been recorded.  相似文献

Uteroglobin is a steroid-binding protein of 15,800 molecular weight, composed of two chemically identical subunits which, in the oxidized form, are covalently linked by disulphide bridges. Large crystals have been grown from ammonium sulphate solutions by vapour diffusion as well as by equilibrium dialysis. The crystals are very stable under X-rays, diffract to at least 2.2 Å resolution and belong to space group P21. The unit cell, with dimensions a = 43.3 A?, b = 31.1 A?, c = 34.5 A?, and β = 90.7 °, contains two dimeric molecules. The crystals exhibit a prominent pseudo symmetry corresponding to space group P21212 which indicates that the two subunits should be structurally nearly identical.  相似文献

The lectin from the seeds of Abrus precatorius has been crystallized and the crystals subjected to study by X-ray diffraction and electron microscopy. Three closely related crystal forms were obtained, of orthorhombic space group P212121 with a = 138 A?, b = 142 A?, and c = 173 A?, of tetragonal space group P41212 with a = b = 136 A?, c = 176 A?, and a twinned intermediate of the first two. From electron microscopy and two-dimensional spatial filtering of electron micrographs of the crystals, the molecule appears to consist of four similar domains grouped in a roughly planar diamond-shaped arrangement having a local intramolecular dyad axis. The average diameter of the Abrus lectin molecule is 50 to 60 Å and the individual domains appear to have a diameter of about 25 Å.  相似文献

Purification and crystallisation procedures are reported for azurin and cytochrome c′ from Alcaligenes denitrificans and Alcaligenes sp. NCIB 11015. The azurin crystals from A. denitrificans are suitable for high-resolution X-ray structure analysis. They are orthorhombic, space group C2221 (with marked tetragonal pseudo-symmetry), cell dimensions a = 75.0 A?, b = 74.1 A?, c = 99.5 A?, with two molecules per asymmetric unit. The cytochrome c′ crystals from both species are hexagonal, space group P6122 (or P6522), cell dimensions a = b = 54.7 A?, c ~ 185 A?, γ = 120 °, with one subunit (molecular weight 14,000) in the asymmetric unit.  相似文献

Reassembled alfalfa mosaic virus coat protein was partially digested with trypsin to remove the first 26 amino acids (Bol et al., 1974). These particles are empty icosahedral protein shells built with 60 alfalfa mosaic virus protein subunits. This aggregate has been crystallized in two different crystal forms, one of which diffracts X-rays to at least 3.4 Å resolution. The type I crystals (space group P63, a = 200 A?, c = 314 A?) contain two particles per cell separated by 195 Å with each sitting on a 3-fold axis. The type II crystals contain three particles per cell in space group P31or P32 (a = 201 A?, c = 485 A?). Other T = 1 viral particles have very similar diameters.  相似文献

Beta-lytic protease from Myxobacter 495 was crystallized by dialysis with 1 m-sodium chloride and 0.1 m-sodium citrate (pH 5.95). The crystals were rhombic prisms with space group P212121 and unit cell parameters a = 54.1, b = 99.6 and c = 53.9 A?. tRNACUGLeu from E. Coli was purified by column chromatography on benzoylated DEAE-cellulose, followed by hydroxyapatite prepared by an improved method. Crystals obtained by vapour diffusion gave X-ray diffraction out to 7 Å in the hk0 projection and 10 Å in h0?. The space group was P42212 with a = b = 133 A?, c = 66 A? and 8 molecules in the unit cell. Birefringence showed preferred orientation of RNA helical regions in the ab plane.  相似文献

The crystal structure of the predominant hemoglobin component of blood from the sea lamprey, Petromyzon marinus, has been determined by X-ray diffraction analysis. Crystals for this analysis were grown from cyanide methemoglobin V as crystal type D2. These crystals are in space group P212121 and have unit cell dimensions of a = 44.57 A?, b = 96.62 A? and c = 31.34 A?. Isomorphous heavyatom derivatives were prepared by soaking crystals in solutions of Hg(CN)2, K2Hg(CNS)4 and KAu(CN)2. Diffracted intensities to as far as 2 Å spacings were measured on a diffractometer. Phases were found by means of the isomorphous replacements and anomalous scattering, with supplementary information provided by the tangent formula. An atomic model was fitted to the final electron density map in a Richards optical comparator. The lamprey hemoglobin molecule is generally similar in structure to other globins, but differs in many details. Each molecule is in contact with ten neighboring molecules in the crystal lattice. The nature of the binding of the heavy atoms to lamprey hemoglobin has been interpreted.  相似文献

Tonin is an enzyme isolated from the submaxillary glands of old rats. This enzyme is involved in the direct conversion of angiotensinogen or the tetradecapeptide renin substrate to the octapeptide angiotensin II, the agent implicated in hypertension. Large well-formed crystals of tonin were grown from 2.5 m-(NH4)2SO4, 0.01 m-phosphate buffer (pH 6.2) and 1% methylpentanediol. The crystals are trigonal, space group P3121 (P3221) with unit cell dimensions: a = b = 94 A?, c = 66 A?, γ = 120°. The value of Vm = 2.6 A?3/dalton corresponds to one molecule of molecular weight 32,000 per asymmetric unit. Definitive classification of tonin into one of the four proteolytic enzyme classes (carboxyl protease, serine protease, thiol protease or metalloprotease) has not been firmly established.  相似文献

Pseudomonas aeruginosa cytochrome oxidase (nitrite reductase, cytochrome cd) has been crystallized in space group P21212 with cell dimensions a = 122.8 A?, b = 87.2 A?, c = 73.4 A?. Density measurements suggest that the asymmetric unit contains one 63,000 molecular weight subunit of the dimeric molecule. Crystal data agree well with electron microscopy of single molecules. The X-ray pattern extends beyond 2.5 Å resolution, and structure analysis is in progress.  相似文献
