Three new compounds (1–3) and 20 known compounds were isolated from the rhizomes and roots of Sophora tonkinensis, and all the isolates were tested for their inhibitory activity against IL-6 production in HMC-1 cells stimulated by PMA plus ionophore, A23187. Of the tested compounds, compounds 1, 5, 9, and 21 were found to potently inhibit IL-6 production with IC50 values of 1.62, 0.73, 3.01, and 4.02 μM, respectively. 相似文献

Bioassay directed-fractionation led to isolation of 12 compounds from the roots of Bursera tonkinensis Guillaum (Burseraceae), including burselignan, bursephenylpropane, and burseneolignan. Bioassay-guided phytochemical investigation of the EtOAc fraction (ST-EtOAc) from the roots of Sophora tonkinensis resulted in the isolation of a new compound 6aR,11aR-1-hydroxy-4-isoprenyl-maackiain (1), along with 12 known compounds (2–13). The structure of the new compound was established by 1D and 2D NMR, MS data and circular dichroism analysis. Polyprenylated flavonoids 6–9 and 11–13 increased GLUT-4 translocation by the range of 1.35–2.75 folds. Sophoranone (8) exerted the strongest activity with 2.75 folds GLUT-4 translocation enhancement at the concentration of 10 μM. This is the first report of the GLUT-4 translocation activity of the plant Sophora tonkinensis. 相似文献

A simple, rapid and reliable high-performance capillary electrophoresis method has been developed to determine quantitatively the alkaloid content of Sophora tonkinensis, a Chinese herb commonly known as shan-dou-gen. A total of seven quinolizidine alkaloids (cytisine, sophocarpine, matrine, lehmannine, sophoranol, oxymatrine and oxysophocarpine) could be readily separated within 15 min. The running buffer was 50 mM phosphate buffer (pH 2.5) containing 1% hydroxypropyl-beta-cyclodextrin and 3.3% isopropanol in water. The applied voltage was 25 kV, the capillary temperature was 25 degrees C, the detection wavelength was 200 nm and scopolamine butylbromide was used as internal standard. The method was used to analyse the chemical constituents of two commercial alternatives to shan-dou-gen. The alkaloid constituents of authentic shan-dou-gen gave a specific HPCE electropherogram that could be used to distinguish the drug from potential substitutes. Furthermore, the content of oxymatrine and the total content of the seven quinolizidine alkaloids could be used as quantitative markers in order to assess the quality of S. tonkinensis. 相似文献

This study investigated the antibacterial activities of sophoraflavanone G from Sophora flavescens in combination with two antimicrobial agents against oral bacteria. The combined effect of sophoraflavanone G and the antimicrobial agents was evaluated using the checkerboard method to obtain a fractional inhibitory concentration (FIC) index. The sophoraflavanone G+ampicillin (AM) combination was found to have a synergistic effect against S. mutans, S. sanguinis, S. sobrinus, S. gordonii, A. actinomycetemcomitans, F. nucleatum, P. intermedia, and P. gingivalis, whereas the sophoraflavanone G+gentamicin (GM) combination had a synergistic effect against S. sanguinis, S. criceti, S. anginosus, A. actinomycetemcomitans, F. nucleatum, P. intermedia, and P. gingivalis. Neither combination exhibited any antagonistic interactions (FIC index >4). In particular, the MICs/MBCs for all the bacteria were reduced to one-half - one-sixteenth as a result of the drug combinations. A synergistic interaction was also confirmed by time-kill studies for nine bacteria where the checkerboard suggested synergy. Thus, a strong bactericidal effect was exerted through the drug combinations, plus in vitro data suggested that sophoraflavanone G combined with other antibiotics may be microbiologically beneficial rather than antagonistic. 相似文献

以大肠杆ATCC25922、金黄色葡萄球菌ATCC25923及其临床多重耐药分离株ETEC-AD9、MRSA-DF12共4株人体病原细菌为靶标菌,采用琼脂块法从越南槐内生真菌中筛选抗菌活性菌株,并以改良的微量肉汤稀释法测定其代谢产物的最小抑菌浓度;根据形态和分子特征鉴定拮抗菌株的分类学地位。结果表明,菌株TRPH-35的活菌块对各病原细菌的抑菌圈直径与阳性对照相当;其代谢产物对各病原菌的最小抑菌浓度为20~40μg/mL;初步鉴定该菌株为刺盘孢。本研究结果表明,菌株TRPH-35鉴定为刺盘孢,对参试病原细菌均显示强的抗菌活性,具有较大的开发应用潜力。 相似文献

Nine oxygenated cyclohexanoids, speciosins L-T (1-9) as well as a 5H-furan-2-one metabolite, 5′-O-acetylaporpinone A (10), together with known analogs, speciosins A, B, D, E, F, I and K (11-17), and aporpinone A (18), were isolated from a scale-up cultures of the basidiomycete Hexagonia speciosa. Their structures were elucidated by analysis of spectroscopic data, including 1D and 2D NMR. With three different methods, the total flavonoids from the roots of cultivated Gtycyrrhiza pallidiflora Maxim. were determined and compared by ultraviolet-visible spectrophotometry. The results showed that the extract yield of total flavonoids in crude extracts was 4.54% by direct determination method, 4.27% by AI( NO3)3 colorimetry method and 2.09% by AlCl3 colorimetry method. The content of total flavonoids in roots of cultivated G. pallidiflora was only 0.05%. The cultivated G. pallidiflora in Lanzhou is not unfit for the medicine source of flavonoids. 相似文献
