Classifications of Theaceae have usually placed the endangered monotypic genus Apterosperma in tribe Schimeae (x=18), whereas recent molecular phylogenetic evidence supports its transfer to tribe Theeae (x=15). Molecular data have not resolved the phylogenetic position of Apterosperma within Theeae. We investigated the chromosome number and karyotype of Apterosperma in the context of molecular and morphological phylogenetic evidence to provide further insight into the placement of Apterosperma within Theaceae. The chromosome number and karyotype was found to be 2n = 30 = 26m + 4sm, consistent with the transfer of Apterosperma to tribe Theeae. When the chromosome data were incorporated into a data set of 46 other nonmolecular characters, Apterosperma was placed as the first-diverging lineage within the clade comprising tribe Theeae. This supports its placement based on molecular data. The low intrachromosomal asymmetry (type 1A) of Apterosperma, presumably ancestral for the family, is also consistent with this placement. Character optimization strongly supports a base chromosome number of x=15 for tribe Theeae. Because of variable and sometimes conflicting chromosome count reports of species in tribes Schimeae and Stewartieae, the base chromosome number of Theaceae could be either x=15 or 17.  相似文献

为了解扇脉杓兰(Cypripedium japonicum Thunb.)和无距虾脊兰(Calanthe tsoongiana T. Tang et F. T. Wang)的核型,采用根尖压片法对扇脉杓兰和无距虾脊兰的染色体数目和核型进行了研究。结果表明,扇脉杓兰体细胞的染色体数为22,核型公式为2n=2x=22=16m+2sm+2st+2t,染色体相对长度组成为2n=22=2L+6M2+12M1+2S,核不对称系数为60.01%,核型分类为2B型;而无距虾脊兰体细胞的染色体数为40,核型公式为2n=2x=40=28m+10sm+2st,染色体相对长度组成为2n=40=8L+10M2+16M1+6S,核不对称系数为59.84%,核型分类为2B型;两者核型都较为对称。其中,无距虾脊兰的核型为首次报道。这为扇脉杓兰和无距虾脊兰的进化地位和种质保护提供了细胞学证据。  相似文献

中国黄连属6种1变种分属叶掌状三全裂和叶掌状五全裂2个类群,前者包括三角叶黄连、峨眉黄连、云南黄连、黄连和其变种短萼黄连;而分布于滇东南的五裂黄连和产于台湾的五叶黄连则归于后者。采用根尖压片法和卡宝品红染色法,对除五叶黄连外的中国黄连属5种1变种,以及日本黄连进行染色体核型比较分析,从细胞学角度为探讨中国黄连属植物的系统分类提供新的线索和证据。结果表明:(1)五裂黄连(2n=2x=18=2M+16m)、短萼黄连(2n=2x=18=8m+10sm)和日本黄连(2n=2x=18=12m+6sm)的染色体数目和核型均为首次报道。(2)7个材料的染色体基数均为9,除三角叶黄连为三倍体外,其余均为二倍体。(3)叶为掌状三全裂的二倍体种核型一致,为2A型,染色体类型以及不对称系数均很相似;叶为掌状五全裂的五裂黄连与五叶黄连的核型更接近,为1A型,核型特征的共性表明了这2个类群的自然属性。(4)三倍体三角叶黄连的不对称程度较高,核型为3A型,其染色体大小与峨眉黄连最接近。(5)根据核型不对称程度和染色体大小,结合地理分布,推测叶掌状五全裂种为本属的原始类群。  相似文献

为探究黄藤(Daemonoropsjenkinsiana)染色体核型和基因组的大小,采用体细胞染色体常规制片法与显微摄影技术相结合的方法,对黄藤染色体进行了核型分析,同时以番茄(Lycopersicon esculentum)为内标,应用流式细胞术对黄藤叶片基因组大小、DNA含量和DNA倍性进行了测定。结果表明,黄藤茎尖是理想的染色体制片材料;黄藤的染色体数为2n=24,核型公式为K(2n)=1M+17m+5sm+1st,核型类型为2C;核型不对称系数61.20%;黄藤的DNA含量为1.57 pg,基因组大小为1 539.53 Mb,黄藤的DNA倍性为二倍体(2n)。这是首次报道黄藤的核型和基因组大小,为深入开展黄藤属及其近缘属植物的核型和基因组比较分析提供了参考依据。  相似文献

Despite extensive literature on the diversity of karyotypes in Allium is available, no attempt to analyse these data together, within a robust phylogenetic framework, has been carried out so far. Thus, we examined patterns and trends in chromosome evolution across the genus. Based on literature survey, karyo-morphometric features for 207 species belonging to 12 subgenera of Allium were obtained. Included in the data-set were basic chromosome number (x), somatic chromosome number (2n), total haploid (monoploid) chromosome length (THL) and three different measures defining karyotype structure: CVCI, measuring how heterogeneous are centromeres positions in a karyotype, CVCL and MCA, quantifying interchromosomal and intrachromosomal karyotype asymmetry, respectively. Trends in karyotype evolution were analysed by phylogenetic regressions and independent contrasts. Mean karyotypes highlighted differences and similarities in karyotype structure between the 12 subgenera. Further differences were noted when the two parameters for analysing karyotype asymmetry were assessed. In addition, by examining the effects of increasing karyotype dimensions (a proxy for genome size) on karyotype structure and asymmetry, it was shown that in Allium species, the DNA was added proportionally to their arm lengths. Overall, p = 8 and somehow intermediate karyotype asymmetry levels seem to represent plesiomorphic character-states in Allium.  相似文献

The localization of NORs by fluorescent in situ hybridization on metaphase spreads of five diploid (Rosa gigantea Coll., Rosa moschata Herrm., Rosa multiflora Thunb., Rosa rugosa Thunb. and Rosa sempervirens L., 2n=2x=14), one triploid (Rosa chinensis’semperflorens’ Koehne., 2n=3x=21) and one tetraploid (Rosa gallica ’versicolor’ L., 2n=4x=28) ancestral species of modern roses was studied. Two terminal hybridization signals were observed in all diploid species corresponding to a single NOR per genome in these species. Triploid R. chinensis showed three hybridization sites on the short arm of three morphologically similar chromosomes. Six hybridization sites, located at terminal positions of the short arms of three chromosome pairs, were observed in the tetraploid species. These signals corresponded to three pairs of NORs and all of them were located in chromosome pairs of different size. These observations, together with the analysis of meiotic pairing in PMCs, support the view that our specimen of R. chinensis is an autotriploid and that R. gallica’versicolor’ has an alloploidy nature.  We have isolated and sequenced 263 microsatellite-containing clones from two small insert libraries of Actinidia chinensis enriched for (AC/GT) and (AG/CT) repeats, respectively. Primer pairs were designed for 203 microsatellite loci and successfully amplified from both plasmid and A. chinensis genomic DNA. In this paper we report the sequences of 40 primer pairs for which we have demonstrated Mendelian segregation in the progeny from controlled crosses. The polymorphism of ten microsatellites of each type was evaluated in four diploid and six tetraploid genotypes of A. chinensis. All microsatellites proved to be polymorphic, the number of alleles per locus detected in polyacrylamide sequencing gels ranging from 9 to 17. The high degree of polymorphism in Actinidia renders these markers useful either for mapping in A. chinensis or for fingerprinting cultivars of both domesticated kiwifruit species (A. chinensis and A. deliciosa). While most primer pairs produced single amplification products, about 20% generated banding patterns consistent with the amplification of two different loci. This supports the hypothesis that diploid species of Actinidia (2n=2x=58) are polyploid in origin with a basic chromosome number x=14/15 and that chromosome duplication may have occurred during the evolution of the genus. Finally, we have assayed the cross-species transportability of primer pairs designed from A. chinensis sequences and have found extensive cross-species amplification within the genus Actinidia; 75% of primer pairs gave successful amplification in the eight species assayed (A. arguta, A. rufa, A. polygama, A. chrysantha, A. callosa, A. hemsleyana, A. eriantha, and A. deliciosa), which are representative of the four sections into which the genus is currently split. Cytogenetic characterization by karyotyping and determination of DNA content by flow cytometry of five species of Chrysolaena (Vernonieae, Asteraceae) was performed. This is the first study of nuclear DNA content realized in the genus. The 2C-values were compared with the ploidy level and the total karyotype length (TKL) of each species. Mitotic analysis revealed a base chromosome number x = 10 for all entities and different ploidy levels, from diploid (2n = 2x = 20) to octoploid (2n = 8x = 80). All species showed bimodal karyotypes composed of metacentric and submetacentric chromosomes. The average chromosome size (ML) varied from 1.86 μm to 2.70 μm, while the TKL ranged from 18.65 μm to 80.55 μm. The intrachromosomal asymmetry index (A1) varied from 0.27 to 0.38, while the interchromosomal asymmetry index (A2) ranged from 0.19 to 0.25. A new cytotype is reported for the first time for C. propinqua. Accessory chromosomes found in C. verbascifolia, C. cognata, C. flexuosa, and C. propinqua are also reported as new.  相似文献

Abstract Cytological characters of four species in Syncalathium (Asteraceae: Lactuceae), a small genus with six identified species endemic to alpine scree of the Sino‐Himalayan region, are surveyed in this report. Three species (Syncalathium pilosum, Syncalathium chrysocephalum, and Syncalathium disciforme) are examined for the first time. Combined with our previous counts, five species have been cytologically investigated from the genus and the results indicated that all species are diploid with the basic somatic chromosome number of x=8. The karyotype asymmetry of Syncalathium souliei is 2A, distinct from the other four species of 1A, and the remaining species are divided into two subgroups with different karyotypes, consistent with their morphological features. The significance of the cytological evolution of Syncalathium is briefly discussed.  相似文献

以2份角堇与4份大花三色堇自交系为试验材料,采用染色体常规压片方法,观察和分析了它们的细胞染色体数目、相对长度、平均臂比等核型指标,以明确两种植物细胞学特点,为分类以及育种提供理论依据。结果表明:(1)2份角堇自交系染色体数目均为2n=2x=26,染色体基数为x=13,染色体核型公式分别为2n=2x=26=8m+12sm+6st、2n=2x=26=4m+16sm+6st,核型不对称系数为67.20%~70.10%,核型分类均属于3B。(2)4份大花三色堇自交系均为四倍体,其中2份(EYO-1-2-1-4、DSRFY-1-1-2)染色体数目为44,核型公式为2n=4x=44=4m+16sm+6st、2n=4x=44=16m+24sm+4st;2份(G10-1-3-1-4、XXL-YB-1-1-1-1)染色体数目为48,核型公式分别为2n=4x=48=8m+20sm+20st、2n=4x=48=4m+36sm+8st,核型不对称系数为66.74%~71.77%,核型分类属于2B、3B。  相似文献

采用常规压片法,对风毛菊属(Saussurea)5种植物的染色体数目和核型类型进行分析。结果表明:大耳叶风毛菊(S.macrota)核型公式为2n=2x=26=10m+12sm+4st,属2A型;长梗风毛菊(S.dolichopoda)核型公式为2n=2x=26=14m+8sm+4st,属2A型;川陕风毛菊(S.licentiana)核型公式为2n=2x=28=12m+16sm,属2B型;杨叶风毛菊(S.populifolia)核型公式为2n=2x=28=6m+18sm+4st,属2B型;尾叶风毛菊(S.caudata)核型公式为2n=2x=30=14m+14sm+2st,属2A型。这5种风毛菊属植物中,除大耳叶风毛菊染色体数目和核型类型与前人报道的一致外,其余4种植物的染色体数目和核型类型均为首次报道,并在川陕风毛菊中发现1对B染色体。  相似文献

采用植物细胞学研究方法对主要采自中国西南地区横断山的6种植物:扁核木(Prinsepia utilis)、小叶金露梅(Potentilla parvifolia)、峨眉繁缕(Stellaria omeiensis)、金铁锁(Psammosilene tunicoides)、山卷耳(Cerastium pusillum)和独尾草(Eremurus chinensis)进行染色体数目和核型研究。研究表明:(1)6个种的核型公式和不对称性如下:扁核木2n=2x=30=1M+20m+9sm,2A;小叶金露梅2n=2x=28=21m+7sm,2B;峨眉繁缕2n=8x=72=1M+71m,1B;金铁锁在同居群下有倍性变化,2n=2x=14=14m,1A和2n=4x=28=28m,1A;山卷耳2n=2x=24=24m(2sat),1A;独尾草2n=2x=14=2m+2sm+8st+2t,4B。(2)讨论了染色体资料在这些物种分类和系统上的意义,支持扁核木为李亚科下的扁核木属;位于委陵菜属木本系的小叶金露梅主要以二倍体和四倍体为主,而草本系的委陵菜属植物多为多倍体。(3)该研究首次报道峨眉繁缕为八倍体,是繁缕属发现的最高倍性的物种;对比金铁锁属与蝇子草属的染色体研究发现,金铁锁属可能是由它们的共同祖先通过非整倍化产生;该研究首次发现山卷耳存在染色体数目2n=24的情况,且有1对随体;独尾草的研究进一步证明该物种可能为二型核。  相似文献

Protoplasts isolated from cotyledon-derived callus of Actinidia chinensisPlanch. var. chinensis (2N=2x=58) were fused with mesophyll protoplasts of Actiniadia kolomikta(Maxim. et Rupr.) Maxim (2N=2x=58) using a PEG method. Plantlets were regenerated from the fusion product clone 11. RAPD analyses, chromosome numbers of root tip cells and fluorescence peak position of leaf nuclei confirmed that clone 11 was an interspecific somatic hybrid (2N=4x=116) between A. chinensis and A. kolomikta. The chilling tolerance of the somatic hybrid was tested with in vitro leaves at low temperatures. Based on data of leaf thickness, electroconductivity, proline levels, malondialdehyde content and activity of superoxide dismutase, dendrogram cluster analysis suggested that the interspecific somatic hybrid was similar to A. kolomikta, and might have a higher capacity of cold resistance than A. chinensis.  相似文献

Cytological analyses were performed in nineteen accessions belonging to seven Achillea species. The results indicate that two species, A. filipendulina and A. tenuifolia, are diploid (2n = 2x = 18) and three species, A. bieberstinii, A. pachycephala, and A. aucheri are tetraploid (2n = 4x = 36). For two latter species, the chromosome numbers are new. Two ploidy levels of 2n = 6x = 54 and 2n = 8x = 72 in A. millefolium and two ploidy levels of 2n = 2x = 18 and 2n = 4x = 36 in A. santolina species were found. Cluster analysis based on chromosomal characteristics and karyotype asymmetry indices clustered the studied genotypes into three major groups. The first group included the diploid species (A. filipendulina, A. santolina and A. tenufolia), while the second group comprised tetraploid species (A. santolina, A. aucheri and A. pachycephala). A. millefolium (hexaploid, octaploid) and A. bieberstinii (tetraploid) were classified in the third group. Total form percentage (TF%) in groups 1, 2 and 3 were 42.03, 42.15 and 41.08, respectively. Group 1 possessed the highest average of symmetry index (S% = 70.8). Stebbins classification method grouped all accessions in class A. Moreover, group 1 (diploid level species), had the most symmetric karyotype from point of view of centromere and chromosome length. Therefore, it can be concluded that the genotypes belonging to group 1 are the earliest evolutionary form.  相似文献

This study focuses on karyological investigation of four taxa of Dianthus (Caryophyllaceae) including D. crossopetalus, D. orientalis subsp. stenocalyx, D. orientalis subsp. gorganicus, and Dianthus sp. distributed in Northeast of Iran. The karyotype asymmetry/symmetry was evaluated using two methods: 1) CVCL (coefficient of variation of chromosome length) and CVCI (heterogeneity of the centromeric index); and 2) MCA (mean centromeric asymmetry). The karyotype asymmetry was also used to investigate the relationships among the taxa. Results obtained from the current study revealed that there are two different ploidy levels (2n = 2x = 30 and 2n = 4x = 60) among the investigated taxa. The indices CVCL and MCA described karyotype asymmetry correctly based on variation in chromosome length. Diagram of CVCL vs. MCA seems to be appropriate for karyological delimitation and taxonomic relationships among the Dianthus taxa under study. (© 2014 WILEY‐VCH Verlag GmbH & Co. Chromosome numbers are determined from 37 populations attributed to 22 taxa of JapaneseArisaema. Of them, chromosome numbers ofA. limbatum var.conspicuum (2n=26),A. minus (2n=26),A. nambae (2n=28) andA. seppikoense (2n=26) are determined for the first time. New chromosome numbers, 2n=26, are reported forA. aequinoctiale, A. limbatum, A. stenophyllum, A. undulatifolium andA. yoshinagae. Three modes of basic chromosome numbers,x=14,x=13 andx=12, occur in JapaneseArisaema. Precise karyotypic comparisons of 20 taxa reveal that taxa withx=14 andx=13 share 26 major chromosome arms and have an obvious chromosomal relationship. One of two submeta-centric chromosomes inx=13 corresponds to two telo-centric chromosomes inx=14. InA. ternatipartitum with 2n=6x=72, ten out of 12 basic chromosomes are the most similar in size and arm ratio with larger ten chromosomes ofA. ringens among JapaneseArisaema examined. A basic chromosome number ofx=14 is the commonest in the genusArisaema and the remaining basic chromosome numbers,x=13 andx=12, seem to be derived through dysploidal reduction by translocating large segments of major arm of telo-centric chromosome onto other minor arm of telo-centric followed by loss of the remainings including a centromere, and by loss of two telo-centrics fromx=14, respectively. Some systematic problems of JapaneseArisaema are discussed based on new cytological data.Arisaema hatizyoense, A. minus andA. nambae are accepted as independent species.  相似文献

The genus Hypochaeris (Asteraceae, Lactuceae) contains ten species in Europe, three in Asia, and approximately 50 in South America. Previous cytotaxonomic studies have shown two groups of taxa: (1) European species with different basic chromosome numbers and differentiated karyotypes, and (2) South American species with x=4 and uniform asymmetric and bimodal karyotypes. Karyotypic data are synthesized for South American species of Hypochaeris with new information for six Chilean species: H. acaulis, H. apargioides, H. palustris, H. spathulata, H. tenuifolia and H. thrincioides. Four main groups can be distinguished based on presence and localization of secondary constrictions – SCs (bearing Nucleolar Organizer Regions – NORs) on chromosomes 2 and 3, and 18S–25S and 5S rDNA loci number, localization, and activity. We propose karyotypic evolution of South American Hypochaeris (x=4) from H. maculata-like (x=5) European ancestors. The original South American karyotype would have possessed two SCs, one on the long arm of chromosome 2, and the other on the short arm of chromosome 3 (in terminal position). Further evolution would have involved inversion within the short arm of chromosome 3 and inactivation/loss of the SC on chromosome 2.  相似文献

首次报道了华南地区兔儿风属(Ainsliaea DC.)(菊科-帚菊木族Asteraceae-Pertyeae)3种植物共4个居群的染色体数目和核型。其中长穗兔儿风(A.henryi Diels)的染色体数目为2n=24,核型公式为2n=16m+8sm;三脉兔儿风(A.trinervis Y.C.Tseng)的染色体数目为2n=26,核型公式为2n=16m+10sm;莲沱兔儿风(A.ramosa Hemsl.)2个居群的染色体数目均为2n=26,核型公式为2n=26=22m+4sm。所有居群的染色体由大到小逐渐变化,核型没有明显的二型性。这些结果表明兔儿风属植物确有x=12和x=13两个基数,其中x=13可能是该属的原始基数。  相似文献

Twenty species of shrubbyOxalis species native in South America were arranged in six groups according to their karyotype similarities. The first group includes more than half of the species studied, all with 2n = 12 relatively small metacentric chromosomes. The rest of the species from the other groups has different basic chromosome numbers, and exhibits various degrees of chromosome asymmetry and size increase. These latter groups apparently have specialized to habitats with low light intensity such as the forest undergrowth in contrast to the frist group whose members live in open areas (i.e., grasslands, savannas etc.).  相似文献
