Hydrazones of a 6-hydrazinonicotinyl-modified cyclic peptide IIb/IIIa receptor antagonist were prepared in order to protect the hydrazine moiety from reaction with trace aldehyde and ketone impurities encountered during the process of manufacturing and compounding lyophilized kits used in radiolabeling with (99m)Tc. Hydrazones were prepared by either a direct reaction of the 6-hydrazinonicotinyl-modified cyclic peptide with carbonyl compounds or by conjugation of the cyclic peptide with hydrazones of succinimidyl 6-hydrazinonicotinate. Stability of the hydrazones was evaluated by treatment with formaldehyde. Hydrazones derived from simple aliphatic aldehydes underwent an exchange reaction with formaldehyde, while hydrazones of aromatic aldehydes and ketones provided the greatest level of stability when challenged with formaldehyde. We have been successful in protecting 6-hydrazinonicotinyl-modified cyclic peptides from reacting with formaldehyde, while still allowing sufficient reactivity for radiolabeling with (99m)Tc. The hydrazones of succinimidyl 6-hydrazinonicotinate are convenient and general reagents for forming 6-hydrazinonicotinyl conjugates with amino-functionalized bioactive molecules.  相似文献

Methods for the determination of carbonyl compounds of biological origin by high-performance liquid chromatography were improved by the use of new fluorescent derivatizing agents. Eight fluorescent hydrazides were either synthesized or obtained commercially and compared to dansyl hydrazine (1-dimethylaminonaphthalene-5-sulfonylohydrazide). Four of the compounds yielded carbonyl hydrazones with a higher relative fluorescence quantum yield than dansyl hydrazine in acetonitrile:water mixtures. Darpsyl hydrazide [(3-phenylpyrazoline-1-yl)-4-phenylsulfonylohydrazide] and apmayl hydrazide [N-(2-aminophenyl-6-methylbenzthiazole)-acetylohydrazide] both yielded an increase of greater than 20-fold in sensitivity over dansyl hydrazine in determinations of abscisic acid and jasmonic acid from plant tissues. Different hydrazides and derivatizing conditions were found to be optimum for the determination of different carbonyl compounds. Also, a simple method for precolumn purification of the hydrazones of acidic carbonyls was developed to remove contaminants arising during derivatization and from the tissue source.  相似文献

Zine(II) forms highly fluorescent chelates with the aroyl hydrazones of furfural and 5-hydroxymethylfurfural, with the latter being more fluorescent than the former. The choice of aromatic acid hydrazide (aroyl hydrazine) as analytical reagents for furfurals was examined; 4-toluenesulfonic acid hydrazine was the most sensitive reagent examined, giving a uv fluorescence 10 times as sensitive as 4-hydroxygenzoic acid hydrazide (PAHBAH). More convenient visible fluorescence was given by PAHBAH and related compounds, and these are capable of detecting less than 500 pmol 5-hydroxymethylfurfural. The condensation reaction is complete in 30 mm ethanolic hydrochloric acid within 2 min at 60°C, and the stable product forms the fluorescent chelate on mixing with a zinc-diethanolamine solution in ethanol.  相似文献

Human leukocyte and lymphoblastoid (Namalva) interferons were purified by a modified acid ethanol extraction procedure and chromatography of the dilute ethanolic interferon solution on Blue Sepharose. A method is described for determining carbonyl groups that is especially suitable for use with proteins and peptides. It involves the determination of the extinction at 370nm of a sample solution after adding 2,4-dinitrophenylhydrazine. The reaction of 2,4-dinitrophenylhydrazine with pyruvoylglycine and with transaminated ribonuclease T(1) is presented; the isolation of protein hydrazones is discussed.  相似文献

The present study showed that the ethanolic extracts from the bark of Acacia confusa exhibited a strong antioxidant activity. Among all the fractions from ethanolic extracts of bark, the EtOAc soluble fraction exhibited the best antioxidant performance. Furthermore, following by CC and HPLC, 11 pure phenolic compounds were isolated and identified from the ethanolic extracts.  相似文献

An efficient, simple, and small-scale procedure for isolating functional ribonucleic acid (RNA) was successfully applied to many different tissues of grape and apple. These woody plants are rich in polyphenolic compounds and polysaccharides that could impair the RNA extraction. The method chosen is based on the use of hot borate buffer at alkaline pH supplemented with several adjuvants and followed by selective precipitations. Starting with only 0.4 g of fresh tissue and working with small tubes (2 mL), we were able to obtain good yields of high-quality RNA suitable for further applications. The procedure can be proposed for many applications, and it is particularly highly recommended when isolating RNA from a large number of samples.  相似文献

The butanol extraction method of Morton (1950), a routine step in enzyme purification, is discussed with special reference to a hydrophobic form of alkaline phosphatase from human liver tissue. This form slowly precipitates from butanol-extracted liver tissue homogenates stored at 4 degrees C. Furthermore, it is lost when acetone precipitation is applied as a purification procedure. The soluble form in liver tissue is shown to have a higher relative hydrophobicity than the serum liver/bone isoenzyme. The use of sodium cholate in the isolation of the hydrophobic form produces an artefact in isoelectric focusing, which can be abolished by dialysis prior to focusing.  相似文献

Additional extraction of biologically active compounds from a chaga shred has been carried out after completion of the aqueous extraction. The extraction was carried out using ethanol. Peroxidation of lipids produces low-molecular-weight carbonyl compounds, which are reactive with biological nucleophiles. The analysis of these compounds is often difficult. A multicomponent method for the determination of 11 of them in biological samples is reported. The samples are subjected to a pretreatment-derivatization procedure followed by gas chromatographic analysis with either electron-capture detection (ECD) or mass-selective detection (MSD) in the selected-ion monitoring mode. The procedure involves derivatization of the analyte with 2,4,6-trichlorophenylhydrazine, extraction with n-hexane, and separation of the derivatization products on a nonpolar gas chromatographic column. The concentration of the derivatization reagent, pH, reaction time, temperature, and presence of extraneous ions were investigated to determine the optimal derivatization conditions. Under these conditions, the method allows for the selective detection of low-molecular-weight carbonyl compounds at femtomole levels in several biological materials such as plasma, urine, and bovine serum albumin without interferences. The limits of detection were in the ranges 0.01-0.2 microM for ECD and 0.15-1.5 microM for MSD. The mean procedural recoveries obtained during the method validation were within the range 85-95% and the intra- and interassay standard deviations do not exceed 4.6 and 6.1%, respectively.  相似文献

The results of the comparative analysis of LPS isolated by different methods of extraction from the cultures of several Brucella species differing in their virulence are presented. Purified LPS preparations have been obtained from Brucella virulent and vaccine strains by using such methods as water-phenol extraction, Boivin's method, mild alkaline hydrolysis of the antigen according to Boivin's procedure. The presence of certain relationship between the method used for the extraction of Brucella LPS to be compared and their chemical composition, immunological characteristics and serological activity has been established. As shown in this investigation, in the process of the preparation of a highly sensitive diagnosticum for the passive hemagglutination test the use of LPS obtained from Brucella virulent strains, but not from the vaccine strain, by the method of mild alkaline hydrolysis according to Boivin's procedure is expedient. The data presented in this work indicate that the soluble complex of lipid A obtained from Brucella LPS has been found to possess serological activity. The results of the study of the serological properties of lipid A indicate that the lipid component may also play a certain role in the manifestation of the serological activity of Brucella LPS.  相似文献

Non-structural phenolic compounds perform a variety of functions in plants, including acting as antioxidants. We describe a microplate-adapted colorimetric total phenolics assay that utilizes Folin-Ciocalteu (F-C) reagent. The F-C assay relies on the transfer of electrons in alkaline medium from phenolic compounds to phosphomolybdic/phosphotungstic acid complexes, which are determined spectroscopically at 765 nm. Although the electron transfer reaction is not specific for phenolic compounds, the extraction procedure eliminates approximately 85% of ascorbic acid and other potentially interfering compounds. This assay is performed in microcentrifuge tubes and assessed in a 96-well plate reader. At least 64 samples can be processed in 1 d.  相似文献

Curdlan is an extracellular polysaccharide produced from soil microorganism Alcaligens faecalis var. 10C3K, and the linear structure consists of β-1,3-glycoside linkages. Curdlan is not soluble in water but it is soluble in alkaline aqueous solution, and we can obtain the gel when curdlan alkaline solution is heated above 60°C or neutralized by acids. In the present study, the gelation mechanism and dispersing structure of curdlan in the alkaline solutions are studied in terms of correlation between the molecular association structure and viscoelastic properties, using static light scattering and rheological measurements. The degree of association for the curdlan molecules in dilute solution increases with decreasing alkaline concentration. The viscoelastic properties also depend strongly on the alkaline concentration. The concentrated curdlan solution shows almost Newtonian flow at high alkaline concentrations and shows a gel-like behavior at low alkaline concentrations. It was elucidated that the molecular association in the dilute solution reflects on the viscoelastic properties of the concentrated solution and that the gelation mechanism is related to the association structure of curdlan molecules. In the case of lower NaOH concentration systems, the molecular association is likely to consist of a hydrophobic core and hydrophilic surface. The gelation mechanism above 60°C is considered to include the dissociation process of the molecular association and reformation of the network structure. © 1997 John Wiley & Sons, Inc. Supercritical fluids (SCFs) are alternative solvents in the field of Green Chemistry that are being developed as advanced separation techniques due to their combined properties such as the penetrability of a gas and the solvent power of a liquid. These characteristics are used in the supercritical fluid extraction (SFE), where compounds of interest can be extracted after mixing a SCF as a solvent with a matrix in a pressurized vessel. Supercritical antisolvent fractionation (SAF) uses the SCF as antisolvent, allowing for the precipitation of insoluble compounds in the SCF-organic solution mixture. An updated overview of SFE and SAF of natural products is presented in this article. Additionally we compare the results of SFE, SAF and organic soxhlet extraction (OSE) of Persea indica, a Macaronesian paleoendemism with strong insecticidal components. The composition of the extracts was analyzed by HPLC-MS, showing in the SFE extracts as major components ryanodol, cinnzeylanol and alkyl-γ-lactones, with their abundance varying with the extraction conditions. On the other hand, the SAF allowed for the fractionation of a liquid solution to give a ryanodol enriched extract (31 % more ryanodol than the initial ethanolic solution). The antifeedant and postingestive effects of these extracts on Spodoptera littoralis showed that the SAF extracts were the most active followed by SFE and OSE and their different effects can be partially explained by their composition.  相似文献

Rapid quantitative extraction of corn endosperm proteins is accomplished by presolubilization of the zeins in 70% ethanol (vv), followed by addition of dilute NaOH solution to the ethanolic slurry. The effectiveness of this technique rests primarily on the rapid solubilization of the zeins in weakly ethanolic dilute NaOH solution and on the prescribed sequence of addition of reagents. Chlorine containing ruthenium bipyridine carbonyl compounds react readily in dilute alkaline solutions under a CO atmosphere affording a poorly soluble and air sensitive product that is suggested to have a polymeric nature. Various analysis methods (MS and TPD) were used in the characterisation of the product. The replacement of the axial chloride ligands in trans(Cl), cis(CO)[Ru(bpy)(CO)2Cl2] and [Ru(bpy)(CO)2Cl]2 is proposed to be the initial step in the polymerisation. The replacement of chlorides in methanolic solution was confirmed by isolating and characterising the dimeric intermediate [Ru(bpy)(CO)2(COOCH3)]2.  相似文献

对黑蚂蚁采用盐酸水解、氢氧化钠溶液提取、盐酸沉淀,以及多种有机溶剂洗涤等工艺手段提取分离、纯化得到黑蚂蚁黑色素。研究发现黑蚂蚁黑色素溶于碱性溶液,不溶于水、酸及有机溶剂;其紫外吸收光谱与红外光谱具有吲哚型黑色素典型光谱性质。利用分光光度法测定了黑蚂蚁黑色素的总抗氧化能力、羟基自由基清除能力及还原力,试验结果表明:黑蚂蚁黑色素有一定的抗氧化能力,但在试验范围内其抗氧化能力比维生素C低。  相似文献

An extraction technique is described for the separation of iron- and calcium-bound phosphate. The iron-bound phosphate is extracted with a 0.05 M solution of Ca-NTA under reducing conditions. Iron, also, is brought into solution, which is an advantage over the NaOH extraction. The calcium-bound phosphate is extracted with a 0.05 M solution of Na-NTA. The NTA also extracts humic compounds. Organic phosphate compounds can be measured in the NTA extracts, unlike the NaOH or H2SO4 extracts such as are used in the (modified) Jackson procedure.Examples of some test compound extractions and of a calcareous sediment are given.  相似文献

A chemiluminescent serotonin assay is described, which is based on a reaction given by some indolic compounds in alkaline dimethyl sulfoxide (DMS).
