Putrescine at 0.6 mM stimulated protocorm-like body growth and polysaccharide synthesis in suspension cultures of Dendrobium huoshanense. The specific growth rate of protocorm-like body increased from 0.047 to 0.056 day−1, and the maximum dry weight and polysaccharide production reached 33.2 and 2.94 g l−1, respectively, while they were 24.6 and 2.12 g l−1, respectively, in the control. The administration of polyamine inhibitor, α-dl-difluoromethylarginine, at 1 mM, decreased protocorm-like body growth and polysaccharide production to 21.4 and 1.76 g l−1, respectively.  相似文献

A strategy that optimization of medium compositions for maximum biomass followed by feeding of sucrose for maximum polysaccharide synthesis was developed for enhancing polysaccharide production in suspension culture of protocorm-like bodies (PLBs) of Dendrobium huoshanense C.Z. Tang et S.J. Cheng. In growth stage, the original half-strength MS medium was optimized with carbon sources, nitrogen sources and metal ion combinations. The effects of different carbon sources on PLBs growth were remarkable and sucrose at 35 g l−1 was the most suitable. Sole nitrate nitrogen of 30 mmol l−1 was the best for PLBs growth. Metal ions (Ca2+, Fe2+, Mn2+ and Zn2+) showed different influences on PLBs growth. The optimal concentration of Ca2+, Fe2+, Mn2+ and Zn2+ was 4.5 mmol l−1, 0.1 mmol l−1, 0.5 mmol l−1 and 0.06 mmol l−1, respectively. In the optimized medium (sucrose, nitrate, Ca2+, Fe2+, Mn2+ and Zn2+ concentration as described above, the other component concentration seen in half-strength MS), 33.9 g DW l−1 PLBs were harvested after 30 days of culture and biomass increase was improved 245% as compared with that in the original medium. In production stage, polysaccharide synthesis was significantly improved by the feeding sucrose. The maximum polysaccharide production (22 g l−1) was obtained in the case of 50 g l−1 sucrose feeding at day 30 of culture, which was about 109-fold higher than that in the original medium without feeding of sucrose.  相似文献

The addition of 1.5 mM putrescine to the culture medium facilitated the conversion of protocorm-like bodies (PLBs) of Dendrobium officinale to shoots. The administration of 1.0 mM of the polyamine inhibitor, α-DL-difluoromethylarginine, decreased the conversion of PLBs to shoots. Compared to the control, the PLBs treated with 1.5 mM putrescine had higher levels of total intracellular free polyamines. The enhanced conversion of PLBs to shoots following the addition of putrescine was accompanied by an increase in the ratio of total cytokinins (CTKs) to indole-3-acetic acid (IAA). Analysis of enzyme activities indicated that the increased endogenous level of total CTKs driven by putrescine was associated with the inhibition of CTK decomposition by CTK oxidase, while the decreased endogenous level of IAA was associated with the promotion of IAA decomposition by IAA oxidase. In addition, putrescine increased the ratio of reduced glutathione to oxidized glutathione in the conversion process of PLBs to shoots.  相似文献

Genetically transformed plants of Cymbidium were regenerated after cocultivating protocorm-like bodies (PLB) with Agrobacterium tumefaciens strain EHA101 (pIG121Hm) that harbored genes for β-glucuronidase (gus), hygromycin phosphotransferase (hpt) and neomycin phosphotransferase II (nptII). PLB of three genotypes maintained in liquid new Dogashima medium (NDM), were subjected to transformation experiments. The PLB inoculated with Agrobacterium produced secondary PLB, 4 weeks after transfer onto 2.5 g L−1 gellan gum-solidified NDM containing 10 g L−1 sucrose, 20 mg L−1 hygromycin and 40 mg L−1 meropenem. Transformation efficiency was affected by genotype and the presence of acetosyringone during cocultivation. The highest transformation efficiency was obtained when PLB from the genotype L4 were infected and cocultivated with Agrobacterium on medium containing 100 μM acetosyringone. Transformation of the hygromycin-resistant plantlets regenerated from different sites of inoculated PLB was confirmed by histochemical GUS assay, PCR analysis and Southern blot hybridization.  相似文献

本研究考察了水杨酸对霍山石斛类原球茎悬浮培养细胞生长、多糖合成、碳代谢的影响,并研究了细胞生长、多糖合成以及蔗糖消耗的动力学。结果表明,水杨酸对霍山石斛类原球茎细胞生长有轻微的抑制作用,但能够显著改善类原球茎对碳源的利用,提高胞内可溶性糖的含量,从而促进多糖的合成,其中以添加100μmol/L浓度的水杨酸效果最好,培养18d时,多糖产量达到3.129g/L,为对照的1.63倍。建立的培养动力学模型能较好地反映水杨酸调控霍山石斛类原球茎悬浮培养过程中细胞生长、多糖合成和蔗糖消耗特性。  相似文献

本研究从铁皮石斛(Dendrobium officinale)根、茎和叶中分离得到内生真菌67株,经形态学和分子生物学鉴定分别归属于16个属,Fusarium和Alternaria为铁皮石斛内生真菌的优势种群。研究发现,铁皮石斛根、茎、叶中内生真菌分布存在较大差别,具有一定的组织差异性,不同石斛植物中内生真菌的存在情况差别较大,具有一定的宿主差异性。本研究丰富了真菌资源,为石斛属药用植物内生真菌资源的开发利用提供了基础理论依据。  相似文献

研究不同培养基和光照条件对铁皮石斛愈伤组织诱导的影响。结果表明,外植体直接接种于培养基上,最适宜培养条件是MS 5.92 g·L-1+2,4-D 5 mg·L-1+IAA 1.5 mg·L-1+KT 0.62 mg·L-1+蔗糖37.5 g·L-1+琼脂0.8% (pH 5.9~6.0),暗培养15 d后再光培养;外植体捣碎后平铺于培养基上,最适宜培养条件是MS 4.74 g·L-1+2,4-D 1 mg·L-1+IAA 1.5 mg·L-1+KT 0.25 mg·L-1+蔗糖30 g·L-1+琼脂0.8% (pH 5.9~6.0),25 ℃持续光培养。  相似文献

用微波消解和马弗炉灰化消解分别处理云南、浙江、霍山三种产地的铁皮石斛干花,再用电感耦合等离子体发射光谱仪(ICP-OES)定量分析多种元素含量,构建一种快速测定石斛干花元素含量的方法。结果表明,微波消解结合ICP-OES测出三产地石斛花存在常量元素K、Ca、Na、Mg和微量元素Se、Co、Cu、Mn、Fe、Cr、Sr、Zn、B,且含量均高于马弗炉消解结合ICP-OES测定结果;有害元素As、Pb、Cd、Hg和潜在有害元素Sb、Ba、Sn、Al、Ni均未检出;内标回收率均在92.5%-110.0%之间,相对标准偏差(RSD)<10%。试验探究出微波消解和ICP-OES的结合不仅效率高,检测成本低,运行稳定且测定结果准确可靠,可用于铁皮石斛干花中多元素分析研究。  相似文献

Dendrobium officinale has been considered over past centuries to be extremely valuable for use as an herbal medicine in South Asian countries. In this work, the chemical profiles of D. officinale from different habitats were systematically characterized using ultra-performance liquid chromatography time-of-flight mass spectrometry (UPLC-Q-TOF-MS) and multivariate analysis. The principal component analysis (PCA), partial least squares discriminate analysis (PLS-DA) and orthogonal partial least squares analysis (OPLS-DA) of UPLC-Q-TOF-MS data displayed an obvious separation. Several flavonoids and terpenoids derivatives contribute to the quantitative chemotypic variation within and between the samples as observed. These findings lead to a better understanding of the phytochemical variation of D. officinale which can aid in quality control of raw material.  相似文献

Proteins and activities of antioxidant enzymes, including polyphenol oxidase (PPO), peroxidase (POX) and catalase (CAT), were evaluated in Dendrobium hookerianum during different stages of development of both protocorms and protocorm-like bodies (PLB) derived from seeds and axillary buds, respectively. The changes in the protein contents and the enzyme activities along with their isozyme patterns were compared between these two culture systems. It was found that the protein contents and the enzyme activities increased steadily over time during different stages of development in both the systems. Protein contents (4.57 mg/g fresh wt.) and activities of POX (21.9 U/mg protein), CAT (9.86 U/mg protein) were observed to be higher in PLB system as compared to protocorm system at stage IV of development. However, although the PPO activity increased gradually till the third stage of development, a decline was observed at stage IV wherein the activity was recorded to be more or less same in both the systems. Also, few proteins (~61, 50, 46, 32, 25, 16, 6 kDa) and new isozymes (POX 7, 8; CAT 2) were expressed only in PLB system of development. In general, high protein content and enzyme activities were detected in PLB system as compared to protocorm system. The results of the present study indicate that few proteins and isozymes could be regarded as specific biochemical markers for different stages of development of this medicinally important orchid.  相似文献

Protocorm-like bodies (PLBs) of Dendrobium candidum Wall. ex Lindl., orchid, were successfully cryopreserved using an encapsulation vitrification method. PLBs were precultured in liquid Murashige and Skoog (MS) medium containing 0.2 mg l−1 α-naphthalene acetic acid and 0.5 mg l−1 6-benzyladenine enriched with 0.75 M sucrose, and grown under continuous light (36 μmol m−2 s−1) at 25 ± 1°C for 5 days. PLBs were osmoprotected with a mixture of 2 M glycerol and 1 M sucrose for 80 min at 25°C and dripped in a 0.5 M CaCl2 solution containing 0.5 M sucrose at 25 ± 1°C and left for 15 min to form Ca-alginate beads (about 4 mm in diameter). Then, these were dehydrated with a plant vitrification solution 2 (PVS2) consisting of 30% (w/v) glycerol, 15% (w/v) ethylene glycol, and 15% (w/v) dimethyl sulfoxide in 0.5 M sucrose, pH 5.8, for 150 min at 0°C. Encapsulated and dehydrated PLBs were plunged directly into liquid nitrogen for 1 h. Cryopreserved PLBs were then rapidly re-warmed in a water bath at 40°C for 3 min and then washed with MS medium containing 1.2 M sucrose for three times at 10 min intervals. Within 60 days, plantlets with the cryopreserved PLBs developed normal shoots and roots, and without any observed morphological abnormalities, were obtained. The survival rate of encapsulated-vitrified PLBs was above 85%. Thus, this encapsulation-vitrification method was deemed promising for cryopreservation of PLBs of D. candidum.  相似文献

Summary Protocorm-like bodies (plbs) of the orchid Dendrobium ‘Sonia’ were obtained from fractionated plb explants on Murashiga and Skoog (1962) medium (MS) supplemented with 4.44 μM 6-benzylaminopurine (BA). The developmental stage of plbs to be encapsulated, concentrations of sodium alginate (2–5%) and calcium chloride (25–100 mM), and concentration of MS salts in the matrix were assessed. Fractionated plbs 13–15 d after culture were at the leaf primordia stage and found suitable for encapsulation studies. The best encapsulation response was observed with 3% sodium alginate upon complexation with 75 mM CaCl2.2H2O. An encapsulation matrix prepared with MS medium (three-quarter strength) supplemented with 0.44 μM BA and 0.54 μM α-naphthaleneacetic acid gave 100% conversion of encapsulated plbs to plants after storage. A viability of >85% of encapsulated plbs of Dendrobium ‘Sonia’ was observed for 75 d at 4°C. Encapsulated plbs of orchids Dendrobium, Oncidium, and Cattleya were stored at 4°C for 75, 60 and 30 d, respectively, with >88% germination. All plants survived potting in media having either wood charcoal pieces alone or in combination with brick pieces.  相似文献

为解决霍山石斛类原球茎液体培养细胞生长缓慢和代谢水平低下的问题,研究了不同浓度的锗(GeO2)对霍山石斛类原球茎增殖、多糖合成及碳氮利用的影响,分析了类原球茎细胞内还原糖、可溶性蛋白质含量、抗氧化酶活性以及细胞氧化还原态的变化。结果表明,适当浓度的二氧化锗(4.0mg/L)显著促进霍山石斛类原球茎的增殖和多糖的积累,最大细胞干重为32.6g/L,最大多糖产量为3.78g/L;显著提高胞内还原糖和可溶性蛋白质含量,超氧化物歧化酶和过氧化氢酶的活性明显升高,而过氧化物酶的活性则有所降低;氧化还原态分析发现,二氧化锗处理的细胞内还原型谷胱甘肽/氧化型谷胱甘肽的值明显提高,谷胱甘肽还原酶活性升高。添加适量的二氧化锗有利于细胞生长和多糖合成。  相似文献

目的探讨铁皮石斛对部分酵母菌、益生细菌和有害菌生长的影响,为铁皮石斛调节微生态平衡提供实验依据。方法通过液体培养比浊法检测铁皮石斛对各供试菌株生长的抑促情况。结果铁皮石斛对白色假丝酵母菌、酿酒酵母菌、汉逊德巴利酵母菌的生长具有明显的促进作用:铁皮石斛对保加利亚乳杆菌和枯草芽胞杆菌等有益细菌的生长也具有明显的促进作用:能够抑制金黄色葡萄球菌、大肠埃希菌和沙门菌等(条件)致病菌的生长:铁皮石斛对产气杆菌、铜绿假单胞菌、嗜热链球菌和地衣芽胞杆菌的生长没有明显的影响。结论铁皮石斛能促进部分有益菌的生长,对部分(条件)致病菌有抑制作用,通过抑制有害菌、促进有益菌调控微生态平衡。  相似文献

目的利用组织培养技术建立铁皮石斛的高效快繁体系,克隆繁殖种苗,为野生居群的恢复提供材料。方法以铁皮石斛腋芽为外植体,研究不同浓度和配比的基本培养基、激素以及天然提取物对腋芽萌发生长,丛生芽的诱导增殖以及壮苗生根的影响,比较不同培养阶段的增殖倍数,筛选各时期的最适培养基。并对试管苗的形态学特征进行了调查。结果1/2MS+NAA 0.2 mg/L+BA 1 mg/L适合腋芽的成活生长。BA与香蕉汁组合有利于丛生芽的诱导增殖,其最适比例为1/2MS+NAA 0.5 mg/L+BA 2.0 mg/L+香蕉汁100 g/L,增殖倍数最大可达14.0,平均增殖倍数为6.4。培养基中添加100 g/L香蕉汁或200 g/L马铃薯汁均有利于壮苗生根。结论铁皮石斛通过腋芽形成丛生芽途径建立高效的快繁技术体系获得优质种苗是可行的。  相似文献

以铁皮石斛当年生茎段为材料,在MS+6-BA 0.1 mg/L+NAA 0.02 mg/L培养基上进行不定芽诱导培养;在MS+6-BA 2.5 mg/L+NAA 0.05 mg/L+AC(活性炭)1g/L培养基上进行丛生芽诱导与增殖培养,50 d为一个继代周期,繁殖系数为5~8;在1/2 MS+6-BA 0.5 mg/L+NAA 0.05 mg/L+椰子100 g/L+AC 1g/L培养基上进行壮苗培养;在1/2 MS+IBA 1.0 mg/L+AC 1g/L培养基上进行生根培养;最后移栽到小颗粒化树皮中,成活率可达95%。  相似文献
