Effects of plant maturity on apparent ruminal synthesis and post-ruminal supply of B vitamins were evaluated in two feeding trials. Diets containing alfalfa (Trial 1) or orchardgrass (Trial 2) silages harvested either (1) early cut, less mature (EC) or (2) late cut, more mature (LC) as the sole forage were offered to ruminally and duodenally cannulated lactating Holstein cows in crossover design experiments. In Trial 1, conducted with 16 cows (569±43 kg of empty BW (ruminal content removed) and 43.7±8.6 kg/day of 3.5% fat-corrected milk yield; mean±SD) in two 17-day treatment periods, both diets provided ~22% forage NDF and 27% total NDF, and the forage-to-concentrate ratios were 53 : 47 and 42 : 58 for EC and LC, respectively. In Trial 2, conducted with 13 cows (588±55 kg of empty BW and 43.7±7.7 kg/day of 3.5% fat-corrected milk yield; mean±SD) in two 18-day treatment periods, both diets provided ~25% forage NDF and 31% total NDF; the forage-to-concentrate ratios were 58 : 42 and 46 : 54 for EC and LC, respectively. Thiamin, riboflavin, niacin, vitamin B6, folates and vitamin B12 were measured in feed and duodenal content. Apparent ruminal synthesis was calculated as the duodenal flow minus the intake. Diets based on EC alfalfa decreased the amounts of thiamin, niacin and folates reaching the duodenum, whereas diets based on EC orchardgrass increased riboflavin duodenal flow. Daily apparent ruminal synthesis of thiamin, riboflavin, niacin and vitamin B6 were correlated negatively with their intake, suggesting a microbial regulation of their concentration in the rumen. Vitamin B12 apparent ruminal synthesis was correlated negatively with total volatile fatty acids concentration, but positively with ruminal pH and microbial N duodenal flow.  相似文献

Corn silage (CS) is associated with a reduction in milk fat content. The fact that CS is constituted of a grain and a forage fraction could explain this effect. This experiment evaluated the effect of grain fraction of CS on rumen fermentation, production performance and milk composition. Earless CS (ECS) was harvested after manually removing corn ears from the plant. Whole CS (WCS) was harvested from the same field on the same day. Eight (four ruminally fistulated) multiparous Holstein cows (84 days in milk) were utilized in a double 4 × 4 Latin square with 21-day periods. Treatments were (dry matter (DM) basis) (1) 23.0% WCS; (2) 12.4% ECS plus 10.6% high moisture corn (HMC) to obtain reconstituted CS (RCS); (3) 23.0% ECS; and (4) 23.0% timothy silage (TS). Diets were formulated to be isonitrogenous and were fed as total mixed ration once a day. DM intake (DMI), milk yield, 4.0% fat-corrected milk (FCM), as well as protein concentration and yield were higher for WCS than ECS. Compared with WCS, cows tended to eat less with RCS, and produced less milk and milk protein. However, yield of FCM was similar between WCS and RCS. Milk fat concentration and yield, as well as the specific ratio of t11 18:1 to t10 18:1 in milk fat did not differ among diets. Milk urea-N tended to be higher for ECS than WCS and TS, whereas ruminal NH3-N was higher with ECS than TS. Rumen pH decreased linearly with time after feeding but was not different between treatments. Higher acetate and lower propionate concentration resulted in greater acetate to propionate ratio with ECS compared with WCS. In conclusion, removing grain fraction from CS decreased milk production and modified rumen fermentation without affecting milk fat concentration and yield. Moreover, despite some differences in DMI and total ruminal volatile fatty acid concentration between WCS and RCS, the restoration of FCM yield, using HMC in RCS diets, to a level of production similar to WCS highlights the importance of energy and nutrients supplied by the grain fraction of CS to support milk yield.  相似文献

Grass silage is typically fed to dairy cows in temperate regions. However, in vivo information on methane (CH4) emission from grass silage of varying quality is limited. We evaluated the effect of two rates of nitrogen (N) fertilisation of grassland (low fertilisation (LF), 65 kg of N/ha; and high fertilisation (HF), 150 kg of N/ha) and of three stages of maturity of grass at cutting: early maturity (EM; 28 days of regrowth), mid maturity (MM; 41 days of regrowth) and late maturity (LM; 62 days of regrowth) on CH4 production by lactating dairy cows. In a randomised block design, 54 lactating Holstein–Friesian dairy cows (168±11 days in milk; mean±standard error of mean) received grass silage (mainly ryegrass) and compound feed at 80 : 20 on dry matter basis. Cows were adapted to the diet for 12 days and CH4 production was measured in climate respiration chambers for 5 days. Dry matter intake (DMI; 14.9±0.56 kg/day) decreased with increasing N fertilisation and grass maturity. Production of fat- and protein-corrected milk (FPCM; 24.0±1.57 kg/day) decreased with advancing grass maturity but was not affected by N fertilisation. Apparent total-tract feed digestibility decreased with advancing grass maturity but was unaffected by N fertilisation except for an increase and decrease in N and fat digestibility with increasing N fertilisation, respectively. Total CH4 production per cow (347±13.6 g/day) decreased with increasing N fertilisation by 4% and grass maturity by 6%. The smaller CH4 production with advancing grass maturity was offset by a smaller FPCM and lower feed digestibility. As a result, with advancing grass maturity CH4 emission intensity increased per units of FPCM (15.0±1.00 g CH4/kg) by 31% and digestible organic matter intake (33.1±0.78 g CH4/kg) by 15%. In addition, emission intensity increased per units of DMI (23.5±0.43 g CH4/kg) by 7% and gross energy intake (7.0±0.14% CH4) by 9%, implying an increased loss of dietary energy with advancing grass maturity. Rate of N fertilisation had no effect on CH4 emissions per units of FPCM, DMI and gross energy intake. These results suggest that despite a lower absolute daily CH4 production with a higher N fertilisation rate, CH4 emission intensity remains unchanged. A significant reduction of CH4 emission intensity can be achieved by feeding dairy cows silage of grass harvested at an earlier stage of maturity.  相似文献

A proteomic approach was used to investigate the effects of the processing method of corn grain and soybean meal on the milk protein expression profile in lactating dairy cows. A total of 12 multiparous Holstein dairy cows were used in a 4×4 Latin square design with a 2×2 factorial arrangement. The primary factors examined were corn (finely ground (FGC) v. steam-flaked (SFC)) and soybean meal (solvent-extracted (SSBM) v. heat-treated (HSBM)), which were used to formulate four diets with the same basal ingredient: 27% FGC and 9% SSBM; 27% SFC and 9% SSBM; 27% FGC and 9% HSBM; and 27% SFC and 9% HSBM. Each period lasted for 21 days. Milk samples were collected on days 18, 19 and 20 of each period. Changes in the milk proteins were assessed by two-dimensional (2D) electrophoresis and ImageMaster 2D Platinum 6.0 software. A total of 13 spots displayed variations in protein spot abundance according to the statistical analysis. These spots were identified by a matrix-assisted laser desorption/ionization-time of flight/time of flight MS. According to the gels, the relative abundance of αs2-casein (CN) fragments was higher in the cows fed the SFC-HSBM than that for SFC-SSBM, whereas β-CN, α-lactalbumin and zinc-alpha-2-glycoprotein fragments were down-regulated in HSBM-fed cows. The relative decrease of β-CN expression was validated by western blot and agreed with the MS data. These results suggested that the method used to process soybean meal modified the synthesis and secretion of milk proteins in lactating dairy cows’ mammary glands.  相似文献

Cobalamin (CBL), the biologically active form of vitamin B12, and its analogs, are produced by bacteria only if cobalt supply is adequate. The analogs differ generally by the nucleotide moiety of the molecule. In CBL, 5,6-dimethylbenzimidazole (5,6-DMB) is the base in the nucleotide moiety. The present study aimed to determine if a supplement of 5,6-DMB could increase utilization of dietary cobalt for synthesis of CBL and change ruminal fermentation, nutrient digestibility, omasal flow of nutrients and ruminal protozoa counts. Eight ruminally cannulated multiparous Holstein cows (mean±standard deviation=238±21 days in milk and 736±47 kg of BW) were used in a crossover design. Cows were randomly assigned to a daily supplement of a gelatin capsule containing 1.5 g of 5,6-DMB via the rumen cannula or no supplement. Each period lasted 29 days and consisted of 21 days for treatment adaptation and 8 days for data and samples collection. Five corrinoids, CBL and four cobamides were detected in the total mixed ration and the omasal digesta from both treatments. The dietary supplement of 5,6-DMB increased (P=0.02) apparent ruminal synthesis of CBL from 14.6 to 19.6 (s.e.m. 0.8) mg/day but had no effect (P>0.1) on apparent ruminal synthesis of the four analogs. The supplement of 5,6-DMB had no effect (P>0.1) on milk production and composition, or on protozoal count, ruminal pH and concentrations of volatile fatty acids and ammonia nitrogen in rumen content. The supplement had also no effect (P>0.1) on intake, omasal flow and apparent ruminal digestibility of dry matter, organic matter, NDF, ADF and nitrogenous fractions. Plasma concentration of CBL was not affected by treatments (P=0.98). Providing a preformed part of the CBL molecule, that is, 5,6-DMB, increased by 34% the apparent ruminal synthesis of CBL by ruminal bacteria but had no effect on ruminal fermentation or protozoa count and it was not sufficient to increase plasma concentrations of the vitamin. Even though the efficiency of cobalt utilization for apparent synthesis of CBL was increased from 2.0% to 2.7% by the 5,6-DMB supplement, this improved efficiency was still very low. Further research is needed to identify the factors affecting efficiency of utilization of cobalt for synthesis of CBL by the bacterial populations in rumen.  相似文献

There are differences in grass-clover proportions and chemical composition between herbage from primary growth (PG) and regrowth (RG) in grass-clover leys. Mixing silages made from PG and RG may provide a more optimal diet to dairy cows than when fed separately. We tested the hypotheses that increasing dietary proportions of grass-clover silage made from RG compared with PG would increase digestion rate of potentially degradable NDF (pdNDF), and increase ruminal accumulation of indigestible NDF (iNDF). Eight rumen cannulated Norwegian Red cows were used in two replicated 4×4 Latin squares with 21-day periods. Silages were prepared from PG and RG of an organically cultivated ley, where PG and RG silages were fed ad libitum in treatments with RG replacing PG in ratios of 0, 0.33, 0.67 and 1 on dry matter basis in addition to 8 kg concentrate. We evaluated the effect of the four diets with emphasis on rumen- and total tract fiber digestibility. Increasing RG proportions decreased silage intake by 7%. Omasal flow of pdNDF decreased, whereas iNDF flow increased with increasing RG proportions. Increasing RG proportions decreased rumen pool sizes of NDF and pdNDF, whereas pool sizes of iNDF and CP increased. Increasing RG proportions increased digestion rate of NDF, which resulted in greater total tract digestion of NDF. Pure PG diet had the highest calculated energy intake, but the improved rumen digestion of NDF by cows offered 0.33 and 0.67 of RG leveled out milk fat and protein yields among the three PG containing diets.  相似文献

ABSTRACT

The study evaluated the effects of soybean oil (SO) and dietary copper levels on nutrient digestion, ruminal fermentation, enzyme activity, microflora and microbial protein synthesis in dairy bulls. Eight Holstein rumen-cannulated bulls (14 ± 0.2 months of age and 326 ± 8.9 kg of body weight) were allocated into a replicated 4 × 4 Latin square design in a 2 × 2 factorial arrangement with factors being 0 or 40 g/kg dietary dry matter (DM) of SO and 0 or 7.68 mg/kg DM of Cu from copper sulphate (CS). The basal diet contained per kg DM 500 g of corn silage, 500 g of concentrate, 28 g of ether extract (EE) and 7.5 mg of Cu. The SO × CS interaction was significant (p < 0.05) for ruminal propionate proportion and acetate to propionate ratio. Dietary SO addition increased (p < 0.05) intake and total tract digestibility of EE but did not affect average daily gain (ADG) of bulls. Dietary CS addition did not affect nutrient intake but increased (p < 0.05) ADG and total tract digestibility of DM, organic matter, crude protein and neutral detergent fibre. Ruminal pH was not affected by treatments. Dietary SO addition did not affect ruminal total volatile fatty acids (VFA) concentration, decreased (p < 0.05) acetate proportion and ammonia N and increased (p < 0.05) propionate proportion. Dietary CS addition did not affect ammonia N, increased (p < 0.05) total VFA concentration and acetate proportion and decreased (p < 0.05) propionate proportion. Acetate to propionate ratio decreased (p < 0.05) with SO addition and increased (p < 0.05) with CS addition. Dietary SO addition decreased (p < 0.05) activity of carboxymethyl cellulase, cellobiase and xylanase as well as population of fungi, protozoa, methanogens, Ruminococcus albus and R. flavefaciens but increased (p < 0.05) α-amylase activity and population of Prevotella ruminicola and Ruminobacter amylophilus. Dietary CS addition increased (p < 0.05) activity of cellulolytic enzyme and protease as well as population of total bacteria, fungi, protozoa, methanogens, primary cellulolytic and proteolytic bacteria. Microbial protein synthesis was unchanged with SO addition but increased (p < 0.05) with CS addition. The results indicated that the addition of CS promoted nutrient digestion and ruminal fermentation by stimulating microbial growth and enzyme activity but did not relieve the negative effects of SO addition on ruminal fermentation in dairy bulls.  相似文献

Two experiments were conducted to evaluate the effects of Bacillus subtilis natto, which was initially isolated from fermented soybeans on milk production, rumen fermentation and ruminal microbiome in dairy cows. In Experiment 1, 36 early lactation Chinese Holstein dairy cows (56 ± 23 days in milk) were randomly assigned to three groups: Control, cows were fed total mixed ration (TMR); BSNLOW, TMR plus 0.5 × 1011 colony-forming units (cfu) of B. subtilis natto/cow per day; and BSNHIGH, TMR plus 1.0 × 1011 cfu of B. subtilis natto/cow per day. During the 70-day treatment period, daily milk production and daily milk composition were determined in individual cows. The results showed that supplementing dairy cows with 0.5 × 1011 and 1.0 × 1011 cfu of B. subtilis natto linearly increased (P < 0.01) milk production (25.2 and 26.4 kg/day v. 23.0 kg/day), 4% fat-corrected milk (27.3 and 28.1 kg/day v. 24.2 kg/day), energy-corrected milk (27.3 and 28.2 kg/day v. 24.2 kg/day), as well as milk fat (1.01 and 1.03 kg/day v. 0.88 kg/day), protein (0.77 and 0.82 kg/day v. 0.69 kg/day) and lactose yield (1.16 and 1.22 kg/day v. 1.06 kg/day) but decreased milk somatic cell counts (SCC) by 3.4% to 5.5% (P < 0.01) in BSNLOW and BSNHIGH treatments compared with Control. In Experiment 2, four rumen-cannulated dairy cows were fed the basal diet from 1 to 7 days (pre-trial period) and rumen samples were collected on days 6 and 7; the same cows then were fed 1.0 × 1011 cfu/day B. subtilis natto from days 8 to 21 (trial period) and rumen samples were collected on days 20 and 21. B. subtilis natto was discontinued from days 22 to 28 (post-trial period) and rumen samples were collected on days 27 and 28. Compared with the pre- and post-periods, ruminal pH decreased by 2.7% to 3.0% during the trial period (P < 0.01), whereas ammonia nitrogen (NH3-N), total volatile fatty acids and molar proportion of propionate (P < 0.01) and valerate (P < 0.05) increased. Molar proportion of acetate decreased and the acetate to propionate ratio was lower (P < 0.01) during the trial period. However, no differences for 24-h in sacco dry matter digestibility were detected among different periods (treatments) though NDF digestibility was reduced in the trial and post-trial periods (P < 0.01). Compared with pre-trial period, total ruminal bacteria, proteolytic and amylolytic bacteria in rumen enumerated by culture methods increased by 15.0%, 16.2% and 11.7%, respectively (P < 0.01) but protozoa decreased to 5.35 log10 cfu/ml (P < 0.01) during the trial period. These results demonstrate that B. subtilis natto improves milk production and milk components yield, decreases SCC and promotes the growth of total ruminal bacteria, proteolytic and amylolytic bacteria, which indicate that B. subtilis natto has potential to be applied as a probiotic for dairy cows.  相似文献

Cellulolytic micro-organisms are potent silage inoculants that decrease the fibrous content in silage and increase the fibre digestibility and nutritional value of silage. This study aimed to evaluate the effects of Bacillus subtilis CCMA 0087 and its enzyme β-glucosidase on the nutritional value and aerobic stability of corn silage after 30 and 60 days of storage. We compared the results among silage without inoculant (SC) and silages inoculated with B. subtilis 8 log10 CFU per kg forage (SB8), 9 log10 CFU per kg forage (SB9) and 9·84 log10 CFU per kg forage + β-glucosidase enzyme (SBE). No differences were observed in the levels of dry matter, crude protein and neutral detergent fibre due to the different treatments or storage times of the silos. Notably, the population of spore-forming bacteria increased in the SB9-treated silage. At 60 days of ensiling, the largest populations of lactic acid bacteria were found in silages treated with SB8 and SBE. Yeast populations were low for all silages, irrespective of the different treatments, and the presence of filamentous fungi was observed only in the SBE-treated silage. Among all silage treatments, SB9 treatment resulted in the highest aerobic stability.  相似文献

Twenty-four lactating Holstein dairy cows (12 first lactation and 12 multiparous; day in milk = 11 ± 5 days) were allotted to a randomised complete block design in a 2 × 3 factorial with four replicates per treatment to evaluate the effects of two methods of alfalfa feeding (dry and reconstituted to achieve a theoretical dry matter (DM) content of 350 g/kg) and three geometric mean (GM) particle sizes of alfalfa (9.13, 4.51 and 1.20 mm) on performance of dairy cows for a period of 28 days. Diets were offered for ad libitum intake as total mixed rations (TMR). The GM particle size, its standard deviation, and the values of physical effectiveness factor of alfalfa and TMR decreased as alfalfa particle size decreased. Reduction of particle size and reconstitution of alfalfa increased the bulk density and the functional specific gravity of alfalfa and rations. Reduction of particle size decreased insoluble dry matter, water-holding capacity, and hydration rate of alfalfa. As particle size decreased, the amount of physically effective NDF in the ration (g/kg) decreased but the daily intake of physically effective NDF (kg/day) increased. Reduction of particle size and reconstitution increased dry matter intake (DMI) and ruminal passage rate, but reduced NDF and ash digestibilities, ruminal pH, N-NH3, milk fat, total chewing activity, rumination and eating time, total and ruminal mean retention time, and time delay of marker. Increased functional specific gravity, from reduced forage particle size and the reconstitution of alfalfa, was the most important factor influencing DMI, milk composition, and chewing activity.  相似文献

The objective of this study was to investigate the intestinal microbiota of growing kittens fed moderate- or high-protein diets using DNA-based qualitative and quantitative techniques. Kittens were weaned to a high-protein (HP; n = 7) or moderate-protein (MP; n = 10) diet at 8 weeks of age. Fresh faecal samples were collected at 8, 12, and 16 weeks of age. DNA was extracted and quantitative PCR used to quantify Bifidobacterium, Lactobacillus, Clostridium perfringens, and Escherichia coli concentrations. Denaturing gradient gel electrophoresis was performed to create a dendrogram and unrooted trees using Bionumerics 5.0 to identify similarity due to litter, age, or diet. Kittens fed HP had lower (p = 0.02) Bifidobacteria and Lactobacillus counts than MP-fed kittens. E. coli was lower (p = 0.02) in HP-fed kittens and tended to be affected by age (p = 0.09). Kittens were clustered by litter at 8 weeks of age, and then clustered by diet at 12 and 16 weeks of age. Our data suggest that faecal microbiota of growing kittens change after weaning and that dietary protein concentration affects E. coli, Bifidobacterium, and Lactobacillus populations. The relevance of these data in terms of intestinal health and disease remain to be determined and justifies further study.  相似文献

This study is an exploratory analysis for understanding the effect of a duodenal infusion of an α-linolenic acid (LNA) on the plasma and milk proteome of lactating dairy cows. Four primiparous Holstein cows were fitted with duodenal cannulas and received 0, 100, 200, 300 and 400 g/day of LNA in a two-treatment crossover design. Blood and milk were collected for determination of protein composition by two-dimensional gel electrophoresis. Alteration of protein spots was detected and identified using matrix-assisted laser desorption/ionization time-of-flight/time-of-flight tandem mass spectrometry (MALDI-TOF-TOF MS). Plasma haptoglobin levels, and milk β-casein A2, αs1-casein variant and albumin, did not differ in cows after infusion of 0, 100, 200 and 300 g/day of LNA, but were increased after the cows received duodenal infusion of 400 g/day of LNA. Western blot analysis of haptoglobin expression in plasma confirmed the alterations in protein expression seen using MS. This study demonstrated that infusion of high doses of LNA by duodenal cannula can result in metabolic stress within the bovine intestine and in changes in milk composition.  相似文献

This experiment evaluated different strategies for allocating first-cut grass silages to dry dairy cows that had low body-condition score (BCS) at drying off. A total of 48 moderately yielding Holstein-Friesian cows were used, receiving one of three dietary treatments in the dry period and a single lactation diet based on a flat-rate of concentrates and grass silage ad libitum. Throughout the dry period, one group received a low-digestibility silage (harvested 15 June 1998; LL; metabolisable energy (ME) = 10.3 MJ/kg dry matter (DM)) and a second group received a high-digestibility silage (harvested 9 May 1998, HH; ME = 11.7 MJ/kg DM). A third strategy (LH) offered the low-digestibility silage in the early dry period and the high-digestibility silage in the final 3 weeks before calving. The silages had very different crude protein concentrations (144 and 201 g/kg DM) and intakes were widely divergent (10.1 v. 13.5 kg DM/day) across the dry period. No concentrates were fed during the dry period. Silage quality had a very large effect on liveweight change, with treatment means of 0.32 and 1.75 kg/day for LL and HH, respectively. BCS changes followed a similar pattern, though no cows became over-conditioned and blood metabolites were within normal ranges. Increased silage digestibility in the late dry period led to a substantial increase in milk fat concentration and a smaller increase in milk protein concentration, the latter confined to the first full week of lactation. Depression of milk fat appears related to low blood glucose when dry cows in low body condition are fed at a low level. The LH strategy avoided the tendency for lower milk yields and fat concentration that resulted from feeding the low-digestibility silage until calving. This strategy also avoided the higher calf weights that resulted from feeding the high-digestibility silage in the early dry period.  相似文献

Whole crop third cut alfalfa, brown mid-rib (bmr) corn, and corn were chopped and inoculated with one of four microbial inoculants used. Uninoculated silage was the control treatment. Each crop was ensiled in four mini-silos (1 L glass jars) per treatment. All silos were fermented for 60 days at room temperature (22 °C), and then they were opened and analyzed for fermentation products, fiber constituents and N fractions. A fraction of wet silage was ground with a blender for 30 s. In vitro gas production was measured in 160 ml sealed serum vials at 3, 6, 9, 24, and 48 h using the wet ground silage. At 9 and 48 h, rumen fluid was analyzed for volatile fatty acids (VFA) and microbial biomass yield (MBY). In all the three crops, the four inoculants produced only minor changes in pH and fermentation products during ensiling. Of the variables measured, soluble nonprotein N fractions were the characteristics most often affected by some inoculants. At 9 h incubation, in vitro gas production and VFA did not differ between control and inoculated silages, but MBY did. Among crops, alfalfa and corn silages had higher MBY than did bmr corn silage. Among inoculants, three of the four inoculated silages produced more MBY than did control. At 48 h, alfalfa silage produced higher MBY than did corn or bmr silage, and two of the inoculated silages had more MBY than did the control. There was no inoculant by crop interaction. Results suggest that some silage inoculants are capable of altering rumen fermentation, even in cases where effects on silage fermentation are small, and that this effect may be linked to better preservation of crop protein during ensiling.  相似文献

为了探明促分解菌剂的应用对还田玉米秸秆的促分解效果及对土壤微生物群落结构的影响,选用3组促分解菌剂,编号依次为ND、NK和NS,于2009年10月至2010年4月期间,在河北省农林科学院辛集实验站冬小麦-玉米轮作田对玉米秸秆还田地进行了接种试验。在接种后的15、25、145和160 d分别测定秸秆残重率和秸秆残渣中C/N,结果表明与未施菌剂对照(CK)相比,3组菌剂均在一定程度上加快了玉米秸秆的分解,其中以菌剂ND促分解效果最好,NK次之,NS较差,三者的最高促分解效果分别比CK提高了14.3%、7.7%和1.6%,主要促分解效果都出现在早期(前25 d),且菌剂促进秸秆残渣中C/N降低的效果也在早期明显。采用变性梯度凝胶电泳技术(DGGE)检测菌剂对玉米秸秆降解过程中土壤细菌和真菌群落结构的影响,结果表明,与不接种CK相比,接种菌剂主要在早期对土壤细菌和真菌的群落结构产生较大的影响,而后期对土壤微生物群的影响不明显。秸秆还田后接种促分解菌剂,能在接种早期有效加快秸秆分解,而随接种后时间的推进,其促进效果逐渐减弱,与之对应,土壤微生物群落结构早期差异明显,其后差异逐渐减小。  相似文献

The objective of this study was to evaluate the effect of supplemented condensed tannins (CT) from the bark of the Black Wattle tree (Acacia mearnsii) on production variables and N use efficiency in high yielding dairy cows. A feeding trial with 96 lactating German Holstein cows was conducted for a total of 169 days, divided into four periods. The animals were allotted to two groups (control (CON) and experimental (EXP) group) according to milk yield in previous lactation, days in milk (98), number of lactations and BW. The trial started and finished with a period (period 1 and 4) where both groups received the same ration (total-mixed ration based on grass and maize silage, ensiled sugar beet pulp, lucerne hay, mineral premix and concentrate, calculated for 37 kg energy-corrected milk). In between, the ration of EXP cows was supplemented with 1% (CT1, period 2) and 3% of dry matter (DM) (CT3, period 3) of a commercial A. mearnsii extract (containing 0.203 g CT/g DM) which was mixed into the concentrate. In period 3, samples of urine and faeces were collected from 10 cows of each group and analyzed to estimate N excretion. Except for a tendency for a reduced milk urea concentration with CT1, there was no difference between groups in period 2 (CON v. CT1; P>0.05). The CT3 significantly reduced (P<0.05) milk protein yield, the apparent N efficiency (kg milk N/k feed N) and milk urea concentration; but total milk yield and energy-corrected milk yield were not affected by treatment. Furthermore, as estimated from 10 cows per group and using urinary K as a marker to estimate the daily amount of urine voided, CT3 caused a minor shift of N compounds from urine to faeces, as urea-N in urine was reduced, whereas the N concentration in faeces increased. As an improvement in productivity was not achieved and N use efficiency was decreased by adding the CT product it can be concluded that under current circumstances the use in high yielding dairy cows is not advantageous.  相似文献

Eight multiparous lactating Holstein-Friesian cows were used to evaluate the partitioning of dietary nitrogen (N) from diets based on mixtures of red clover and maize silages in comparison with diets based on ryegrass silage. All cows received 4 kg/day of a standard dairy concentrate with one of four forage treatments in an incomplete changeover design with three 4-week periods. Three treatments were based on mixtures of red clover and maize silage. N intake was altered both by varying the ratio of these silages (40/60 and 25/75 on a dry matter (DM) basis) and by an additional treatment for which the DM intake of the 40/60 mixture was restricted to the level achieved with grass silage. Rumen passage rates were estimated from faecal excretion curves following a pulse oral dose of Dysprosium-labeled silage and urinary excretion of purine derivatives (PD) was used as an index of rumen microbial protein synthesis. Red clover silage mixtures led to significantly increased feed intake (21.5, 20.7 and 15.2 kg DM/day for 40/60 and 25/75 red clover/maize silage mixtures and grass silage, respectively), milk production (25.8, 27.8 and 20.0 kg/day for the same treatments, respectively) and milk component yields, but were without effect on milk fat and protein concentrations. The large increase in the yield of milk (24.5 kg/day) and milk components for the restricted red clover/maize silage treatment, in comparison with the grass silage treatment, was proportionately greater than the increase in DM intake (16.6 kg DM/day). There were no significant treatment effects on diet digestibility, while the higher intakes of red clover silage mixtures were associated with higher rumen passage rates (5.82%, 6.24% and 4.55%/h, respectively). There were significant effects of both N intake and forage source on the partitioning of dietary N between milk and urine. When dietary protein was diluted by the inclusion of maize silage, red clover silage led to increased milk N and reduced urinary N in comparison with grass silage. Improvements in N utilisation may be related to increased dietary starch and/or rumen passage rates leading to increased microbial protein synthesis for these treatments. Urinary excretion of PD was significantly higher for all diets based on mixtures of red clover and maize silages, in comparison with grass silage. Urinary N output was close to literature predictions based on N intake for the diet based on ryegrass silage, but 40 to 80 g/day (25% to 30%) less than predicted for the diets based on the mixtures of red clover and maize silages.  相似文献

Abstract

Biotin is present in nature either free or as biocytin, which is only degraded under the action of a specific enzyme: biotinidase. This enzyme is not included in analytical assays generally used. A method for sample preparation using biotinidase was developed in our laboratory before analysis by ELISA. Three cows equipped with duodenal and ileal cannulae were used to compare the effects of methods of sample preparation on calculations of apparent ruminal synthesis and intestinal absorption of biotin. There was no apparent ruminal synthesis of biotin, no matter whether free or total biotin was measured (p = 0.84). Results also suggested that rumen microbes cannot utilize nor degrade biocytin present in the feed. Estimates of apparent intestinal absorption were influenced by the sample preparation method (p = 0.002). Analysis of free biotin caused an artefact, suggesting intestinal synthesis of this vitamin; whereas determination of total biotin concentrations showed that absorption was taking place in the small intestine.  相似文献

Three experiments were conducted to determine (1) the dose of glutamate needed to alter fermentation and nitrogen (N) partitioning in a continuous culture system, (2) the effect of supplemental glutamate in diets varying in rumen-undegradable protein on fermentation and N partitioning in a continuous culture system, and (3) the effect of dietary supplemental glutamate on the lactational performance of mid-lactation dairy cows, total tract nutrient digestibility, and ruminal microbial N synthesis. In experiment 1, the equivalent of 0, 40, or 80 g of supplemental glutamate per cow per day was added to a basal diet. The dietary treatments were evaluated in a continuous culture system. Glutamate decreased protein digestion and microbial growth while increasing non-ammonia, non-microbial N. Within the doses tested, the equivalent of 80 g glutamate per cow per day most effectively increased non-ammonia, non-microbial N. In experiment 2, dietary treatments consisted of diets formulated to have low rumen-undegradable protein (LRUP; 62 g/kg DM), low rumen-undegradable protein plus the equivalent of 80 g glutamate per cow per day (LRUP + G), and high rumen-undegradable protein [HRUP; 68 g/kg dry matter (DM)]. The dietary treatments were evaluated in a continuous culture system. When added to a diet low in rumen-undegradable protein, glutamate tended to decrease DM and organic matter (OM) digestibility, decreased total volatile fatty acid (VFA) production, increased fermenter pH, increased feed N converted to microbial N, and had no effect on microbial N production. The LRUP + G diet was similar to the HRUP diet and different from the LRUP diet in feed N converted to microbial N and ammonia N concentration. In experiment 3, 40 Holstein cows were utilized in a crossover study to test the effects of two dietary treatments: 0 or 80 g of supplemental glutamate per cow per day. The addition of glutamate to the diet of lactating dairy cows did not improve lactational performance or nutrient digestibility. Based on the results from these in vitro and in vivo experiments, the addition of glutamate to lactating cow diets is not recommended.  相似文献
