Hydrogen gas production from cellulose was investigated using an integrated hydrogen production process consisting of a dark fermentation reactor and microbial fuel cells (MFCs) as power sources for a microbial electrolysis cell (MEC). Two MFCs (each 25 mL) connected in series to an MEC (72 mL) produced a maximum of 0.43 V using fermentation effluent as a feed, achieving a hydrogen production rate from the MEC of 0.48 m3 H2/m3/d (based on the MEC volume), and a yield of 33.2 mmol H2/g COD removed in the MEC. The overall hydrogen production for the integrated system (fermentation, MFC and MEC) was increased by 41% compared with fermentation alone to 14.3 mmol H2/g cellulose, with a total hydrogen production rate of 0.24 m3 H2/m3/d and an overall energy recovery efficiency of 23% (based on cellulose removed) without the need for any external electrical energy input.  相似文献

Vegetable based market waste was evaluated as a fermentable substrate for hydrogen (H2) production with simultaneous stabilization by dark-fermentation process using selectively enriched acidogenic mixed consortia under acidophilic microenvironment. Experiments were performed at different substrate/organic loading conditions in concurrence with two types of feed compositions (with and without pulp). Study depicted the feasibility of H2 production from vegetable waste stabilization process. H2 production was found to be dependent on the concentration of the substrate and composition. Higher H2 production and substrate degradation were observed in experiments performed without pulp (23.96 mmol/day (30.0 kg COD/m3); 13.96 mol/kg CODR (4.8 kg COD/m3)) than with pulp (22.46 mmol/day (32.0 kg COD/m3); 12.24 mol/kg CODR (4.4 kg COD/m3)). Generation of higher concentrations of acetic acid and butyric acid was observed in experiments performed without pulp. Data enveloping analysis (DEA) was employed to study the combined process efficiency of system by integrating H2 production and substrate degradation.  相似文献

In this study, a semi-continuous operation of photo-fermentative H2-producing reactor was attempted at various decanting volume ratios (DVR, decanting volume per day/total working volume, %), ranging 30-70%, using Rhodobacter sphaeroides KD131. H2 production was not efficient with showing low H2 yields of 0.2 and 0.5 mol H2/mol succinateadded at 30% and 40% DVR, respectively. The low performance ascribed to the fact that over 70% of substrate electrons were diverted towards cell growth under these conditions. Meanwhile, cell growth was limited at DVR ? 50%; therefore, higher H2 yields (>2.0 mol H2/mol succinateadded) were observed. Both the highest H2 yield of 3.7 mol H2/mol succinateadded and production rate of 1494 mL H2/L-reactor/d were achieved at 60% DVR. The content of soluble microbial products (SMPs) was measured, which accounted for 3-15% of substrate electrons. It was found that the largest (65-75%) portion of SMPs comprised low molecular-weight (<3 kDa).  相似文献

This study evaluated how different types of industrial wastewaters (bakery, brewery, paper and dairy) affect the performance of identical microbial fuel cells (MFCs); and the microbial composition and electrochemistry of MFC anodes. MFCs fed with paper wastewater produced the highest current density (125 ± 2 mA/m2) at least five times higher than dairy (25 ± 1 mA/m2), brewery and bakery wastewaters (10 ± 1 mA/m2). Such high current production was independent of substrate degradability. A comprehensive study was conducted to determine the factor driving current production when using the paper effluent. The microbial composition of anodic biofilms differed according to the type of wastewater used, and only MFC anodes fed with paper wastewater showed redox activity at −134 ± 5 mV vs NHE. Electrochemical analysis of this redox activity indicated that anodic bacteria produced a putative electron shuttling compound that increased the electron transfer rate through diffusion, and as a result the overall MFC performance.  相似文献

Functional role of biomolecules viz., carbohydrates and proteins on acidogenic biohydrogen (H2) production was studied through the treatment of canteen based composite food waste. The performance was evaluated in an anaerobic sequencing batch reactor (AnSBR) at pH 6 with five variable organic loading conditions (OLR1, 0.854; OLR2, 1.69; OLR3, 3.38; OLR4, 6.54 and OLR5, 9.85 kg COD/m3-day). Experimental data depicted the feasibility of H2 production from the stabilization of food waste and was found to depend on the substrate load. Among the five loading conditions studied, OLR4 documented maximum H2 production (69.95 mmol), while higher substrate degradation (3.99 kg COD/m3-day) was observed with OLR5. Specific hydrogen yield (SHY) vary with the removal of different biomolecules and was found to decrease with increase in the OLR. Maximum SHY was observed with hexose removal at OLR1 (139.24 mol/kg HexoseR at 24 h), followed by pentoses (OLR1, 108.26 mol/kg PentoseR at 48 h), proteins (OLR1, 109.71 mol/kg ProteinR at 48 h) and total carbohydrates (OLR1, 58.31 mol/kg CHOR at 24 h). Proteins present in wastewater helped to maintain the buffering capacity but also enhanced the H2 production by supplying readily available organic nitrogen to the consortia. Along with carbohydrates and proteins, total solids also registered good removal.  相似文献

Ammonium recovery using a two chamber microbial fuel cell (MFC) was investigated at high ammonium concentration. Increasing the ammonium concentration (from 0.07 to 4 g ammonium-nitrogen/L) by addition of ammonium chloride did not affect the performance of the MFC. The obtained current densities by DC-voltammetry were higher than 6 A/m2 for both operated MFCs. Also continuous operation at lower external resistance (250 Ω) showed an increased current density (0.9 A/m2). Effective ammonium recovery can be achieved by migrational ion flux through the cation exchange membrane to the cathode chamber, driven by the electron production from degradation of organic substrate. The charge transport was proportional to the concentration of ions. Nonetheless, a concentration gradient will influence the charge transport. Furthermore, a charge exchange process can influence the charge transport and therefore the recovery of specific ions.  相似文献

Electricity production and modeling of microbial fuel cell (MFC) from continuous beer brewery wastewater was studied in this paper. A single air-cathode MFC was constructed, carbon fiber was used as anode and diluted brewery wastewater (COD = 626.58 mg/L) as substrate. The MFC displayed an open-circuit voltage of 0.578 V and a maximum power density of 9.52 W/m3 (264 mW/m2). Using the model based on polarization curve, various voltage losses were quantified. At current density of 1.79 A/m2, reaction kinetic loss and mass transport loss both achieved to 0.248 V; while ohmic loss was 0.046 V. Results demonstrated that it was feasible and stable for producing bioelectricity from brewery wastewater; while the most important factors which influenced the performance of the MFC are reaction kinetic loss and mass transport loss.  相似文献

The effect of sewage sludge (SWS) addition on the H2 fermentation of food waste (FW) was investigated. It was found that a slight addition of SWS (10:1 = FW:SWS on a COD basis) significantly enhanced the H2 fermentation performance, not only increasing the total amount of H2 produced but accelerating the whole reaction, shortening the lag period, and increasing the H2 production rate. Substrate degradation and microbial germination were also facilitated by SWS addition. A simple calculation reveals that the increased amount of H2 production derived mostly from FW, indicating that SWS addition synergistically enhanced H2 fermentation performance. This was attributed to the existence of Fe and Ca at much higher concentrations in the SWS compared to the FW. The batch process treating a mixture of FW and SWS was repeated and showed an average H2 yield of 2.11 ± 0.20 mol H2/mol hexoseadded, which was 13% higher than that of FW treated alone.  相似文献

Bio (microbial) fuel cell (microbial fuel cell) with Saccharomyces cerevisiae as anodic biocatalyst was evaluated in terms of power generation and substrate degradation at three redox conditions (5.0, 6.0 and 7.0). Fuel cell was operated in single chamber (open-air cathode) configuration without mediators using non-catalyzed graphite as electrodes. The performance was further studied with increasing loading rate (OLRI, 0.91 kg COD/m3-day; OLRII, 1.43 kg COD/m3). Higher current density was observed at pH 6.0 [160.36 mA/m2 (OLRI); 282.83 mA/m2 (OLRII)] than pH 5.0 (137.24 mA/m2) and pH 7.0 (129.25 mA/m2). Bio-electrochemical behavior of fuel cell was evaluated using cyclic voltammetry which showed the presence of redox mediators (NADH/NAD+; FADH/FAD+). Higher electron discharge was observed at pH 6.0, suggesting higher proton shuttling through the involvement of different redox mediators. The application of yeast based fuel cell can be extended to treat high strength wastewaters with simultaneous power generation.  相似文献

The effect of electrodialytic treatment in terms of a current density, pH and Na2H2EDTA addition on the methanogenic activity of copper-amended anaerobic granular sludge taken from the UASB reactor from paper mill was evaluated. Moreover, the specific energy consumption and simplified operational and treatment costs were calculated. Addition of Na2H2EDTA (at pH 7.7) to copper-amended sludge resulted in the highest microbial activity (62 mg CH4-COD g VSS−1 day−1) suggesting that Na2H2EDTA decreased the toxic effects of copper on the methanogenic activity of the anaerobic granular sludge. The highest methane production (159 %) was also observed upon Na2H2EDTA addition and simultaneous electricity application (pH 7.7). The energy consumption during the treatment was 560, 840, 1400 and 1680 kW h m−3 at current densities of 0.23, 0.34, 0.57 and 0.69 mA cm−2, respectively. This corresponded to a treatment costs in terms of electricity expenditure from 39.2 to 117.6 € per cubic meter of sludge.  相似文献

Cellulosic plant and waste materials are potential resources for fermentative hydrogen production. In this study, hydrogen producing, cellulolytic cultures were enriched from compost material at 52, 60 and 70 °C. Highest cellulose degradation and highest H2 yield were 57% and 1.4 mol-H2 mol-hexose−1 (2.4 mol-H2 mol-hexose-degraded−1), respectively, obtained at 52 °C with the heat-treated (80 °C for 20 min) enrichment culture. Heat-treatments as well as the sequential enrichments decreased the diversity of microbial communities. The enrichments contained mainly bacteria from families Thermoanaerobacteriaceae and Clostridiaceae, from which a bacterium closely related to Thermoanaerobium thermosaccharolyticum was mainly responsible for hydrogen production and bacteria closely related to Clostridium cellulosi and Clostridium stercorarium were responsible for cellulose degradation.  相似文献

This work describes the first step towards combination of the bioreactor with a starch-degrading microbial consortium and hydrogenase electrode (HE) in one unit for electricity generation. For this purpose, the bioreactor for microbial fermentation was designed with a set of electrodes (pH-sensor, Ag|AgCl reference electrode, Pt-electrode, and HE) inside the bioreactor. Potentials of all electrodes and H2 accumulation were monitored in the system under the precise pH control. Results obtained with the hydrogen-producing microbial consortium indicated that HE generates the potential equal to the H2|2H+ equilibrium potential. Furthermore, HE was able to catalyze the current generation (200 μA) by consuming H2 gas produced in the microbial consortium from starch. After 220 h of operation, HE retained at least 81% of the initial activity. Calculations of carbon balance indicated that fermentation products were similar in microbial cells without HE and with HE generating the current due to H2 consumption.  相似文献

A two-step anaerobic baffled reactor (ABR-1 and ABR-2) for H2 production from municipal food waste (MFW) was investigated at a temperature of 26 °C. In ABR-1, the average yield of H2 at an HRT of 26 h and OLR of 58 kg COD/m3 d was 250 ml H2/g VS removed. As unexpected; the H2 production in the ABR-2 was further increased up to 370 ml H2/gVS removed at a HRT of 26 h and OLR of 35 kgCOD/m3 d. The total H2 yield in the two-step process was estimated to be 4.9 mol H2/mol hexose. The major part of H2 production in the ABR-1 was due to the conversion of CODparticulate (36%). In the ABR-2 the H2 yield was mainly due to the conversion of COD in the soluble form (76%). Based on these results MFW could be ideal substrate for H2 production in a two-step ABR processes.  相似文献

Ethylene–vinyl acetate (EVA) copolymer was used to immobilize H2-producing sewage sludge for H2 production in a three-phase fluidized bed reactor (FBR). The FBR with an immobilized cell packing ratio of 10% (v/v) and a liquid recycle rate of 5 l/min (23% bed expansion) was optimal for dark H2 fermentation. The performance of the FBR reactor fed with sucrose-based synthetic medium was examined under various sucrose concentration (Cso) and hydraulic retention time (HRT). The best volumetric H2 production rate of 1.80 ± 0.02 H2 l/h/l occurred at Cso = 40 g COD/l and 2 h HRT, while the optimal H2 yield (4.26 ± 0.04 mol H2/mol sucrose) was obtained at Cso = 20 g COD/l and 6 h HRT. The H2 content in the biogas was stably maintained at 40% or above. The primary soluble metabolites were butyric acid and acetic acid, as both products together accounted for 74–83% of total soluble microbial products formed during dark H2 fermentation.  相似文献

To investigate the potential role of microbial community composition in soil carbon and nitrogen cycling, we transplanted soil cores between a grassland and a conifer ecosystem in the Sierra Nevada California and measured soil process rates (N-mineralization, nitrous oxide and carbondioxide flux, nitrification potential), soil water and temperature, and microbial community parameters (PLFA and substrate utilization profiles) over a 2 year period. Our goal was to assess whether microbial community composition could be related to soil process rates independent of soil temperature and water content. We performed multiple regression analyses using microbial community parameters and soil water and temperature as X-variables and soil process rates and inorganic N concentrations as Y-variables. We found that field soil temperature had the strongest relationship with CO2 production and soil NH4+ concentration, while microbial community characteristics correlated with N2O production, nitrification potential, gross N-mineralization, and soil NO3 concentration, independent of environmentalcontrollers. We observed a relationship between specific components of the microbial community (as determined by PLFA) and soil processes,particularly processes tightly linked to microbial phylogeny (e.g. nitrification). The most apparent change in microbial community composition in response to the 2 year transplant was a change in relative abundance of fungi (there was only one significant change in PLFA biomarkers for bacteria during 2 years). The relationship between microbial community composition and soil processes suggests that prediction of ecosystem response to environmental change may be improved by recognizing and accounting for changes in microbial community composition and physiological ecology.  相似文献

The effects of light/dark cycle, mixing pattern and partial pressure of H2 on the growth and hydrogen production of Rhodobacter sphaeroides ZX-5 were investigated. The results from light/dark cycle culture showed that little or no hydrogen production was observed during the dark periods, and the hydrogen production immediately recovered once illumination was resumed. Also, it was found that the optimum condition of shaking velocity was 120 rpm for hydrogen photo-fermentation. Meanwhile, shaking during H2 production phase (i.e., cell growth stationary phase) of photo-fermentation played a crucial role on effectively enhancing the phototrophic hydrogen production, rather than that during cell exponential growth phase. The other factor evaluated was hydrogen partial pressure in the culture system. The substrate conversion efficiency increased from 86.07% to 95.56% along with the decrease of the total pressure in the photobioreactor from 1.082 × 105 to 0.944 × 105 Pa, which indicated that reduction of H2 partial pressure by lowering the operating pressure substantially improved H2 production in an anaerobic, photo-fermentation process.  相似文献

Copper(II) complexes [Cu(satp)(L)] (1-3) of a Schiff base thiolate (salicylidene-2-aminothiophenol, H2satp) and phenanthroline bases (L), viz. 1,10-phenanthroline (phen in 1), dipyrido[3,2-d:2′,3′-f]quinoxaline (dpq in 2) and dipyrido[3,2-a:2′,3′-c]phenazine (dppz in 3), were prepared, characterized and their anaerobic DNA photocleavage activity and hypoxic photocytotoxicity studied. The redox active complexes show the Cu(II)-Cu(I) couple near − 0.5 V for 1 and near 0.0 V vs. SCE (saturated calomel electrode) for 2 and 3. The one-electron paramagnetic complexes (~ 1.85 μB) are avid DNA binders giving Kb values within 1.0 × 105 − 8.0 × 105 M− 1. Thermal melting and viscosity data along with molecular docking calculations suggest DNA groove and/or partial intercalative binding of the complexes. The complexes show anaerobic DNA cleavage activity in red light under argon via type-I pathway, while DNA photocleavage in air proceeds via hydroxyl radical pathway. The DFT (density functional theory) calculations reveal a thyil radical pathway for the anaerobic DNA photocleavage activity and suggest the possibility of generation of a transient copper(I) species due to bond breakage between the copper and sulfur to generate the thyil radical. An oxidation of the copper(I) species is likely by oxygen in an aerobic medium or by the buffer medium in an anaerobic condition. Complex 3 exhibits significant photocytotoxicity in HeLa cells (IC50 = 8.3(± 1.0) μM) in visible light, while showing lower dark toxicity (IC50 = 17.2(± 1.0) μM). A significant reduction in the dark toxicity is observed under hypoxic cellular conditions (IC50 = 30.0(± 1.0) μM in dark), while retaining its photocytotoxicity (IC50 = 8.0(± 1.0) μM).  相似文献

Homo-acetogens in the anode of a microbial electrolysis cell (MEC) fed with H2 as sole electron donor allowed current densities similar to acetate-fed biofilm anodes (∼10 A/m2). Evidence for homo-acetogens included accumulation of acetate at high concentrations (up to 18 mM) in the anode compartment; detection of formate, a known intermediate during reductive acetogenesis by the acetyl-CoA pathway; and detection of formyl tetrahydrofolate synthetase (FTHFS) genes by quantitative real-time PCR. Current production and acetate accumulation increased in parallel in batch and continuous mode, while both values decreased simultaneously at short hydraulic retention times (1 h) in the anode compartment, which limited suspended homo-acetogens. Acetate produced by homo-acetogens accounted for about 88% of the current density of 10 A/m2, but the current density was sustained at 4 A/m2 at short hydraulic retention time because of a robust partnership of homo-acetogens and anode respiring bacteria (ARB) in the biofilm anode.  相似文献

ABPS-1, a new water-soluble polysaccharide with molecular weight of 26 kDa and a specific optical rotation of +170° (c 1.0, H2O), was extracted from the roots of Acanthophyllum bracteatum by warm water and further successively purified through DEAE-cellulose A52 and Sephadex G-100 columns. Monosaccharide analysis revealed that the ABPS-1 was composed of Glc, Gal and Ara with a relative molar ratio of 1.4:5.2:1.0. Its structural features were elucidated by a combination of FT-IR, methylation and GC-MS analysis, periodate oxidation and Smith degradation, partial acid hydrolysis and 13C and 1H NMR spectroscopy. The data obtained indicate that ABPS-1 possessed a backbone of α-(1 → 6)-linked Gal with branches attached to O-2 by α-1 → linked Glc and at O-3 by α-1 → linked Gal and by α-(1 → 3)-linked Ara. The in vitro antioxidant activity showed that ABPS-1 possesses DPPH radical-scavenging activity in a concentration-dependent manner with an EC50 value of 2.6 mg/ml.  相似文献
